Introduction MRI of excised hearts at ultra-high field strengths ($${\mathrm{B}}_{0}$$ B 0 ≥7 T) can provide high-resolution, high-fidelity ground truth data for biomedical studies, imaging science, and artificial intelligence. In this study, we demonstrate the capabilities of a custom-built, multiple-element transceiver array customized for high-resolution imaging of excised hearts. Method A dedicated 16-element transceiver loop array was implemented for operation in parallel transmit (pTx) mode (8Tx/16Rx) of a clinical whole-body 7 T MRI system. The initial adjustment of the array was performed using full-wave 3D-electromagnetic simulation with subsequent final fine-tuning on the bench. Results We report the results of testing the implemented array in tissue-mimicking liquid phantoms and excised porcine hearts. The array demonstrated high efficiency of parallel transmits characteristics enabling efficient pTX-based B1+-shimming. Conclusion The receive sensitivity and parallel imaging capability of the dedicated coil were superior to that of a commercial 1Tx/32Rx head coil in both SNR and T2*-mapping. The array was successfully tested to acquire ultra-high-resolution (0.1 × 0.1 × 0.8 mm voxel) images of post-infarction scar tissue. High-resolution (isotropic 1.6 mm3 voxel) diffusion tensor imaging-based tractography provided high-resolution information about normal myocardial fiber orientation.

Two 16-element radio frequency (RF) transceiver antisymmetric coil arrays with two different implementation designs aiming for improved characteristics for static phase $B_{1}^{+}$ shimming and parallel receive were developed for human cardiac MRI at 7 T. Both cardiac arrays consist of a weakly curved lightweight housing fit to an average human thorax shape as an anterior array in combination with a flat posterior array. The first array (Design 1) comprised eight identical antisymmetric loops for both anterior and posterior sections. Based on initial testing and to improve parallel imaging capabilities and $B_{1}^{+}$ field homogeneity, the second array (Design 2) was composed of 12-loops for the anterior section and four antisymmetric loops of larger size for the posterior section. Electromagnetic-field (EMF) simulations were carried out for both antisymmetric arrays loaded with an elliptical-shaped thorax phantom and two human voxel models (Duke and Ella). Static phase $B_{1}^{+}$ shimming has been carried out within Duke and Ella models using two optimization cost functions aiming to maximize the transmit efficiency and weighted combination of both $B_{1}^{+}$ field homogeneity and efficiency. The hardware and imaging performance of the two developed antisymmetric cardiac arrays was validated through EMF simulations, benchtop measurements, and MR measurements in a thorax phantom. Both antisymmetric arrays were compared to two commercial 16-element transceiver arrays (a 1Tx/16Rx in single transmit mode and an 8Tx/16Rx in parallel transmit mode). MRI measurements were performed with Design 2 using a 70-kg fresh pig cadaver (10–15 min postmortem). Parallel imaging with an acceleration factor up to $R = 6$ was possible using Design 2 while maintaining a mean g-factor of 1.47 within the pig heart. $T_{2}^{\ast }$ -weighted images of the pig heart were acquired using up to $R = 5$ with a spatial resolution of $0.35 \times 0.35 \times 4$ mm3.

ObjectivesMicro-computed tomography (μ-CT) and histology, the current gold standard methods for assessing the formation of new bone and blood vessels, are invasive and/or destructive. With that in mind, a more conservative tool, dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI), was tested for its accuracy and reproducibility in monitoring neovascularization during bone regeneration. Additionally, the suitability of blood perfusion as a surrogate of the efficacy of osteoplastic materials was evaluated.Materials and methodsSixteen rabbits were used and equally divided into four groups, according to the time of euthanasia (2, 3, 4, and 6 weeks after surgery). The animals were submitted to two 8-mm craniotomies that were filled with blood or autogenous bone. Neovascularization was assessed in vivo through DCE-MRI, and bone regeneration, ex vivo, through μ-CT and histology.ResultsThe defects could be consistently identified, and their blood perfusion measured through DCE-MRI, there being statistically significant differences within the blood clot group between 3 and 6 weeks (p= 0.029), and between the former and autogenous bone at six weeks (p= 0.017). Nonetheless, no significant correlations between DCE-MRI findings on neovascularization and μ-CT (r=−0.101, 95% CI [−0.445; 0.268]) or histology (r= 0.305, 95% CI [−0.133; 0.644]) findings on bone regeneration were observed.ConclusionsThese results support the hypothesis that DCE-MRI can be used to monitor neovascularization but contradict the premise that it could predict bone regeneration as well.

