Marine phytoplankton contribute about one half of global primary production and play a key role in global biogeochemical cycles. High cell densities in extensive phytoplankton blooms are expected to be modified by global changes in ocean circulation and stratification, acidification and carbonation, solar radiation, temperature, and eutrophication. Although photochemical gas production from chromophoric dissolved organic matter (CDOM) has been widely studied, ultraviolet (UV) effects on emissions from phytoplankton cells themselves have not been fully explored. We therefore investigated UV-driven emissions of carbon monoxide (CO), carbon dioxide (CO2), methane (CH4), ethene (C2H4), ethane (C2H6), and nitrous oxide (N2O) from cell suspensions of 16 phytoplankton species and their filtrates under controlled experimental conditions. These gases make direct or indirect contributions to radiative forcing of the atmosphere or contribute to atmospheric chemistry including stratospheric ozone (O3) depletion. We observed production of CH4, CO, CO2, C2H4, and N2O from cell suspensions and CO, CO2, and N2O after 0.45 μm-filtration to remove phytoplankton cells. CH4 production was only observed with cells present, whereas N2O was still produced after filtration. Production of CO from filtrates was 30%–90% of that from cell suspensions in all but two species with a CO2:CO mole ratio from filtrates always below one. Our results clearly demonstrate a need to quantify the production potentials of these climate-relevant gases in situ under natural sunlight using key phytoplankton species, especially those forming blooms which are predicted to change in prevalence and distribution with future global change scenarios.

Temperate shelf seas are productive areas with the potential to export high quantities of particulate organic carbon (POC), as sinking particles, to the sediments or off-shelf to the open ocean. The amount of carbon which can be exported depends partly on the amount of POC produced and on the remineralization processes occurring on the sinking material. Here, we assessed the relative seasonal importance of microbial respiration and bacterial production associated with suspended, slow- and fast-sinking particle fractions. The three fractions were collected in the Celtic Sea above and below the seasonal thermocline in November 2014, April and July 2015 using Marine Snow Catchers. The slow-sinking fraction had higher microbial respiration and bacterial production rates than the fast-sinking fractions, and these two fractions sustained rates of microbial respiration and bacterial production between 1 and 3 orders of magnitude lower than the suspended fraction. This low contribution of the slow- and fast-sinking fractions was consistent with their low contribution to the POC concentration at the two depths sampled. The POC-specific respiration rates associated with the slow- and fast-sinking fractions were low (median 0.17 and 0.08 d−1, respectively), indicating low-sinking particle degradation. Our results indicate that ∼5% of the POC in surface waters can be exported below the thermocline.

Fjordic coastlines provide an ideal protected environment for both finfish and shellfish aquaculture operations. This study reports the results of a cruise to the Scottish Clyde Sea, and associated fjordic sea lochs, that coincided with blooms of the diarrhetic shellfish toxin producing dinoflagellate Dinophysis acuta and the diatom genus Chaetoceros, that can generate finfish mortalities. Unusually, D. acuta reached one order of magnitude higher cell abundance in the water column (2840 cells L−1) than the more common Dinophysis acuminata (200 cells L−1) and was linked with elevated shellfish toxicity (maximum 601 ± 237 μg OA eq/kg shellfish flesh) which caused shellfish harvesting closures in the region. Significant correlations between D. acuta abundance and that of Mesodinium rubrum were also observed across the cruise transect potentially supporting bloom formation of the mixotrophic D. acuta. Significant spatial variability in phytoplankton that was related to physical characteristics of the water column was observed, with a temperature-driven frontal region at the mouth of Loch Fyne being important in the development of the D. acuta, but not the Chaetoceros bloom. The front also provided important protection to the aquaculture located within the loch, with neither of the blooms encroaching within it. Analysis based on a particle-tracking model confirms the importance of the front to cell transport and shows significant inter-annual differences in advection within the region, that are important to the harmful algal bloom risk therein.

