Purpose Traditional organic formulations are widely used as a plant growth promoters; however, the knowledge on the microbial aspect of traditional organic formulations is still limited. The aim of this study was to illustrate the cultivable bacterial diversity of various traditional organic formulations and their potential for early plant growth promotion. Methods Five different traditional organic formulations such as 100 % panchagavya, 33 % panchagavya, plant extract with native microorganisms, commercial organic fertilizer extract with two percent leaf soil extract and commercial organic fertilizer extract with 2 % yogurt were prepared and used in this study. The liquid fraction of these traditional organic formulations were used to analyze the beneficial effect on plant growth by seed treatment and foliar applications. Results Bacterial 16SrDNA analysis revealed that the isolates fell into forty-three different genera, which can be grouped into seven different classes, such as Alphaproteobacteria, Actinobacteria, Bacilli (or Firmibacteria), Betaproteobacteria, Cytophagia, Flavobacteriia and Gammaproteobacteria. Higher bacterial diversity was observed in cow dung followed by 33 and 100 % panchagavya. Radish and Chinese cabbage seed germination and growth were significantly improved by traditional organic formulations compared to control. Conclusion The results of this study showed that the bacterial diversity changes depend on the type and concentration of ingredients used in traditional organic formulations. Substantial increase in plant growth by the traditional organic formulations indicates the suitability of using these organic preparations in eco-friendly agriculture.

Plant-associated microbiota plays an important role in plant disease resistance. Bacterial wilt resistance of tomato is a function of the quantitative trait of tomato plants; however, the mechanism underlying quantitative resistance is unexplored. In this study, we hypothesized that rhizosphere microbiota affects the resistance of tomato plants against soil-borne bacterial wilt caused by Ralstonia solanacearum. This hypothesis was tested using a tomato cultivar grown in a defined soil with various microbiota transplants. The bacterial wilt-resistant Hawaii 7996 tomato cultivar exhibited marked suppression and induction of disease severity after treatment with upland soil-derived and forest soil-derived microbiotas, respectively, whereas the transplants did not affect the disease severity in the susceptible tomato cultivar Moneymaker. The differential resistance of Hawaii 7996 to bacterial wilt was abolished by diluted or heat-killed microbiota transplantation. Microbial community analysis revealed the transplant-specific distinct community structure in the tomato rhizosphere and the significant enrichment of specific microbial operational taxonomic units (OTUs) in the rhizosphere of the upland soil microbiota-treated Hawaii 7996. These results suggest that the specific transplanted microbiota alters the bacterial wilt resistance in the resistant cultivar potentially through a priority effect.

Plant growth-promoting bacteria improve plant growth under abiotic stress conditions. However, their effects on microbial succession in the rhizosphere are poorly understood. In this study, the inoculants of Bacillus mesonae strain H20-5 were administered to tomato plants grown in soils with different salinity levels (EC of 2, 4, and 6 dS/m). The bacterial communities in the bulk and rhizosphere soils were examined 14 days after H20-5 treatment using Illumina MiSeq sequencing of the bacterial 16S rRNA gene. Although the abundance of H20-5 rapidly decreased in the bulk and rhizosphere soils, a shift in the bacterial community was observed following H20-5 treatment. The variation in bacterial communities due to H20-5 treatment was higher in the rhizosphere than in the bulk soils. Additionally, the bacterial species richness and diversity were greater in the H20-5 treated rhizosphere than in the control. The composition and structure of the bacterial communities varied with soil salinity levels, and those in the H20-5 treated rhizosphere soil were clustered. The members of Actinobacteria genera, including Kineosporia, Virgisporangium, Actinoplanes, Gaiella, Blastococcus, and Solirubrobacter, were enriched in the H20-5 treated rhizosphere soils. The microbial co-occurrence network of the bacterial community in the H20-5 treated rhizosphere soils had more modules and keystone taxa compared to the control. These findings revealed that the strain H20-5 induced systemic tolerance in tomato plants and influenced the diversity, composition, structure, and network of bacterial communities. The bacterial community in the H20-5 treated rhizosphere soils also appeared to be relatively stable to soil salinity changes.

