Background: Avian trichomoniasis is an important disease that causes bird mortality, both wild and captive, around the world. This study evaluated the in vitro cytotoxicity, antioxidant, and antiparasitic activity of citral (3.7-Dimetil-2.6-octadienal) and geraniol (trans-3.7-Dimetil-2.6-octadien-1-ol) against Trichomonas gallinae trophozoites. Methods: In vitro assays were conducted at the Laboratory of Protozoology and Entomology (LAPEN) at the Federal University of Pelotas (UFPel), Brazil in 2019 using tests with 106 parasites and citral and geraniol at concentrations ranging from 10 to 100 μM and four controls: NC (culture medium and trophozoites), MTZ (trophozoites plus 100 μM of metronidazole), and TW (trophozoites plus vehicles used for solubilizing derivatives (0.01% Tween). Results: The citral (60 μM) and geraniol (50 μM) concentrations reduced the trophozoitesʼs viability by 100%. The molecular docking experiment demonstrated that citral and geraniol might inhibit a hydrogen enzyme for T. gallinae survival. Conclusion: The major compounds of lemongrass have potential antitrichomonal activity against T. gallinae in vitro.

Introduction: Echinococcus granulosus is the causative agent of hydatid cysts, which are distributed worldwide. Nowadays, much research is being done to inactivate hydatid cyst protoscoleces. This study was conducted with the aim to compare the scolicidal effect of two chemical compounds 12 and 2 on hydatid cyst protoscoleces in vitro. Material & Methods: The scolicidal activity of two amini-hydrox-phenylcoumarin and amini-bromo-phenylcoumarin at concentrations of 1.2, 1.5, 1.10 and 1.100 after 15, 30 and 60 minutes of incubation was tested separately and each of the experiments was repeated three times. The viability of protoscoleces was checked with 0.1% eosin dye under a light microscope. Subsequently data were analyzed using GraphPad version 5 software and two-way repeated measures ANOVA and values (P > 0.05) was considered a significant level. Findings: The results of this study showed that the chemical compound amini-bromophenylcoumarin at a concentration of 1.10 destroyed 100% of protoscoleces after 60 minutes of exposure compared to the control group, but amini-hydroxophenyl-coumarin, at a concentration of 1.100 could destroy 98% of protoscolecia after 60 minutes of exposure. Discussion & Conclusion: The results of the present study showed that amini-bromophenylcoumarin had stronger protoscolicidal effects than amini-hydroxophenyl-coumarin. However, further studies and in vivo studies are needed to evaluate the efficacy of these chemical compounds.

Background Propolis is a natural resinous product from a variety of plants and combines it with beeswax and salivary enzymes to create bee glue. Its potentials have been employed in the treatment of many diseases and conditions for centuries. This study, therefore, aims to determine the anti-Acanthamoeba properties of the propolis extract, eye drops coupled to some mechanisms such as inhibition of encystation and excystation. Methods The minimum inhibitory concentrations (MICs) of the most active propolis extract were assessed in trophozoites and cysts of Acanthamoeba triangularis (A. triangularis) at 0.256 and 1 mg/mL, respectively. Results Active eye drops inhibited trophozoites and cysts with a MIC value of 100%. At concentrations lower than their MICs values (1/2–1/16 MIC), propolis and eye drops revealed inhibition of encystation. In addition, at 1/2 MIC, both the propolis extract and eye drops showed potential inhibition of excystation. In combined sets of propolis extract and eye drops, they showed slightly increasing inhibition of encystation and excystation. Surprisingly, the MIC of chlorhexidine decreased when combined with the propolis and eye drops. SEM and TEM images displayed morphological changes in A. triangularis treated with combined sets of propolis extract and eye drops. Conclusion From this study, a new combined eye drop solution with propolis extract was found to be helpful in reducing encystation and excystation in A. triangularis. Therefore, this combination is an alternative for the treatment of eye diseases in early stages along with reducing the entry into the cyst stage of Acanthamoeba. The results of this study revealed new anti-Acanthamoeba inhibitors with promising combined alternative therapeutic potential for the treatment of Acanthamoeba keratitis.

Background: Trichomonas vaginalis, a parasitic flagellated protozoan, is one of the main non-viral sexually transmitted diseases worldwide. Treatment options for trichomoniasis are limited to nitroimidazole compounds. However, resistance to these drugs has been reported, which requires the development of new anti-Trichomonas agents that confer suitable efficacy and less toxicity. Methods: In the present work, we assessed the effectiveness of the liposomal system containing essential oils of Bunium persicum and Trachyspermum ammi against T. vaginalis in vitro. The chemical composition of B. persicum and T. ammi were analyzed using gas chromatography-mass spectrometry (GC–MS). Liposomal vesicles were prepared with phosphatidylcholine) 70%) and cholesterol)30%) using the thin-film method. The essential oils of B. persicum and T. ammi were loaded into the liposomes using the inactive loading method. Liposomal vesicles were made for two plants separately. Their physicochemical features were tested using Zeta-Sizer, AFM and SEM. The anti-Trichomonas activity was determined after 12 and 24 h of parasite cultures in TYI-S-33 medium. Results: After 12 and 24 h of administration, the IC50 of the B. persicum essential oil nano-liposomes induced 14.41 μg/mL and 45.19 μg/mL, respectively. The IC50 of T. ammi essential oil nano-liposomes induced 8.08 μg/mL and 25.81 μg/mL, respectively. Conclusions: These data suggested that nano-liposomes of the essential oils of B. persicum and T. ammi may be a promising alternative to current treatments for Trichomonas infection.

