We found the main regularities of quantitative changes in energy-dispersive spectra in the determination of the elemental composition of powdered materials. At certain energy of probing electrons, the calculation of the relative weight fractions of elements in powder samples gives results corresponding to those obtained for reference polished sample surfaces. Generalizing the experimental data yielded an empirical dependence that relates the parameters of characteristic photons to the value of accelerating voltage required to obtain the proper ratio of the weight concentrations of elements in the analysis of powdered materials. A new technique is described for the concentration and separation of ionic species in solution within microchannels or capillaries. Concentration is achieved by balancing the electrophoretic velocity of an analyte against the bulk flow of solution in the presence of a temperature gradient. With an appropriate buffer, the temperature gradient can generate a corresponding gradient in the electrophoretic velocity, so that the electrophoretic and bulk velocities sum to zero at a unique point, and the analyte will be focused at that point. The technique is demonstrated for a variety of analytes, including fluorescent dyes, amino acids, DNA, proteins, and particles, and is shown to be capable of greater than 10 000-fold concentration of a dilute analyte.

Complaints due to odors are an important problem for the wastewater, composting, and animal agriculture industries. Accurate, objective measurement techniques are needed to monitor emissions, to develop new waste handling procedures, and to reduce the production of these volatile gases. Solid-phase microextraction was investigated as a technique for the determination of representative odorous gases. A flow-through Teflon chamber was used to expose the fibers to certified gas standards. A 75-[micro] m carboxen-poly(dimethylsiloxane) (Car-PDMS) coating was used for trimethylamine (TMA), carbon disulfide (C[S.sub.2]), dimethylsulfide (DMS), and dimethyl disulfide (DMDS), and an 85-[micro] m polyacrylate coating was used for propionic acid (PA) and butyric acid (BA). Using a 1-h fiber exposure time and a flow rate through the chamber of 72 mL/min, method detection limits were 2.38, 0.074, 0.150, 0.063, 1.85, and 1.32 ppbv for TMA, DMS, C[S.sub.2], DMDS, PA, and BA, respectively. Enhanced detector signal was observed for all analytes under flow conditions, as compared to static conditions, and the porous nature of the Car-PDMS coating appears to increase the time needed for analytes to reach equilibrium under flow conditions.

Oligonucleotide microarrays are used to analyze target sequences on the basis of differences in hybridization stability between matched and mismatched probe-target duplexes. DNA microarray manufacture via photolithographic synthesis generates a minority of full-length oligonucleotide probes along with a series of 5'-truncated contaminants. In a model experiment, we now investigate the effect of truncated oligonucleotides on the ability to distinguish target sequence variants that differ in a single nucleotide position. A series of oligonucleotides, mixed in proportions simulating stepwise synthetic yields of between 82 and 100%, were bound to a solid support and allowed to hybridize to a target molecule. The extent of hybridization was monitored over a range of temperatures via the fluorescence of a double-strand-specific dye. The discriminatory power of pure oligonucleotide probes was found to be significantly greater than that of a population of truncated probes, but only over a limited temperature interval. We conclude that at optimal temperatures greater oligonucleotide quality can improve the performance of oligonucleotide hybridization microarrays.

A preformed T-microchannel imprinted in polycarbonate was postmodified with a pulsed UV excimer laser (KrF, 248 nm) to create a series of slanted wells at the junction. The presence of the wells leads to a high degree of lateral transport within the channel and rapid mixing of two confluent streams undergoing electroosmotic flow. Several mixer designs were fabricated and investigated. All designs were relatively successful at low flow rates (0.06 cm/s, [greater than or equal to] 75% mixing), but had varying degrees of success at high flow rates (0.81 cm/s, 45-80% mixing). For example, one design operating at high flow rates was able to split an incoming fluorescent stream into two streams of varying concentrations depending on the number of slanted wells present. The final mixer design was able to overcome stream splitting at high flow rates, and it was shown that the two incoming streams were 80% mixed within 443 [micro]m of the T-junction for a flow rate of 0.81 cm/s. Without the presence of the mixer and at the same high flow rate, a channel length of 2.3 cm would be required to achieve the same extent of mixing when relying upon molecular diffusion entirely, while 6.9 cm would be required for 99% mixing.

Capillary electrophoresis was employed in examining rat airway surface fluid samples. The fluid was found hypotonic as indicated by Cl-, K+ and Na+ values. Airway surface fluid nitrite and nitrate levels, however, were found higher than plasma values. Results demonstrate the use of the microanalytical approach in examining airway surface fluid composition in vivo regulation.

We describe the application of infrared optical tweezers to the in situ microparticle analysis of marine phytoplankton cells. A Nd:YAG laser ([Lambda] = 1064 nm) trap is used to confine and manipulate single Nannochloris and Synechococcus cells in an enriched seawater medium while spectral fluorescence and Lorenz-Mie backscatter signals are simultaneously acquired under a variety of excitation and trapping conditions. Variations in the measured fluorescence intensities of chlorophyll a (Chl a) and phyco-erythrin pigments in phytoplankton cells are observed. These variations are related, in part, to basic intrasample variability, but they also indicate that increasing ultraviolet-exposure time and infrared trapping power may have short-term effects on cellular physiology that are related to Chl a photobleaching and laser-induced heating, respectively. The use of optical tweezers to study the factors that affect marine cell physiology and the processes of absorption, scattering, and attenuation by individual cells, organisms, and particulate matter that contribute to optical closure on a microscopic scale are also described.

Leucine aminopeptidase, a clinically significant enzyme, was assayed in complex biological samples using a new technique termed electrophoretically mediated microanalysis. The assay was performed in capillary electrophoresis columns using time-resolved laser-induced fluorescence detection. Human serum, human urine, and Escherichia coli supernatant samples were assayed using this method. Results for serum and urine were within the ranges of expected values found in the literature. A low concentration of 6 X 10 to the -13 M enzyme in buffer was detected using this method. A detection limit (3-Sigma) of 400 enzyme molecules in buffer was determined.

A new concept in reaction-based chemical analysis is introduced and theoretically described. By utilization of the variability in electrophoretic mobilities among charged species, spatially distinct zones of chemical reagents can be electrophoretically merged under the influence of an applied electric field. Electrophoretically mediated microanalysis (EMMA) exploits this phenomenon as a basis for chemical analysis utilizing capillary electrophoretic systems. EMMA is described in terms of the four stages required for reaction-based analysis: (1) analyte and analytical reagent metering; (2) initiation of reaction; (3) control of reaction conditions and product formation; (4) detection of species whose production or depletion is indicative of the concentration or quantity of the analyte of interest. The method is illustrated by the enzymatic oxidation of ethanol to acetaldehyde by alcohol dehydrogenase with the concurrent reduction of NAD(super +) to NADH monitored at 340 nm. Experimental results for both substrate and enzyme determinations are shown to agree with the presented theory.

