Disaster victim identification (DVI) refers to the forensic identification of unknown individuals following a mass disaster event. Human dental structures can contain viable DNA sources when other soft tissues are compromised. However, labor‐intensive sample preparation performed by extensively trained personnel is needed to expose the nuclear material for traditional forensic DNA workflows. With this in mind, we evaluated two simplified sample preparation protocols for processing tooth samples using either a conventional forensic DNA workflow or the Applied Biosystems® RapidHIT™ ID instrument. Briefly, sample sets for both protocols included 10 deciduous teeth that were cleaned prior to either fragmentation with a claw hammer (for RapidHIT™ ID processing) or fine‐powder pulverization with a consumer‐grade coffee grinder (for traditional workflows). The average percentage of expected STR alleles that were detected above analytical threshold for these tooth samples were comparable between methods: RapidHIT™ ID = 99.0% and GlobalFiler™ = 99.8%. Average intralocus heterozygote peak height ratios (PHRs) were comparable: RapidHIT™ ID = 0.80 and GlobalFiler™ = 0.86. Importantly, 9 of 10 samples analyzed via the RapidHIT™ ID required analyst review for flagged artifact peaks and quality issues. Across all profiles, 91% of alleles passed quality metrics for the RapidHIT™ workflow versus 100% for conventional GlobalFiler™ analysis. Collectively, these results suggest that quick, low‐tech tooth sample fragmentation followed by analysis with the RapidHIT™ ID instrument can produce complete STR profiles from aged tooth samples. Insects have long played a role in forensic investigations and can be used to estimate minimum time since death, corpse translocation, and link an individual to a crime scene. Bed bugs (Cimex lectularius) are wingless ectoparasitic insects of potential forensic utility, given that all mobile life stages feed on vertebrate blood. Successful profiling of autosomal short tandem repeats (STRs) from human DNA isolated from bed bugs has been previously reported. This proof‐of‐concept study looked to expand this work and determine any possible limitations of using bed bugs for both rapid stain identification (RSID™) for human blood and Y‐STR profiling. To achieve this, bed bugs were fed either human male only or human pooled (female:male) blood for 30 min and subsequently collected at 12‐h intervals up to 108 h post‐blood meal (PBM). RSID™ blood testing was successful from the bed bug carcass remaining after DNA isolation, regardless of blood meal type and time of collection PBM. Complete Y‐STR profiles were generated from bed bugs <60 h PBM. As the time PBM increased, DNA quantity decreased, while the degradation index increased. Collection of bed bugs at a crime scene could provide a valuable source of human blood for Y STR profiling and be used to link an individual to a crime scene or for potential male suspect exclusion. Future studies should look to replicate the results of this proof‐of‐concept study with larger numbers of bed bugs, more diverse blood donors, and additional STR profiling kits. The investigation of criminal offenses requires modern, up-to-date, practical means. The ANDE Rapid DNA Identification System is a new technology that generates a DNA ID from forensic samples in less than two hours. In today’s conditions, its use in forensics, practical activities of law enforcement agencies on a permanent basis is very relevant. ANDE technology is specially designed and proven to be robust, mobile and suitable for use in harsh environments. The National Police of Ukraine is tasked with the enormous task of identifying victims of horrific crimes related to the ongoing war with russia. Over the past year, it has been proven that a significant percentage of these victims cannot be identified through forensic DNA alone, given her critical condition. The enormous amount of forensic DNA processing work required to address this situation far exceeds the capabilities of traditional DNA laboratory infrastructure. In addition, the logistical overhead associated with managing the efficient storage, movement, tracking, and provenance of large numbers of bodies or even DNA samples across wide geographic areas causes significant wastage of critical resources and long delays. ANDE technology accelerates DNA processing from months or even years to less than two hours. Forensic specialists use mobile DNA laboratories to identify dead. Today, the National Police operates 32 mobile laboratories. Rapid DNA’s secure, field-ready mobile systems provide the most accurate and efficient DNA identification results. Thus, the main goal of this study is to provide practical recommendations for forensic support for the use of rapid DNA profiling technology, using Rapid DNA as an example, in pre-trial investigation. This study measures the similarity of the short tandem repeat (STR) profile of human DNA. The similarity measurement had been done to the STR value of the allele loci in DNA profile between the query's DNA to the reference's DNA profile. The measurements were conducted on 27 DNA profile loci including the Y chromosome loci (YSTR). The YSTR loci were used as the main comparison of similarity measurements to determine the biological kinship relationship between the query DNA profile and the alleged male biological family. To measure the similarity of two STR values that have shifted due to several factors in the DNA source extraction process, a fuzzy similarity measure was used. The STR values of the DNA profile loci are described as triangular fuzzy numbers. Similarity value of the STR is the intersection of two isosecle that been compared. To conclude that the query has a biological relationship with the male reference, the similarity of the YSTR locus is equal or more than 0.75 and the similarity value of the other 24 DNA profile loci is greater or equal to 0.5. Background. Traditionally, the description of grape varieties is a task of ampelographic studies. However, several different grape cultivars have similar phenotypic traits. Molecular genetic characterization is the most accurate