Arginine rich, mutated in early stage of tumours (Armet), is a well-characterized bifunctional protein as an unfolded protein response component intracellularly and a neurotrophic factor extracellularly in mammals. Recently, a new role of Armet as an effector protein mediating insect-plant interactions has been reported; however, its molecular mechanisms underlying the regulation of plant defences remain unclear. We investigated the molecular mechanisms underlying whitefly-secreted Armet-mediated regulation of insect-plant interaction by agrobacterium-mediated transient expression, RNA interference, electrical penetration graph, protein-protein interaction studies, virus-induced gene silencing assay, phytohormone analysis and whitefly bioassays. Armet, secreted by Bemisia tabaci whitefly, is highly expressed in the primary salivary gland and is delivered into tobacco plants during feeding. Overexpression of the BtArmet gene in tobacco enhanced whitefly performance, while silencing the BtArmet gene in whitefly interrupted whitefly feeding and suppressed whitefly performance on tobacco plants. BtArmet was shown to interact with NtCYS6, a cystatin protein essential for tobacco anti-whitefly resistance, and counteract the negative effects of NtCYS6 on whitefly. These results indicate that BtArmet is a salivary effector and acts to promote whitefly performance on tobacco plants through binding to the tobacco cystatin NtCYS6. Our findings provide novel insight into whitefly-plant interactions.

Aims: Embedded system plays a vital role in today’s life. Hence, our interest is in areadelay- energy efficient embedded system design in post-CMOS technology, i.e., QCA. Objectives: The research is focused on efficient area-delay-energy Configurable Logic Block (CLB) design for Field-Programmable Gate Array architecture (FPGA) with successful simulation-based on next-generation technology, Quantum-dot cellular automata. Methods: Each proposed circuit is designed on post CMOS 4 dot 2 electron technology, i.e. QCA (Quantum dot Cellular Automata), is adopted in circuit implementation due to low power dissipation, high clock frequency and high package density. QCADesigner is used to verify the functionality of every circuit. QCAPro tool is used for determining power dissipation. Results: In contrast, a new approach of using de-multiplexer replacing the decoder has been introduced that results in the reduction of the average energy dissipation by almost 57%. A NOR based D flip-flop memory architecture and multiplexer are also used in the lookup table for the configurable logic block. The proposed architecture thus reduces the overall latency. The proposed CLB consists of 6356 QCA cells covering 7.44 um2 area. Write and read latency of proposed CLB are 12 and 7.25 QCA clock, respectively. Conclusion: The present paper concludes that read and write latency reduction occurs; average energy dissipation, leakage, and switching energy dissipation are reduced in a large amount resulting in an advantage of the overall minimization of the latency for the proposed CLB in the process.

Polychlorinated dibenzo-p-furans and dioxins (PCDD/Fs) that pose a great threat to human health are commonly found during the incineration of municipal solid waste. In this study, industrial urea was injected into a commercial MSW incinerator flue gas to evaluate the suppression performances of PCDD/Fs, and the possible inhibition mechanisms were proposed. The results show that the use of urea dramatically reduced the PCDD/Fs emission concentration from 8.87 to 0.63 ng/Nm3, along with a significant decrease in the I-TEQ value (0.26 → 0.047 ng I-TEQ/Nm3), below the Chinese national standard of 0.1 ng I-TEQ/Nm3. The emission reduction cost of the industrial urea was over 41 % lower than that of using activated carbon. Urea molecule poisoned the metal ions, thus seriously inhibiting the de novo synthesis of PCDD/Fs. Furthermore, the decomposition products of urea reduced the concentration of HCl in the flue gas, thereby reducing the formation of Cl2 and hindering the chlorination reaction, which was identified from the reduction of chlorination degree and lower distribution of high-chlorinated PCDD/Fs. This research provided some practice basis and experience to reduce the emission of PCDD/Fs from municipal solid waste incineration for future commercial promotion and application.

