We describe the isolation and characterisation of 17 microsatellite loci for the red-billed chough ( Pyrrhocorax pyrrhocorax, Corvidae). Sixteen loci were polymorphic in 269 individuals from across Western Europe, with a mean allele number of 8.75 ± 3.73 SD. Observed (H) and expected (H) heterozygosity ranged from 0.11 to 0.71 and 0.15 to 0.70, respectively. No evidence was found for null-alleles or linkage-disequilibrium. Cross-species utility was tested on 15 Alpine choughs ( Pyrrhocorax graculus) and five jackdaws ( Corvus monedula). Sixteen loci amplified for Alpine chough and fifteen loci amplified for jackdaw, indicating useful application within and beyond the Pyrrhocorax genus. [ABSTRACT FROM AUTHOR]

Black grouse in Britain have faced contraction of their range and have declined in numbers during the recent decades. As such, the species is a conservation concern in the UK. In order to aid conservation decisions, the terms Evolutionary Significant Unit (ESU) and Management Unit (MU) have been proposed. An ESU is an independently evolving evolutionary lineage defined by being reciprocally monophyletic for mitochondrial alleles, and which is significantly different from other lineages with regard to nuclear alleles, whereas an MU is operationally defined by only the latter criterion. Using mitochondrial sequences and nuclear microsatellite loci, we failed to find evidence that British black grouse is an ESU. However, British black grouse are sufficiently different from continental black grouse both with respect to mitochondrial and nuclear data to regard them as a separate MU. Furthermore, we present genetic data which suggest that British black grouse presently occur in what are probably three demographically independent units (roughly corresponding to Wales, northern England/southern Scotland and northern Scotland), which are genetically differentiated. Microsatellites are DNA-fragments containing short repetitive motifs with 2–10 bp. They are highly variable in most species and distributed throughout the whole genome. It is broadly accepted that their high degree of variability is closely associated with mispairing of DNA-strands during the replication phase, termed slippage, although recombination is also observed. The aim of this study is to demonstrate evidence that non-reciprocal recombination processes changing the total genomic structure are common in microsatellites and flanking regions. We sequenced DNA fragments from birds in which microsatellites are located, and analyzed the structure of the microsatellites and their flanking regions. Additionally, other data and those from literature of three microsatellite regions of primates coding for the Ataxin-2, the Huntingtin and the TATA-box binding protein were analyzed. The structures of seven avian and three primate microsatellites support the hypothesis that non-reciprocal recombination is a common process that may also contribute considerably to the variation at microsatellite loci. We conclude that results of population genetic studies that are analyzed statistically with methods based on stepwise mutation models should be interpreted with caution if no detailed information on the allelic variation of microsatellites is available. Genetic heterogeneity is considered an important parameter for individual fitness and reproductive success. In 1999 and 2000, we studied the population genetics of great tit ( Parus major L., 1758) in southwestern Germany from two different forest types (deciduous and mixed-coniferous), which may significantly differ in prey diversity and/or food supply. Adults of 99 families were genotyped at four enzyme and eight microsatellite loci, in order to estimate individual heterozygosity. In the mixed-coniferous forest, a significant positive correlation between the genetic heterogeneity of females and early egg-laying date and clutch size was detected. Early egg-laying date and increased clutch size are conditions that positively affect the number of fledglings. This effect of individual heterozygosity was not observed in the deciduous woodland. Maternal genetic heterogeneity, however, did not correlate with fledgling condition, and individual heterozygosity of fathers had no impact on breeding success in either habitat. The positive effect of female genetic heterogeneity on brood size of great tits in mixed-coniferous forests is attributed to early egg-laying date, i.e. a maternal effect, rather than to a specific mating strategy that optimizes fitness through an increased brood size and the quality of offspring. Abstract Genetic diversity and population structure were studied in eight populations of the kestrel Falco tinnunculus to identify the genetic consequences of spatial distribution and to infer the colonization patterns of the Cape Verde archipelago. We studied genetic differentiation and gene flow among seven island populations and one mainland population using nine microsatellite loci. Within the archipelago, differentiation was strong and genetic diversity and heterozygosity were low but variable among populations. Two subspecies F. tinnunculus neglectus on the northwestern islands and F. tinnunculus alexandri on all the other islands were identified as genetically distinct units. F. t. alexandri could be further separated into two groups on eastern and southern islands. Populations are probably founded by birds originating from the mainland. Immigration is more likely to the eastern and southern populations, whereas the northwestern islands with the lowest genetic diversity and highest differentiation are likely to exhibit fewer founding events by immigrants. The number of founding events on each island may depend not only on geographical distance to neighbouring populations, but also on directional immigration due to the northeastern trade winds. This may explain differences in genetic differentiation and diversity between populations and subspecies and may enable allopatric speciation. [ABSTRACT FROM AUTHOR]