The objective of this scoping review was to use an established methodology and the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) reporting framework to map the evidence for associations between marine HABs and observed both acute and chronic human health effects. A systematic and reproducible search of publications from 1985 until May 2019 was conducted using diverse electronic databases. Following de-duplication, 5,301 records were identified, of which 380 were included in the final qualitative synthesis. The majority of studies (220; 57.9%) related to Ciguatera Poisoning. Anecdotal and case reports made up the vast majority of study types (242; 63.7%), whereas there were fewer formal epidemiological studies (35; 9.2%). Only four studies related to chronic exposure to HABs. A low proportion of studies reported the use of human specimens for confirmation of the cause of illness (32; 8.4%). This study highlighted gaps in the evidence base including a lack of formal surveillance and epidemiological studies, limited use of toxin measurements in human samples, and a scarcity of studies of chronic exposure. Fjordic coastlines provide sheltered locations for finfish and shellfish aquaculture, and are often subject to harmful algal blooms (HABs) some of which develop offshore and are then advected to impact nearshore aquaculture. Numerical models are a potentially important tool for providing early warning of such HAB events. However, the complex topography of fjordic shelf regions is a significant challenge to modelling. This is frequently compounded by complex bathymetry and local weather patterns. Existing structured grid models do not provide the resolution needed to represent these coastlines in their wider shelf context. In a number of locations advectively transported blooms of the ichthyotoxic dinoflagellate Karenia mikimotoi are of particular concern for the finfish industry. Here were present a novel hydrodynamic model of the coastal waters to the west of Scotland that is based on unstructured finite volume methodology, providing a sufficiently high resolution hydrodynamical structure to realistically simulate the transport of particles (such as K. mikimotoi cells) within nearshore waters where aquaculture sites are sited. Model–observation comparisons reveal close correspondence of tidal elevations for major semidiurnal and diurnal tidal constituents. The thermohaline structure of the model and its current fields are also in good agreement with a number of existing observational datasets. Simulations of the transport of Lagrangian drifting buoys, along with the incorporation of an individual-based biological model, based on a bloom of K. mikimotoi, demonstrate that unstructured grid models have considerable potential for HAB prediction in Scotland and in complex topographical regions elsewhere.

The “Applied Simulations and Integrated Modelling for the Understanding of Harmful Algal Blooms” (Asimuth) project sought to develop a harmful algal bloom (HAB) alert system for Atlantic Europe. This was approached by combining, at a national or regional level, regulatory monitoring phytoplankton and biotoxin data with satellite remote sensing and other information on current marine conditions, coupled with regional scale models that included a representation of HAB transport. Synthesis of these products was achieved by expert interpretation within HAB risk alert bulletins that were prepared on a regular basis (typically weekly) for use by the aquaculture industry. In this preface to the Asimuth Special Issue we outline the main HAB species of concern in the region and the strengths and limitations of different methodologies to provide early warning of their blooms.

The ‘English’ have an ambiguous relationship with the language of their nearest neighbour, France. There is a history, ‘1066 and all that’. Christmas Day 2016 saw the 950th anniversary of the crowning at Westminster Abbey of the French Duke William of Normandy as King of England (not, please note, of ‘Britain’); William having defeated King Harold – and so the very last ‘English’ monarch – at the battle of Hastings. (The other ‘nations’ of these European off-shore islands have different trajectories.) Sooner or later the English at least would have to come to terms with French.

Cyclic imines (CIs) are a group of phytoplankton produced toxins related to shellfish food products, some of which are already present in UK and European waters. Their risk to shellfish consumers is poorly understood, as while no human intoxication has been definitively related to this group, their fast acting toxicity following intraperitoneal injection in mice has led to concern over their human health implications. A request was therefore made by UK food safety authorities to examine these toxins more closely to aid possible management strategies. Of the CI producers only the spirolide producer Alexandrium ostenfeldii is known to exist in UK waters at present but trends in climate change may lead to increased risk from other organisms/CI toxins currently present elsewhere in Europe and in similar environments worldwide. This paper reviews evidence concerning the prevalence of CIs and CI-producing phytoplankton, together with testing methodologies. Chemical, biological and biomolecular methods are reviewed, including recommendations for further work to enable effective testing. Although the focus here is on the UK, from a strategic standpoint many of the topics discussed will also be of interest in other parts of the world since new and emerging marine biotoxins are of global concern.

Harmful algal blooms (HABs) are thought to be increasing in coastal waters worldwide. Anthropogenic nutrient enrichment has been proposed as a principal causative factor of this increase through elevated inorganic and/or organic nutrient concentrations and modified nutrient ratios. We assess: 1) the level of understanding of the link between the amount, form and ratio of anthropogenic nutrients and HABs; 2) the evidence for a link between anthropogenically generated HABs and negative impacts on human health; and 3) the economic implications of anthropogenic nutrient/HAB interactions. We demonstrate that an anthropogenic nutrient-HAB link is far from universal, and where it has been demonstrated, it is most frequently associated with high biomass rather than low biomass (biotoxin producing) HABs. While organic nutrients have been shown to support the growth of a range of HAB species, insufficient evidence exists to clearly establish if these nutrients specifically promote the growth of harmful species in preference to benign ones, or if/how they influence toxicity of harmful species. We conclude that the role of anthropogenic nutrients in promoting HABs is site-specific, with hydrodynamic processes often determining whether blooms occur. We also find a lack of evidence of widespread significant adverse health impacts from anthropogenic nutrient-generated HABs, although this may be partly due to a lack of human/animal health and HAB monitoring. Detailed economic evaluation and cost/benefit analysis of the impact of anthropogenically generated HABs, or nutrient reduction schemes to alleviate them, is also frequently lacking.