Resistance in iron ore undergoes a sharp change of up to several orders of magnitude when the sintered solid phase changes to liquid phase. In view of the insufficiency of existing assimilation detection methods, a timing-of-assimilation reaction is proposed, which was judged by continuously detecting the changes in resistance at the reaction interface. Effects of pole position and additional amounts of iron ore on assimilation reaction timing were investigated. The results showed that the suitable depth of pole groove was about 2 mm, and there was no obvious impact when the distance of the poles changed from 4 to 6 mm, or the amount of iron ore changed from 0.4 to 0.6 g. The temperature of sudden change of resistance in the temperature-resistant image was considered to be the lowest assimilation temperature of iron ore. The accuracy of this resistance method was clarified by X-ray diffraction, optical microscope, and scanning electron microscope/energy dispersive spectrometer (SEM/EDS) analyses.

PCDD/Fs are typical toxic persistent aromatic compounds that greatly reduce air quality and harm human health. In this study, urea’s suppressive effect on PCDD/Fs and their congener emissions was investigated via sintering pot tests, and the inhibition mechanisms were studied. The results showed that the I-TEQ values for the total PCDD/Fs decreased from 0.50 to 0.20, 0.12 and 0.20 ng I-TEQ Nm–3 after adding solid 0.02, 0.035 and 0.05 wt.% urea particles, respectively, to the iron ore sintering mixture, but these values increased for the low-chlorinated TeCDFs (from 0.003 to 0.009, 0.006 and 0.006 ng I-TEQ Nm–3) and TeCDDs (from 0.014 to 0.020, 0.018 and 0.020 ng I-TEQ Nm–3). Moreover, the average I-TEQ values for the chlorine substituents in the PCDFs and PCDDs decreased, indicating that urea inhibited chlorination or enhanced dechlorination. The potential mechanisms by which urea suppresses the total PCDD/Fs and hydrodechlorinates the high-chlorinated PCDD/Fs are discussed.

Alkaline phosphatases (ALPs: EC 3.1.3.1) are ubiquitous enzymes and play crucial roles in the fundamental phosphate uptake and secretory processes. Although insects are regarded as the most diverse group of organisms, the current understanding of ALP roles in insects is limited. As one type of destructive agricultural pest, whitefly Bemisia tabaci, a phloem feeder and invasive species, can cause extensive crop damage through feeding and transmitting plant diseases. In this study, we retrieved five ALP genes in MEAM1 whitefly, nine ALP genes in MED whitefly via comparative genomics approaches. Compared with nine other insects, whiteflies’ ALP gene family members did not undergo significant expansion during insect evolution, and whiteflies’ ALP genes were dispersed. Moreover, whiteflies’ ALP gene family was conserved among insects and emerged before speciation via phylogenetic analysis. Whiteflies’ ALP gene expression profiles presented that most ALP genes have different expression patterns after feeding on cotton or tobacco plants. Female/male MED whiteflies possessed higher ALP activities on both cotton and tobacco plants irrespective of sex, relative to MEAM1 whiteflies. Meanwhile, adult MED whiteflies possessed higher ALP activity in both whole insect and salivary samples, relative to MEAM1 whiteflies. We also found that both MED and MEAM1 whiteflies could upregulate ALP activities after feeding on cotton compared with feeding on tobacco plants. These findings demonstrated the functions of whiteflies ALPs and will assist the further study of the genomic evolution of insect ALPs.

The high-temperature characteristics of iron ores play important roles in optimizing ore proportion of sintering, which are tested by using iron-ore fines and analytical reagent CaO as raw materials. Two calculation methods of CaO addition amount based on binary basicity (basicity method) and n(Fe2O3)/n(CaO) (mole ratio method), respectively, were employed to evaluate the liquid phase fluidity (LPF) and the capability of calcium ferrite formation (CCFF) of iron ores. The results show that the rule of LPF of iron ores under the mole ratio method is different from that with basicity method. The LPF measured by basicity method has a linear positive correlation with the SiO2 content, and there is no linear relationship between LPF and Al2O3 content or mass loss on ignition, which are inconsistent with the results of the previous study. And the results of CCFF with low SiO2 content ( 7 wt.%) based on basicity method cannot reflect the true CCFF. The mole ratio method could successfully solve this problem by reducing the effect of CaO addition amount changes caused by SiO2 content of iron ores.

