G protein-coupled receptors (GPCRs) are an ancient family of signal transducers that are both abundant and consequential in metazoan endocrinology. The evolutionary history and function of the GPCRs of the decapod superfamilies of gonadotropin-releasing hormone (GnRH) are yet to be fully elucidated. As part of which, the use of traditional phylogenetics and the recycling of a diminutive set of mis-annotated databases has proven insufficient. To address this, we have collated and revised eight existing and three novel GPCR repertoires for GnRH of decapod species. We developed a novel bioinformatic workflow that included clustering analysis to capture likely GnRH receptor-like proteins, followed by phylogenetic analysis of the seven transmembrane-spanning domains. A high degree of conservation of the sequences and topology of the domains and motifs allowed the identification of species-specific variation (up to ~70%, especially in the extracellular loops) that is thought to be influential to ligand-binding and function. Given the key functional role of the DRY motif across GPCRs, the classification of receptors based on the variation of this motif can be universally applied to resolve cryptic GPCR families, as was achieved in this work. Our results contribute to the resolution of the evolutionary history of invertebrate GnRH receptors and inform the design of bioassays in their deorphanization and functional annotation. Cytochrome P450s (CYP450s) are a versatile superfamily of enzymes known to undergo rapid evolution. They have important roles across growth and development pathways in crustaceans, although it is difficult to characterise orthologs between species due to their sequence diversity. Conserved CYP450s enzymes in crustaceans are those associated with ecdysteroidogenesis: synthesising and breaking down the active moult hormone, 20-hydroxyecdysone. The complex life cycle of the ornate spiny lobster, Panulirus ornatus, relies on moulting in order to grow and develop. Many of these diverse life stages have been analysed to establish a comprehensive transcriptomic database for this species. The transcripts putatively encoding for CYP450s were mapped using transcriptomic analysis and identified across growth and development stages. With the aid of phylogeny, 28 transcripts of 42 putative P. ornatus CYP450s were annotated, including the well conserved Halloween genes, which are involved in ecdysteroidogenesis. Expression patterns across the life stages determined that only a subset of the CYP450s can be detected in each life stage or tissue. Four Shed transcripts show overlapping expression between metamorphosis and adult tissues, suggesting pleotropic functions of the multiple Shed orthologs within P. ornatus. [ABSTRACT FROM AUTHOR]

The "messy" feeding behaviour of spiny lobsters remains an obstacle for formulated feed development. This study examined the relationship between feeding efficiency and feed waste by juvenile spiny lobster, Sagmariasus verreauxi, fed different formulated pellet diameters or lengths across two separate experiments. Feed texture (hard and dry pellet, HDP; soft and moist pellet, SMP) was also examined. Juvenile lobsters were fed experimental feeds at 0.5% BW daily over a 6 h duration. The resulting feed waste was categorised as either feeding-related waste (FRW) or non-feeding-related waste (NFRW). For all feed types, the FRW increased with increasing pellet diameter and pellet length. The increase in FRW corresponded with a decrease in NFRW, particularly for HDP, resulting in no difference in total feed waste in any treatment investigated. Thus, even with improved feeding efficiency with small feed dimensions, feed intake was not improved. Feed leaching rate decreased with increasing pellet size, suggesting a more rapid decline in feed attractiveness for smaller pellets. This finding indicates that currently a counteractive interaction exists between pellet size and feed attractiveness and suggests improving attractiveness would further enhance feeding. RNA interference (RNAi) has been widely utilised in many invertebrate models since its discovery, and in a majority of instances presents as a highly efficient and potent gene silencing mechanism. This is emphasized in crustaceans with almost all taxa having the capacity to trigger effective silencing, with a notable exception in the spiny lobsters where repeated attempts at dsRNA induced RNAi have demonstrated extremely ineffective gene knockdown. A comparison of the core RNAi machinery in transcriptomic data from spiny lobsters (Panulirus ornatus) and the closely related slipper lobsters (Thenus australiensis, where silencing is highly effective) revealed that both lobsters possess all proteins involved in the small interfering and microRNA pathways, and that there was little difference at both the sequence and domain architecture level. Comparing the expression of these genes however demonstrated that T. australiensis had significantly higher expression in the transcripts encoding proteins which directly interact with dsRNA when compared to P. ornatus, validated via qPCR. These results suggest that low expression of the core RNAi genes may be hindering the silencing response in P. ornatus, and suggest that it