U.S. forests, particularly in the eastern states, provide an important offset to greenhouse gas (GHG) emissions. Some have proposed that forest‐based natural climate solutions can be strengthened via a number of strategies, including increases in the production of forest biomass energy. We used output from a forest dynamics model (SORTIE‐ND) in combination with a GHG accounting tool (ForGATE) to estimate the carbon consequences of current and intensified timber harvest regimes in the Northeastern United States. We considered a range of carbon pools including forest ecosystem pools, forest product pools, and waste pools, along with different scenarios of feedstock production for biomass energy. The business‐as‐usual (BAU) scenario, which represents current harvest practices derived from the analysis of U.S. Forest Service Forest Inventory and Analysis data, sequestered more net CO2 equivalents than any of the intensified harvest and feedstock utilization scenarios over the next decade, the most important time period for combatting climate change. Increasing the intensity of timber harvest increased total emissions and reduced landscape average forest carbon stocks, resulting in reduced net carbon sequestration relative to current harvest regimes. Net carbon sequestration "parity points," where the regional cumulative net carbon sequestration from alternate intensified harvest scenarios converge with and then exceed the BAU baseline, ranged from 12 to 40 years. A "no harvest" scenario provides an estimate of an upper bound on forest carbon sequestration in the region given the expected successional dynamics of the region's forests but ignores leakage. Regional net carbon sequestration is primarily influenced by (1) the harvest regime and amount of forest biomass removal, (2) the degree to which bioenergy displaces fossil fuel use, and (3) the proportion of biomass diverted to energy feedstocks versus wood products. Non‐obstructive azoospermia is one of the most common causes of male infertility, but there is still no specific treatment drug. Given that the Oct4 (Octamer‐binding transcription factor 4) has an important regulatory effect on spermatogenesis, activating it can effectively promote spermatogenesis, so it is of great value to develop Oct4‐targeted drug design and elucidating its mechanism of action. Here, we screened out the Oct4‐targeted drug molecule NBMA (N‐benzyl‐4‐methoxy‐2‐(1‐(4‐(trifluoromethyl)phenyl)vinyl)aniline) by computer‐assisted technology, and found that it has a significant promoting effect on spermatogenesis in the established mouse azoospermia model. Subsequently, through transcriptome sequencing and enrichment analysis, real‐time fluorescent quantitative PCR (qPCR) and western blot experiments revealed that NBMA promotes the differentiation of spermatogonial stem cells by activating the Oct4 pathway, thereby promoting spermatogenesis. This study proves that NBMA is a molecule with great potential to be developed as a therapeutic drug for azoospermia. Coarse woody debris (CWD) is a meaningful contributor to forest carbon cycles, wildlife habitat, and biodiversity and can influence wildfire behavior. Using airborne laser scanning (ALS), we map CWD across a range of natural forest stand types in north-central British Columbia, Canada, providing forest managers with spatially detailed information on the presence and volume of ground-level woody biomass. We describe a novel methodology that isolates CWD returns from large diameter logs (>30cm) using a refined grounding algorithm, a mixture of height and pulse-based filters and linear pattern recognition, to transform ALS returns into measurable, vectorized shapes. We then assess the accuracy of CWD detection at the individual log level and predict CWD volume at the plot level. We detected 64% of CWD logs and 79% of CWD volume within our plots. Increased elevation of CWD significantly aided detection (P  = 0.04), whereas advanced stages of decay hindered detection (P  = 0.04). ALS-predicted CWD volume totals were compared against field-measured CWD and displayed a strong correlation (R = 0.81), allowing us to expand the methodology to map CWD over a larger region. The expanded CWD volume map compared ALS volume predictions between stands and suggests greater volume in stands with older and more heterogeneous stand structure. [ABSTRACT FROM AUTHOR]

