Abstract Background Megakaryocytic differentiation is a complicated process regulated by a series of transcription factors in a context- and stage-dependent manner. Recent studies have suggested that krüppel-like transcription factor 2 (KLF2) is involved in the control of embryonic erythroid precursor cell differentiation and maturation. However, the function and mechanism of KLF2 in regulating megakaryocytic differentiation remain unclear. Methods The expression patterns of krüppel-like transcription factors (KLFs) during megakaryocytic differentiation were identified from public databases. Phorbol 12-myristate 13-acetate (PMA) treatment of the myeloid-erythroid-leukemic cell lines K562 and HEL were used as cellular megakaryocytic differentiation models. A lentiviral transduction system was utilized to achieve the goal of amplifying or reducing KLF2. The expression of KLF2 was examined using real-time PCR and western blot. The impact of KLF2 on the megakaryocytic differentiation of K562 cells was examined by flow cytometry, Giemsa staining, Phalloidin staining and western blot. RNA-sequencing (RNA-seq) and chromatin immunoprecipitation-sequencing (ChIP-seq) technologies were used to identify the KLF2-regulated targets. Results KLF2 is increased in the maturation process of megakaryocytes. KLF2 overexpression accelerated the PMA-induced megakaryocytic differentiation, as reflected by an increased percentage of CD41/CD61 cells, an increased number of polyploid cells, and an elevated expression of P21 and P27. KLF2 knockdown exhibited the opposite results, indicating that KLF2 knockdown suppressed the megakaryocytic differentiation. Further, combination of the RNA-seq and ChIP-seq results suggested that chimerin 1 (CHN1) and potassium voltage-gated channel subfamily Q member 5 (KCNQ5) may be target genes regulated of KLF2. Both CHN1 and KCNQ5 knockdown could block the megakaryocytic differentiation to some content. Conclusion This study implicated a regulatory role of KLF2 in megakaryocytic differentiation, which may suggest KLF2 as a target for illness with abnormal megakaryocytic differentiation.

Cyclic loads generated by environmental factors, such as winds, waves, and trains, will likely lead to performance degradation in pile foundations, resulting in issues like permanent displacement accumulation and bearing capacity attenuation. This paper presents a semi-analytical solution for predicting the axial cyclic behavior of piles in sands. The solution relies on two enhanced nonlinear load-transfer models considering stress-strain hysteresis and cyclic degradation in the pile-soil interaction. Model parameters are calibrated through cyclic shear tests of the sand-steel interface and laboratory geotechnical testing of sands. A novel aspect involves the meticulous formulation of the shaft load-transfer function using an interface constitutive model, which inherently inherits the interface model's advantages, such as capturing hysteresis, hardening, degradation, and particle breakage. The semi-analytical solution is computed numerically using the matrix displacement method, and the calculated values are validated through model tests performed on non-displacement and displacement piles in sands. The results demonstrate that the predicted values show excellent agreement with the measured values for both the static and cyclic responses of piles in sands. The displacement pile response, including factors such as bearing capacity, mobilized shaft resistance, and convergence rate of permanent settlement, exhibit improvements compared to non-displacement piles attributed to the soil squeezing effect. This methodology presents an innovative analytical framework, allowing for integrating cyclic interface models into the theoretical investigation of pile responses.

Abstract Background Limited evidence exists regarding the link between platelet count and 30-day in-hospital mortality in acute respiratory failure (ARF) patients. Thus, this study aims to investigate this association among ICU patients experiencing acute respiratory failure. Methods We conducted a retrospective cohort study across multiple centers, utilizing data from the US eICU-CRD v2.0 database covering 22,262 patients with ARF in the ICU from 2014 to 2015. Our aim was to investigate the correlation between platelet count and 30-day in-hospital mortality using binary logistic regression, subgroup analyses, and smooth curve fitting. Results The 30-day in-hospital mortality rate was 19.73% (4393 out of 22,262), with a median platelet count of 213 × 109/L. After adjusting for covariates, our analysis revealed an inverse association between platelet count and 30-day in-hospital mortality (OR = 0.99, 95% CI 0.99, 0.99). Subgroup analyses supported the robustness of these findings. Furthermore, a nonlinear relationship was identified between platelet count and 30-day in-hospital mortality, with the inflection point at 120 × 109/L. Below the inflection point, the effect size (OR) was 0.89 (0.87, 0.91), indicating a significant association. However, beyond this point, the relationship was not statistically significant. Conclusion This study establishes a clear negative association between platelet count and 30-day in-hospital mortality among ICU patients with ARF. Furthermore, we have identified a nonlinear relationship with saturation effects, indicating that among ICU patients with acute respiratory failure, the lowest 30-day in-hospital mortality rate occurs when the baseline platelet count is approximately 120 × 109/L.

Abstract Phosphatase and tensin homolog deleted on chromosome 10 (PTEN) is a critical tumor suppressor protein that regulates various biological processes such as cell proliferation, apoptosis, and inflammatory responses by controlling the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (PI3K/AKT) signaling pathway. PTEN plays a crucial role in the pathogenesis of rheumatoid arthritis (RA). Loss of PTEN may contribute to survival, proliferation, and pro-inflammatory cytokine release of fibroblast-like synoviocytes (FLS). Also, persistent PI3K signaling increases myeloid cells’ osteoclastic potential, enhancing localized bone destruction. Recent studies have shown that the expression of PTEN protein in the synovial lining of RA patients with aggressive FLS is minimal. Experimental upregulation of PTEN protein expression could reduce the damage caused by RA. Nonetheless, a complete comprehension of aberrant PTEN drives RA progression and its interactions with other crucial molecules remains elusive. This review is dedicated to promoting a thorough understanding of the signaling mechanisms of aberrant PTEN in RA and aims to furnish pertinent theoretical support for forthcoming endeavors in both basic and clinical research within this domain.

Abstract Microbiota have an important function in shaping and priming neonatal immunity, although the cellular and molecular mechanisms underlying these effects remain obscure. Here we report that prenatal antibiotic exposure causes significant elevation of group 2 innate lymphoid cells (ILC2s) in neonatal lungs, in both cell numbers and functionality. Downregulation of type 1 interferon signaling in ILC2s due to diminished production of microbiota-derived butyrate represents the underlying mechanism. Mice lacking butyrate receptor GPR41 (Gpr41 -/- ) or type 1 interferon receptor IFNAR1 (Ifnar1 -/- ) recapitulate the phenotype of neonatal ILC2s upon maternal antibiotic exposure. Furthermore, prenatal antibiotic exposure induces epigenetic changes in ILC2s and has a long-lasting deteriorative effect on allergic airway inflammation in adult offspring. Prenatal supplementation of butyrate ameliorates airway inflammation in adult mice born to antibiotic-exposed dams. These observations demonstrate an essential role for the microbiota in the control of type 2 innate immunity at the neonatal stage, which suggests a therapeutic window for treating asthma in early life.

