Abstract Global developments such as climate change, a growing world population and the depletion of fossil resources make the sustainable use of biogenic resources in chemical production inevitable. This would also provide a final product with a higher added value than just utilizing the raw materials for applications in energy generation. In recent years, many researchers have shown that e.g., grass clippings, carrots and potato peels can be biotechnologically converted into high-value chemicals thereby increasing resource efficiency. A particular challenge, however, is the decentralized production of such biogenic raw materials as well as degradation affecting the composition and quality within short periods of time. Therefore, appropriate logistics concepts must be developed and evaluated to economically valorize biogenic raw materials. Such concepts differ significantly in terms of material utilization for the production of chemicals, composting or energetic valorization. This overview presents relevant examples of the conversion of biogenic residues into chemicals investigating basic logistic concepts and highlighting major challenges along bio-based value chains.

Abstract Biotransformation with enzymes and de novo syntheses with whole-cell biocatalysts each have specific advantages. These can be combined to achieve processes with optimal performance. A recent approach is to perform bioconversion processes and enzymatic catalysis simultaneously in one-pot. This is a well-established process in the biorefinery, where starchy or cellulosic material is degraded enzymatically and simultaneously used as substrate for microbial cultivations. This procedure leads to a number of advantages like saving in time but also in the needed equipment (e.g., reaction vessels). In addition, the inhibition or side-reaction of high sugar concentrations can be overcome by combining the processes. These benefits of coupling microbial conversion and enzymatic biotransformation can also be transferred to other processes for example in the sector of biofuel production or in the food industry. However, finding a compromise between the different requirements of the two processes is challenging in some cases. This article summarises the latest developments and process variations. Abstract Cable bacteria, characterized by their multicellular filamentous growth, are prevalent in both freshwater and marine sediments. They possess the unique ability to transport electrons over distances of centimeters. Coupled with their capacity to fix CO2 and their record‐breaking conductivity for biological materials, these bacteria present promising prospects for bioprocess engineering, including potential electrochemical applications. However, the cultivation of cable bacteria has been limited to their natural sediment, constraining their utility in production processes. To address this, our study designs synthetic sediment, drawing on ion exchange chromatography data from natural sediments and existing literature on the requirements of cable bacteria. We examined the effects of varying bentonite concentrations on water retention and the impacts of different sands. For the first time, we cultivated cable bacteria on synthetic sediment, specifically the freshwater strain Electronema aureum GS. This cultivation was conducted over 10 weeks in a specially developed sediment bioreactor, resulting in an increased density of cable bacteria in the sediment and growth up to a depth of 5 cm. The creation of this synthetic sediment paves the way for the reproducible cultivation of cable bacteria. It also opens up possibilities for future process scale‐up using readily available components. This advancement holds significant implications for the broader field of bioprocess engineering.

One major obstacle to the commercialization of electrobiotechnological systems is the cost of materials, including expensive electrodes. Smart recycling as well as the use of renewable resources can contribute to producing electrodes more ecologically and economically. Green waste is a biogenic residual material that occurs mainly in urban areas and is currently not recycled to a sufficient extent. Here we show the fabrication of electrodes from carbonized grass clippings and their application in microbial electrosynthesis as well as microbial fuel cells. While the electrodes cannot compete with established metal competitors for water electrolysis in microbial electrosynthesis, they perform comparably to commercial graphite electrodes in microbial fuel cells. With Geobacter sulfurreducens, a current response can be recorded for more than six weeks. To the best of our knowledge, this is the first time that carbonized green waste has been used as an electrode material for bioelectrochemical systems. This demonstrates the potential of carbonized biological materials as a raw material for electrodes and presents a recycling alternative for green waste.

The aim of this work was to produce the biopolymer polyhydroxybutyrate (PHB) from industrial flue gas as CO2 source and electrolysis originating hydrogen via microbial electrosynthesis. Besides the laboratory experiments, the experiments were carried out under industrial conditions directly on-site in a cogeneration plant. We were able to demonstrate that the use of flue gas as a CO2 source has no detectable negative effect on bacterial growth and PHB production in comparison to a pure gas mixture. In an electrochemical H-cell 333 ± 44 mg L−1 PHB were obtained, which corresponds to a PHB content of 43 ± 3 % of the cell dry weight. By using flue gas for the production of the biopolymer PHB, not only CO2 emissions are reduced, but also possible environmental pollution from non-biodegradable plastics. To the best of our knowledge, this is the first time that the production of PHB from flue gas has been demonstrated using Cupriavidus necator.

