ABSTRACT Antibiotic Resistance (ABR) is a global concern and while many students are aware of this issue, many of them are unclear on the mechanisms by which ABR may emerge. The mechanism of horizontal gene transfer is something many students are not familiar with. In this curriculum contribution we present 2 versions of an ‘interrupted case study’ that is designed as an introduction to horizontal gene transfer for early major students and as a review case for advanced major students in biology and life sciences. The case is based on an authentic patient who developed infections with both methicillin resistant Staphylococcus aureus and vancomycin resistant S. aureus. The interrupted case study is appropriate for small and large groups and engages students while content is introduced in a highly structured way. This type of case study can be done by novice and seasoned instructors and lead to considerable learning gains in both introductory and intermediate microbiology courses.

A digitally enabled environment is a setting which incorporates sensors coupled with reporting and analytics tools for understanding, observing or managing that environment. Large scale data collection and analysis are a part of the emerging digitally enabled approach for the characterisation and understanding of our environment. It is recognised as offering an effective methodology for addressing a range of complex and interrelated social, economic and environmental concerns. The development and construction of the approach requires advances in analytics control linked with a clear definition of the issues pertaining to the interaction between elements of these systems. This paper presents an analysis of selected issues in the field of analytics control. It also discusses areas of progress, and areas in need of further investigation as sensing networks evolve. Three case studies are described to illustrate these points. The first is a physical analytics test kit developed as a part of the “Reinvent the Toilet Challenge” (RTTC) for process control in a range of environments. The second case study is the Cranfield Urban Observatory that builds on elements of the RTTC and is designed to allow users to develop user interfaces to monitor, characterise and compare a variety of environmental and infrastructure systems plus behaviours (e.g., water distribution, power grids). The third is the Data and Analytics Facility for National Infrastructure, a cloud-based high-performance computing cluster, developed to receive, store and present such data to advanced analytical and visualisation tools.

Control of the reproduction of domesticated stocks is considered a prerequisite for aquaculture development of pikeperch. However, knowledge about the physiology of the captive pikeperch male reproductive system and the biology of semen is very limited, especially regarding protein characteristics. The aims of our study were to characterize pikeperch sperm quantity and quality parameters and to analyze changes in the proteome of the same males spawned for the first and second times. Moreover, attempts were made to generate the first proteomic library of seminal plasma proteins. Semen collected during the first spawning season was characterized by lower sperm concentration and volume than for the second season. Using mass spectrometry-based label-free quantitative proteomics, we identified 850 proteins in the seminal plasma of pikeperch from both spawning seasons, and 65 seminal proteins were found to be differentially abundant between the first and second spawning seasons. The majority of differentially abundant proteins were involved in stress and immune responses, developmental processes, cofactor metabolic processes, proteolysis, cellular oxidant detoxification and organization of the extracellular matrix (ECM). In addition, several proteins unique to pikeperch seminal plasma were identified, including antifreeze proteins, hibernation-specific plasma proteins, lectins and vitellogenin. In summary, our results indicate that males that spawned for the first time were characterized by incompletely mature gonads and the expression of proteins associated with the early phase of spermatogenesis and ECM organization. On the other hand, males that spawned for the second time exhibited advanced gonadal maturation and expression of proteins related to the late stage of spermatogenesis and sperm maturation, including regulation of reactive oxygen species generation, bicarbonate production, sperm elongation and separation. The identification of a large number of seminal plasma proteins provides a valuable resource for understanding the functions of seminal plasma and the molecular mechanisms involved in testicular development and maturation in domesticated fish, which is a prerequisite for better control of reproduction in captivity.

Society relies on infrastructure, but as infrastructure systems are often collocated and interdependent, they are vulnerable to cascading failures. This study investigated cross-infrastructure and societal impacts of burst water mains, with the hypothesis that multi-infrastructure failures triggered by burst water mains are more common in sandy soils. When water mains in sandy soils burst, pressurised water can create subsurface voids and abrasive slurries, contributing to further infrastructure failures. Three spatial data investigations, at nested scales, were used to assess the influence that soil sand content has on the frequency and damage caused by burst water mains (1) to roads in the county of Lincolnshire, (2) to other proximal water mains in East Anglia and (3) to other proximal infrastructure and wider society across England and Wales. These investigations used infrastructure network and failure data, media reports and soil maps, and were supported by workshop discussions and structured interviews with infrastructure industry experts. The workshop, interviews and media reports produced a greater depth of information on the infrastructure and societal impacts of cascading failures than the analysis of infrastructure data. Cross-infrastructure impacts were most common on roads, built structures and gas pipes, and they occurred at a higher rate in soils with very high sand contents.