BackgroundEvidence of the relationship between platelet count and 30-day in-hospital mortality in ICU stroke patients is still scarce. Therefore, the purpose of this study was to explore the relationship between platelet count and 30-day in-hospital mortality among ICU stroke patients.MethodsWe conducted a multicenter retrospective cohort study using data from 8,029 ICU stroke patients in the US eICU-CRD v2.0 database from 2014 to 2015. Utilizing binary logistic regression, smooth curve fitting, and subgroup analyses, we examined the link between platelet count and 30-day in-hospital mortality.ResultsThe 30-day in-hospital mortality prevalence was 14.02%, and the mean platelet count of 223 × 109/L. Adjusting for covariates, our findings revealed an inverse association between platelet count and 30-day in-hospital mortality (OR = 0.975, 95% CI: 0.966, 0.984). Subgroup analyses supported the robustness of these results. Moreover, a nonlinear relationship was observed between platelet count and 30-day in-hospital mortality, with the inflection point at 163 × 109/L. On the left side of the inflection point, the effect size (OR) was 0.92 (0.89, 0.95), while on the right side, the relationship was not statistically significant.ConclusionThis study establishes an independent negative association between platelet count and 30-day in-hospital mortality in ICU stroke patients. Furthermore, a nonlinear relationship with a saturation effect was identified, suggesting that maintaining the platelet count around 163 × 109/L can reduce 30-day in-hospital mortality in these patients.

The impact of climate warming on soil microbial communities can significantly influence the global carbon cycle. Coastal wetlands, in particular, are susceptible to changes in soil microbial community structure due to climate warming and the presence of invasive plant species. However, there is limited knowledge about how native and invasive plant wetland soil microbes differ in their response to warming. In this study, we investigated the temporal dynamics of soil microbes (prokaryotes and fungi) under experimental warming in two coastal wetlands dominated by native Phragmites australis (P. australis) and invasive Spartina alterniflora (S. alterniflora). Our research indicated that short-term warming had minimal effects on microbial abundance, diversity, and composition. However, it did accelerate the succession of soil microbial communities, with potentially greater impacts on fungi than prokaryotes. Furthermore, in the S. alterniflora wetland, experimental warming notably increased the complexity and connectivity of the microbial networks. While in the P. australis wetland, it decreased these factors. Analysis of robustness showed that experimental warming stabilized the co-occurrence network of the microbial community in the P. australis wetland, but destabilized it in the S. alterniflora wetland. The dependable and efficient identification of Qin seal script characters is pivotal in the discovery, preservation, and inheritance of the distinctive cultural values embodied by these artifacts. This paper uses image histograms of oriented gradients (HOG) features and an SVM model to discuss a character recognition model for identifying partial and blurred Qin seal script characters. The model achieves accurate recognition on a small, imbalanced dataset. Firstly, a dataset of Qin seal script image samples is established, and Gaussian filtering is employed to remove image noise. Subsequently, the gamma transformation algorithm adjusts the image brightness and enhances the contrast between font structures and image backgrounds. After a series of preprocessing operations, the oriented gradient histograms (HOG) features are extracted from the images. During model training, different weights are assigned to classes with varying sample quantities to address the issue of class imbalance and improve the model's classification accuracy. Results show that the model achieves an accuracy of 95.30%. This research can help historians quickly identify and extract the text content on newly discovered Qin slip cultural relics, shortening the cycle of building a historical database.

Data-driven models perform poorly on part-of-speech tagging problems with the square Hmong language, a low-resource corpus. This paper designs a weight evaluation function to reduce the influence of unknown words. It proposes an improved harmony search algorithm utilizing the roulette and local evaluation strategies for handling the square Hmong part-of-speech tagging problem. The experiment shows that the average accuracy of the proposed model is 6%, 8% more than HMM and BiLSTM-CRF models, respectively. Meanwhile, the average F1 of the proposed model is also 6%, 3% more than HMM and BiLSTM-CRF models, respectively.

Statement of problem: Bibliometric analysis methods were used to evaluate pediatric dental sedation research and to identify topical hotspots using quantitative and qualitative methodologies. Purpose: To conduct bibliometric analysis on the retrieved data and to foresee the development of trends and hotspots in this research area. Material and methods: We retrieved appropriate research articles from the Web of Science Core Collection on January 1, 2023. VOSviewer, Citespace and the Bibliometrics website were used to conduct bibliometric analysis on the retrieved data. GraphPad Prism 10.0 (GraphPad, San Diego, CA, USA) was used to conduct the statistical analysis. Results: A total of 396 publications on pediatric sedation in dentistry, published between 1993 and 2022, were retrieved from online databases. The USA published most papers. Furthermore, the most frequent countries who cooperated were the USA and Canada. Six of the top ten publishing establishments were USA based. Papers on the research have appeared primarily in the journals of Dentistry and Anesthesiology. Keyword co-occurrence and co-citation cluster analysis revealed that the most common topics mainly were: dental anxiety; conscious sedation; dental caries; midazolam; propofol; hypoxemia. Conclusions: During the three decades, the focus of pediatric sedation research has been on drugs, dental anxiety and procedural sedation. Keyword burst detection indicated that procedural sedation; adverse event; respiratory depression is an emerging research hotspot.

ObjectiveHuman brucellosis causes serious public health concerns in Ningxia, China.MethodsThis study employed epidemiological, bacteriological, and multiple-locus variable-number tandem repeat analysis (MLVA) methods to conduct an epidemiological investigation, which is necessary for devising tailored control strategies.ResultsBetween 1958 and 2022, 29,892 cases were reported, with an average annual number of cases and incidence of 467 and 7.1/100,000, respectively. The epidemic situation gradually worsened, with cases escalating from 26 cases in 2005 to 6,292 in 2022, with the incidence rate rising from 0.441 in 2005 to 86.83 in 2022. Geographically, the disease spread from a single affected county in 2004 to encompass all 22 counties in 2022. Yanchi County had the highest incidence, followed by the Hongsibao and Tongxin counties. These data suggest that Brucella infection has become a rampant regional concern in human brucellosis. Between 1958 and 2019, a total of 230 Brucella strains were identified across four studied hosts. These strains comprised four species with 12 biovars, including B. melitensis bv. 1, bv. 2, bv. 3, B. abortus bv. 1, bv. 3, bv. 4, bv. 5, bv. 6, bv. 7, B. suis bv. 1 and bv. 3, and B. canis. These data highlight the high species/biovars and host diversity of the Brucella population, posing a substantial challenge to brucellosis surveillance. There was an apparent transition from multiple species/biovars historically to the current dominance of a single species, B. melitensis, emphasizing the requirement for strengthening surveillance of B. melitensis. Genotypes 42 and 116, constituting 96.2% of the total number of genotypes, predominated in panel 1 and MLVA-11, indicating that all strains belong to the East Mediterranean lineage. MLVA cluster analysis revealed persistent transmission of dominant circulating genotypes, presenting an epidemic pattern characterized primarily by epidemiologically related cases with a few sporadic cases. Strains in this study exhibited high genetic homogeneity with strains from the Northwest, and those from Kazakhstan and Mongolia.ConclusionThe epidemic situation of human brucellosis has gradually worsened; the rampant epidemic of the disease has become a regional concern. Carbon dots (CDs) have photoconversion performance and have a wide range of applications in agriculture. However, the solution of CDs is susceptible to quench under evaporation of sunlight and is easy to run-off when influenced by the external environment. Here, PVA-coated CDs were used to overcome the solid-state quenching and to prepare CDs microcapsules-polyurethane Film for plant cultivation. In this work, CDs microcapsules which can convert ultraviolet light into blue light for plant growth, were prepared by the “water/oil/water” method, and light-converting CDs microcapsules polyurethane film (CMPF) was prepared by doping CDs microcapsules with polyurethane. The study showed that polyurethane film has good mechanical properties and self-healing ability, the CMPF was able to effectively promote the growth of lettuce, and the effect was most significant at the CDs microcapsules concentration of 4000 mg/L. Compared with the blank group, the fresh and dry weights were increased by 177 % and 143.5 %, respectively, and the content of chlorophyll a, chlorophyll b, carotenoids, and soluble protein were increased by 14.5 %, 188.5 %, 43.3 %, and 17.9 %, respectively. Extracellular vesicles (EVs) contain the characteristics of their cell of origin and mediate cell-to-cell communication. Platelet-derived extracellular vesicles (PEVs) not only have procoagulant activity but also contain platelet-derived inflammatory factors (CD40L and mtDNA) that mediate inflammatory responses. Studies have shown that platelets are activated during storage to produce large amounts of PEVs, which may have implications for platelet transfusion therapy. Compared to platelets, PEVs have a longer storage time and greater procoagulant activity, making them an ideal alternative to platelets. This review describes the reasons and mechanisms by which PEVs may have a role in blood transfusion therapy.

