Aim: This study aimed to assess the shaping ability of Reciproc Blue in the apical third and apical foramen of moderately curved canals at different working lengths (WLs), by micro-computed tomography. Methods: Thirty-six mesial roots (mesiobuccal and mesiolingual canals) were included, each with 2 separate root canals and independent apical foramina, according to type IV of Vertucci's classification of first and second mandibular molars. The canals were instrumented at three different WLs: G-1, 1mm short of the major apical foramen; G0, at the major apical foramen; G+1, 1mm beyond the major apical foramen. The groups were assessed for changes in root canal volume and untouched wall area in the apical third. Groups G0 and G+1 were also compared for percentage of untouched walls at the apical foramen. One-way ANOVA (post hoc Tukey test) and Student's t-test adopted a 5% level of significance. Results: Root canal volumes (mm³) in the apical third were 22.86±10.46, 44.48±24.91, and 55.71±21.32 in G-1, G0 and G+1, respectively. G-1 volume following instrumentation increased significantly less than that of G0 or G+1 (P>.05); G0 did not differ from G+1. The percentage of untouched wall area in the apical third did not differ among the three groups (P>.05). G0 and G+1 did not differ regarding untouched walls in the major apical foramem walls. Conclusion: Extending the WL from 1mm short of the apical foramen to a point at and beyond the WL increases the apical third volume without increasing the prepared area. Pilosocereus gounellei A. Weber ex K. Schum. Bly. Ex Rowl (xique-xique) is a cactus native to the Brazilian Caatinga biome and considered a non-conventional food plant. This study evaluated the physicochemical characteristics, antioxidant capacity, and impacts on the composition and metabolic activity of human intestinal microbiota of freeze-dried P. gounellei cladodes (FDPg) during 48 h of in vitro colonic fermentation. FDPg had a high nutritional value, the presence of bioactive compounds, and antioxidant capacity. The colonic fermentation of FDPg decreased the Firmicutes/Bacteroidetes ratio and increased bacterial diversity. It also increased the relative abundance of bacterial groups considered beneficial, such as Mitsuokella , Bacteroides , Megasphaera , and Acidaminococcus , and decreased the relative abundance of bacterial groups considered non-beneficial, such as Succinivibrio , Citrobacter , Escherichia-Shigella , Faecalibacterium , and Blautia. The colonic fermentation of FDPg induced the production of various metabolites related to host health promotion. FDOd had high levels of total fiber (52.36 g/100 g), especially soluble fiber (33.36 g/100 g), as well as minerals, such as potassium (4415.49 mg/100 g), calcium (1593.25 mg/100 g), magnesium (853.55 mg/100 g), and phosphorus (209.04 mg/100 g), high total chlorophyll content (912.00 mg/100 g), presence of various phenolic compounds, such as catechin (41.00 mg/100 g), myricetin (785.00 mg/100 g), isorhamnetin (5.00 mg/100 g), rutin (42 mg/100 g), and antioxidant activity (ABTS•+: 0.51 µmol TEAC/g; DPPH•: 0.32 µmol TEAC/g; FRAP: 2.75 µmol FeSO4/g). Fermentation of FDOd using human fecal inoculum changed the composition and metabolic activity of intestinal microbiota, increasing the relative abundance of Ligilactobacillus (0.03 %–16.44 %), Lactiplantibacillus (0.00 %–1.11 %), and Agathobacter (1.80 %–4.23 %), and decreasing the relative abundance of Prevotella_9 (25.08 %–0.15 %) and Succinivibrio (24.77 %–1.54 %). FDOd influenced the metabolic profile of the intestinal microbiota with the production of various metabolites, including short- and medium-chain fatty acids, organic acids, essential amino acids, and other compound classes with health-promoting properties. The purpose of this study was to determine differences in the neuromechanical properties of knee extensors and vertical jump height between pre-pubertal and pubertal children, as well as to investigate the influence of these properties on the performance of the children's squat jump (SJ). The sample consisted of 13 pre-pubertal and 13 pubertal children of both sexes. The vertical SJ, a maximal isometric voluntary contraction (MVC) of the knee extensors, concomitant with the electromyographic activity (EA) of the vastus lateralis (VL), rectus femoris, vastus medialis (VM), and biceps femoris were collected. We compared the neural-mechanical variables of the CVM as Maximal Force (FMax), EA, Rate of Force Development (RFD), Force Max (FMAX), Rise of Electromyographic Rate (RER), Electromechanical Delay (EMD), and Co-activation. The findings showed that during the MVC of the knee extensors of the pubertal children versus the pre-pubertal children, they presented the following characteristics: (a) higher muscle capacity EA, lower EMD, higher RER, and similar co-activation; and (b) higher RFD at the beginning of MVC. Regarding the performance in the SJ, the pubertal children presented higher performance than the pre-pubertal children. In addition, linear regression identified an influence of the EMD of the VL and VM muscles on the performance of the SJ. [ABSTRACT FROM AUTHOR]

Objective: The study's purpose has been to delineate the clinical-epidemiological profile of patients undergoing cardiac catheterization; furthermore, to propose a management technology to create a database with information of epidemiological relevance. Methods: It is a retrospective study with a quantitative approach, which considers the databases and medical records of 1,890 patients who underwent cardiac catheterization at a hemodynamic unit from April 2014 to April 2016. Microsoft Office Excel ® software was used to both organize and analyze the data. CAAE No. 55615616.0.0000.5282. Results: The average age was 61.45 years old. The majority of the assisted population is indicated by the National Regulation System (63%) with the following distribution: (52.86%) male and (47.14%) female. It was identified that 79.5% of the users have high blood pressure. It was found that the current strategy for monitoring the assisted users shows information deficiencies. Conclusion: The nursing consultation contributes to the improvement of the Sistema Único de Saúde (SUS) [Unified Health