tool for cultivar identification. The development of DNA fingerprinting of varieties is the first step in this direction. An extensive database of DNA profiles of grape genotypes makes it possible to determine the varietal affiliation of unknown forms, confirm or refute the varietal correspondence of planting material. ‘Khatmi’ is an autochthonous grape variety from Dagestan. The profile of ‘Khatmi’ is presented in the VIVC international database of DNA fingerprints for grape varieties. However, an application of DNA analysis methods in grape variety studies has determined that several ancient varieties were cultivated under one name, while for others a certain variability of genotypes was observed. The objectives of the work were to study samples of ‘Khatmi’ plants from different places of growth in Dagestan by standard microsatellite loci used for grape genotyping, to assess the level of genetic similarity of the samples, and to refine the DNA profile of ‘Khatmi’. Materials and methods. Molecular genetic study was carried out on 10 samples from different ‘Khatmi’ populations. The material was picked from the collection sites of Dagestan breeding experimental station of viticulture and vegetable growing and the Dagestan Experiment Station of VIR, as well as from production plantations. DNA was extracted from herbarium specimens of young grape shoot tips by the CTAB method. The samples were genotyped by polymerase chain reaction using a standard set of primers for 9 microsatellite markers VVS2, VVMD5, VVMD7, VVMD25, VVMD27, VVMD28, VVMD32, VrZAG62 and VrZAG79 recommended by the International Organization of Grapes and Wine (OIV) for grapevine DNA fingerprinting. Amplification products were separated, and their sizes were assessed using capillary electrophoresis on an ABI Prism 3130 genetic analyzer. Results. Genotyping was done for 10 samples of grapes growing in Dagestan under the name ‘Khatmi’, including samples from different collections and places of industrial cultivation, as well as clonal variations of this variety and putative clonal variations. The two base pair differences within one of the loci distinguished the DNA profiles of the analyzed samples from that of ‘Khatmi’ presented in the international grape varieties database VIVC. It was determined that the sample under the name ‘Khatmi krupnoyagodnyi’ is closely related to ‘Khatmi’ variety by its genotype, but probably represents a clonal variation of ‘Koz uzyum’, another local variety of Dagestan. Conclusion. The DNA profile of the local Dagestan grape variety ‘Khatmi’ has been refined.

The letter to the Editor is in response to the manuscript by Hertzog et al. (2023) titled "Data management instruments to Protect the personal information of Children and Adolescents in sub-Saharan Africa." The letter elaborates on personal data protection, particularly the POPI Act's data management requirements; the DNA Act mandates specific measures to ensure the data integrity and security of the NFDD's information. In addition, it criminalises the misuse or compromise of the data's integrity within the NFDD. In addition, the DNA Act established the National Forensic Oversight and Ethical Board (NFOEB), which is responsible for overseeing ethical compliance, implementing the Act, and preserving data integrity within the NFDD. The NFOEB is also responsible for investigating any complaints regarding DNA forensics and the management of the NFDD.

Forensic DNA profiles are established by multiplex PCR amplification of a set of highly variable short tandem repeat (STR) loci followed by capillary electrophoresis (CE) as a means to assign alleles to PCR products of differential length. Recently, CE analysis of STR amplicons has been supplemented by high-throughput next generation sequencing (NGS) techniques that are able to detect isoalleles bearing sequence polymorphisms and allow for an improved analysis of degraded DNA. Several such assays have been commercialised and validated for forensic applications. However, these systems are cost-effective only when applied to high numbers of samples. We report here an alternative, cost-efficient shallow-sequence output NGS assay called maSTR assay that, in conjunction with a dedicated bioinformatics pipeline called SNiPSTR, can be implemented with standard NGS instrumentation. In a back-to-back comparison with a CE-based, commercial forensic STR kit, we find that for samples with low DNA content, with mixed DNA from different individuals, or containing PCR inhibitors, the maSTR assay performs equally well, and with degraded DNA is superior to CE-based analysis. Thus, the maSTR assay is a simple, robust and cost-efficient NGS-based STR typing method applicable for human identification in forensic and biomedical contexts. [ABSTRACT FROM AUTHOR]

Criminal activity is on the rise in Nigeria. The nefarious activity of bandits is on the increase with kidnapping of students and attack on farmers. This creates fear in the terrorized communities. Armed robbery as well as rape of citizens going about their normal duties is also on the increase. The road is becoming increasingly unsafe for the citizenry as well as government officials. The government has mandated individuals to register in the National Identification Number NIN which is linked to the Bank Verification Number (BVN) (a database established to curb crime in the banking sector) and the Subscriber Identity module (SIM). The Establishment of such databases is pertinent however the NIN, BVN, SIM-NIN linkage and fingerprints may not be sufficient in solving criminal cases without DNA profiling of convicted individuals, establishment of forensic laboratories and the creation of criminal DNA profile database CRODER -- a Criminal offender DNA Exert Repertoire. The role of DNA profiling as an additional measure is to keep a record of all convicted individuals and the CRODER will be an additional useful measure in profiling criminals and individuals can be adopted as a forensic tool in addition to the captured databases of NIN, BVN and NIN-SIM and will be on the watch list of security operatives. The CRODER forensic tool can be done stepwise by availability