This work examines the electrochemically enhanced production of succinic acid using the bacterium Actinobacillus succinogenes. The principal objective is to enhance the metabolic potential of glucose and CO2 utilization via the C4 pathway in order to synthesize succinic acid. We report on the development of an electro-bioreactor system to increase succinic acid production in a power-2-X approach. The use of activated carbon fibers as electrode surfaces and contact areas allows A. succinogenes to self-initiate biofilm formation. The integration of an electrical potential into the system shifts the redox balance from NAD+ to NADH, increasing the efficiency of metabolic processes. Mediators such as neutral red facilitate electron transfer within the system and optimize the redox reactions that are crucial for increased succinic acid production. Furthermore, the role of carbon nanotubes (CNTs) in electron transfer was investigated. The electro-bioreactor system developed here was operated in batch mode for 48 h and showed improvements in succinic acid yield and concentration. In particular, a run with 100 µM neutral red and a voltage of −600 mV achieved a yield of 0.7 gsuccinate·gglucose−1. In the absence of neutral red, a higher yield of 0.72 gsuccinate·gglucose−1 was achieved, which represents an increase of 14% compared to the control. When a potential of −600 mV was used in conjunction with 500 µg∙L−1 CNTs, a 21% increase in succinate concentration was observed after 48 h. An increase of 33% was achieved in the same batch by increasing the stirring speed. These results underscore the potential of the electro-bioreactor system to markedly enhance succinic acid production.

Abstract In Germany alone, more than 5·106 tons of municipal green waste is produced each year. So far, this material is not used in an economically worthwhile way. In this work, grass clippings and tree pruning as examples of municipal green waste were utilized as feedstock for the microbial production of platform chemicals. A pretreatment procedure depending on the moisture and lignin content of the biomass was developed. The suitability of grass press juice and enzymatic hydrolysate of lignocellulosic biomass pretreated with an organosolv process as fermentation medium or medium supplement for the cultivation of Saccharomyces cerevisiae, Lactobacillus delbrueckii subsp. lactis, Ustilago maydis, and Clostridium acetobutylicum was demonstrated. Product concentrations of 9.4 gethanol L−1, 16.9 glactic acid L−1, 20.0 gitaconic acid L−1, and 15.5 gsolvents L−1 were achieved in the different processes. Yields were in the same range as or higher than those of reference processes grown in established standard media. By reducing the waste arising in cities and using municipal green waste as feedstock to produce platform chemicals, this work contributes to the UN sustainability goals and supports the transition toward a circular bioeconomy. Digitale Medien sind elementarer Bestandteil in Lehr-Lern-Prozessen und stehen im Einklang mit den physischen, medialen und virtuellen Lebensräumen der Schüler:innen indem sie Gegenwarts- und insbesondere auch eine Zukunftsbedeutung für die Heranwachsenden haben. Die didaktisch-methodische Reflektion und Bewertung von technologischen Innovationen aus dem Internet of Things (IoT) oder der Industrie 4.0 ermöglichen es, grundlegende Aussagen zu fächerspezifischen sowie fächerübergreifenden Potentialen zum Einsatz im schulischen Kontext zu formulieren. In diesem Beitrag werden die Potentiale für ein gelingendes Lehren und Lernen in der naturwissenschaftlichen Bildung von digitalen Zwillingen zusammenfassend betrachtet. Digitale Zwilling sind bereits in der industriellen Prozessentwicklung und -optimierung implementiert. Grundsätzlich nehmen die, in digitalen Zwillingen, repräsentierten Vernetzungsstrukturen zwischen digitaler und realer Welt und deren Potential in den Bereichen künstliche Intelligenz, maschinelles Lernen und Data-Mining-Prozessen einen wesentlichen Fokus ein. Um Bildungsperspektiven für den naturwissenschaftlichen Unterricht abzuleiten, werden die eingehenden und ausgehenden Datenstrukturen und deren Funktionsweisen in digitalen Zwillingen näher betrachtet. Der Beitrag thematisiert den Blick auf digitale Zwillinge als intelligente neue Bildungstechnologie in Lehr-Lern-Prozessen, welche auch im industriellen Sektor noch nicht an ihrem Entwicklungsendpunkt steht und vielfältige Anwendungspotentiale im Schulkontext, viele offene Forschungsfragen und Entwicklungspotentiale im Rahmen der Bildungsforschung bereithalten.