Phloem-feeding insects feed on plant phloem using their stylets. While ingesting phloem sap, these insects secrete saliva to circumvent plant defenses. Previous studies have shown that, to facilitate their feeding, many phloem-feeding insects can elicit the salicylic acid- (SA-) signaling pathway and thus suppress effective jasmonic acid defenses. However, the molecular basis for the regulation of the plant's defense by phloem-feeding insects remains largely unknown. Here, we show that Bt56, a whitefly-secreted low molecular weight salivary protein, is highly expressed in the whitefly primary salivary gland and is delivered into host plants during feeding. Overexpression of the Bt56 gene in planta promotes susceptibility of tobacco to the whitefly and elicits the SA-signaling pathway. In contrast, silencing the whitefly Bt56 gene significantly decreases whitefly performance on host plants and interrupts whitefly phloem feeding with whiteflies losing the ability to activate the SA pathway. Protein-protein interaction assays show that the Bt56 protein directly interacts with a tobacco KNOTTED 1-like homeobox transcription factor that decreases whitefly performance and suppresses whitefly-induced SA accumulation. The Bt56 orthologous genes are highly conserved but differentially expressed in different species of whiteflies. In conclusion, Bt56 is a key salivary effector that promotes whitefly performance by eliciting salicylic acid-signaling pathway.

Iron ore sintering process is an important sector for iron and steel industry as well as a major pollution emission source of PCDD/Fs. The PCDD/Fs emission of sintering process has not been properly controlled because the flue gas presents the following characteristics, including large amount, remarkable flow fluctuations and lower concentration. The generation mechanism of PCDD/Fs in the iron ore sintering was discussed systematically and the new developments and technologies of PCDD/Fs emission reduction were also summarized from the source, process and the end treatments. Commonly, the PCDD/Fs formed in drying and preheating zone is transferred to the lower part of the material layer which is a chemical transfer process in the iron ore sintering. Finally, the potential future development of PCDD/Fs emission reduction in the iron ore sintering was also pointed out.

Iron ore sintering is the main SO2 emission process in iron and steel industry. The flue gas of iron ore sintering has been cleaned by desulfurization equipment, which causes high capital and operation cost. A novel technology of reducing SO2 emission of flue gas during iron ore sintering process by adding urea into granules bed was investigated in this paper. The results show that the emission concentration of SO2 in flue gas decreased significantly by adding urea loaded in the bed. The desulfurization rate higher than 85% was obtained when 0.05% urea was added in the bed at the height of 70–150 mm from the bed bottom, and there is no adverse influence on sintering indexes and final sinter quality.

Background Plant viruses in agricultural crops are of great concern worldwide, and over 75% of them are transmitted from infected to healthy plants by insect vectors. Tomato yellow leaf curl virus (TYLCV) is a begomovirus, which is the largest and most economically important group of plant viruses, transmitted by the whitefly Bemisia tabaci. The circulation of TYLCV in the insect involves complex insect-virus interactions, whereas the molecular mechanisms of these interactions remain ambiguous. The insect gut as a barrier for viral entry and dissemination is thought to regulate the vector specificity. However, due to its tiny size, information for the responses of whitefly gut to virus infection is limited. Methods We investigated the transcriptional response of the gut of B. tabaci Middle East-Asia Minor 1 species to TYLCV infection using Illumina sequencing. Results A total of 5207 differentially expressed genes (DEGs) between viruliferous and non-viruliferous whitefly guts were identified. Enrichment analyses showed that cargo receptor and ATP-binding cassette (ABC) transporters were enriched in DEGs, and might help the virus to cross gut barrier. TYLCV could perturb cell cycle and DNA repair as a possible result of its replication in the whitefly. Our data also demonstrated that TYLCV can activate whitefly defense responses, such as antimicrobial peptides. Meanwhile, a number of genes involved in intracellular signaling were activated by TYLCV infection. Conclusions Our results reveal the complex insect-virus relationship in whitefly gut and provide substantial molecular information for the role of insect midguts in virus transmission. To solve the problem that the sintered liquid phase fluidity characteristic of low silica (w(SiO2) 7 wt%)) iron ores were different from actual sintering production with using the common method based on w(CaO)/w(SiO2), a new improved method based on n(Fe2O3)/n(CaO) was proposed. The differences between two methods for iron ore fluidity detection were studied. The results show that when the common method is used to detect the liquid phase fluidity, as the content of SiO2 increased from 2.12 to 11.68 wt%, the amount of CaO addition augmented from 7.88 to 32.65 wt% and the corresponding fluidity index changed from 0 to 5.68. The fluidity index has a linearly positive correlation with the content of SiO2. The improved method could significantly reduce the effect of the change of CaO addition on the fluidity characteristic of iron ore which caused by the difference of SiO2.