Argon ion milling is the conventional means by which mineral sections are thinned to electron transparency for transmission electron microscope (TEM) analysis, but this technique exhibits significant shortcomings. In particular, selective thinning and imaging of submicrometer inclusions during sample milling are highly problematic. We have achieved successful results using the focused ion beam (FIB) lift-out technique, which utilizes a 30 kV [Ga.sup.+] ion beam to extract electron transparent specimens with nanometer scale precision. Using this procedure, we have prepared a number of Earth materials representing a range of structures and compositions for TEM analysis. We believe that FIB milling will create major new opportunities in the field of Earth and planetary materials microanalysis, particularly with respect to ultraprecious mineral and rock samples.

A class of 'lab-on-a-chip' devices use external air pressure for pumping discrete drops in a microchannel network. External air connectors can be cumbersome and are real-estate intensive. We have developed an on-chip technique to generate pressures required for metering and pumping of nanoliter-volume discrete drops. This is achieved by heating of trapped air in a pressure-generating chamber. The pressure-generating chamber is connected to the point of pressure application in the liquid-conveying microchannel through an air-delivery channel. The trapped air volume on the order of 100 nL is heated by resistive metal heaters by tens of degrees celcius to generate air pressures on the order of 7.5 kN/[m.sup.2]. The rate of discrete drop pumping is electronically controlled in the microchannel device by controlling the rate of air heating. Flow rates on the order of 20 nL/s are obtained in the microchannel (300 [micro]m x 30 [micro]m) by heating the air chamber at the rate of ~6 [degrees] C/s. In this paper, we describe the design, fabrication, and operation of this new technique of generating on-chip air pressure, used for metering and pumping nanoliter discrete drops in microchannels.

Spatially resolved chemical imaging is achieved by combining a fiber-optic scanning probe microscope with laser-induced breakdown spectroscopy in a single instrument, TOPOLIBS. Elemental composition of surfaces can be mapped and correlated with topographical data. The experiment is conducted in air with minimal sample preparation. In a typical experiment, surface topography is analyzed by scanning a sharp fiber-optic probe across the sample using shear force feedback. The probe is then positioned over a feature of interest and pulsed radiation is delivered to the surface using a nitrogen laser. The pulse vaporizes material from the surface and generates a localized plasma plume. Optical emission from the plume is analyzed with a compact UV/visible spectrometer. Ablation crater size is controlled by the amount of laser power coupled into the probe. Sampling areas with submicrometer dimensions are achieved by using reduced laser power.

Seahorses (Hippocampus spp.; Family Syngnathidae) are mainly targeted by Traditional Chinese Medicine, curio and ornamental trade, as dried or live specimens. Traceability tools may help fill the gaps on supply chains, securing information on geographic origin and identification of traded specimens. Fin-clipping, a non-lethal and well-established method in seahorse research, offers a potential approach to trace the geographic origin and certify the aquaculture of these flagship species. As such, this study aimed to investigate the existence of differences in isotopic profiles of four seahorse species cultured at research centers located in southern Portugal and northern Spain, as well as between cultured Hippocampus guttulatus sourced from two research centers, and between wild and cultured specimens of this species. This research also evaluated the potential of combining isotopic and elemental fingerprints for seahorse species discrimination, through inductively continuous-flow isotope ratio mass spectrometry (IR-MS) and plasma mass spectrometry (ICP-MS). Species cultured at the same research centers exhibited similar stable isotope composition (δ13C and δ15N), except in the case of temperate H. guttulatus from northern Spain, which differed significantly from tropical species H. kuda and H. reidi. These differences could be due to phylogenetic dissimilarities and differences in seawater temperature. The δ15N composition allowed to discriminate between cultured H. guttulatus from the two research centers and between cultured and wild specimens. While dorsal fin isotopes alone did not prove to be a reliable tool for the discrimination of different cultured species, combining them with elemental profiles from seahorses' whole-body allowed to successfully discriminate between H. kuda and H. reidi. This preliminary research demonstrates the potential of stable isotope and elemental analyses for tracing seahorses' geographic origin and species identification. However, further research should be performed to validate these findings for wild specimens, particularly those from illegal, unreported and unregulated (IUU) fisheries and trade. In marine fishes of commercial interest, defining habitat use and migration strategies through ontogeny can help better understand the structure and dynamics of harvested populations and guide their management. The present study relied on otolith chemistry to identify three contingents within the Atlantic halibut (Hippoglossus hippoglossus) stock in the Gulf of St. Lawrence (GSL). We differentiated two chemical signatures from otolith edges, one for shallow (<100 m) and another one for deep (>100 m) waters. By identifying transitions between the deep and shallow habitats, we found that most halibut display migrations from the deep waters to shallow waters during the first 3 years of life. After reaching maturity, most halibut distributing in northern regions of the GSL became full-time residents in deep areas of the GSL. In contrast, halibut found in summer on the shallow plateau of the southern GSL displayed migrating behaviour between shallow (summer) and deep (winter) waters throughout their lives, either on an annual or irregular basis. Overall, our results demonstrate that otolith chemical signatures serve as natural markers of geographically distinct marine environments, facilitating the identification and reconstruction of environmental histories of long-lived marine fishes. To tackle the question of the reliability of otoliths as recorders of individual life events, we compared the information enclosed in otolith pairs: the sagittae pair and the sagitta/lapillus pair. We used the synchrotron XRF scanning imaging method, which enabled the comparison of this information at both global and hyperfine scales. Using otoliths of diadromous pipefish, we compared element incorporation in each pair with a focus on (i) environment and transition between water bodies with strontium (Sr) and heavy metals, (ii) temporal information and age estimation based on sulphur (S) incorporation, and (iii) otolith growth and biomineralization processes with zinc (Zn). Results show that the global information in terms of Sr and heavy metals given by both otoliths of a pair is the same and that any otolith may be used to retrieve such global data. In terms of S-based growth increment counts, the numbers are the same between two otoliths of the same kind, but the sagitta/lapillus pairs show a significant difference. Hyperfine-scale analysis of element distribution reveals that a given otolith is under the control of specific growth mechanisms, which can lead to heterogeneous elemental incorporation. Chronological records of elemental concentrations in fish otoliths are a widely used tool to infer the environmental conditions experienced by individual fish. To interpret elemental signals within the otolith, it is