of an emergency number to dial for emergency, a dispatch personnel to respond to the call, and vigilante groups or other adhoc security officer in the vicinity of the emergency call to get to the vicinity and preserve the crime scene environment for investigators and detectives to work on. CRODER Forensic tool can then be adopted to solve the crime. Of fenders trying to escape enroute the airport, can be checked via CRODER and fingerprint compared and matched with the collection in the database. The CRODER may be a necessary stride to bring kidnappers, bandits, and armed robbers to book in record time and curb the rise in criminal activity, insurgency, banditry, and insecurity in general. Rapid DNA technology is being utilized for reference profiles worldwide. There is also strong data in the literature to support its use for high‐template DNA sources, the same is not true for low‐template sources, such as touch DNA; this is a requirement before wider implementation to forensic casework is considered. We report on the Rapid HIT Intel cartridge's ability to facilitate successful amplification of touch DNA to obtain profiles from template deposited on items commonly encountered in forensic casework. Eight items were touched in ten replicates— two were tapelifted, three swabbed, and three directly inserted. Significance was observed in the alleles amplified and RFU with respect to sample type. Three samples performed well: cable tie, fabric, and matchstick. As two of these were directly inserted, this should be considered for any sample small enough. Placement of highly absorbent substrates into the cartridge is not advised as it can cause a lysate‐pull error. Heterozygote loci often presented as homozygous (32%–78% loci per profile); this was influenced by substrate type and profile RFU. Loci with larger masses exhibited higher false homozygosity also. Comparison of the donor's profile analyzed was performed against previous datasets analyzing touch DNA through standard workflow, including manual DNA extraction, PCR, and CE separation. These data show that for all substrates, except for a fabric swatch, standard processing is preferential to Rapid HIT analysis. The article deals with the topical issue of the current information age, related to the legal protection of genetic data as a special category of personal data. This article presents an analysis of the current legal regulation of DNA processing for forensic purposes in the context of the legal protection of genetic data. The processing of DNA profiles for forensic purposes is an issue to which the Office for Personal Data Protection has paid considerable attention, especially with regard to the constant topicality and sensitivity of this topic. Therefore, the article also focuses on the Office's decision-making practice in matters of processing DNA profiles for forensic purposes in the light of the case law of administrative courts. The author considered both some particular consequences of the impact of current legal regulation as well as the subjective perception of these impacts. The aim of the article is to contribute to raising the awareness about the risks of the processing of DNA profiles for forensic purposes based on the current legal regulation and to explain the importance of applying the principle of proportionality as one of the key principles of personal data protection in the processing of genetic data. Based on the analysis, the article concludes that in the case of DNA processing for forensic purposes by the Czech Police, it is therefore necessary to balance the interest of the police and the general public in clarifying crime with the right to privacy, especially personal data protection. DNA amplification is known to be the most expensive step during forensic DNA analysis. This study evaluated the half-reaction amplification protocol (12.5 µL PCR product) using DNA amplification kits from Promega PowerPlex® (PowerPlex® 18D System, PowerPlex ®21System, PowerPlex® Fusion System and PowerPlex® Y23 System), which might aid in reducing sample analysis cost by half and allow the analysis of more samples. A sensitivity study (15 samples) along with testing of various blood stain samples (n=100) that were submitted to the Medico-Legal Directorate laboratory for DNA testing was accomplished to compare the DNA profiles resulting from half-reaction volume procedure to those with full-reaction volume procedure, using three differed methods along with standard protocol to evaluate the effect of half reaction volume with some variables. Results demonstrated the use of half-reaction amplification protocol preceded by washing step for all aforementioned DNA amplification kits gave a robust and reliable amplification result that aid to increase the number of samples analyzed and decreased the test cost for each kit without compromising the quality of 3DNA profiles obtained.

In recent years, several forensic laboratories have noted an increase in the number of sexual assault cases submitted for testing, often leading to longer turnaround times. In that context, forensic laboratories may be interested in reviewing their procedures to increase productivity. Here, we present two different strategies that were put in place in our laboratory. First, we changed the way sexual assault evidence kits (SAEK) are processed by implementing an optimized workflow that prioritizes the internal samples (vaginal, anal, and oral). This new procedure allowed for a drastic decrease in turnaround time, while maintaining a similar investigative power.Secondly, we used data from casework to target cases and samples that were likely to yield biological material from the perpetrator, in an attempt to avoid dedicating time and effort to cases for which there is a very low probability of obtaining foreign DNA evidence. Among other things, we looked at the likelihood of obtaining DNA from the perpetrator when the complainant reported the use of a condom, has showered after the assault or when the complainant has no memory of the assault. Results show that those circumstances do not dramatically decrease the probability of finding DNA from the perpetrator.