International audience; GMZ bentonite has been considered as a possible material for engineered barrier in the Chinese program of nuclear waste disposal at great depth. In the present work, the hydraulic conductivity of this bentonite was determined by simultaneous profile method. A specific infiltration cell equipped with five resistive relative humidity probes was designed for this purpose. The water retention properties were studied under both confined and unconfined conditions; the results shows that at high suctions (> 4 MPa) the water retention capacity is independent of the confining condition, and by contrast, at low suctions (< 4MPa) the confined condition resulted in significant low water retention. Furthermore, the microstructure was investigated at Mercury Intrusion Porosimetry (MIP) and Environmental Scanning Electron Microscope (ESEM) in different states: on oven-dried powder, bentonite slurry, as-compacted and wetted samples. It has been observed that the soil powder is constituted of aggregates of various sizes; this aggregates are destroyed by fully saturation at a water content equal to the liquid limit; compaction at the initial water content of 11-12% and a dry density of 1.7 – 1.75 Mg/m3 led to a microstructure characterized by an dense assembly of relatively well preserved aggregates; saturation of the compacted sample under constant volume condition defined a non-homogeneous microstructure with the presence of well preserved aggregates. This non-homogeneous microstructure would be due to the non uniform distribution of the generated swelling pressure within the soil sample upon wetting. The hydraulic conductivity determined has been found decreasing firstly and then increasing with suction decrease from the initial value of about 80 MPa to zero; the decrease can be attributed to the large pore clogging due to soft gel creation by exfoliation process, as observed at ESEM.

In this paper, we introduce a problem of the optimization of approximate solutions of operator equations in the probabilistic case setting, and prove a general result which connects the relation between the optimal approximation order of operator equations with the asymptotic order of the probabilistic width. Moreover, using this result, we determine the exact orders on the optimal approximate solutions of multivariate Freldholm integral equations of the second kind with the kernels belonging to the multivariate Sobolev class with the mixed derivative in the probabilistic case setting.

Treatment with injectable testosterone undecanoate (TU) alone or in combination with oral levonorgestrel (LNG) resulted in marked decreases in sperm concentrations. In this study, we used proteomic analyses to examine the cellular/molecular events occurring in the human testis after TU or TU + LNG treatment. We conducted a global proteomic analysis of the human testicular biopsies before and at 2 weeks after TU alone or TU + LNG treatment. Proteins showing significant changes in expression were identified and analyzed. As a result, 17 and 46 protein spots were found with significant differential expression after the treatment with TU alone and TU + LNG, respectively. TU treatment changed the expression of heterogeneous nuclear ribonucleoprotein K (hnRNP K), proteasome inhibitor PI31 subunit (PSMF1), and superoxide dismutase [Mn] mitochondrial precursor (SOD2). These proteins inhibit "assembly", induce cell death, and promote compensatory "cell survival" in the testis. After TU + LNG treatment, "proliferation/cell survival" and "apoptosis/death" were the predominant responses in the testis. TU + LNG treatment inhibited the expression of Prolyl 4-hydroxylase beta subunit (P4HB) and Annexin A2 (Annexin II). These proteins are involved in apoptosis and cell proliferation, respectively. TU + LNG treatment also enhanced the expression of SOD2 and Parvalbumin alpha (Pvalb). These two proteins may protect testicular cells against apoptosis/death and promote cell survival. In conclusion, TU and TU + LNG treatments suppress spermatogenesis through different pathways by changing the expression of different proteins. hnRNP K, PSMF1, SOD2, P4HB, Annexin II, and Pvalb, are key proteins that may be early molecular targets responsible for spermatogenesis suppression induced by hormone treatment.

