Abstract Background Understanding the lineage differentiation of human prostate not only is crucial for basic research on human developmental biology but also significantly contributes to the management of prostate-related disorders. Current knowledge mainly relies on studies on rodent models, lacking human-derived alternatives despite clinical samples may provide a snapshot at certain stage. Human embryonic stem cells can generate all the embryonic lineages including the prostate, and indeed a few studies demonstrate such possibility based on co-culture or co-transplantation with urogenital mesenchyme into mouse renal capsule. Methods To establish a stepwise protocol to obtain prostatic organoids in vitro from human embryonic stem cells, we apply chemicals and growth factors by mimicking the regulation network of transcription factors and signal transduction pathways, and construct cell lines carrying an inducible NKX3-1 expressing cassette, together with three-dimensional culture system. Unpaired t test was applied for statistical analyses. Results We first successfully generate the definitive endoderm, hindgut, and urogenital sinus cells. The embryonic stem cell-derived urogenital sinus cells express prostatic key transcription factors AR and FOXA1, but fail to express NKX3-1. Therefore, we construct NKX3-1-inducible cell line by homologous recombination, which is eventually able to yield AR, FOXA1, and NKX3-1 triple-positive urogenital prostatic lineage cells through stepwise differentiation. Finally, combined with 3D culture we successfully derive prostate-like organoids with certain structures and prostatic cell populations. Conclusions This study reveals the crucial role of NKX3-1 in prostatic differentiation and offers the inducible NKX3-1 cell line, as well as provides a stepwise differentiation protocol to generate human prostate-like organoids, which should facilitate the studies on prostate development and disease pathogenesis.

Abstract With the increase of mining scope, rockburst occurs frequently, but its generation mechanism has not been understood comprehensively. Based on a rockburst in the coal pillar area of high tectonic stress zones (HTSZs), this study analyzed the distribution characteristics of large-energy microseismic (MS) events by using data statistics. The mechanical cause of the MS event that induced the rockburst was revealed by means of seismic moment tensor inversion. On this basis, by using numerical simulation, this study explored the distribution characteristics of static load in rockburst area and the effect of dynamic load in the floor, and then proposed the rockburst mechanism. The results show that under the squeezing action, the floor strata in HTSZs implode and transmit energy outward in the form of stress waves. This causes the cumulative damage and stress of the coal body in the fast track of coal pillar area increase in a short time. Since the coal in this area has already been in the critical stress state, small stress changes may lead to coal failure and rockburst. In this case of rockburst, the high static load of coal is the main force source, and the dynamic load plays a role in increasing coal body damage and inducing rockburst. Combined with seismic moment tensor inversion and numerical simulation, this paper proposes a rockburst research scheme, which makes the simulation of dynamic load more reasonable. The results provide the theoretical basis for rockburst control under similar conditions.

The rock burst frequently occurs in the roadways with top coal, and the roof disasters are particularly severe under dynamic stress in China. In order to explore the classification and instability mechanisms of anchoring structures in the roadways with top-coal, analog simulation experiments, theoretical analysis, and data statistics were used under the engineering background of the large-area rock burst roof failure in the Working Face 301 of a mine in Shaanxi Province, China. The dynamic load response characteristics of the stress, displacement, and surface acceleration of the surrounding rock in the roadways with top coal were analyzed. The classification characteristics of the roof anchoring structure under the influence of top coal thickness and cables were studied. The stress response mechanism of the roadways with top coal under elastic waves was explored, and the instability mechanism of the anchoring “beam arch” structure was proposed. The rock burst resistance of the air-return roadway in the Working Face 301 was evaluated, and the corresponding optimization schemes for the “beam-arch” composite structure were proposed. The results show that ① the monitoring data of stress and displacement of the surrounding rock in the roadways with top coal under dynamic stress verify the existence of internal and external beam or arch anchoring structures in the roof. Due to the increase in the thickness of top coal, there is a transition from “beam” to “arch” in the anchoring structure of the inner layer of the roof; ② based on the relative relationship between the thickness of the top coal and the cables, the roof anchoring structure is divided into “superimposed-beam and arch” for thin top coal, “composite-beam and arch” for thick top coal, and “combined arch” for extra thick top coal. A critical thickness index for the transformation of thick coal beams from “beam” to “arch” is established; ③ the instability mechanism of the beam structure and the arch structure is that they fail after reaching their tensile and shear strength limits under dynamic and static stress, respectively. The amplification effect of the “beam-arch” composite structure on the load stress is significantly affected by its size; ④ the bearing strength of the inner arch in the "combined arch" structure is relatively low. Increasing the length of cables can increase the thickness of the inner arch, and the corresponding “combined arch” structure’s bearing strength increases significantly, which is consistent with the evaluation results of the rock burst resistance.

