A recent study demonstrated that in small-scale prepolarized surface nuclear magnetic resonance (SNMR-PP) measurements with a footprint of a few square meters, customized PP switch-off ramps can serve as an efficient excitation mechanism, eliminating the requirement for a conventional oscillating excitation pulse. This approach enables the detection of short relaxation signals from the unsaturated soil zone and can, therefore, be used to directly provide soil moisture and pore geometry information. Because ultimately such small-scale SNMR-PP setups are intended for a mobile application, it is necessary to develop strategies that allow for speedy measurement progress and do not require noise cancellation protocols based on reference stations. Hence, we developed a new concentric figure-of-eight (cFOE) loop layout that combines the direction independence of a circular loop with the intrinsic noise cancellation properties of a classical FOE-loop. This approach significantly decreases the measurement time because suitable signal-to-noise ratios are reached much faster compared to a classical circular loop and will bring us one step further toward fast and non-invasive soil moisture mapping applications.

Hesperomyces virescens (Ascomycota, Laboulbeniales), a fungal ectoparasite, is thus far reported on Harmonia axyridis from five continents: North and South America, Europe, Africa, and Asia. While it is known that He. virescens can cause mortality of Ha. axyridis under laboratory conditions, the role of biotic and abiotic factors in influencing the distribution of He. virescens in the field is unknown. We collected and screened 3,568 adult Ha. axyridis from 23 locations in seven countries in Central Europe between October and November 2018 to test the effect of selected host characters and climate and landscape variables on the infection probability with He. virescens. Mean parasite prevalence of He. virescens on Ha. axyridis was 17.9%, ranging among samples from 0 to 46.4%. Host sex, climate, and landscape composition did not have any significant effect on the infection probability of He. virescens on Ha. axyridis. Two color forms, f. conspicua and f. spectabilis, had a significantly lower parasite prevalence compared to the common Ha. axyridis f. novemdecimsignata.

Abstract Geophysical nuclear magnetic resonance (NMR) applications are used to estimate pore size distributions (PSDs) of rocks and sediments. This is commonly realized by empirical calibration using information about the surface‐to‐volume ratio of the material. Recent research has developed joint inversion concepts for NMR relaxation data that provides the PSD with a minimum of information. The application requires the NMR signal of a sample at saturation and at least one at partial saturation and at known suction. The new inversion concept physically simulates the desaturation process as part of the forward operator. The cross‐section of the model capillaries in the underlying bundle can be either circular or triangular. Our study investigates the performance of the NMR joint inversion to predict water retention function (WRF) and capillary‐based hydraulic conductivity (Kcap) as functions of saturation for different sands. The angularity of the pores has no significant impact on the estimated WRF but affects the Kcap estimation significantly. Our study shows that the WRF is predicted reliably for sand samples under fast diffusion conditions. The Kcap estimations are also plausible but tend to systematic overestimation, for which we identified the tortuosity being the main reason. Because NMR relaxation data generally do not provide tortuosity information, a plausible tortuosity model remains an issue of classical calibration. Further development of the approach will thus consider tortuosity measurements (e.g., by electrical resistivity measurements and/or gradient NMR) and will consider the relaxation mechanisms outside fast diffusion conditions to enhance its applicability for coarse soils.

Harmonia axyridis is an invasive alien ladybird in North America and Europe. Studies show that multiple natural enemies are using Ha. axyridis as a new host. However, thus far, no research has been undertaken to study the effects of simultaneous infection by multiple natural enemies on Ha. axyridis. We hypothesized that high thallus densities of the ectoparasitic fungus Hesperomyces virescens on a ladybird weaken the host’s defenses, thereby making it more susceptible to infection by other natural enemies. We examined mortality of the North American-native Olla v-nigrum and Ha. axyridis co-infected with He. virescens and an entomopathogenic fungus—either Beauveria bassiana or Metarhizium brunneum. Laboratory assays revealed that He. virescens-infected O. v-nigrum individuals are more susceptible to entomopathogenic fungi, but Ha. axyridis does not suffer the same effects. This is in line with the enemy release hypothesis, which predicts that invasive alien species in new geographic areas experience reduced regulatory effects from natural enemies compared to native species. Considering our results, we can ask how He. virescens affects survival when confronted by other pathogens that previously had little impact on Ha. axyridis.

Harmonia axyridis (Coleoptera, Coccinellidae) is a globally invasive ladybird. It has been intentionally introduced in many countries as a biological control agent, whereas it has been unintentionally released in many others. Climatic factors are important in limiting the spread of H. axyridis. For example, very few records are known from tropical or desert regions. Currently, no published reports are known from Central America. Here, we report H. axyridis from Costa Rica, Guatemala, Honduras, Panama, and Puerto Rico. Specimens were either observed by the authors, discovered in dried insect collections, or retrieved from searching through online photographs available from the citizen science project iNaturalist and the photo-sharing website Flickr. These new records and the wide distribution of H. axyridis in Latin America suggest several invasion events, which have gone unnoticed until now. We stress the need for further, large-scale monitoring and show the advantage of citizen science to assess the presence of invasive alien species.