To study the etiopathogenesis, clinical manifestations, diagnosis and management of persistent Müllerian duct syndrome (PMDS).Two cases of PMDS were reported, one accompanied by transverse testicular ectopia and the other associated with cryptorchidism. Corporeal hysterectomy and orchidopexy were given to both the patients and cryptorchidectory the latter.Vascular supply and texture of the testis were normal in both the 2 patients after 1.5-2 years' follow-up.PMDS is male pseudohermaphroditism, for which means should be taken to preserve the blood supply and fertility function of the testis in surgical management, and attention should be paid to possible development of testis tumor in follow-up.

Context: In rodents and monkeys, a combination of hormonal and physical agents accelerates germ cell death. Objective: A “proof of concept” study was performed to investigate whether addition of heat exposure or a progestin to an androgen induces germ cell death and more complete and rapid spermatogenesis suppression. Design and Settings: A randomized clinical trial was performed at academic medical centers. Participants: We treated four groups of healthy male volunteers (18 per group) for 18 wk: 1) testosterone undecanoate (TU) 1000 mg im (first dose), followed by 500 mg im every 6 wk; 2) submersion of scrota at 43 C in water for 30 min/d for 6 consecutive days; 3) TU plus heat; and 4) TU plus oral levonorgestrel (LNG) 250 μg/d. Main Outcome Measures: Semen parameters, testicular histology, and germ cell apoptosis were the main outcome measures. Results: Heat alone and TU plus heat suppressed sperm counts more than TU alone by wk 6. By wk 9, recovery began in the heat only group, whereas spermatogenesis remained suppressed in the TU plus heat group. Oral LNG plus TU suppressed spermatogenesis earlier and more severely than TU alone. At wk 2, significantly greater germ cell apoptosis occurred in heat and heat plus TU subjects, but not in subjects without heat treatment, compared with pretreatment subjects. By 9 wk, markedly smaller seminiferous tubule diameters and fewer spermatocytes and spermatids were noted in all 12 biopsies from men receiving TU, TU plus LNG, with most dramatic differences for the TU plus heat group, whereas no differences from pretreatment biopsies were observed in men who received heat treatment only. Conclusions: Heat causes a rapid and transient suppression of spermatogenesis. TU plus heat resulted in low-sperm output that was maintained by continuous treatment with TU. Addition of an oral progestin accelerated spermatogenesis suppression by TU alone. Increased germ cell apoptosis contributed to suppression of spermatogenesis.

To investigate the association of XRCC1 Arg399Gln polymorphism, smoking and drinking with the risk of prostate cancer (PCa) in the population of Han nationality in Jiangsu and Anhui.A case-control study including 207 PCa patients and 235 age-matched controls was conducted. The polymorphisms of XRCC1 Arg399Gln sites were analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique using genomic DNA isolated from peripheral blood lymphocytes. We compared the correlations between the susceptibility to PCa and different genotypes, and investigated the effect of smoking and drinking.The heterozygous Arg/Gln genotype was associated with statistically significantly increased risk of PCa (OR = 1.55, 95% CI: 1.01-2.39) compared with those with Arg/Arg wild-type homozygote. An increased susceptibility to PCa was shown to be associated with the 399Gln allele (either the heterozygous Arg/Gln or the homozygous Gin/Gln genotypes, OR = 1.61, 95% CI: 1.07-2.44) , and heavy smokers (smoking index;or =20) (OR = 1.94, 95% CI: 1.02-3.71) and superficial smokers (taking smoke into the mouth only) (OR = 2.44, 95% CI: 1.02-5.80) with 399Gln allele demonstrated a significantly increased risk in comparison with those carrying wild genotype.XRCC1 Arg399Gln polymorphism might contribute to the susceptibility to PCa. The Arg/Gln and Gln/Gln genotypes might increase the risk of PCa and have synergistic effect with smoking.

