Background Investigations into alternative treatments for hypertension are necessary because current treatments cannot fully reduce the risk for the development of cardiovascular diseases. Chronic activation of unfolded protein response attributable to the endoplasmic reticulum stress has been proposed as a potential therapeutic target for hypertension and associated vascular remodeling. Triggered by the accumulation of misfolded proteins, chronic unfolded protein response leads to downstream signaling of cellular inflammation and dysfunction. Here, we have tested our hypothesis that a novel chemical chaperone, 3‐hydroxy‐2‐naphthoic acid (3HNA) can attenuate angiotensin II (AngII)‐induced vascular remodeling and hypertension. Methods and Results Mice were infused with AngII for 2 weeks to induce vascular remodeling and hypertension with or without 3HNA treatment. We found that injections of 3HNA prevented hypertension and increase in heart weight body weight ratio induced by AngII infusion. Histological assessment revealed that 3HNA treatment prevented vascular medial thickening as well as perivascular fibrosis in response to AngII infusion. In cultured vascular smooth muscle cells, 3HNA attenuated enhancement in protein synthesis induced by AngII. In vascular adventitial fibroblasts, 3HNA prevented induction of unfolded protein response markers. Conclusions We present evidence that a chemical chaperone 3HNA prevents vascular remodeling and hypertension in mice with AngII infusion, and 3HNA further prevents increase in protein synthesis in AngII‐stimulated vascular smooth muscle cells. Using 3HNA may represent a novel therapy for hypertension with multiple benefits by preserving protein homeostasis under cardiovascular stress.

Summary: Activated factor X is a key component of the coagulation cascade, but whether it directly regulates pathological cardiac remodeling is unclear. In mice subjected to pressure overload stress, cardiac factor X mRNA expression and activity increased concurrently with cardiac hypertrophy, fibrosis, inflammation and diastolic dysfunction, and responses blocked with a low coagulation-independent dose of rivaroxaban. In vitro, neurohormone stressors increased activated factor X expression in both cardiac myocytes and fibroblasts, resulting in activated factor X-mediated activation of protease-activated receptors and pro-hypertrophic and -fibrotic responses, respectively. Thus, inhibition of cardiac-expressed activated factor X could provide an effective therapy for the prevention of adverse cardiac remodeling in hypertensive patients. Abstract Ecosystems across the United States are changing in complex ways that are difficult to predict. Coordinated long‐term research and analysis are required to assess how these changes will affect a diverse array of ecosystem services. This paper is part of a series that is a product of a synthesis effort of the U.S. National Science Foundation’s Long Term Ecological Research (LTER) network. This effort revealed that each LTER site had at least one compelling scientific case study about “what their site would look like” in 50 or 100 yr. As the site results were prepared, themes emerged, and the case studies were grouped into separate papers along five themes: state change, connectivity, resilience, time lags, and cascading effects and compiled into this special issue. This paper addresses the time lags theme with five examples from diverse biomes including tundra (Arctic), coastal upwelling (California Current Ecosystem), montane forests (Coweeta), and Everglades freshwater and coastal wetlands (Florida Coastal Everglades) LTER sites. Its objective is to demonstrate the importance of different types of time lags, in different kinds of ecosystems, as drivers of ecosystem structure and function and how these can effectively be addressed with long‐term studies. The concept that slow, interactive, compounded changes can have dramatic effects on ecosystem structure, function, services, and future scenarios is apparent in many systems, but they are difficult to quantify and predict. The case studies presented here illustrate the expanding scope of thinking about time lags within the LTER network and beyond. Specifically, they examine what variables are best indicators of lagged changes in arctic tundra, how progressive ocean warming can have profound effects on zooplankton and phytoplankton in waters off the California coast, how a series of species changes over many decades can affect Eastern deciduous forests, and how infrequent, extreme cold spells and storms can have enduring effects on fish populations and wetland vegetation along the Southeast coast and the Gulf of Mexico. The case studies highlight the need for a diverse set of LTER (and other research networks) sites to sort out the multiple components of time lag effects in ecosystems.

