1. Study on characteristics of ferrate coupled peracetic acid for advanced treatment of metronidazole in water

Abstract

Metronidazole(MNZ), prone to accumulate in the environment, is a nitroimidazole antibiotic and its metabolites have potential carcinogenicity and mutagenicity. In this paper, the degradation characteristics of MNZ by coupled ferrate/peroxyacetic acid〔Fe(Ⅵ)/PAA〕technology, as well as the simulation of MNZ degradation and reaction mechanism, were investigated. When the dosages of Fe(Ⅵ) as 500 μmol/L and PAA as 100 μmol/L, the degradation of MNZ reached 94.85% at pH of 8 in 60 min, and the degradation process followed pseudo-first-order kinetics. Variable experiments showed that the degradation of MNZ was positively correlated with the dosage of Fe(Ⅵ) and PAA, and the best effect was achieved at pH=8. Chloride (Cl-) and nitrate (NO3-) in the water matrix had little effect on the degradation of MNZ. Ammonium salt (NH4+) could promote the degradation of MNZ,while bicarbonate (HCO3-) and humic acid (HA) inhibited the degradation. The removal of MNZ in simulated medical wastewater by Fe(Ⅳ)/PAA was similar to that in ultra-pure water, which could achieve better removal effect. Probe experiments and quenching experiments showed that PAA accelerated the removal of MNZ from active species. H2O2 coexisting with PAA was the main activator of Fe(Ⅵ) while the main active species of Fe(Ⅵ)/PAA system were Fe(V) and Fe(Ⅳ). The results were expected to provide theoretical reference and data support for the application of Fe(Ⅵ)/PAA system in the deep treatment of MNZ in medical wastewater.

Abstract

The continuously increasing bank sizes have received much attention from both policymakers and researchers. We present new estimates of returns to scale in cost, revenue, and profit for European banks over 2000–2020 based on nonparametric, local‐linear methods. We find that most banks faced increasing returns to scale in cost and decreasing returns to scale in revenue and profit across years, across European countries, and across bank size quartiles. Our results suggest that restricting the size of banks may not bring too much loss of revenue and profit, but could prevent European banks from exploiting increasing returns to scale in cost. Abstract

Summary: Hexaploid wheat (Triticum aestivum), a major staple crop, has a remarkably large genome of ~14.4 Gb (containing 106 913 high‐confidence [HC] and 159 840 low‐confidence [LC] genes in the Chinese Spring v2.1 reference genome), which poses a major challenge for functional genomics studies. To overcome this hurdle, we performed whole‐exome sequencing to generate a nearly saturated wheat mutant database containing 18 025 209 mutations induced by ethyl methanesulfonate (EMS), carbon (C)‐ion beams, or γ‐ray mutagenesis. This database contains an average of 47.1 mutations per kb in each gene‐coding sequence: the potential functional mutations were predicted to cover 96.7% of HC genes and 70.5% of LC genes. Comparative analysis of mutations induced by EMS, γ‐rays, or C‐ion beam irradiation revealed that γ‐ray and C‐ion beam mutagenesis induced a more diverse array of variations than EMS, including large‐fragment deletions, small insertions/deletions, and various non‐synonymous single nucleotide polymorphisms. As a test case, we combined mutation analysis with phenotypic screening and rapidly mapped the candidate gene responsible for the phenotype of a yellow‐green leaf mutant to a 2.8‐Mb chromosomal region. Furthermore, a proof‐of‐concept reverse genetics study revealed that mutations in gibberellic acid biosynthesis and signalling genes could be associated with negative impacts on plant height. Finally, we built a publically available database of these mutations with the corresponding germplasm (seed stock) repository to facilitate advanced functional genomics studies in wheat for the broad plant research community. Abstract

A simple yet easy to implement method is proposed to further improve the finite sample approximation of the recently developed central limit theorems for aggregates of envelopment estimators. Focusing on the simple mean efficiency, we propose using the bias-corrected individual efficiency estimate to improve the variance estimator. The extensive Monte-Carlo experiments confirm that, for relatively small sample sizes (≤ 100), with both low dimensions and especially for high dimensions, our new method combined with the data sharpening method generally provides better ‘coverage’ (of the true values by the estimated confidence intervals) than the previously developed approaches.

