In the chicken three types of T-cell receptors can be defined by monoclonal antibodies TCRI, TCR2 and TCR3. which recognize γδ T cells, and Vβl- and Vβ2-expressing αβ T cells, respectively. In the present report we have analysed means of selectively depleting the γδ T cells and the V β1+ αβ T cells. γδ T cells, which represent up to 66% of ail T cells in blood of a 6-month-old chicken, can be effectively depleted by neonatal thymectomy (Tx) to levels as low as 1 %. Immunohistology demonstrates a similar depletion in lymphoid organs while intestinal epithelium-associated γδ T cells are affected by Tx to a lesser extent. Vβ1 -bearing αβ T cells, which comprise about 80% of the αβ T cells, were depleted by embryonic and neonatal injection of the TCR2 antibody. In the thymus such treatment depleted only the V β1+ αβ T cells with high density expression of T-cell receptor. Therefore, we thymectomized TCR2-treated animals in order to prevent development of mature V β1+ αβ T cells from the low density immature thymocyte's. Treatment of chickens with a total of 22 mg of TCR2 antibody plus Tx reduced V β1+ αβ T cells from an average of 65% to 10% of all T cells. The pathogenic effect of enterotoxogenic E. coli is mainly to be found in the jejunum. Hence, an effective immunoprophylaxis should be focussed at this site. This can be done effectively by an oral application of specific antibodies. The present study describes the effectiveness of specific antibodies against E. coli. The antibodies were gained from the yolk of E. coli immunized hens and tested for their in vitro activity. The tests showed a significantly reduced adhesion of enterotoxogenic E. coli onto isolated pig enterocytes as long as the bacteria had been incubated with the specific yolk antibodies beforehand. For this reason, yolk immunoglobulins from immunized hens prove to be an effective and handy source of specific antibodies which offers good protection against adhesion of E. coli onto pig enterocytes. A field study and a controlled infection trial showed the protective effect of egg yolk lyophilisate and whole egg lyophilisate against enterotoxic E. coli germs. The lyophilisates were gained from eggs of hens immunized against pilus antigen of porcin-enterotoxic E. coli. In a first field study using egg yolk antibodies, 92 % of 299 diarrhea affected piglets were cured. In a further field study diarrhea affected piglets were cured after 3 days by application of egg yolk lyophilisate from immunized hens. Piglets treated only with egg yolk of not immunized hens showed no signs of recovery. The infection trial showed, that whole egg lyophilisate of immunized hens was as successful as a common antibiotic therapy in curing piglets, orally infected with 5 times 1010 E. coli / feeding and animal. The present data show that chicken egg antibodies can be used for treatment of infectious diarrheal diseases in young animals. Functions of porcine polymorphonuclear neutrophils are evaluated with in vitro test systems. Results are compared with those from human PMN and the relevance for in vivo conditions is discussed. Ethanol was inhibitory to all porcine PMN functions investigated here. Influex, a combination product, containing extracts of Echinacea, Aconitum, Apis and Lachesis stimulated adherence, chemotaxis, and phagocytosis, but inhibited chemiluminescence. These results suggest an effect of the product in the generation of reactive oxygen species. The complement-dependence of polymorphonuclear yeast cell phagocytosis of sheep, goat, cattle, horse, dog, pig and man is determined by comparing the opsonizing abilities of untreated sera with complement-inactivated autologous sera at a serum concentration of 2.5 % and at different phagocytosis periods. The performed phagocytosis assays only detects incorporated particles and allows for the differentiation of granulocytes with different quantities of phagocytosed particles. In sheep, horse and man the addition of