Abstract This study introduced an automated long‐term fermentation process for fungals grown in pellet form. The goal was to reduce the overgrowth of bioreactor internals and sensors while better rheological properties in the fermentation broth, such as oxygen transfer and mixing time, can be achieved. Because this could not be accomplished with continuous culture and fed‐batch fermentation, repeated‐batch fermentation was implemented with the help of additional bioreactor internals (“sporulation supports”). This should capture some biomass during fermentation. After harvesting the suspended biomass, intermediate cleaning was performed using a cleaning device. The biomass retained on the sporulation support went through the sporulation phase. The spores were subsequently used as inocula for the next batch. The reason for this approach was that the retained pellets could otherwise cause problems (e.g., overgrowth on sensors) in subsequent batches because the fungus would then show undesirable hyphal growth. Various sporulation supports were tested for sufficient biomass fixation to start the next batch. A reproducible spore concentration within the range of the requirements could be achieved by adjusting the sporulation support (design and construction material), and an intermediate cleaning adapted to this.

Fucoidan is a class of multifunctional polysaccharides derived from marine organisms. Its unique and diversified physicochemical and chemical properties have qualified them for potential and promising pharmacological uses in human diseases, including inflammation, tumors, immunity disorders, kidney diseases, and diabetes. Physicochemical and chemical properties are the main contributors to these bioactivities. The previous literature has attributed such activities to its ability to target key enzymes and receptors involved in potential disease pathways, either directly or indirectly, where the anionic sulfate ester groups are mainly involved in these interactions. These findings also confirm the advantageous pharmacological uses of sulfated versus non-sulfated polysaccharides. The current review shall highlight the molecular targets of fucoidans, especially enzymes, and the subsequent responses via either the upregulation or downregulation of mediators’ expression in various tissue abnormalities. In addition, in silico studies will be applied to support the previous findings and show the significant contributors. The current review may help in understanding the molecular mechanisms of fucoidan. Also, the findings of this review may be utilized in the design of specific oligomers inspired by fucoidan with the purpose of treating life-threatening human diseases effectively.

Cupriavidus necator is a facultative chemolithotrophic organism that grows under both heterotrophic and autotrophic conditions. It is becoming increasingly important due to its ability to convert CO2 into industrially valuable chemicals. To translate the potential of C. necator into technical applications, it is necessary to optimize and scale up production processes. A previous proof-of-principle study showed that C. necator can be used for the de novo production of the terpene α-humulene from CO2 up to concentrations of 11 mg L−1 in septum flasks. However, an increase in final product titer and space–time yield will be necessary to establish an economically viable industrial process. To ensure optimized growth and production conditions, the application of an improved process design in a gas bioreactor with the control of pH, dissolved oxygen and temperature including a controlled gas supply was investigated. In the controlled gas bioreactor, the concentration of α-humulene was improved by a factor of 6.6 and the space–time yield was improved by a factor of 13.2. These results represent an important step toward the autotrophic production of high-value chemicals from CO2. In addition, the in situ product removal of α-humulene was investigated and important indications of the critical logP value were obtained, which was in the range of 3.0–4.2.

Abstract Laboratory protocols using magnetic beads have gained importance in the purification of mRNA for vaccines. Here, the produced mRNA hybridizes specifically to oligo(dT)‐functionalized magnetic beads after cell lysis. The mRNA‐loaded magnetic beads can be selectively separated using a magnet. Subsequently, impurities are removed by washing steps and the mRNA is eluted. Magnetic separation is utilized in each step, using different buffers such as the lysis/binding buffer. To reduce the time required for purification of larger amounts of mRNA vaccine for clinical trials, high‐gradient magnetic separation (HGMS) is suitable. Thereby, magnetic beads are selectively retained in a flow‐through separation chamber. To meet the requirements of biopharmaceutical production, a disposable HGMS separation chamber with a certified material (United States Pharmacopeia Class VI) was developed which can be manufactured using 3D printing. Due to the special design, the filter matrix itself is not in contact with the product. The separation chamber was tested with suspensions of oligo(dT)‐functionalized Dynabeads MyOne loaded with synthetic mRNA. At a concentration of cB = 1.6–2.1 g·L–1 in lysis/binding buffer, these 1 μm magnetic particles are retained to more than 99.39% at volumetric flows of up to 150 mL·min–1 with the developed SU‐HGMS separation chamber. When using the separation chamber with volumetric flow rates below 50 mL·min–1, the retained particle mass is even more than 99.99%.

Abstract Purification of mRNA with oligo(dT)‐functionalized magnetic particles involves a series of magnetic separations for buffer exchange and washing. Magnetic particles interact and agglomerate with each other when a magnetic field is applied, which can result in a decreased total surface area and thus a decreased yield of mRNA. In addition, agglomeration may also be caused by mRNA loading on the magnetic particles. Therefore, it is of interest how the individual steps of magnetic separation and subsequent redispersion in the buffers used affect the particle size distribution. The lysis/binding buffer is the most important buffer for the separation of mRNA from the multicomponent suspension of cell lysate. Therefore, monodisperse magnetic particles loaded with mRNA were dispersed in the lysis/binding buffer and in the reference system deionized water, and the particle size distributions were measured. A concentration‐dependent agglomeration tendency was observed in deionized water. In contrast, no significant agglomeration was detected in the lysis/binding buffer. With regard to magnetic particle recycling, the influence of different storage and drying processes on particle size distribution was investigated. Agglomeration occurred in all process alternatives. For de‐agglomeration, ultrasonic treatment was examined. It represents a suitable method for reproducible restoration of the original particle size distribution.