Rhizobacteria play important roles in plant growth and health enhancement and render them resistant to not only biotic stresses but also abiotic stresses, such as low/high temperature, drought, and salinity. This study aimed to select plant growth promoting rhizobacteria (PGPR) with the capability to mitigate biotic and abiotic stress effects on tomato plants. We isolated a novel PGPR strain, Variovorax sp. PMC12 from tomato rhizosphere. An in vitro assay indicated that strain PMC12 produced ammonia, indole-3-acetic acid (IAA), siderophore, and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase, which are well-known traits of PGPR. The aboveground fresh weight was significantly higher in tomato plants treated with strain PMC12 than in non-treated tomato plants under various abiotic stress conditions including salinity, low temperature, and drought. Furthermore, strain PMC12 also enhanced the resistance to bacterial wilt disease caused by Ralstonia solanacearum. Taken together, these results indicated that strain PMC12 is a promising biocontrol agent and a biostimulant to reduce the susceptibility of plants to both abiotic and biotic stresses.

During a survey of endophytic diazotrophic bacteria associated with different rice varieties in Tamilnadu, some “endophytes” were obtained. Thirteen bacterial isolates from surface-sterilized roots and shoots were obtained in pure culture, which produced indole acetic acid (IAA) and reduced acetylene to ethylene. Polymerase chain reaction (PCR) amplification confirmed the presence of nif-H gene in all the isolates. Morphological, biochemical, and molecular characteristics indicated that all of them belonged to the genus Burkholderia One of them, MGK3, was consistently more active in reducing acetylene, and 16S rDNA sequences of isolate MGK3 confirmed its identification as Burkholderia vietnamiensis. Colonization of rice root was confirmed by strain MGK3 marked with gusA gene. The inoculated roots showed a blue color, which was most intense at the points of lateral root emergence and at the root tip. Transverse sections of roots, 15 days after inoculation, revealed beta-glucuronidase (GUS) activity within many of the cortical intercellular spaces next to the stele and within the aerenchyma. Nitrogen fixation was quantified by using 15N isotope dilution method with two different cultivars grown in pot and field experiments. Higher nitrogen fixation was observed in variety Ponni than in ADT-43, where nearly 42% (field) and 40% (pot) of the nitrogen was derived from the atmosphere (% Ndfa). Isolate MGK3 was used to inoculate rice seedlings in a comparison with four other diazotrophs, viz., Gluconacetobacter diazotrophicus LMG7603, Herbaspirillum seropedicae LMG6513, Azospirillum lipoferum 4B LMG4348, and B. vietnamiensis LMG10929. They were used to conduct two pot and four field inoculation experiments. MGK3 alone, and combined with other diazotrophs, performed best under both pot and field conditions: combined inoculation produced yield increases between 9.5 and 23.6%, while MGK3 alone increased yield by 5.6 to 12.16% over the uninoculated control treatment. Abstract  A group of endophytic diazotrophs were isolated from surface-sterilized roots and stems of different sugarcane varieties in the Tamilnadu region of India. From these, four isolates were selected, based on the highest acetylene reduction activity. Gene-specific PCR amplification confirmed the presence of nif-D genes in those isolates. The 16S rRNA sequence of isolates GR4 and GR7 had a 99.5% sequence similarity to the Pseudomonas sp. pDL01 (AF125317) and 16S rDNA sequence of isolate GR3 had a 100% similarity to that of Burkholderia vietnamiensis (AY973820). The 16S rDNA sequence of isolate GR9 was 99.79% similar to that of the Klebsiella pneumoniae type strain (KPY17657). Colonization by the isolates was confirmed using micropropagated sugarcane and sterile rice seedlings. Isolate GR9, identified as Klebsiella pneumoniae, was consistently more active in reducing acetylene as compared with the other isolates. The effects of GR9 and the sugarcane diazotroph Gluconacetobacter diazotrophicus were compared in inoculated micropropagated sugarcane plantlets. The effects of K. pneumoniae GR9, and four other diazotrophs, G. diazotrophicus, Herbaspirillum seropedicae, Azospirillum lipoferum 4B, and Burkholderia vietnamiensis in inoculated rice seedlings were compared. GR9 alone or in combination with the other diazotrophs performed best under pot conditions. The combined effects of nitrogen fixation and endophytic colonization of this diazotroph may be useful for the development of bio-inoculants. [ABSTRACT FROM AUTHOR]