Background: Nitrite and nitrate salts are used as preservatives in meat products to prevent the growth of pathogen organisms such as Clostridium botulinum. Nitrogen salts additives create the desired color in meat products. However, the use of these salts accelerates nitrosamine production and increases the risk of carcinogenesis. Hence, nitrate and nitrite consumption rate should be according to the approved standards. This analytical study is designed to determine nitrate and nitrite residue of non-heat-treated meat products including hamburger and kebab in East Azerbaijan Province, Iran during 2015-2020. Methods: 120 samples of non-heat-treated meat products from different brands were randomly collected. They consisted of 60 products manufactured in East Azerbaijan province and 60 products imported from other provinces. Nitrate and Nitrite residues in non-heat-treated meat products including hamburger and kebab were determined by spectrophotometric method. Results: Results showed that the residue levels of nitrate and nitrite content regarding non-heat- treated meat products were less than maximum residual limits (MRLs) of national standards. The study also indicated that frozen non-heated meat products did not have any nitrogenous additive. Conclusion: The results of this research revealed that the frozen non-heat treated meat products including hamburger and kebab, produced or consumed in East Azerbaijan province during 2015-2020 did not have nitrate and nitrite additives. Continuous monitoring to ensure the compliance of meat products with the standards is necessary.

Cystic echinococcosis (CE) is a zoonotic disease and with has a global distribution. Today, much research carries out to inactivate hydatid cyst protoscoleces. In particular, herbal compounds have received more attention due to their cheapness, easy access, low toxicity, and side effects. This study aimed to compare the scolicidal effect of hydroalcoholic extract of Allium sativum (garlic) and Ferula asafoetida (angozeh) on hydatid cyst protoscoleces in vitro. The scolicidal activity of A. sativum and F. asafoetida extracts were evaluated at concentrations of 50, 100, 150, 200, and 250 mg/ml following 10, 30, and 60 minutes of exposure. Each reaction was repeated three times. The viability of protoscoleces was examined with a 0.1% eosin stain under a light microscope. The chemical composition of two extracts was analyzed using Gas ChromatographyMass Spectrometry (GC–MS). Statistical analyses were performed with GraphPad software version 5.0. The results of this study showed that F. asafoetida extract, at a concentration of 250 mg/ml after 60 minutes of exposure, killed 100% of the protoscoleces compared to the control group, but the hydroalcoholic extract of A. sativum at the same concentration and time, it was able to kill 98% of protoscoleces. The main chemical components of A. sativum and F. asafoetida identified as allyl methyl trisulfide (12.8%) and methyl ester (13.9%), respectively. The findings of the present study showed that F. asafoetida has more potent scolicidal effects than A. sativum. However, further studies are needed to evaluate the effectiveness of the F. asafoetida plant.

Background and Objectives: Cutaneous leishmaniasis is one of the most important health-threatening diseases in Iran and other countries. Glucantime is currently used to treat this disease, but due to its side effects and high resistance, alternative therapies such as the use of nanoparticles have been considered by researchers. This study aims to investigate the anti-leishmania activity of silver nanoparticles on Leishmania major in vitro. Methods: This is an experimental study on the anti-leishmania activities of silver nanoparticles at different concentrations of 0.75-0.96 µg/ml after 24, 48 and 72 hours of exposure to 106 live Leishmania major promastigotes. The numbers of live parasites were counted by Trypan Blue on a neobar slide using optical microscope (Hemocytometer method). Glucantime and distilled water were considered as positive and negative controls, respectively. The half-maximal inhibitory concentration (IC50) was calculated by SigmaPlot™ software, v‌ersion 13. All reactions were done three times and their average was considered as final result. Results: All concentrations of silver nanoparticles had anti-leishmania activity, where the concentration of 96 µg/ml had the highest effect (100%) 72 hours after exposure. The IC50 was obtained 36.67, 27.2 and 21.08 µg/ml after 24, 48 and 72 hours of exposure, respectively. Conclusion: Silver nanoparticles have an inhibitory effect on the growth of Leishmania major in different concentrations. However, further in-vivo studies are needed to determine the effictivness of silver nanoparticles.

The aims of this study to assess the efficiency of AGL against acetaminophen (APAP)-induced hepatic toxicity that was generated by mitochondrial oxidative stress and glutathione depletion. Free radical scavenging potentiality was analyzed by using 2, 2-diphenyl-1-picrylhydrazyl (DPPH), hydrogen peroxide, nitric oxide, and hydroxyl radical scavenging assays. APAP-induced liver toxicity was formed at a dose level of 640 mg/kg mg/kg BW each, p.o. for 14 days for all experimental rats except the vehicle control group. AGL (5 and 10 mg/kg) were treated orally with negative control and negative control silymarin (50 mg/kg) group. To assess the protective effect, we looked at the levels of serum biochemical markers, liver histoarchitecture, and hepatic antioxidant enzyme activity. AGL showed in vitro anti-oxidant potentialities by scavenging radicals in the respective assays. As evidenced by serum biochemical indicators and relative liver weight, AGL co-administration substantially reduced toxicant-induced hepatic damage. APAP-intoxication increased the malondialdehyde (MDA) level and declined in cellular endogenous antioxidant enzymes such as reduced catalase, superoxide dismutase, and glutathione, where, AGL treatment amended their level. In the same way, histopathological evaluation further verified that AGL protected the hepatocyte from APAP-induced damage. As AGL scavenges toxic free radicals, thereby protects mitochondria and other organelles from reactive oxygen and nitrogen species-mediated stress and its eventual consequence necrosis. Therefore, we propose the hepatoprotective activity of AGL through its antioxidant mechanism.