important to understand how both external and internal factors impact ion uptake, transport and incorporation. In this study, we have combined chronological records from otoliths and archival data storage tags to quantify the influence of internal (sex, size, age, growth) and external (temperature, depth, salinity) conditions on otolith elemental chemistry of cod (Gadus morhua) in natural settings of the Baltic Sea. This study focused on elements primarily under physiological control: Phosphorus (P), magnesium (Mg) and zinc (Zn); and elements under environmental control: Strontium (Sr), barium (Ba) and manganese (Mn). Based on known spatial and temporal patterns in environmental conditions and fish size, growth, and maturity, we posed a series of hypotheses of expected otolith element patterns. Partial effects of internal and external drivers on element concentration were analyzed using a Linear Mixed Model approach with random variables (fish and year). Predicted effects of otolith concentrations of all elements under physiological control (P, Mg, Zn) showed similar trends, with distinct seasonal patterns (lowest concentration in late spring, highest concentrations in winter), and a positive correlation with water temperature, in addition to higher Zn and lower P in spawning individuals. Predicted effects of otolith concentrations of elements expected to be predominantly under environmental control showed the predicted geographic and depth-related trends based on ambient salinity (Ba) and coastal hypoxia (Mn). However, contrary to expectation, Sr was unrelated to salinity. Predicted otolith Ba, Sr and Mn concentrations also exhibited pronounced seasonal patterns that were out of phase with each other but appeared to be partly explained by spawning/feeding migrations. While performing laboratory validation studies for adult fish is typically not possible, these results highlight the importance of assessing local water chemistry and freshwater endmembers in one's study system before otolith elemental chemistry can be reliably used to reconstruct fish habitat use and environmental histories. The spatial and temporal uncertainty in availability and distribution of wild mussel seed is a major obstacle to sustainable production in mussel aquaculture globally. An improved understanding of patterns of dispersal of wild mussel spat could help to resolve this uncertainty to progress the management of wild spat resources and increase mussel seed supply. In this study, the natal locations and patterns of dispersal for recently settled green-lipped mussels, Perna canaliculus , at an offshore mussel farm in northern New Zealand were estimated using a combination of analysis of shell microchemistry of arriving spat and biophysical modelling. The shell microchemistry from mussel spat settling on the offshore mussel farm were compared with those from mussels cultured in situ at 22 surrounding sites (∼2–6 km apart) arranged around the Bay of Plenty, New Zealand, over two consecutive months. Six elemental signatures (Li, Co, Ni, Ba, La and U) were the primary drivers for correctly reassigning shells back to their individually generated sites with an overall 57.4% of Leave-one-out cross-validated (LOOCV) cases correctly assigned, ranging from a low of 10% for one coastal site (near Hikuwai Beach) and up to 78% for an offshore mussel spat collection site (near mussel farm). The microchemistry signatures were subsequently employed to backtrack ∼160 spat arriving on the mussel farm to two main natal sites (i.e., 7.5 and 8.0 km away) near Ōhiwa Harbour and known to have wild mussel reefs, which accounted for 85% of the mussels arriving on the mussel farm. Forward and backward simulations of movements of mussel spat using high resolution (i.e., 1 km) hydrodynamic particle tracking models indicated that most spat arriving on the mussel farm during the sampling period originated from the same stretch of coast identified by the shell microchemistry. The concordance of results from the two methods of determining natal sources provide mutual validation of the results and highlight the importance of managing coastal populations of wild mussels that are under increasing pressure from human activities, especially where they provide a critical seed source for aquaculture. • Microchemistry methods can effectively identify wild spat source in an open embayment. • Most of mussel spat settling on the farm area during Nov to Jan are larger than 1 mm. • The majority (85%) of mussel spat arriving at the farm came from two possible natal sites. • Biophysical modelling suggests age-dependant wild mussel seed dispersal pattern. • A multidisciplinary approach is valuable in addressing the shortage of wild mussel spat. Using strontium (Sr) and barium (Ba) in otoliths to determine natal origins and understand patterns of fish movements is based on the fundamental assumption that otoliths record water chemistry signals without any major alterations. Although prior studies highlighted that fish physiology can modify the water signal in otoliths, studies for freshwater fish are scarce. We exposed different groups of Atlantic salmon parr Salmo salar to different scenarios of ambient-level variations in Sr/Ca and Ba/Ca ratios and then combined otolith chemical profiles with environmental data (water chemistry and temperature), Fulton's index, and otolith growth rates to assess what factors explain/influence the elemental ratios of Sr and Ba in otoliths. Generalized additive mixed models (GAMMs) using water-based otolith composition, temperature, Fulton's index, and "individual" as explanatory variables allow to demonstrate that water chemistry alone cannot fully explain measured ratios in otoliths, except in scenarios involving significant changes in water chemistry. Other factors (physiological effects) should be accounted for reproducing short and minimal seasonal variations in water composition, considering that inter-individual variability contributes quite significantly in most scenarios. A Femtosecond Laser Ablation-Inductively Coupled Plasma-Time of Flight Mass Spectrometer (fs-LA-ICP-TOFMS) was evaluated for multi-elemental mapping of complex otoliths; in particular, for the analysis of a sample of 8 year-old freshwater fish otolith with irregular growth ring distribution pattern. The fundamental wavelength (λ ∼ 1028 nm) and the 4th harmonic (λ ∼ 257 nm) of a diode pumped Yb:KGW fs-laser (<290 fs) were successfully employed for otolith elemental mapping (map area ∼6000 μm2), at high spatial resolution (spot sizes ≤5 μm) and with negligible thermal effects. Fast, sensitive multi-elemental analysis was achieved combining the use of high laser repetition rates (≥200 Hz), high sample scanning speeds (100 μm s−1), a fast response time ablation cell (<40 ms), and the use of a ICP-TOFMS equipped with a reaction/collision cell (operated in He and H2) to remove plasma gas ion related species and polyatomic interferences. The multi-elemental (from 23Na+ to 238U+) fs-LA-ICP-TOFMS mapping of the Prochilodus lineatus lapillus otolith allowed the identification of the appropriate growth axis based on the chronological chemical record associated with the endogenous and exogenous changes experienced by the fish throughout its life. Moreover, each of the detected and quantified elements provided useful biochemical information about environmental and endogenous factors. The Lebranche mullet (Mugil liza) is an economically important estuarine species found along the coast of Brazil. The southern population of this species ranges from the coast of the state of São Paulo (23°S) to the coast of Argentina (36°S). It migrates annually among estuaries in Argentina, Uruguay, and southern Brazil to reproduce in their marine spawning grounds (~26°S). We evaluated whether variations in the otolith chemical composition for fish acquired at nursery grounds