Abstract The capercaillie inhabits a continuous range in large parts of the Palearctic boreal forest, but is patchily distributed in temperate Europe. An ongoing population decline, largely related to human land use changes, has been most pronounced in central and western Europe, where some local populations have become extinct. In this study, we document the genetic differentiation of capercaillie populations at different stages along a gradient of spatial structuring from high connectivity (continuous range in the boreal forest) to a metapopulation systems (Alps) and recent (central Europe) and historic (Pyrenees) isolation. Four hundred and sixty individuals from 14 sample sites were genotyped at 10 polymorphic microsatellite loci to assess genetic structure and variation of capercaillie populations across its European range. As expected, differentiation was least pronounced within the continuous range in the boreal forest. Within the metapopulation system of the Alps, differentiation was less than among the isolated populations of central Europe (Black Forest, Fichtelgebirge, Thuringia, Vosges). In the long-isolated population of the Pyrenees, and the recently isolated populations of central Europe, genetic diversity was significantly reduced compared with the Alps and boreal forest. Our results agree with the concept of a gradual increase in genetic differentiation from connectivity to isolation, and from recent to historic isolation. Anthropogenic habitat deterioration and fragmentation thus not only leads to range contractions and extinctions, but may also have significant genetic and evolutionary consequences for surviving populations. Noninvasive samples are useful for molecular genetic analysis of free-ranging animals. I tested whether moulted feathers collected in the field are a reliable source of DNA for genotyping microsatellite loci. I prescreened extracts for DNA quantity and, using only samples with higher amounts of DNA, obtained reliable genotyping results. Polymerase chain reaction (PCR) amplification success was higher from extracts of plucked feathers than moulted feathers. DNA quantity in larger feathers was higher than that in smaller feathers. Abstract Background While the gene flow in some organisms is strongly affected by physical barriers and geographical distance, other highly mobile species are able to overcome such constraints. In southern South America, the Andes (here up to 6,900 m) may constitute a formidable barrier to dispersal. In addition, this region was affected by cycles of intercalating arid/moist periods during the Upper/Late Pleistocene and Holocene. These factors may have been crucial in driving the phylogeographic structure of the vertebrate fauna of the region. Here we test these hypotheses in the burrowing parrot Cyanoliseus patagonus (Aves, Psittaciformes) across its wide distributional range in Chile and Argentina. Results Our data show a Chilean origin for this species, with a single migration event across the Andes during the Upper/Late Pleistocene, which gave rise to all extant Argentinean mitochondrial lineages. Analyses suggest a complex population structure for burrowing parrots in Argentina, which includes a hybrid zone that has remained stable for several thousand years. Within this zone, introgression by expanding haplotypes has resulted in the evolution of an intermediate phenotype. Multivariate regressions show that present day climatic variables have a strong influence on the distribution of genetic heterogeneity, accounting for almost half of the variation in the data. Conclusions Here we show how huge barriers like the Andes and the regional environmental conditions imposed constraints on the ability of a parrot species to colonise new habitats, affecting the way in which populations diverged and thus, genetic structure. When contact between divergent populations was re-established, a stable hybrid zone was formed, functioning as a channel for genetic exchange between populations.

Abstract Background Mating outside the pair-bond is surprisingly common in socially monogamous birds, but rates of extra-pair paternity (EPP) vary widely between species. Although differences in life-history and contemporary ecological factors may explain some interspecific variation, evolutionary forces driving extra-pair (EP) mating remain largely obscure. Also, since there is a large phylogenetic component to the frequency of EPP, evolutionary inertia may contribute substantially to observed EP mating patterns. However, the relative importance of plasticity and phylogenetic constraints on the incidence of EP mating remains largely unknown. Results We here demonstrate very low levels of EPP (4.4% of offspring) in the purple-crowned fairy-wren Malurus coronatus, a member of the genus with the highest known levels of EPP in birds. In addition, we show absence of the suite of distinctive behavioral and morphological adaptations associated with EP mating that characterize other fairy-wrens. Phylogenetic parsimony implies that these characteristics were lost in one speciation event. Nonetheless, many life-history and breeding parameters that are hypothesized to drive interspecific variation in EPP are not different in the purple-crowned fairy-wren compared to its promiscuous congeners. Conclusion Such radical loss of an extreme EP mating system with all associated adaptations from a lineage of biologically very similar species indicates that evolutionary inertia does not necessarily constrain interspecific variation in EPP. Moreover, if apparently minor interspecific differences regularly cause large differences in EPP, this may be one reason why the evolution of EP mating is still poorly understood.
