Abstract

A hybrid photodetector (HPD) consisting of a photocathode and a multipixel avalanche diode (MP-AD) was developed a few years ago. Our previous studies showed that its inherent potential for high resolution photon counting could be further enhanced by reducing fluctuations in charge loss in the dead layer at the entrance of the MP-AD. In this paper, we report on the improvement with the newly developed HPD whose encapsulated MP-AD has a thinner dead layer than before. It is demonstrated that the new HPD has much better energy resolution, which enables clearer counting up to nine photoelectrons. Further enhancement of the photocathode sensitivity of the HPD is also discussed. Index Terms--Hybrid photodetctor, HPD, multipixel, photon sensor, single photon.

Abstract

A large cylindrical transition radiation detector has been constructed as a part of the Venus detector for the TRISTAN |e.sup.+~|e.sup.-~ collider. It was designed to supplement the e/|Pi~ separation capability of the lead glass calorimeter. It is one of the largest transition radiation detector in the world at present. The detector has a total of 2,688 channels of readout electronics. The electronics employs the method of charge sensitive preamplifier-trapezoidal shaping. Each TKO board contains 32 channels of shaper-gated integrators and a 12-bit ADC. This paper describes full details of the readout electronics, the electronics for wire-gain control and the performance of the detector.

Abstract

We made a series of surveys to obtain neutrino beam line direction toward SuperKamiokande (SK) at a distance of 250 km for the long-baseline neutrino oscillation experiment at KEK. We found that the beam line is directed to SK within 0.03 mr and 0.09 mr (in sigma) in the horizontal and vertical directions, respectively. During beam operation, we monitored the muon distribution from secondary pions produced at the target and collected by a magnetic born system. We found that the horn system functions like a lens of a point-to-parallel optics with magnification of approximately -100 and the focal length of 2.3 m. Namely, a small displacement of the primary beam position at the target is magnified about a factor -100 at the muon centroid, while the centroid position is almost stable against a change of the incident angle of the primary beam. Therefore, the muon centroid can be a useful monitor of the neutrino beam direction. We could determine the muon centroid Within 6 mm and 12 mm in horizontal and vertical directions, respectively. This means that the neutrino beam direction could be controlled within 0.03 mr and 0.06 mr (in sigma) In the horizontal and vertical directions, respectively. Abstract

Invasive potentials of malignant cancer cells are regulated by cell motility factors. To examine the regulation of motility and invasiveness in oral squamous carcinoma, we investigated autocrine- and/or paracrine-αcting cell motility factors, using a newly established human cell line (IF cells) from oral squamous cell carcinoma, which has highly invasive and metastatic characteristics. Conditioned medium derived from IF cells stimulated cell scattering and migration of GB-d1 gallbladder carcinoma cells, indicating that IF cells secreted cell motility factors. Using antibodies, IF-derived cell motility factors proved to be transforming growth factor (TGF)-α and TGF-β1. Antibodies against TGF-α and TGF-β1 inhibited autonomous migration of the IF cells. On the other hand, in vitro invasion of IF cells was strongly enhanced by hepatocyte growth factor (HGF) but only slightly by TGF-α and TGF-β1. The conditioned medium from fibroblasts enhanced in vitro invasion of IF cells, an event abrogated by anti-HGF antibody, but not by antibodies against TGF-α and TGF-β1. Importantly, IF cells secreted a factor inducing HGF production in fibroblasts and the factor was identified as interleukin-1, which means that a mutual interaction exists between tumour cells and fibroblasts, as mediated by the HGF/HGF-inducer loop. These results indicate that IF cells utilize TGF-α and TGF-β1 as autocrine-αcting motility factors and HGF as a paracrine-αcting motility factor, and that invasiveness of IF cells is particularly stimulated by HGF derived from stromal fibroblasts. Utilization of multiple cell motility/invasion factors that act in distinct pathways may confer highly invasive and metastatic potentials in IF oral squamous carcinoma cells. [ABSTRACT FROM AUTHOR]

