Background. Inducible nitric oxide synthase (iNOS) plays a crucial role in neuroinflammation, especially microglial activity, and may potentially represent a useful biomarker of neuroinflammation. In this study, we carefully defined a strategic plan to develop iNOS-targeted molecular PET imaging using (4 ′ -amino-5 ′ ,8 ′ -difluoro-1 ′ H-spiro[piperidine-4,2 ′ -quinazolin]-1-yl)(4-fluorophenyl)methanone ([18F]FBAT) as a tracer in a mouse model of lipopolysaccharide- (LPS-) induced brain inflammation. Methods. An in vitro model, murine microglial BV2 cell line, was used to assess the uptake of [18F]FBAT in response to iNOS induction at the cellular level. In vivo whole-body dynamic PET/MR imaging was acquired in LPS-treated (5 mg/kg) and control mice. Standard uptake value (SUV), total volume of distribution ( V t ), and area under the curve (AUC) based on the [18F]FBAT PET signals were determined. The expression of iNOS was confirmed by immunohistochemistry (IHC) of brain tissues. Results. At the end of synthesis, the yield of [18F]FBAT was 2.2–3.1% (EOS), radiochemical purity was >99%, and molar radioactivity was 125–137 GBq/ μ mol. In vitro, [18F]FBAT rapidly and progressively accumulated in murine microglial BV2 cells exposed to LPS; however, [18F]FBAT accumulation was inhibited by aminoguanidine, a selective iNOS inhibitor. In vivo biodistribution studies of [18F]FBAT showed a significant increase in the liver and kidney on LPS-treated mice. At 3 h postinjection of LPS, in vivo, the [18F]FBAT accumulation ratios at 30 min post intravenous (i.v.) radiotracer injection for the whole brain, cortex, cerebellum, and brainstem were 2.16 ± 0.18 , 1.53 ± 0.25 , 1.41 ± 0.21 , and 1.90 ± 0.12 , respectively, compared to those of mice not injected with LPS. The mean area under the curve (AUC0-30min), total volume of distribution ( V t , mL/cm3), and K i (influx rate) of [18F]FBAT were 1.9 ± 0.21 - and 1.4 ± 0.22 -fold higher in the 3 h LPS group, respectively, than in the control group. In the pharmacokinetic two-compartment model, the whole brain K i of [18F]FBAT was significantly higher in mice injected with LPS compared to the control group. Aminoguanidine, selective iNOS inhibitor, pretreatment significantly reduced the AUC0-30min and V t values in LPS-induced mice. Quantitative analysis of immunohistochemically stained brain sections confirmed iNOS was preferentially upregulated in the cerebellum and cortex of mice injected with LPS. Conclusion. An automated robotic method was established for radiosynthesis of [18F]FBAT, and the preliminary in vitro and in vivo results demonstrated the feasibility of detecting iNOS activity/expression in LPS-treated neuroinflammation by noninvasive imaging with [18F]FBAT PET/MRI. Purpose: To determine the feasibility of ultra-short echo time (UTE) MRA for assessment of inferior vena cava (IVC) filters and evaluate the impact of different imaging protocols at 3.0 T, using conventional Cartesian MRA (cMRA) as the reference standard. Methods: Patients with IVC-filters were recruited for this prospective IRB-approved, HIPAA-compliant study. Subjects underwent contrast-enhanced breath-held and a free-breathing 3D radial acquisition UTE-MRA (bhUTE, fbUTE) at three different flip angles (FA: 10°, 15°, 20°) to optimize T1-weighted image quality. Two radiologists performed a direct comparison consensus reading to assess the optimal FA. Image quality (IQ) of both UTE techniques at the best FA was rated independently on a 4-point Likert scale (0 = non-diagnostic, 3 = excellent) and compared to 3D T1-weighted breath-held cMRA. Results: Nine subjects were recruited. Low FAs of 10° were rated best for both UTE techniques. fbUTE was excellent (3, IQR: 2; 3) and significantly better for IVC-filter depiction than cMRA (2, IQR: 0.75; 2, p = 0.001) and bhUTE (1.5, IQR: 0.75; 2, p < 0.001). Both UTE techniques showed significantly less filter-related artifacts (fbUTE: 28%, bhUTE: 33%) than cMRA (89%, p = 0.001 and p = 0.002, respectively). However, IQ of bhUTE was generally degraded due to high image noise and low image contrast. IQ of the IVC venogram was best with cMRA. Clinically relevant signal voids were only observed with the cage-shaped OptEase filter. Conclusion: UTE-MRA is feasible at 3.0 T for the assessment of IVC-filters, particularly using a free-breathing protocol. Previous neuroimaging research indicates that English verbs and nouns are represented in frontal and posterior brain regions, respectively. For Chinese monolinguals, however, nouns and verbs are found to be associated with a wide range of overlapping areas without significant differences in neural signatures. This different pattern of findings led us to ask the question of where nouns and verbs of two different languages are represented in various areas in the brain in Chinese–English bilinguals. In this study, we utilized functional magnetic resonance imaging (fMRI) and a lexical decision paradigm involving Chinese and English verbs and nouns to address this question. We found that while Chinese nouns and verbs involved activation of common brain areas, the processing of English verbs engaged many more regions than did the processing of English nouns. Specifically, compared to English nouns, English verb presentation was associated with stronger activation of the left putamen and cerebellum, which are responsible for motor function, suggesting the involvement of the motor system in the processing of English verbs. Our findings are consistent with the theory that neural circuits for linguistic dimensions are weighted and modulated by the characteristics of a language. [ABSTRACT FROM AUTHOR]