Abstract The implementation of single‐use technologies offers several major advantages, e.g. prevention of cross‐contamination, especially when spore‐forming microorganisms are present. This study investigated the application of a single‐use bioreactor in batch fermentation of filamentous fungus Penicillium sp. (IBWF 040‐09) from the Institute of Biotechnology and Drug Research (IBWF), which is capable of intracellular production of a protease inhibitor against parasitic proteases as a secondary metabolite. Several modifications to the SU bioreactor were suggested in this study to allow the fermentation in which the fungus forms pellets. Simultaneously, fermentations in conventional glass bioreactor were also conducted as reference. Although there are significant differences in the construction material and gassing system, the similarity of the two types of bioreactors in terms of fungal metabolic activity and the reproducibility of fermentations could be demonstrated using statistic methods. Under the selected cultivation conditions, growth rate, yield coefficient, substrate uptake rate, and formation of intracellular protease‐inhibiting substance in the single‐use bioreactor were similar to those in the glass bioreactor.

Abstract Diazotrophic cyanobacteria are able to fix N2 from the atmosphere and release it as bioavailable nitrogen what other organisms can utilize. Thus, they could be used as living nitrogen supplier whereby the use of fertilizer could be reduced in agricultural industry what results in a decrease of laughing gas released during fertilizer production. The diazotroph cyanobacterium Desmonostoc muscorum (D. muscorum) was characterized in shake flasks cultivated in nitrogen‐free and nitrogen‐containing medium. Similar growth rates were reached in both cultivations and the release of ammonium by D. muscorum was detected under nitrogen depletion. Subsequently, D. muscorum was co‐cultivated with Arabidopsis thaliana (A. thaliana) in nitrogen‐free medium. Additionally, the plant was cultivated in nitrogen containing and nitrogen‐free medium without D. muscorum as reference. A co‐cultivation led to higher growth rates of the cyanobacterium and similar growth of A. thaliana with similar maximum photochemical efficiency of photosystem II compared to the growth of nitrogen containing medium. Further, accumulation of cyanobacterial cells around the roots of A. thaliana was detected, indicating a successfully induced artificial symbiosis. Based on these results, D. muscorum could be a promising cyanobacterium as living nitrogen supplier for plants.

Abstract Considering global developments like climate change and the depletion of fossil resources, the use of new and sustainable feedstocks such as lignocellulosic biomass becomes inevitable. Green waste comprises heterogeneous lignocellulosic biomass with low lignin content, which does not stem from agricultural processes or purposeful cultivation and therefore mainly arises in urban areas. So far, the majority of green waste is being composted or serves as feedstock for energy production. Here, the hitherto untapped potential of green waste for material utilization instead of conventional recycling is reviewed. Green waste is a promising starting material for the direct extraction of valuable compounds, the chemical and fermentative conversion into basic chemicals as well as the manufacturing of functional materials like electrodes for electro-biotechnological applications through carbonization. This review serves as a solid foundation for further work on the valorization of green waste.

With regard to the emerging health threat by antibiotic resistant bacteria, bacteriocins are considered as a promising way to overcome this urgent problem. In this work, a hitherto unknown bacteriocin from the terrestrial cyanobacterium Chroococcidiopsis cubana was heterologously expressed in Escherichia coli, purified to homogeneity and tested for activity against several bacteria and one yeast species. The compound showed potent bacteriolytic activity against several Gram-positive bacteria and slight activity against one Gram-negative strain. The bacteriocin had no cytotoxic impact on mouse neuroblastoma N2a cells, indicating its potential for treatment against Gram-positive bacterial pathogens in human diseases.

Abstract Due to the emerging rise of multi‐drug resistant bacteria, the discovery of novel antibiotics is of high scientific interest. Through their high chemodiversity of bioactive secondary metabolites, cyanobacteria have proven to be promising microorganisms for the discovery of antibacterial compounds. These aspects make appropriate antibacterial screening approaches for cyanobacteria crucial. Up to date, screenings are mostly carried out using a phenotypic methodology, consisting of cyanobacterial cultivation, extraction, and inhibitory assays. However, the parameters of these methods highly vary within the literature. Therefore, the common choices of parameters and inhibitory assays are summarized in this review. Nevertheless, less frequently used method variants are highlighted, which lead to hits from antimicrobial compounds. In addition to the considerations of phenotypic methods, this study provides an overview of developments in the genome‐based screening area, be it in vivo using PCR technique or in silico using the recent genome‐mining method. Though, up to date, these techniques are not applied as much as phenotypic screening.

