Direct detection of highly reactive, atmospheric hydroxyl radicals (OH) is widely accomplished by laser-induced fluorescence (LIF) instruments. The technique is also suitable for the indirect measurement of HO2 and RO2 peroxy radicals by chemical conversion to OH. It requires sampling of ambient air into a low pressure cell, where OH fluorescence is detected after excitation by 308 nm laser radiation. Although the residence time of air inside the fluorescence cell is typically only on the order of milliseconds, there is potential that additional OH is internally produced, which would artificially increase the measured OH concentration. Here, we present experimental studies investigating potential interferences in the detection of OH and peroxy radicals for the LIF instruments of Forschungszentrum Jülich for nighttime conditions. For laboratory experiments, the inlet of the instrument was overflown by excess synthetic air containing one or more reactants. In order to distinguish between OH produced by reactions upstream of the inlet and artificial signals produced inside the instrument, a chemical titration for OH was applied. Additional experiments were performed in the simulation chamber SAPHIR where simultaneous measurements by an open-path differential optical absorption spectrometer (DOAS) served as reference for OH to quantify potential artifacts in the LIF instrument. Experiments included the investigation of potential interferences related to the nitrate radical (NO3, N2O5), related to the ozonolysis of alkenes (ethene, propene, 1-butene, 2,3-dimethyl-2-butene, α-pinene, limonene, isoprene), and the laser photolysis of acetone. Experiments studying the laser photolysis of acetone yield OH signals in the fluorescence cell, which are equivalent to 0.05 x 106 cm-3 OH for a mixing ratio of 5 ppbv acetone. Under most atmospheric conditions, this interference is negligible. No significant interferences were found for atmospheric concentrations of reactants during ozonolysis experiments. Only for α-pinene, limonene, and isoprene at reactant concentrations which are orders of magnitude higher than in the atmosphere artificial OH could be detected. The value of the interference depends on the turnover rate of the ozonolysis reaction. For example, an apparent OH concentration of approximately 1 x 106cm-3 is observed, if 5.8ppbv limonene reacts with 600ppbv ozone. Experiments with the nitrate radical NO3 reveal a small interference signal in the OH, HO2 and RO2 detection. Dependencies on experimental parameters point to artificial OH formation by surface reactions at the chamber walls or in molecular clusters in the gas expansion. The signal scales with the presence of NO3 giving equivalent radical concentrations of 1.1 x 105 cm-3 OH, 1 x 107 cm-3 HO2, and 1.7 x 107 cm-3 RO2 per 10 pptv NO3. [ABSTRACT FROM AUTHOR]

Ozone concentrations in the Po Valley of northern Italy often exceed international regulations. As both a source of radicals and an intermediate in the oxidation of most volatile organic compounds (VOCs), formaldehyde (HCHO) is a useful tracer for the oxidative processing of hydrocarbons that leads to ozone production. We investigate the sources of HCHO in the Po Valley using vertical profile measurements acquired from the airship Zeppelin NT over an agricultural region during the PEGASOS 2012 campaign. Using a 1-D model, the total VOC oxidation rate is examined and discussed in the context of formaldehyde and ozone production in the early morning. While model and measurement discrepancies in OH reactivity are small (on average 3.4 ± 13%), HCHO concentrations are underestimated by as much as 1.5 ppb (45%) in the convective mixed layer. A similar underestimate in HCHO was seen in the 2002-2003 FORMAT Po Valley measurements, though the additional source of HCHO was not identified. Oxidation of unmeasured VOC precursors cannot explain the missing HCHO source, as measured OH reactivity is explained by measured VOCs and their calculated oxidation products. We conclude that local direct emissions from agricultural land are the most likely source of missing HCHO. Model calculations demonstrate that radicals from degradation of this non-photochemical HCHO source increase model ozone production rates by as much as 0.6 ppb h-1 (12 %) before noon. A simple and easy liquid chromatographic procedure was developed for the separation and analysis of α,β-arteether. The developed method uses a reversed-phase Luna C18 column (250 mm × 4.6 mm), a hybrid micellar mobile phase containing 0.15 M SDS–6% (v/v) butanol–0.01 M phosphate buffered at pH 7, and detection at 218 nm. Under these conditions, the analysis time was less than 6 min. Validation studies were performed according to the International Conference on Harmonization guideline, including linearity (R2 > 0.9998) in the 0.1–50 µg/mL range, accuracy (99.8–100.8%), repeatability [RSD (%) 1.8–2.8], intermediate precision [RSD (%) 1.8–2.9], and robustness, which were found satisfactory. Method was applied to the determination of α,β-arteether in different pharmaceutical preparations and wrapper/label free ampoules obtaining recoveries in the 97.7–102.2% range. Compared to other chromatographic techniques, micellar liquid chromatography has the advantages of avoiding sample extraction from matrices, thus reducing the global analysis time and environmental toxicity. Micellar liquid chromatography (MLC) was first proposed as an attractive alternative to avoid the use of organic solvents. It was soon apparent that pure micellar solutions yield poor efficiencies. This problem was remediated by the addition of a small amount of an organic solvent. However, the general opinion of the poor peak shape has prevailed as a handicap for MLC, in spite of the fact that the hybrid mode often offers similar or even improved efficiencies (for basic compounds) relative to that attained in the hydro-organic mode. Only the efficiencies for apolar non-ionizable compounds are still clearly inferior. This work describes the type of interactions and polarity