Abstract Sea ice is a critical component of the Earth’s Climate System and a unique habitat. Sea-ice changes prior to the satellite era are poorly documented, and proxy methods are needed to constrain its past variability. Here, we demonstrate the potential of sedimentary DNA from Polarella glacialis, a sea-ice microalga, for tracing past sea-ice conditions. We quantified P. glacialis DNA (targeting the nuclear ribosomal ITS1 region) in Arctic marine and fjord surface sediments and a sediment core from northern Baffin Bay spanning 12,000 years. Sea ice and sediment trap samples confirmed that cysts of P. glacialis are common in first-year sea ice and sinking particulate matter following sea-ice melt. Its detection is more efficient with our molecular approach than standard micropaleontological methods. Given that the species inhabits coastal and marine environments in the Arctic and Antarctic, P. glacialis DNA has the potential to become a useful tool for circum-polar sea-ice reconstructions.

Abstract Background Evidence suggests that pathophysiological conditions such as obesity and type 2 diabetes (T2D) are associated with morphologic and metabolic alterations in the small intestinal mucosa. Exploring these alterations generally requires invasive methods, limiting data acquisition to subjects with enteropathies or undergoing bariatric surgery. We aimed to evaluate small intestine epithelial cell homeostasis in a cohort of men covering a wide range of adiposity and glucose homoeostasis statuses. Methods Plasma levels of citrulline, a biomarker of enterocyte mass, and I-FABP, a biomarker of enterocyte death, were measured by UHPLC‑MS and ELISA in 154 nondiabetic men and 67 men with a T2D diagnosis. Results Plasma citrulline was significantly reduced in men with insulin resistance and T2D compared to insulin sensitive men. Decreased citrulline levels were, however, not observed in men with uncontrolled metabolic parameters during T2D. Plasma I-FABP was significantly higher in men with T2D, especially in presence of uncontrolled glycemic and lipid profile parameters. Integration of both parameters, which estimate enterocyte turnover, was associated with glucose homeostasis as well as with T2D diagnosis. Differences in biomarkers levels were independent of age and BMI and glucose filtration rates. Conclusions Our study supports a decreased functional enterocyte mass and an increased enterocyte death rate in presence of metabolic alterations but emphasizes that epithelial cell homeostasis is especially altered in presence of severe insulin resistance and T2D. The marked changes in small intestine cellularity observed in obesity and diabetes are thus suggested to be part of gut dysfunctions, mainly at an advanced stage of the disease.

Unusually warm conditions recently observed in the Pacific Arctic region included a dramatic loss of sea ice cover and an enhanced inflow of warmer Pacific-derived waters. Moored sediment traps deployed at three biological hotspots of the Distributed Biological Observatory (DBO) during this anomalously warm period collected sinking particles nearly continuously from June 2017 to July 2019 in the northern Bering Sea (DBO2) and in the southern Chukchi Sea (DBO3), and from August 2018 to July 2019 in the northern Chukchi Sea (DBO4). Fluxes of living algal cells, chlorophyll a (chl a), total particulate matter (TPM), particulate organic carbon (POC), and zooplankton fecal pellets, along with zooplankton and meroplankton collected in the traps, were used to evaluate spatial and temporal variations in the development and composition of the phytoplankton and zooplankton communities in relation to sea ice cover and water temperature. The unprecedented sea ice loss of 2018 in the northern Bering Sea led to the export of a large bloom dominated by the exclusively pelagic diatoms Chaetoceros spp. at DBO2. Despite this intense bloom, early sea ice breakup resulted in shorter periods of enhanced chl a and diatom fluxes at all DBO sites, suggesting a weaker biological pump under reduced ice cover in the Pacific Arctic region, while the coincident increase or decrease in TPM and POC fluxes likely reflected variations in resuspension events. Meanwhile, the highest transport of warm Pacific waters during 2017-2018 led to a dominance of the small copepods Pseudocalanus at all sites. Whereas the export of ice-associated diatoms during 2019 suggested a return to more typical conditions in the northern Bering Sea, the impact on copepods persisted under the continuously enhanced transport of warm Pacific waters. Regardless, the biological pump remained strong on the shallow Pacific Arctic shelves.