Abstract Parkinson's disease (PD) is a mitochondria‐related neurodegenerative disease characterized by locomotor deficits and loss of dopaminergic (DA) neurons in the substantia nigra pars compacta (SNc). Majority of PD research primarily focused on neuronal dysfunction, while the roles of astrocytes and their mitochondria remain largely unexplored. To bridge the gap and investigate the roles of astrocytic mitochondria in PD progression, we constructed a specialized optogenetic tool, mitochondrial‐targeted anion channelrhodopsin, to manipulate mitochondrial membrane potential in astrocytes. Utilizing this tool, the depolarization of astrocytic mitochondria within the SNc in vivo led to the accumulation of γ‐aminobutyric acid (GABA) and glutamate in SNc, subsequently resulting in excitatory/inhibitory imbalance and locomotor deficits. Consequently, in vivo calcium imaging and interventions of neurotransmitter antagonists demonstrated that GABA accumulation mediated movement deficits of mice. Furthermore, 1 h/day intermittent astrocytic mitochondrial depolarization for 2 weeks triggered spontaneous locomotor dysfunction, α‐synuclein aggregation, and the loss of DA neurons, suggesting that astrocytic mitochondrial depolarization was sufficient to induce a PD‐like phenotype. In summary, our findings suggest the maintenance of proper astrocytic mitochondrial function and the reinstatement of a balanced neurotransmitter profile may provide a new angle for mitigating neuronal dysfunction during the initial phases of PD.

AbstractWith the increase in coal mining depth, many rock bursts triggered by fold structures have been observed, but the disaster mechanism is not clear. This research aims at investigating the influence of upright fold structure(UFS) on rock burst mechanisms. The mechanical model and numerical model of the UFS are constructed, and the mining stress evolution law and rock burst mechanism of working face in a UFS area are studied by utilizing engineering practice. The research shows that the occurrence of rock burst disasters in the fold structure area has obvious regional characteristics. The horizontal and vertical stress of the UFS in the horizontal distribution is similar to the periodic variation characteristics of the sine (cosine) curve. The stress state of the UFS area is divided into five areas. The horizontal and vertical stress concentration factors increase first and then decrease with the working face gradually approaching and away from the upright fold syncline axis. A roadway impact risk index I0 is proposed to determine whether the roadway is damaged, and the mechanism of inducing ground pressure during mining in UFS areas of deep mines is expounded. The control plan effectively reduces the stress levels of surrounding rock in working faces.

AbstractCoal bursts caused by mining-induced seismicity occur in the working face in steeply inclined and extra thick coal seams (SIETCSs). This article presents a case study in SIETCS through discrete element simulation as well as field investigation. The evolution of the coal burst was studied from the perspective of stress distribution, crack propagation and coal fragment ejection. The results showed that dynamic failure mainly occurred in the coal near the roof. The deep bottom coal and the top coal near the roof are under high static stress due to the clamping effect of the roof and floor and the rotation of the deep roof. The coal burst in the working face was triggered by mining-induced seismicity. The coal burst occurred when the sum of the static stress and the dynamic stress caused by the mining-induced seismicity exceeds the sum of the ultimate strength of the coal and the support strength, which was called the ‘clamping-rotation-dynamic loads’ mechanism. During the coal burst, the cracks developed rapidly into the deep bottom coal and the top coal on the roof side. The results can serve as an important reference for the coal burst failure mechanism in the working face in SIETCSs.