IntroductionEpidemiological and clinical analyses of brucellosis are vital for public health leaders to reinforce disease surveillance and case management strategies.MethodsIn this study, we aimed to analyse the epidemiology and clinical features of 1,590 cases of human brucellosis.ResultsApproximately 72.08% (1,146) of the patients were male and 27.92% (444) were female. At least 88.18% (1,402/1,590) of the patients had a history of contact with sheep/goats and cattle, which was identified as the main risk factor for infection. The most common age group affected was 30–69 years, comprising 83.90% of all cases, with a median age of 47.3 years. Meanwhile, 75.03% (1,193/1,590) of the patients were farmers, followed by workers (10.50%, 167/1,590). The spectrum of clinical manifestations varied, and the major symptoms were fatigue (42.96%), joint pain (37.30%), and fever (23.33%). Arthritis was diagnosed in 989 patients, spondylitis was diagnosed in 469 patients, and external genital complications were found in at least 53.96% (858/1,590) of patients. In addition, approximately 41.25% (625/1,515) and 24.53% (390/1,590) of cases exhibited elevated CRP and D-dimer levels, respectively. Conversely, a significant decrease was observed in fibrinogen, total protein, and albumin levels, affecting 48.36% (769/1,590), 77.30% (1,226/1,586), and 91.80% (1,456/1,586) of the patients, respectively. These data demonstrate that brucellosis is a severe wasting disease that leads to an imbalance in nutritional metabolism and a decline in immunity. In total, 86.73% (1,379/1,590) of patients showed improvement with antibiotic therapy, while 13.27% (211/1,590) of patients experienced relapses or treatment failure.ConclusionBrucellosis often presents with non-specific symptoms and laboratory findings, accompanied by multiple organ invasions, as well as being a vital challenge for diagnosis and treatment; thus, it is essential for a high degree of suspicion to be placed on brucellosis for a timely diagnosis and treatment. This study provides basic data and resources for developing tailored countermeasures to curb its further spread.

The metastasis of cancer cells is the main cause of death in patients with gastric cancer (GC). Mounting evidence has demonstrated the vital importance of tumor-associated macrophages in promoting tumor invasion and metastasis; however, the interaction between tumor cells and macrophages in GC is largely unknown. In this study, we demonstrated that cyclase-associated protein 2 (CAP2) was upregulated in GC, especially in cases with lymph node metastasis, and was correlated with a poorer prognosis. The transcription factor JUN directly bound to the promoter region of CAP2 and activated CAP2 transcription. The N-terminal domain of CAP2 bound to the WD5 to WD7 domains of receptor for activated C kinase 1 (RACK1) and induced M2 macrophage polarization by activating the SRC/focal adhesion kinase (FAK)/ERK signaling pathway, which resulted in IL-4 and IL-10 secretion. Polarized M2 macrophages induced premetastatic niche formation and promoted GC metastasis by secreting TGFB1, which created a TGFB1/JUN/CAP2 positive-feedback loop to activate CAP2 expression continuously. Furthermore, we identified salvianolic acid B as an inhibitor of CAP2, which effectively inhibited GC cell invasion capabilities by suppressing the SRC/FAK/ERK signaling pathway. Our data suggest that CAP2, a key molecule mediating the interaction between GC cells and tumor-associated macrophages, may be a promising therapeutic target for suppressing tumor metastasis in GC.

China’s mountainous village relocation planning faces the problems of extensive survey ranges and high travel costs, and current planning still depends on traditional field visits and census data survey method. To improve efficiency and control costs, digital survey methods are proposed to support efficient data collection and produce data with accurate spatial reference . This research discusses a specific survey method for relocation planning using virtual imaging space modeling based on 3D GIS combined with neural network algorithm. A digital village model of a pilot village is established by oblique photography and WEBGIS technology, while the improved Mask R-CNN algorithm is adopted to identify and classify different buildings in the village. A relocation plan including the number of buildings in each class and the reclamation area is provided and the cost and value of the plan is fully evaluated. The results demonstrate that compared with the traditional survey method, the proposed method of digital survey can better address the issue of remote mountain areas, including provide more accurate, complete and sustainable information, make the surveying and planning more convenient, efficient and smart and improve the objectivity of village relocation planning.

Abstract In the treatment of spinal cord injury (SCI), the complex process of secondary injury is mainly responsible for preventing SCI repair or even exacerbating the injury. In this experiment, we constructed the 8-gingerol (8G)-loaded mesoporous polydopamine (M-PDA), M@8G, as the in vivo targeting nano-delivery platform, and investigated the therapeutic effects of M@8G in secondary SCI and its related mechanisms. The results indicated that M@8G could penetrate the blood-spinal cord barrier to enrich the spinal cord injury site. Mechanism research has shown that all of the M-PDA,8G and M@8G displayed the anti-lipid peroxidation effect, and then M@8G can inhibit the secondary SCI by suppressing the ferroptosis and inflammation. In vivo assays showed that M@8G significantly diminished the local injury area, reduced axonal and myelin loss, thus improving the neurological and motor recovery in rats. Based on the analysis of cerebrospinal fluid samples from patients, ferroptosis occurred locally in SCI and continued to progress in patients during the acute phase of SCI as well as the stage after their clinical surgery. This study showcases effective treatment of SCI through the aggregation and synergistic effect of M@8G in focal areas, providing a safe and promising strategy for the clinical treatment of SCI.