Fucoidan is a heterogeneous group of polysaccharides isolated from marine organisms, including brown algae and marine invertebrates. The physicochemical characteristics and potential bioactivities of fucoidan have attracted substantial interest in pharmaceutical industries in the past few decades. These polysaccharides are characterized by possessing sulfate ester groups that impart negatively charged surfaces, low/high molecular weight, and water solubility. In addition, various promising bioactivities have been reported, such as antitumor, immunomodulatory, and antiviral effects. Hence, the formulation of fucoidan has been investigated in the past few years in diverse pharmaceutical dosage forms to be able to reach their site of action effectively. Moreover, they can act as carriers for various drugs in value-added drug delivery systems. The current work highlights the attractive biopharmaceutical properties of fucoidan being formulated in oral, inhalable, topical, injectable, and other advanced formulations treating life-quality-affecting diseases. Therefore, the present work points out the current status of fucoidan pharmaceutical formulations for future research transferring their application from in vitro and in vivo studies to clinical application and market availability.

Digitization offers new perspectives for educational research to identify the effects of visualizations regarding cognitive processing. In addition, new types of data can be generated, expanding the possibilities for visualizing cognitive processes and understanding human learning. Digital twins are already used in Industry 4.0, as an additional visualization to a real object, for data mining and data analysis for process optimization. The increasing integration of digital twins in the industrial sector requires the formulation of corresponding educational goals to ensure high-quality and future-oriented education. Therefore, future generations must be introduced to technologies from industry during their education. In this paper, an intelligent photometric measurement system called SmaEPho with a digital twin for science, technology, engineering, and mathematics (STEM) learning is presented. In addition to its function as a photometric measurement device, an intelligent sensor technology allows for data generation on the user’s usage behavior. The digital twin reflects and visualizes these data in real-time. This enables a variety of new didactic and methodological approaches in teaching. A first study evaluating the hardware and tracking components of SmaEPho shows that the deviation accuracy of the measurement system is sufficient for experimental applications in schools. Another study with n=52 students confirmed the excellent usability of the SmaEPho hardware platform. These research results lay the foundation for a variety of future research questions on data analysis and machine learning algorithms with the aim of increasing the quality of education. The use of intelligent digital twins as an element of digitization in educational contexts offers the extended possibility of identifying cognitive processing steps using this technology.

Microwave-assisted extraction (MAE) is recognized as a green method for extraction of natural products. The current research aimed to explore the MAE for fucoidans extraction from different brown seaweeds, including Fucus vesiculosus, F. spiralis, and Laminaria saccharina. Following several solvent-extraction pre-treatment steps and MAE optimization, the algal biomasses were extracted in a ratio of 1:25 in 0.1 M HCl containing 2 M CaCl2 for 1.0 min. The results showed that L. saccharina’s extract was different from the others, regarding the highest sugar content reached 0.47 mg glucose equivalent/mg extract being confirmed by monosaccharide composition analysis and the lowest fucoidan content and sulfation degree at 0.09 mg/mg extract and 0.13, respectively. Moreover, these findings were confirmed by tentative structural elucidation based on Fourier-transform infrared spectrometry which also showed a different spectrum. However, the MAE enhanced melanoidins formation in products, which was confirmed by the intense band at 1420 cm−1. Interestingly, the results of monomeric composition showed that fucoidan extract by MAE from F. vesiculosus belonged to sulfated galactofucans which are known for their potential bioactivities. Furthermore, the cytotoxic activity of the four fucoidans in concentrations ranging from 4.9 µg/mL to 2500 µg/mL was investigated and correlated with the chemical characterization showing that F. vesiculosus_MAE fucoidan was the most potent and safest. The current research revealed the chemical heterogeneity of fucoidans regarding taxonomical class and used greener extraction method of fucoidans toward the achievement of the UN sustainability goals.

