Abstract

One of the most serious public health problems in Peru and worldwide is Tuberculosis (TB), which is produced by a bacterium known as Mycobacterium tuberculosis. The purpose of this work is to facilitate and automate the diagnosis of tuberculosis using the MODS method and using lens-free microscopy, as it is easier to calibrate and easier to use by untrained personnel compared to lens microscopy. Therefore, we employed a U-Net network on our collected data set to perform automatic segmentation of cord shape bacterial accumulation and then predict tuberculosis. Abstract

Early diagnosis of autism spectrum disorder (ASD) is known to improve the quality of life of affected individuals. However, diagnosis is often delayed even in wealthier countries including the US, largely due to the fact that gold standard diagnostic tools such as the Autism Diagnostic Observation Schedule (ADOS) and the Autism Diagnostic Interview-Revised (ADI-R) are time consuming and require expertise to administer. This trend is even more pronounced lower resources settings due to a lack of trained experts. As a result, alternative, less technical methods that leverage the unique ways in which children with ASD react to visual stimulation in a controlled environment have been developed to help facilitate early diagnosis. Previous studies have shown that, when exposed to a video that presents both social and abstract scenes side by side, a child with ASD will focus their attention towards the abstract images on the screen to a greater extent than a child without ASD. Such differential responses make it possible to implement an algorithm for the rapid diagnosis of ASD based on eye tracking against different visual stimuli. Here we propose a convolutional neural network (CNN) algorithm for gaze prediction using images extracted from a one-minute stimulus video. Our model achieved a high accuracy rate and robustness for prediction of gaze direction with independent persons and employing a different camera than the one used during testing. In addition to this, the proposed algorithm achieves a fast response time, providing a near real-time evaluation of ASD. Autism is a developmental disorder that affects social interaction and communication of children. The gold standard diagnostic tools are very difficult to use and time consuming. However, diagnostic could be deduced from child gaze preferences by looking a video with social and abstract scenes. Abstract

Pyrazinamide (PZA) is a key drug in the treatment of Mycobacterium tuberculosis. Although not completely understood yet, the bactericidal mechanism of PZA starts with its diffusion into the cell and subsequent conversion into pyrazinoic acid (POA) after the hydrolysis of ammonia group. This leads to the acidification cycle, which involves: (1) POA extrusion into the extracellular environment, (2) reentry of protonated POA, and (3) release of a proton into the cytoplasm, resulting in acidification of the cytoplasm and accumulation of intracellular POA. To better understand this process, we developed a system of coupled non-linear differential equations, which successfully recapitulates the kinetics of PZA/POA observed in M. tuberculosis. The parametric space was explored, assessing the impact of different PZA and pH concentrations and variations in the kinetic parameters, finding scenarios of PZA susceptibility and resistance. Furthermore, our predictions show that the acidification cycle alone is not enough to result in significant intracellular accumulation of POA in experimental time scales when compared to other neutral pH scenarios. Thus, revealing the need of novel hypotheses and experimental evidence to determine the missing mechanisms that may explain the pH-dependent intracellular accumulation of POA and their subsequent effects. Abstract

Anemia is a common problem that affects a significant part of the world's population, especially in impoverished countries. This work aims to improve the accessibility of remote diagnostic tools for underserved populations. Our proposal involves implementing algorithms to estimate hemoglobin levels using images of the eyelid conjunctiva and a calibration label captured with a mid-range cell phone. We propose three algorithms: one for calibration label segmentation, another for palpebral conjunctiva segmentation, and the last one for estimating hemoglobin levels based on the segmented images from the previous algorithms. Experiments were performed using a data set of children's eyelid images and calibration stickers. An L1 norm error of 0.72 g/dL was achieved using the SLIC-GAT model to estimate the hemoglobin level. In conclusion, the integration of these segmentation and regression methods improved the estimation accuracy compared to current approaches, considering that the source of the images was a mid-range commercial camera. The proposed method has the potential for mass screening in low-income rural populations as it is non-invasive, and its simplicity makes it feasible for community health workers with basic training to perform the test. Abstract

Within the framework of the current COVID-19 pandemic, there is a race against time to find therapies for the outbreak to be controlled. Since vaccines are still tedious to develop and partially available for low-income countries, passive immunity based on egg-yolk antibodies (IgY) is presented as a suitable approach to preclude potential death of infected patients, based on its high specificity/avidity/production yield, cost-effective manufacture, and ease of administration. In the present study, IgY antibodies against a recombinant RBD protein of SARS-CoV-2 were produced in specific-pathogen-free chickens and purified from eggs using a biocompatible method. In vitro immunoreactivity was tested, finding high recognition and neutralization values. Safety was also demonstrated prior to efficacy evaluation, in which body weight, kinematics, and histopathological assessments of hamsters challenged with SARS-CoV-2 were performed, showing a protective effect administering IgY intranasally both as a prophylactic treatment or a post-infection treatment. The results of this study showed that intranasally delivered IgY has the potential to both aid in prevention and in overcoming COVID-19 infection, which should be very useful to control the advance of the current pandemic and the associated mortality. La resistencia a PZA (PZAr) está presente en la mitad de las cepas resistentes a fármacos; sin embargo, PZAr no se prueba rutinariamente debido a la baja confiabilidad de las pruebas de susceptibilidad.