can be used to make distinctions among 4 nursery areas of the southern population. Analysis of otolith microchemistry included concentrations of 9 elements: lithium (Li), magnesium (Mg), copper, zinc, strontium (Sr), cadmium, barium, lanthanum, and lead. When using random forest classification algorithms, the maximum accuracy of 96% in assignment of nursery habitat for Lebranche mullet between 2 groups (fish caught at nurseries in Brazil and fish migrating from waters of Uruguay and Argentina) was achieved with the combination of Li, Mg, and Sr. Our results indicate that the elemental composition of otoliths can be an important tool for establishing connectivity between nursery areas used by Lebranche mullet. We discuss the implications of this result for the structure of the population and the management of the mullet fishery in southern Brazil relative to the limitations of the methods we employed. To clarify potential trans-oceanic connectivity and variation in the natal origin of albacore tuna (Thunnus alalunga) from the southwest Indian Ocean (SWI) and the southeast Atlantic (SA), lifetime otolith elemental signatures were assessed from 46 adults sampled from Reunion Island, and 26 juveniles(group 2+) sampled from two locations along the Atlantic coast of South Africa. LA-ICP-MS analysis was used to assess the multi-elemental composition in B, Ba, Mg, P, Sr, and Zn along the otolith edge (chemical signatures of the capture area), but also near the otolith primordium (spawning origin) and in an area located at 1400–1600 µm from it (nursery origin). Two groups of distinct near-primordium multi-elemental signatures, denoting potentially discrete spawning origins (SpO), were identified using hierarchical clustering. Each of the two SpO was found to contribute to the albacore stocks from all the areas sampled, suggesting a common spawning origin in some fish from the SWI and from the SA, and complex migrations between the two oceans. Three potentially discrete primary nursery sites were identified, each contributing to SA juvenile and SWI adult capture sites differently. Anguillid eels were once considered to be the classic example of catadromy. However, alternative life cycles have been reported, including skipping the freshwater phase and habitat shifting between fresh, brackish, and saltwater throughout the growth phase. There is a lack of knowledge regarding these alternate life strategies, for example, the proportion of individuals in the population that adopt them compared to classic catadromy. We provide a description of these alternate life cycle strategies in temperate anguillids, their possible drivers, and the methods available to investigate them. These methods (lethal and non-lethal), include otolith microchemistry, fatty acid and stable isotope analyses, parasite identification, blood transcriptomics, and electronic tags. We argue that since the current management framework for the European eel and other temperate eels is based mainly on the freshwater component of the population, it ignores eels growing in saline waters. Many of the factors that are thought to be responsible for the precipitous decline of the eel population are more prevalent in freshwater systems. Therefore, the contribution of saline eels may be more important than currently estimated. The habitat-shifting ability of eels may be all the more crucial for the persistence and recovery of those species that are endangered. [ABSTRACT FROM AUTHOR]

Chemical analysis of calcified structures continues to flourish, as analytical and technological advances enable researchers to tap into trace elements and isotopes taken up in otoliths and other archival tissues at ever greater resolution. Increasingly, these tracers are applied to refine age estimation and interpretation, and to chronicle responses to environmental stressors, linking these to ecological, physiological, and life-history processes. Here, we review emerging approaches and innovative research directions in otolith chemistry, as well as in the chemistry of other archival tissues, outlining their value for fisheries and ecosystem-based management, turning the spotlight on areas where such biomarkers can support decision making. We summarise recent milestones and the challenges that lie ahead to using otoliths and archival tissues as biomarkers, grouped into seven, rapidly expanding and application-oriented research areas that apply chemical analysis in a variety of contexts, namely: (1) supporting fish age estimation; (2) evaluating environmental stress, ecophysiology and individual performance; (3) confirming seafood provenance; (4) resolving connectivity and movement pathways; (5) characterising food webs and trophic interactions; (6) reconstructing reproductive life histories; and (7) tracing stock enhancement efforts. Emerging research directions that apply hard part chemistry to combat seafood fraud, quantify past food webs, as well as to reconcile growth, movement, thermal, metabolic, stress and reproductive life-histories provide opportunities to examine how harvesting and global change impact fish health and fisheries productivity. Ultimately, improved appreciation of the many practical benefits of archival tissue chemistry to fisheries and ecosystem-based management will support their increased implementation into routine monitoring. The thread herring Opisthonema oglinum (Lesueur, 1818) is a small pelagic fish distributed in the western margin of the Atlantic Ocean from the United States to Argentina. It is a target species for diverse commercial fisheries, including the Brazilian industrial purse seine fleets that operate in the Southeastern Brazilian Bight. To investigate the geographic variation in the thread herring populations in this fishing ground, sagittal otoliths were collected from two areas: Rio de Janeiro (RJ: 23°04′ S, 44°03′ W) and Santa Catarina (SC: 26°05′ S, 48°18′ W). Otolith shape analyses and multi-elemental signatures were statistically evaluated using elliptical Fourier descriptors and elemental/Ca ratios. Remarkable differences in the thread herring otoliths between the two areas were found. The previous scenario in which the thread herring constitutes a single panmictic population in the Atlantic Ocean is now debatable. Scientists use otoliths to trace fish life history, especially fish migrations. Otoliths incorporate signatures of individual growth and environmental use. For many species, distinct increment patterns in the otolith are difficult to discern; thus, questions remain about crucial life history information. To unravel the history of such species, we use synchrotron-based scanning X-ray fluorescence. It allows the mapping of elements on the entire otolith at a high spatial resolution. It gives access to precise fish migration history by tagging landmark signature for environmental transition and it also characterises localised growth processes at a mineral level. Freshwater pipefish, which are of conservation concern, have otoliths that are small and fragile. Growth increments are impossible to identify and count; therefore, there is a major lack of knowledge about their life history. We confirm for the first time, by mapping strontium that the two tropical pipefish species studied are diadromous (transition freshwater/marine/freshwater). Mapping of other elements uncovered the existence of different migratory routes during the marine phase. Another major breakthrough is that we can chemically count growth increments solely based on sulphur signal as it is implicated in biomineralization processes. This novel method circumvents reader bias issues and enables age estimation even for otoliths with seemingly untraceable increments. Understanding the diversity of life history, life stage connectivity and population is essential to determine the spatial scale over which fish populations operate. Otolith