Abstract

Cheek pressure during swallowing was measured on six operated complete cleft lip and palate cases (4-8 years of age) before and alter expansion of buccal segments of maxillary arch. As control, seven non-cleft subjects with acceptable occlusion of the same age level were used. Before expansion, the pressures in tile cleft eases were much higher than the controls. Immediately alter expansion, increase in pressure was observed in eases which received rapid expansion. However, a few months after expansion, most cases showed a decrease in pressure. This is explained as being due to the adaptation of the perioral musculature to the expanded dental arch and/or the disappearance of hyperactivity of the perioral musculature during swallowing. Abstract

It has often been hypothesized that a large tongue leads to an enlargement of the mandible and therefore contributes to the development of mandibular prognathism. We examined (1) whether the tongue volume in human subjects with mandibular prognathism was larger than that in subjects with good occlusion and (2) whether the tongue volume and the pharyngeal capacity correlated with the morphological characteristics of dento-skeletal structures. Magnetic resonance images of the tongue and its surrounding structures were recorded for female adult volunteers with good occlusion (control group, n = 10) and patients with mandibular prognathism (test group, n = 16). Lateral cephalograms were obtained for the patients. No significant differences were determined for the tongue volume or the pharyngeal capacity between the two groups. The tongue volume did not correlate with the pharyngeal capacity (r =0.280, p = 0.166). The tongue volume correlated with the facial angle (r = 0.548, p = 0.028), the Y-axis (r = 0.539, p = 0.031), and the angle nasion-A point-pogonion (r = 0.540, p = 0.031). These results suggest that the tongue volume is accounted for by the combined horizontal and vertical location of the chin and symphysis, but do not support the conventional clinical surmise that large tongue volume is inherent in patients with mandibular prognathism. Abstract

The electromyographic (EMG) activity of the human genioglossus (GG) muscle during chewing efforts is not fully understood. In this study, the EMG activity of the human GG muscle during unilateral gum chewing was illustrated and correlated with the activities in the anterior temporalis (AT), the anterior digastric (DC), and the inferior orbicularis oris (OI) muscles. GG muscle activity was measured with customized surface electrodes, while other muscles were recorded with conventional surface electrodes. EMG activities during tongue displacement and the articulation of long vowels, recorded by the customized electrodes, were consistent with the recordings obtained by fine wire electrodes placed in the GG muscle. Jaw displacement was monitored by means of a kinesiograph with a transducer attached to the mandibular central incisors. Mean normalized GG muscle activity showed an onset in the last one-fifth of the intercuspal phase, gradually increasing during jaw-opening, and at its greatest immediately before the maximum jaw-opening position. It then decreased during jaw-closing and ceased in intercuspation but showed a small rebound in the third fifth of the intercuspal phase. The GG muscle burst showed phase lags with the DG and OI muscles and an opposite phase with the AT muscle (all P < 0.0001). All correlations were statistically significant (all P < 0.0001, r values between 0.88 and 0.97). Abstract

The effect of biomechanical force on growth of skeletal tissue was studied in monolayer cultures of mouse osteoblastic MC3T3-E1 cells which were centrifuged at 320 g for 15 min to 72 h in a CO2 incubator. Centrifugation of the cells for 30 min in low concentrations (0.3 or 1%) of fetal bovine serum (FBS) caused a two-fold increase of [³H]thymidine incorporation at 20 h from the start of centrifugation. However, centrifugation under 10% FBS caused no increase in [³H]thymidine incorporation into DNA. Under 0.3% FBS, [³H]thymidine incorporation increased in a manner dependent on the period of centrifugation and reached a maximum when the cells were centrifuged for 3 h. Stimulation of DNA synthesis by centrifugation was abolished in the presence of H-7, an inhibitor of protein kinase C. Moreover, conditioned medium collected from the centrifuged cultures increased [³H]thymidine incorporation by two-fold over the basal when added to a quiescent culture of MC3T3-E1 cells. These results suggest that centrifugal force stimulates growth of osteoblastic cells through autocrine secretion of some diffusible growth-promoting activity. On the other hand, centrifugation of the cells inhibited induction by FBS of alkaline phosphatase activity and calcium-uptake, two indices of the differentiated phenotype of osteoblasts. Abstract