heat-inactivated serum reduces the phagocytosis index to less than −80% of the value obtained at untreated serum addition at a phagocytosis period of 60 min. The other species show a smaller reduction ranging from −66.9% (dog) to −41.4% (cattle). The evaluation of the distribution pattern of granulocytes incorporating a certain number of yeasts offers significant differences in the investigated species. Zusammenfassung Komplementabhängigkeit der Hefezell-Phagozytose polymorphnukleärer Leukozyten: Vergleichende Untersuchungen am Menschen und an verschiedenen Haustieren Die Komplementabhängigkeit der Hefezell-Phagozytose durch polymorphnukleäre Leukozyten von Schaf, Ziege, Rind, Pferd, Hund, Schwein und vom Menschen wird durch den Vergleich der opsonisierenden Fähigkeiten unbehandelten Serums mit Komplement-inaktiviertem autologen Serum bestimmt - bei einer Serumkonzentration von 2,5 % und unterschiedlich langen Inkubationszeiten. Der vorgestellte Phagozytose-Test erfaßt nur inkorporierte Partikel und läßt die sichere Differenzierung von Granulozyten mit unterschiedlicher Anzahl phagozytierter Partikel zu. Beim Schaf, Pferd und beim Menschen verringert der Zusatz hitzeinaktivierten Serums den Phagozytose-Index auf weniger als −80% des Kontrollwertes mit unbehandeltem Serum (Inkubationszeit: 60 Minuten). Die anderen Spezies zeigen eine weniger starke Herabsetzung von −66,9 % (Hund) bis −41,4% (Rind). Large quantities of specific egg antibodies can be produced with little effort by immunization of laying hens. Several antibody containing egg preparations were subjected to a tenacity test against digestive activity taking piglets with artificial jejunum fistulas as a model. Even with high doses of orally administered yolk lyophilisate no antibody activity could be found in the distal jejunum. The additional feeding of egg white, however, showed a significant protective effect on the yolks antibodies during the digestive act. No difference between egg white of immunized hens and egg white of unimmunized hens could be detected. Buffering the acidic gastric environment with sodium bicarbonate increased the antibody activity in the intestine by an average of 40 %. Eggs administered in hot (70 °C) beef stock had higher antibody titers than eggs that were boiled for 4 minutes. Tenacity in antibodies fed in pellets was significantly lower than in powdered feed. In vivo-Tenazitätstest bei Ferkeln mit künstlicher Jejunalfistel Durch Immunisierung von Legehennen lassen sich kostengünstig große Mengen von spezifischen Eiantikörpern produzieren. Am Modell des Ferkels mit Dünndarmfistel wurden verschiedene antikörperhaltige Eizubereitungen auf ihre Tenazität gegenüber Verdauungsvorgängen hin überprüft. Selbst bei hohen Dosen von oral verabreichtem Dotterlyophilisat konnte im distalen Jejunum keine Antikörperaktivität festgestellt werden. Zufütterung von Eiklar dagegen zeigte eine erhebliche Schutz-wirkung auf die Dotterantikörper während des Verdauungsvorganges, unabhängig davon, ob das Eiklar von immunisierten oder nicht immunisierten Hühnern stammte. Eine Pufferung des sauren Magenmilieus durch Natriumbicarbonat bewirkte eine durchschnittliche 40%ige Erhöhung der Antikörperaktivität im Darminhalt. Eier in 70 °C heißer Bouillon verabreicht wiesen höhere Antikörpertiter auf als 4 Minuten gekochte Eier. The present study describes the application of egg yolk antibodies in immunohistochemistry. In order to obtain specific antibodies against canine distemper virus (CDV), chickens were immunized with attenuated virus. Distinct antibody titres in serum and yolk could be detected by means of a modified plaque / focus immunoassay (ELISA) two weeks after a second immunization. The lower concentrations in corresponding yolk globulin preparations are attributed to the loss of antibodies caused by the isolation procedure (dextrane and ammonium sulfate precipitation). After verification of the antibody specificity by indirect immunofluorescence