microchemistry analysis is a powerful tool to elucidate the life history and population connectivity of fish, providing important insights to the natal origin and population structure. In this study, we used laser ablation inductively coupled plasma mass spectrometry to analyze the chemical composition of otoliths throughout the entire lifetime of endangered fourfinger threadfin species, Eleutheronema tetradactylum. We reconstructed the life history of E. tetradactylum from Southern China collected from different locations over a spatial scale of 1200 km. Sr:Ca and Ba:Ca ratios profiles from otolith core-to-edge analysis suggested two contrasting life history patterns. Based on the differences in early life stages, we identified some fish spending their first year in an estuarine environment with subsequent movement to marine coastal systems, while some fish remaining in the coastal systems throughout their entire early life history stages. The non-metric multi-dimensional scaling showed a strong overlap in otolith core elemental composition, indicating a large-scale connectivity in the life history of E. tetradactylum. The immature fish from different natal origins mixed to a large extent when they fed and overwintered in the extensive offshore waters. Clustering of near core chemistry pointed to three possible sources of nursery for the threadfin fish. This study demonstrated the diversity of life history patterns of E. tetradactylum in Southern Chinese waters. Invasive Silver Carp (Hypophthalmichthys molitrix) have established populations throughout the Missouri River basin. The main-stem Missouri River has undergone a multitude of alterations, creating a channel with greater mean depths and velocities, limiting optimal habitat for Silver Carp. Tributaries to the Missouri River may provide refuge from the swift flows within the main-stem Missouri River and, therefore, may play a vital role in the life-cycle of Silver Carp throughout the basin. Understanding the spatial extent under which these invasive fish function in this large, open river system is crucial to inform management efforts. Here, we used otolith microchemistry of Silver Carp from the Kansas River, a major tributary to the Missouri River, to reconstruct environmental histories as a means to assess the proportions of resident (individuals who never left the Kansas River system) and transient (individuals who at some point occupied the Missouri River) individuals. Silver Carp within the Kansas River were predominantly residents (adults = 54%; juveniles = 65%) with the majority of reproduction coming from within the Kansas River itself. These results suggest removal efforts in the Kansas River may be effective means of managing this invasive fish species. Transient fish exhibited short durations of signatures indicative of the Missouri River (mean percent of data points for adults = 10% and juveniles = 36%), suggesting movements into the Missouri River were brief. Though the narrow-barred Spanish mackerel (Scomberomorus commerson) is considered to be migratory, the species is nevertheless thought to be locally overexploited in the northwest Indian Ocean. At the regional level, this local depletion is a major concern for food security. As the population structure and connectivity between sub-populations are poorly understood for this species, we examined the spatio-temporal dynamics of narrow-barred Spanish mackerel via elemental concentrations (P, Mg, Sr and Ba) along otolith transects using LA-ICPMS for samples from 6 sites: Egypt, Djibouti North and South, Somalia, Mozambique and South Africa. For homogeneous size class samples (70–90 cm), otolith chemical signatures immediately preceding capture were used to accurately group individuals sharing a spatial proximity and/or season of capture. Notable differences in otolith edge signatures were found among individuals from north and south of the equator and contrasting cluster compositions from nearby sites in the Gulf of Aden of individuals captured in summer versus winter. Otolith core chemistry identified two spawning chemical compositions. The first common composition was characterized by relatively high concentrations of Sr and lower concentrations of P, Ba and Mg. The second less common spawning chemical composition was particularly rich in P, Ba and Mg and corresponded primarily to individuals caught off Mozambique, Somalia and Djibouti. These results are broadly consistent on one hand with patterns of water mass circulation in the Red Sea and western Indian Ocean and on the other hand with the observed spawning seasons. Though further research using, for example, archival tagging is needed to clarify the mechanisms behind these patterns, these results reveal the potential of otolith chemistry to provide insights into the spatio-temporal dynamics of narrow-barred Spanish mackerel. Fish otoliths encode information on surrounding environmental conditions. This study used otolith chemistry to investigate population structure and potential connectivity in stout whiting (Sillago robusta) along the east coast of Australia. Otoliths were analysed for both minor and trace elements, targeting the otolith core (location of birth) and otolith edge (location of capture), along with Ba:Ca transects to investigate life histories. Significant differences were found between core and edge signatures as well as among sample zones for select trace and minor elements. Otolith edge signatures showed evidence of some heterogeneity among zones, likely influenced by both environmental and physiological factors. This combination of factors is reflected in poor separation of sample zones from the multielement analysis, suggesting that future research may benefit from incorporating additional natural markers. Transects reflected some degree of individuality in Ba:Ca profiles, with evidence for potential exposure to upwelling events, or movement from shallow, inshore habitats into deeper waters. The variation in Ba:Ca profiles among fish suggests that populations are comprised of both resident and migratory individuals. While analysis of otolith chemistry could not reliably separate capture locations, patterns in the data suggest that inshore juvenile habitats are likely to be important in supporting exploited populations. Strontium marking technology is commonly used for extensive marking in stock enhancement and releasing activities. In order to explore the feasibility of this technology for juvenile Takifugu obscurus, five different concentrations of strontium (0, 12, 18, 36, and 72 mg/L) were set up by strontium chloride hexahydrate (SrCl2·6H2O) and T. obscurus were immersed for 7 days. Then, T. obscurus were reared in non-additive water for 25 days. The results showed that the survival rate of all groups was 100%, except the 72 mg/L group, after 7 days of immersion. Moreover, the growths of all the marked groups were better than that of the control group. There was no significant difference between the control and marked groups, except for the 18 mg/L group, indicating that the appropriate concentration of strontium might have positive effect for T. obscurus. The strontium to calcium (Sr/Ca) ratios of otoliths in the marked groups increased with increasing concentration and time, which were higher than the baseline, respectively. Then, the Sr/Ca ratios returned to the original level, but the peak value was still retained, suggesting that the success rate of the strontium marking reached 100%. Notably, the residual strontium content of 18 mg/L group was insufficient for causing negative effects for T. obscurus after 25 days. Therefore, in consideration of the growth, survival, and effect of strontium marking on otolith, 18 mg/L is an appropriate concentration for strontium marking of juvenile T. obscurus. This study confirmed the feasibility of strontium marking for T. obscurus and provided a