Background: Propolis is a natural resinous mixture produced by bees. It provides beneficial effects on human health in the treatment/management of many diseases. The present study was performed to demonstrate the anti-Acanthamoeba activity of ethanolic extracts of Propolis samples from Iran. The interactions of the compounds and essential proteins of Acanthamoeba were also visualized through docking simulation. Methods: The minimal inhibitory concentrations (MICs) of Propolis extract against Acanthamoeba trophozoites and cysts was determined in vitro. In addition, two-fold dilutions of each of agents were tested for encystment, excystment and adhesion inhibitions. Three major compounds of Propolis extract such as chrysin, tectochrysin and pinocembrin have been selected in molecular docking approach to predict the compounds that might be responsible for encystment, excystment and adhesion inhibitions of A. castellanii. Furthermore, to confirm the docking results, molecular dynamics (MD) simulations were also carried out for the most promising two ligand-pocket complexes from docking studies. Results: The minimal inhibitory concentrations (MICs) 62.5 and 125 µg/mL of the most active Propolis extract were assessed in trophozoites stage of Acanthamoeba castellanii ATCC30010 and ATCC50739, respectively. At concentrations lower than their MICs values (1/16 MIC), Propolis extract revealed inhibition of encystation. However, at 1/2 MIC, it showed a potential inhibition of excystation and anti-adhesion. The molecular docking and dynamic simulation revealed the potential capability of Pinocembrin to form hydrogen bonds with A. castellanii Sir2 family protein (AcSir2), an encystation protein of high relevance for this process in Acanthamoeba. Conclusions: The results provided a candidate for the development of therapeutic drugs against Acanthamoeba infection. In vivo experiments and clinical trials are necessary to support this claim.

Due to its high side effects and resistance, the use of alternative therapies, especially plants and natural compounds, has been highly recommended by researchers. The aim of this study is to investigate the effect of the methanolic extract of dandelion root (Taraxacum officinale) on Leishmania major in vitro techniques. Methods: In this experimental study, the extract was prepared by the Soxhlet method and to evaluate the effect of methanolic extract of dandelion root on L. major parasite at concentrations of 300-1200 mg/ml. Amphotericin B and distilled water were considered as positive and negative controls, respectively. Then, 106 live parasites were added to all tubes, and all groups were kept at 25 ±1 °C. At 24, 48, and 72 hours, the number of live parasites was counted by Trypan Blue using a neobar slide and light microscope (Hemocytometer method). Then Half-maximal inhibitory concentration (IC50) value for the above extract was calculated using SigmaPlot™ 13 software. All steps of the experiment were done in triplicate and the results were considered as average. Results: The IC50 content of methanolic extract of dandelion roots after 24, 48, and 72 hours on L. major were calculated to be 1.04, 0.9, and 0.68 µg/ml. The highest growth inhibition (100%) was observed at a concentration of 1200 μg/ml after 72 hours of exposure. Background and objectives: Soil contamination with Toxocara spp. eggs is the main source of human toxocariasis. The aim of this study was to determine rate of soil contamination with Toxocara spp. eggs in Yazd, central Iran. Methods: During September 2014 to February 2015, a total of 220 soil samples were collected from public parks, primary schools, green areas, kindergartens, suburban areas, streets, residential complexes, backyards and a bus terminal in the city of Yazd, Yazd Province. Soil samples were investigated for the presences of Toxocara eggs by flotation technique using sucrose solution (1.2 g/cm3) and using light microscopy. Data analysis was carried out by SPSS (version 23) software package. Results: A total of 220 soil samples from nine different places were examined. Of these samples, 51 (23.2%) were contaminated with Toxocara spp. eggs. The contamination rate was 26.3% in public parks, 19.2% in primary schools, 28.6% in green areas, 22.2% in kindergartens, 24% in suburban areas, 8.3% in streets, 18.8% in residential complexes, 19% in backyards and 28.6% in the bus terminal. There was no significant difference in the soil contamination rate with Toxocara spp. eggs between different sampling places (χ2 = 3.096, d.f = 8 and P= 0.92) and sampling areas (χ2= 2.289, d.f = 2 and P= 0.318). Conclusion: The high rate of soil contamination with Toxocara spp. eggs in Yazd emphasize on the need for preventive measures, such as improving sanitation, providing public health education and controlling stray dogs and cats near children play areas.