AbstractWith the increase of mining depth and mining intensity of the large inclined extra-thick coal seam (LIETCS), the number of rock bursts increases significantly. In this paper, based on a rock burst during upper roadway driving of the LIETCS, the distribution characteristics and fracture mechanism of microseismic (MS) events are analyzed using data statistics and seismic moment tensor inversion. By means of mechanical analysis and numerical simulation, this paper studies the static load distribution characteristics and dynamic load action effect of the LIETCS under upper slice mining condition. Based on the theory of dynamic and static combined load, the mechanism of rock bursts is proposed. The results show that the coal and rock mass of the LIETCS are mainly destroyed by shearing. The “shear-clamping” stress produced by the joint action of the roof and floor is the static load source. In upper slice mining, the shear stress and energy of the coal pillar behind the slice and the coal body below the coal pillar always remain high. Under the superposition of floor dynamic load, the regional stress environment changes, which induces rock bursts. This study provides a theoretical basis for rock burst prevention under similar conditions.

The variation of hard roof thickness is an essential contributor in triggering rock bursts during longwall mining. Case analysis and numerical modeling were used to study the stress and energy characteristics of the coal and rock mass and its fracture behaviour in the roof thickness variation zone (RTVZ). The results show that the coal seam has higher initial stress if overlain by a thicker hard roof, whose stress monitoring value is 1.8–2.6 times that of the thin zone. The increasing variation in the roof thickness or the roof properties causes a greater initial stress change in the coal seam. In the thick roof zones, the superposition of the advanced abutment pressure and the increased initial stress will result in a high-stress concentration area, where the stress mutation coefficient value can be up to 1.08–1.15. A higher rock burst risk might thus present in the roadway near the longwall in the thick roof zone, where more intensive elastic energy was released in the coal/rock mass. Also, it is more likely to have a significant dynamic load in the thin roof zone due to the higher possibility of roof breakage, and the total microseismic energy can reach 1.8–3.2E + 08J. HighlightsCase analysis and numerical modelling were used to study the formation mechanism of stress anomaly in coal seams, energy evolution characteristics of the coal/rock mass and its fracture behaviour in the RTVZ.The mechanism of rock bursts induced by coal mining in the RTVZ is determined. The thick roof zone has high-stress concentration, where more intensive elastic energy is released in the coal/rock mass. Due to easier roof breakage, it is more likely to have a significant dynamic load in the thin roof zone.The prevention and control method of rock bursts in the RTVZ is put forward. The rock bursts can be relieved by reducing the initial stress increased in the thick roof zone. Strengthening roadway support can reduce the influence of dynamic load on the roadway.

Seismic energy analysis is the basis of coal burst risk assessment. The prerequisite for utilizing seismic energy to quantify coal burst hazards is the accurate understanding of seismic energy attenuation. Thereby the characteristics of seismic energy attenuation were first systematically investigated in this paper. Based on the seismic data and 24 coal burst records during the period of sixteen months, a risk assessment method including three indexes (Static Intensity Index (SII), Dynamic Intensity Index (DII), and Risk Assessment Index (RAI)) was derived from seismic energy attenuation. In consideration of the static and dynamic response, the superposition effects of seismic energy were proposed to improve the performance of risk assessment. Results show that residual seismic energy (RSE) is relatively correlated with seismic energy, source radius, and energy attenuation coefficient; the higher seismic energy, source radius, and lower energy attenuation coefficient would result in higher coal burst risk. The case study demonstrated that SII could effectively predict the damaged area, but its prediction efficiency of high-magnitude events (HMEs) is relatively low; DII could predict damaged areas and HMEs, but may lead to risk overestimation; and RAI could efficiently predict the damaged area and HMEs. The sensitivity of the monitoring system and seismicity activeness could be the two main restrictions of the proposed method, leading to low efficiency or even false risk assessment. This paper sheds light on seismic energy utilization to risk prediction in underground coal mines. HighlightsThe attenuation response characteristics of seismic energy to different factors including seismic energy, source radius, and attenuation coefficient were investigated.A new assessment method based on seismic energy attenuation calculation was proposed to assess coal burst risks. Three seismic-based indexes considering static and dynamic response and their superposition effects were proposed to assess coal burst risk.Sensitivity of monitoring system and seismicity activeness could be the two main restrictions leading to low accuracy or even false in risk assessing, which implies that the proposed method could also have false judgements in the place where rare seismicity is monitored.