The privatization exercise that led to the creation of 11 distribution companies (Discos) is yet to yield the result of meeting the electricity supply needs of Nigeria. It is in realization of this fact that the Federal Government of Nigeria recently made different proposals and regulations to increase efficiency, availability, and competitiveness within the power industry. This article establishes a technical gain ratio to measure the impact of scaling up smart metering on the aggregated technical commercial and collection (ATC&C) losses. The ATC&C loss is the difference between the amount of electricity received by a distribution company from the transmission company and the amount of electricity for which it invoices its customers plus the adjusted collections loss. To achieve this objective, reviews of historical data of the Discos ATC&C losses and customers’ metering records as published by the Nigerian Electricity Regulatory Commission (NERC) for 2015, 2016, 2017, 2018, and 2020 were carried out. In addition, efforts were made to mathematically model the relationship between % metering and % ATC&C losses as this was the framework to help carry out effective forecasts and analyses of the study in order to show the impact level of the strategy employed. One of the salient technical contributions of this article was that it established that for every 1% increase in metering installation, there is a proportionate 0.8% decrease in ATC&C losses, provided all other factors responsible for technical and commercial losses remain constant. Consequently, improved ATC&C loss reduction would be achieved if Discos adopt a combination of other strategies that would ensure reduction in technical and commercial losses in addition to aggressive deployment of meter assets. However, in practice, factors causing technical and commercial losses are never constant as system components depreciate/burn out with time, energy theft, and pilferage, and meter tamper/bypass is on the increase daily; meter deployment is not aggressive enough to match utility customers’ growth. Hence, the adoption of combined modern strategies in addition to aggressive metering in tandem with customers’ growth has to be employed by Discos in a deliberate attempt to reduce ATC&C losses.

Green waste, e.g., grass clippings, is currently insufficiently recycled and has untapped potential as a valuable resource. Our aim was to use juice from grass clippings as a growth medium for microorganisms. Herein, we demonstrate the production of the sesquiterpene α-humulene with the versatile organism Cupriavidus necator pKR-hum on a growth medium from grass clippings. The medium was compared with established media in terms of microbial growth and terpene production. C. necator pKR-hum shows a maximum growth rate of 0.43 h−1 in the grass medium and 0.50 h−1 in a lysogeny broth (LB) medium. With the grass medium, 2 mg/L of α-humulene were produced compared to 10 mg/L with the LB medium. By concentrating the grass medium and using a controlled bioreactor in combination with an optimized in situ product removal, comparable product concentrations could likely be achieved. To the best of our knowledge, this is the first time that juice from grass clippings has been used as a growth medium without any further additives for microbial product synthesis. This use of green waste as a material represents a new bioeconomic utilization option of waste materials and could contribute to improving the economics of grass biorefineries.

A potential fucoidan-based PEGylated PLGA nanoparticles (NPs) offering a proper delivery of N-methyl anthranilic acid (MA, a model of hydrophobic anti-inflammatory drug) have been developed via the formation of fucoidan aqueous coating surrounding PEGylated PLGA NPs. The optimum formulation (FuP2) composed of fucoidan:m-PEG-PLGA (1:0.5 w/w) with particle size (365 ± 20.76 nm), zeta potential (−22.30 ± 2.56 mV), % entrapment efficiency (85.45 ± 7.41), drug loading (51.36 ± 4.75 µg/mg of NPs), % initial burst (47.91 ± 5.89), and % cumulative release (102.79 ± 6.89) has been further investigated for the anti-inflammatory in vivo study. This effect of FuP2 was assessed in rats’ carrageenan-induced acute inflammation model. The average weight of the paw edema was significantly lowered (p ≤ 0.05) by treatment with FuP2. Moreover, cyclooxygenase-2 and tumor necrosis factor-alpha immunostaining were decreased in FuP2 treated group compared to the other groups. The levels of prostaglandin E2, nitric oxide, and malondialdehyde were significantly reduced (p ≤ 0.05) in the FuP2-treated group. A significant reduction (p ≤ 0.05) in the expression of interleukins (IL-1β and IL-6) with an improvement of the histological findings of the paw tissues was observed in the FuP2-treated group. Thus, fucoidan-based PEGylated PLGA–MA NPs are a promising anti-inflammatory delivery system that can be applied for other similar drugs potentiating their pharmacological and pharmacokinetic properties.

A novel portable low-cost Arduino-controlled photo- and fluorimeter for on-site measurements has been developed. The device uses LEDs as a light source and a phototransistor as a light sensor. The circuit is based on the discharge of a capacitor with the photocurrent from the phototransistor. Validation experiments for absorbance measurements were performed by measuring protein concentration using the Bradford method and measuring phosphate ions in water using a commercial test kit. The emission light of the excited fluorescent dyes rhodamine 6G and calcofluor white was measured to validate the usability of the device as a fluorescence photometer. In all validation experiments, similar correlation coefficients and limit of detection could be achieved with the portable photo- and fluorimeter and a laboratory spectrometer and fluorimeter. Real sample analysis was performed, measuring phosphate concentration in freshwater and concentration of green fluorescent protein, extracted from Escherichia coli.