Abstract Acanthamoeba spp. can cause amoebic keratitis (AK). Chlorhexidine is effective for AK treatment as monotherapy, but with a relative failure on drug bioavailability in the deep corneal stroma. The combination of chlorhexidine and propamidine isethionate is recommended in the current AK treatment. However, the effectiveness of treatment depends on the parasite and virulence strains. This study aims to determine the potential of Garcinia mangostana pericarp extract and α-mangostin against Acanthamoeba triangularis, as well as the combination with chlorhexidine in the treatment of Acanthamoeba infection. The minimal inhibitory concentrations (MICs) of the extract and α-mangostin were assessed in trophozoites with 0.25 and 0.5 mg/mL, for cysts with 4 and 1 mg/mL, respectively. The MIC of the extract and α-mangostin inhibited the growth of A. triangularis trophozoites and cysts for up to 72 h. The extract and α-mangostin combined with chlorhexidine demonstrated good synergism, resulting in a reduction of 1/4–1/16 of the MIC. The SEM results showed that Acanthamoeba cells treated with a single drug and its combination caused damage to the cell membrane and irregular cell shapes. A good combination displayed by the extract or α-mangostin and chlorhexidine, described for the first time. Therefore, this approach is promising as an alternative method for the management of Acanthamoeba infection in the future.

Background and Objectives: Trichomonas vaginalis is a flagellate protozoan that is the most common sexually parasitic transmitted infection in the world. Metronidazole is currently used for the treatment of trichomoniasis, which has side effects. In recent years, the use of plants and natural compounds has been considered by researchers. This study was conducted to investigate the anti-trichomonal activity of methanolic extract of Hyssopus officinalis on Trichomonas vaginalis in vitro. Methods: In this experimental study, the extract was prepared by Soxhlet method, and to evaluate the effect of methanolic extract of Hyssopus officinalis on Trichomonas vaginalis parasites, the concentrations of 50, 100, 200, 400, 600, 800, 1,000, 1,200, and 1,400 μg/ml was prepared. Metronidazole and distilled water were considered as positive and negative controls, respectively. Afterward, 105 live parasites were added to all tubes, and all groups were kept at 37°C. Live parasites were counted at 24, 48, and 72 h intervals by Trypan Blue using a neobar slide and light microscope (Hemocytometer). Subsequently, the IC50 value for the above extract was calculated using SigmaPlot ™ software (version 13). It should be noted that all steps of the experiment were performed in triplicate and the results were considered as average. Results: The IC50 of methanolic extract of Hyssopus officinalis on Trichomonas vaginalis parasite was calculated at 965.8, 730.4, and 542.1 μg/ml after 24, 48, and 72 h, respectively. The highest mortality rate (100%) was observed at a concentration of 1,400 μg/ml after 72 h of exposure. Conclusion: Methanolic extract of Hyssopus officinalis in different concentrations had an inhibitory effect on the growth of the Trichomonas vaginalis parasite. Further and more comprehensive studies are suggested to investigate the constituents of this plant and the lethal effect of the parasite in vivo conditions.

Background and Aim: Giardia duodenalis is one of the most common enteric protozoan parasites in vertebrates, such as humans, domestic and wild animals, causing giardiasis. To the best of our knowledge, little is known about the genetic diversity of G. duodenalis assemblages. This study aimed to identify genetic diversity of G. duodenalis assemblages in Iranian stray dogs. Materials and Methods: A total of 450 fecal samples were collected from 2015 to 2016 from stray dogs of Northwest Iran. All specimens were observed microscopically following concentration and flotation techniques. Subsequently, DNA samples were extracted, amplified, and sequenced targeting the glutamate dehydrogenase gene. Results: The overall prevalence of G. duodenalis in infected dogs was estimated at 1.6%, based on microscopic and molecular diagnoses. Sequencing and phylogenetic analyses indicated a high level of genetic diversity of assemblage C (haplotype diversity; 0.802). Conclusion: The pairwise sequence distances between the identified isolates of assemblage C showed an intradiversity of 0.3%-1.3% and identity of 98.7%-100%. Current findings indicate that a significant genetic diversity of G. duodenalis assemblage C haplotypes is unequivocally circulates among stray dogs in Northwest Iran.

Background and objectives: Head louse, also known as Pediculus humanus capitis, is an obligate ectoparasite that is a distributed all around the world. The aim of present study was to determine the prevalence of head lice infestation in primary school students in Tabriz, East Azerbaijan Province, Iran. Methods: Overall, 3,032 students (1,515 boys and 1,517 girls) from 33 primary schools in Tabriz were selected via multi-stage cluster sampling in 2018. The students were examined individually and privately by experts. Head lice infestation was confirmed by detecting adult or nymph or nits less than 1 cm from base of hair. Data analysis was carried out in SPSS software package (version 23). Association of participants’ characteristics with pediculosis infestation was evaluated using logistic regression and chi-square test at significance level of 0.05. Results: Head lice infestation was present in 130 (4.29%) students, while the prevalence of this condition was higher in girls (2.11%) than in boys. The highest prevalence (6.05 %) was observed in children age 6 years old regardless of gender. The rate of head lice infestation was lowest among students aged ≥12 years (3.17%). There was a significant relationship between head louse infestation and parent’s occupation, family size, the frequency of combing and hair style. Conclusion: Although the prevalence of head lice infestation in Tabriz is relatively low, screening and treating schoolchildren should be done continuously in order to reduce the infestation rate.