AbstractWith the increase of mining depth, rock burst disasters frequently occur in steeply inclined coal seams. Firstly, this paper analyzes the rock burst of 5521-20 working face in Yaojie No. 3 coal mine and summarizes the characteristics of rock burst in horizontal section mining of steeply inclined extra-thick coal seam (SIETCS). Then, the static load distribution characteristics and the influence of dynamic load in the horizontal section mining of SIETCS are systematically studied by combining theoretical analysis with numerical simulation. On this basis, the mechanism of rock burst in horizontal section mining of SIETCS is put forward, verified by actual measurement. The results show that the SIETCS is “clamped” under the combined action of the same change trend of roof and floor. The maximum principal stress peak values on the roof and floor sides reach 22.0 MPa and 20.5 MPa. The maximum shear stress earned 8.7 MPa and 8.4 MPa, which makes the shear stress concentration in the coal body high and tends to “shear dislocation.” Under this “shear-clamping” action, an approximate “trapezoidal” plastic zone and a “rectangular” stress concentration zone are formed under the section. With the increase of mining depth, the “shear-clamping” action of SIETCS becomes more and more intense. When the roof cantilever reaches the ultimate span and breaks, the intense dynamic load increases the shear stress and failure of coal, which is easy to induce rock burst. The superimposed load greatly affects the area from the roof side to the middle of the working face, and the rock burst is intense. The rock burst is weak on the floor side due to the pressure relief of the surrounding plastic zone. The monitoring results show that the supports pressure and MS events activity on the roof side and near the middle part of the working face is considerable, while the floor side is opposite, which verifies the research results.

Strong mining tremors (SMTs) frequently occur in super-thick strata near the goaf when mining. Since 2021, there have been three consecutive SMTs with magnitude greater than 2.0 in longwall 1208 of the Shilawusu Coal Mine. These SMTs caused mine production to be suspended for more than 290 days and affected over 100 households located on the shaking ground, and seriously threatened the safety of underground workers and restricted production capacity. Therefore, it is essential to investigate the occurrence mechanism of SMTs in super-thick strata goaf mining in order to understand the phenomenon, how the disaster of mining tremors occurs, and the prevention and control of mining tremor disasters. In this study, field observation, numerical analysis, and theoretical calculation were used to study the occurrence mechanism of three SMTs in the Shilawusu Coal Mine. The results show that the super-thick strata fracture induced by the SMTs is generally higher by one to three orders of magnitude in some of the source mechanical parameters compared to other mining tremors, and so is more likely to cause ground shaking. Field observations revealed that before and after the occurrence of SMTs, the maximum surface subsidence suddenly increased by about 0.1 m and showed a “stepped” increase, and the super-thick strata began to experience fractures. The following theoretical mechanics model of super-thick strata was established: at the goaf stage of mining, with the increase in the area of the hanging roof, the super-thick strata will experience initial and periodic fractures, which can easily induce SMTs. The relative moment tensor inversion method was used to calculate the source mechanism of SMTs, which was found to be caused by the tensile rupture resulting from the initial and periodic ruptures of super-thick strata, in addition to the shear rupture generated by the adjustment of unstable strata structures. As the mining continues on the longwall face, there is still a possibility of SMT occurrence. This paper provides some insights into the mechanism and prevention of SMT in underground coal mines.

Coal burst is a severe and dynamic hazard, and understanding its mechanism is crucial in preventing such incidents. Strong tremors during the working face mining in the stress anomaly zone of the pinch-out coal seam are frequent. Theoretical analysis, numerical simulation, and field measurement methods are used to analyze the energy evolution law for mining at the working face and mechanism of coal burst. Mechanical models of the inclined and strike overhang structures are established, and the theoretical analysis of the strike and inclined energy distribution characteristics of the working face roof is carried out. The two key areas with a high overhang bending deformation energy accumulation are identified at the lower end and middle-upper part of the working face. The simulation results show that the energy accumulation area of the roof in the inclined coal seam has prominent asymmetric distribution characteristics. The roof energy accumulates in the lower end and middle-upper area of the working face. The floor energy accumulates in the lower end area of the working face, and the peak position of the overhead energy of the working face in the direction shifts toward the coal-wall side. Influenced by the local absence of the No. 8 coal seam, the vertical stress of the surrounding rock at the working face of the massive, inclined coal seam increased by 12.7%; the peak of roof energy at the working face inclination and strike increased by 46.2% and 32.2%; and the range of roof energy accumulation expanded. A deep directional hole blasting plan to prevent the phenomenon at the working face roof is developed, which effectively reduces the stress and energy level of the inclined hanging roof and avoids the occurrence of coal bursts in the abnormal stress area of the working face.

