Objective: This research intended to compare histological impacts of various antioxidants in preventing doxorubicin-induced testicular toxicity and subsequent male infertility in rats. Study Design: Cross-sectional study. Place and Duration of Study: This study was conducted at the Animal House and Histopathology Laboratory of Peshawar Medical College, Peshawar, Pakistan from May 2023 to April 2024. Methods: An overall of 120 male rats were included in the research. The rats were divided into six groups: the control group, the doxorubicin only group, and four treatment groups receiving doxorubicin along with different antioxidants. The antioxidants administered were Vitamin C, Vitamin E, Coenzyme Q10, and Selenium. Histological analyses were performed on testicular tissues post-treatment to assess the extent of damage and protective effects of the antioxidants. Results: The doxorubicin only group showed statistically significant histological damage, including marked reductions in spermatogenesis and degeneration of seminiferous tubules. Antioxidant treated groups demonstrated significant protective effects, with the Selenium group exhibiting the highest level of protection, closely resembling the control group, followed by Vitamin E and Coenzyme Q10, which also provided substantial preservation of testicular structure. Conclusion: The study concluded that antioxidants, particularly Selenium, Vitamin E, and Coenzyme Q10, provided significant protection against doxorubicin induced testicular toxicity in rats. These findings suggested possible use of these antioxidants in mitigating male infertility associated with doxorubicin therapy. Objective: To determine the immunohistochemical expression of CK 13 and CK 17 in Oral Epithelial Dysplasia and Oral Squamous Cell Carcinoma. Methods: A total of 170 cases were retrieved from record files of Histopathology Department, to conduct a cross sectional study at Armed Forces Institute of Pathology, Rawalpindi, over a period of one year from June 2018 to June 2019 along with their formalin-fixed, paraffin embedded blocks comprising 85 cases of each oral squamous cell carcinoma and oral epithelial dysplasia. Blocks were trimmed and cut into very thin sections of 5 microns using microtome and mounted on slides. Tissue mounted slides were stained with routine haematoxylin and eosin followed by immunohistochemical staining of cytokeratin 13 and cytokeratin 17. New histological diagnosis of each case was made. Mean and standard deviation were calculated for quantitative variables. Frequency and percentages were calculated for qualitative variables. Objective: To determine the Immunohistochemical expression of BRAF V600E and Epidermal growth factor receptor (EGFR) in ameloblastoma and correlate the expression with age and gender of patients, and patterns and types of ameloblastoma. Material & Methods: 39 cases were retrieved with their formalin-fixed, paraffin embedded blocks, trimmed and cut into 5 microns sections. They were mounted on slides after staining with routine hematoxylin and eosin followed by Immunohistochemical staining of BRAF V600E and EGFR. Mean and standard deviation were calculated for quantitative variables. Frequency and percentages were calculated for qualitative variables. Chi-square test was employed to assess the significance of difference. The p-value ≤ 0.05 was considered significant. Results: There were 19 (41.2%) males and 20 (48.7%) female patients. The mean age of patients at which they presented was 39.97 ± 15.505 (mean ± SD) with an age range between 12 to 65 years. 25 (64.1 %) cases showed positive expression of BRAF V600E and 14 (35.8 %) cases showed negative expression of BRAF V600E. 27 (69.2 %) cases showed positive expression of EGFR whereas 6 (15.3 %) cases showed negative expression of EGFR. The p-value was ≤ 0.05 for expression of BRAF V600E with respect to patterns of ameloblastoma and tumor site and expression of EGFR with respect to sub-types of ameloblastoma. Conclusion: There is positive expression of BRAF V600E (64.1%) and EGFR (74.4%) in different sub-types and patterns of ameloblastoma. Correct assessment with the help of these markers can lead to early diagnosis and use of adjuvant treatment protocols

Objective: To check the expression pattern of cyclin-D1 in stage I head and neck squamous cell carcinoma (H.N.S.C.C.) cases (evaluated based on history, clinical, medical, and radiological records) in our setting and evaluate the role of cyclin-D1 as a prognostic marker of H.N.S.C.C. (considering recurrence within five years and metastasis). Study Design: Retrospective cohort study. Place and Duration of Study: The study was carried out at Armed Forces Institute of Pathology (A.F.I.P.)Rawalpindi working in conjunction with the Department of Radiation Oncology, Rawalpindi. The duration of study was from 7 March 2022-14 October 2022. Materials and Methods: The study comprised 92 patients from year 2015-16 of stage I diagnosed with squamous cell carcinoma of head and neck with complete five-year follow-up after fulfilling the inclusion and exclusion criteria. After collecting and matching the data on follow-up with record of the patients from Department of Radiation Oncology, C.M.H., Rawalpindi; paraffin-embedded blocks of these patients from year 2015/2016 were retrieved. Fresh Hematoxylin and Eosin (H&E) slides were prepared, and diagnosis was reconfirmed, vimentin stain was applied to check the antigenicity of the tissue followed by application of cyclin-D1 immuno-histochemical marker (I.H.C.) marker with controls to check the expression pattern. Results: In our study we included a total of 92 diagnosed cases of stage I H.N.S.C.C. with a complete 5-year follow-up. Out of 92 cases male to female ratio was almost 3:1 with males comprising a total of 68 (73.9%) and females comprising 24 (26.1%). Mean age of patients was 62.48+ 11.439 years. Larynx was the most frequently involved site 34.8% (n=32), followed by tongue 23.9% (n=22) and buccal mucosa 13.0% (n=12). Cyclin-D1expression and tumor grade were revealed to have a statistically significant clinicopathological relationship (p value 0.001). A statistically significant association between the expression of cyclin-D1 and tumor recurrence (p value < 0.001) and duration of tumor recurrence (p value 0.003) was seen. The cyclin-D1 expression was also compared with metastasis which was also statistically significant (p value 0.003). Conclusion: An increased risk of metastasis and recurrence is linked to increased levels of cyclin-D1 expression in the early years. Hence it is valuable to include cyclin-D1 expression in the initial diagnostic work up of H.N.S.C.C. for an early prognostic assessment. Further use of these results could be made for targeted treatment of head and neck malignancies.