Background and Aim: There is a large amount of information on intestinal parasites in stray dogs and golden jackals (Canis aureus) in Mazandaran Province, Iran. However, there is little information about foxes and wolves, which have a potential role in the spread of dangerous parasitic diseases, such as echinococcosis and toxocariasis. The aim of the present study was to identify the genus or species of parasitic worms in stool samples obtained from carnivores in Mazandaran Province, Iran, from August 2017 to April 2018. Materials and Methods: A total of 274 fecal samples were collected from carnivores, including dog, fox, wolf, and C. aureus in three areas of Mazandaran Province, Iran. All specimens were examined by centrifugal fecal flotation using a solution of Sheather's sugar to detect helminths eggs. Then, all samples were assessed using a light microscope. Data analysis was performed by SPSS version. 18 (Chicago, IL, USA). Results: In this study, seven genera of helminths were observed, including Ancylostoma, Uncinaria, Toxocara, Dipylidium, Toxascaris, Taenia, and Spirocerca. The prevalence of helminth infections was 97.7% (127 out of 130), 56.7% (51 out of 90), 51.4% (18 out of 35), and 52.6% (10 out of 19), among dogs, C. aureus, foxes, and wolves, respectively. The highest prevalence of Ancylostoma and Toxocara infections occurred in the eastern and central areas of the province (42.1% and 35.7%, respectively). Conclusion: Based on the results of this study, the infection with intestinal zoonotic helminths in carnivores was an important public health factor in Mazandaran. Therefore, these infections can be potentially harmful to humans and other animals.

Hyalomma spp. is responsible for the transmission of bacterial, protozoan, rickettsial and viral pathogens in animals and humans.The aim of this study was to evaluate the acaricidal activity of iron oxide nanoparticles (Fe2O3 and Fe3O4 NPs) size 15 nm against Hyalomma spp. in vitro. The acaricidal activity of Fe-NPs was evaluated at concentrations of 50, 125, and 250 µg/ml and controls (distilled water and Cypermethrin) following 10, 30, and 60 min of exposure in triplicate and the experiments were performed two spraying and contact methods. The results of this study showed that all concentrations of Fe-NPs had acaricidal activity, and a concentration of 250 µg/ml at an exposure time of 10 min had the highest acaricidal effect (85.7%). The median lethal concentration (LC50) values were 50 µg/ml in 60 min, and (LC99) values were 150 mg/ml in 30 min for Hyalomma spp.. The results showed that the spray method was more effective than the contact method. Statistically, there was no difference between the acaricidal effect of trivalent iron (Fe2O3) and quadrivalent (Fe3O4) iron nanoparticles. The findings of the present study showed that Fe-NPs had potent acaricidal activity. However, further in vivo studies are required to evaluate the efficacy of this nanoparticle.

ABSTRACT: Background: Bacterial resistance to multiple drugs has recently emerged as a serious health problem. Concomitantly, the characterization of new substances with potential antimicrobial activity has been less frequent in the drug development industry. The overexpression of genes encoding efflux pumps that expel antimicrobial drugs from the intracellular environment, lowering these to subinhibitory concentrations, are among the resistance mechanisms predisposing microorganisms to high drug resistance. Staphylococcus aureus is a bacterium found in the normal microbiota of the skin and mucous membranes, and is an opportunistic microorganism capable of causing infections with high rates of morbidity and mortality. TetK is an efflux pump characterized by its ability to provide bacterial resistance to antibiotics from the tetracycline class. This study aimed to evaluate the inhibitory effect of ferulic acid and four of its esterified derivatives against resistant Staphylococcus aureus strains. Method: Ferulic acid derivatives were obtained by esterification and then characterized by hydrogen and carbon-13 nuclear magnetic resonance analysis. The minimum inhibitory concentrations (MIC) of ferulic acid and its esterified derivatives, ethidium bromide, and antibiotics were obtained using the microdilution test, while the efflux pump inhibition test was conducted by examining reduction in the MICs. Results: Propylferulate was seen to reduce the minimum inhibitory concentration (MIC) of both the control substance ethidium bromide and the tested antibiotic, indicating that this compound is promising for the use of efflux pump inhibition of IS-58 strains. Conclusions: This study provides strong evidence that the molecular basis for this activity is potentially due to the MsrA and TetK efflux pumps. However, further investigations are necessary to prove this hypothesis and elucidate the potentiating mechanism of the modulatory effect.

Objective(s): Hyalomma spp. is responsible for transmission of protozoan, bacterial, rickettsial and viral diseases in humans and animals. Recently, there was a wide number of promising attempts to evaluate and use of nanoparticles for the control of ticks. Methods: The aim of this study was to evaluate the acaricidal activity of zinc oxide nanoparticles (ZnO NPs) size 15 nm against Hyalomma spp. in vitro. The arcaricidal activity of Zn NPs were evaluated at concentrations of 50, 125 and 250 mg/ml and controls (distilled water and Cypermethrin) following 10, 30 and 60 min of exposure in triplicate and the experiments were performed two spraying and contact methods. Results: The results of this study showed that all concentrations of Zn NPs had acaricidal activity and concentration of 125 mg/ml at exposure time of 30 min and concentration of 250 mg/ml at all exposure times had highest acaricidal effect (100%). The median lethal concentration (LC50) values were 50 mg/ml in 60 min and (LC99) values were 150 mg/ml in 30 min for Hyalomma spp.. The results showed that the spray method was more effective than the contact method. Conclusions: The findings of present study showed that Zn NPs had potent acaricidal effect and recommended as an effective acaricidal agent. However, further in vivo studies are required to evaluate the efficacy of this nanoparticle.