technique high titred globulin fractions were employed in immunohistochemistry using the Avidin-Biotin-Complex method. A specific and distinct immunostaining in formalin fixed and paraffin embedded brain sections of CDV-infected dogs was obtained. The advantages of egg yolk antibodies for immunological purposes are discussed in detail. Um spezifische Antikörper gegen das Hundestaupevirus (CDV) zu gewinnen, wurden Hühner mit attenuiertem Virus immunisiert. Mit Hilfe eines modifizierten Plaque/Focus ELISA konnten bereits 14 Tage nach der zweiten Immunisierung deutliche Antikörpertiter in Serum und Dotter festgestellt werden. Die geringeren Konzentrationen in den entsprechenden Dotterglobulinpräparationen können auf Antikörperverluste, bedingt durch das Isolierungsverfahren (Dextransulfat- und Ammoniumsulfatfällung), zurückgeführt werden. Nach der Überprüfung der Antikörperspezifität mittels indirekter Immunfluoreszenztechnik wurden die hochtitrigen Globulinfraktionen in der Immunhistochemie unter Verwendung der Avidin-Biotin-Complex-Methode eingesetzt. In formalin-fixierten und paraffineingebetteten Hirnschnitten staupeinfizierter Hunde konnte Virusantigen spezifisch markiert werden. Influence of a medicinal complex drug (Influex) and Echinacea angustifolia extract on avian humoral immune reactions Medicinal complex drugs as well as single ethanolic or aqueous extracts of several plants are commonly used to increase the natural resistance to various infections, though their efficacy and mechanism of action are not yet well elucidated. In the present study, we investigated two problems: firstly, whether the complex drug (Influex) and Echinacea angustifolia extract do stimulate the immunoglobulin and antibody synthesis in chickens immunized with human serum albumin; and secondly, whether a restoration of IgG-synthesis in immunodefective (dysgammaglobulinemic) UM-B 19 chickens is possible with these plant preparations, i. e. if the BG cells which may possibly be present can be polyclonally or antigen specifically stimulated. The preparations were administered orally in two doses, after which the complete immunoglobulin concentration was determined by rocket immunoelectrophoresis and the antibody production by ELISA. The effect of ethanolic solvent was taken into account. The administration of the complex drug to normal Leghorn chickens induced a rise in the serum immunoglobulin concentration, as well as an increase in the three classes of antibody. By the immunodeficient chickens (IgG concentration was below the level of test sensitivity at the start), the administration of the drug led to a slight production in IgG and antibody. Zusammenfassung Zur Unterstützung der körpereigenen Immunabwehr werden in der Medizin verschiedene pflanzliche Kombinationspräparate in alkoholischer oder in wäßriger Lösung verabreicht, obwohl ihre Wirksamkeit und die Wirkungsmechanismen noch nicht experimentell untersucht wurden. In der vorliegenden Arbeit wurde untersucht, ob erstens das Kombinationspräparat Influex und Echinacea angustifolia Extrakt die Immunglobulin- und Antikörpersynthese beim mit Humanserum-albumin immunisierten Huhn stimulieren können, und zweitens, ob es möglich ist, bei immundefizienten (dysgammaglobulinämischen) UM-B19 Hühnern eine Restaurierung der Immunglobulin-Synthese zu bewirken, d. h. ob die noch vorhandenen BG-Zellen polyklonal oder antigenspezifisch stimuliert werden können. Die Präparate wurden oral in zwei Dosierungen verabreicht. Die Immunglobulinkonzentration wurde mit der Rocket-Immunelektrophorese, die Antikörper-Produktion mit dem ELISA-Test bestimmt. Der Einfluß durch den Alkohol als Lösungsmittel wurde berücksichtigt. Die Verabreichung des Kombinationspräparates an normale Leghorn-Hühner induzierte einen Anstieg in der Serum-Immunglobulinkonzentration, ebenso einen Anstieg aller drei Anti-HSA-Antikörperklassen. A simple two step procedure for the isolation of caprine, ovine, bovine, equine, canine, porcine