A 2-mm non-healing bony defect was prepared in the premaxilla of male Wistar rats weighing about 180 g as a simulation of an alveolar cleft, for determination of whether a pulsing electromagnetic field (PEMF) could promote regeneration of bone induced by demineralized bone matrix (DBM). The defect was either treated with 7 mg DBM or was left as a non-grafted control. The rats were exposed to a PEMF with a frequency of 100 Hz, a 10-ms-wideburst with 100 µs-wide quasi-rectangular pulses, repeating at 15 Hz, and magnetic field strength of 1.5-1.8 G. Alkaline phosphatase activity increased significantly from day 7 in the DBM-graft-plus-PEMF group and from day 10 in the DBM-graft group, reaching a maximum on day 14. A greater-than-two-fold rise in alkaline phosphatase activity and a three-fold rise in the amount of 45Ca incorporation in the DBM-graft- plus-PEMF group were attained compared with those of the DBM-graft group. The DBM-graft-plus-PEMF group produced more bone with almost complete osseous bridging in the defect sites than did the group treated with DBM only on day 35. The findings indicate that PEMF had an enhancing effect on the bone-inductive properties of the DBM through the stimulation of osteoblast differentiation induced by DBM. [ABSTRACT FROM AUTHOR]

Abstract

Orthodontic tooth movement requires remodeling of periodontal tissues, especially alveolar bone. 1,25-(OH)2D3, the active form of vitamin D3, is known to be a potent stimulator of osteoclastic bone resorption. The purpose of this study was to investigate the effect of local application of 1,25-(OH) )2D3 on osteoclast numbers induced by experimental tooth movement. A piece of orthodontic elastic band was inserted between the first and second upper molars of male Wistar rats weighing about 200 g each. Twenty μL of 1,25-(OH) 2D3 (10-12-10-7 mol/L) was injected locally into the submucosal palatal area of the root bifurcation of the right first molar. The left side was injected with phosphate-buffered saline (PBS). The number of osteoclasts was counted in a 700 x 1050 µm² area of the inter-radicular septum. The local injection of 1,25-(OH) )2D3 caused a dose-dependent increase in osteoclast number. The effect of 1,25-(OH) 2D3 reached a response plateau at 10-10 mol/L when greater than a three-fold rise in osteoclast number was attained compared with the PBS-treated controls. While the insertion of a piece of elastic band for three days induced a significant increase in osteoclasts in the alveolar bone, daily injections of 20µL of 10-10 mol/M 1,25-(OH) 2D3 for three days markedly stimulated the numbers of osteoclasts induced by the insertion of an elastic band. 1,25-(OH)2D3 was apparently synergistic with mechanical stimuli, resulting in enhancement of the numbers of osteoclasts induced by mechanical stimuli alone. These findings suggest that the local application of 1,25-(OH) )2D3 acted directly to increase osteoclast number and to potentiate osteoclastic bone resorption induced by mechanical stimuli. Abstract

The present study was designed to measure the elastic properties of temporomandibular joint (TMJ) discs from six adult dogs. Each disc was divided mediolaterally into medial, middle, and lateral parts. Under tension, the articular disc exhibited a non-linear stress-strain relationship, which could be represented as two lines (two moduli of elasticity) connected at a point of stress around 1.5 MPa. These two elastic moduli of the disc were approximately 44 Mpa and 92 MPa in the lower- and higher-stress regions, respectively. Elastic moduli of the articular disc in the middle area were significantly different from that in the lateral area of the disc. Abstract