may be critical to enhance the expression of these genes to induce efficient silencing in spiny lobsters. Simple Summary: Sensory stimuli including olfactory, visual, acoustic and tactile input, are important aspects of feed exposure and optimisation of feeding schedules. These aspects were investigated in our study, with a focus on the effect of feed frequency (one, two, four, eight, sixteen and thirty-two feed supplies per day) on survival, growth and behaviour of juvenile P. ornatus. Increasing daily feed frequency from one to sixteen improved feed intake and growth. Spiny lobsters have a range of complex chemical communication pathways that contribute to feeding behaviour. Feed intake is modulated by feed availability and feed characteristics, such as attractiveness and palatability, with behavioural factors, such as social competition and circadian rhythm, providing an extra layer of complexity. In this study, we investigated the effect of feed frequency on survival and growth of early-stage (instar 2–6) juvenile Palunirus ornatus. In addition, we investigated the interactive effect of feed frequency and circadian rhythm on lobster feed response. Lobsters were fed a set ration at a frequency of either one, two, four, eight, sixteen or thirty-two times per day over 49 days. The effect of feed frequency on growth and survival was determined. Circadian feeding activity under these feeding treatments was assessed by time-lapse photography. Increased feed frequency from one to sixteen feeds daily improved growth by increasing apparent feed intake (AFI) and feed attraction, as confirmed by the increased presence of lobsters in the feeding area. The rapid leaching of feed attractant, particularly free amino acid, suggests a beneficial effect of multiple feeding frequencies on feed intake and growth. However, more than sixteen feeds per day resulted in decreased feed intake and a subsequent reduction in growth. The decrease in feed intake is thought to be associated with saturation of the culture environment with attractants, resulting in a reduced behavioural response to feed supplies. This may indicate the need for depletion of attractants to retrigger a feeding response. As lobsters were grown communally, faster growth at sixteen rations per day was also coupled with increased cannibalism, likely driven by increased vulnerability with the occurrence of more frequent ecdysis events. The respiratory quotient (RQ) has been used extensively as an index to evaluate metabolic energy expenditure in terrestrial animals including humans. In contrast, RQ use in understanding physiology and nutrition of aquatic ectotherms has been restricted due to technical challenges in measuring total CO2 in water. With technical advances in measuring total CO2 in water, RQ in aquatic ectotherms can be accurately determined and is potentially available as a valuable method. Here, we provide a comprehensive review of studies on RQ and metabolic energy substrate use in aquatic ectotherms. Metabolic energy substrate use is evaluated by a reliable stoichiometric bioenergetic approach, based on measuring RQ and nitrogen quotient (NQ) simultaneously. Stoichiometry provides a non‐destructive and unequivocal way to quantify the instantaneous oxidation of each major energy substrate (protein, lipid or carbohydrate). This review aims to refine knowledge about bioenergetics of aquatic ectotherms under different conditions including nutritional aspects of sustainable aquaculture. Notably, stoichiometry provides a promising approach to optimize feeds and feeding regimes to realize sustainable aquaculture under differing conditions and with differing feed ingredients. It also provides an approach to consider climate change impacts and physiological adaptation mechanisms for survival and development in farmed environments and natural ecosystems. Stoichiometric bioenergetics knowledge in aquatic ectotherms has relevance to commercial impacts in the face of overfishing and food security, and ecological significance in the face of environmental change scenarios. Climate‐driven species redistribution is pervasive and accelerating, yet the complex mechanisms at play remain poorly understood. The implications of large‐scale species redistribution for natural systems and human societies have resulted in a large number of studies exploring the effects on individual species and ecological communities worldwide. Whilst many studies have investigated discrete components of species redistribution, the integration required for a more complete mechanistic understanding is lacking. In this paper, we provide a framework for synthesising approaches to more robustly understand and predict marine species redistributions. We conceptualise the stages and processes involved in climate‐driven species redistribution at increasing levels of biological organisation, and synthesize the laboratory, field and modelling approaches used to study redistribution related processes at individual, population and community levels. We then summarise links between scales of biological organisation and methodological approaches in a hierarchical framework that represents an integrated mechanistic assessment of climate‐driven species redistributions. In a rapidly expanding