Abstract: The mangrove oyster, Crassostrea rhizophorae (Bivalvia, Ostreidae) is commonly collected by fisherwomen in the estuaries of the Ceará State (CE), Northeastern Brazil. Despite the socioeconomic importance of this natural resource, there are few studies on the health of the oysters in this region. This study aimed to survey pathological changes in the mangrove oyster C. rhizophorae in the estuary of the Pacoti River, CE. Adult oysters were collected in August 2008 (N =450) and December 2009 (N =450) at three sites of the Pacoti estuary and in 2010 (N =600) samplings were done quarterly at one site which has showed the higher prevalence de Perkinsus. Macroscopical and histological analyses were used to evaluate pathological changes, Ray’s Fluid Thioglycollate Medium (RFTM) to detect Perkinsus spp. and polymerase chain reactions (PCR) and DNA sequencing to identify Perkinsus species. In 2009, RFTM assay detected Perkinsus sp. infecting the tissues of C. rhizophorae with low prevalences of 1.3%, 6.7% e 7.3% in sites 1, 2 and 3, respectively, and in 2010, in site 3, prevalence was 2% (12 of 600 oysters). PCR did not confirm any positive case in 2009 and only 5 in 2010. The phylogenetic analyses strongly indicate that the Perkinsus species infecting oysters C. rhizophorae of this study belongs to Perkinsus beihaiensis. The histology confirmed 11 cases of Perkinsus sp. infecting the C. rhizophorae in 2009, and only two cases in 2010. Nematopsis sp. was the protozoan observed with greater prevalence (up 96.7%). Other found protozoa were: Trichodina, Sphenophrya, Ancistrocoma – like and an unknown ovarian parasite. The metazoa found were the polychaete Polydora with high prevalences, a turbellarian, possibly of the genus Urastoma, an unidentified digenean metacercariae and larvae of cestode Tylocephalum. A continuous monitoring of diseases in bivalves from this natural population is recommended, since the phylogenetic analyses indicate the occurrence of P. beihaiensis infecting oysters C. rhizophorae whose pathogenic potential is unknown. Background: The milk production chain is an important component of the agribusiness in Western Santa Catarina, Brazil. However, this activity is facing problems regarding health of the herd that are often difficult to control because they can be caused by different etiologic agents such as viruses, bacteria, fungi and parasites. The damage caused by endoparasites may occur directly and indirectly, reflecting on weight gain, milk production, expenses with antiparasitic drugs, and mortality of untreated animals. To minimize these losses, all properties should implement a strategic program to monitor the herd. As a result, the objective of this study was to monitor dairy cattle properties from the West of Santa Catarina, by collecting information about the herd and parasitological stool examinations. Materials, Methods & Results: This study was conducted on farms from 12 cities of the Western region of Santa Catarina state during the months of November and December of 2010 and January and February of 2011 (summer). The study included 23 properties that have part of their farm income from dairy cattle activity; these animals had not been treated with anti-parasitic for at least 30 days. To assist research, a questionnaire was given to the farmers. For parasitological examination, fecal material was collected from at least 20% of the herd up to 100%, that is, was standardized the analysis of 8-10 samples per property for the McMaster technique using saturated salt solution to determine the degree of infection (EPG: eggs per gram of faeces). The total number of cows present in the properties was 648 animals, and of these 569 was lactating. All properties fed their animals with cultivated pasture and ration, whose amount ranged between 1 to 8 kg ration/day according to the productivity. In the interview, it was found that 100% of properties used anti-helmintic treatment. Of these, 70% of all treatments were performed usually with six months of interval and 75% conducted antiparasite rotations. Stool examination showed that only five farms had animals positive for gastrointestinal parasites the of Trichostrongylidae family (degree of infection between 100-300 EPG). Therefore, from 215 samples processed only 11 were positive for helminth eggs, which correspond to 5.16%. Discussion: Cattle raised on natural pastures and raised in Brazil are exposed to infection by larvae of gastrointestinal nematodes. The occurrence and distribution of endoparasites present regional and seasonal variations. Young animals are generally more susceptible to helminths, however, adult animals may be involved in situations such as post-partum, in association with other diseases, high exposure to the agent and stress conditions, like during the lactation period during the summer. Milk production has gained importance in rural properties of West Santa Catarina and in recent years has modernized and invested in technology and genetics. Based on the data it is possible to conclude that the occurrence of gastrointestinal parasites in dairy cows, in the summer, is low. This fact can be attributed to efficient parasite control achieved by owners; by adult animals with good immunity and probably low exposure to parasites in the environment due the excellent parasite control with drugs, held by owners. Protective factors associated with atopy or asthma in rural areas include socioeconomic level, overcrowding, and helminth infection. However, little