Drought stress is considered as one of major abiotic stresses; it leads to reduce plant growth and crop productivity. In this study, we selected bacterial strains for alleviating drought stress in chili pepper plants. As drought-tolerant bacteria, 28 among 447 strains were pre-selected by in vitro assays including growth in drought condition with polyethylene glycol and plant growth-promoting traits including production of 1-aminocyclopropane-1-carboxylate deaminase, indole-3-acetic acid and exopolysaccharide. Sequentially, 7 among pre-selected 28 strains were screened based on relative water content (RWC); GLC02 and KJ40, among seven strains were finally selected by RWC and malondialdehyde (MDA) in planta trials under an artificial drought condition by polyethylene glycol solution. Two strains GLC02 and KJ40 reduced drought stress in a natural drought condition as well as an artificial condition. Strains GLC02 or KJ40 increased shoot fresh weight, chlorophyll and stomatal conductance while they decreased MDA in chili pepper plants under a natural drought condition. However, two strains did not show biocontrol activity against diseases caused by Phytophthora capsici and Xanthomonas campestris pv. vesicatoria in chili pepper plants. Taken together, strains GLC02 or KJ40 can be used as bio-fertilizer for alleviation of drought stress in chili pepper plants.

A variety of microorganisms in rhizosphere affect plant health by plant growth promotion, mitigation of abiotic stresses as well as protection from pathogen attacks. In our previous study, we selected a bacterium, Bacillus mesonae H20-5, for alleviation of salinity stress in tomato plants. In this study, we verified the effect of a liquid formulation of B. mesonae H20-5 (TP-H20-5) on fruit production and phytochemical accumulation including lycopene and polyphenol in cherry tomato and strawberry fruits in on-farm tests of protected cultivation under salinity stress. When vegetables including tomato, cherry tomato, strawberry, and cucumber were treated with TP-H20-5 by irrigated systems, final marketable yields were increased by 21.4% (cherry tomato), 9.3% (ripen tomato), 120.6% (strawberry), and 14.5% (cucumber) compared to untreated control. Moreover, treatment of TP-H20-5 was showed increase of phytochemicals such as lycopene and total polyphenol compared to untreated control in cherry tomato and strawberry. Therefore, these results indicated that a formulant of B. mesonae H20-5 can be used as a potential biofertilizer for increasing fruit production and quality.

Plants are exposed to biotic stresses caused by pathogen attack and complex abiotic stresses including heat and drought by dynamic climate changes. To alleviate these stresses, we investigated two bacterial stains, H26-2 and H30-3 in two cultivars (‘Ryeokkwang’ and ‘Buram-3-ho’) of Chinese cabbage in plastic pots in a greenhouse. We evaluated effects of bacterial strains on plant growth-promotion and mitigation of heat and drought stresses; the role of exopolysaccharides as one of bacterial determinants on alleviating stresses; biocontrol activity against soft rot caused by Pectobacterium carotovorum subsp. carotovorum PCC21. Strains H26-2 and H30-3 significantly increased fresh weights compared to a MgSO4 solution; reduced leaf wilting and promoted recovery after re-watering under heat and drought stresses. Chinese cabbages treated with H26-2 and H30-3 increased leaf abscisic acid (ABA) content and reduced stomatal opening after stresses treatments, in addition, these strains stably colonized and maintained their populations in rhizosphere during heat and drought stresses. As well as tested bacterial cells, exopolysaccharides (EPS) of H30-3 could be one of bacterial determinants for alleviation of tested stresses in Chinese cabbages, however, the effects were different to cultivars of Chinese cabbages. In addition to bacterial activity to abiotic stresses, H30-3 could suppress incidence (%) of soft rot in ‘Buram-3-ho’. The tested strains were identified as Bacillus aryabhattai H26-2 and B. siamensis H30-3 based on 16S rRNA gene sequence analysis. Taken together, H26-2 and H30-3 could be candidates for both plant growth promotion and mitigation of heat and drought stresses in Chinese cabbage.