field of research, this framework provides direction for: 1) guiding future research, 2) highlighting key knowledge gaps, 3) fostering data exchange and collaboration between disciplines and 4) improving shared capacity to predict and therefore, inform the proactive management of climate impacts on natural systems. Predation risk can strongly shape prey ecological traits, with specific anti-predator responses displayed to reduce encounters with predators. Key environmental drivers, such as temperature, can profoundly modulate prey energetic costs in ectotherms, although we currently lack knowledge of how both temperature and predation risk can challenge prey physiology and ecology. Such uncertainties in predator–prey interactions are particularly relevant for marine regions experiencing rapid environmental changes due to climate change. Using the octopus (Octopus maorum)–spiny lobster (Jasus edwardsii) interaction as a predator–prey model, we examined different metabolic traits of sub adult spiny lobsters under predation risk in combination with two thermal scenarios: 'current' (20°C) and 'warming' (23°C), based on projections of sea-surface temperature under climate change. We examined lobster standard metabolic rates to define the energetic requirements at specific temperatures. Routine metabolic rates (RMRs) within a respirometer were used as a proxy of lobster activity during night and day time, and active metabolic rates, aerobic scope and excess post-exercise oxygen consumption were used to assess the energetic costs associated with escape responses (i.e. tail-flipping) in both thermal scenarios. Lobster standard metabolic rate increased at 23°C, suggesting an elevated energetic requirement (39%) compared to 20°C. Unthreatened lobsters displayed a strong circadian pattern in RMR with higher rates during the night compared with the day, which were strongly magnified at 23°C. Once exposed to predation risk, lobsters at 20°C quickly reduced their RMR by ~29%, suggesting an immobility or 'freezing' response to avoid predators. Conversely, lobsters acclimated to 23°C did not display such an anti-predator response. These findings suggest that warmer temperatures may induce a change to the typical immobility predation risk response of lobsters. It is hypothesized that heightened energetic maintenance requirements at higher temperatures may act to override the normal predator-risk responses under climate-change scenarios. Extensions of species' geographical distributions, or range extensions, are among the primary ecological responses to climate change in the oceans. Considerable variation across the rates at which species' ranges change with temperature hinders our ability to forecast range extensions based on climate data alone. To better manage the consequences of ongoing and future range extensions for global marine biodiversity, more information is needed on the biological mechanisms that link temperatures to range limits. This is especially important at understudied, low relative temperatures relevant to poleward range extensions, which appear to outpace warm range edge contractions four times over. Here, we capitalized on the ongoing range extension of a teleost predator, the Australasian snapper Chrysophrys auratus , to examine multiple measures of ecologically relevant physiological performance at the population's poleward range extension front. Swim tunnel respirometry was used to determine how mid-range and poleward range edge winter acclimation temperatures affect metabolic rate, aerobic scope, swimming performance and efficiency and recovery from exercise. Relative to 'optimal' mid-range temperature acclimation, subsequent range edge minimum temperature acclimation resulted in absolute aerobic scope decreasing while factorial aerobic scope increased; efficiency of swimming increased while maximum sustainable swimming speed decreased; and recovery from exercise required a longer duration despite lower oxygen payback. Cold-acclimated swimming faster than 0.9 body lengths sec−1 required a greater proportion of aerobic scope despite decreased cost of transport. Reduced aerobic scope did not account for declines in recovery and lower maximum sustainable swimming speed. These results suggest that while performances decline at range edge minimum temperatures, cold-acclimated snapper are optimized for energy savings and range edge limitation may arise from suboptimal temperature exposure throughout the year rather than acute minimum temperature exposure. We propose incorporating performance data with in situ behaviour and environmental data in bioenergetic models to better understand how thermal tolerance determines range limits. Lobsters are fished world-wide due to their status as a high value, luxury seafood. A large proportion of the product is sold via live export, with lobsters subject to a range of stressors during holding post-capture. Improving the current understanding of the immune response to these stressors assists in improving efficiency and reducing loss in the chain between capture and consumption. In this study, the immune status of four treatment groups of Southern Rock Lobster (Jasus edwardsii) were studied: controls recently landed from a fishing boat, lobsters displaying