This article shows a first step in the development of an immersive virtual tour of the Cathedral of Palermo, entering the fields of Digital Cultural Heritage and Edutainment. Its purpose is to help people to gain knowledge about the site, highlighting the complex stratifications that have characterized its history.The development of the project has been possible thanks to different phases of work: surveys were initially carried out by laser scanning, then assembled and processed to obtain the 3D model of the current state; at the same time, the model of reconstruction was processed in several phases, based on historical, archival and iconographic sources; both models were, later, subject to post-processing, preparatory to the development of virtual navigation. The collection of tree-ring data from living trees is widespread and highly valuable in ecological and dendro-climatological research, yet there is concern that coring injures trees, potentially contributing to mortality. Unlike resinous conifers that can quickly compartmentalize wounds, less decay-resistant angiosperms may face more pronounced risk of injury from coring. To test if coring increases mortality rates in temperate hardwood trees, we leverage a unique dataset tracking the mortality of cored and uncored hardwood trees representing 19 species from 10 genera in a 26-ha plot in Virginia, USA. We compare mortality rates between 935 cored trees and 8605 uncored trees for seven years following coring. Annual mortality rates did not differ between cored trees (1.71% yr-1; 95% CI 1.40 to 2.20) and uncored trees (1.85% yr-1; 95% CI 1.70 to 2.28) across the full dataset, nor were there differences by genus or size class. These results indicate that the risk of hardwood mortality caused by increment coring is probably lower than previously assumed. However, these results cannot rule out the possibility that coring elevates hardwood mortality in other climates or when trees facemultiple additional stressors that were not influential over the course of our study. The increasing interest in forest biomass for energy or carbon cycle purposes has raised the need for forest resource managers to refine their understanding of downed woody debris (DWD) dynamics. We developed a DWD forecasting tool using field measurements (mean size and stage of stage of decay) for three common forest types across the eastern United States using field observations of DWD from the US Department of Agriculture Forest Service's Forest Inventory and Analysis program. Residence times for DWD ranged from as short as 20.3 ± 9.7 years (mean ± SD) for loblolly pine biomass in the southeastern United States to as long as 41.7 ± 20.9 years for aspen biomass in the Lake States. Although we suggest numerous improvements through refined DWD measurements and models, our proposed tool can be considered a rapid assessment technique to assist natural resource managers in forecasting DWD populations. Beginning with using log size and decay stage to understand deadwood dynamics, future researchers should seek to refine DWD metrics through implementing enhanced forest inventory information and exploring the impacts of forest disturbances on inputs to the DWD pool. In the eastern United States, American chestnut (Castanea dentata) was historically a major component of forest communities, but was functionally extirpated in the early 20th century by an introduced pathogen, chestnut blight (Cryphonectria parasitica). Because chestnut is fast-growing, long-lived, and resistant to decay, restoration of American chestnut using blight-resistant stock could have the potential to increase carbon sequestration or storage in forested landscapes. However, carbon dynamics are also affected by interspecific competition, succession, natural disturbance, and forest management activities, and it is unknown how chestnut restoration might interact with these other processes. We used the PnET-Succession extension of the LANDIS-II forest landscape model to study the implications of chestnut restoration on forest composition and carbon storage in the context of other disturbances, including timber harvest and insect pest outbreaks. Our results imply that it could take a millennium or more for chestnut to fully occupy landscapes without aggressive restoration efforts. When successful, chestnut restoration activities displaced other species approximately in proportion to their abundance on the landscape, rather than replacing a single species or genus (e.g., Quercus). Insect pests increased the rate of chestnut colonization by reducing the abundance of competitors, and also had a dominant effect on carbon dynamics. Although chestnut is fast-growing, moderately shade-tolerant, and decomposes very slowly, our results suggest that it can only modestly increase the carbon storage potential of eastern forests. However, our results also demonstrate that compositional changes in forest communities can have noticeable effects on biomass accumulation, even with the large uncertainties introduced by invasive pests. Forest management practices emphasizing stand structural complexity are of interest across the northern forest region of the United States because of their potential to enhance carbon storage. Our research is part of a long-term study evaluating silvicultural treatments that promote late-successional forest characteristics in northern hardwood-conifer forests. We are testing the hypothesis that aboveground biomass development (carbon storage) is greater in structural complexity enhancement (SCE) treatments when compared to conventional uneven-aged treatments. Structural complexity enhancement treatments were compared against selection systems (single-tree and group) modified to retain elevated structure. Manipulations and