epidemiological information was originated from schistosomiasis areas. This study aimed to investigate factors associated with asthma in a schistosomiasis endemic area. A questionnaire was used to obtain information on demographics, socioeconomic, and environmental features. The ISAAC questionnaire was used to identify individuals with asthma. Parasitological exam was done in all participants and skin prick test to aeroallergens in all asthmatics. Prevalence of Schistosoma mansoni infection was 57.4% and Ascaris lumbricoides, 30.8%. Asthma was found in 13.1% of the population, and 35.1% of them had a positive SPT. Active and passive smoking was positively associated with asthma, whereas A. lumbricoides was negatively associated. In a schistosomiasis hyperendemic region, current infection with A. lumbricoides is protective against asthma. However, we cannot rule out the involvement of S. mansoni infection in this process. Abstract: A parasitological survey was conducted among children and adolescents in the municipality of Jaboatão dos Guararapes, Brazil to describe the occurrence and spatial distribution of lymphatic filariasis. Microfilaraemia was investigated through the thick smear technique, using 50μl of capillary blood that was collected at night. The spatial analysis used a digital base map of the municipality, divided into districts, which were classified as hypoendemic, mesoendemic or hyperendemic. 8670 children were examined and 96 cases of microfilaraemia were identified (1.1%). The prevalence rate did not differ significantly between the sexes. Occurrences of filarial infection increased with increasing age: the greatest prevalence was recorded between 15 and 18 years of age (P <0.05). There were 49 reports of clinical manifestations. The spatial distribution of microfilaraemia according to residential district showed that 13 (54.2%) of the 24 districts investigated were positive. Approximately 33% of the districts were hyperendemic. The results demonstrated that the pediatric population had intense early exposure to the parasite, thus characterizing filariasis as endemic in the municipality. The spatial analysis allowed identification of areas with greater occurrence of infection among children, and showed localities where the populations most exposed to transmission were concentrated. Epidemiological surveillance of microfilaraemia among children and spatial analysis are important local transmission indicators and form instruments for planning actions within the Global Program to Eliminate Lymphatic Filariasis, since they make it possible to identify priority areas. American tegumentary leishmaniasis (ATL) is an infectious disease that presents a wide spectrum of clinical manifestations making parasitological tests important for its diagnosis. Direct examination, although considered of low sensitivity is still employed mainly in areas with poor laboratory infrastructure. The aim of this study was to standardize the method of collecting and reading the scraping procedure and to then compare sensitivity of this procedure on two sites of the lesion (outer edge-OE and inner edge-IE) and of the imprint against the reference method (isolation in culture) in a group of 110 patients treated at a Referral Center in Rio de Janeiro, Brazil. ATL diagnosis was confirmed in 40 patients (36.4%), 39 cases were caused by L. braziliensis and 1 by L. amazonensis. Imprint was positive in 28 patients and scraping in OE in 17 and in IE in 25 patients, resulting in sensitivity of 70%, 42.5%, and 62.5% respectively. When the three direct examinations were combined, sensitivity value attained 77.5%. Aspects related to ease and quality of the collected material, pain intensity and frequency of bleeding in the scraping procedure were also broached and discussed in this study. The parameters of accuracy presented indicate that the direct methods can be safely used in ATL diagnosis, principally in IE scraping, as it is easy to produce and the examination is not costly, which allows the procedure to be repeated at different moments which, in turn, increases the possibility of finding the parasite. Despite that the direct methods are technically widespread, they are not standardized and the parameters of accuracy are unknown. If we consider the high incidence of leishmaniasis in low-income areas, the implantation of standardized and selective methods would provide advances in the diagnosis of leishmaniasis. Objective: In the city of Salvador, a large urban centre in Northeast Brazil, a city-wide sanitation intervention started in 1997, aimed at improving the sewerage coverage of households from 26% to 80%. Our aim was to study the impact of the intervention on the prevalence and incidence of geohelminths in the school-aged population. Methods: The study comprised two comparable cohorts: the first assembled in 1997, before the intervention, and thesecond assembled in 2003, after the intervention. Both were sampled from 24 sentinel areas chosen to represent the different environmental conditions throughout the city. Copro-parasitological examinations were carried out on everyindividual from both cohorts, at baseline and nine months later. Demographic, socio-economic, and environmental data were collected using semi-structured questionnaires and environmental surveys. A hierarchical modelling approach fitting a sequence of Poisson multivariate linear models was undertaken to test the effect of the intervention variables on the prevalence and incidence rate ratios. Findings: 729 and 890 children aged 7-14 years (mean = 10.4 y, SD = 0.05 y) were analysed over the first and the second cohorts, respectively. The adjusted reductions of the prevalence and incidence rates at the second in relation to the first cohort were 27% and 34%, 25% and 32%, 33% and 26%, and 82% and 42% for geohelminths overall, Ascaris lumbricoides, Trichuris trichiura, and hookworm, respectively. Hierarchical modelling showed that a major part of each of these reductions was explained by the intervention. Conclusion: Our results show that a city-wide sanitation program may reduce significantly the prevalence and incidence of geohelminths. [ABSTRACT FROM AUTHOR]