Summary: The domestic cat (Felis silvestris catus) is a valued companion animal throughout the world. Over 60 different cat breeds are accepted for competition by the cat fancy registries in different countries. Genetic markers, including short tandem repeats and SNPs, are available to evaluate and manage levels of inbreeding and genetic diversity, population and breed structure relationships, and individual identification for forensic and registration purposes. The International Society of Animal Genetics (ISAG) hosts the Applied Genetics in Companion Animals Workshop, which supports the standardization of genetic marker panels and genotyping for the identification of cats via comparison testing. SNP panels have been in development for many species, including the domestic cat. An ISAG approved core panel of SNPs for use in cat identification and parentage analyses is presented. SNPs (n = 121) were evaluated by different university‐based and commercial laboratories using 20 DNA samples as part of the ISAG comparison testing procedures. Different SNP genotyping technologies were examined, including DNA arrays, genotyping‐by‐sequencing and mass spectroscopy, to select a robust and efficient panel of 101 SNPs as the ISAG core panel for cats. The SNPs are distributed across all chromosomes including two on the X chromosome and an XY pseudo‐autosomal sexing marker (zinc‐finger XY; ZFXY). A population study demonstrated that the markers have an average polymorphic information content of 0.354 and a power of exclusion greater than 0.9999. The SNP panel should keep testing affordable while also allowing for the development of additional panels to monitor health, phenotypic traits, hybrid cats and highly inbred cats. Incest is a sexual activity that happens between close family members that are not allowed to get married due to customs or law. In a case of incest between a brother and his married sister, a paternity test and a statistical analysis were performed at the laboratories of the Paternity and Kinship Division of the Medico-legal Directorate Baghdad, Iraq. Blood samples were taken from the concerned subjects in this case (newborn baby, mother, alleged father, and the husband) and placed on FTA cards. DNA extraction was done using Chelex®, then the amplification of extracted DNA was carried out using an AmpFlSTR® Identifiler kit. PCR products were run with a 3130xl Genetic Analyzer, and the data were analyzed with Gene-Mapper ID® Analysis Software V.3.2 software.The analysis of DNA profiles using 15 loci as well as the statistical analysis for calculating the paternity index confirmed the allegation of the brother-sister incest, since the baby inherited all the obligate alleles from the alleged father (suspected brother). With a 99.9998% probability of paternity, these results showed that even in the case of brother-sister incest, paternity could be proved using 15 DNA locus with a high rate of certainty.

The article discusses recent judgments of the European Court of Human Rights on the collection of personal data for preventing, detecting or investigating criminal offences. The article delves into three national frameworks permitting the police to process, for a period or indefinitely, several categories of data in respect of persons who were suspected of the offences of varying gravity and were (not) convicted of them. It transpires from the case law that the States' margin of appreciation (i) was narrowed for indefinite processing of a convicted person's DNA profile, (ii) was not overstepped on account of processing – for several years – a non-convicted person's less sensitive data, where the decision-making was based on that person's previous criminal convictions and where the law contained adequate safeguards. Relying on their polymorphic nature, short tandem repeats (STRs) have been extensively studied and routinely utilized as human identity markers in forensic genetics. However, even the most comprehensive STR multiplexes in use today are limited in their ability to determine the number and appropriation of component contributor alleles in DNA mixtures. The difficulty in parsing out individuals in DNA mixtures is a consequence of overlapping length‐based alleles genotyped using the polymerase chain reaction (PCR) coupled with capillary electrophoresis (CE). Many challenges exist in the resolution of minor alleles (i.e., the alleles originating from a minor contributor) from stutter and stochastic effects (e.g., inherent heterozygote peak imbalance, undetected alleles [drop out]) in a given DNA profile, in addition to the complex statistical models and algorithms necessary to render DNA mixtures interpretable from a forensic casework standpoint. Therefore, we can either adapt to complex interpretation methods, or pursue new bench science approaches to DNA mixture deconvolution. One promising area of research includes the incorporation of additional highly polymorphic STR loci to compliment current marker multiplexes and offer great potential to improve the forensic genetic analysis of DNA mixtures. The introduction of molecular markers, such as STR (Short Tandem Repeats), was a fundamental event in Forensic Genetics. It was possible to obtain individual genetic profiles that allowed to properly identify individuals with small probability of error. Furthermore, police officers soon discovered the investigative value provided by DNA databanks, defined as a collection of individual DNA profiles usually used to investigate crimes and identify suspects. In this article, the authors present the most common use of these police databanks, comment their drawbacks and how to overcome them. The usage and value of police DNA databanks in special criminal situations are also explained: cold cases, familial searches and dragnets. These three particular situations are described and commented in this article. In sum­mary, although several technical, ethical-legal and internation­al aspects have to be improved, police DNA databanks are really powerful investigation tools that allow solving a large number of criminal cases.