System] when capable of obtaining both relevant and determinant information for the professional nursing performance. Evidence shows that physical exercise carried out systematically promotes numerous health benefits, including improvements in glucose homeostasis. Of all types of physical activity, resistance training is effective in promoting increased strength, speed, power, hypertrophy, and metabolic effects such as increased insulin sensitivity and control in glycogen metabolism. Since there are still gaps requiring attention in regards to the molecular mechanisms leading to effective insulin signaling, this paper attempts to review the effects and implications of resistance exercise on the major intracellular protein activities. In this regard, we set out to understand the role of resistance training on IRS1, IRS2, PI3K, Akt/PKB, and GLUT4 activity, which can lead to the development of possible future interventions to improve physical performance and mitigate the effects of altered metabolic conditions such as obesity, insulin resistance, and type 2 diabetes mellitus. Resumo Justificativa e objetivos A dor crônica persistente pós‐operatória (DCPO) constitui um grave problema de saúde, incapacitante, mina a qualidade de vida dos pacientes acometidos. Apesar de mais estudos e pesquisas terem sido desenvolvidos a respeito dos possíveis mecanismos da evolução da dor aguda para dor crônica pós‐operatória, ainda não existem dados consistentes a respeito de seus fatores de risco e prevenção. Este artigo se propõe a trazer o que há no panorama da literatura atual disponível. Conteúdo Esta revisão descreve a definição, os fatores de risco e os mecanismos da DCPO, sua prevenção e seus tratamentos. Os principais medicamentos e técnicas são expostos de forma compreensiva. Conclusão A dor crônica persistente pós‐operatória é uma entidade complexa e de etiologia ainda não esclarecida, que interfere intensamente na vida do sujeito. A dor neuropática decorrente do trauma cirúrgico ainda é a expressão mais comum dessa entidade. Técnicas que evitem a lesão de nervos estão recomendadas e devem ser usadas sempre que possível. Apesar dos esforços para entender e selecionar os pacientes de risco, o manuseio e a prevenção dessa síndrome continuam desafiantes e inapropriados. Background and objectives Postoperative persistent chronic pain (POCP) is a serious health problem, disabling, undermining the quality of life of affected patients. Although more studies and research have addressed the possible mechanisms of the evolution from acute pain to chronic postoperatively, there are still no consistent data about the risk factors and prevention. This article aims to bring what is in the panorama of the current literature available. Content This review describes the definition, risk factors, and mechanisms of POCD, its prevention and treatment. The main drugs and techniques are exposed comprehensively. Conclusion Postoperative persistent chronic pain is a complex and still unclear etiology entity, which interferes heavily in the life of the subject. Neuropathic pain resulting from surgical trauma is still the most common expression of this entity. Techniques to prevent nerve injury are recommended and should be used whenever possible. Dehydroepiandrosterone (DHEA) and the dehydroepiandrosterone sulfate (DHEA-S) are steroids produced mainly by the adrenal cortex. There is evidence from both human and animal models suggesting beneficial effects of these steroids for obesity, diabetes mellitus, hypertension, and osteoporosis, conditions associated with the post-menopausal period. Accordingly, we hypothesized that DHEA supplementation in ovariectomized (OVX) female rats fed a high-fat diet would maintain glucose-induced insulin secretion (GSIS) and pancreatic islet function. OVX resulted in a 30% enlargement of the pancreatic islets area compared to the control rats, which was accompanied by a 50% reduction in the phosphorylation of AKT protein in the pancreatic islets. However, a short-term high-fat diet induced insulin resistance, accompanied by impaired GSIS in isolated pancreatic islets. These effects were reversed by DHEA treatment, with improved insulin sensitivity to levels similar to the control group, and with increased serine phosphorylation of the AKT protein. These data confirm the protective effect of DHEA on the endocrine pancreas in a situation of diet-induced overweight and low estrogen concentrations, a phenotype similar to that of the post-menopausal period. Background/objectives: This study investigates the metabolic profile of a single dose of etodolac in healthy volunteers, focusing on pharmacokinetics, clinical parameters, and metabolomic variations to identify biomarkers and pathways linked to drug response, efficacy, and safety., Methods: Thirty-seven healthy volunteers, enrolled after rigorous health assessments, received a single dose of etodolac (Flancox ® 500 mg). Pharmacokinetic profiles were determined using tandem mass spectrometry analysis, and the metabolomic profiling was conducted using baseline samples (pre-dose) and samples at maximum drug concentration (post-dose) via liquid chromatography coupled with a quadrupole time-of-flight mass spectrometer. Network analysis was employed to interpret the data., Results: Correlations were observed between metabolomic profiles and pharmacokinetic parameters as well as clinical characteristics. Notably, metabolites derived from arachidonic acid, such as prostaglandins and leukotrienes, were linked to etodolac's pharmacokinetics. Other metabolites involved in pathways like cholesterol biosynthesis, bile salts, riboflavin, and retinoic acid signaling were correlated with hematological and liver function parameters. These findings are consistent with the infrequent adverse events reported by participants, including hematological and biochemical changes in liver function., Conclusions: A set of metabolites was identified in possible associations between specific pathways and unusual side effects, comparing the metabolic profiles before and after doses of etodolac. Our results highlight the importance of optimizing drug therapy and minimizing adverse events by taking into account individual metabolic profile information.