and human peripheral blood granulocytes is described. After enrichment of granulocytes by centrifugation, contaminating erythrocytes are lysed hypotonically. Recovery, purity, and viability of the granulocyte suspensions are determined. FACScan analysis of the cell suspensions measuring cellular size by forward and sideward light scatter is compared with the corresponding analysis of whole blood leukocytes. Constituencies of the isolated cell suspensions and loss of granulocyte subpopulations through isolation procedure is discussed with regard to granulocyte function assays. Nach Anreicherung der Granulozyten durch Zentrifugation werden die Erythrozyten durch hypotone Lyse zerstört. Ausbeute, Reinheit und Vitalität der gewonnenen Granulozytensuspensionen werden bestimmt. In FACScan-Analysen werden die Vorwärts- und Seitwärts-Lichtstreuung der isolierten Zellen mit der von Vollblutleukozyten verglichen. Specific antibodies directed against enterotoxic E. coli strains from the yolk of immunized hens were exposed in vitro to the influence of varying digestive phenomena like reduction of pH and proteolytic digestion. The remaining antibody activity was tested in a specific ELISA system. It could be shown that already within the stomach a considerable loss in antibody activity caused by a lowering of pH and peptic cleavage can be expected. A further loss in antibody activity is due to the proteolytic effect of the pancreas proteases trypsin and chymotrypsin. It was found that antibodies in protein rich solutions like egg or yolk suspensions were more resistant than mere globulin fractions or antibodies isolated by affinity chromatography. In vitro-Studien über die Verdauung von Dotter-Antikörpern, spezifisch für Escherichia coli-Stämme, im Magen-Darmtrakt Spezifische gegen enterotoxische E. coli-Stämme gerichtete Antikörper aus dem Eidotter immunisierter Hennen wurden in vitro dem Einfluß unterschiedlicher Verdauungsphänomene wie erhöhter (H+)-Konzentration und proteolytischer Digestion ausgesetzt. Die verbleibende Antikörperaktivität wurde in einem spezifischen ELISA bestimmt. Es zeigte sich, daß bereits im Magen ein bedeutender Verlust der Antikörperaktivität durch hohe (H+)-Konzentrationen und peptische Spaltung zu erwarten ist. Ein weiterer Verlust der Antikörperaktivität ist auf die proteolytische Wirkung der Pankreasenzyme Trypsin und Chymotrypsin zurückzuführen. Es stellte sich heraus, daß Antikörper in proteinreichen Lösungen wie Vollei- oder Dottersuspension widerstandsfähiger waren als reine Globulinfraktionen oder affinitätschromatographisch isolierte Antikörper. A light- and a fluorescence-microscopic method for quantitative assessment of yeast cell incorporation in phagocytes were developed. The light-microscopic method offers methylene blue prestained yeast cells as phagocytosis particles and counterstains nonincorporated yeasts with eosine. The fluorescence-microscopic method works by acridine orange staining of phagocytosis assays. Fluorescence of nonincorporated yeast cells is suppressed by addition of methylene blue. Different ways of evaluating the results of microscopic quantitation of phagocytosis are discussed. Die lichtmikroskopische Methode bietet mit Methylenblau vorgefärbte Hefe als Phagozytosepartikel an und färbt nichtinkorporierte Hefe mit Eosinrot. Bei der fluoreszenzmikroskopischen Methode wird den Phagozytoseansätzen Akridin-orange zugegeben, wobei die Fluoreszenz nichtinkorporierter Hefe durch Methylenblau unterdrückt wird. Antibody response of dysgammaglobulinemic chickens and of immunologically intact controls (LSL) after immunization with haptens (DNP, MATP) coupled to carrier proteins was investigated. Only one group of dysgammaglobulinemic chickens showed an IgG-antibody response comparable to that of control chickens. Carrier preimmunization caused a strong antibody response of all the chickens. The mitogenic LPS effect resulted in increased serum IgG-level of the