Freight transport accounts for about half of all distances travelled in Europe. Therefore, freight transport is one of the decisive factors for reducing greenhouse gases and air pollutants. For this reason, the electrification of road freight transport is being promoted as part of the project “BEV Goes eHighway—[BEE]”. The data basis for the modelling used in this project is an electric drive axle for a heavy commercial vehicle, which was developed in the “Concept-ELV2” project. Based on the results of the previous project, the methodological tools that were developed are presented in this paper. These allow a wide range of possible powertrain topologies to be considered at the concept stage of development based on an estimation of future system characteristics. For this purpose, the components are automatically designed taking into account the mutual influence of the requirements and are evaluated in the context of the holistic system. This publication focuses on the efficiency and thermal evaluation of the transmission stages of the addressed electric drive units and validates the developed models using a pototypically designed electric commercial vehicle axle.

Fucoidans encompass versatile and heterogeneous sulfated biopolysaccharides of marine origin, specifically brown algae and marine invertebrates. Their chemistry and bioactivities have been extensively investigated in the last few decades. The reported studies revealed diverse chemical skeletons in which l-fucose is the main sugar monomer. However, other sugars, i.e., galactose, mannose, etc., have been identified to be interspersed, forming several heteropolymers, including galactofucans/fucogalactans (G-fucoidans). Particularly, sulfated galactofucans are associated with rich chemistry contributing to more promising bioactivities than fucans and other marine polysaccharides. The previous reports in the last 20 years showed that G-fucoidans derived from Undaria pinnatifida were the most studied; 21 bioactivities were investigated, especially antitumor and antiviral activities, and unique biomedical applications compared to other marine polysaccharides were demonstrated. Hence, the current article specifically reviews the biogenic sources, chemistry, and outstanding bioactivities of G-fucoidans providing the opportunity to discover novel drug candidates.

Unspecific peroxygenases (UPOs) catalyze the selective transfer of single oxygen atoms from peroxides to a broad range of substrates such as un-activated hydrocarbons. Since specific oxyfunctionalizations are among the most-desired reactions in synthetic chemistry, UPOs are of high industrial interest. To broaden the number of available enzymes, computational and experimental methods were combined in this study. After a comparative alignment and homology modelling, the enzymes were expressed directly in P. pastoris. Out of ten initially selected sequences, three enzymes (one from Aspergillus niger and two from Candolleomyces aberdarensis) were actively expressed. Cultivation of respective expression clones in a bioreactor led to production titers of up to 300 mg L−1. Enzymes were purified to near homogeneity and characterized regarding their specific activities and pH-optima for typical UPO substrates. This work demonstrated that directed evolution is not necessarily required to produce UPOs in P. pastoris at respective titers. The heterologous producibility of these three UPOs will expand the toolbox of available enzymes and help to advance their synthetic application.

Phototrophic biofilms, in particular terrestrial cyanobacteria, offer a variety of biotechnologically interesting products such as natural dyes, antibiotics or dietary supplements. However, phototrophic biofilms are difficult to cultivate in submerged bioreactors. A new generation of biofilm photobioreactors imitates the natural habitat resulting in higher productivity. In this work, an aerosol-based photobioreactor is presented that was characterized for the cultivation of phototrophic biofilms. Experiments and simulation of aerosol distribution showed a uniform aerosol supply to biofilms. Compared to previous prototypes, the growth of the terrestrial cyanobacterium Nostoc sp. could be almost tripled. Different surfaces for biofilm growth were investigated regarding hydrophobicity, contact angle, light- and temperature distribution. Further, the results were successfully simulated. Finally, the growth of Nostoc sp. was investigated on different surfaces and the biofilm thickness was measured noninvasively using optical coherence tomography. It could be shown that the cultivation surface had no influence on biomass production, but did affect biofilm thickness.