advanced shell necrosis, lobsters in an unexplained moribund state and lobsters held in a processing facility for 10 weeks in standard conditions (i.e. high density, fasted). A total of 15 immune parameters and 19 haemolymph biochemical parameters were assayed. Phenoloxidase activity was only sporadically observed in haemocyte lysate and was consistently observed at a low level in the plasma with no difference between treatments for either. Haemocyte lysate prophenoloxidase activity was detected in most individuals, with no differences found between treatments. Prophenoloxidase in the plasma showed the highest level of activity, with the shell necrosis treatment demonstrating an elevated activity level relative to the other three treatments. Cell viability was not affected in any treatment. Lobsters with shell necrosis had a reduced capacity for phagocytosis, a significantly higher total haemocyte count, fewer hyalinocytes and more granulocytes and semigranulocytes. Fasted lobsters showed an opposite shift, with significantly more hyalinocytes compared to the other treatments and very few granulocytes and semigranulocytes. The balance of a range electrolytes, minerals metabolites and enzymes were affected in shell necrosis and fasted treatments, raising them as potential markers for immunocompromised lobsters. Multivariate analysis of all assayed parameters showed that all individuals in the necrosis treatment showed a similar, distinct immune response and that the fasted treatment, along with one control and one moribund individual, showed a separate intermediate response. The remainder of the control and moribund lobsters demonstrated a distinct "non-response" in comparison. Metamorphosis involves a complex network of genes that orchestrate a perfectly timed reorganization of one body form to another. The molecular pathways that start to unravel for an increasing number of species show that there exists great diversity among different species, as would be expected by their wide range of life histories and transformation strategies. The metamorphosis process could account for a considerably high percentile of transcribed sequences over a short period of time, with the genome encoding for different life forms. Such important changes in expression patterns for a high number of genes pose a challenge for accurately assign each gene to a function. Several key conserved factors are consistently expressed and can be placed at the center of metamorphosis, including the mechanisms involving the molt hormone, 20 Hydroxy-Ecdysone, and the juvenile hormone. Yet, many additional factors are not characterized, remain unannotated, or do not have a function assigned. This manuscript provides several examples of how an integrated omics approach can develop further insights into crustacean metamorphosis and eventually lead to discovery of key factors for metamorphosis. There is a critical knowledge gap regarding the impacts of seismic air gun signals on the physiology of adult crustaceans. We conducted four controlled field experiments to examine the impact of seismic acoustic signals on spiny lobster, Jasus edwardsii . Seismic air gun exposure suppressed total haemocyte count (THC) for up to 120 days post-exposure, suggesting a chronic negative impact of immune competency. THC levels after 365 days post-exposure, were elevated two fold, potentially indicating an immune response to infection. Haemolymph refractive index was reduced after 120 days post exposure in one experiment, suggesting a chronic impairment of nutritional condition. There was no effect of air gun exposure on 24 haemolymph biochemical parameters, hepatopancreas index or survival. Collectively these results indicate that the biochemical haematological homeostasis of J. edwardsii is reasonably resilient to seismic acoustic signals, however, air gun exposure may negatively influence the lobster's nutritional condition and immunological capacity. Sex determination pathways are extensively diverse across species, with the master sex-determinants being the most variable element. Despite this, there is a family of DM-domain transcription factors ( Dmrts ), which hold a highly conserved function in sexual development. This work is the first to describe a heterogametic sex-linked Dmrt in an invertebrate species, the Eastern spiny lobster, Sagmariasus verreauxi . We have termed the Y-linked, truncated paralogue of the autosomal iDmrt1, Sv-iDMY. Considering the master sex-determining function of both DMY in medaka and DM-W in frog, we hypothesised a similar function of Sv-iDMY. By conducting temporal expression analyses during embryogenesis we have identified a putative male sex-determining period during which iDMY > iDmrt1 . Employing a GAL4-transactivation assay we then demonstrate the dominant negative suppression of iDMY over its autosomal iDmrt1 paralogue, suggesting the mechanism with which iDMY determines sex. Comparative analyses of Sv-iDMY, DM-W and medaka DMY, highlight the C′-mediated features of oligomerisation and transactivation as central to the mechanism that each exerts. Indeed, these features may underpin the plasticity facilitating the convergent emergence of these three sporadic sex-linked master-Dmrts. [ABSTRACT FROM AUTHOR]