Abstract

We expand the recently developed framework for the inference for aggregate efficiency, by extending the existing theory and providing guidelines for practitioners. In particular, we develop the central limit theorems (CLTs) for aggregate input-oriented efficiency, analogous to the output-oriented framework established by Simar and Zelenyuk (2018). To further improve the finite sample performance of the developed CLTs, we propose a simple yet easy to implement method through using the bias- corrected individual efficiency estimate to improve the variance estimator. The extensive Monte-Carlo experiments confirmed the developed CLTs for aggregate input- oriented efficiency and also confirmed the better performance of our proposed method in the finite sample sizes. Finally, we use two well-known empirical data sets to illustrate the differences between all the existing methods to facilitate the use by practitioners.

Abstract

Plant height is a key agronomic trait that is closely to the plant morphology and lodging resistance in wheat. However, at present, the few dwarf genes widely used in wheat breeding have narrowed wheat genetic diversity. In this study, we selected a semi-dwarf wheat mutant dwarf33 that exhibits decreased plant height with little serious negative impact on other agronomic traits. Genetic analysis and mutant gene mapping indicated that dwarf33 contains a new recessive semi-dwarf gene Rht-SN33d, which was mapped into ~1.3 Mb interval on the 3DL chromosome. The gibberellin metabolism-related gene TraesCS3D02G542800, which encodes gibberellin 2-beta-dioxygenase, is considered a potential candidate gene of Rht-SN33d. Rht-SN33d reduced plant height by approximately 22.4% in mutant dwarf33. Further study revealed that shorter stem cell length may be the main factor causing plant height decrease. In addition, the coleoptile length of dwarf33 was just 9.3% shorter than that of wild-type Shaannong33. 42. Fine mapping of two recessive genes TaFLA1and TaSPL8controlling flag leaf angle in bread wheat

Abstract

Flag leaf angle is one of the key target traits in high yield wheat breeding. A smaller flag leaf angle reduces shading and enables plants to grow at a higher density, which increases yield. Here we identified a mutant, je0407, with an 84.34%–89.35% smaller flag leaf angle compared with the wild type. The mutant also had an abnormal lamina joint and no ligule or auricle. Genetic analysis indicated that the ligule was controlled by two recessive genes, which were mapped to chromosomes 2AS and 2DL. The mutant allele on chromosome 2AS was named Tafla1b, and it was fine mapped to a 1 Mb physical interval. The mutant allele on chr. 2DL was identified as Taspl8b, a novel allele of TaSPL8with a missense mutation in the second exon, which was used to develop a cleaved amplified polymorphic sequence marker. F3and F4lines derived from crosses between Jing411 and je0407were genotyped to investigate interactions between the Tafla1band Taspl8balleles. Plants with the Tafla1b/Taspl8agenotype had 58.41%–82.76% smaller flag leaf angles, 6.4%–24.9% shorter spikes, and a greater spikelet density (0.382 more spikelets per cm) compared with the wild type. Plants with the Tafla1a/Taspl8bgenotype had 52.62%–82.24% smaller flag leaf angles and no differences in plant height or spikelet density compared with the wild type. Tafla1b/Taspl8bplants produced erect leaves with an abnormal lamina joint. The two alleles had dosage effects on ligule formation and flag leaf angle, but no significant effect on thousand-grain weight. The mutant alleles provide novel resources for improvement of wheat plant architecture.

Abstract

Induced mutation is an important approach for creating novel plant germplasms. The introduction of dwarf or semi-dwarf genes into wheat has led to great advancements in yield improvement. In this study, four elite dwarf wheat mutants, named dm1 – dm4 , induced from γ-ray irradiation or ethyl methanesulfonate (EMS) mutagenesis, were used to identify transcriptome variations and dwarfing mechanisms. The results showed that the hotspot regions of mutations distributed on the chromosomes were consistent among the four mutant lines and these regions were mainly located around the 50, 360 and 400 Mb positions of chromosome 1A and the distal regions of chromosomes 2A and 2BL. Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses suggested that 'protein processing in endoplasmic reticulum' was the most common significantly enriched pathway based on the differentially expressed genes (DEGs) between wildtype (WT) and the mutants. Notably, 18 out of 20 genes involved in this process encode heat shock proteins (HSPs). The results implied that HSPs might participate in wheat dwarfism response and function in the dwarfism process through protein folding and/or degradation. Moreover, seven genes in dm4 involved in modulating auxin levels were down-regulated and dm4 was more sensitive to auxin treatment compared with WT, indicating the important roles of auxin in regulation of dwarf phenotype in dm4. This study not only identified transcriptome sequence variation induced by physical and chemical mutagenesis but also revealed potential dwarfing mechanisms in the wheat mutant lines.