Background:Notopterygium incisum K.C. Ting ex H.T. Chang, a synonym of Hansenia weberbaueriana (Fedde ex H. Wolff) Pimenov & Kljuykov, is an anti-inflammatory medicinal plant. Although abrnotopterol has been reported to be its primary active metabolite, the other metabolites and their mechanisms of action remain unclear. This study aims to investigate the potential mechanisms by which its active metabolites treat Obstructive Sleep Apnea Syndrome (OSAS) through network analysis and experimental assessment.Methods: The metabolites and potential targets of Notopterygium incisum were extracted from public databases. We searched for OSAS-related genes in the Genecards, OMIM, PharmGkb, TTD, and DrugBank databases. Cytoscape 3.9.0 was used to construct the drug-target-disease network and screen for hub genes. Human bronchial epithelial (HBE) cells were cultivated in normoxia and chronic intermittent hypoxia (CIH) medium for 24 h. Interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), and prostaglandin E2 (PGE2) were quantified using enzyme-linked immunosorbent assay (ELISA). Prostaglandin-endoperoxide synthase 2(PTGS2) mRNA was detected using RT-qPCR, while PTGS2 and nuclear factor-kappa B (NF-κB) proteins were identified using Western blot analysis. Co-Immunoprecipitation (CoIP) and Western blotting were utilized to evaluate the ubiquitination of PTGS2 in HBE cells.Results: Pterostilbene and notopterol, isolated from Notopterygium incisum, had potential therapeutic effects on OSAS. The PTGS2 and estrogen receptor alpha (ESR1) hub genes were associated with OSAS. The pathway enrichment analysis focuses on the NF-κB, apoptosis, and HIF-1A pathways. In response to CIH, pterostilbene and notopterol decreased IL-6, TNF-α, and PGE2 levels. The NF-κB pathway was activated by an increase in PTGS2 levels. Pterostilbene promoted proteasome-mediated ubiquitination of PTGS2 protein and reduced PTGS2 levels, inhibiting the NF-κB pathway.Conclusion: This study reveals the active metabolites of Notopterygium incisum and hub genes involved in treating OSAS, which provide a basis for the follow-up development and exploitation of the botanical drug.

Abstract Background Self-repair of lower limb wounds has always been one of the research hotspots. Flaps and skin graft are the preferred treatment for lower extremity wound reconstruction. However, these treatments have many disadvantages, such as secondary damage, poor healing quality. In recent years, the use of acellular dermal matrix has emerged as an alternative treatment option for extremity ulcers. Methods This study aimed to explore whether acellular dermal matrix can be used as a single treatment to promote wound healing. 7 patients with lower extremities cutaneous deficiency exposing bone or tendon, were covered by Pelnac, which was an acellular dermal matrix product approved by China Food and Drug Administration. All the wound was treated by Pelnac without flaps and skin graft. The external dressing was changed every 10 days. Results After a maximum of 20 weeks, all the wounds were completely healed. During the 12 months follow-up period none of the patients developed skin wear on the treatment area. All patients maintained their postoperative ambulatory ability. All patients were satisfied with the appearance and feeling after wound healing. Conclusion These findings may mean acellular dermal matrix is a novel method offering opportunity for treatment of lower extremities cutaneous deficiency exposing bone or tendon. It also has the potential to close wounds of all uninfected, non-ischemic, full-thickness cutaneous deficiency.

Federated edge learning (FEL) has recently attracted great interest due to its real-time response and energy-efficient characteristics. Most existing work focuses on designing algorithms to improve model performance, ignoring the malicious behavior and personal decision-making of self-interested edge servers. Although some efforts have been devoted to incentivizing honest edge server engagement by compensating training costs, this rarely considers resource efficiency and often assumes that edge servers provide complete information to the platform, which may lead to the risk of private attribute leakage. Hence, we aim to achieve an incentive mechanism that promotes secure and efficient model training between the platform and edge servers. However, edge servers’ multi-dimensional private attributes and training strategies make the optimization problem nonconvex, and incomplete information further increases the complexity of the analysis. In order to address these challenges and by integrating contract theory and exponential mechanism, we propose an efficiency-aware incentive mechanism, FedNow, which enables edge servers to personally determine their local training rounds while motivating their participation without giving access to their true training strategies and private attributes. Specifically, we enabld edge servers to add noise to their submitted training strategy to hide their true training rounds; then, we carefully designed an efficiency score function to select honest and efficient edge servers without disclosing their private attributes. In order to demonstrate that FedNow strictly outperforms existing schemes in terms of total costs, we theoretically derived sufficient conditions for making the total costs of FedNow lower than existing schemes and designed a greedy algorithm that uses the Monte Carlo method to find feasible near-optimal solutions in polynomial time. Our extensive experimental assessment using synthetic and real datasets shows the superiority of FedNow.

A successful case of crossover replantation of the left foot to the stump of the right leg was described. The lower extremities were amputated at different levels. On the left side, there was a complete amputation in the distal part of the lower leg with comminuted fracture of the distal tibia. On the right side, there was a complete amputation in the hindfoot with comminuted fracture of both the talus and calcaneus. Since anatomical replantation was impossible, we performed crossover replantation of the left foot to the right lower leg. At the latest follow-up examination, forty-six months after the accident, the patient walked independently with a prosthesis on the stump of the left leg. Crossover replantation should be considered in bilateral amputations for the salvage of at least one extremity.

The microstructural evolution and corrosion behaviour of railroad flash-butt-welded U71Mn joints and the effect of heat treatment were investigated via scanning electron microscopy and electrochemical measurements. The joint structures were found to mainly comprise pearlite and a few ferrites. The grains became finer and more homogeneous after heat treatment. Additionally, there was a decrease in the corrosion current density (1.71 × 10−5 A cm−2) and increases in the absolute corrosion potential (0.86 mV) and corrosion resistance (1088.83 Ω−1cm2). This was primarily attributed to the fewer Cl− ions at the homogeneous grain boundaries and fewer oxidation reactions on the joints after heat treatment. The findings of this study explain corrosion failure and will guide the development of corrosion-resistant joints for improved railroad quality.