To improve parallel transmit (pTx) and receive performance for cardiac MRI (cMRI) in pigs at 7 T, a dedicated transmit/receive (Tx/Rx), 16-element antisymmetric dipole antenna array, which combines L-shaped and straight dipoles, was designed, implemented, and evaluated in both cadavers and animals in vivo. Electromagnetic-field simulations were performed with the new 16-element dipole antenna array loaded with a pig thorax-shaped phantom and compared with an eight-element array of straight dipoles. The new dipole array was interfaced to a 7 T scanner in pTx mode (8Tx/16Rx). Imaging performance of the novel array was validated through MRI measurements in a pig phantom, an 85 kg pig cadaver, and two pigs in vivo (74 and 81 kg). Due to the improved decoupling between interleaved L-shaped and straight dipole elements, the 16-element dipole array fits within the same outer dimensions as an eight-element array of straight dipoles. A dedicated, novel, multichannel transmit/receive (Tx/Rx) coil array was designed, simulated, and tested for cardiac magnetic resonance imaging (cMRI) of pigs at 7 T. The cardiac array was composed of 16 elements with physically independent anterior and posterior parts. The array was connected to the MRI scanner in the parallel transmit (pTx) mode forming an 8-channel transmit and 16-channel receive (8Tx/16Rx) coil configuration. The anterior array was composed of four central octagonal elements, which were decoupled using a common central ring (CCR) and shared decoupling capacitors. The surrounding four triangular elements were decoupled from the central four elements using gaps and capacitive decoupling. In total, the eight elements of the anterior array resembled a coil with a symmetrical circular shape. Due to the advantageous arrangement of the elements in conjunction with the novel decoupling method of a CCR, the coupling between the adjacent loop elements was minimized. The posterior array was built using a $4 \times 2$ standard rectangular, symmetric element configuration. The dedicated cardiac array was tested in phantom and in ex vivo MR measurements with a female pig of 46 kg. The dedicated coil array enhanced the signal-to-noise ratio (40 ± 27) in ex vivo measurements of the pig heart by about three times compared to a coil prototype for cardiac human imaging (11.8 ± 7). High-resolution, ex vivo cardiac images were acquired with an in-plane resolution of up to 0.3 mm $\times0.3$ mm using the dedicated pig coil array. The novel cardiac array supports parallel imaging with an acceleration factor (R) of 4 without increasing the mean $g$ -factor within the heart region beyond 1.15.

The design, simulation, assembly and testing of a novel dedicated antisymmetric transmit/receive (Tx/Rx) coil array to demonstrate the feasibility of cardiac magnetic resonance imaging (cMRI) in pigs at 7 T was described. The novel antisymmetric array is composed of eight elements based on mirrored and reversed loop orientations to generate varying B 1 + field harmonics for RF shimming. The central four loop elements formed together a pair of antisymmetric L-shaped channels to allow good decoupling between all neighboring elements of the entire array. The antisymmetric array was compared to a standard symmetric rectilinear loop array with an identical housing dimension. Both arrays were driven in the parallel transmit (pTx) mode forming an 8-channel transmit and 16-channel receive (8Tx/16Rx) coil array, where the same posterior array was combined with both anterior arrays. The hardware and imaging performance of the dedicated cardiac arrays were validated and compared by means of electromagnetic (EM) simulations, bench-top measurements, phantom, and ex-vivo MRI experiments with 46 kg female pig. Combined signal-to-noise ratio (SNR), geometry factor (g-factor), noise correlation maps, and high resolution ex-vivo cardiac images were acquired with an in-plane resolution of 0.3 mm × 0.3 mm using both arrays. The novel antisymmetric array enhanced the SNR within the heart by about two times and demonstrated good decoupling and improved control of the B 1 + field distributions for RF shimming compared to the standard coil array. Parallel imaging with acceleration factor (R) up to 4 was possible using the novel antisymmetric coil array while maintaining the mean g-factor within the heart region of 1.13.

In this study we aimed to assess the effects of continuous formalin fixation on diffusion and relaxation metrics of the ex vivo porcine heart at 7 T. Magnetic resonance imaging was performed on eight piglet hearts using a 7 T whole body system. Hearts were measured fresh within 3 hours of cardiac arrest followed by immersion in 10% neutral buffered formalin. T\(_{2}\)* and T\(_{2}\) were assessed using a gradient multi‐echo and multi‐echo spin echo sequence, respectively. A spin echo and a custom stimulated echo sequence were employed to assess diffusion time‐dependent changes in metrics of cardiac diffusion tensor imaging. SNR was determined for b = 0 images. Scans were performed for 5 mm thick apical, midcavity and basal slices (in‐plane resolution: 1 mm) and repeated 7, 15, 50, 100 and 200 days postfixation. Eigenvalues of the apparent diffusion coefficient (ADC) and fractional anisotropy (FA) decreased significantly (P < 0.05) following fixation. Relative to fresh hearts, FA values 7 and 200 days postfixation were 90% and 80%, while respective relative ADC values at those fixation stages were 78% and 92%. Statistical helix and sheetlet angle distributions as well as respective mean and median values showed no systematic influence of continuous formalin fixation. Similar to changes in the ADC, values for T\(_{2}\), T\(_{2}\)* and SNR dropped initially postfixation. Respective relative values compared with fresh hearts at day 7 were 64%, 79% and 68%, whereas continuous fixation restored T\(_{2}\), T\(_{2}\)* and SNR leading to relative values of 74%, 100%, and 81% at day 200, respectively. Relaxation parameters and diffusion metrics are significantly altered by continuous formalin fixation. The preservation of microstructure metrics following prolonged fixation is a key finding that may enable future studies of ventricular remodeling in cardiac pathologies.