Kaempferol (KPF) is a flavonoid antioxidant found in fruits and vegetables. Many studies have described the beneficial effects of dietary KPF in reducing the risk of chronic diseases, especially cancer. Nevertheless, little is known about the cellular and molecular mechanisms underlying KPF actions in the central nervous system (CNS). Also, the relationship between KPF structural properties and their glycosylation and the biological benefits of these compounds is unclear. The aim of this study was to review studies published in the PubMed database during the last 10 years (2010-2020), considering only experimental articles that addressed the isolated cell effect of KPF (C 15 H 10 O 6 ) and its derivatives in neurological diseases such as Alzheimer's disease, Parkinson, ischemia stroke, epilepsy, major depressive disorder, anxiety disorders, neuropathic pain, and glioblastoma. 27 publications were included in the present review, which presented recent advances in the effects of KPF on the nervous system. KPF has presented a multipotential neuroprotective action through the modulation of several proinflammatory signaling pathways such as the nuclear factor kappa B (NF-kB), p38 mitogen-activated protein kinases (p38MAPK), serine/threonine kinase (AKT), and β-catenin cascade. In addition, there are different biological benefits and pharmacokinetic behaviors between KPF aglycone and its glycosides. The antioxidant nature of KPF was observed in all neurological diseases through MMP2, MMP3, and MMP9 metalloproteinase inhibition; reactive oxygen species generation inhibition; endogenous antioxidants modulation as superoxide dismutase and glutathione; formation and aggregation of beta-amyloid (β-A) protein inhibition; and brain protective action through the modulation of brain-derived neurotrophic factor (BDNF), important for neural plasticity. In conclusion, we suggest that KPF and some glycosylated derivatives (KPF-3-O-rhamnoside, KPF-3-O-glucoside, KPF-7-O-rutinoside, and KPF-4'-methyl ether) have a multipotential neuroprotective action in CNS diseases, and further studies may make the KPF effect mechanisms in those pathologies clearer. Matricaria chamomilla L. contains antioxidant flavonoids that can have their bioactivity enhanced by enzymatic hydrolysis of specific glycosyl groups. This study implements an untargeted metabolomics approach based on ultra-performance liquid chromatography coupled with electrospray ionisation quadrupole time-of-flight mass spectrometry technique operating in MS E mode (UPLC-QTOF-MS E ) and spectrophotometric analysis of chamomile aqueous infusions, before and after hydrolysis by hesperidinase and β-galactosidase. Several phenolic compounds were altered in the enzymatically treated infusion, with the majority being flavonoid derivatives of apigenin, esculetin, and quercetin. Although enzymatically modifying the infusion only led to a small increase in antioxidant activity (DPPH • method), its inhibitory effect on pancreatic lipase was of particular interest. The enzymatically treated infusion exhibited a greater inhibitory effect (EC 50 of 35.6 µM) than unmodified infusion and kinetic analysis suggested mixed inhibition of pancreatic lipase. These results are of great relevance due to the potential of enzymatically treated functional foods in human health.

Background: Sepsis remains the primary cause of death from infection, despite advances in modern medicine. The identification of reliable diagnostic biomarkers for the early detection of this disease is critical and may reduce the mortality rate as it could allow early treatment. The purpose of this study was to describe the changes in the plasma and red cells blood lipidome profiling of patients diagnosed with sepsis and septic shock with the aim to identify potentially useful metabolic markers., Methods: Lipids from plasma and erythrocytes from septic patients (n = 20) and healthy controls (n = 20) were evaluated by electrospray ionization quadrupole time-of-flight mass spectrometry, and the fatty acid composition of the phospholipids fraction of erythrocytes was determined by gas chromatography. The data were treated with multivariate data analysis, including principal component analysis and (orthogonal) partial least squares discriminant analysis., Results: Potential biomarkers including lysophosphatidylcholines (lyso-PCs) and sphingomyelin (SMs) with specific fatty acid chains were identified. Both Lyso-PCs and SMs were downregulated, whereas the saturated and unsaturated phosphatidylcholines (PCs) were upregulated in the plasma and erythrocytes of septic patients.