Abstract Stem cell-derived exosomes have recently been regarded as potential drugs for treating spinal cord injury (SCI) by reducing reactive oxygen species (ROS) and suppressing M1 macrophage polarization. However, the roles of ROS and exosomes in the process of M1 macrophage polarization are not known. Herein, we demonstrated that ROS can induce M1 macrophage polarization and have a concentration-dependent effect. ROS can induce M1 macrophage polarization through the MAPK-NFκB P65 signaling pathway. Dental pulp stem cell (DPSC)-derived exosomes can reduce macrophage M1 polarization through the ROS-MAPK-NFκB P65 signaling pathway in treating SCI. This study suggested that DPSC-derived exosomes might be a potential drug for treating SCI. Disruption of the cycle between ROS and M1 macrophage polarization might also be a potential effective treatment by reducing secondary damage. Graphical Abstract

Chronic intermittent hypoxia (CIH) induces oxidative stress in the brain, causing sleep disorders. Herein, we investigated the role of pterostilbene (Pte) in CIH-mediated oxidative stress in the brain tissue. A CIH mouse model was constructed by alternately reducing and increasing oxygen concentration in a sealed box containing the mouse; brain tissue and serum were then collected after intragastric administration of Pte. Neurological function was evaluated through field experiments. The trajectory of the CIH mice to the central region initially decreased and then increased after Pte intervention. Pte increased the number of neuronal Nissl bodies in the hippocampus of CIH mice, upregulated the protein levels of Bcl-2, occludin, and ZO-1 as well as the mRNA and protein levels of cAMP-response element binding protein (CREB) and p-BDNF, and reduced the number of neuronal apoptotic cells, Bax protein levels, IBA-1, and GFAP levels. Simultaneously, Pte reversed the decreased levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), and BDNF and increased levels of malondialdehyde (MDA) in the serum of CIH mice. Pte increased Th2 cells, Treg cells, IL-4, IL-10, and TGF-β1 levels and decreased Th1 cells, Th17 cells, IFN-γ, IL-6, and IL- 17A levels in activated BV2 cells and hippocampus in CIH mice. The protein levels of p-ERK1/2, TLR4, p-p38, p-p65, and Bax, apoptosis rate, MDA concentration, Bcl-2 protein level, cell viability, and SOD and GSH-PX concentrations decreased after the activation of BV2 cells. Pte inhibited gliocytes from activating T-cell immune imbalance through p-ERK signaling to alleviate oxidative stress injury in nerve cells.

ObjectiveThe burden of both general and drug-resistant tuberculosis in rural areas is higher than that in urban areas in China. To characterize the genetic structure and transmission risk of Mycobacterium tuberculosis in rural China, we used whole genome sequencing to analyze clinical strains collected from patients in two counties of Yichang for three consecutive years.MethodsFrom 2018 to 2020, sputum samples were collected for cultures from patients with suspected tuberculosis in Yidu and Zigui county, and DNA was extracted from the positive strains for genome sequencing. The online SAM-TB platform was used to identify the genotypes and drug resistance-related mutations of each strain, establish a phylogenetic tree, and calculated the genetic distances between pairwise strains. Twelve single nucleotide polymorphisms (SNPs) were used as thresholds to identify transmission clusters. The risk of related factors was estimated by univariable and multivariable logistic regression.ResultsA total of 161 out of the collected 231 positive strains were enrolled for analysis, excluding non-tuberculous mycobacterium and duplicate strains from the same patient. These strains belonged to Lineage 2 (92, 57.1%) and Lineage 4 (69, 42.9%), respectively. A total of 49 (30.4%) strains were detected with known drug resistance-related mutations, including 6 (3.7%) multidrug-resistant-TB (MDR-TB) strains and 11 (6.8%) RIF-resistant INH-susceptible TB (Rr-TB) strains. Six of the MDR/Rr-TB (35.3%) were also resistant to fluoroquinolones, which made them pre-extensively drug-resistant TB (pre-XDR-TB). There were another seven strains with mono-resistance to fluoroquinolones and one strain with resistance to both INH and fluoroquinolones, making the overall rate of fluoroquinolones resistance 8.7% (14/161). A total of 50 strains (31.1%) were identified as transmission clusters. Patients under 45 years old (adjusted odds ratio 3.46 [95% confidential intervals 1.28–9.35]), treatment-naive patients (6.14 [1.39–27.07]) and patients infected by lineage 4 strains (2.22 [1.00–4.91]) had a higher risk of transmission.ConclusionThe drug resistance of tuberculosis in rural China, especially to the second-line drug fluoroquinolones, is relatively serious. The standardized treatment for patients and the clinical use of fluoroquinolones warrant attention. At the same time, the recent transmission risk of tuberculosis is high, and rapid diagnosis and treatment management at the primary care needs to be strengthened.

Compared with quartz sand, recycled aggregate sand is characterized by the high liquefaction and shear resistance because of its irregular particle shape. However, the interrelationship between its particle shape and shear behaviors is seldom investigated. This paper investigates the role of the particle shape on the shear behaviors of recycled aggregate sand by conducting a series of monotonic and cyclic simple shear tests under undrained constant volume conditions. The particles’ morphology parameters are assessed in terms of sphericity, roundness and regularity with microscopic observation and image processing. According to the experimental results, the recycled aggregate sand with irregular shapes mobilizes greater gradients of the flow liquefaction lines under monotonic shear conditions. The irregularity of the particle shape also leads to greater peak shear stress and dynamic shear modulus, indicating stronger shear and liquefaction resistances under the cyclic shearing. Furthermore, the interrelationships between the peak shear stress, dynamic shear modulus and particle shape of the samples are revealed and established by well fitted functions, which could serve as references for the selection of recycled aggregate sand and corresponding designs of the foundation.

Background: The use of single cell protein (SCP) has become a method for alleviating the shortage of protein feed that microorganisms propagate in a suitable culture medium. In this study, SCP was produced by yeast to use the nutrition contained in the biogas slurry of chicken manure. Results: The results showed that Galactomyces candidum was the most efficient at producing SCP among the seven yeasts studied. The maximum cell dry weight (CDW) 6.79 g/L and protein content 39.39%, were obtained under the fermentation conditions of initial NH4+-N concentration of 2000 mg/L and a C/N ratio of 6:1 with acetate as the pH regulator. The total CDW increased to 9.24 g/L after secondary fermentation. Metal elements had a little effect on the growth of G. candidum. The addition of sulfur not only promoted the synthesis of sulfur-containing amino acid cysteine but also increased protein content by promoting the synthesis of glutamic acid and glutamine. Conclusions: Future experiments should focus more on achieving high-density cultivation and more efficient utilization of ammonia nitrogen in the biogas slurry.How to cite: Zhou Pan, Zhang Lu, Ding Hongxia, et al. Microneedles have garnered significant interest as transdermal drug delivery route owing to the advantages of nonselective loading capacity, minimal invasiveness, simple operation, and good biocompatibility. A number of therapeutics can be loaded into microneedles, including hydrophilic and hydrophobic small molecular drugs, and macromolecular drugs (proteins, mRNA, peptides, vaccines) for treatment of miscellaneous diseases. Microneedles feature with special benefits for cutaneous diseases owing to the direct transdermal delivery of therapeutics to the skin. This review mainly introduces microneedles fabricated with different technologies and transdermal delivery of various therapeutics for cutaneous diseases, such as psoriasis, atopic dermatitis, skin and soft tissue infection, superficial tumors, axillary hyperhidrosis, and plantar warts.