Abstract Background The larval stage of the tapeworm Echinococcus granulosus is the causative agent of hydatid disease in humans. This zoonotic parasitic infection remains a major health problem in certain areas of the world where is still endemic. In view of the ineffectiveness of some drug treatments, the surgical removal of cysts remains the preferred treatment option together with the administration of albendazole and mebendazole. However, severe side effects of these drugs have been reported which demands developing new scolicidal agents that confer suitable efficacy and fewer side effects during surgery. Methods To that purpose, in the present work we assessed the effectiveness of ivermectin (IVM), a macrocyclic lactone endectocide that has shown to be an effective nematocidal drug against other important parasitic infections. To overcome the limitations observed in some drug formulations and resistance, we used nano lipid carriers (NLCs) as a targeted and sustained drug delivery system for IVM. We evaluated the in vitro cestocidal and apoptotic effects of NLCs-loaded IVM versus IVM by quantifying the expression of caspase-3 mRNA. Results We found that after 60 and 120 min of administration, 800 μg/ml and 400 μg/ml NLCs-loaded IVM induced 100% mortality, respectively. On the other hand, the 800 μg/ml of IVM induced 100% mortality rate 150 min after administration. Additionally, we found that NLCs-loaded IVM induced higher mRNA caspase-3 expression suggesting a more potent apoptotic effect on the parasite. Conclusions These data suggest that NLCs-loaded IVM may be a promising alternative to current treatments although in vivo studies are needed.

Plants with medicinal properties have been used in the treatment of several infectious diseases, including Acanthamoeba infections. The medicinal properties of Cambodian plant extracts; Annona muricata and Combretum trifoliatum were investigated against Acanthamoeba triangularis. A total of 39 plant extracts were evaluated and, as a result, 22 extracts showed positive anti-Acanthamoeba activity. Of the 22 extracts, 9 and 4 extracts showed anti-Acanthamoeba activity against trophozoites and cysts of A. triangularis, respectively. The minimum inhibitory concentration of A. muricata and C. trifoliatum extracts against trophozoites and cysts was 500 and 1,000 μg/mL, respectively. The combination of A. muricata at 1/4×MIC with chlorhexidine at 1/8×MIC demonstrated a synergistic effect against trophozoites, but partial synergy against cysts. A 40% reduction in trophozoites and 60% of cysts adhered to the plastic surface treated with both extracts at 1/2×MIC were noted comparing to the control (P < 0.05). Furthermore, a reduction of 80% and 90% of trophozoites adhered to the surface was observed after pre-treatment with A. muricata and C. trifoliatum extracts, respectively. A 90% of cysts adhered to the surface was decreased with pre-treatment of A. muricata at 1/2×MIC (P < 0.05). A 75% of trophozoites and cysts from Acanthamoeba adhered to the surface were removed after treatment with both extracts at 4×MIC (P < 0.05). In the model of contact lens, 1 log cells/mL of trophozoites and cysts was significantly decreased post-treatment with both extracts compared to the control. Trophozoites showed strong loss of acanthopodia and thorn-like projection pseudopodia, while cysts demonstrated retraction and folded appearance treated with both extracts when observed by SEM, which suggests the potential benefits of the medicinal plants A. muricata and C. trifoliatum as an option treatment against Acanthamoeba infections.

The evaluation and confirmation of healing properties of several plant species of genus Terminalia based on their traditional uses and the clinical claims are of utmost importance. Genus Terminalia has received more attention to assess and validate the therapeutic potential and clinical approval due to its immense folk medicinal and traditional applications. Various species of Terminalia genus are used in the form of herbal medicine and formulations, in treatment of diseases, including headache, fever, pneumonia, flu, geriatric, cancer, to improve memory, abdominal and back pain, cough and cold, conjunctivitis, diarrhea, heart disorder, leprosy, sexually transmitted diseases, and urinary tract disorders. These are reported to possess numerous biological properties, counting: antibacterial, antifungal, antiinflammatory, antiviral, antiretroviral, antioxidant, and antipa7rasitic. This current research review aims to update the detailed biological activities, pre-clinical and clinical studies of various extracts and secondary metabolites from several plant species under the genus Terminalia, along with information on the traditional uses and chemical composition to develop a promising strategy for their potential applications in the form of medicine or use in modern drug formulations for treating diseases like pneumonia, flu, and other types of viral infections or controlling human contagions.