Alkaptonuria is a rare genetic disorder characterized by a high level of circulating (and urine) homogentisic acid (HGA), which contributes to ochronosis when it is deposited in connective tissue as a pigmented polymer. In an observational study carried out by National AKU Centre (NAC) in Liverpool, a total of thirty-nine AKU patients attended yearly visits in varying numbers. At each visit a mixture of clinical, joint and spinal assessments were carried out and the results calculated to yield an AKUSSI (Alkaptonuria Severity Score Index), see “Nitisinone arrests ochronosis and decreases rate of progression of Alkaptonuria: evaluation of the effect of nitisinone in the United Kingdom National Alkaptonuria Centre” (Ranganath at el., 2018). The aim of this data article is to produce visual representation of the change in the components of AKUSSI over 3 years, through radar charts. The metabolic effect of nitisinone is shown through box plots.

In this study, the quality of northern pike eggs collected by the traditional method (hand stripping) and the pneumatic method (air stripping) was compared. Additionally, different activation solutions (Billard solution, hatchery water, and Woynarovich solution) were tested for their effects on egg fertilization under artificial conditions. After the eggs were collected, the Pseudo-Gonado-Somatic Index (PGSI) was measured. Although the values of the PGSI in the samples obtained with use of air stripping were lower (13.8 ± 3.9%), they did not differ statistically from those obtained by hand stripping (16.5 ± 5.4%). Hatchery water and Woynarovich solution were found to be the most suitable solutions for sperm activation using the Computer-Assisted Sperm Analysis (CASA) system as compared to the Billard solution. Hand stripping was found to negatively affect the percentage of fertilized eggs and the percentage of hatched larvae in samples fertilized in hatchery water and Woynarovich solution. When the traditional method of egg collection was used, there were no differences in the percentage of fertilization and the percentage of hatched larvae between Billard solution (54.0 ± 21.5% and 44.1 ± 21.9%, respectively), hatchery water (60.0 ± 22.5% and 55.9 ± 22.8%, respectively), and Woynarovich solution (72.0 ± 25.8% and 69.0 ± 23.9%, respectively) treatments. Air stripped eggs showed a higher fertilization rate when hatchery water or Woynarovich solution was applied (86.2 ± 9.3% and 92.4 ± 3.9%, respectively). Also hatching rate was the highest in these samples (83.0 ± 8.4% and 88.3 ± 6.2%). The application of the pneumatic method and Woynarovich solution to northern pike artificial fertilization resulted in higher fertilization and hatching rates as compared to other techniques. Because this was successful in northern pike, the use of air stripping is a promising option for artificial reproduction in other fish species.

The drier parts of the Mediterranean biome of southwest Australia contain the largest remaining Mediterranean woodlands and shrublands on Earth. Despite this, there has been no formal, comprehensive assessment of their biodiversity. The region abuts the southwest Australian floristic region which has received much scientific attention. The aim of this paper is to provide the first general overview of the biodiversity of part of this intact, yet relatively unknown, Mediterranean ecosystem. We do this by synthesizing data from State Government agencies and published research. We found that, like other parts of southwest Australia, the region has globally significant levels of plant species diversity. More than 2400 plant species, including 291 species considered threatened, have been recorded, representing one-sixth of all Australia’s vascular plant species. Eleven of Australia’s 23 major vegetation groups are represented even though the region covers less than 1% of continental Australia. We documented 170 vertebrate species, including 31 threatened species, with a particularly high richness of reptile species (n = 46). We highlight how little is known about this region. For example, 116 vertebrate species not recorded in the region probably occur there based on their habitat requirements and known distributions. An examination of plant and vertebrate diversity in the region, using a half degree latitude and longitude grid cells, showed a highly heterogeneous pattern of species richness and vulnerability, with a general decline in species richness from southwest to northeast. Conservation strategies that rely on capturing the highest levels of biodiversity in a series of protected areas are unlikely to guarantee protection for all species given these high levels of heterogeneity. Instead, a region-wide conservation plan should involve targeted ecological research, consideration of ecological processes and stakeholder consultation.

Extended-spectrum beta-lactamase-producing Enterobacteriaceae (ESBL-E) among animals and humans are a public health threat. This study analyzed the occurrence of ESBL-E in a high-risk environment in a companion animal clinic and two animal patients’ households. In an intensive care unit (ICU), rectal swabs from 74 dogs and cats, 74 hand swabs from staff and 298 swabs from surfaces were analyzed for ESBL-E. Seventeen hospitalized patients (23%) and ten (3%) surfaces in the ICU tested ESBL-E positive. Transmission chains for Klebsiella pneumoniae ST307 blaCTX-M-15 and Escherichia coli ST38 blaCTX-M-14, ST88 blaCTX-M-14 and ST224 blaCTX-M-1 were observed over extended periods of time (14 to 30 days) with similar strains isolated from patients and the clinical environment. After discharge, two colonized dogs (dogs 7 and 12) and their household contacts were resampled. Dog 7 tested repeatedly positive for 77 days, dog 12 tested negative; six (24%) surfaces in the household of the persistently colonized dog tested ESBL-E positive. The owner of dog 7 and one of the owners of dog 12 were colonized. Based on whole genome sequencing, isolates from the owners, their dogs and other ICU patients belonged to the same clusters, highlighting the public health importance of ESBL-E in companion animal clinics.