High-frequency oscillatory ventilation is looked upon as a lung-protective ventilation strategy. For a further clarification of the physical processes promoting gas transport, a visualization of gas flow and the distribution of ventilation are of considerable interest. Therefore, fluorine-19 magnetic resonance imaging of the imaging gas octafluorocyclobutane (C(4) F(8) ) during high-frequency oscillatory ventilation was performed in five healthy pigs. For that, a mutually compatible ventilation-imaging system was set up and transverse images were acquired every 5 sec using FLASH sequences on a 1.5 T scanner. Despite a drop in signal-to-noise ratio after the onset of high-frequency oscillatory ventilation, for each pig, the four experiments could be analyzed. A mean wash-out time (τ) at 5 Hz of 52.7 ± 18 sec and 125.9 ± 39 sec at 10 Hz, respectively, were found for regions of interest including the whole lung. This is in agreement with the clinical findings, in that wash-out of respiratory gases is significantly prolonged for increased high-frequency oscillatory ventilation frequencies. Our study could be a good starting-point for a further optimization of high-frequency oscillatory ventilation.

Imaging with hyperpolarized 3-helium is becoming an increasingly important technique for MRI diagnostics of the lung but is hampered by long breath holds (>20 sec), which are not always applicable in patients with severe lung disease like chronic obstructive pulmonary disease (COPD) or α-1-anti-trypsin deficiency. Additionally, oxygen-induced depolarization decay during the long breath holds complicates interpretation of functional data such as apparent diffusion coefficients. To address these issues, we describe and validate a 1.5-T, 32-channel array coil for accelerated 3He lung imaging and demonstrate its ability to speed up imaging 3He. A signal-to-noise ratio increase of up to a factor of 17 was observed compared to a conventional double-resonant birdcage for unaccelerated imaging, potentially allowing increased image resolution or decreased gas production requirements. Accelerated imaging of the whole lung with one-dimensional and two-dimensional acceleration factors of 4 and 4 × 2, respectively, was achieved while still retaining excellent image quality. Here, design of the first pathogen-mimicking metal oxide nanoparticles with the ability to enter cancer cells and to selectively target and activate the TLR9 pathway, and with optical and MR imaging capabilities, is reported. The immobilization of ssDNA (CpG ODN 2006) on MnO nanoparticles is performed via the phosphoramidite route using a multifunctional polymer. The multifunctional polymer used for the nanoparticle surface modification not only affords a protective organic biocompatible shell but also provides an efficient and convenient means for loading immunostimulatory oligonucleotides. Since fluorescent molecules are amenable to photodetection, a chromophore (Rhodamine) is introduced into the polymer chain to trace the nanoparticles in Caki-1 (human kidney cancer) cells. The ssDNA coupled nanoparticles are used to target Toll-like receptors 9 (TLR9) receptors inside the cells and to activate the classical TLR cascade. The presence of TLR9 is demonstrated independently in the Caki-1 cell line by western blotting and immunostaining techniques. The magnetic properties of the MnO core make functionalized MnO nanoparticles potential diagnostic agents for magnetic resonance imaging (MRI) thereby enabling multimodal detection by a combination of MR and optical imaging methods. The trimodal nanoparticles allow the imaging of cellular trafficking by different means and simultaneously are an effective drug carrier system.

The effectiveness of solid-state MRI with dipolar refocusing techniques was tested to improve the sensitivity and spatial resolution for the investigation of various solid polymers. The comparison of dipolar refocusing methods with conventional SPI was undertaken. As an alternative, gas-phase MRI experiments with thermally polarized SF6 and hyperpolarized xenon were performed. The performance of SF6 was found to be equivalent to solid-state methods. The possibility of achieving a significant increase in imaging efficiency by use of the hyperpolarization technique is demonstrated. The effects of embedding gaseous SF6 into EPDM rubber were investigated using NMR methods. A strong correlation was found between EPDM cross-linking and the transversal relaxation time of embedded SF6.

For our low-field resistive MRI-System we have developed suitable rf-probes to be able to image fingers with a possibly high spatial resolution. To reduce the measurement time and to improve the signal/noiseratio, which is inherently bad in a low field system, we used a multi-echo sequence, accumulating up to six echo signals for each image. For a 256 x 256 pixel matrix we achieved with this technique a spatial resolution of 176 microns/pixel in measuring times between 4 and 5 minutes. In comparison to healthy persons also first MR-images of patients suffering on arthrosis at the fingers and small joint injures were acquired. Using a 512 x 512 pixel matrix and a field of view of 5 cm images with a resolution of 80 microns/pixel were recorded in a total measuring time of less than 10 minutes.