To investigate the changes of antisperm antibodies (AsAb), sexual hormones, and inhibin B (INH B) in patients before and after testicular torsion, as well as the effects of these factors on testicular function and reproduction.Ten patients with single acute testicular torsion (left side 9 and right side 1), aged 16-45 years (19.6 on average), disease course of 3-6 days (averaging 4.7 days), underwent surgical removal of the damaged testis. Before and after the operation, serum AsAb (IgG, IgM, IgA) and INH B were measured by ELISA, and serum follicle-stimulating hormone (FSH), luteotropic hormone (LH), and testosterone (T) determined by chemoluminescence autoanalyzer.After the operation, the AsAb levels rose significantly and remained high for at least 26 weeks. The level of INH B was the lowest in the 3rd week and restored to normal in the 12th week, with significant difference between preoperation and the 3rd or the 6th week after the operation. The levels of LH and INH B in the 26th week were elevated significantly compared with the 6th.Testicular injury induced the elevation of AsAb, which would last a very long time. The change of INH B was closely related with the injury of the testis, which reflected the degree of testicular injury and functional restoration of the patients after the operation. Our study showed that AsAb and INH B can be used as useful tools for monitoring testicular function and reproduction.

To investigate the association among the polymorphisms of the cytochrome P450 1A1 and 2E1 genes, smoking, drinking and the risk of prostate cancer (PCa) in a Han nationality population in Southern China.A case-control study including 225 PCa patients and 250 age-matched controls was conducted. The six polymorphic sites of the CYP 1A1 and CYP2E1 genes were analysed by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) or allele-specific PCR technique using genomic DNA isolated from peripheral blood lymphocytes.We found that the CYP1A1 Val/Val genotype significantly increased the risk for PCa (OR, 2.26; 95% CI, 1.09-4.68). In contrast, the CYP2E1 C1/C2 (OR, 0.67; 95% CI, 0.46-0.99) or C2/C2 genotype (OR, 0.31; 95% CI, 0.10-1.00) significantly decreased the risk. Furthermore, the individuals carrying the CYP1A1 Val allele and the CYP2E1 C1/C1 genotype showed the highest risk (OR, 2.50; 95% CI, 1.45-4.29). Though there was no significant difference with smoking history (P = 0.237) or drinking habit (P = 0.499) between cases and controls, a deep smoking habit (OR, 2.02; 95% CI, 1.28-3.17) and heavy smoking history (OR, 1.61; 95% CI, 1.04-2.50) significantly increased the susceptibility of PCa after stratification by smoking method and accumulative smoking amount. Moreover, both the CYP1A1 Val allele (OR, 2.82; 95% CI, 1.49-5.35) and CYP2E1 C1/C1 genotype (OR, 2.57; 95% CI, 1.31-5.02) had obvious interaction with heavy smoking history that significantly raised the risk. We also discovered a significant interaction between the CYP2E1 C1/C1 genotype and drinking (OR, 1.85; 95% CI, 1.04-3.28).Individuals carrying the CYP1A1 Val allele or the CYP2E1 C1/C1 genotype with a smoking or drinking habit were at increased risk of PCa, which also showed a positive correlation with exposure dose of tobacco.

To evaluate the correlation of epididymal protease inhibitor(Eppin) and Semenogelin(Sg) on human ejaculated spermatozoa.The experimental approaches include: (1) Immunoprecipitation of Eppin with anti-Eppin from semen; (2) Colocalization of Eppin and Sg by immunofluorescence; (3) Immunoprecipitation of rEppin and rSg;(4) Far-Western blotting of rEppin and rSg;(5) Competition of saturated 125I-rSg binding to rEppin with unlabeled Sg, and direct binding of 125I-rSg to rEppin on a blot; (6) Autoradiography of 125I-rSg with rEppin.Eppin-Sg complex present on the surface of human ejaculated spermatozoa, Cys-239 is the only cystein for rEppin binding rSg. Reduction and carboxymethylation of Cys-239 blocks binding of 125I-rEppin to rSg.Our study demonstrates that Eppin and Sg bind to each other on human ejaculated spermatozoa. A disulfide linkage occurs between Sg and Eppin, indicating the specificity of binding.

