The most suitable drug delivery route is oral delivery due to its easily administration, patient adherence/ patient capacitance etc. Several approaches have been made for maximizing the G.R.T such as high-density system, floating system, swelling & expanding system and mucoadhesive & bio adhesive system etc. the main motive of reviewing the article is to focus on the mechanism of HBS system, classification with new system such as raft forming system and hollow microsphere, its application, marketed preparation and evaluation study. The procedure of gastric emptying is a complex and may leads to uncertainty for in vivo performance of the DDS. To prevent this type of complex formation and uncertainty, hard work has been done to expand the retention time of DDS for half of the day. The FDDS are beneficial in such process. Oral drug delivery system is the most preferred route of administration for drug delivery. In the development of the drug delivery system many components play important role. Polymers are amongst those components which have evolved with the drug delivery system. Polymers are the macromolecule compound containing many monomer units joined to each other by bonds. The floating drug delivery systems (FDDS) become an additional advantage for drugs that are absorbed primarily in the upper segments of gastrointestinal (GI) tract, i.e., the stomach, duodenum and jejunum. The purpose of writing this review on floating drug delivery systems (FDDS) was to focus on the types of floating drug delivery systems, principal and mechanism of floatation to achieve gastric retention and polymers used in floating Drug delivery systems. Polymers used in the drug delivery system are of two types Natural and Synthetic based on their origin. Both types of the polymers have some advantages and disadvantages. This particular article gives information about the different types of natural and synthetic polymer used in the drug delivery system. Natural polymers like guar gum, chitosan, xanthan gum, Gellan gum and sodium alginate are mentioned in the article. Synthetic polymers mentioned are HPMC, Eudragit, and Ethylcellulose. In this study, a multiparticulate matrix system was produced, containing two different active pharmaceutical ingredients (APIs): enalapril-maleate and hydrochlorothiazide. The critical control points of the process were investigated by means of factorial design. Beside the generally used microcrystalline cellulose, ethylcellulose was used as matrix former to achieve modified drug release ensured by diffusion. The matrix pellets were made by extrusion-spheronization using a twin-screw extruder. Some pellet properties (aspect ratio, 10% interval fraction, hardness, deformation process) were determined. The aim of our study was to investigate how the two different APIs with different solubility and particle size influence the process. The amount of the granulation liquid plays a key role in the pellet shaping. A higher liquid feed rate is preferred in the pelletization process. [ABSTRACT FROM PUBLISHER]
