There continues to be debate regarding the timing of the collision between the Indian and Eurasian plates and the uplift of the Tibetan Plateau. This study presents zircon U–Pb geochronology, whole-rock geochemistry, and Lu–Hf isotopic data for the Saiduopugangri granite of the Qiangtang Terrane, located within the core of the Tibetan Plateau. These data provide the basis for the geodynamic setting, petrogenesis, and characteristics of its magma source. Zircons from the Saiduopugangri granite yield a weighted-mean 206Pb/238U age of 62.72 ± 0.06 Ma, indicating that these rocks formed during the early Palaeocene. The rocks are members of the highly calc-alkaline to shoshonitic series, with weak peraluminous characteristics. Trace elements are characterised by high Sr (483–616 ppm), and low Y (6–10 ppm) and Yb (1 ppm) content, typical of a high Sr and low Yb granite. The εHf(t) of zircon ranges from −2.14 to 2.35, with two-stage Hf model ages (TDM2) ranging from 1182 to 895 Ma. These data suggest that the Saiduopugangri granite magma was derived from the melting of lower-crustal clastic meta-sedimentary rocks and mantle-derived basalts. The high Sr and low Yb granite characteristics and experimental results indicate that melting occurred at >1.2 GPa and >750 °C, consistent with a crustal thickness greater than 50 km. Magmatism occurred from the Late Cretaceous to the early Palaeogene and is broadly synchronous with the collision timing between the Indian and Eurasian plates. The Saiduopugangri granite provides evidence of crustal thickening of the Tibetan Plateau and its age and petrogenesis constrain the timing of the initial uplift. [ABSTRACT FROM AUTHOR]

Ophiocordyceps sinensis, endemic to the Tibetan Plateau, is one of the most important medicinal fungi with a huge economic value. In the present study, specific primer pairs were designed based on a comprehensive ITS sequence dataset of O. sinensis and its related fungi, and tested for specificity and sensitivity through PCR experiments using 27 individuals of O. sinensis from different geographical origins and 40 other related fungal species in terms of phylogeny or ecology. A primer pair highly specific to O. sinensis was obtained, yielding a 275 bp PCR product from all the individuals of O. sinensis but no product from the other fungi tested. The detection limit of the primers was demonstrated to be 10 ng of pure O. sinensis DNA for conventional PCR and 10 pg for nested PCR in a 25 µl reaction system. Soil samples collected from the habitat of O. sinensis were also tested using this PCR assay. The results showed that the primer pair and PCR-based assays developed in this study can be applied to the rapid detection of O. sinensis in its natural habitat. [ABSTRACT FROM AUTHOR]

Background and Aims: Fenced enclosures have become an important method for re-establishing degraded grassland on the Tibetan plateau, and examination of soil seed banks may provide useful insights to understanding the effects and mechanisms of fencing enclosure on the restoration. Methods: An investigation was conducted into the effects of enclosure for 3 years on the soil seed banks of degraded natural and sown grasslands at eight study sites. Species composition, soil seed bank density and the relationships with above-ground vegetation were analysed based on 4800 soil core samples and counting of seeds extracted from soil samples. Results: After 3 years of fencing enclosure, soil seed banks differed between the different communities across the study sites. Species numbers and seed density in soil seed banks decreased from natural grassland to sown grassland, with most seeds occurring in the upper 5 cm soil layer. In these alpine grasslands, relatively few species produced high numbers of seeds, although their occurrence across the eight study sites was variable. Total vegetation cover increased with enclosure due to the colonization capacity of the vegetation rather than soil seed banks. Conclusions: This study provided evidence that soil seed banks do not play an important role in the restoration of degraded alpine grassland when using fencing enclosures.