The aim of this research is to investigate the relationship between DNA profiles of schools and the levels of teachers’ academic optimism. The population of this research which is a correlational design is composed of the teachers working in Uşak and the sample of the study is consisted of 333 teachers. Convenience sampling was used in the research. The data were collected through ‘Organizational DNA Scale’ and ‘School Academic Optimism Scale.’ The data were analyzed by using arithmetic mean, one- way ANOVA, Pearson correlation coefficient and multiple regression analysis. The results of the study indicated that teachers thought that schools had the DNA profiles of resilient organization at high level, military organization at medium level and full- time organization. In addition, it was revealed that the level of teachers’ academic optimism was at medium level. There was a medium- level, positive- oriented, significant relationship between teachers’ perceptions of resilient and just in time organization profiles regarding the schools and the level of academic optimism. Moreover, it was found in the research that teachers’ perceptions of military and just in time organization profiles concerning the schools predicted their academic optimism levels. Accordingly, it has been foreseen that academic optimism perceptions of teachers who considered their organizations as healthy DNA profiles may increase. Therefore, it has been recommended that DNA profiles of the schools must be identified and structured as healthy DNA profiles in order to enhance the perception of academic optimism representing teachers’ beliefs towards student achievement. The purpose of this research by the way of investigating the molecular genetic structure of a highly variable fungal phytopathogen Pyricularia oryzae Cav., to determine effective genes for the development of a strategy for immunogenetic protection against rice blast in conditions of epiphytotic development of the disease in the south of Russia, which would combine high efficacy with both environmental friendliness and resource and energy saving, to ensure country's food security. The knowledge of local pathotype diversity of Pyricularia oryzae Cav. and the (a)virulence genes in rice-growing regions of Russia may allow the prediction of new races and its interaction in local agro-ecology. The identification of virulence gene may become an indispensable theoretical basis for the development of genetic sources with long-lasting resistance to rice blast. Based on molecular and genetic approaches, the genetic structure and biodiversity of the phytopathogenic fungus Pyricularia oryzae Cav. in the south of Russia were considered. The monitoring was studied and it isolated 57 strains of the pathogen from the damaged herbal material collected from the fields in eight agro-ecological rice-growing regions of the Krasnodar Region (Russian Federation): Krasnoarmeysky, Kalininsky, Krymsky, Abinsky, Temryuksky, Seversky, Slavyansky districts, Krasnodar, Rostov Region (Russian Federation)—Proletarsky district and the Republic of Adygea (Russian Federation). A multiplex PCR technique was applied on the basis of fragment analysis to identify the virulent fungal isolates. 33 fungal genotypes with unique genetic profiles were identified among the studied races of Pyricularia oryzae Cav. Their DNA profiles were created. The studied isolates of the pathogen of rice blast were classified using morphological and microbiology cultural features. Based on the phytopathological test using differentiation rice varieties, the quantitative and qualitative composition of (a)virulence genes in fungal races was established. Effective genes for pathogen resistance, which are recommended for breeding programs for the development of rice varieties resistant to rice blast, were identified in the south of Russia. A high yielding redgram culture CRG 10-01 was a cross derivative of APK 1x LRG 41 and matures in 170 to 180 days. The culture recorded an average grain yield of 1600 kg/ha under rainfed condition and 1800 kg/ha under irrigated condition with a yield increase of 19 percent over CO 6 and 22 percent over VBN 2. Drought tolerant CO 8 variety shown resistance to Sterility Mosaic Disease, root rot and moderately resistant to pod borer complex. This culture CRG 10-01 is released as CO 8 redgram for general cultivation in Tamil Nadu during 2017. The variety notification proposal has been submitted to the PPV&FRA with DNA finger printing profile for approval.

