Pachyphlodes (Pezizaceae) is a genus of truffle-like fungi that is distributed across the Northern Hemisphere. These fungi form ectomycorrhizae primarily with trees in the Fagaceae family, and occasionally with other host plants. The genus Plicariella (= Scabropezia) is phylogenetically inferred as an ally of, or within, the Pachyphlodes lineage. Despite molecular phylogenetic analyses that show the close relationships of species in these two genera, morphological differences in ascomata shape and color, spore ornamentation, and ascus shape are profound. Here, we studied spore wall development to better understand affinities within the Pachyphlodes-Plicariella lineages. Electron microscopy studies indicate that the initial spore wall development is similar across six Pachyphlodes species and a Plicariella species, despite striking differences in mature spore ornamentation among species. Ultrastructural analyses reveal that differences in spore ornamentation among Pachyphlodes species are due to unique developmental events at the final stages of spore wall deposition. Septal pore ultrastructure in Pachyphlodes species is similar to other Pezizaceae that have been studied. Molecular analyses of the five species studied indicate that four of them have not been previously described. The new species Pachyphlodes annagardnerae is here described, and the ultrastructural features of species of Pachyphlodes, Plicariella, and other Pezizales are compared and discussed. Male-sterile, female-fertile plants were used to produce hybrid soybean seed. Manual cross-pollination using male-sterile plants to produce large quantities of hybrid seed is difficult and time-consuming because of the low success rate in cross-pollination. Insect pollinators may be suitable vectors to transfer pollen, but the most suitable vector for pollen transfer from the male parent to the female parent has not been identified for soybean. The objective of the present study was to evaluate seed-set on four male-sterile, female-fertile soybean lines by using alfalfa leafcutting bees (Megachile rotundata (F.)) and honey bees (Apis mellifera (L.)) as pollinators. Seed-set was evaluated in summers 2003 and 2005 near Ames, Iowa, USA and in summers 2003, 2004, and 2005 near Wooster, Ohio, USA. Neither the effect of pollinator species nor the interaction effect of pollinator species×location was significant for any year. Honey bees performed similarly to alfalfa leafcutting bees at both locations. The results indicated significant differences for seed-set among male-sterile lines, suggesting preferential pollination. Male-sterile lines, ms1 (Urbana) and ms2 (Ames 2), had higher cross-pollinated seed-set compared to ms6 (Ames 1), and ms6 (Corsoy 79). At the Ames location, ms1ms1 (Urbana) plants had the highest seed-set (50·16 seeds per male-sterile plant in 2005). At the Wooster location, ms1ms1 (Urbana) plants also had the highest seed-set (92·04 seeds per male-sterile plant) in 2005. Crystal macropatterns in Fagaceae and Nothofagaceae were examined from cleared herbarium leaves (eight genera, 122 spp.) by polarizing microscopy. Prisms and druses dominate, but previously undescribed intermediate forms occur in 25 species of Fagaceae. Sixty-two species had only prisms associated with vein orders 1–4, but smallest orders 5 and 6 had few or no crystals. In mesophyll, 98 species had small druses in spongy parenchyma and larger but fewer druses in palisade parenchyma. Subfamily Fagoideae conformed closely to this macropattern, Castaneoideae less so. Six Castaneoideae species lacked vein crystals. Two Fagoideae species and seven Nothofagus species lacked mesophyll crystals. Druses with prominent cores occur throughout Fagaceae but none were seen in Nothofagaceae. Tabular presentation of results shows significant although overlapping trends from Fagoideae to Castaneoideae to Nothofagaceae, with the latter taxon deviating most from Fagaceae. Calcium excretion and deposition were studied during sporogenesis in the myxomycete, Physarella oblonga. Prior to fruiting, the migrating vegetative plasmodium contains numerous, large channels connected to the external environment. During early sporangial development, these channels become smaller and are isolated as channel remnants. Plasmodial microfilament bundles also disappear at this time. Internally, two types of spheres, each about 0.5-1.5 μm in diameter, are formed in the channel remnants. A dense sphere appears to originate from small particles initially secreted from the mitochondria. These particles then enter the channels and channel remnants by reverse pinocytosis and fuse to form the dense spheres. A second, less dense sphere has no detectable cytoplasmic origin but also appears to form in the channels and channel remnants. Both types of spheres are concentrated by the fusion of channel remnants to form enlarged regions (knots and spikes) of the developing capillitium. Some of the channel remnants also fuse with the outer surface of the sporangium, depositing both spheres to the outside. Both spheres have been studied by various light and electron microscopic techniques and with specific histochemical and analytical procedures. The less dense spheres appear to contain a major portion of calcium in the form of calcium carbonate. These results are compared to other recent studies on calcium deposition in the Myxomycetes and a different mechanism for calcium deposition is proposed. [ABSTRACT FROM AUTHOR]

A cytological study of Texas cytoplasmic male sterile (Tcms) and normal (N) anther tapetal protoplasts of Zea mays was undertaken to determine whether there were any differences prior to Tcms male cell abortion not noted in previous published studies. Squash preparations, tapetal protoplast separation via flow cytometry, image analysis, and electron microscopy were utilized. Chemically preserved tapetal protoplasts from both lines were prominently angular in shape and typically smaller than any other cell type in the anthers. The tapetum from both lines consisted of a mixture of uninucleate and binucleate protoplasts. The Tcms tapetum consistently had a higher proportion of binucleate protoplasts during all stages of microsporogenesis prior to abortion. The size of Tcms uniand binucleate tapetal protoplasts was more variable than the N tapetal protoplasts and was largest during the microspore stage when male cells abort. Tapetal nuclear size in both lines was less variable. Uni- and binucleate tapetal protoplasts from each line could be separated from the other anther cells and from each other by filtration and then by flow cytometry, based on intensity of nuclear fluorescence. These results suggest that Tcms uninucleate tapetal protoplasts have a higher level of DNA than N uninucleate tapetal protoplasts. Both fluorescence microscopy and electron microscopy confirmed pure populations of intact uni- and binucleate tapetal protoplasts using flow cytometry. The results from this study indicate that the methodology presented here could be used for a variety of further studies to better understand the cellular and molecular basis of male sterility in maize, and in other taxa, where the tapetum is the primary target that leads to male sterility. The development and distribution of calcium oxalate crystals, stomates and hairs were studied in the first trifoliolate leaf of Rhynchosia caribaea (Leguminosae: Papilionoideae, Phaseoleae). Using light and transmission electron microscopy, the crystals were shown to occur in both bundle sheath and mesophyll cells. Crystal distribution and shapes are characteristic for Rhynchosia. Crystals develop late in leaf development in contrast to the stomates and hairs. As these latter two structures decrease in number per unit area with leaf age, crystal number increases.