The co-occurrence of morphologically identical toxic and nontoxic ribotypes of the biotoxin producing marine dinoflagellate Alexandrium tamarense presents a significant problem for its identification and enumeration, particularly in a regulatory monitoring context. To address this, we have developed a fluorescence in situ hybridization-flow cytometry (FISH-FC)—based method of cell identification and enumeration. This employed the taxa specific oligonucleotide probes TamToxC and TamA to fluorescently label (with the fluorochromes CY.3 and FITC) Group I (toxic) and Group III (nontoxic) A. tamarense ribotypes, respectively. Detection was by fluorescence activated flow cytometric analysis. The FISH-FC method allowed effective discrimination between laboratory cultures of Group I and Group III ribotypes, with toxic and nontoxic cells creating distinct, easily identifiable, clusters in a flow cytometer bi-plot of side scatter (SSC) versus the green (FL1) fluorescence detection channel. Comparison of estimates of cell abundance obtained by the FISH-FC technique with those obtained by microscopy (Sedgwick Rafter technique) showed no statistically significant difference across a range of concentrations. Subsequently, the methodology was successfully applied on natural seawater samples, spiked with known concentrations of toxic and nontoxic A. tamarense cells at environmentally relevant concentrations.

The abundance and productivity of phytoplankton and bacterioplankton in the region of Jones Bank (Celtic Sea) were assessed in relation to potential physical and chemical drivers of the marine microbial community during July 2008 on RRS James Cook cruise JC25. A major storm dominated the early part of the cruise; this influenced the microbial community with a decrease in diatom abundance being evident due to mixing of cells to sub euphotic zone depths. In the euphotic zone, phytoplankton was dominated by Phaeocystis cells which, with time, became constrained within a chlorophyll maximum that was located near the top of the nutricline. Multivariate statistical analyses found that temporal changes in diatom and dinoflagellate communities were significantly related to water temperature (diatoms) and nitrate concentration (dinoflagellates). Post storm, primary productivity was greatest in the Phaeocystis dominated 2–20 μm size fraction, but with no obvious topographically generated differences between on and off bank stations. Use of a mathematical model demonstrated the likely enhancement of productivity by the storm event, and also that this storm would have masked any topographic influence on productivity. The primary production:bacterial production ratio decreased significantly with time during the cruise, suggesting active competition by bacteria with phytoplankton for mineral nutrients. This was confirmed by a deck board enrichment experiment that demonstrated a rapid increase in bacterial productivity following supplementation of the euphotic zone microbial community with nutrient rich water collected below the mixed layer.

There is a perception that anthropogenically-driven changes in nutrient supply to coastal waters influences the abundance, frequency and toxicity of harmful algal blooms (HABs) through a change in the form or ratio of nutrient that limits phytoplankton growth. If nutrient concentrations are not limiting for growth, then ratios do not influence floristic composition. At non-limiting concentrations, evidence that alteration of nitrogen: phosphorus (N:P) ratios has stimulated HABs is limited, and primarily based on hypothesised relationships in relatively few locations (in particular: Tolo Harbour Hong Kong and Dutch Coastal Waters). In all cases, an unequivocal causal link between an increase in HABs (frequency, magnitude or duration) and change in N or P as the limiting nutrient is difficult to establish. The silicon (Si) limitation hypothesis is generally supported by experimental evidence and field data on the nuisance flagellate Phaeocystis. We found little evidence that high N:Si ratios preferentially promote harmful dinoflagellates over benign species. Laboratory studies demonstrate that nutrient ratios can influence toxin production, but genus and species specific differences and environmental control make extrapolation of these data to the field difficult. Studies of the role of dissolved and particulate organic nutrients in the growth of HAB species, while limited, demonstrate the potential for organic nutrients (especially organic N) to support the growth of a range of HAB species. There is a clear need for better understanding of the role of mixotrophy in the formation of HABs and for studies of HAB and non-HAB species in competition for environmentally realistic concentrations of organic nutrients.