new approach to rationally and scientifically evaluate the stock enhancement and releasing efficiency of T. obscurus. Background: The scales serve as an ideal model for studying the regulatory mechanism of bone homeostasis in fish. To explore the effect of salinity acclimation on bone metabolism of juvenile rainbow trout (Oncorhynchus mykiss), three sampling time points during salinity acclimation (7D, 14D and 21D) were selected to detect variations in histological characteristics. In the histological analysis, osteoblast marker enzymes alkaline phosphatase (ALP), osteoclast marker tartrate-resistant acid phosphatase (TRAcP) and calcium salt deposit areas (Von Kossa′s) were detected. Changes in calcium (Ca), phosphorus (P) and the molar mass ratio of calcium to phosphorus (Ca/P) in the scales were also detected by Inductively Coupled Plasma Mass Spectrometry (ICP-MS). In addition, the global MicroRNA (miRNA) expression profiles during salinity acclimation were examined using Illumina sequencing platform because of their important regulatory roles in teleost biological processes. Results: Twelve independent miRNA libraries were constructed, a total of 664 known and 92 putative novel miRNAs were identified. A total of 290 differentially expressed (DE) miRNAs were found in clusters with significant trends in the cluster analysis, and five types of clustering patterns were obtained; 22,374 DE predicted target genes of the aforementioned 290 DE miRNAs were obtained, 5957 of which clustered in six types of clustering patterns with a significant trend. To better understand the functions of the DE miRNAs, GO and KEGG analysis was performed on the 5957 target genes, as a result, they were significantly enriched in bone metabolism related signaling pathways such as MAPK signaling pathway, Calcium signaling pathway, Wnt signaling pathway, Mineral absorption and NF-kappa B signaling pathway. Six DE miRNAs were randomly selected and their expression were verified by quantitative real-time PCR (qRT-PCR), the expression trends were consistent with the results of transcriptome sequencing. Conclusions: The DE miRNAs and DE target genes identified in this study might play an important role in regulation of bone metabolism during salinity acclimation, relative genes or pathways could serve as key candidates for further studies to elucidate molecular mechanism of teleost bone metabolism, and help performing salinity acclimation and developing marine culture of salmonid species. Understanding biological and environmental factors that influence movement behaviors and population connectivity of highly migratory fishes is essential for cooperative international management and conservation of exploited populations, like bluefin tuna. Pacific bluefin tuna Thunnus orientalis (PBT) spawn in the western Pacific Ocean and then juveniles disperse to foraging grounds across the North Pacific. Several techniques have been used to characterize the distribution and movement of PBT, but few methods can provide complete records across ontogeny from larvae to adult in individual fish. Here, otolith biominerals of large PBT collected from the western, eastern, and south Pacific Ocean, were analyzed for a suite of trace elements across calcified/proteinaceous growth zones to investigate patterns across ontogeny. Three element:Ca ratios, Li:Ca, Mg:Ca, and Mn:Ca displayed enrichment in the otolith core, then decreased to low stable levels after age 1–2 years. Thermal and metabolic physiologies, common diets, or ambient water chemistry likely influenced otolith crystallization, protein content, and elemental incorporation in early life. Although similar patterns were also exhibited for otolith Sr:Ca, Ba:Ca and Zn:Ca in the first year, variability in these elements differed significantly after age-2 and in the otolith edges by capture region, suggesting ocean-specific environmental factors or growth-related physiologies affected otolith mineralization across ontogeny. Fish eye lenses are a protein‐based chronological recorder of microchemical constituents that are a potentially useful tool for interpretations of environmental, ecological and life‐history experienced by fish. Here, we present the first study with data on the chemical composition of eye lenses from Baltic cod examined using laser ablation inductively coupled plasma mass spectrometry (LA‐ICPMS) and compare these spatially resolved data to otoliths from the same fish also analysed by LA‐ICPMS, measuring the isotopes 27Al, 137Ba, 43Ca, 52Cr, 65Cu, 57Fe, 39K, 7Li, 25Mg, 55Mn, 31P, 208Pb, 85Rb, 45Sc, 29Si, 88Sr, 47Ti, 50V, 149Yb, 66Zn and 90Zr. Comparison of the variation in element concentrations between eye lenses and otoliths from the same individuals showed minor similarities, suggesting a different governance in the uptake processes. A strong overlap between the concentric growth rings in the eye lenses and the otolith Sr periodicity was observed, where each consecutive minima in the chemical profile with high accuracy correspond to the width of each lens ring. No comparable trends were seen between growth rings and all other elements measured from both lenses and otoliths. The characteristic rings observed in cod eye lenses do not seem to represent seasonal fluctuation nor are they found to be directly linked to age. With this research, we provide a baseline study identifying elements in corresponding eye lenses and otoliths that show potential for unravelling the environmental and biological conditions experienced by fish. The strontium-to-calcium ratio (Sr/Ca) in aragonitic shells and statoliths often depends on temperature, and has been investigated for its potential availability as a sclerochronological record of the temperatures experienced during the life of the animal. Only a few cephalopod species have been subjected to rearing experiments to validate the temperature dependence of statolith Sr/Ca despite the strong demand for estimating their historical ecology, particularly of migratory squid populations. In this context, we examined the relationship between temperature and statolith Sr/Ca in the spear squid, Heterololigo bleekeri , by maintaining wild-caught immature individuals at one of three constant temperatures: 12, 14 or 16°C. A portion of statolith precipitated during the experiment was optically identified by daily increment analysis, and then subjected to Sr/Ca measurement using an electron probe micro-analyser. Regression analysis (N = 29) demonstrated the dependence of statolith Sr/Ca on temperature and the relationship was estimated as an equation Y = 9.93 (±0.29) − 0.11 (±0.02) X. However, ~90% of the total residual variance was accounted for by the among-individual variation of statolith Sr/Ca within each temperature group. Consequently, the 95% confidence interval ranged over ± 7.85°C when the temperature was estimated by inserting a Sr/Ca value into this equation. The statolith Sr/Ca values are unlikely to provide reliable estimates for absolute temperatures, but it may allow reconstruction of a time-series of relative temperatures experienced by a particular individual. The otolith chemistry is one of the most commonly used tools to elucidate the structure of fish stocks. The correct application of this methodology depends on the fulfillment of several assumptions. One of the most relevant assumptions is that the otoliths and water chemistries vary at appropriate spatio-temporal scale in relation to the hypotheses being addressed. Due to the increase in the number of publications where this assumption is not properly considered, it is essential to discuss and order different concepts to contribute to the correct interpretation of the information provided by the otolith chemistry applied to the delimitation of stocks. Herein, the opportunity for dispersal concepts in otolith chemistry-based stock discrimination methods is discussed