Cardiovascular disease (CVD) is a prevalent issue in the global aging population. Premature vascular aging such as elevated arterial stiffness appears to be a major risk factor for CVD. Vascular smooth muscle cells (VSMCs) are one of the essential parts of arterial pathology and prone to stress-induced senescence. The pervasiveness of senescent VSMCs in the vasculature increases with age and can be further expedited by various stressing events such as oxidative stress, mitochondria dysfunction, endoplasmic reticulum stress, and chronic inflammation. Angiotensin II (AngII) can induce many of these responses in VSMCs and is thus considered a key regulator of VSMC senescence associated with CVD. Understanding the precise mechanisms and consequences of senescent cell accumulation may uncover a new generation of therapies including senolytic and senomorphic compounds against CVD. Accordingly, in this review article, we discuss potential molecular mechanisms of VSMC senescence such as those induced by AngII and the therapeutic manipulations of senescence to control age-related CVD and associated conditions such as by senolytic.

Investigations of vascular smooth muscle cell (VSMC) phenotypic modulation due to angiotensin II (AngII) stimulation are important for understanding molecular mechanisms contributing to hypertension and associated vascular pathology. AngII induces endoplasmic reticulum (ER) stress in VSMCs, which has been implicated in hypertensive vascular remodeling. Under ER stress, 78 kDa glucose-regulated protein (GRP78) acts as an endogenous chaperone, as well as a master controller of unfolded protein response (UPR) to maintain protein quality control. However, the potential downstream consequences of ER stress induced by AngII on protein quality control and pro-inflammatory phenotype in VSMCs remain elusive. This study aims to identify protein aggregation as evidence of the disruption of protein quality control in VSMCs, and to test the hypothesis that preservation of proteostasis by overexpression of GRP78 can attenuate the AngII-induced pro-inflammatory phenotype in VSMCs. Increases in protein aggregation and enhanced UPR were observed in VSMCs exposed to AngII, which were mitigated by overexpression of GRP78. Moreover, GRP78 overexpression attenuated enhanced monocyte adhesion to VSMCs induced by AngII. Our results thus indicate that the prevention of protein aggregation can potentially mitigate an inflammatory phenotype in VSMCs, which may suggest an alternative therapy for the treatment of AngII-associated vascular disorders.

Angiotensin II (AngII) has a crucial role in cardiovascular pathologies, including endothelial inflammation and premature vascular aging. However, the precise molecular mechanism underlying aging-related endothelial inflammation induced by AngII remains elusive. Here, we have tested a hypothesis in cultured rat aortic endothelial cells (ECs) that the removal of AngII-induced senescent cells, preservation of proteostasis, or inhibition of mitochondrial fission attenuates the pro-inflammatory EC phenotype. AngII stimulation in ECs resulted in cellular senescence assessed by senescence-associated β galactosidase activity. The number of β galactosidase-positive ECs induced by AngII was attenuated by treatment with a senolytic drug ABT737 or the chemical chaperone 4-phenylbutyrate. Monocyte adhesion assay revealed that the pro-inflammatory phenotype in ECs induced by AngII was alleviated by these treatments. AngII stimulation also increased mitochondrial fission in ECs, which was mitigated by mitochondrial division inhibitor-1. Pretreatment with mitochondrial division inhibitor-1 attenuated AngII-induced senescence and monocyte adhesion in ECs. These findings suggest that mitochondrial fission and endoplasmic reticulum stress have causative roles in endothelial senescence-associated inflammatory phenotype induced by AngII exposure, thus providing potential therapeutic targets in age-related cardiovascular diseases.

Abstract Overstory forest structure responds to terrain‐related abiotic factors and to biotic interactions among overstory and understory plants. Unlike species abundance, tree height, biomass, and leaf area in many regions have been poorly quantified in relation to terrain‐driven environmental gradients. In addition, the magnitude of understory influences on overstory structure has been poorly characterized for many forest systems. Our primary goal was to identify relationships between terrain (elevation, convexity, exposure), evergreen understory, and overstory structure (height, aboveground biomass, leaf area) in mature deciduous forests of the southern Appalachian Mountains. We used a combination of field point and plot measurements, LiDAR, and satellite image data to sample little‐disturbed deciduous forest stands. Height, biomass, and gap frequency were significantly related to changes in elevation, exposure (aspect), and convexity (cove to ridge). Higher evergreen understory density was strongly correlated with decreases in forest height and biomass, with an impact observed across moisture, elevation, and exposure gradients. Canopies on ridges averaged half as tall at the highest evergreen understory densities when compared to those without evergreen shrubs (10 vs. 19 m), and overstory canopy height averaged 6 m shorter on sideslopes with high evergreen understory density compared to those with low evergreen understory density. Canopy height declined from low to high elevations, with larger relative decreases on ridges, but biomass increased from low to high elevations, due primarily to high biomass in coves at mid‐ to upper elevations. Biomass and canopy height declined from cove to ridge and north‐ to south‐facing slopes. Responses in canopy height and aboveground biomass associated with changes in understory evergreen density were similar to impacts due to terrain. Gaps were more frequent on south‐facing slopes. Previous studies at this site and others identify soil moisture and soil N competition as the most plausible mechanisms by which understory shrubs might influence overstory canopy structure, with low light limiting seedling recruitment as an additional mechanism. Our work suggests evergreen understory density, particularly on sideslope and ridge locations, substantially affects overstory canopy height and biomass.