The concept of combining bioenergy with carbon capture and storage (BECCS) has been identified as one mechanism to achieve energy production with a net negative atmospheric carbon emission. Conventional geological carbon capture and storage involves large capital outlay and recurring expense. Therefore there is increased interest in coupling biological carbon capture and sequestration with biomass production. Systems such as low-input high-diversity native grasslands and biochar production have been heralded as carbon negative biofuels. Another potential production system for bioenergy with biological carbon capture and storage (BEBCCS) is the large-scale cultivation of seaweed (macroalgae) for biofuels. Marine biofuels offer considerable advantages over terrestrial first-generation biofuels in terms of their low land use, fresh water and fertilizer requirements. In addition, they also offer considerable potential in the field of BEBCCS. Macroalgae rapidly accumulate biomass and as part of this process, a significant proportion (estimates range up to 60%) of the carbon they fix photosynthetically is released into the water. A proportion of this released dissolved organic carbon (DOC) is highly labile and enters the bacterial loop and is rapidly remineralized back to CO2. However, a proportion is known to be resistant to biological degradation and enters the refractory DOC pool. The marine DOC pool is the largest organic carbon pool on the planet and the refractory component is known to have a turnover period of 100s-1000s years. The release of refractory DOC from macroalgae combined with biofuel production offers the potential for large scale carbon negative fuel production. © 2012 Society of Chemical Industry and John Wiley & Sons, Ltd

Marine aquaculture relies on coastal habitats that will be affected by climate change. This review assesses current knowledge of the threats and opportunities of climate change for aquaculture in the UK and Ireland, focusing on the most commonly farmed species, blue mussels (Mytilus edulis) and Atlantic salmon (Salmo salar). There is sparse evidence to indicate that climate change is affecting aquaculture in the UK and Ireland. Impacts to date have been difficult to discern from natural environmental variability, and the pace of technological development in aquaculture overshadows effects of climatic change. However, this review of broader aquaculture literature and the likely effects of climate change suggests that over the next century, climate change has the potential to directly impact the industry. Impacts are related to the industry's dependence on the marine environment for suitable biophysical conditions. For instance, changes in the frequency and strength of storms pose a risk to infrastructure, such as salmon cages. Sea-level rise will shift shoreline morphology, reducing the areal extent of some habitats that are suitable for the industry. Changes in rainfall patterns will increase the turbidity and nutrient loading of rivers, potentially triggering harmful algal blooms and negatively affecting bivalve farming. In addition, ocean acidification may disrupt the early developmental stages of shellfish. Some of the most damaging but least predictable effects of climate change relate to the emergence, translocation and virulence of diseases, parasites and pathogens, although parasites and diseases in finfish aquaculture may be controlled through intervention. The spread of nuisance and non-native species is also potentially damaging. Rising temperatures may create the opportunity to rear warmer water species in the UK and Ireland. Market forces, rather than technical feasibility, are likely to determine whether existing farmed species are displaced by new ones.

Among common speculations about the ultimate demise of English as the world's lingua franca (see Jeffrey Gil in ET 105, March 2011, reconsidering Chinese as a possible replacement) Nicholas Ostler (2010) is one more to project ‘the breakdown of English-speaking hegemony’, but his case is more curious than most. After an exhaustive, not to say exhausting, survey of ancient empires and modes of communication, in which Latin as the last lingua franca has but a late bit part, he arrives at an unrelated conclusion: ready machine translation sooner or later rendering a global ‘lingua-franca’ irrelevant (his hyphenation to legitimise an English plural – ‘lingua-francas’). Our springtime island-hopping pilgrimage rather gave the lie to this.

High-biomass harmful algal blooms can kill farmed fish through toxicity, physical effects or de-oxygenation of the water column. These blooms often form over spatially large areas meaning that Earth observation is well placed to monitor and study them. In this letter, we present a statistical-based background subtraction technique that has been modified to detect high-biomass algal blooms. The method builds upon previous work and uses a statistical framework to combine spatial and temporal information to produce maps of bloom extent. Its statistical nature allows the approach to characterize the region of interest meaning that region-specific tuning is not needed. The accuracy of the approach has been evaluated using Moderate Resolution Imaging Spectroradiometer (MODIS) data and an in situ cell concentration dataset, resulting in a correct classification rate of 68.0% with a false alarm rate of 0.24 (n = 25). 7 páginas, 2 figuras, 4 tablas, Production of dissolvedorganicmatter (DOM) by heterotrophic microbial communities isolated from Loch Creran (Scotland) was studied in time course incubations in which cells were re-suspended in artificial seawater amended with variable proportions of glucose, ammonium and phosphate. The incubation experiments demonstrated that microheterotrophs released part of the substrate as new DOM, with a production efficiency of 11 ± 1% for DOC, 18 ± 2% for DON and 17 ± 2% for DOP. Estimating the impact of this production in Loch Creran, showed that from 3 ± 1% (DOC) to 72 ± 16% (DOP) of DOM could originate from the heterotrophic microbial community. The produced DOM (PDOM) was both bioavailable (BDOM) and refractory (RDOM). Bioavailability as assessed by the difference between the maximum and the end DOM concentration, was generally higher than found in natural systems, with DOP (73 ± 15%, average ± SD) more bioavailable than DON (70 ± 15%), and DON than DOC (34 ± 13%). The stoichiometry of PDOM was linked to both nutrient uptake and BDOM ratios. Absorption and fluorescence of DOM increased significantly during the incubation time, indicating that microheterotrophs were also a source of coloured DOM (CDOM) and that they produce both bioavailable protein-like and refractory humic-like fluorophores, This study was funded by a fellowship to C.L from the Early Training site ECOSUMMER (Marie Curie action 20501)

