Objective: To forecast the visceral leishmaniasis cases using autoregress integrated moving average (ARIMA) and hybrid ARIMA-EGARCH model, which offers a scientific basis to control visceral leishmaniasis spread in Kashgar Prefecture of Xinjiang, China. Methods: The data used in this paper are monthly visceral leishmaniasis cases in the Kashgar Prefecture of Xinjiang from 2004 to 2016. The sample data between 2004 and 2015 were used for the estimation to choose the best model and the sample data in 2016 were used for the forecast. Time series of visceral leishmaniasis started on 1 January 2004 and ended on 31 December 2016, consisting of 1 790 observations reported in Kashgar Prefecture. Results: For Xinjiang, the total number of reported cases were 2 187, the male-to-female ratio of cases was 1:1.42. Patients aged between 0 and 10 years accounted for 82.72% of all reported cases and the largest percentage of visceral leishmaniasis cases was detected among scattered children who accounted for 68.82%. The monthly incidences fitted by ARIMA (2, 1, 2) (1, 1, 1)12 model were consistent with the real data collected from 2004 to 2015. However, the predicted cases failed to comply with the observed case number; we then attempted to establish a hybrid ARIMA-EGARCH model to fit visceral leishmaniasis. Finally, the ARIMA (2, 1, 2) (1, 1, 1)12- EGARCH (1, 1) model showed a good estimation when dealing with volatility clustering in the data series. Conclusions: The combined model has been determined as the best prediction model with the root-mean-square error (RMSE) of 7.23% in the validation phase, which means that this model has high validity and rationality and can be used for short-term prediction of visceral leishmaniasis and could be applied to the prevention and control of the disease.

We have investigated the thickness-dependent transport properties of SrMoO 3 thin films deposited on LaAlO 3 substrates. Metal–insulator transitions (MITs) were observed in SrMoO 3 thin films with thickness below 10 nm. The low-temperature resistivity of these films can be explained by quantum corrections of the conductivity. An insulating behavior is observed when the thickness becomes 3.5 nm, and the resistivity can be described by the variable range hopping model with 2D fitting. The magneto-transport measurement of an SrMoO 3 thin film with small positive magnetoresistance confirms that the driving force behind MIT is the renormalized electron–electron interaction. Comprehensive Summary: Dehydration of serine/threonine residues necessitates the activity of a dehydratase enzyme (domain) during the biosynthesis of RiPP. Recently, it was reported that dehydration in the thioviridamide pathway relies on a distinct dehydratase complex that showcases the activities of a phosphotransferase TvaC for serine/threonine phosphorylation and a lyase TvaD for subsequent phosphate elimination. Herein, we report that dehydration reactions in the pathway of lantibiotic cacaoidin involves a similar dehydratase complex, CaoK/CaoY. Remarkably, this dehydratase complex exhibits flexible enzymatic activity and tolerates significant variations in its substrate peptide sequence. By binding with the leader peptide (LP) sequence of precursor peptide CaoA, the dehydration reactions proceed in a directional manner from the C‐terminus of the core peptide (CP) to the N‐terminus, and C‐terminally truncated variants of CP are acceptable. We show that fusing CaoK to CaoY in a 1 : 1 molar ratio enables the resulting enzyme CaoYK to exert enhanced dehydration activity. CaoK binds with the LP to improve its own solubility and to ensure the phosphate transfer activity, while CaoY functions in a manner independently of LP. This work advances our understanding of the dehydration process during cacaoidin formation, and provides useful enzymes and methods for the studies of the rapidly emerging RiPPs. Objectives: High reactive oxygen species (ROS) levels lead to cell death, and the testes are among the most vulnerable organs to oxidative damage. Rg1, an active ingredient extracted from the natural medicine ginseng, has potential anti-inflammatory, antioxidant and antiapoptotic properties. Our previous studies showed that Rg1 can effectively improve spermatogenic function in mice, but the specific mechanism remains unclear. The purpose of this study was to investigate the effect of Rg1 on oxidative stress and spermatogonium apoptosis in D-gal-induced testicular toxicity and elucidate the associated mechanism. Methods: Male C57BL/6 mice at 6–8 weeks of age were intraperitoneally injected with D-gal (200 mg/kg) for 42 days to establish a testicular injury model, and on day 16, 40 mg/kg Rg1-rich saline was injected intraperitoneally. Concurrently, we established an in vitro model of D-gal-damaged spermatogonia, which was treated with Rg1. Results: We found that treatment with the ginsenoside Rg1 reduced D-gal-induced oxidative stress and spermatogonium apoptosis in vivo and in vitro. Mechanistically, we found that Rg1 activated Akt/bad signaling and reduced D-gal-induced spermatogonium apoptosis. Discussion: We provide evidence showing that the antioxidant effect of Rg1 is mediated by the Akt/GSK-3β/NRF2 axis. Objective To build a construction strategy for RNA-seq library for small quantity of cells by optimizing Tn5 transposase assembly and purification workflow. Methods Recombinant plasmid pSUMO-Tn5 was constructed by homologous recombination. After Tn5 protein was induced to express, the product was purified with affinity chromatography followed by optimization of Tn5 assembly. Then we constructed RNA-seq library using RAW264.7 cells and analyzed the number of genes detected and the ratio of unmapped reads. Results Soluble Tn5 protein was expressed in Codon Plus BL21 with the recombinant plasmid pSUMO-Tn5. Tn5 with high purity was obtained through affinity purification. RNA-seq library for small quantity of RAW264.7 cells was successfully constructed. After the optimization of Tn5 assembly, nonspecific amplification was remarkably reduced compared with the control (P < 0.05). Conclusion Tn5 protein is effectively expressed and purified. Ultra-filtered Tn5 transposase can reduce the nonspecific amplification in RNA-seq, providing a new strategy of RNA-seq for small quantity of cells.