So that the effects of biomechanical forces on the proliferation of chondrocytes and their proteoglycan synthesis could be studied, growth-plate and articular chondrocytes were maintained separately as packed masses in centrifuge tubes in the presence of 10% serum. In these conditions, the cells became re-organized into cartilaginous tissue in seven days. After ten days, they were centrifuged at gravities (g) of 1.3-27 for 24 h in a CO2 incubator. Control cells were maintained in the CO2 incubator without centrifugation. Centrifugation of growth-plate chondrocytes at 3 g resulted in a two-fold increase in incorporation of [35S]sulfate into proteoglycans, but had little effect on their [³H]thymidine incorporation into DNA. On the other hand, centrifugation of articular chondrocytes at 3 g for 24 h caused 1.5-fold increases in both [35S]sulfate incorporation into proteoglycans and [³H]thymidine incorporation into DNA. Abstract

Previously, we reported methods for isolating chondrocytes from the craniofacial complex and their culture in vitro. The response of these chondrocyte cultures to bovine parathyroid hormone (1-34) (PTH) has now been investigated. PTH stimulated glycosaminoglycan (GAG) synthesis, a characteristic of the cartilage phenotype in cultured chondrocytes isolated from mandibular condylar cartilage (MCC), nasal septal cartilage (NSC), and spheno-occipital synchondrosis (SOS). These stimulations of GAG synthesis by PTH were dose-dependent. PTH also increased accumulation of cyclic AMP (cAMP) and the activity of ornithine decarboxylase (ODC), a rate-limiting enzyme in polyamine biosynthesis. However, PTH did not stimulate DNA synthesis. The increases in the cAMP level, ODC activity, and GAG synthesis after addition of PTH(10-7 mol/L) were greatest in MCC-chondrocytes and least in NSC-chondrocytes. Abstract

Cross-sectional data were obtained from 1182 Japanese high school girls of 12 and 15 years old. The data were analysed for the prevalence of specific occlusal features and the prevalence of cardinal Temporomandibular Joint Dysfunction (TMJD) symptoms in different occlusion groups. The associations between occlusal features and TMJD symptoms were analysed by the Z-analysis. The results were as follows: the prevalence of TMJD symptoms (total) was about 23% in both age groups examined. Noise had the highest prevalence of the specific TMJD symptoms in both age groups (16% and 11%). With age and for different occlusion groups, there was a significant increase in deviation on opening (P< 0.001) and a decrease in pain and noise (P < 005). No significant associations were found between the occurrence of TMJD symptoms (total) and occlusal features in all the examined occlusion groups. With respect to specific TMJD symptoms, however, the investigation of the different groups revealed a significant association (P < 0.05) of intra-arch occlusal (crowding) features with the occurrence of deviation on opening and with the occurrence of the combined TMJD symptoms which include deviation on opening. This indicates that crowding may be an important sign for predisposing TMJD. For the other single disorder symptoms (except pain), we did not find a significant relationship (P<0.05) with aspects of occlusion, which may confirm a multifactorial TMJD problem wherein occlusal features can have a contribution to TMJD. Abstract

Lactobacillus -fermented milk can stimulate anabolic effects in skeletal muscle. Fermented milk containing Lactobacillus produces aqueous molecules, such as free AA and lactate. This study aimed to investigate how processing fermented milk by centrifugation and removal of supernatant affects AA absorption and postprandial skeletal muscle protein synthesis (MPS) when mice are fed fermented milk. We gavaged male Sprague-Dawley rats with skim milk (S), fermented milk (F), or processed fermented milk (P), and examined the total AA content in portal vein blood (reflecting AA absorption) and plantaris muscle MPS at 30, 60, and 90 min following administration. Relative to fasted rats, at 30 min the total AA concentration in portal vein blood from rats in the P groups was significantly higher, followed by F and S, respectively. The MPS rates were higher for the F or P groups compared with the S group. Phosphorylation levels of p70S6 kinase in the P and F groups were significantly higher than those for the S group 30 min after administration, although the level of Akt phosphorylation was similar among the groups. These results suggested that fermentation improves AA absorption that in turn enhances postprandial MPS via Akt-independent mechanisms, and that processed fermented milk retains these favorable effects on MPS.