A prolonged bloom of Karenia mikimotoi was observed during 2006 in Scottish waters. This bloom is thought to be unique in the region in terms of its large spatial extent. From its first detection in the west of the country, the bloom moved clockwise around the coast eventually reaching the east coast and the Shetland Isles to the north. The bloom resulted in extensive mortalities of benthic organisms including annelids and molluscs and some species of fish. Farmed fish mortalities were absent but gill damage was reported. The availability of satellite remote sensing, phytoplankton counts from multiple sites, meteorological data and some water chemistry, as well as information on the physical characteristics of the sampling sites, provided an extensive spatial and temporal data set. Analysis of remotely sensed chlorophyll-a data from Aqua-MODIS indicated that this parameter is a useful early warning indicator of K. mikimotoi in shelf waters off the Scottish west coast, and suggested that the bloom developed in this region prior to its advection to coastal waters. An earth observation (EO) based harmful bloom classifier for K. mikimotoi recognised areas of elevated K. mikimotoi cell density but generated false positives in areas of high reflectance. Data were also used to evaluate, in Scottish waters, various hypotheses that exist to explain the formation of K. mikimotoi blooms including phototaxis, nutrient availability, cell transport and elevated water temperature. Specifically, we sought to evaluate if routinely collected environmental data (water temperature, insolation, wind strength and direction, and sea-loch aspect) could be used as a predictor of bloom magnitude near aquaculture facility locations, which typically lie within fjordic sea lochs. Path analysis was used to derive intuitive models linking environmental drivers to bloom magnitude. Once the effects of latitude such as northward water cooling were taken into account, only rainfall was a significant predictor of bloom magnitude at the sampling sites. Therefore, while the offshore development and progression of a bloom may be predicted from satellite information, it is likely that local hydrodynamic influences are crucial in determining its magnitude at coastal aquaculture sites.

Through their ability to regulate production of the key lipid messenger PtdIns(3,4,5)P(3), the class I phosphatidylinositol-3-OH kinases (PI(3)Ks) support many critical cell responses. They, in turn, can be regulated by cell-surface receptors through signals acting on either their adaptor subunits (for example, through phosphotyrosine or Gbetagammas) or their catalytic subunits (for example, through GTP-Ras). The relative significance of these controlling inputs is undefined in vivo. Here, we have studied the roles of Gbetagammas, the adaptor p101, Ras and the Ras binding domain (RBD) in the control of the class I PI(3)K, PI(3)Kgamma, in mouse neutrophils. Loss of p101 leads to major reductions in the accumulation of PtdIns(3,4,5)P(3), activation of protein kinase B (PKB) and in migration towards G-protein activating ligands in vitro, and to an aseptically inflamed peritoneum in vivo. Loss of sensitivity of PI(3)Kgamma to Ras unexpectedly caused similar reductions, but additionally caused a substantial loss in production of reactive oxygen species (ROS). We conclude that Gbetagammas, p101 and the Ras-RBD interaction all have important roles in the regulation of PI(3)Kgamma in vivo and that they can simultaneously, but differentially, control distinct PI(3)Kgamma effectors.

Pseudo-nitzschia seriata (Cleve) H. Peragallo isolated from Scottish west coast waters was studied in laboratory batch culture. The neurotoxin domoic acid was produced in both P- and Si-limited stationary phase, but in greater quantities when Si was the limiting nutrient. A mathematical model based on cell quota kinetics was used to simulate P. seriata growth and toxin production. The model was capable of simulating changes in cell density and carbon biomass in the independent Si- and P-limited experiments using a single parameter set. In the model, toxin production was related to C cell−1 and triggered by nutrient stress, defined by low values of the carbon-based cell quota of the limiting nutrient. The study therefore suggests that simple models, based on easily measured quantities, are capable of simulating Pseudo-nitzschia growth and toxin production.