Abstract  An unindentified microfilaria sharing characteristics with Mansonella ozzardi and Onchocerca volvulus was detected in blood samples from seven human volunteers, inhabitants of a community in the border of Amazonas and Acre State. They were detected during epidemiological studies carried out in some communities along Antimary, Acre, and Purus Rivers in the Brazilian Amazon. The most striking difference was presented in the shape of the cephalic space from this microfilaria which was different from those of M. ozzardi and with similarities to O. volvulus in this region, but no remarkable differences were observed at the caudal region. More accurate studies are being carried out in order to provide additional data and supporting evidences before establishment of a new species can be done. [ABSTRACT FROM AUTHOR]

Abstract  We report the finding, the isolation by hemoculture, and the characterization of Trypanosoma rangeli stocks from two chronic Chagas’ disease patients who received ambulatory care at the Evandro Chagas Clinical Research Institute (IPEC, FIOCRUZ). Both patients proceeded from Bahia State (Brazil). One of them presented the cardiac form of the disease and the other indeterminate symptomalogy. Giemsa-stained smears of the hemocultures from these patients evidenced that they were coinfected with T. rangeli and Trypanosoma cruzi, with predominance of the former species. These isolates could only be successfully grown in Novy–MacNeal–Nicolle + liver infusion-tryptose supplemented with 20–30% fetal calf serum. After 6 months of serial maintenance, rich and apparently pure cultures of T. rangeli were obtained. Both stocks were analyzed with different approaches and compared with two T. cruzi isolates also from chagasic patients under care at IPEC, besides T. rangeli and T. cruzi reference strains. All stocks were characterized by morphology, biometry, electrophoresis of isoenzymes, and products of kDNA minicircle amplified by polymerase chain reaction. The identification of T. rangeli was largely confirmed by all techniques. Taken together, these data represent the third report on T. rangeli in human hosts in Brazil. [ABSTRACT FROM AUTHOR]

Abstract  The in vitro antileishmanial activity of Brazilian green propolis hydroalcoholic extract (BPE) were carried out on Leishmania (Viannia) braziliensis against both promastigote (doses ranging from 1 to 750 μg mL−1) and amastigote (10, 100, and 250 μg mL−1) assays in comparison with the positive (amphotericin B) and negative (dimethyl sulfoxide at 1% in physiologic solution) control groups. BPE displayed in vitro antileishmanial activities against promastigote forms of the parasite (p p-coumaric acid, aromadendrine-4′-methyl-ether, 3-prenyl-p-coumaric acid (drupanin), and 3,5-diprenil-p-cumárico acid (artepillin C) as major compounds of BPE. In the in vivo assay, using a Balb/C lineage of Mus musculus male mice, groups of ten animals each were treated (1.5 mg kg day−1) with BPE orally (group 1), BPE topically (group 2), BPE orally and topically (group 3), and glucantime (group 4), using NaCl 0.9% (group 5) as the negative control group. Groups 1, 2, and 3 displayed a decrease on lesion development, after 90 days of treatment, by 78.6%, 84.3%, and 90.0%, respectively, while the glucantime-treated group showed 57.7% of decrease, all in comparison with the negative control group. It is the first time that the in vivo antileishmanial activity has been reported for Brazilian green propolis. [ABSTRACT FROM AUTHOR]

Abstract  The Brazilian Pantanal has been considered one of the richest and most diverse wetland ecosystems in the world. It is occupied by cattle ranching, and a variety of wildlife species share the same habitats with domestic livestock. We investigated infections of Trypanosoma evansi and Trypanosoma cruzi in the sympatric suiformes-collared peccary (Tayassu tajacu), white-lipped peccary (Tayassu pecari), and feral pig (Sus scrofa) by parasitological, serological, and molecular tests. Additionally, we evaluated the health status of both positive and negative suiformes by hematological and biochemical parameters. The results show that peccaries and feral pigs play an important role on the maintenance of both T. evansi and T. cruzi in the Brazilian Pantanal. Health impairment was observed only in the white-lipped peccary infected with T. evansi. Despite presenting low T. evansi parasitemia, all infected white-lipped peccaries displayed low hematocrit values and marked leucopenia. The hematological values showed that the T. evansi infection is more severe in young white-lipped peccaries. The presented data show that feral pigs and peccaries are immersed in the transmission net of both trypanosome species, T. cruzi and T. evansi, in the Pantanal region. [ABSTRACT FROM AUTHOR]