In this work, we propose a symmetry approach and design a convolutional neural network for mouse pose estimation under scale variation. The backbone adopts the UNet structure, uses the residual network to extract features, and adds the ASPP module into the appropriate residual units to expand the perceptual field, and uses the deep and shallow feature fusion to fuse and process the features at multiple scales to capture the various spatial relationships related to body parts to improve the recognition accuracy of the model. Finally, a set of prediction results based on heat map and coordinate offset is generated. We used our own built mouse dataset and obtained state-of-the-art results on the dataset.

Pseudomonas chlororaphis P3 has been well-engineered as a platform organism for biologicals production due to enhanced shikimate pathway and excellent physiological and genetic characteristics. Gentisate displays high antiradical and antioxidant activities and is an important intermediate that can be used as a precursor for drugs. Herein, a plasmid-free biosynthetic pathway of gentisate was constructed by connecting the endogenous degradation pathway from 3-hydroxybenzoate in Pseudomonas for the first time. As a result, the production of gentisate reached 365 mg/L from 3-HBA via blocking gentisate conversion and enhancing the gentisate precursors supply through the overexpression of the rate-limiting step. With a close-up at the future perspectives, a series of bioactive compounds could be achieved by constructing synthetic pathways in conventional Pseudomonas to establish a cell factory.

Coal pillar bursts continue to be a severe dynamic hazard. Understanding its mechanism is of paramount importance and crucial in preventing and controlling its occurrence. The extreme roadway deformations from the asymmetric isolated coal pillars in the central mining district of YaoQiao Coal Mine have responded with frequent intense tremors, with risky isolated coal pillar bursts. The theoretical analysis, numerical simulation, and field measurements were done to research the impact of spatial overburden structure and stress distribution characteristics on the isolated coal pillar area, aiming to reveal the mechanism of coal pillar burst leading to the practice of prevention and control in the asymmetric isolated coal pillar area. The study shows that the overburden structure of the asymmetric is an asymmetric “T” structure in the strike-profile, and the stress in the coal pillar is mostly asymmetric “saddle-shaped” distribution, with the peak stress in the east side of the coal pillar, and the coal pillar is a “high stress serrated isolated coal pillar.” Numerical simulation results showed that the support pressure in the isolated coal pillar area on the strike profile was asymmetrically “saddle-shaped” distribution. The peak vertical stress in the coal pillar area continued to rise and gradually shifted to the mining district's deep part. As a result, the response of the roadway sides to the dynamic load disturbance was more pronounced. They developed a coal burst prevention and control program of deep-hole blasting in the roof of asymmetrical isolated coal pillar roof and unloading pressure from coal seam borehole. Monitored data confirmed that the stress concentration was influential in the roadway’s surrounding rock in the asymmetric isolated coal pillar area, circumventing coal pillar burst accidents. The research outcomes reference the prevention and control of coal bursts at isolated working faces of coal pillars under similar conditions.

Coal mine pillar burst frequently occurs in Western China, which seriously restricts safe production. This paper takes the 35 m coal pillar of the 3102 working face of MKQ coal mine as the engineering background. The mechanism and evolution control of pillar bursts in multithick key strata are studied using field investigation, theoretical analysis, and numerical simulation. The mechanism of dynamic and static stress-induced pillar bursts was revealed combining the “O-X” broken features for key strata and numerical simulation of pillar stress evolution. A prevention scheme is put forward for strata presplit blasting and adjusting coal pillar width to minimize the dynamic and static stresses. The results demonstrate the following. (1) In the multithick strata, the first and second near-field subkey strata have perpendicular “O-X” broken features, whereas the third far-field subkey has parallel “O-X” broken features. The working face has three kinds of periodic weighting phenomena: long, medium, and short. (2) The simulated vertical stress curve of 35 m coal pillar goes through three states: two-peak, asymmetric trapezoidal and symmetrical trapezoidal shape with the different advancing position of working face. The stress concentration is extensively promoting a high-risk area for rock burst. (3) The coal pillar burst was induced by the superposition of energy released by the key strata breaking and the elastic energy accumulated in the wide coal pillar. (4) The monitoring data showed that the long, medium, and short periodic weighting steps of multithick key strata are 141.6 m, 43.2–49.6 m, and 17.6–27.2 m, respectively. The microseismic events energy, frequency, and stress of hydraulic support increment are the highest during the long periodic weighting, and the spatial distribution of microseismic events coincides with the stress concentration area. The theoretical analysis is confirmed with the field practice.