integrating the space-time scales, and biogeographic foundations. Additionally, a graphic guide was developed to facilitate hypothesis testing and their respective interpretations. This article encourages further use and attention on age and growth data, dispersal ability and life history, but further integration of the issue of biology versus management, as well as how the integration of other approaches may assist, is recommended, as most likely there will be no single approach to resolve stock structure. Simple Summary: Otolith geochemical signatures were important tools used to investigate the population of commercially exploited fish species. Historical and contemporary otolith samples of Isopisthus parvipinnis, Bigtooth corvina, an economically and ecologically important Brazilian fish species, collected in five subareas [São Paulo: North—NSP, Center—CSP and South—SSP; Paraná (PR) and Santa Catarina (SC)] of the shallow waters off the coast of the South Brazil Bight were used in this study. Univariate and multivariate statistical analyses showed spatial differences in otolith chemical composition over the years, suggesting that long-term temporal variability in oceanographic conditions, anthropogenic influence, and climate change on this coastal ecosystem influenced the geochemical signatures. Moreover, these results also confirm that I. parvipinnis is not a single and homogeneous fish stock in this geographic area, supporting the existence of a metapopulation structure scenario and corroborating previous studies that used alternative, complementary phenotypic tags. In this study, otolith geochemical signatures (Element:Ca ratios) were used to investigate the long-term spatial shifts of the population structure of Isopisthus parvipinnis, Bigtooth corvina, an economically and ecologically important Brazilian fish species. Two-hundred and ninety-seven juvenile individuals from historical (1975) and contemporary (2018/2019) samples were collected in five subareas [São Paulo: North—NSP, Center—CSP and South—SSP; Paraná (PR) and Santa Catarina (SC)] of the shallow waters off the coast of the South Brazil Bight were analyzed. The main informative single elements were Co:Ca, Cu:Ca, Li:Ca, Mg:Ca, Mn:Ca, Ni:Ca, Na:Ca, and Rb:Ca. Multivariate analysis showed spatial differences in otolith chemical composition over the years. Samples from 1975 presented an overall low reclassification rate (58%), suggesting the existence of two population units: (1) SP + PR; and (2) SC. However, samples from 2018/2019 discriminated four distinct population units with a good overall reclassification (80%): (1) NSP; (2) CSP; (3) SSP + PR; and (4) SC. This spatial differentiation on the geochemical signatures probably reflects the effects of long-term temporal variability in oceanographic conditions, anthropogenic influence, and climate change on this coastal ecosystem. The release of hatchery-reared fish fry for restocking is important for the enrichment of fishery resources; however, the effective evaluation of the success rate of marking such fish is challenging. We exposed juvenile crucian carp (Carassius carassius) to a single concentration of SrCl2·6H2O for 5 d and evaluated the efficiency of Sr marking of the fish otoliths (sagittae, asterisci, and lapilli) using an electron probe micro-analyzer. Sr marking signatures formed a peak in all otolith types, with a marking success rate of 100%. The ratio of Sr to Ca in the lapilli and sagittae was higher than that in the asterisci. It took 2 d from the beginning of immersion to the deposition of Sr on the lapilli and sagittae, and the time delay for asterisci was 1 d. For the lapilli and sagittae, it took 16 d to terminate Sr marking and fully recover to the pre-marking Sr level, whereas it was 12 d for the asterisci. The application of the Sr dose had no effect on the survival or growth of the carp. This study demonstrated that the lapilli are the most suitable otolith type for Sr marking observations in crucian carp and provides a theoretical basis and technical support for carp restocking using the Sr marking approach. Expansion of aquatic deoxygenation has altered the quality and availability of habitats and worsened body condition for many fish species through past decades. Identifying complementary chemical redox proxies in fish otoliths, in addition to Mn/Ca, would strengthen the ability to identify hypoxia exposure in a diversity of aquatic habitats. I/Ca ratios have been used in marine sediments and bio-mineralized materials for reconstruction of past redox conditions. In order to explore influences from various endogenous and exogenous processes, a large data set of I/Ca ratios from cores of otoliths, including fishes from fresh, estuarine, and coastal waters across 30 species within 10 taxonomic orders, were reported with other chemical proxies (Mn/Ca and Sr/Ca) using LA-ICP-MS. Our results suggest no single abiotic factor, including redox condition and salinity derived from Mn/Ca and Sr/Ca, predicts I/Ca values, while they may be correlated for specific fish species. Iodine may be related to organic matter in the cores of otoliths for some species. Maternal transfer, habitat change, dietary source, phylogeny, and ecology may also influence I/Ca ratios. Based on these exploratory results, we suggest a range of future research directions to further evaluate the factors controlling biomineralization of otolith iodine and its utility as a redox proxy. Longitudinal movement plays fundamental role in habitat colonization and population establishment of many riverine fish species. Movement patterns of amphidromous fish species at fine-scales that would allow characterizing the direction of movement and factors associated with the establishment of specific life-history strategies (resident or amphidromous) in rivers are still poorly understood. We assess fine-scale longitudinal movement variability patterns of facultative amphidromous fish species Galaxias maculatus in order to unfold its life-history variation and associated recruitment habitats. Specifically, we analyzed multi-elemental composition along core to edge transects in ear-bones (otoliths) of each fish using recursive partitions that divides the transect along signal discontinuities. Fine-scale movement assessment in five free-flowing river systems allowed us to identify movement direction and potential recruitment habitats. As such, resident recruitment of G. maculatus in freshwater (71%) and estuarine (24%) habitats was more frequent than amphidromous recruitment (5%), and was linked to availability of slow-flowing lotic or lentic habitats that produce or retain small-bodied prey consumed by their larvae. We postulate that life-history variation and successful recruitment of facultative amphidromous fish such as G. maculatus in river systems is driven by availability of suitable recruitment habitats and natural hydrologic connectivity that allows fish movement to these habitats. Acquisition of biological information on invasive fishes during the early stages of invasion could be critical in orienting subsequent management strategies. To achieve such a task, biological invasion researchers and practitioners take advantage of numerous technologies (e.g. genomics and acoustic telemetry). Surprisingly, the study of invasive fish ecology by analysis of the chemical composition of calcium carbonate hard parts (e.g. otoliths and scales) remains underutilized, despite some convincing examples of successful applications in the scientific literature. Among its most common applications in invasion biology, otolith chemistry has been used to identify natal origins, reconstruct migratory behaviour and assess mixed-stock structure. In this literature review, we provide a general overview of those previous applications but more importantly identify some gaps and obstacles to applications of otolith chemistry