Objective: To evaluate the biological function of cervical subtotal discectomy prosthesis (CSDP) implantation in a non-human primate model.Methods: A CSDP was tested for cytocompatibility and osseointegration capacity before implantation in non-human primates. Subsequently, the CSDP was improved based on three-dimensional CT measurements of the non-human primate cervical spine. Eight cynomolgus monkeys were selected for removal of the intervertebral disc and lower endplate of the C5/6 segment to complete the model construction for CSDP implantation. In 18-month follow-up, physiological indices, radiology, and kinematics were assessed to estimate the biological function of the CSDP in non-human primates, including biosafety, osseointegration, and biomechanics.Results: Co-cultured with the CSDP constituent titanium alloy (Ti6Al4V-AO), the mouse embryo osteoblast precursor cell MC3T3-E1 obtained extended adhesion, remarkable viability status, and cell proliferation. After implantation in the mouse femur for 28 days, the surface of Ti6Al4V-AO was covered by a large amount of new cancellous bone, which formed further connections with the femur cortical bone, and no toxicity was detected by blood physiology indices or histopathology. After completing implantation in primate models, no infection or osteolysis was observed, nor was any subsidence or displacement of the CSDP observed in CT scans in the 18-month follow-up. In particular, the interior of the cervical vertebra fixation structure was gradually filled with new trabecular bone, and the CSDP had achieved fixation and bony fusion in the vertebral body at 1 year post-operation. Meanwhile, no signs of inflammation, spinal cord compression, adjacent segment degeneration, or force line changes were observed in subsequent MRI observations. Moreover, there were no pathological changes of the joint trajectory, joint motion range, stride length, or the stance phase ratio revealed in the kinematics analysis at 3, 6, 12, or 18 months after CSDP implantation.Conclusion: We successfully designed a new cervical subtotal discectomy prosthesis and constructed an excellent non-human primate implantation model for the evaluation of subtotal disc replacement arthroplasty. Furthermore, we demonstrated that CSDP had outstanding safety, osseointegration capacity, and biomechanical stability in a non-human primate model, which might be a new choice in the treatment of cervical disc diseases and potentially change future outcomes of degenerative cervical diseases.

A transition-metal-free method for the construction of 3-substituted or 3,4-disubstituted quinolines from readily available N,N-dimethyl enaminones and o-aminobenzyl alcohols is reported. The direct oxidative cyclocondensation reaction tolerates broad functional groups, allowing the efficient synthesis of various quinolines in moderate to excellent yields. The reaction involves a C (sp3)-O bond cleavage and a C=N bind and a C=C bond formation during the oxidative cyclization process, and the mechanism was proposed.

Dietary yeast culture supplementation can contribute to the performance and health of sows and piglets, but few studies have focused on the relationships between the effects of yeast culture and gut microbiota. This study investigated the effect of yeast culture (Saccharomyces cerevisiae) supplementation from late gestation to weaning on the reproductive performance of lactating sows and their faecal microbiota. One hundred and six purebred Landrace sows, of parities two to six were selected and randomly assigned to a control (CON) and yeast culture supplementation (YC) groups based on parity and back fat thickness. The YC sows were individually fed with yeast culture at a dose of 24 g/d from day 90 of gestation to parturition and 40 g/d during lactational period. Blood samples were collected from sows on d 110 of gestation and at weaning at day 21 of lactation for plasma hormone and immunoglobulin analysis. Colostrum and milk on day 20 of lactation were collected for composition analysis. Faecal samples from sows on day 110 of gestation and day 20 of lactation were collected for short-chain fatty acid and faecal microbial analysis. Results showed that the farrowing performance of YC sows did not differ significantly from the CON group (P > 0.05). The average daily feed intake by the YC group during the lactation period was significantly increased by 9.98% (P = 0.004), the weaning-to-oestrus interval was shortened by 0.96 d (P = 0.046) and average daily weight gain of piglets increased by 7.14% (P = 0.036) compared with the CON group. Aiming at the problem of sparse measurement points due to test conditions in engineering, a smoothing method based on zero-padding in the wavenumber domain is proposed to increase data density. Firstly, the principle of data extension and smoothing is introduced. The core idea of this principle is to extend the discrete data series by zero-padding in the wavenumber domain. The conversion between the spatial and wavenumber domains is achieved using the Discrete Fourier Transform (DFT) and the Inverse Discrete Fourier Transform (IDFT). Then, two sets of two-dimensional discrete random data are extended and smoothed, respectively, and the results verify the effectiveness and feasibility of the algorithm. The method can effectively increase the density of test data in engineering tests, achieve smoothing and extend the application to areas related to data processing.

Multilabel data share important features, including label imbalance, which has a significant influence on the performance of classifiers. Because of this problem, a widely used multilabel classification algorithm, the multilabel k-nearest neighbor (ML-kNN) algorithm, has poor performance on imbalanced multilabel data. To address this problem, this study proposes an improved ML-kNN algorithm based on value and weight. In this improved algorithm, labels are divided into minority and majority, and different strategies are adopted for different labels. By considering the label of latent information carried by the nearest neighbors, a value calculation method is proposed and used to directly classify majority labels. Additionally, to address the misclassification problem caused by a lack of nearest neighbor information for minority labels, weight calculation is proposed. The proposed weight calculation converts distance information with and without label sets in the nearest neighbors into weights. The experimental results on multilabel datasets from different benchmarks demonstrate the performance of the algorithm, especially for datasets with high imbalance. Different evaluation metrics show that the results are improved by approximately 2–10%. The verified algorithm could be applied to a multilabel classification of various fields involving label imbalance, such as drug molecule identification, building identification, and text categorization.

Multiple myeloma (MM) is a plasma cell malignancy that remains incurable. The protein tyrosine phosphatase SHP2 is a central node regulating RAS/mitogen-activated protein kinase (MAPK)/extracellular signal regulated kinase (ERK) signaling pathway which plays a crucial role in the pathogenesis and proteasome inhibitor (PI) resistance of MM. Several preclinical studies have demonstrated that SHP2 inhibitors exerted antitumor activity in cancer-harboring diverse mutations in the RAS pathway, offering the potential for targeting myeloma. In this study, we showed that pharmacological inhibition of SHP2 activity using SHP099 and RMC-4550 efficiently inhibited the proliferation of MM cells by inducing apoptosis and cell cycle arrest. As per the mechanism, SHP2 inhibitors activated the level of cleaved caspase3, BAK, and P21 and downregulated ERK phosphorylation in MM cells. Moreover, the blockade of SHP2 exhibited anti-myeloma effect in vivo in a mouse xenograft model. In addition, SHP2 inhibitors synergized the antineoplastic effect of bortezomib in bortezomib-sensitive MM cells and showed identical efficacy in targeting bortezomib-resistant MM cells. Overall, our findings suggest that SHP2-specific inhibitors trigger anti-myeloma activity in vitro and in vivo by regulating the ERK pathway and enhancing cytotoxicity of bortezomib, providing therapeutic benefits for both bortezomib naïve and resistant MM.