The shallow Pacific Arctic shelf has historically acted as an effective carbon sink, characterized by tight benthic pelagic coupling. However, the strength of the biological carbon pump in the Arctic has been predicted to weaken with climate change due to increased duration of the open-water period for primary production, enhanced nutrient limitation, and increased pelagic heterotrophy. In order to gain insights into how the biological carbon pump is functioning under the recent conditions of extreme warming and sea ice loss on the Pacific Arctic shelf, we measured sinking particulate organic carbon (POC) fluxes with drifting and moored sediment traps, as well as rates of primary production and particle-associated microbial respiration during June 2018. In Bering Shelf/Anadyr Water masses, sinking POC fluxes ranged from 0.8 to 2.3 g C m–2 day–1, making them among the highest fluxes ever documented in the global oceans. Furthermore, high export ratios averaging 82% and low rates of particle-associated microbial respiration also indicated negligible recycling of sinking POC in the water column. These results highlight the extraordinary strength of the biological carbon pump on the Pacific Arctic shelf during an unusually warm and low-sea ice year. While additional measurements and time are needed to confirm the ultimate trajectory of these fluxes in response to ongoing climate change, these results do not support the prevailing hypothesis that the strength of the biological carbon pump in the Pacific Arctic will weaken under these conditions.

Sea ice in the Arctic Ocean (AO) has been undergoing dramatic changes during the last two decades. In addition, the water temperature of the inflow of Atlantic water masses at the gateway Fram Strait has recently increased. Long-term data may help to evaluate the impact of these physical changes on the biological processes in surface waters. Over a 25-year period, and mostly in summer, water samples were collected at discrete depths within the uppermost 100 m of the Fram Strait and other regions of the AO to investigate chlorophyll a (Chl a) and particulate organic carbon (POC) standing stocks. Stations sampled from 1991 to 2015 were located in the Fram Strait, Barents Sea (BS), on the Eurasian shelf, and over the Nansen, Amundsen, and parts of the Amerasian basins (AB). Discrete Chl a and POC measurements obtained during 33 and 24 expeditions, respectively, were integrated over the upper 100 m of the water column to monitor spatial and interannual variations in distribution patterns of standing stocks. In general, the highest Chl a and POC standing stocks were observed in the eastern Fram Strait (EFS) and in the BS, while the lowest biomasses were observed in the heavily ice-covered regions of the central AO, mainly in the Amundsen and ABs. Whereas summertime Chl a stocks sharply decreased northward from the Fram Strait and Barents Sea toward high latitudes, the decline in POC standing stocks was less pronounced. Over the sampling period, summertime Chl a stocks slightly increased in the EFS but remained more or less constant in the central AO. In contrast to Chl a, standing stocks of POC eventually increased over the last 25 years in the central AO, possibly as an effect of increasing air temperatures, decreasing sea ice extent and thickness, and increasing light availability. Moreover, variations in riverine discharge and in sea ice export within the Transpolar Drift may have contributed to the enhanced POC stock in the central AO surface waters. Overall, the objective of the present study was to provide baseline datasets of Chl a and POC to better track the effects of environmental changes due to global warming on the Arctic pelagic system.

Baffin Bay, located at the Arctic Ocean’s ‘doorstep’, is a heterogeneous environment where a warm and salty eastern current flows northwards in the opposite direction of a cold and relatively fresh Arctic current flowing along the west coast of the bay. This circulation affects the physical and biogeochemical environment on both sides of the bay. The phytoplanktonic species composition is driven by its environment and, in turn, shapes carbon transfer through the planktonic food web. This study aims at determining the effects of such contrasting environments on ecosystem structure and functioning and the consequences for the carbon cycle. Ecological indices calculated from food web flow values provide ecosystem properties that are not accessible by direct in situ measurement. From new biological data gathered during the Green Edge project, we built a planktonic food web model for each side of Baffin Bay, considering several biological processes involved in the carbon cycle, notably in the gravitational, lipid, and microbial carbon pumps. Missing flow values were estimated by linear inverse modeling. Calculated ecological network analysis indices revealed significant differences in the functioning of each ecosystem. The eastern Baffin Bay food web presents a more specialized food web that constrains carbon through specific and efficient pathways, leading to segregation of the microbial loop from the classical grazing chain. In contrast, the western food web showed redundant and shorter pathways that caused a higher carbon export, especially via lipid and microbial pumps, and thus promoted carbon sequestration. Moreover, indirect effects resulting from bottom-up and top-down control impacted pairwise relations between species differently and led to the dominance of mutualism in the eastern food web. These differences in pairwise relations affect the dynamics and evolution of each food web and thus might lead to contrasting responses to ongoing climate change.

