Simple Summary: Marek's disease virus is a pathogen capable of inducing tumors, immunosuppression, and paralysis in birds. The degree of virulence is associated with oncogenes such as the meq gene, which is constantly subjected to mutations that can exacerbate clinical manifestations and aid immune escape. This study reports the detection of this virus in backyard and ornamental birds from Brazil and Peru. Through molecular and evolutionary analyses, we determined the circulation of various strains with distinct genetic virulence profiles. Additionally, we identified potential sources of introduction and distribution of this virus among the studied birds, as well as a link between backyard and commercial birds. Given that different bird species exhibit varying degrees of susceptibility or resistance to this virus, the detection and characterization of these strains are crucial for monitoring and preventing potential epidemiological outbreaks in other species. This study highlights the importance of monitoring and characterizing circulating MDV in backyard and ornamental birds, as they can act as reservoirs for future epidemiological outbreaks. Silkie chicken, an important chicken breed with high medicinal and nutritional value, has a long history of being used as a dietary supplement in China. However, the compounds with health-promoting effects in Silkie chickens remain unclear. In the present study, we conducted a comprehensive analysis of metabolic and lipidomic profiles to identify the characteristic bioactive compounds in Silkie chickens, using a common chicken breed as control. The results showed that the levels of 13 metabolites including estradiol, four lipid subclasses including cardiolipin (CL), eight lipid molecules, and three fatty acids including docosahexaenoic acid (C22:6) were significantly increased in Silkie chickens, which have physiological activities such as resisting chronic diseases and improving cognition. These characteristic bioactive compounds have effects on meat quality characteristics, including improving its water-holding capacity and umami taste and increasing the content of aromatic compounds and phenols. The differentially expressed genes (DEGs) between the two chicken breeds revealed the regulatory network for these characteristic bioactive compounds. Fifteen DEGs, including HSD17B1, are involved in the synthesis of characteristic metabolites. Eleven DEGs, including ELOVL2, were involved in the synthesis and transport of characteristic lipids and fatty acids. In summary, we identified characteristic bioactive compounds in Silkie chickens, and analyzed their effects on meat quality characteristics. The Silkie chicken (Gallus gallus domesticus Brisson) is one of domestic chicken breeds with commercial rearing and breeding potentials for egg production. Prolactin (PRL) and dopamine D2 receptor (DRD2) are potential genes associated with reproductive traits in chickens. This study was conducted to analyze the association of PRL and DRD2 insertion/deletion (Indel) polymorphisms with chicken reproductive traits in Silkie chickens. A total of 380 hens from 16-40 weeks of age were used, with each one being placed in a separate cage. DNA isolation was performed using feather samples, and genotypes were detected using the Indel technique. Two polymorphisms consisting of 24 base pair (bp) Indel in the promoter region of the PRL gene and 22 bp Indel in the promoter region of the DRD2 gene were identified. At both sites, the Indel polymorphisms did not follow the Hardy-Weinberg equilibrium. In addition, with the exception of total eggs over 23 weeks of laying in the PRL gene, the analysis revealed no association between these polymorphic loci and any traits collected. In conclusion, birds with the DD genotype produced the maximum egg yield (73.6 eggs/hen), whereas those with the II genotype produced approximately 9 fewer eggs (64.1 eggs/hen), resulting in laying rates of 45.7% and 40.1%, respectively. For enhancing the egg-laying capacity of Silkie chickens via selective breeding, opting for DD birds with DD genotype of PRL Indel is highly recommended. [ABSTRACT FROM AUTHOR]

The tetrapod limb has long served as a paradigm to study vertebrate pattern formation and evolutionary diversification. The distal part of the limb, the so-called autopod, is of particular interest in this regard, given the numerous modifications in both its morphology and behavioral motor output. While the underlying alterations in skeletal form have received considerable attention, much less is known about the accompanying changes in the neuromuscular system. However, modifications in the skeleton need to be properly integrated with both muscle and nerve patterns, to result in a fully functional limb. This task is further complicated by the distinct embryonic origins of the three main tissue types involved—skeleton, muscles and nerves—and, accordingly, how they are patterned and connected with one another during development. To evaluate the degree of regulative crosstalk in this complex limb patterning process, here we analyze the developing limb neuromuscular system of Silkie breed chicken. These animals display a preaxial polydactyly, due to a polymorphism in the limb regulatory region of the Sonic Hedgehog gene. Using lightsheet microscopy and 3D-reconstructions, we investigate the neuromuscular patterns of extra digits in Silkie wings and legs, and compare our results to Retinoic Acid-induced polydactylies. Contrary to previous findings, Silkie autopod muscle patterns do not adjust to alterations in the underlying skeletal topology, while nerves show partial responsiveness. We discuss the implications of tissue-specific sensitivities to global limb patterning cues for our understanding of the evolution of novel forms and functions in the distal tetrapod limb.