controls were replicated across 2-ha treatment units at two study areas in Vermont, United States. Data on aboveground biomass pools (live trees, standing dead, and downed wood) were collected pre- and post-treatment, then again a decade later. Species group-specific allometric equations were used to estimate live and standing dead biomass, and downed log biomass was estimated volumetrically. We used the Forest Vegetation Simulator to project "no-treatment" baselines specific to treatment units, allowing measured carbon responses to be normalized against differences in site characteristics affecting tree growth and pre-treatment stand structure. Results indicate that biomass development and carbon storage 10 yr post-treatment were greatest in SCE treatments compared to conventional treatments, with the greatest increases in coarse woody material (CWM) pools. Structural complexity enhancement treatments contained 12.67 Mg/ha carbon in CWM compared to 6.62 Mg/ha in conventional treatments and 8.84 Mg/ha in areas with no treatment. Percentage differences between post-treatment carbon and simulated/projected baseline values indicate that carbon pool values in SCE treatments returned closest to pre-harvest or untreated levels over conventional treatments. Total carbon storage in SCE aboveground pools was 15.90% less than that projected for no-treatment compared to 44.94% less in conventionally treated areas. Results from classification and regression tree models indicated treatment as the strongest predictor of aboveground C storage followed by site-specific variables, suggesting a strong influence of both on carbon pools. Structural enhancement treatments have the potential to increase carbon storage in managed northern hardwoods. We report three cases of effective management of infertility in women with a history of repeated unsuccessful IVF attempts, who have developed antibodies to hCG. A novel approach to conservative treatment of immunologic reproductive failure, suggested for selected patients, included membrane plasmapheresis, combined prednisolone, and intravenous immunoglobulin therapy. No adverse side effects were observed; all cases resulted in pregnancy and subsequent life births. In order to be given an adequate efficient treatment, women with recurrent implantation failure should be suspected for autoimmune factor of infertility and its possible association with anti-hCG autoimmunity. This article deals with enantioselective gas chromatography (Es-GC) utilizing cyclodextrin derivatives as chiral stationary phases (CSPs) for chiral recognition in the flavour and fragrance field. It consists of two main parts. The first one concerns cyclodextrins and their use in Es-GC, and it deals with their introduction and evolution as CSPs in GC, together with some theoretical aspects involved with enantiomeric separation and their consequent influence on routine chiral recognition. The second part reports on the strategy of chiral recognition in routine analysis with cyclodextrin derivatives as CSP, illustrated with examples from the authors' everyday experiences. This part describes how to identify enantiomers or to measure their excess or ratio determination in complex mixtures; in particular, it discusses the opportunities offered by multidimensional and fast GC techniques, the role of mass spectrometry, and the application of total analysis systems to chiral recognition. Trait analysis aims to understand relationships between traits, species diversity, and the environment. Current methods could benefit from a model-based probabilistic framework that accommodates covariance between traits and quantifies contributions from inherent trait syndromes, species interactions, and responses to the environment. I develop a model-based approach that separates these effects on trait diversity. Application to USDA Forest Inventory and Analysis ( FIA) data in the eastern United States demonstrates an apparent paradox, that the analysis of species better explains and predicts traits than does direct analysis of the traits themselves; trait data contain less, not more, information than species on environmental responses. Whereas variation in some traits is dominated by inherent syndromes (tendency for certain traits to be associated with others within an individual and species), others are strongly controlled by variation in species diversity. There is substantial variation in environmental control on trait patterns, between traits and regionally. We study the effect of maize zeins on cellular immune response as compared to that of wheat gliadins for exacerbating celiac disease due to a hypothetical similar response in some patients.In vitroactivation of celiac duodenal mucosa with gliadin or zein immunoreactive peptides and pro-inflammatory response of CACO-2 cells to digested gliadin or zein fractions were evaluated as indicators of adaptive and innate response, respectively. In 3/5 biopsies, zein increased production of IFN-γ, whereas gliadin has done it in all the patient biopsies. In CACO-2 cells, two zein fractions (3–5 and 1–3 kDa), similar to gliadin fractions, stimulated the production of IL-8, p38 MAPK, COX2, and release of ZO-1 as compared to medium alone. The 3–5 kDa zein fraction increased permeability in cell monolayers, although less than gliadin. Zein peptides are immunogenic for some patients and induce a similar innate response, but to a lesser extent than gliadin peptides. [ABSTRACT FROM PUBLISHER]