Objective(s): Nanoparticles (NPs) are particles with the size range approximately from 1 to 100 nanometers that are made in different shapes. Nanotechnology is an emerging technology that expected to open some new opportunities in order to destroy and control of microorganisms using of materials and systems at the scale of the atom. Echinococcus granulosus is the agent of cystic echinococcosis (CE), which has a cosmopolitan distribution. This parasite causes hydatid cysts that can afflicted at various organs of mammalian host such as liver, lung even in heart, brain and bone which may lead to death. The current methods for treatment of human CE include surgery. Methods: This study was undertaken for the first time to evaluate the scolicidal effect of zinc oxide nanoparticles (ZnO- NPs) against hydatid cyst protoscolices in vitro. The scolicidal activities of the zinc oxide nanoparticles were tested in concentrations of 50,100 and 150 mg/ml following 10, 30 and 60 min of incubation and tested were repeated at three times. Data were analyzed by SAS software. Results: The results showed that the zinc oxide nanoparticles at the concentration of 50 mg/ml leads to killing 19/6% protoscolices at 10 minutes. In 150 mg/ml concentration, the black ZnO particles were covered on all protoscolices, and they could not be seen or counted. Conclusions: This investigation showed that ZnO-NPs a statistically significant difference in the protoscolicidal activity with different dilutions but, is not recommended as a powerful scolicidal agent.

ABSTRACT Background and objectives: Sarcoptes scabiei is a microscopic parasitic arthropod that penetrates into epidermis of the skin and causes scabies, a contagious disease that is one of the most common causes of dermatosis in the world. Methods: We herein summarize published papers about Sarcoptes scabiei infestation in Iran. Databases including MEDLINE, Pubmed and Google scholar were searched using the following terms: Sarcoptes scabiei, mange, prevalence of Sarcoptes scabiei and scabies status in Iran. Results: Overall, we found approximately 15 articles related to the prevalence of Sarcoptes scabiei infestation in Iran. According to these studies, the prevalence of Sarcoptes scabiei infestation has been rather high in Iran during war and in prisons and humid areas of the country. Conclusion: In spite of the low prevalence of scabies, it is necessary to implant prevention programs continuously by reducing overcrowding, improving health education, personal hygiene, diagnostics tools, and screening and treatment of high-risk populations. Cymbopogon winterianus, known as “citronella grass”, is an important aromatic and medicinal tropical herbaceous plant. The essential oil of C. winterianus (EOCw) is popularly used to play an important role in improving human health due to its potential as a bioactive component. The present study aimed to identify the components of the essential oil of C. winterianus and verify its leishmanicidal and trypanocidal potential, as well as the cytotoxicity in mammalian cells, in vitro. The EOCw had geraniol (42.13%), citronellal (17.31%), and citronellol (16.91%) as major constituents. The essential oil only exhibited significant cytotoxicity in mammalian fibroblasts at concentrations greater than 250 μg/mL, while regarding antipromastigote and antiepimastigote activities, they presented values considered clinically relevant, since both had LC50 < 62.5 μg/mL. It can be concluded that this is a pioneer study on the potential of the essential oil of C. winterianus and its use against the parasites T. cruzi and L. brasiliensis, and its importance is also based on this fact. Additionally, according to the results, C. winterianus was effective in presenting values of clinical relevance and low toxicity and, therefore, an indicator of popular use.

As our recent society is turning to senility, this research tries to analyze meaningful factors on elders' lives according to social interpretation approach. In this research, it is tried to reach a background theory through qualitative method and deep interview. Using systematic sampling and theoretical saturation scale,21 people among 68 to 81-years-old elders in Tehran, district 21 (Tehransar) were interviewed and the information received coded(open and axial)and concepts and categories reached, were analysed. The data using background theory contains 10 categories. Categorizing in this research is family adaptability that generally covered all other categories. Based on nuclear category, two cases out of data, are convergence and disparity and the most important achievement of research is the effect of convergence (family and social) on life's meaningfulness and the effect of disparity on life's meaninglessness. Factors like health or sickness, financial situation and fidelity of elders are considered underlie that shows their approach to life. Finally, basic reached theory presented in the form of paradigm, selective, adaptive and marriage.

Parasitic infections are a threat to the wildlife by endangering the health of wild animals and serving as a source of transmission of infectious agents to livestock and humans. Despite the distribution of wild boars (Sus scrofa), wild goat (Capra aegagrus) and red deer (Cervus elaphus) throughout the world, information on their parasitic infections is limited. Knowledge of the parasitic fauna of animals in protected areas may be used in protection of endangered species. The present study aimed to investigate the status of gastrointestinal parasitic infections in wild boar, wild goat and red deer in the Aynaloo protected area, Northwest of Kaleybar city, East Azerbaijan province. For this purpose, in spring 2015, 30 fecal samples of wild boar, 30 fecal samples of wild goat and 30 fecal samples of red deer were collected. The samples were processed using direct smear and Clayton lane floatation technique. Five species of helminths were detected from wild boar fecal samples consisting of Trichuris suis, Strongyloides suis, Physocephalus sexalatus, Ascaris suum and Metastrongylus apri. Among them, one species (Strongyloides suis) was reported for the first time in Iran. Protozoa were not detected in the fecal samples of the wild boar. Wild goats were infected with Oesophagostomum columbianum, Muellerius capillaris and Eimeria spp. Respiratory larva and Eimeria spp. were detected from red deer samples.