Abstract Southern Appalachian riparian forests have undergone changes in composition and function from invasive pathogens and pests. Castanea dentata mortality in the 1930s from chestnut blight (Cryphonectria parasitica) and Tsuga canadensis mortality in the 2000s from the hemlock woolly adelgid (Adelges tsugae) have led to the expansion and increased growth of Rhododendron maximum, an evergreen subcanopy shrub. A better understanding of seed bank characteristics and the various abiotic and biotic factors that affect the seed bank may be useful in determining the restoration potential of forest communities following invasion‐related disturbances. We compared the seed bank of two deciduous forest types: hardwood forests with a dense R. maximum subcanopy (hereafter, RR) and hardwood forests without R. maximum (hereafter, HWD). We evaluated numerous microenvironmental variables through principal component analysis (PCA) and correlated the derived PCA axes scores to seed bank density and richness across forest types. We found that seed bank density was comparable between the forests types; however, seed bank richness was much lower in RR than HWD and the species composition was dissimilar between forest types. Twenty‐eight of 64 (44%) species in the seed bank of HWD were not found in the seed bank of RR. Species that were represented in both forest types were often found in contrasting densities. Most notably, seed bank densities of several woody species were considerably higher in RR (85%) than HWD (45%), while herbaceous seed bank density was lower in RR (11%) than HWD (50%). Mineral soil pH, soil nutrient availability, and soil moisture were lower, and organic soil (Oi + Oe + Oa) depth and mass were greater in the RR than HWD forest type. PCA correlations revealed that PCA4 (represented by understory density and Oe + Oa phosphorus and carbon/nitrogen ratio) was negatively correlated with total seed bank density. PCA1 (represented by Oe + Oa cations and phosphorus, understory richness, ground‐layer cover, and mineral soil pH) and PCA4 were positively correlated with total seed bank richness. These results suggest that the soil seed bank will not be the primary mode of recruitment to establish a diverse and herbaceous‐rich community if a RR is present.

Although AngII (angiotensin II) and its receptor AT1R (AngII type 1 receptor) have been implicated in AAA (abdominal aortic aneurysm) formation, the proximal signalling events primarily responsible for AAA formation remain uncertain. Caveolae are cholesterol-rich membrane microdomains that serve as a signalling platform to facilitate the temporal and spatial localization of signal transduction events, including those stimulated by AngII. Cav1 (caveolin 1)-enriched caveolae in vascular smooth muscle cells mediate ADAM17 (a disintegrin and metalloproteinase 17)-dependent EGFR (epidermal growth factor receptor) transactivation, which is linked to vascular remodelling induced by AngII. In the present study, we have tested our hypothesis that Cav1 plays a critical role for the development of AAA at least in part via its specific alteration of AngII signalling within caveolae. Cav1−/− mice and the control wild-type mice were co-infused with AngII and β-aminopropionitrile to induce AAA. We found that Cav1−/− mice with the co-infusion did not develop AAA compared with control mice in spite of hypertension. We found an increased expression of ADAM17 and enhanced phosphorylation of EGFR in AAA. These events were markedly attenuated in Cav1−/− aortas with the co-infusion. Furthermore, aortas from Cav1−/− mice with the co-infusion showed less endoplasmic reticulum stress, oxidative stress and inflammatory responses compared with aortas from control mice. Cav1 silencing in cultured vascular smooth muscle cells prevented AngII-induced ADAM17 induction and activation. In conclusion, Cav1 appears to play a critical role in the formation of AAA and associated endoplasmic reticulum/oxidative stress, presumably through the regulation of caveolae compartmentalized signals induced by AngII.