New advances into the chirality effect in the self-assembly of block copolymers (BCPs) have been achieved by tuning the helicity of the chiral-core-forming blocks. The chiral BCPs {[NP( R)-O2C20H12]200− x[NP(OC5H4N)2] x}- b- [NPMePh]50 (( R)-O2C20H12=( R)-1,1′-binaphthyl-2,2′-dioxy, OC5H4N=4-pyridinoxy (OPy); x=10, 30, 60, 100 for 3 a- d, respectively), in which the [NP(OPy)2] units are randomly distributed within the chiral block, have been synthesised. The chiroptical properties of the BCPs ([ α]D vs. T and CD) demonstrated that the helicity of the BCP chains may be simply controlled by the relative proportion of the chiral and achiral (i.e., [NP( R)-O2C20H12] and [NP(OPy)2], respectively) units. Thus, although 3 a only contained only 5 % [NP(OPy)2] units and exhibited a preferential helical sense, 3 d with 50 % of this unit adopted non-preferred helical conformations. This gradual variation of the helicity allowed us to examine the chirality effect on the self-assembly of chiral and helical BCPs (i.e., 3 a- c) and chiral but non-helical BCPs (i.e., 3 d). The very significant influence of the helicity on the self-assembly of these materials resulted in a variety of morphologies that extend from helical nanostructures to pearl-necklace aggregates and nanospheres (i.e., 3 b and 3 d, respectively). This contribution explores the effect of the bridging diamine upon the aggregation properties of a ZnII Schiff-base complex, 1, both in the solid state and in solution. The X-ray structure of 1, resulting from the harvest of good quality crystals using chloroform and diethyl ether as solvents, shows the presence of a densely packed dimer in the solid state which pentacoordinates two Zn atoms involved in a μ-phenoxo bridge. Detailed studies in solution, through 1H NMR, DOSY NMR, and optical spectroscopic investigations, indicate the typical aggregation/deaggregation behaviour on switching from non-coordinating to coordinating solvents, in relation to the Lewis acidic character of such ZnII complexes. Thus, while in DMSO-d6 both 1H NMR and DOSY studies suggest the existence of monomeric species, in chloroform solution experimental data support the existence of aggregates. However, unlike our previous studies, 1H NMR data in chloroform solution indicate the existence of an asymmetric dimer, as observed in the X-ray crystal structure. This further evidences a very rigid backbone of the dimeric aggregate and can be related to the defined stereochemistry of the chelate cis-1,2-diaminocyclohexane bridge. [ABSTRACT FROM AUTHOR]

Forest ecosystems are an important component in the global carbon cycle. Coarse woody debris (CWD), which is a substantial portion of the aboveground forest biomass, can be an indicator of historical disturbance events and is a key variable for estimating carbon storage in forests. This study focused on identifying spatial and temporal dynamics in CWD pools within an old-growth temperate deciduous forest. The CWD in the study site was measured three times (1989, 1999, and 2012) following identical methods. Volumes of CWD were relatively stable over the measurement intervals, with the only significant difference being a higher CWD load in the midslope (mesophytic) community in 1989. The spatial arrangement of CWD across the topographically complex landscape was assessed using a geographical information systems (GIS) model that tested for physiographic predictors of CWD volume. Topographic patterns were not discovered, suggesting that CWD deposition was spatially stochastic. The relative temporal and spatial uniformity in CWD matches expectations for old-growth temperate forests; however, oncoming pests and pathogens as well as climate effects have the potential to drive future shifts in CWD loads. Complexation of racemic citalopram with β-cyclodextrin ( β-CD) in aqueous medium was investigated to determine atom-accurate structure of the inclusion complexes. 1H-NMR chemical shift change data of β-CD cavity protons in the presence of citalopram confirmed the formation of 1 : 1 inclusion complexes. ROESY spectrum confirmed the presence of aromatic ring in the β-CD cavity but whether one of the two or both rings was not clear. Molecular mechanics and molecular dynamic calculations showed the entry of fluoro-ring from wider side of β-CD cavity as the most favored mode of inclusion. Minimum energy computational models were analyzed for their accuracy in atomic coordinates by comparison of calculated and experimental intermolecular ROESY peak intensities, which were not found in agreement. Several least energy computational models were refined and analyzed till calculated and experimental intensities were compatible. The results