Acute promyelocytic leukemia (APL) usually presents with a series of coagulation-anticoagulation disturbance. Early administration of All-trans retinoic acid (ATRA) can reduce the risk of bleeding, but the potential for thrombosis needs to be addressed in some cases. The role of arsenic agent in correcting coagulation disorder remains to be studied, but oral arsenic agent shows potential advantages in coagulation recovery compared with intravenous agent, and chemotherapy can aggravate the progress of coagulation disease. In addition to early application of ATRA, avoiding invasive procedures and transfusion support can reduce the risk of bleeding. Whether the administration of heparin, thrombomodulin, recombinant factor VIIa or antifibrinolytics reduces the risk of bleeding and thrombosis associated with APL remains to be further explored, and their routine use outside of clinical trials is not recommended. This article reviews the effects of related drugs on coagulation-anticoagulation balance in APL patients.

Federated Learning (FL) has recently attracted considerable attention in internet of things, due to its capability of enabling mobile clients to collaboratively learn a global prediction model without sharing their privacy-sensitive data to the server. Despite its great potential, a main challenge of FL is that the training data are usually non-Independent, Identically Distributed (non-IID) on the clients, which may bring the biases in the model training and cause possible accuracy degradation. To address this issue, this paper aims to propose a novel FL algorithm to alleviate the accuracy degradation caused by non-IID data at clients. Firstly, we observe that the clients with different degrees of non-IID data present heterogeneous weight divergence with the clients owning IID data. Inspired by this, we utilize weight divergence to recognize the non-IID degrees of clients. Then, we propose an efficient FL algorithm, named CSFedAvg, in which the clients with lower degree of non-IID data will be chosen to train the models with higher frequency. Finally, we conduct simulations using publicly-available datasets to train deep neural networks. Simulation results show that the proposed FL algorithm improves the training performance compared with existing FL protocol.

Graph Neural Networks (GNNs) have been applied in many fields of semi-supervised node classification for non-Euclidean data. However, some GNNs cannot make good use of positive information brought by nodes which are far away from each central node for aggregation operations. These remote nodes with positive information can enhance the representation of the central node. Some GNNs also ignore rich structure information around each central node’s surroundings or entire network. Besides, most of GNNs have a fixed architecture and cannot change their components to adapt to different tasks. In this article, we propose a semi-supervised learning platform ATPGNN with three variable components to overcome the above shortcomings. This novel model can fully adapt to different tasks by changing its components and support inductive learning. The key idea is that we first create a high-order topology graph, which is from similarity of node structure information. Specifically, we reconstruct the relationships between nodes in a potential space obtained by network embedding in graph. Second, we introduce graph representation learning methods to extract representation information of remote nodes on the high-order topology graph. Third, we use some network embedding methods to get graph structure information of each node. Finally, we combine the representation information of remote nodes, graph structure information and feature for each node by attention mechanism, and apply them to learning node representation in graph. Extensive experiments on real attributed networks demonstrate the superiority of the proposed model against traditional GNNs.

N-Acylethanolamine acid amidase (NAAA) is a lysosomal enzyme responsible for the hydrolysis of fatty acid ethanolamides (FAEs). However, the role of NAAA in FAEs metabolism and regulation of pain and inflammation remains mostly unknown. Here, we generated NAAA-deficient (NAAA-/-) mice using CRISPR-Cas9 technique, and found that deletion of NAAA increased PEA and AEA levels in bone marrow (BM) and macrophages, and elevated AEA levels in lungs. Unexpectedly, genetic blockade of NAAA caused moderately effective anti-inflammatory effects in lipopolysaccharides (LPS)-induced acute lung injury (ALI), and poor analgesic effects in carrageenan-induced hyperalgesia and sciatic nerve injury (SNI)-induced mechanical allodynia. These data contrasted with acute (single dose) or chronic NAAA inhibition by F96, which produced marked anti-inflammation and analgesia in these models. BM chimera experiments indicated that these phenotypes were associated with the absence of NAAA in non-BM cells, whereas deletion of NAAA in BM or BM-derived cells in rodent models resulted in potent analgesic and anti-inflammatory phenotypes. When combined, current study suggested that genetic blockade of NAAA regulated FAEs metabolism and inflammatory responses in a cell-specifical manner.

This study investigates the impact of coronavirus disease 2019 (COVID-19) on economic development of China by measuring the HP financial index as an alternative variable of the digital economy. This study shows that economy of China developed further with the dissemination of COVID-19. Furthermore, the digital economy increased the level of economic development more prominently at the onset of COVID-19 pandemic. Moreover, an analysis of regional heterogeneity reveals that the eastern region maintained economic stability through its digital economy during COVID-19, while the central region improved its digital economy during COVID-19 pandemic. Although the economically underdeveloped western region has not suffered too seriously from COVID-19 pandemic, considering the sustained impact of disease and the uncertainty of its transmission speed, the region should vigorously develop its digital economy to manage public risk.

Mobile edge computing (MEC) brings a breakthrough for Internet of Things (IoT) for its ability of offloading tasks from user equipments (UEs) to nearby servers which have rich computation resource. 5G network brings a huge breakthrough on transmission rate. Together with MEC and 5G, both execution delay of tasks and time delay from downloading would be shorter and the quality of experience (QoE) of UEs can be improved. Considering practical conditions, the computation resource of an MEC server is finite to some extent. Therefore, how to prevent the abuse of MEC resource and further allocate the resource reasonably becomes a key point for an MEC system. In this paper, an MEC system with multi-user is considered where a base station (BS) with an MEC server, which can not only provide computation offloading service but also data cache service. Especially, we take the charge for both data transmission and task computation as one part of total cost of UEs, and then explore a joint optimization for downlink resource allocation, offloading decision and computation resource allocation to minimize the total cost in terms of the time delay and the charge to UEs. The proposed problem is formulated as a mixed integer programming (MIP) one which is NP-hard. Therefore, we decouple the original problem into two subproblems which are downlink resource allocation problem and joint offloading decision and computation resource allocation problem. Then we address these two subproblems by using convex and nonconvex optimization techniques, respectively. An iterative algorithm is proposed to obtain a suboptimal solution in polynomial time. Simulation results show that our proposed algorithm performs better than benchmark algorithms.

Reversible data hiding (RDH) methods are widely used in many privacy-sensitive real-time applications for digital images. As an efficient RDH method, prediction-error histogram (PEH) shifting technique has found wide application for its high efficiency in increasing embedding capacity. Nowadays, the performance of most PEH-based methods is evaluated only by Peak Signal to Noise Ratio (PSNR) or Structural Similarity (SSIM) Index. However, more and more applications require high quality in both PSNR and SSIM. Therefore, in this paper, we propose a new RDH method to concentrate on both PSNR and SSIM performance, which involves two key techniques: 1) an SSIM-based block selection technique, 2) a PEH-based optimal expansion bins selection technique. Through block selection, the original image is divided into two parts: smooth blocks and rough blocks. We select pixels of smooth block for double embedding and leave pixels of rough block unchanged. According to the optimal expansion bins, reasonable pixels are chosen for embedding such that the embedding distortion can be reduced. With these improvements, our proposed method has higher SSIM and PSNR after embedding compared to other PEH-based methods. The experimental results demonstrate its superiority over some state-of-the-art counterpart and other conventional PEH-based works.