Neurons responding sensitively to motions in several rather than all directions have been identified in many sensory systems. Although this directional preference has been demonstrated by previous studies to exist in the isthmi pars magnocellularis (Imc) of pigeon (Columba livia), which plays a key role in the midbrain saliency computing network, the dynamic response characteristics and the physiological basis underlying this phenomenon are unclear. Herein, dots moving in 16 directions and a biologically plausible computational model were used. We found that pigeon Imc’s significant responses for objects moving in preferred directions benefit the long response duration and high instantaneous firing rate. Furthermore, the receptive field structures predicted by a computational model, which captures the actual directional tuning curves, agree with the real data collected from population Imc units. These results suggested that directional preference in Imc may be internally prebuilt by elongating the vertical axis of the receptive field, making predators attack from the dorsal-ventral direction and conspecifics flying away in the ventral-dorsal direction, more salient for avians, which is of great ecological and physiological significance for survival.

Abstract Background Arbutin is a plant-derived glycoside with potential antioxidant, antibacterial and anti-inflammatory activities. Currently, it is mainly produced by plant extraction or enzymatic processes, which suffers from expensive processing cost and low product yield. Metabolic engineering of microbes is an increasingly powerful method for the high-level production of valuable biologicals. Since Pseudomonas chlororaphis has been widely engineered as a phenazine-producing platform organism due to its well-characterized genetics and physiology, and faster growth rate using glycerol as a renewable carbon source, it can also be engineered as the cell factory using strong shikimate pathway on the basis of synthetic biology. Results In this work, a plasmid-free biosynthetic pathway was constructed in P. chlororaphis P3 for elevated biosynthesis of arbutin from sustainable carbon sources. The arbutin biosynthetic pathway was expressed under the native promoter P phz using chromosomal integration. Instead of being plasmid and inducer dependent, the metabolic engineering approach used to fine-tune the biosynthetic pathway significantly enhanced the arbutin production with a 22.4-fold increase. On the basis of medium factor optimization and mixed fed-batch fermentation of glucose and 4-hydroxybenzoic acid, the engineered P. chlororaphis P3-Ar5 strain led to the highest arbutin production of 6.79 g/L with the productivity of 0.094 g/L/h, with a 54-fold improvement over the initial strain. Conclusions The results suggested that the construction of plasmid-free synthetic pathway displays a high potential for improved biosynthesis of arbutin and other shikimate pathway derived biologicals in P. chlororaphis.

DAHP synthase catalyzes the first step in the shikimate pathway, deriving the biosynthesis of aromatic amino acids (Trp, Phe and Tyr), phenazine-1-carboxamide, folic acid, and ubiquinone in Pseudomonas chlororaphis. In this study, we identified and characterized one DAHP synthase encoding gene phzC, which differs from the reported DAHP synthase encoding genes aroF, aroG and aroH in E. coli. PhzC accounts for approximately 90% of the total DAHP synthase activities in P. chlororaphis HT66 and plays the most critical role in four DAHP synthases in the shikimate pathway. Inactivation of phzC resulted in the reduction of PCN production by more than 90%, while the absence of genes aroF, aroG and aroH reduced PCN yield by less than 15%, and the production of PCN was restored after the complementation of gene phzC. Moreover, the results showed that phzC in P. chlororaphis HT66 is not sensitive to feedback inhibition. This study demonstrated that gene phzC is essential for PCN biosynthesis. The expression level of both phzC and phzE genes are not inhibited in feedback by PCN production due to the absence of a loop region required for allosteric control reaction. This study highlighted the importance of PhzC and applying P. chlororaphis for shikimate pathway-derived high-value biological production.