ABSTRACT: The use of antibiotics leads to antibiotic residues in livestock and poultry products, adversely affecting human health. Ciprofloxacin (CFX) is a broad-spectrum antibiotic shared between animals and humans that is useful in treatments besides infections. However, changes in the gut microbiota caused by CFX and the possible link with the elimination of CFX residues have not been investigated. Herein, we used the Silkie chicken model to study the changes in the gut microbiota during the entire CFX-metabolic repertoire. We detected CFX residues in different tissues and showed that the elimination time of CFX from different tissues was dissimilar (liver > kidney > chest muscle > skin). Analysis of liver and kidney injury biomarkers and plasma antioxidant indices indicated slight hepatotoxicity and nephrotoxicity in the Silkie chickens. Importantly, the changes in the gut microbial community predominantly occurred early in the metabolic process. Correlation analysis revealed that the particular bacterial microbiota were associated with the pharmacokinetics of CFX in different Silkie chicken tissues (e.g., aerobic bacteria, including Escherichia and Coprococcus, and anaerobic bacteria, including Fusobacterium, Ruminococcus, Bifidobacterium, and Eubacterium). Collectively, certain microbiota may boost antibiotic metabolism and participate in restoring the microbial consortia after CFX is metabolized. Therefore, regulating the core intestinal microbiota may reduce foodborne antibiotics and accelerate the development of drug resistance.

The dermal hyperpigmentation phenotype in chickens is controlled by the dominant fibromelanosis allele. One of the ten unique characteristics of Silkie chickens is the fibromelanosis phenotype, which is pigmentation in the dermal layer of the skin and connective tissue. In this study, we found a mutation of fibromelanosis, a genomic rearrangement that included an inverted duplication of endothelin3 (EDN3), is responsible. We show that, as a stimulator of melanoblast proliferation, EDN3 expression was increased in silkie embryos and in both skin and muscle throughout adulthood. EDN3 expression led to an increase in expression of the downstream genes EDNRB2 and TYRP2, and was closely relate with the hyperpigmentation phenotype. We examined eight different Chinese chicken breeds showing hyperpigmentation and conclude that this structural genetic variant exists in all fibromelanosis chicken breeds.

Summary: The crest in chicken consists of elongated and upraised feathers, as seen in various breeds such as the Silkie chicken. Recently, the still unknown causative mutation for the crest phenotype was assigned to chromosome 33 and an ectopic expression of HOXC8 was shown. The aim this study was to evaluate whether the crest phenotype in a local Swiss chicken breed, the Appenzeller Spitzhaubenhuhn, is associated with HOXC8. Three previously reported crest‐associated flanking markers at the HOXC8 locus were genotyped in cohorts of this breed and two other local breeds without the crest phenotype. For the Appenzeller Spitzhaubenhuhn chicken showing the crest phenotype, no clear association of the reported markers could be revealed. Furthermore, the two exons of HOXC8 were sequenced in crested chicken of the Appenzeller Spitzhaubenhuhn and Silkie breeds and revealed no evidence of polymorphisms within the coding region of HOXC8. Therefore, the molecular genetic etiology for the crest phenotype in the investigated breeds remains unclear. [ABSTRACT FROM AUTHOR]

This experiment was to investigate the differences in egg quality between silkie chickens with high and low egg production numbers (EPNs) groups. The EPNs were recorded from first laying to 40 weeks of age of 240 silkie chickens. Based on the total average EPNs at 40 weeks of age, the chickens were divided into two groups which were the H group with higher EPNs and the L group with lower EPNs. The egg weight, egg shape index, eggshell traits, Haugh units, yolk index, and color were conducted at 40 weeks of age in each group. The results showed that the total average EPNs of 240 silkie chickens was 83.69 at 40 weeks of age. 160 silkie chickens in the H group had an average EPNs of 94.50, which was significantly higher (P < 0.05) than the L group of 80 silkie chickens with an average EPNs of 68.89. The egg shape index, eggshell strength, height of thick egg white, Haugh units, and yolk index of the H group were significantly higher (P < 0.05) than the L group. The L value (Lightness) and a value (Redness) of eggshell and egg yolk ratio were significantly (P < 0.05) lower than the L group. There were no significant differences between the H and L groups in egg weight, eggshell ratio, egg white ratio, and egg yolk color. In conclusion, the H group had better performance in the quality of eggshell, egg white, and egg yolk and the color degree of eggshell. [ABSTRACT FROM AUTHOR]