demonstrate that computational models of CD complexes need to be analyzed for atom-accuracy and quantitative ROESY analysis is a promising method. Moreover, the study also validates that the quantitative use of ROESY is feasible even with longer mixing times if peak intensity ratios instead of absolute intensities are used. Ovarian follicular growth and development are regulated by extraovarian and intraovarian factors, which influence granulosa cell proliferation and differentiation. However, the molecular mechanisms that drive follicular growth are not completely understood. Ovarian follicular cysts are one of the most common causes of reproductive failure in dairy cattle. Nevertheless, the primary cause of cyst formation has not been clearly established. A gene expression comparison may aid in elucidating the causes of ovarian cyst disease. Our objective was to identify differentially expressed genes in ovarian granulosa cells between normal dominant and cystic follicles of cattle. Granulosa cells and follicular fluid were isolated from dominant and cystic follicles collected via either ultrasound-guided aspiration from dairy cows (n = 24) or slaughterhouse ovaries from beef cows (n = 23). Hormonal analysis for progesterone, estradiol, and androstenedione in follicular fluid was performed by RIA. Total RNA was extracted and hybridized to 6 Affymetrix GeneChip Bovine Genome Arrays (Affymetrix, Santa Clara, CA). Abundance of mRNA for differentially expressed selected genes was determined through quantitative real-time reverse-transcription PCR. Follicular cysts showed greater (P < 0.05) progesterone, lesser (P < 0.05) estradiol, and no differences (P > 0.10) in androstenedione concentrations compared with noncystic follicles. A total of 163 gene sequences were differentially expressed (P < 0.01), with 19 upregulated and 144 downregulated. From selected target genes, quantitative real-time reverse-transcription PCR confirmed angiogenin, PGE2 receptor 4, and G-protein coupled receptor 34 genes as upregulated in cystic follicles, and Indian hedgehog protein precursor and secreted frizzled-related protein 4 genes as downregulated in cystic follicles. Current methods used for analyzing biomarkers involve expensive and time consuming techniques like the Sandwich ELISA which require lengthy incubation times, high reagent costs, and bulky optical equipment. We have developed a technique involving the use of a micro-channel with integrated electrodes, functionalized with receptors specific to target biomarkers. We have applied our biochip to the rapid electrical detection and quantification of target protein biomarkers using protein functionalized micro-channels. We successfully demonstrate detection of anti-hCG antibody, at a concentration of 1 ng ml−1and a dynamic range of three orders of magnitude, in less than one hour. We envision the use of this technique in a handheld device for multiplex high throughput analysis using an array of micro-channels for probing various protein biomarkers in clinically relevant samples such as human serum for cancer detection. In order to determine the clinical and laboratory features of X-linked agammaglobulinemia, the records of 33 male patients with XLA were reviewed during 22 years (1980–2002) in the Iranian referral center of primary immunodeficiency disorders. The patients' ages ranged from 20 to 360 months (median 113 months). The median age at the onset of the disease was 8 months and the median age of diagnosis was 48 months, with a median diagnosis delay of 33 months. Almost all of the patients presented common infectious diseases, which were: pneumonia, otitis, diarrhea, sinusitis, and arthritis. During the course of illness, infections in the respiratory tract, gastrointestinal tract, central nervous system, and musculoskeletal system were seen in 93.9%, 75.8%, 33.3%, and 21.2% of XLA patients, respectively. The most common complications of these patients were chronic infections in 75.8% of them, including: chronic otitis media, chronic sinusitis, chronic diarrhea, and bronchiectasis. Muntjac deer (Muntiacinae, Cervidae) are of great interest in evolutionary studies because of their dramatic chromosome variations and recent discoveries of several new species. In this paper, we analyze the evolution of karyotypes of muntjac deer in the context of a phylogeny which is based on 1,844-bp mitochondrial DNA sequences of seven generally recognized species in the muntjac subfamily. The phylogenetic results support the hypothesis that karyotypic evolution in muntjac deer has proceeded via reduction in diploid number. However, the reduction in number is not always linear, i.e., not strictly following the order: 46→14/13→8/9→6/7. For example, Muntiacus muntjak (2n = 6/7) shares a common ancestor with Muntiacus feae (2n = 13/14), which indicates that its karyotype was derived in parallel with M. feae's from an ancestral karyotype of 2n ≥ 13/14. The newly discovered giant muntjac (Muntiacus vuquangensis) may represent another parallel reduction lineage from the ancestral 2n = 46 karyotype. Our phylogenetic results indicate that the giant muntjac is relatively closer to Muntiacus reevesi than to other muntjacs and may be placed in the genus Muntiacus. Analyses of sequence divergence reveal that the rate of change in chromosome number in muntjac deer is one of the fastest in vertebrates. Within the muntjac subfamily, the fastest evolutionary rate is found in the Fea's lineage, in which two species with different karyotypes diverged in around 0.5 Myr. [ABSTRACT FROM PUBLISHER]