Abstract: Temporal clustering analysis (TCA) and independent component analysis (ICA) are promising data-driven techniques in functional magnetic resonance imaging (fMRI) experiments to obtain brain activation maps in conditions with unknown temporal information regarding the neuronal activity. Although comparable to ICA in detecting transient neuronal activities, TCA fails to detect prolonged plateau brain activations. To eliminate this pitfall, a novel derivative TCA (DTCA) method was introduced and its algorithms with different subtraction intervals were tested on simulated data with a pattern of prolonged plateau brain activation. It was found that the best performance of DTCA method in generating functional maps could be obtained if the subtraction interval is equal to or larger than the length of the rising time of the fMRI response. The DTCA method and its theoretical predication were further investigated and validated using in vivo fMRI data sets. By removing the limitations in the previous TCA, DTCA has shown its powerful capability in detecting prolonged plateau neuronal activities. [Copyright &y& Elsevier]

Abstract: Signal changes can be detected by proton density-weighted functional imaging in both the brain and the spinal cord. These are attributed to changes in extravascular water proton (signal enhancement by extravascular protons) density during neuronal activation. In this study, we used this technique to detect correlations between acupoint stimulation and neural activity in the spinal cord. Stimulation of acupoints associated with treatment of sensorimotor deficits (LI4 and LI11) was performed on 11 volunteers. During stimulation, 8 of the 11 subjects had consistent functional activations in C6/C7. A bilateral activation pattern was common. Our findings show that acupoint stimulation modulates activity in the spinal cord. [Copyright &y& Elsevier]

Sixty-seven healthy right-handed subjects were studied using a 1.5-T MRI. Activations, which were absent during stimulation of the adjacent nonacupoints over the right arm or leg, were consistently seen over the right premotor area during stimulation of the sensorimotor-implicated acupoints over the left arm or leg. The left premotor area was also activated during stimulation of the arm or leg acupoints. The right precentral and postcentral gyri showed activations during stimulation of the arm acupoints. The volume and location of activations vary when different combinations of the arm acupoints were stimulated. Our results indicate the existence of sensorimotor-implicated acupoints as sensitive sites to obtain somatotopic activations over the sensory cortices with co-activation of the motor cortices. Abstract: Proton-density-weighted fMRI at low field (0.2 T) was carried out in the cervical spinal cord of healthy volunteers in this study to examine the feasibility of detecting proton density alteration accompanying activation in the spinal cord. Subjects were asked to grip both hands simultaneously, providing sensorimotor simulation for spinal fMRI. Over 70% subjects recruited had activation localized at C6–C7 spinal levels with discrete activation detected in both the anterior and posterior horns of the cervical spinal cord, and the average fractional signal change was 4.06%. The 0.2 T low magnetic field and the 24 ms short TE used in this study diminished the BOLD effect to a negligible level, thus the observed signal change was believed to be mainly attributable to proton density increase during neuronal stimulation. Real-time or near-real-time acquisition plays a key role in providing immediate image guidance for interventional magnetic resonance imaging (iMRI). However, the requirement of accurate needle tip localization has made several accelerating techniques, like Keyhole imaging or sliding window reconstruction, difficult to be applied to iMRI. The purpose of this work was to further explore the possible ways of applying view sharing techniques to iMRI. Inspired by Keyhole imaging, we present an easy-to-implement accelerating strategy called “Alternate update of shifted extended keyholes (AUSEK)”. In this method, the keyhole views are not only extended but also shifted towards either high-frequency edge to form two divisions in k-space. The divisions which are mirrored to each other along the center are alternately updated following a reference scan. By using simulations and experiments, we demonstrate that AUSEK could effectively preserve the spatial resolution of the image, especially of the needle, at a temporal acceleration rate of about 2.5.