An assessment of the production, distribution and fate of highly branched isoprenoid (HBI) biomarkers produced by sea ice and pelagic diatoms is necessary to interpret their detection and proportions in the northern Bering and Chukchi Seas. HBIs measured in surface sediments collected from 2012 to 2017 were used to determine the distribution and seasonality of the biomarkers relative to sea ice patterns. A northward gradient of increasing ice algae deposition was observed with localized occurrences of elevated IP25 (sympagic HBI) concentrations from 68-70°N and consistently strong sympagic signatures from 71-72.5°N. A declining sympagic signature was observed from 2012 to 2017 in the northeast Chukchi Sea, coincident with declining sea ice concentrations. HBI fluxes were investigated on the northeast Chukchi shelf with a moored sediment trap deployed from August 2015 to July 2016. Fluxes of sea ice exclusive diatoms (Nitzschia frigida and Melosira arctica) and HBI-producing taxa (Pleurosigma, Haslea and Rhizosolenia spp.) were measured to confirm HBI sources and ice associations. IP25 was detected year-round, increasing in March 2016 (10 ng m-2 d-1) and reaching a maximum in July 2016 (1331 ng m-2 d-1). Snowmelt triggered the release of sea ice algae into the water column in May 2016, while under-ice pelagic production contributed to the diatom export in June and July 2016. Sea ice diatom fluxes were strongly correlated with the IP25 flux, however associations between pelagic diatoms and HBI fluxes were inconclusive. Bioturbation likely facilitates sustained burial of sympagic organic matter on the shelf despite the occurrence of pelagic diatom blooms. These results suggest that sympagic diatoms may sustain the food web through winter on the northeast Chukchi shelf. The reduced relative proportions of sympagic HBIs in the northern Bering Sea are likely driven by sea ice persistence in the region.

Between Greenland and Spitsbergen, Fram Strait is a region where cold ice-covered Polar Water exits the Arctic Ocean with the East Greenland Current (EGC) and warm Atlantic Water enters the Arctic Ocean with the West Spitsbergen Current (WSC). In this compilation, we present two different data sets from plankton ecological observations in Fram Strait: (1) long-term measurements of satellite-derived (1998–2012) and in situ chlorophyll a (chl a) measurements (mainly summer cruises, 1991–2012) plus protist compositions (a station in WSC, eight summer cruises, 1998–2011); and (2) short-term measurements of a multidisciplinary approach that includes traditional plankton investigations, remote sensing, zooplankton, microbiological and molecular studies, and biogeochemical analyses carried out during two expeditions in June/July in the years 2010 and 2011. Both summer satellite-derived and in situ chl a concentrations showed slight trends towards higher values in the WSC since 1998 and 1991, respectively. In contrast, no trends were visible in the EGC. The protist composition in the WSC showed differences for the summer months: a dominance of diatoms was replaced by a dominance of Phaeocystis pouchetii and other small pico- and nanoplankton species. The observed differences in eastern Fram Strait were partially due to a warm anomaly in the WSC. Although changes associated with warmer water temperatures were observed, further long-term investigations are needed to distinguish between natural variability and climate change in Fram Strait. Results of two summer studies in 2010 and 2011 revealed the variability in plankton ecology in Fram Strait.

Abstract: Purpose: To validate quantitative and qualitative criteria of normal healthy skin using high-resolution MR imaging. Materials and methods: FIESTA and spin echo sequences of the skin of the heel, back and calf were obtained in 31 healthy volunteers. A dedicated 3-in. coil for study of the skin was used to optimize reception sensitivity. Quantitative analysis was performed to measure skin thickness in these three sites and qualititative analysis aimed to differentiate the various skin layers. Results: With both sequences, the stratum corneum appeared as a hyperintense layer. The epidermis was visualized appeared as a fine, homogeneous, hyperintense line. The dermoepidermal junction was clearer particularly in the calf. The dermis was also identified in each sequence and in each anatomic site. The signal was hypointense in both sequences, homogeneous with spin echo and more heterogeneous with FIESTA. Moreover the interface between the papillary and the reticular dermis could be distinguished. The hypodermis was visualized in both sequences and in all sites and hypodermal inclusions in the dermis were seen particularly in the back and calf. Lastly, the pilosebaceous follicles and the deep vascular network of the reticular dermis were more clearly seen with FIESTA sequence. Measures of overall skin thickness and for each layer according to site, sex and MRI sequence were performed. Statistical analysis was then performed to seek significant differences between the results according to these parameters. Conclusion: MR imaging provides global analysis with high resolution of the various skin layers down to the hypodermis and the muscular fascia. [Copyright &y& Elsevier]
