A rapid method based on pressurized liquid extraction followed by high-performance liquid chromatography coupled with evaporative light scattering detection was firstly developed for the quantitative analysis of two bioactive triterpenoids (acankoreoside A and acankoreagenin) in the leaves of Schefflera octophylla and Schefflera actinophylla. The analysis was performed on an Agilent Zorbax SB-Aq column (4.6 × 50 mm, 3.5 μm) with gradient elution of 0.1% formic acid and acetonitrile. Calibration curves of two analytes showed good linearity ( R2 > 0.9990) within the tested ranges. This novel method is simple, rapid and accurate, and the results of quantification showed that contents of each investigated compound is significant high in natural S. octophylla (6.36-14.83%), which indicated that natural S. octophylla as potential medicinal resource. Furthermore, hierarchical clustering analysis based on the typical peaks of acankoreoside A and acankoreagenin from the 17 tested samples showed that natural and cultured Schefflera species were in different clusters, which could provide a means of discriminating between Schefflera species from different origins. A high-performance anion-exchange chromatography coupled with diode array detection method was developed for the determination of dencichine in Panax notoginseng and related species. The analysis was performed on an Eprogen Synchropak WAX column (4.6 × 250 mm, 6 μm) with 50 mM NaH2PO4 aqueous solution isocratic elution. The method was validated in terms of linearity, sensitivity, precision, stability, and accuracy. It was found that the calibration curve for dencichine showed good linearity ( R2 = 0.9999) within the test range. The LOD and LOQ were 0.77 and 3.06 ng, respectively. The RSD for intra- and interday repeatability was 0.2 and 0.5%, respectively. The test solution of dencichine is stable at least for three days at room temperature and for seven days at 4°C. The mean recovery of dencichine was 102.0%. The established method was successfully applied to determine dencichine in the raw root of P. nogoginseng, P. ginseng, and P. quinquefolium as well as the steamed root of P. notoginseng. Introduction - Aconites, with aconite alkaloids as the major therapeutic and toxic components, are used for the treatment of analgesic, antirheumatic and neurological symptoms. Quantification of the aconite alkaloids is important for the quality control of aconite-containing drugs. Objective - To establish a validated capillary zone electrophoresis (CZE) method for the simultaneous determination of six major alkaloids, namely aconitine, mesaconitine, hypaconitine, benzoylaconine, benzoylmesaconine and benzoylhypaconine, in crude and processed aconite roots. Methodology - The CZE method was optimised and validated using a stability-indicating method. The optimised running buffer was a mixture of 200 m m Tris, 150 m m perchloric acid and 40% 1,4-dioxane (pH 7.8) with the capillary thermostated at 25°C. Results - Using the optimised method, six aconite alkaloids were well separated. The established method showed good precision, accuracy and recovery. Contents of these alkaloids in crude and processed aconites were determined and it was observed that the levels of individual alkaloids varied between samples. Conclusion - The developed CZE method was reliable for the quality control of aconites contained in herbal medicines. The method could also be used as an approach for toxicological studies. The potential anti-tumor activity of gamboges, a herbal medicine derived from Garcinia hanburyi, has increasingly gained the interest of scientist worldwide. The major components of gamboges are cytotoxic caged xanthones. In the present study, an improved HPLC method was developed to simultaneously quantify 12 caged xanthones, including three pairs of epimers and four pairs of trans- cis isomers, i.e. forbesione, isomorellic acid, morellic acid, R-30-hydroxygambogic acid, S-30-hydroxygambogic acid, isogambogenic acid, gambogenic acid, gambogellic acid, R-isogambogic acid, S-isogambogic acid, R-gambogic acid and S-gambogic acid. This method was validated to be sensitive, precise and accurate with limits of detection of 0.03-0.08 µg/mL, overall intra-day and inter-day variations less than 7.9% and overall recovery over 93.2%. The correlation coefficients ( r2) of the calibration curves were higher than 0.995 for all analytes. The newly established method was successfully applied to reveal the difference in the chemical profiles and contents of these analytes in gamboges from different origins. It can be concluded that this method was not only an effective quality control method to ensure the safety and efficacy consistency of gamboges, but also a useful tool for screening and determining more potent cytotoxic xanthones with potential anticancer activity. Abstract: A recycling counter-current chromatographic system was first set up with a high-speed counter-current chromatography instrument coupled with a column switching valve. This system was first successfully applied to the preparative separation of epimers, gambogic acid and epigambogic acid from Garcinia hanburyi using n-hexane–methanol–water (5:4:1, v/v/v) as the two-phase solvent system. As a result, 28.2mg gambogic acid and 18.4mg epigambogic acid were separated from 50mg of mixture. Their purities were both above 97% as determined by HPLC. Abstract: Four novel polyisoprenylated benzophenone derivatives, paucinones A–D (1–4), were isolated from the leaves of the plant Garcinia paucinervis. Paucinones A–C (1–3) contained an unexpected cyclohexane-spiro-tetrahydrofuran moiety. A 1-methylene-3,3-dimethylcyclohexane group never reported before was found in the structure of paucinone D (4). The structures of these compounds were elucidated with spectroscopic evidences. The relative stereochemistries of 1–4 were determined by NOESY correlations. These compounds showed significant cytotoxicities against HeLa cells. [Copyright &y& Elsevier]

Abstract: A simple, rapid, reverse-phase high performance liquid chromatographic method was developed for the quantitative analysis of pseudolaric acids in Cortex Pseudolaricis and its related medicinal products. With a C18 analytical column (4.6mm×150mm i.d.), five pseudolaric acids, namely pseudolaric acids A–C, pseudolaric acid A-O-β-d-glucopyranoside and pseudolaric acid B-O-β-d-glucopyranoside, were well separated within 7min. Acetonitrile and 0.10% acetic acid were used as the mobile phase in a gradient program. The UV detection wavelength was set at 260nm. The detection limits and quantification limits ranged in 8.26–16.66ng/ml and 27.54–55.53ng/ml, respectively. The intra- and inter-day variations were less than 1% for all five compounds. The recovery of all spiked pseudolaric acids ranged from 99.1% to 101.9%. Compared to existing analytical methods, this new method not only used two more important chemical markers but also provided a fivefold reduction in analysis time. In addition, the extraction method of herb sample was also modified by an orthogonal array experiment on three variable parameters: extraction time, solvent volume, and extraction cycles. The optimized extraction method was much simpler and could be efficiently used to analyse large set of herbal materials and related medicinal products. Nineteen herb samples collected from different regions of China and five related products were examined with this new analytical method.