To study the diagnosis and treatment of acute prostatitis.The data of 35 cases of acute prostatitis who were admitted from January 2001 to March 2004 were reviewed. The main clinical manifestations were chills, fever, frequency, urgency and dysuria. All patients were treated with antibiotics and supportive measures. Two patients underwent surgical drainage for prostate abscess. Three patients were indwelled catheter for acute urinary retention.All patients'temperatures returned to normal within 3 to 5 days. Blood and urine routine tests, urine culture and transurethral ultrasound examination results returned to normal 2 weeks later. Q maximal urinary flow rate improved in patients with dysuria.After diagnosis of acute prostatitis, full-dose of sensitive antibiotics should be given to all patients for some time as early as possible. At the same time, supportive therapy may be important to some patients. Surgical drainage should be used for patients with prostate abscess.

To investigate the clinical characteristics, diagnosis and therapeutic strategies of congenital absence of the vas deferens (CAVD).We summarized the clinical data of 81 cases of CAVD and investigated clinical features, diagnosis and management of the disease.Seventy-nine cases of infertility were diagnosed as CAVD at the clinic, and 2 were diagnosed during surgical exploration. The population consisted of 40 cases of congenital bilateral absence of the vas deferens (CBAVD), 25 cases of congenital unilateral absence of the vas deferens (CUAVD), and 16 cases of segmental agenesis of the vas deferens. Seventy-four spouses received the treatment of assisted reproductive techniques, including intracytoplasmic sperm injection (ICSI) in 12 cases, and 4 of them achieved pregnancy.CAVD frequently presents with infertility. CBAVD may manifest as obstructive azoospermia, and CUAVD and segmental vasal agenesis as oligospermia, asthenospermia, or obstructive azoospermia. CAVD is usually not difficult to diagnose, but may be missed due to careless examination. Assisted reproductive technology (ART) plays a key role in the management of CAVD.

This case-control study was aimed to detect the single nucleotide polymorphisms (SNPs) in JWA promoter region, to assess the effect of SNP on transcriptional activity, and to probe the relationship between SNP and the risk of bladder cancer.The design of one control per case was adopted. The JWA gene promoter region in 155 patients with bladder cancer and in 155 cancer-free controls was amplified by PCR-SSCP technique, and the SNP were confirmed by direct DNA sequencing. The recombinant plasmids of JWA promoter fragment which contain the SNP were constructed as CAT reporter gene and were transfected transiently into NIH 3T3 cells for disclosing whether SNP changes the transcriptional activity of the promoter.A novel SNP -76 G--C at promoter region of JWA gene was found. The frequencies of the C allele and GC genotype at JWA promoter -76G--C in bladder cancer group (10.00% and 20.00% respectively) were significantly higher than those in control group (5.16% and 10.32% respectively) (P0.05). The transcriptional activity of -76GC allele genotype was significantly down-regulated as compared with that of -76GG allele genotype (P0.01). Multivariate logistic regression analysis revealed that JWA polymorphism at promoter -76G--C is an independent novel risk factor for bladder cancer.The JWA -76G--C variant genotype may play an important role in transcription regulation of JWA gene and in the susceptibility to bladder cancer.

Congenital agenesis of the seminal vesicle (CASV) is frequently associated with congenital absence of the vas deferens (CAVD) or ipsilateral congenital vasoureteral communication. We reported two cases of a rare condition that the vas deferens open ectopically into Mullerian duct cyst associated with agenesis of the ipsilateral seminal vesicle. The diagnosis was confirmed by vasography. Transurethral unroofing of the Mullerian duct cyst was performed in both patients with favourable results, however, assisted reproductive technology (ART) was still necessary for them to father children.

To evaluate the clinical efficacy of high intensity focused ultrasound (HIFU) combined with endocrine therapy in the treatment of patients with prostate cancer.Twenty patients with prostate cancer were treated with extracorporeal HIFU device( model FEP-BY01 ) and androgen ablation, of whom 15 received orchiectomy and 5 LHRH-a. Fourteen patients of the total number were given flutamide in addition to castration.The mean follow-up was 13.5 months (ranging 6 to approximately 30). Before and after the treatment, the prostate volume, prostate specific antigen (PSA), international prostate symptom score (IPSS) and maximal flow rate (Qmax) of the patients were (36.4 +/- 16.2) ml and (20.6 +/- 11.8) ml (P0.05), (32.2 +/- 10.4) ng/ml and (2.4 +/- 0.8) ng/ml (P0.01), 20. 5 +/- 6.5 and 13.6 +/- 7.5 (P0.05), (10.6 +/- 6.3) ml/s and (14.2 +/- 4.6) ml/s (P0.05), respectively. To study the effect of combined androgen block therapy on hemoglobin and hematocrit values in patients with prostate cancer.One hundred and thirty-six patients with adenocarcinoma of prostate were treated with combined androgen block (orchiectomy and flutamide 250 mg, tid). Complete blood counts were determined before and after 1, 2, 3, 6, 9 and 12 months of therapy.The hemoglobin and hematocrit levels declined significantly in all patients and at all the time points after treatment (P0.05).Prostate cancer patients treated with combined androgen block would develop obvious anemia. Recombinant human erythropoietin can be used to treat patients with severe anemia.