Ependymoma is an uncommon neuroepithelial tumor that may arise anywhere within the neuroaxis, both in children and in adults. It has been classically graded upon histopathological features, yet with limited clinical utility. Recently, DNA methylation profiling has provided a novel classification of ependymoma in nine molecular subgroups. This stratification method harbors prognostic value with supratentorial RELA-fusion and posterior fossa group A tumors showing a significantly shorter survival compared to the rest. Currently, the treatment of choice involves maximal safe resection and, in cases of residual disease, adjuvant conformal radiotherapy. Second-look surgery is also a feasible and recommended option for incompletely resected tumors. The role of chemotherapy is not yet established and can be considered in infants and children with relapsing disease or prior to re-intervention. Abstract We compare two open-source programs for the evaluation of evidential weight arising from complex DNA profiles recovered in a crime investigation. Here, "complex" means one or more of: low-template, degraded and mixed-source. Although software for complex DNA profile analysis has made great strides in recent years, the ability of courts to effectively scrutinise and challenge the reliability of the resulting evidence remains problematic. One key step is to compare different software on the same evidence, but there are currently few published comparisons in part because of the problems posed by restricted access to commercial software. We present here an extensive comparison between two open-source software, LikeLTD and EuroForMix. We find that despite different modelling assumptions the two programs generate similar results. Deoxyribose Nucleic Acid (DNA) are the basic elements that make up a whole section of an individual. The basic elements store information that is unique to each individual and will be passed down the generations. DNA also helps in identifying the father in paternity testing, locating missing person investigations, identifying victims in mass disasters. Identification of the victims has a problem if the comparison the father and mother no instance the victim’s parents have died or are very far away from where the victim. Therefore, it is necessary to try to identify Short Tandem Repeat (STR) Inference of live family such as sibling, grandfather/grandmother, uncle/aunt, cousin and nephew. In this paper, we performed a method to measure the similarity of human DNA profiles using fuzzy similarity. In this fuzzy system, DNA profile data is used as an input that stores human identity along with its DNA profile. The data entered is the result of polymerase chain reaction (PCR) identification which is an electropherogram consisting of 16 loci with two alleles for each locus. Population allele frequency is an indicator of population genetic diversity and plays significant roles in DNA profiling interpretation in human identification, kinship, and forensic testing. The Lebanese population is a mosaic of 18 religious communities with high rates of consanguinity and endogamy. Allele frequencies for 23 STR loci were estimated and analyzed in the seven major Lebanese religious subcommunities (Muslims Shiaa, Sunni, and Druze, and Christians Orthodox, Maronite, and Catholics), to assess possible significant differences among their STR allele frequencies. Population representative short tandem repeat (STR) allele frequencies are crucial for proper probabilistic interpretation of DNA forensic evidence. STR allele frequencies also provide information about the genetic diversity of a given population. The Lebanese population is characterized by the presence of more than 18 religious communities that have high recorded rates of endogamous and consanguineous marriages, where the choice of marriages is mainly influenced by their respective geographical distributions and religious affiliation. These factors could have led to an increase in complex DNA cases, whereby high numbers of STR markers are needed to help clear the case. Allele frequencies for 23 STR loci in the Lebanese population were recently estimated and published. The present study aims at estimating the STR allele frequencies for five supplementary STRs, namely, the LPL, F13B, FESFPS, F13A01, and Penta C STR markers, which are mainly used to resolve complex kinship investigations. A DNA database representative of the Lebanese population that comprises 505 Lebanese unrelated individuals was used to amplify the five STR systems, using the CS7 kit released by Promega. The allele frequencies for these STR systems were estimated, and a significant departure from the Hardy–Weinberg equilibrium was detected after Bonferroni correction for multiple tests at locus LPL, F13A01, and Penta C. The newly added systems appeared to have variation in their discriminative power among Lebanese individuals. Penta C showed the highest power of discrimination, whereas FESFPS showed the lowest power of discrimination. Employed for the first time in 1986, DNA profiling is nowadays established as one of the most widely used forensic techniques worldwide. However, until today, no efficient sampling technique existed to collect DNA from human skin cells from a large area, not to say from the floor of an entire room. This has been extremely unfortunate, as there is enormous forensic potential in these DNA traces from the ground to provide clues as to who has been present at a particular location, i.e. at the crime scene. By desquamation, humans loose several millions of skin cells per day, everywhere they stand, sit or walk; and they can do little about it. We developed a fast and simple method by which we can make use of all those lost skin cells. We use a vacuum cleaner equipped with a specialized filter cartridge to sample the ground. Fragmentation of the filter membrane and subsequent parallel processing of the filter fragments, using a modified Chelex® 100 extraction protocol, significantly reduce the complexity of the dust mixture. In this way, a large number of interpretable major contributor DNA profiles can be generated from individuals who have been present on the sampled surface. Overall, at least 38 % of the generated DNA profiles from all sampled test areas fulfilled the criteria for submission of single major contributor profiles to the Swiss DNA database. As demonstrated through a mock crime scene scenario simulating an indoor stabbing event, the perpetrator's DNA could be found on the floor even after a very short stay in the room of less than one minute. Furthermore, already the first application of the method at a real crime scene led to relevant case information for the police. Touch DNA, also known as trace DNA, represents one of the most frequent biological traces that are often found at crime scenes. It involves the transfer of biological material from a donor to an object or to another person during physical contact. Allows