Marine planktonic diatoms of the genus Pseudo-nitzschia Peragallo have been responsible for amnesic shellfish poisoning (ASP) events worldwide through the production of the neurotoxin domoic acid (DA). The appearance and toxicity of Pseudo-nitzschia species is variable throughout the year and potentially linked to changes in environmental parameters; many ASP events occur in relatively high latitudes where day length is particularly variable with season. In UK waters, shellfish monitoring has prevented any impact on human health but has led to long-term closures of fisheries, with severe economic consequences. Laboratory experiments on two Pseudo-nitzschia species typically found in Scottish West Coast waters during spring (short photoperiod (SP)) and summer (long photoperiod (LP)) conditions were conducted to determine the influence of photoperiod on their growth and toxicity. Results indicated that non-toxic P. delicatissima (Cleve) Heiden achieved a greater cell density under SP (9-h light:15-h dark (L:D) cycle). For toxin-producing P. seriata (Cleve) H. Peragallo, a LP (18-h L:6-h D cycle) resulted in an enhanced growth rate, cell yield and total toxin production, but it decreased the toxin production per cell. A better understanding of the response of Pseudo-nitzschia species to photoperiod and other foreseeable environmental variables may help predict the appearance of toxic strains.

Nitrate loading to coastal waters has increased over recent decades while silicon loading has remained relatively constant or decreased. As the N:Si ratio in coastal waters shifts due to these anthropogenic influences, silicate limitation of diatom biomass may become a feature of the biogeochemistry in coastal waters especially in regions of reduced exchange. Two sets of nutrient enrichment mesocosm experiments were conducted in successive years using a natural planktonic assemblage obtained from the Trondheimsfjord, Norway. The inorganic nutrient concentrations at the start of the experiments were manipulated to give a variety of N:Si concentrations at ratios representative of current and possible future values, should N loading continue. In June 1999 experiments were conducted with a gradient of inorganic N:Si ratios (1:2, 1:1, 2:1, 4:1) to investigate the influence of low and high N:Si ratio conditions and to determine the conditions that would generate Si limitation of diatom growth. In June 2000, based on 1999 data, highly replicated experiments were conducted at N:Si ratios of 1:1 and 4:1 which were expected to result in N and Si limitation of diatom growth, respectively; statistical differences in cellular composition were recorded. N limitation of diatom biomass increase was observed under the three lowest N:Si ratios: particulate carbon (C) accumulation continued to occur following N exhaustion resulting in an increase in the organic C:N ratio. Silicate limitation of diatom biomass increase only occurred at the highest N:Si ratio of 4:1. Silicate exhaustion was followed by continued nitrate uptake for several days, at a slower rate than previously. The resulting increase in organic N was accompanied by an increase in organic C such that the C:N ratio of the organic material at the highest N:Si ratio failed to increase to the extent observed under the N limited conditions. Statistically significant differences in chlorophyll-a yield per unit nitrate, C:chlorophyll-a ratios, C:N ratio and diatom cell yield per unit nitrate or Si were observed in Si compared to N limited conditions. All mesocosms became dominated numerically and in terms of biomass by the diatom Skeletonema costatum. The potential implications of changing N and Si regimes in coastal waters are discussed.

These notes are largely based on Lodge (2009), and the IPA transcriptions on Wells (2000).Michael Bulley has singled out for discussion one variant in the articulation of the /r/ phoneme in British English which he characterises as ‘w-for-r’, ‘close to /w/, but distinct from it’ (Bulley, 2014: 46). The focus is on /r/ in syllable-initial position, including /r/ in initial consonant clusters. He describes this particular, not to say singular, variant as ‘/w/-like’, which begs a number of questions.