Thrust disk as auxiliary impeller has a vital effect on the big power pump unit with wet motor. It can balance the axial force of the unit, provide power for internal cooling circulation circuit of motor, and further reduce the axial length of the unit. Due to the motor chamber space is limited, and the transmission medium is liquid which needs to consider the effect of cavitation, the research of thrust disk of auxiliary impeller is different from the previous studies of rotating channel. In this paper, the hydraulic performance and cavitation characteristics of thrust disk as auxiliary impeller are investigated experimentally under different conditions. A thrust disk test rig was established to obtain data under different rotation speeds and flow points. Three types of volutes with different outlet angles were designed to match the thrust disk as auxiliary impeller in experiments. Results indicate that the law of speed proportion for the traditional centrifugal pump is not applicable to the head of thrust disk as auxiliary impeller. In addition, the flow coverage increases slightly with rotation speed. Furthermore, the higher the speed is, the larger the NPSH s is, and the narrower the range of NPSH is. Besides, the smaller the outlet angle is, the higher the head of the thrust disk as auxiliary impeller is, the worse anti-cavitation performance is. The research could provide reference and guidance for the design of thrust disk as auxiliary impeller.

Humic acid (HA) in makeup water is one of the important safety issues of high-parameter power plants. Herein, the Zr-based metal organic frameworks (Zr-MOFs) were applied to remove humic acid in water. The mesoporous and active sites of Zr-MOFs were controlled by different ratios of ligands to increase the adsorption of HA. The maximum adsorption capacity was 150.15 mg g−1. The morphology and adsorption properties of the Zr-MOFs were characterized using scanning electron microscopy (SEM), X-ray diffraction (XRD), surface charge, Fourier Transform infrared (FT-IR), N2 adsorption-desorption and adsorption test. The adsorption process of HA accorded with the pseudo-second-order kinetics, while the adsorption isotherm conformed to the Langmuir model and the adsorption was proved to be a spontaneous and endothermic process. Physical adsorption by the mesoporous materials and the hydrogen bonding interactions between the Zr-MOFs and HA were the driving forces of HA adsorption. These results provided useful information for the effective removal of HA and enhanced our understanding of the adsorption mechanism of HA on Zr-MOFs.

Retinal organoids generated from human embryonic stem cells or iPSCs recreate the key structural and functional features of mammalian retinal tissue in vitro. However, the differences in the development of retinal organoids and normal retina in vivo are not well defined. Thus, in the present study, we analyzed the development of retinal organoids and zebrafish retina after inhibition of CXCR4, a key role in neurogenesis and optic nerve development, with the antagonist AMD3100. Our data indicated that CXCR4 was mainly expressed in ganglion cells in retinal organoids and was rarely expressed in amacrine or photoreceptor cells. AMD3100 treatment reduced the retinal organoid generation ratio, impaired differentiation, and induced morphological changes. Ganglion cells, amacrine cells, and photoreceptors were decreased and abnormal locations were observed in organoids treated with AMD3100. Neuronal axon outgrowth was also damaged in retinal organoids. Similarly, a decrease of ganglion cells, amacrine cells, and photoreceptors and the distribution of neural outgrowth was induced by AMD3100 treatment in zebrafish retina. However, abnormal photoreceptor ensembles induced by AMD3100 treatment in the organoids were not detected in zebrafish retina. Therefore, our study suggests that although retinal organoids might provide a reliable model for reproducing a retinal developmental model, there is a difference between the organoids and the retina in vivo.

The aerodynamic interference between the different components of quad-tiltrotor (QTR) aircraft were considered to analyze its influence on trim characteristics. A comprehensive method with the fixed-wake model was developed for multiple aerodynamic interactions, improving the accuracy of the flight dynamics analysis. Additionally, a more general control strategy was developed to tackle the redundant control issue of the QTR, improving its control efficiency by coordinating the authority relationship of various control surfaces across the flight range. Then, the trim features were calculated in the helicopter mode, conversion mode, and airplane mode, and the relevant results with and without interaction were compared. The results show that the aerodynamic interaction mainly influences the body’s vertical force, longitudinal force, and pitching moment. Furthermore, there are significant differences between collective and longitudinal sticks and pitch attitudes. The interference plays a major role in helicopter and conversion modes with a less-than-30-degree tilt angle.

Chemoradiotherapy and autoimmune disorder often lead to secondary thrombocytopenia in cancer patients, and thus, platelet transfusion is needed to stop or prevent bleeding. However, the effect of platelet transfusion remains controversial for the lack of agreement on transfusion strategies. Before being transfused, platelets are stored in blood banks, and their activation is usually stimulated. Increasing evidence shows activated platelets may promote metastasis and the proliferation of cancer cells, while cancer cells also induce platelet activation. Such a vicious cycle of interaction between activated platelets and cancer cells is harmful for the prognosis of cancer patients, which results in an increased tumor recurrence rate and decreased five-year survival rate. Therefore, it is important to explore platelet transfusion strategies, summarize mechanisms of interaction between platelets and tumor cells, and carefully evaluate the pros and cons of platelet transfusion for better treatment and prognosis for patients with cancer with secondary thrombocytopenia.

Abstract PRDM family proteins are dysregulated in many human diseases, especially hematological malignancies and solid cancers, and share a unique N‐terminal PR domain followed by zinc fingers toward the C terminus. With a high frequency of DNA promoter hypermethylation, PRDM5 is primarily considered as a tumor suppressor in solid tumors. However, little is known about the function of PRDM5 in blood malignancies, especially acute myeloid leukemia (AML). In this study, we showed that high PRDM5 expression levels were independently correlated with poor overall survival in AML patients. PRDM5 overexpression promoted cell proliferation, colony formation, and migration in vitro and enhanced tumorigenesis in an in vivo xenograft model. Furthermore, we found that PRDM5 overexpression promoted cell cycle progression with the decreased level of cell cycle inhibitors such as p16 and p21, and regulated the expression of epithelial‐mesenchymal transition markers ZO‐1 and Vimentin to promote migration. Moreover, we observed that PRDM5 upregulated the Jun N‐terminal kinase (JNK) signaling pathway and downregulated c‐Myc expression. Pharmacological inhibition of JNK by SP600125 partially abrogated PRDM5‐induced cell proliferation and migration. Taken together, our findings demonstrate that PRDM5 functions as an oncogenic driver in AML via JNK pathway, suggesting that PRDM5 is a potential therapeutic target for AML.

Finite battery capacity limits the network lifetime of wireless sensor networks, and thus severely impedes the deployment of large scale sensor networks. To prolong the lifetime, researchers utilize mobile chargers to recharge sensors with external power sources. In this paper, we study both periodic charging time scheduling and charging path planning with multiple chargers. First, we present an efficient slot-based periodic charging time scheduling algorithm with both a fine-grained node classification scheme to prevent unnecessary visits of energy-sufficient nodes, and a balanced charging task assignment scheme to avoid charging starvation. To further enhance charging efficiency, we also propose a charging path planning algorithm, which enables parallel power replenishment with multiple chargers. The simulation results show that our algorithms are effective and competitive when compared with existing algorithms.