dysgammaglobulinemic chickens. BG-cell specific tolerance as a consequence of a retarded bursal development is discussed as a possible reason of the dysgammaglobulinemia. Der mitogene Effekt von LPS führte zu einer Erhöhung der Serum-IgG-Konzentration der dysgammaglobulinämischen Hühner. In order to obtain high amounts of specific antibodies against porcine enteropathogenic E. coli strains laying hens were immunized with a vaccine for pregnant sows. The course of antibody concentrations in serum and egg yolk was detected by a specific ELISA system. Advantages of egg yolk antibodies and potential therapeutic applications in human and veterinary medicine are discussed. The different molecular forms of serum IgM and IgA of Leghorn, normo- and dysgamma-globulinaemic UM-B19 chickens were separated by gel filtration and quantified by rocket immunoelectrophoresis, separately for male and female animals. In all three chicken groups the IgM concentrations increased from the second to the sixth month of life. However, the absolute concentrations of monomeric IgM in normogammaglobulinaemic and Leghorn chickens were within the same range for all three age groups, while for the monomer, as a percentage of total IgM, declined from the second to the fourth month of life by approximately 9% to reach 24% and remained roughly constant thereafter. Dysgammaglobulinaemic chickens showed three to five times higher total IgM concentrations as well as two to four times higher monomeric IgM serum concentrations. In two-month-old dysgammaglobulinaemic chickens the monomer percentage was only 25% and fell to 18% by the sixth month. For all chickens investigated the IgA concentrations in serum decreased from the second to the sixth month of life while the absolute concentrations of monomeric IgA remained constant between the fourth and sixth month. Comparing the percentage of monomeric IgA with total IgA, in contrast to the monomeric IgM, a distinct percentage increase of monomeric IgA due to a decrease of polymeric IgA was found in the normal UM-B19 and in the Leghorn chickens. In contrast to the individual distributions of monomeric and polymeric IgM and IgA, sex differences were found for the total IgM concentrations with progressing age. In six-month-old female fowls the IgM levels were significantly higher than in their male counterparts. Dysgamma-globulinaemic roosters showed slightly higher serum IgM concentrations than their female counterparts. Männliche und weibliche Tiere wurden getrennt ausgewertet. Die IgM-Konzentrationen stiegen bei allen drei untersuchten Tiergruppen vom zweiten bis zum sechsten Lebensmonat an. Die absoluten Konzentrationen von monomerem IgM lagen hingegen bei den Leghorn- und normogammaglobulinämischen Hühnern in den drei Altersgruppen im gleichen Bereich, während der prozentuale Anteil der Monomere am Gesamt-IgM vom zweiten zum vierten Lebensmonat um etwa 9% auf 24% absank und dann annähernd konstant blieb. Dysgammaglobulinämische Hühner wiesen je nach Alter nicht nur 2- bis 4fach höhere IgM-Gesamtserumkonzentrationen, sondern auch 2- bis 4fach höhere monomere IgM-Serumspiegel auf. Bei den dysgammaglobulinämischen Tieren wiesen die Monomere bereits im Alter von zwei Monaten lediglich einen Anteil von 25% auf und fielen bis zum sechsten Lebensmonat auf 18% ab. Bei allen untersuchten Hühnern fielen die Serum-IgA-Konzentrationen vom zweiten bis zum sechsten Lebensmonat, während die Gesamtserumkonzentrationen vom monomeren IgA zwischen dem vierten und sechsten Monat konstant blieben. Wird der Monomerenanteil des IgA prozentual zur IgA-Gesamtkonzentration dargestellt, so findet man, im Gegensatz zum monomeren IgM, bei Leghorn und normogammaglobulinämischen Hühnern einen prozentual deutlichen Anstieg von monomerem IgA bei einer gleichzeitigen Abnahme von polymerem IgA. Im Gegensatz zu der individuellen Streuung von monomerem und polymerem IgM und IgA zeichnen sich Geschlechtsunterschiede mit zunehmendem Alter für