Peganum harmala, a well-known medicinal plant, has been used for several therapeutic purposes as it contains numerous pharmacological active compounds. Our study reported an anti-parasitic activity of P. harmala seed extract against Acanthamoeba triangularis. The stress induced by the extract on the surviving trophozoites for Acanthamoeba encystation and vacuolization was examined by microscopy, and transcriptional expression of Acanthamoeba autophagy-related genes was investigated by quantitative PCR. Our results showed that the surviving trophozoites were not transformed into cysts, and the number of trophozoites with enlarged vacuoles were not significantly different from that of untreated control. Molecular analysis data demonstrated that the mRNA expression of tested AcATG genes, i.e., ATG3, ATG8b, and ATG16, was at a basal level along the treatment. However, upregulation of AcATG16 at 24 h post treatment was observed, which may indicate an autophagic activity of this protein in response to the stress. Altogether, these data revealed the anti-Acanthamoeba activity of P. harmala extract and indicated the association of autophagy mRNA expression and cyst formation under the extract stress, representing a promising plant for future drug development. However, further identification of an active compound and a study of autophagy at the protein level are needed.

Cystic hydatid disease (CHD) is a zoonotic disease with different clinical stages caused by the larval stage of the cestode Echinococcus granulosus. It is important to highlight as a public health problem in various regions of the world. In the current study, the efficacy and apoptotic activity of the liposomal system containing juglone (5-hydroxy-1,4-naphthoquinone) were assessed against protoscoleces (PSCs) in vitro. To this aim, firstly, liposomal vesicles were prepared by the thin-film method. Their physico-chemical features were assessed using Zeta-Sizer and Scanning Electron Microscope (SEM). Subsequently, various concentrations (50, 100, 200, 400, and 800 μg/mL) of juglone nanoliposomes at different exposure times (15, 30, 60, and 120 min) were used against PSCs. Results showed that juglone nanoliposomes at all tested concentrations induced scolicidal effect, however, 800 μg/mL and 400 μg/mL of juglone nanoliposomes could reach 100% mortality in 60 and 120 min, respectively. Additionally, we found that caspase-3 mRNA expression was higher in PSCs treated with juglone nanoliposomes compared to control groups (p < 0.001). Therefore, juglone nanoliposomes are suggested to have a more potent apoptotic effect on PSCs. Generally, optimized doses of juglone nanoliposomes could display significant scolicidal effects. Moreover, further in vivo studies are required to evaluate the efficacy of this nanoliposome.

Parasites may affect the dynamics of bird populations. Plasmodium, Leucocytozoon and Haemoproteus are well-known avian haematozoa that can trigger decreased productivity and high mortality in domesticated birds. In this study, we evaluated the prevalence of avian blood parasites (Plasmodium, Leucocytozoon and Haemoproteus) against 335 birds of 8 species in the Yazd province in central Iran. To detect blood parasites, Giemsa-stained blood smears were prepared. Of the birds, 11.64% (39/335) were infected with at least one parasite genus, particularly Haemoproteus (32.6%; 23/335). The total prevalence values for Plasmodium, Haemoproteus and Leucocytozoon were 1.7, 6.8 and 2.9%, respectively. Plasmodium had lower prevalence rates of 1.7% (6/335). Among birds, pigeons, hens and ducks have the highest prevalence of Haemoproteus, Leucocytozoon and Plasmodium parasites at 1.7%, 6.8% and 2.9%, respectively. Results from this research extend our knowledge on the incidence of avian blood parasites in domesticated birds living in central Iran. The overall low incidence of avian blood parasites in birds was found in the Yazd province, Iran.

Myiasis is the infestation of animals or man tissues by parasitic dipterous fly larvae. Wound myiasis is the result of fly egg deposition on decaying flesh or pus discharging wounds. This case report describes a type of wound myiasis caused by Calliphora spp. in a Flamingo (Phoenicopterus ruber) from East Azerbaijan Province, Iran. A 3-yr-old female Flamingo was suffering in its left wing leading to an extensive discharging wound, which was heavily infested by maggots (fly larvae). The examination of external morphological characters of the second and third-instar larvae, posterior spiracles and internal cephalopharyngeal skeleton, led to the identification of the Calliphora spp. fly genus. Treatment consisted of removal of the larvae and surgical debridement, then spray of antibiotic and toxic drug. Following removal of larvae and treatment, the symptoms completely resolved within the last hour and remained asymptomatic several weeks later. This is the first report of wound myiasis in a Flamingo (Phoenicopterus ruber) by the facultative myiasis agent Calliphora spp. in Iran and the world.

Myiasis is an animal or human pathogenic condition initiated by parasitic dipterous fly larvae feeding in the host’s necrotic or living tissue. Here we report a case of external ophthalmomyiasis caused by Lucilia sericata in a 78-yr-old with a vascular tumor of the retina and surgery history, from Bijar City of Kurdistan Province, Iran in 2015. Associated symptoms included right eye pain with mucoid ocular discharge, headache, sensing the presence of a foreign body in the eye and itching. Examination revealed a L. sericata Larva in his right eye. Infestation of ocular tissue by fly larvae (ophthalmomyiasis) progresses after retinal surgery and can destroy orbital tissues within days, especially in patient with poor hygienic conditions. Treatment consists of removal of the larvae and surgical debridement. Following removal of larva, the symptoms completely resolved within a few hours and remained asymptomatic several weeks later.