to be collected as evidence when there is a lack of visible DNA (blood, hair, semen, or saliva) or fingerprints are too stained or incomplete for fingerprint analysis. The collection and interpretation of the results of touch DNA contain very valuable information and represent a very important forensic tool that can lead to the identification of a criminal and help solve the crime. In the presented study, DNA analysis was done on samples collected at a crime scene investigation in Canton Sarajevo. Standard methods for DNA isolation, amplification, and DNA profile generation were used. From a total of 83 samples collected from different substrates at crime scenes, 6 samples with full DNA profiles and 8 with partial DNA profiles were generated. Additionally, 12 mixed DNA profiles were obtained. For 46 samples, no DNA profiles were generated. This research has shown that the accuracy of DNA profiling is significantly influenced by the proper collection of touch DNA from certain substrates. Summary: In dogs and cats, unusual coat colour phenotypes may result from various phenomena, including chimerism. In the domestic cat, the tortoiseshell coat colour that combines red and non‐red hairs is the most obvious way to identify chimeras in males. Several cases of tortoiseshell males have been reported, some of which were diagnosed as chimeras without any molecular confirmation. Here, we report the case of a female feline chimera identified thanks to its coat colour and confirmed through DNA profiling and a coat colour test. DNA typing has been long proposed as a component of food traceability especially for the ability to provide genetic information on processed and/or mixed commercial products. However, the use of DNA testing to deliver legal evidence has been very limited in the agro-food chain, also because in this field, a genetic concordance is rarely sustained with indications on the interpretation and statistics of a DNA profile. Zucchini is the most important and diverse horticultural group belonging to Cucurbita pepo , a plant species that yields a large variety of edible fruits such as squashes and pumpkins. Challenges in genetic discrimination of zucchini material exiting the agro-food chain typify the problems in DNA typing of horticultural food products, because of limited allelic diversity, presence of common alleles, highly processed food and genetic similarity with other edible species. The aims of this work were to identify zucchini varieties in commercial products by a means of Simple Sequence Repeat (SSR) analysis and to provide indications for the statistical interpretation of a match. We assessed the efficacy of SSRs analyzing some of the most diffused hybrids and open pollinated zucchini varieties. SSRs were also screened for transferability in DNA isolated from edible organs of different plant species that are frequently present in food products containing zucchini. Moreover, we demonstrate the possibility to identify the variety in food products of different origins. The statistical evaluation on the rarity of DNA profiles in zucchini indicated that the genetic match presented in this work cannot be attributed to chance alone. Touch DNA recovery from firearms can be central to many criminal investigations, yet the generation of DNA profiles from these items remains poor. Currently in Australia, published casework data highlights extremely poor DNA success from samples recovered from firearms. Only between 5% and 25% of samples result in useful DNA data and therefore increasing the success of DNA recovered from firearms is highly important but has not yet been explored in-depth. This study focused on increasing the recovery of DNA from ten firearm components that were held for 15 s. Multiple recovery methods were used, and the resulting genetic data compared. DNA evidence may be deliberately removed from firearms after discharge to hamper forensic investigations, therefore this study examined the effect of wiping down the components or handling them with gloves. A standard double swab and rinse swab recovery method resulted in an average of 73% cellular recovery. A cumulative swab process had the highest average recovery at 86%, although it was found that increasing the DNA yield led to an increase in mixture complexity. Wiping over the components was observed to remove on average 69% of cellular material, compared with 33% when handed with gloves. However, the size and texture of the components affected the efficiency of cellular material removal. The results from this study allow for prioritisation of areas to sample on firearms, as well as suggesting techniques that can be applied for the optimum process of cellular recovery and subsequent generation of STR DNA data. • Ten firearm components were sampled for touch DNA. • Cellular material recovery was highest using a cumulative swab at 86%. • The trigger component rarely generated an informative STR profile. • Even though one volunteer handled the firearm components, single source STR profiles were rarely obtained. • Deliberate removal of cellular material prior to forensic testing reduced the mixture complexity, but DNA data were still obtained. This lesson presents an interrupted case study based on the true story of the 2002 murder of Christa Worthington in Massachusetts. The case was developed for use in an undergraduate non-majors life science course, but would also be appropriate for a high school biology course or a forensic science course. During this lesson, students examine a crime scene diagram and discuss evidence collection. Students then conduct a hands-on activity extracting DNA from wheat germ to simulate how DNA would be isolated from crime scene samples. Lastly the students will analyze simulated DNA profiles produced using STRs, polymerase chain reaction, and gel electrophoresis to help match a crime scene sample to one of five suspects. The use of biometrics to enhance identification has been explored and utilized to various extents. DNA is the most reliable and stable biometric that remains unchanged throughout an individual's lifetime. Advancements in DNA analysis, in terms of reduced cost and faster processing times, make the use of DNA as a biometric more feasible over time. Since DNA data is of a sensitive nature, privacy and ethical concerns would have to be carefully considered before large-scale adoption for use in identity documents. Birth certificates are a fundamental document used by a person for identification. However, it does not contain any means of authentication beyond possession of the document. This paper examines the security measures that would be required if birth certificates were embedded with DNA profile information. The U.S. FBI CODIS approach is referred to, being an established standard for human DNA profiling and identification. H d true testing is a way of assessing the performance of a model, or DNA profile interpretation system. These tests involve