To investigate the effect of free internal prepuce lamina patch urethroplasty on long-segment urethral stricture.Eight patients of long-segment urethral stricture underwent resection of atresia posterior urethra and/or incision of stricture anterior urethra, free internal prepuce lamina patch as of corresponding length and width were taken for the reconstruction of the urethra, and mean while multi-hole silica ge tract was placed in the urethra.Miction was easy and fluent in 7 cases, and unobstructed in 1 case after two urethral soundings.Free internal prepuce lamina patch urethroplasty was a good method for treating long-segment urethral stricture.

To evaluate the clinical efficacy of alpha-1 A adrenoceptor antagonist (tamsulosin) in the treatment of benign prostatic hyperplasia (BPH) patients with acute urinary retention.Seventy-two BPH patients with acute retention of urine were randomly divided into treatment group and control group of 36 patients each. All the patients were treated with indwelling catheter, oral antibiotics and the patients in treatment group tamsulosin 0.4 mg once a day for 3 days. The catheter was removed after 72 hours of treatment.After removal of the catheter, 44% (32/72) of patients voided successfully. The effect rates were 61% (22/36) in the tamsulosin treatment group and 28% (10/36) in the control group(P0.01).Treatment with tamsulosin was effective in raising the success rate of voiding without catheter after an episode of acute urinary retention. The efficacy of treatment was not influenced by the volume of prostate.

Female sexual dysfunction(FSD) is an age-related progressive disease and may affect up to half of adult women. It has not been paid attention to for a long time. This paper reviews the current information on the anatomy, physiology and pathophysiology of FSD. In addition the diagnosis and treatment of FSD are also described. It aims at improving the knowledge of FSD.

To study the effect of cryopreservative period on the cryosurvival of human spermatozoa and find out the optimal recovery time of cryopreservation.Eighty-eight semen samples were collected from normal donors and divided randomly into 5 groups according to the period of cryopreservative storage (1 d, 7 d, 30 d, 180 d, 300 d) in liquid nitrogen after being frozen by the programized, three-step freezing method. Fresh and frozen-thawed semen were examined by the routine analysis of semen and then the sperm recovery rate were calculated.There were no significant differences in sperm recovery rate between group I and the others (P0.05). The period of cryopreservative storage in liquid nitrogen had no correlation with the cryosurvival of human spermatozoa (r = 0.05, P0.05).It was indicated that freezing-thawing after 24 h would be helpful to the screening of semen donors in batches for donor insemination of human sperm bank.

To assess the effect of transrectal prostatic biopsy (TPB) on the concentrations of serum prostate specific antigen (PSA).Twenty patients with abnormal PSA levels and/or digital rectal examination (DRE) underwent TPB. Serum PSA levels were measured before TPB and at 0.5 h, 1 week, 1 month after TPB, respectively.The serum PSA levels before TPB and 0.5 h, 1 week, 1 month after TPB were (12.23 +/- 8.62), (34.90 +/- 41.53), (23.59 +/- 20.78) and (11.31 +/- 6.95) micrograms/L, respectively. The serum PSA concentration was significantly higher at 0.5 h after TPB than before (P0.05), and then gradually decreased. PSA levels remained higher for at least 1 week in 85% (17/20) patients(P0.05), then returned to the baseline at one month after TPB (P0.05).TPB can lead to a dramatic increase of PSA in serum and keep the PSA value high in one week. Then the PSA in serum decreased gradully. Serum PSA level cannot return to baseline until one month after TPB.
