The pharmacokinetics of the sildenafil 100 mg orodispersible film (IBSA) was compared to that of the conventional marketed 100 mg film-coated tablet (Viagra®) after single-dose administration to 53 healthy male volunteers (aged 18–51 years) in a randomized, open, two-way crossover bioequivalence study. Each subject received a single oral dose of 100 mg of sildenafil as test or reference formulation administered under fasting conditions at each of the two study periods according to a randomized crossover design. There was a washout interval of ≥7 days between the two administrations of the investigational medicinal products. Blood samples for pharmacokinetic analysis were collected up to 24 h post-dosing. The primary objective was to compare the rate (peak plasma concentration; Cmax) and extent (area under the curve [AUC] from administration to last observed concentration time; AUC0–t) of sildenafil absorption after single-dose administration of test and reference. Secondary endpoints were observed to describe the plasma pharmacokinetic profiles of sildenafil and its metabolite N-desmethyl-sildenafil relative bioavailability and safety profile after single-dose administration. The mean sildenafil and N-desmethyl-sildenafil plasma concentration–time profiles up to 24 h after single-dose administration of sildenafil 100 mg orodispersible film and film-coated tablet were nearly superimposable. The bioequivalence test was fully satisfied for sildenafil and N-desmethyl-sildenafil in terms of rate and extent of bioavailability. Adverse events occurred at similar rates for the two formulations and were of mild-to-moderate severity. The results suggest that the new orodispersible film formulation can be used interchangeably with the conventional film-coated formulation. Abstract

The experimental observation we wish to explain and which is common in most of the polymethylene systems with no polar substituents is that the absorption intensity in the infrared of the CH2 wagging motions is extremely weak for solid all-trans chains and becomes much stronger in the liquid phase in which molecules are conformationally distorted. Using current theories for the interpretation of the infrared intensities, it is shown that the observed phenomenon arises from the fact that for all transplanar chains, a sizeable ‘‘σ conjugation’’ along the CC chain takes place. It is shown that ‘‘charge fluxes’’ from second nearest neighbors play the main role in determining such intensity behavior. [ABSTRACT FROM AUTHOR]

Abstract

In this paper the infrared intensities of formaldehyde d0 and d2 are interpreted on the basis of electro-optical parameters. It is shown that infrared intensities can be used as a powerful tool to electro-optical parameters. It is shown that infrared intensities can be used as a powerful tool to choose among different, even if reasonable, force fields: 16 force fields taken from literature have been examined and only a few of them have been found appropriate to fit infrared intensities. Both electrical and potential intramolecular interactions in CH2O support the hypothesis of a backdonation of electronic charge from the oxygen lone pairs to the CH bonds. Abstract

The aim of this work is to understand the molecular structural properties which may justify the great observed stability of doped alkoxy polythiophenes. Infrared frequency and intensity spectroscopy and Raman spectroscopy are used as probes. Suitably synthesized oligomers with increasing chain length and preassigned chemical structure have been studied together with several model molecules. The existence of through-space and through-bond S- - -O interactions has been revealed. The extent of the topologically dependent perturbation induced by the charge injection by the oxygen atoms is detected and measured. The inter- and intraring delocalizations have been monitored by the observation of the softening of the strongly Raman active ‘‘amplitude mode’’ and by the red shift of the electronic spectra. The conformation of the side chains is trans planar in the solid state and collapses in a liquidlike premelting state before the melting of the crystal. © 1996 American Institute of Physics. Abstract

The vibrational infrared and Raman spectra of a series of t-butyl substituted oligomers of para-phenylenevinylene and of the corresponding polymer in the pristine and potassium-doped states have been recorded for the first time and interpreted on the basis of the theory of the effective conjugation coordinate. The Raman spectra of selectively deuteriated materials have also been analyzed. Evidence is found of strong phonon confinement. The information derived from the spectra is that the charge carrier is confined over a few monomer units and that when the polymer is doped with electron donors the benzene ring is preferentially perturbed. The data collected in this work are compared with the results from UV spectroscopy and ESR experiments which indicate a larger dimension of the charge carrier. Using a recently developed method, we show here that solvent effects on the first molecular hyperpolarizabilities of push-pull polyenes can be predicted by evaluation of the relaxation contribution βr to the hyperpolarizability from vibrational infrared and Raman intensities and vibrational frequencies. The molecules studied in this work are a few organic polymethine dyes with different donor and acceptor groups. The analysis of their vibrational spectra confirms the key role played by the structural parameters of the polyene chain in determining the nonlinear optical response. © 1995 American Institute of Physics. Abstract

Mozambique’s tourism sector could play a key role in the country’s socio-economic development, especially in the region of Cabo Delgado where the demand for tourist services is expected to increase. Nonetheless, several constraints (e.g. the lack of adequate training) are hindering the capacity of local people to take full advantage of this opportunity. Qualitative research has been performed in order to align vocational training programmes with the needs of the emerging tourism sector. Local and foreign key informants were interviewed in order to gain a better understanding of training needs and to gain insights into developing training programmes that can enhance local people’s employability. The main findings highlight the lack of symbolisation of tourism and its correlates from local communities. Therefore, rather than just delivering technical skills, training programmes should also promote a ‘culture of tourism’ and a more favourable attitude towards ‘working in the tourism sector’.