In order to study the effect of the nitrogen:silicon (N:Si) ratio on temperate microplankton food webs, mesocosm experiments were conducted in Trondheim (Norway) using two different ratios (molar ratios of 1:1 and 4:1). With the exception of diatoms, the increase in abundance of all microbial groups [phototrophic nanoflagellates, autotrophic dinflagellates, bacteria, heterotrophic nanoflagellates (HNAN), heterotrophic microflagellates and ciliates] was significantly greater in the high N:Si treatment. Midway through the experiment, HNAN biovolume exceeded that of bacteria in the high N:Si treatment, indicating strong top-down grazing control. Heterotrophic microflagellate biovolume exceeded ciliate biovolume under both nutrient regimes. However, heterotrophic plankton failed to respond rapidly to increased diatom biomass. Nutrient manipulation experiments were conducted on a natural planktonic community in outdoor mesocosms. Inorganic nitrogen (N) and silicon (Si) were added to achieve N[ratio ]Si ratios of 1[ratio ]1 and 4[ratio ]1. Total particulate carbon (PC) biomass of the microbial assemblage was determined by elemental analysis. Cell volume measurements by microscope on individual components of the community (bacteria, diatoms, photosynthetic nanoflagellates, heterotrophic nanoflagellates, dinoflagellates and ciliates) were also made. We applied published C[ratio ]volume relationships to determine the volume estimated C content (CBV) of these microbial groups and hence of the total assemblage. The total CBV and total PC were compared to test the applicability of C[ratio ]volume relationships under different nutrient regimes both before and after nutrient exhaustion. For initial N[ratio ]Si ratios of 1[ratio ]1, prior to nutrient exhaustion, the relationship between CBV and PC was linear with a gradient of approximately 1, (0.99±0.06), indicating that the published C[ratio ]volume relationships accurately predicted the C content of the microbial assemblage. For N[ratio ]Si ratios of 4[ratio ]1, a linear relationship was again evident between CBV and PC (slope: 1.36±0.08). However, statistical comparison using a general linear model indicated that the gradient of this relationship differed significantly from that when the N[ratio ]Si ratio was 1[ratio ]1, and hence CBV overestimated elemental C. For both N[ratio ]Si ratios, subsequent to nutrient exhaustion (N or Si), and hence when the diatom fraction of the microbial assemblage was in yield-limited post-exponential phase, the two measures of biomass were not well correlated. This indicated that measured cytoplasmic cell volume was a poor indicator of C biomass within the microbial assemblage in nutrient-deplete conditions.

We show that matrices carrying the tethered homologs of natural phosphoinositides can be used to capture and display multiple phosphoinositide binding proteins in cell and tissue extracts. We present the mass spectrometric identification of over 20 proteins isolated by this method, mostly from leukocyte extracts: they include known and novel proteins with established phosphoinositide binding domains and also known proteins with surprising and unusual phosphoinositide binding properties. One of the novel PtdIns(3,4,5)P3 binding proteins, ARAP3, has an unusual domain structure, including five predicted PH domains. We show that it is a specific PtdIns(3,4,5)P3/PtdIns(3,4)P2-stimulated Arf6 GAP both in vitro and in vivo, and both its Arf GAP and Rho GAP domains cooperate in mediating PI3K-dependent rearrangements in the cell cytoskeleton and cell shape.

The ability of mathematical models to simulate competition for nutrients between three algal species, the diatom Thalassiosira pseudonana, a marine raphidophyte Heterosigma carterae and the dinoflagellate Alexandrium minutum,. was investigated. Transient growth models were parameterized and tested using a number of closely controlled laboratory data sets including batch monocultures, batch competition experiments and semi-continuous culture competition experiments. The cell quota model of algal growth was found to be adequate to simulate growth of both the raphidophyte and the dinoflagellate. Batch monoculture data for diatom growth obtained under either nitrogen (N) or silicon (Si) limitation could also be simulated with a quota-style model, which in this case included feedback inhibition of nutrient uptake. However, to simulate both batch and semicontinuous culture experiments (and competition between the species), it was necessary to consider diatom Si-N metabolism. A model was derived which contains a representation of both intracellular N and Si, and of the interaction of these nutrients within the cell. The model used a co-nutrient limitation based on the perceived functional and structural role of N and Si, respectively, within the cell. Simulations indicated that models capable of adequately representing monoculture growth in batch culture may produce erroneous results when incorporated into models of competition. The co-nutrient model is a first step to producing tractable algal growth models which will represent multiple nutrient stress in transient growth conditions.

In order to test rigorously the transient behaviour of mathematical models of algal growth, detailed laboratory data sets with good temporal resolution are required. A series of algal growth experiments was conducted in transient conditions. Monoculture growth of, and competition for nutrients between, three contrasting species of phytoplankton (the diatom Thalassiosira pseudo- nana, the harmful flagellate Heterosigma carterae and the toxic dinoflagellate Alexandrium minutum) were investigated in different temperature, light and nutrient regimes. Although growth dynamics were qualitatively similar in batch culture, quantitative differences were evident in the growth response of the different species when grown in single yield-limiting nutrient conditions in identical physical conditions. Quantities such as the carbon:nitrogen (C:N) ratio and C and N per cell varied between species and within species under different growth conditions. Such results have particular significance to the development of mathematical models, which commonly represent algal popu- lations as a single homogeneous group using a single currency such as numbers, C or N. Changes in light and temperature regime influenced algal growth: Alexandrium failed to grow at low tempera- tures, while specific growth rates of Thalassiosira were more sensitive to changes in temperature than those of Heterosigma. Changes in the dominant organism(s) and/or its size or nutrient status may influence the transfer of nutrients within the food web. Commonly, mathematical models make cell growth a function of a single yield-limiting nutrient. Decreased growth rates and high residual nutrient concentrations in competition experiments indicate that this approach is unlikely to be successful in conditions of limited supply of more than one nutrient, where multiple nutrient stresses will be significant.