Brain science research often requires accurate localization and quantitative analysis of neuronal activity in different brain regions. The premise of related analysis is to determine the brain region of each site on the brain slice by referring to the Allen Reference Atlas (ARA), namely the regional localization of the brain slice. The image registration methodology can be used to solve the problem of regional localization. However, the conventional multi-modal image registration method is not satisfactory because of the complexity of modality between the brain slice and the ARA. Inspired by the idea that people can automatically ignore noise and establish correspondence based on key regions, we proposed a novel method known as the Joint Enhancement of Multimodal Information (JEMI) network, which is based on a symmetric encoder–decoder. In this way, the brain slice and the ARA are converted into a segmentation map with unified modality, which greatly reduces the difficulty of registration. Furthermore, combined with the diffeomorphic registration algorithm, the existing topological structure was preserved. The results indicate that, compared with the existing methods, the method proposed in this study can effectively overcome the influence of non-unified modal images and achieve accurate and rapid localization of the brain slice.

In order to maximize inventory benefits or minimize costs, reliability and cost of inventory control models need to be identified and analyzed. These importance measures are one important approach to recognize and evaluate system weaknesses. However, importance measures have fewer applications in inventory systems’ reliability. Considering the cost, this paper mainly discusses the reliability change of performance parameters with the importance measures in inventory systems. The calculation methods of differential importance and Birnbaum importance are studied in the inventory control model with shortages. By comparing the importance values of various parameters in the model, the optimization analysis of the inventory model can be used to identify the key parameters, so as to effectively reduce the total inventory cost. The importance order and the identification of key parameters are helpful to increase the operational efficiency of the inventory control and provide effective methods for improving the inventory management. Lastly, a case study with a shortage and limited inventory capacity is used to demonstrate the proposed model.

Detecting moving objects in a video sequence is an important problem in many vision-based applications. In particular, detecting moving objects when the camera is moving is a difficult problem. In this study, we propose a symmetric method for detecting moving objects in the presence of a dynamic background. First, a background compensation method is used to detect the proposed region of motion. Next, in order to accurately locate the moving objects, we propose a convolutional neural network-based method called YOLOv3-SOD for detecting all objects in the image, which is lightweight and specifically designed for small objects. Finally, the moving objects are determined by fusing the results obtained by motion detection and object detection. Missed detections are recalled according to the temporal and spatial information in adjacent frames. A dataset is not currently available specifically for moving object detection and recognition, and thus, we have released the MDR105 dataset comprising three classes with 105 videos. Our experiments demonstrated that the proposed algorithm can accurately detect moving objects in various scenarios with good overall performance.

China is highly rich in diversity of Begonia, and in which Guangxi Zhuangzu Autonomous Region, following Yunnan province, has the second highest number of Begonia species. As a distribution center of Begonia sect. Coelocentrum, Guangxi already had 58 species recognized in this section. In this paper, Begonia ornithopedata, a new taxon of B. sect. Coelocentrum from southwestern Guangxi is described. With the bird-foot-shape lobed leaves, the new species is easily distinguished from any of other taxa, excepting few similarities only in leaf morphology of B. laminariae Irmsch. and B. circumlobata Hance. China has an exceptional diversity in Begonia, holding the second largest number of described species in this genus at the national level. Many new taxa need to be explored and described. This paper reports two new species, Begonia ruchengensis (B. sect. Reichenheimia) and B. auritalata (B. sect. Platycentrum), from Hunan province and Guizhou province, respectively. Besides the detailed morphology description, color illustrations, and line drawings, the diagnoses of these two species distinguished from their allied taxa are provided. Promoter of alcohol oxidase I (PAOX1) is the most efficient promoter involved in the regulation of recombinant protein expression in Pichia pastoris (P. pastoris). PAOX1 is tightly repressed by the presence of glycerol in the culture medium; thus, glycerol must be exhausted before methanol can be taken up by P. pastoris and the expression of the heterologous protein can be induced. In this study, a candidate glycerol transporter (GT1, GeneID: 8197545) was identified, and its role was confirmed by further studies (e.g. bioinformatics analysis, heterologous complementation in Schizosaccharomyces pombe (S. pombe)). When GT1 is co-expressed with enhanced green fluorescent protein (EGFP), it localizes to the membrane and S. pombe carrying gt1 but not the wild-type strain can grow on medium containing glycerol as the sole carbon source. The present study is the first to report that AOX1 in the X-33⧠gt1 mutant can achieve constitutive expression in medium containing glycerol; thus, knocking down gt1 can eliminate the glycerol repression of PAOX1 in P. pastoris.