The Nigeria 330KV integrated power network consisting of 52 buses, 64 transmission lines and sixteen generating stations was studied. The network was subjected to different reactive power loadings ranging from 25%, 50%, 75%, 100%, 125% and 150% respectively with and without UPFC and TCSC FACTS devices using Newton-Raphson power flow algorithm and Genetic Algorithm (GA) for optimally locating these devices on the network. The heavily loaded active power lines were identified. The heavily loaded active power lines are thirteen (13). These include: Katampe-Shiroro, Akangba-Ikejawest, Aiyede-Oshogbo, Alaoji-Onitsha, Egbin-ikejawest, Jebba-Oshogbo, Jebba-Shiroro, Jos-Makurdi, Kaduna-Shiroro, Kaduna-Kano, Ajaokuta-Benin, Benin-Ikejawest, and Benin-Onitsha. The ten(10) heavily loaded reactive power lines include: B-Kebbi-Kainji, Benin-Oshogbo, Benin-Egbin, Benin-Sapele, Damaturu-Gombe, Damaturu-Maiduguri, Gombe-Yola, Jalingo-Yola, Ikeja West-Oshogbo and Jos-New Haven. Weak buses were New-Haven(0.9201pu),Damaturu (0.9283pu), Gombe (0.9405pu), Maiduguri(0.9425pu), Makurdi (0.9260pu), Jalingo (0.9354pu),Erunkan(0.9314pu) and Jos (0.9461pu).Ten(10) heavily loaded reactive power lines include:B-Kebbi-Kainji, Benin-Oshogbo, Benin-Egbin, Benin-Sapele, Damaturu-Gombe, Damaturu-Maiduguri, Gombe-Yola, Jalingo-Yola, Ikeja West-Oshogbo and Jos-New Haven. Using GA for optimal location of these devices, eight (8) of the thirteen (13) lines were installed with UPFC. These are Akangba-Ikejawest, Alaoji-Onitsha, Jebba-Oshogbo, Jos-Makurdi, Kaduna-Kano, Ajaokuta-Benin, Benin-Ikeja west, and Benin-Onitsha . Also of the eight (8) weak buses six (6) had UPFC and the improved bus voltage values are Damaturu (0.9983pu), Gombe (0.9751 pu), Maiduguri (0.9815 pu), Makurdi (0.9804 pu), Jalingo (0.9541 pu) and Jos (0.9646 pu). Six (6) TCSC devices were incorporated on the ten (10) heavily loaded lines, which include Ikeja West-Oshogbo, Damaturu-Maiduguri, Gombe-Yola, Jos-New Haven, Benin-Oshogbo and BKebbi-Kainji. In situ radio-frequency heating (ISRFH) combined with soil vapor extraction was demonstrated at a contaminated field site of a former hydrotreatment plant in Zeitz near Leipzig. The project was carried out in several phases including cold soil vapor extraction for comparison. During the test, a soil volume of about 300 m3 was heated to an average temperature of 54 °C. As expected, the extraction rate for hydrocarbons (especially the main contaminant benzene) was markedly enhanced by soil heating. Furthermore, microbial degradation of organic compounds was supported. Although a total amount of approximately 1.4 t of hydrocarbons was removed from the soil, the demonstration project was not aimed at complete remediation of the site. Conditions limiting the extent of cleanup are discussed in detail and conclusions for an efficient application of ISRFH in soil remediation are derived whereas experiences from other sites are also implied. In situ radio-frequency heating (ISRFH) was successfully applied during remediation of a former petrol station. Using a three-electrode array in combination with extraction wells for soil vapor extraction (SVE), pollution consisting mainly of benzene, toluene, ethylbenzene, xylenes, and mineral oil hydrocarbons (in total about 1100 kg) was eliminated from a chalk soil in the unsaturated zone. Specially designed rod electrodes allowed selective heating of a volume of approximately 480 m3, at a defined depth, to a mean temperature of about 50 °C. The heating drastically increased the extraction rates. After switching off ISRFH, SVE remained highly efficient for some weeks due to the heat-retaining properties of the soil. Comparison of an optimized regime of ISRFH/SVE with conventional 'cold' SVE showed a reduction of remediation time by about 80 % while keeping the total energy consumption almost constant. Abstract: The heterogeneous catalytic wet oxidation of methyl tert-butyl ether (MTBE) with hydrogen peroxide, catalyzed by the iron-containing zeolites Fe-ZSM5 and Fe-Beta, was studied at ambient conditions and pH 7. The kinetics of MTBE degradation could be well-fitted to a pseudo-first-order model. Using Fe-ZSM5, the dependence of the reaction rate constant on hydrogen peroxide and catalyst concentration was determined. Furthermore, the formation and oxidation of tert-butyl alcohol and tert-butyl formate as intermediates of MTBE oxidation were studied. A comparison of the reaction rates of MTBE, trichloroethylene and diethyl ether in the Fe-ZSM5/H2O2 system revealed that adsorption plays a positive role for the degradation reaction. Comparing the two types of Fe-containing zeolites applied in this study, Fe-Beta showed a lower catalytic activity for H2O2 decomposition and also MTBE degradation. However, in terms of utilization of H2O2 for MTBE degradation Fe-Beta is advantageous over Fe-ZSM5. This could be explained by the stronger adsorptive enrichment of MTBE on the Fe-Beta zeolite. This study shows that Fe-containing zeolites are promising catalysts for oxidative degradation of MTBE by H2O2. [Copyright &y& Elsevier]