Notions of grammar, spelling and punctuation in the National Curriculum for schools in England – an introduction to the NATE Position Paper

The fluorescence behaviour of thin films of poly( N-vinyl carbazole), PNVCz, and a series of N-vinyl carbazole/methyl acrylate, NVCz/MA, copolymers has been investigated using both steady-state and time-resolved emission techniques. The fluorescence of PNVCz at 298 K is dominated by emission from two excimeric traps. Trap I has the conventional ‘sandwich’ structure formed from two fully overlapped aromatic rings. The other (trap II) is a species involving two partially overlapped carbazole substituents. The fluorescences of the NVCz/MA copolymers contain contributions from unassociated or monomeric carbazoyl excited states which increase in prominence as the aromatic content of the system is reduced. The influence of intermolecular interactions in creating excimer traps is apparent: even at a carbazole content of 8.3 mol%, excimer emission is evident. In PNVCz and copolymers of higher aromatic contents, the microtacticity of the macromolecule exerts a dominant influence upon the photophysical behaviour of the bulk polymer despite the moderating effects of intermolecular interactions between chromophores. As the temperature of the system is reduced, the contribution from trap II to the overall fluorescence from the polymer films increases relative to that made by emission from trap I. At 77 K, the fluorescence of PNVCz contains a significant contribution from trap II emission. At 298 K, fluorescence from the conventional excimer, trap I, dominates the steady-state emission spectrum. On the basis of observations upon time-resolved fluorescence data, it is proposed that restrictions to the reorientation of carbazole substituents which occur at low temperature serve to inhibit the conversion of a proportion of the trap II sites into fully overlapped excimers, thereby reducing depletion of the trap II population. Implicit to this explanation of the photophysical characteristics of PNVCz and the higher content NVCz/MA copolymers, is the belief that the high concentration of excimer-forming trap sites mitigates against significant energy migration between carbazole substituents which might otherwise populate the excimer traps. These observations are of importance to considerations of the photoconductivity displayed by polymers derived from NVCz.

A discussion of the role of English in a unique Mediterranean island, in relation to Margie Berns's article ‘English in the European Union’ (ET43, Jul 95)

In mathematical models, the decoupling of the kinetics of nutrient assimilation and growth under transient conditions is traditionally handled by using either time lags (which are dynamically unsatisfactory) or multiple intracellular nutrient compartments (which are difficult to paramaterize). We use the estimated time required for nutrient processing to calculate the quantity of processed nutrient held within the cells. We then determine growth rate by the mean cell content of processed nutrient. The resulting model successfully simulates the transient growth of nitrogen-limited cultures of Chlamydomonas reinhardii and Isochrysis galbana. The model can be fully parameterized from relatively simple data sets and generates overshoots and time delays without exhibiting the dynamical instability characteristic of previously published time-lag formulations.

Aromatic onium salts are known compounds with well understood photoactivity. Their use as acid-generating species has, in the last few years, improved the performance of photoresists, particularly for microelectronics applications. There has, however, been little research dedicated to the use of these photoinitators in aqueousrdevelopable resists. This work investigates the feasibility of employing onium salts in the formulation of resists suitable for aqueous development. The ability of the onium salts to photoinitiate the cationic polymerization or cross-linking of epoxide residues was exploited using water-soluble or water-dispersible copolymers containing glycidyl methacrylate (GMA). The comonomers included N-vinyl pyrrolidinone (NVP), NN-dimethylacrylamide (DMAc) and 2-hydroxyethyI methacrylate (HEMA). Various copolymers were prepared by radical copolymerization to low conversion, typically in chloroform. Several onium salts were prepared by literature methods and their efficiency examined using a model copolymer. The photoacid generator (PAG) that performed best was then used in the examination of the aqueous systems.

The molecular mechanisms by which receptors regulate the Ras Binding Domains of the PIP3-generating, class I PI3Ks remain poorly understood, despite their importance in a range of biological settings, including tumorigenesis, activation of neutrophils by pro-inflammatory mediators, chemotaxis of Dictyostelium and cell growth in Drosophila. We provide evidence that G protein-coupled receptors (GPCRs) can stimulate PLCb2/b3 and diacylglycerol- dependent activation of the RasGEF, RasGRP4 in neutrophils. The genetic loss of RasGRP4 phenocopies knock-in of a Ras-insensitive version of PI3Kc in its effects on PI3Kc-dependent PIP3 accumulation, PKB activation, chemokinesis and reactive oxygen species (ROS) formation. These results establish a new mechanism by which GPCRs can stimulate Ras, and the broadly important principle that PLCs can control activation of class I PI3Ks.