Simple Summary: Inborn Errors of Immunity (IEI) are a heterogeneous group of disorders characterized by a defect in the function of at least one, and often more, components of the immune system. The overall risk for cancer in children with IEI ranges from 4 to 25%. Several factors, namely, age of the patient, viral infection status and IEI type can influence the development of different cancer types. Immunologists and oncologists should interact to monitor and promptly diagnose the potential development of cancer in known IEI patients, as well as an underlying IEI in newly diagnosed cancers with suggestive medical history or high rate of therapy-related toxicity. The creation of an international registry of IEI cases with detailed information on the occurrence of cancer is fundamental to optimizing the diagnostic process and to evaluating the outcomes of new therapeutic options, with the aim of improving prognosis and reducing comorbidities. Inborn Errors of Immunity (IEI) are a heterogeneous group of disorders characterized by a defect in the function of at least one, and often more, components of the immune system. The aim of this narrative review is to discuss the epidemiology, the pathogenesis and the correct management of tumours in patients with IEI. PubMed was used to search for all of the studies published over the last 20 years using the keywords: "inborn errors of immunity" or "primary immunodeficiency" and "cancer" or "tumour" or "malignancy". Literature analysis showed that the overall risk for cancer in children with IEI ranges from 4 to 25%. Several factors, namely, age of the patient, viral infection status and IEI type can influence the development of different cancer types. The knowledge of a specific tumour risk in the presence of IEI highlights the importance of a synergistic effort by immunologists and oncologists in tracking down the potential development of cancer in known IEI patients, as well as an underlying IEI in patients with newly diagnosed cancers. The unwanted emergence of neutralizing antibodies (nAbs) against an endogenous or a therapeutic protein can result in deficiency diseases or therapy failure. Here, we developed a cell-free microfluidic method for the sensitive detection and quantification of nAbs in human serum that are associated with infertility. We used cell-derived vesicles containing the luteinizing hormone (LH)/choriogonadotropin receptor (LHHCGR) to detect nAbs against LH. The method exploits the entire cellular signal amplification mechanism, and facilitates the detection of as little as 0.44 nM of LH-nAb (Kd 1.5 nM) in human serum matrix within only 15 minutes. In addition, dose-response curves can be generated in less than 2 hours to evaluate the nAB concentration and dissociation constant. The developed system is devoid of problems associated with cell-based assays and we believe that this simple effect-directed analysis can be used in clinical environments, and is adaptable to other hormones or cytokines and their respective nAbs. Abstract.— The saola, Pseudoryx nghetinhensis , was unknown to science until its formal description in 1993. This endangered species is a member of the ruminant artiodactyl family Bovidae (cattle, sheep, goats, and antelopes). However, given its puzzling combination of morphological traits, the specific affinities of Pseudoryx within Bovidae are controversial. A preliminary genetic investigation suggested that Pseudoryx should be placed in the subfamily Bovinae (cattle, buffaloes, spiral-horned antelopes, and nilgai), but a recent cladistic analysis of skeletal and dental characters allied Pseudoryx with caprine bovids (sheep, goats, musk oxen, goat antelopes, and Pantholops ). The morphological and molecular hypotheses differ in assigning the saola to either of the two most divergent clades of Bovidae. In this report, phylogenetic analyses of DNA sequences from five genes are used to test these alternatives. Protein coding regions, introns, and ribosomal DNAs from the nuclear and mitochondrial genomes discount the hypothesis that Pseudoryx is a close relative of Caprinae. Instead, combined analyses of the DNA data and published morphological evidence place Pseudoryx with Bovini (cattle and buffaloes), a subclade of Bovinae. In a separate analysis, the matrix of morphological characters links Pseudoryx with caprine bovids, but in the context of the molecular data, the gross anatomical evidence strongly supports a grouping of Pseudoryx with Bovinae. Surprisingly, the morphological partition provides the most character support in the combined analysis.