simulating DNA profiles of non-donors to a DNA mixture and calculating a likelihood ratio ( LR ) with one proposition postulating their contribution and the alternative postulating their non-contribution. Following Turing it is possible to predict that “ The average LR for the H d true tests should be one” [1] . This suggests a way of validating softwares. During discussions on the ISFG software validation guidelines [2] it was argued by some that this prediction had not been sufficiently examined experimentally to serve as a criterion for validation. More recently a high profile report [3] has emphasised large scale empirical examination. A limitation with H d true tests, when non-donor profiles are generated at random (or in accordance with expectation from allele frequencies), is that the number of tests required depends on the discrimination power of the evidence profile. If the H d true tests are to fully explore the genotype space that yields non-zero LR s then the number of simulations required could be in the 10 s of orders of magnitude (well outside practical computing limits). We describe here the use of importance sampling, which allows the simulation of rare events to occur more commonly than they would at random, and then adjusting for this bias at the end of the simulation in order to recover all diagnostic values of interest. Importance sampling, whilst having been employed by others for H d true tests, is largely unknown in forensic genetics. We take time in this paper to explain how importance sampling works, the advantages of using it and its application to H d true tests. We conclude by showing that employing an importance sampling scheme brings H d true testing ability to all profiles, regardless of discrimination power. [ABSTRACT FROM AUTHOR]

Entomopathogenic baculoviruses have long been recognized as vital environmentally safe alternatives to chemical pesticides. A study for investigating the virulence and susceptibility of Spodoptera littoralis larvae to S. littoralis nucleopolyhedrovirus (SpliNPV) isolate was carried out. Bioassay experiments on the 1st and 2nd larval instars using droplet feeding method revealed that polyhedral for 50% or 90% mortality of the 1nd larval instar of S. littoralis was significantly lower compared with the 2st instar. While, the 2st instar was killed faster than the 2nd one especially with the higher concentration. The susceptibility of the infected 2nd instar when feeding on different hosts showed that larvae were more susceptible when feeding on eggplant leaves followed by lettuce leaves, artificial diet and tomato leaves as shown by larval weight and reduction in feeding. The reduction in both larval weight and feeding were significantly higher with 1x107 PIB/ml than 1x104 PIB/ml in the majority of cases. S. littoralis larvae showed more than 2 times of tolerant to the 3rd passage of SpliNPV on the level of LC50 and more than 18 times tolerant on the level of LC90. The tolerant levels of S. littoralis larvae for the 3rd passage of virus was associated with more than 20% increase in time taken for kill with no changes in DNA patterns regarding subsequent passages in S. littoralis larvae using a restriction enzyme EcoRI. In this article we present the results of a study to explore if cross-border DNA matches between the Netherlands and Belgium are relatively more likely to occur in areas near the Dutch-Belgian border than in areas at some distance from this border. For this study we used the results of the transnational DNA profile exchange and comparison between the Belgian and Dutch DNA databases, which first took place in 2014. It appears that the Dutch regions adjacent to Belgium, i.e., Zeeland-West-Brabant, Oost-Brabant and Limburg, have relatively more DNA matches with Belgium than the other Dutch regions. In other words, a DNA profile obtained from a crime scene close to the Dutch border with Belgium is more likely to match with the profile of a person whose DNA profile is stored in the Belgian database than a DNA profile that originates from a crime scene further afield. Our data suggest that crimes committed by repeat offenders show a spatial pattern despite the presence of a national border, with crime scenes clustering in relatively close proximity to each other. Many DNA profiles recovered from crime scene samples are of a quality that does not allow them to be searched against, nor entered into, databases. We propose a method for the comparison of profiles arising from two DNA samples, one or both of which can have multiple donors and be affected by low DNA template or degraded DNA. We compute likelihood ratios to evaluate the hypothesis that the two samples have a common DNA donor, and hypotheses specifying the relatedness of two donors. Our method uses a probability distribution for the genotype of the donor of interest in each sample. This distribution can be obtained from a statistical model, or we can exploit the ability of trained human experts to assess genotype probabilities, thus extracting much information that would be discarded by standard interpretation rules. Our method is compatible with established methods in simple settings, but is more widely applicable and can make better use of information than many current methods for the analysis of mixed-source, low-template DNA profiles. It can accommodate uncertainty arising from relatedness instead of or in addition to uncertainty arising from noisy genotyping. Disaster victim identification (DVI) often occurs in remote locations with extremes of temperatures and humidities. Access to mortuary facilities and refrigeration are not always available. An effective and robust DNA sampling and preservation procedure would increase the probability of successful DNA profiling and allow faster repatriation of bodies and body parts. If the act of tissue preservation also released DNA into solution, ready for polymerase chain reaction (PCR), the DVI process could be further streamlined. In this study, we explored the possibility of obtaining DNA profiles without DNA extraction, by adding aliquots of preservative solutions surrounding fresh human muscle and decomposing human muscle and skin tissue samples directly to PCR. The preservatives consisted of two custom preparations and two proprietary solutions. The custom preparations were a salt-saturated solution of dimethyl sulfoxide (DMSO) with ethylenediaminetetraacetic (EDTA) and TENT buffer (Tris, EDTA, NaCl, Tween 20). The proprietary preservatives were DNAgard (Biomatrica) and Tissue Stabilising Kit (DNA Genotek). We obtained full PowerPlex 21 (Promega) and GlobalFiler (Life Technologies) DNA profiles from fresh and decomposed tissue preserved at 35 °C for up to 28 days for all four preservatives. The preservative aliquots removed from the fresh muscle tissue samples had been stored at −80 °C for 4 years, indicating that long-term archival does not diminish the probability of successful DNA typing.