in invasion biology and suggest development for innovative applications, including use in wildlife forensic sciences and reconstruction of the early dynamics of invasions. Directional or stabilising selection should drive the expression of a dominant movement phenotype within a population. Widespread persistence of multiple movement phenotypes within wild populations, however, suggests that individuals that move (movers) and those that do not (residents) can have commensurate performance.The costs and benefits of mover and resident phenotypes remain poorly understood. Here, we explored how the presence and timing of movements are correlated with annual somatic growth rates, a useful proxy for performance because it is easily measured and rapidly reflects environmental changes.We used otolith growth measurements and stable isotope analyses to recreate growth and among‐reach movement histories of a partially migrating, long‐lived freshwater fish, golden perch Macquaria ambigua. We compared the association between movement and growth at two temporal scales: (a) short‐term (annual) differences in growth, in the years preceding, during or following movement; and (b) long‐term (lifetime) differences in growth.Overall, 59% of individuals performed at least one among‐reach movement, with these individuals subsequently more likely to move repeatedly throughout their lives. Movers grew faster than residents, with this difference most pronounced in the juvenile and early adult stages, when most movements occurred. Annual growth did not, however, change immediately prior to or following a specific movement event. Among‐individual variation in growth was initially higher for residents than for movers but decreased with age, at a faster rate for residents than for movers, such that levels conformed after 5 years of age.Our results indicate that lifetime movement is linked to faster growth in the early years of a fish's life. These faster growing movers are likely to be larger at a given age, leading to numerous potential benefits. However, the persistence of resident phenotypes suggests that there is likely a cost–benefit trade‐off to moving. The presence of multiple movement phenotypes may contribute to the resilience of populations by buffering against naturally and anthropogenically exacerbated environmental variability. Specific changes identified in the otolith macrostructure of Northeast Arctic cod as "spawning zones" are presumed to represent spawning events, but recent experimental studies have challenged this relationship. Because these zones are not routinely recorded outside of Norway, otoliths from multiple Atlantic cod populations with different life history and environmental traits were first examined to see if spawning zones could be identified as a general characteristic of cod. Then, a large archival collection of cod otoliths was used to investigate temporal changes in the occurrence of spawning zones and test for correlations between maturity at age derived from otolith spawning zones and gonad maturity stages. This study shows that spawning zones likely are a universal trait of Atlantic cod and not limited to certain environments or migratory behaviors as previously proposed. Maturity at age derived from spawning zone data showed trends consistent with those from gonad examinations. However, spawning zones appear to form with a one- or two-year lag with sexual maturity, which is suspected to reflect a stabilizing of energy partitioning after the first spawning events. Our results illustrate the potential for use of spawning zones, for example in species or populations with limited available maturity data, and highlights the need for addressing the physiological processes behind their formation. Otoliths can be used to examine the migration history of fish individuals based on trace element concentrations in them. In order to verify migration patterns of Clupanodon thrissa and Nematalosa nasus (Clupeiformes: Clupeidae), Sr:Ca ratios in otoliths of 13 individuals (6 C. thrissa and 7 N. nasus) collected from Vietnam were examined. In addition, to provide guidelines for identification, the otolith morphology of the two species was comparatively illustrated. The otolith Sr:Ca ratios from the core to the edge ranged from 0 to 5.7 × 10-3 and 0 to 4.9 × 10-3 for C. thrissa and N. nasus, with an average of 1.8 × 10-3 and 2.3 × 10-3, respectively. The present data do not show typical anadromous profiles as the Sr:Ca ratio of the two species mostly fluctuates evenly from 0 to 4 × 10-3. Nonindigenous lake trout (Salvelinus namaycush) expansion in Yellowstone Lake has led to a large decline in the native Yellowstone cutthroat trout (Oncorhynchus clarkii bouvieri) population. We assessed whether otolith microchemistry could be used to identify subbasin natal origins and long-term use by lake trout as a potential tool for optimizing removal efforts. 87Sr:86Sr and Sr:Ca ratios in otolith cores were used to assess natal origins and 87Sr:86Sr ratios in otolith transects were used to assess movement patterns. Water chemistry was similar throughout the lake, ranging from 0.70634 to 0.70642 and 3.94 to 4.38 mmol/mol for 87Sr:86Sr and Sr:Ca ratios, respectively. Lake trout otoliths also showed little variation in 87Sr:86Sr and Sr:Ca ratios of the natal region and 87Sr:86Sr across otolith transects. Thus, we found that microchemical differences among Sr isotope and Sr:Ca elemental ratios in otoliths were insufficient to detect the natal origin or extensive within-lake movement that has been established from telemetry investigations. Detailed analysis of other elements or isotopes in hard-part microchemistry, in combination with other tools for detecting movement, may improve detection of natal origin and life history movement among fishes within freshwater lentic systems. Element profiles of a 4-year-old, 290 mm long, male Baltic cod from thefirst validation collection, the DECODE samples, where age was obtained from patterns in daily otolith increment widths, including an image of the corresponding otolith viewed under reflected light. Profiles show relative element concentrations (element:Ca ratios divided by the profile mean for each element) from the TET mark to the otolith edge. While it is common knowledge that Red Drum (Sciaenops ocellatus) inhabit oligohaline waters (salinity <5), lifetime reconstructions of salinity histories have been lacking, and this study provides unique insight into interannual and ontogenetic patterns of oligohaline occupancy by this economically valuable sportfish. Growth consequences of oligohaline exposure and the relationship of oligohaline residency with river discharge were also investigated. Oligohaline exposure varied most during years 2 and 3 of life. During this time, 22% (n = 26/120 individuals) of Red Drum were oligohaline residents (≥ 90% of these years spent in oligohaline salinities), 34% (n = 41) were meso-polyhaline residents (< 10% of years 2 and 3 spent in oligohaline waters), and 44% (n = 53) spent time in both oligohaline and meso-polyhaline salinities. Trends in oligohaline residency match putative Red Drum life history. Oligohaline residents were present during years 1–6 of life; however, oligohaline residency peaked during the second year of life (n = 37, 31%) and by year 7 no oligohaline residents remained. Growth of oligohaline resident Red Drum during years 2–3 of life was lower than non-resident fish. However, long-term growth consequences of oligohaline residency were not apparent. The proportion of oligohaline residents during years 2 or 3 of life was positively related to river discharge. This divergence in salinity residency by juvenile Red Drum demonstrates that life history diversity was present in this population and that oligohaline waters provided important nursery habitat for fish that successfully recruited to the adult population.