changes with organic solvent and surfactant adsorption, and the stationary phase architecture and wetting with micellar mobile phases, compared to hydro-organic mobile phases. The predominant term influencing band broadening in MLC appears to be stationary phase mass transfer. Organic solvents produce a thinner surfactant layer on the column, which permits better solute diffusion. Anionic surfactants suppress the silanol effect for basic compounds due to the protecting coating of the stationary phase by the surfactant, which seems to be more effective than the direct electrostatic interaction of amines with free silanols. Stress-induced volatile organic compound (VOC) emissions from transgenic Grey poplar modified in isoprene emission potential were used for the investigation of photochemical secondary organic aerosol (SOA) formation. In poplar, acute ozone stress induces the emission of a wide array of VOCs dominated by sesquiterpenes and aromatic VOCs. Constitutive light-dependent emission of isoprene ranged between 66 nmol m-2 s-1 in non-transgenic controls (wild type WT) and nearly zero (<0.5 nmol m-2 s-1) in isoprene emission-repressed plants (line RA22), respectively. Nucleation rates of up to 3600 cm-3 s-1 were observed in our experiments. In the presence of isoprene new particle formation was suppressed compared to non-isoprene containing VOC mixtures. Compared to isoprene/monoterpene systems emitted from other plants the suppression of nucleation by isoprene was less effective for the VOC mixture emitted from stressed poplar. This is explained by the observed high efficiency of new particle formation for emissions from stressed poplar. Direct measurements of OH in the reaction chamber revealed that the steady state concentration of OH is lower in the presence of isoprene than in the absence of isoprene, supporting the hypothesis that isoprenes' suppressing effect on nucleation is related to radical chemistry. In order to test whether isoprene contributes to SOA mass formation, fully deuterated isoprene (C5D8) was added to the stress-induced emission profile of an isoprene free poplar mutant. Mass spectral analysis showed that, despite the isoprene-induced suppression of particle formation, fractions of deuterated isoprene were incorporated into the SOA. A fractional mass yield of 2.3% of isoprene was observed. Abstract: The addition of the anionic surfactant sodium dodecyl sulphate (SDS) to hydro-organic mixtures of methanol, ethanol, propanol or acetonitrile with water yielded enhanced peak shape (i.e. increased efficiencies and symmetrical peaks) for a group of basic drugs (β-blockers) chromatographed with a Kromasil C18 column. The effect can be explained by the thin layer of surfactant associated to the hydrocarbon chain on the stationary phase in the presence of the organic solvents, which covers the free silanols on the siliceous support avoiding their interaction with the cationic basic drugs. These instead interact with the anionic head of the surfactant increasing their retention and allowing a more facile mass transfer. The peak shape behaviour with the four organic solvents (methanol, ethanol, propanol and acetonitrile) was checked in the presence and absence of SDS. The changes in peak broadening rate and symmetry inside the chromatographic column were assessed through the construction of peak half-width plots (linear relationships between the left and right half-widths at 10% peak height versus the retention time). The examination of the behaviour for a wide range of compositions indicated that the effect of acetonitrile in the presence of SDS is different from ethanol and propanol, which behave similarly. Acetonitrile seems to be superior to the alcohols in terms of peak shape, which can be interpreted by the larger reduction in the adsorbed surfactant layer on the C18 column. However, the decreased efficiencies observed at increasing surfactant concentration in the mobile phase should be explained by the reduction in retention times, more than by a change in the stationary phase nature. Seven commercially available: Zorbax C18, Kromasil C18, C8, cyano, phenyl, monolithic, and amino stationary phase columns, have been characterized and classified into broadly similar types to simplify column choice. The results were evaluated employing cluster analysis, which shows several interesting groups based on distances (Minkowski and Euclidean) in agreement with the manufacturer's claims: chain density of the stationary phase used, and the presence or not of a silica base. Finally, results showed that C18 columns offer the best chromatographic characteristics for separation and quantification of antibiotics in micellar liquid chromatography. [ABSTRACT FROM AUTHOR]

Nifedipine is a photosensitive compound that is converted into its 4-(2-nitrophenyl) pyridine and 4-(2-nitrosophenyl) pyridine homologue. In order to obtain the most adequate conditions for handling nifedipine solutions in the analytical laboratory, a number of studies on the decomposition of this compound were performed. A simple micellar liquid chromatographic procedure was described to determine nifedipine in different biological matrices such as serum and urine, and to control its decomposition. To perform the analysis, nifedipine was dissolved in 0.1 m SDS at pH 3 and chromatographed using a mobile phase containing 0.125 m SDS-3% pentanol, pH 3 on a C18 column and UV detection at 235 nm. The chromatographic analysis time was 8 min. The response of the drug for both biological matrices was linear in the 1-100 µg/mL range, with r2 > 0.997 at all times. Repeatability, intermediate precision (CV, %) and limits of quantification and detection (ng/mL) were 0.19, 4.3, 104 and 31 in serum and 0.81, 2.1, 136 and 41 in urine. The method developed here does not show interferences or matrix effects produced by endogenous compounds. Micellar media and mobile phases have the advantage of stabilising the compounds, thus preventing photodegradation and allowing the direct injection of biological samples.