IgM ab. Sechs Monate alte weibliche Hühner zeigen gegenüber den männlichen Tieren signifikant höhere IgM-Serumkonzentrationen. Antibody titers achieved by the immunization of hens are presented with examples. The transfer of immunoglobulins from the blood or oviduct to the egg and the distribution of these proteins in the various compartments which develop in the egg during incubation are quantitatively recorded. Possible procedures for extracting IgG antibody from the yolk are pointed out. Results concerning acid and temperature resistance of yolk antibodies are presented. An overview of the literature concerning with diagnostic use of yolk antibodies is given. The possible therapeutic application of egg antibodies is discussed. Die Möglichkeiten der IgG-Antikörper-Extraktion aus dem Dotter werden aufgezeigt. Ergebnisse zur Säure- und Temperatur-Resistenz der Dotterantikörper werden vorgelegt. Treatment of chickens as embryos and in the first six days after hatching with mg amounts of anti-TCR2 (αβ T cell antigen receptor) antibody plus thymectomy 6 to 8 days after hatching suppressed by more than 90% the TCR2 and TCR1 (TCRγδ) subpopulations in the blood. Thymectomy alone resulted only in a profound reduction (> 90%) of the absolute number of TCR1 cells, while the absolute number of TCR2 cells was reduced only by 65%. We were interested in the regulation of the immunoglobulin synthesis in these T cell deficient chickens. We determined serum IgG, IgM and IgA levels and found a profound selective reduction (by about 90% on average) of the serum IgA level in anti-TCR2 antibody treated plus thymectomized chickens. In chickens which had been thymectomized only, the serum IgA level was reduced on average only by about 60%. Serum IgG and IgM levels were not reduced in any group. Thus, there was a positive correlation of the serum IgA level and the absolute number of TCR2 cells in all groups. These results indicate that T cells which play a role in IgA specific isotype switching or in IgA B cell differentiation belong to the TCR2 subpopulation. [ABSTRACT FROM AUTHOR]

In order to obtain high amounts of specific antibodies against the hapten methylphosphoric acid, para-aminophenyl-1,2,2-trimethyl-propyldiester (MATP) different animals were immunized with MATP coupled to the carrier protein human serum albumin (MATP12-HSA) using several adjuvants. The best specific immune response in sheep, rabbit and chicken was reached with Freund's complete adjuvant with animal specific differences being tested by an ELISA. The adjuvants aluminium hydroxide (5% and 10%) and diphosphoryl lipid A showed no significant difference compared to the control group (NaCl with MATP12-HSA). In rabbits and chickens MATP12-HSA can be used to reach an immune response without the help of an adjuvant. [ABSTRACT FROM AUTHOR]

Whereas the yolk of freshly-laid eggs contains only IgG, apart from traces of IgA, we were able to measure, on average, 0.145 mg/ml IgM and 0.207 mg/ml IgA in the yolk sac contents of 21-day-old chicken embryos. Up to the 14th embryonic day, IgG is exclusively contained in the yolk sac contents. It was not possible to demonstrate an increase in the amount of immunoglobulins present by comparing the amounts of IgM and IgA in freshly-laid eggs and in the yolk sac contents of 21-day-old embryos. The offspring of hens with experimentally-induced agammaglobulinemia did not begin with the production of the immunoglobulin isotypes IgG, IgM and IgA until after hatching. From these results the conclusion can be drawn that the immunoglobulins IgM and IgA are transferred from the egg white to the yolk sac contents during the last third of embryonic development. Embryonic synthesis of these immunoglobins can be discounted as a result of this study. Synthesis was found to be initiated between the 2nd and 7th day of life for IgG, between the 2nd and 4th day of life for IgM and between the 6th and 13th day of life for IgA. [ABSTRACT FROM AUTHOR]