Parasitism is one of the most diverse and abundant modes of life, and of great ecological and evolutionary importance. Notwithstanding, large groups of parasites remain relatively understudied. One particularly unique form of parasitism is hyperparasitism, where a parasite is parasitized itself. Bats (Chiroptera) may be parasitized by bat flies (Diptera: Hippoboscoidea), obligate blood-sucking parasites, which in turn may be parasitized by hyperparasitic fungi, Laboulbeniales (Ascomycota: Laboulbeniomycetes). In this study, we present the global tritrophic associations among species within these groups and analyze their host specificity patterns. Bats, bat flies, and Laboulbeniales fungi are shown to form complex networks, and sixteen new associations are revealed. Bat flies are highly host-specific compared to Laboulbeniales. We discuss possible future avenues of study with regard to the dispersal of the fungi, abiotic factors influencing the parasite prevalence, and ecomorphology of the bat fly parasites.

Carbonate reservoirs form important exploration targets for the oil and gas industry in many parts of the world. This study aims to differentiate and quantify pore types and their relation to petrophysical properties in the Permo-Triassic Khuff Formation, a major carbonate reservoir in Oman. For that purpose, we have employed a number of laboratory techniques to test their applicability for the characterization of respective rock types. Consequently, a workflow has been established utilizing a combined analysis of petrographic and petrophysical methods which provide the best results for pore-system characterization. Micro-computed tomography (µCT) analysis allows a representative 3D assessment of total porosity, pore connectivity, and effective porosity of the ooid-shoal facies but it cannot resolve the full pore-size spectrum of the highly microporous mud-/wackestone facies. In order to resolve the smallest pores, combined mercury injection capillary pressure (MICP), nuclear magnetic resonance (NMR), and BIB (broad ion beam)-SEM analyses allow covering a large pore size range from millimeter to nanometer scale. Combining these techniques, three different rock types with clearly discernible pore networks can be defined. Moldic porosity in combination with intercrystalline porosity results in the highest effective porosities and permeabilities in shoal facies. In back-shoal facies, dolomitization leads to low total porosity but well-connected and heterogeneously distributed vuggy and intercrystalline pores which improves permeability. Micro- and nanopores are present in all analyzed samples but their contribution to effective porosity depends on the textural context. Our results confirm that each individual rock type requires the application of appropriate laboratory techniques. Additionally, we observe a strong correlation between the inverse formation resistivity factor and permeability suggesting that pore connectivity is the dominating factor for permeability but not pore size. In the future, this relationship should be further investigated as it could potentially be used to predict permeability from wireline resistivity measured in the flushed zone close to the borehole wall.

Harmonia axyridis is an invasive ladybird (Coleoptera, Coccinellidae) with the potential to outcompete native ladybird species in its invasive distribution area. It was introduced as a biological control agent in many countries but has also spread unintentionally in many others. Hesperomyces virescens (Ascomycota, Laboulbeniales) is a minute (200–400 µm in size) biotrophic fungus that infects over 30 species of ladybirds. The aim of this study was to evaluate whether the elytral color pattern, size, and sex of Ha. axyridis affect infection by H. virescens. Coloration in Ha. axyridis has been linked to the presence of an antimicrobial alkaloid (harmonine). In fall 2016, we collected 763 Ha. axyridis individuals in Cambridge, Massaschusetts, of which 119 (16%) bore H. virescens fruiting bodies. We analyzed 160 individuals (80 infected, 80 uninfected) concerning the intensity of infection by H. virescens. Elytral sizes and coloration patterns were quantified using digital photography and analytical methods. Smaller ladybirds had a higher prevalence and higher intensity of parasitism. Additionally, male ladybirds bore more thalli compared to female ladybirds. Elytral color patterns had an effect on neither prevalence nor intensity of infection by Laboulbeniales in our dataset, although we found a slight trend to higher intensity of parasitism in more melanic males. This suggests that the development of Laboulbeniales might be affected by certain insect alkaloids.

We present a 3D numerical karst evolution model describing mechanisms governing the evolution of collapse dolines. The results confirm the current state of the art of the knowledge about the conditions necessary for a collapse doline to grow. We demonstrate that these geological features develop in karst aquifers, above subsurface rivers flowing through active cave systems or mechanically unstable zones. The mechanisms of growth of single/isolated collapse dolines are generally two dimensional and can be modeled with very dense numerical models. On the other hand, our results demonstrate that there can be more than one location, within a karst system, where the conditions allow the development of a collapse doline. For such complex scenarios a 3D modeling approach and a detailed knowledge of the hydro-geological situation is required in order to correctly predict and describe the development of collapse dolines.

Abstract The gene regulatory network (GRN) of a cell executes genetic programs in response to environmental and internal cues. Two distinct classes of methods are used to infer regulatory interactions from gene expression: those that only use observed changes in gene expression, and those that use both the observed changes and the perturbation design, i.e. the targets used to cause the changes in gene expression. Considering that the GRN by definition converts input cues to changes in gene expression, it may be conjectured that the latter methods would yield more accurate inferences but this has not previously been investigated. To address this question, we evaluated a number of popular GRN inference methods that either use the perturbation design or not. For the evaluation we used targeted perturbation knockdown gene expression datasets with varying noise levels generated by two different packages, GeneNetWeaver and GeneSpider. The accuracy was evaluated on each dataset using a variety of measures. The results show that on all datasets, methods using the perturbation design matrix consistently and significantly outperform methods not using it. This was also found to be the case on a smaller experimental dataset from E. coli. Targeted gene perturbations combined with inference methods that use the perturbation design are indispensable for accurate GRN inference.

The regulatory relationships between genes and proteins in a cell form a gene regulatory network (GRN) that controls the cellular response to changes in the environment. A number of inference methods to reverse engineer the original GRN from large-scale expression data have recently been developed. However, the absence of ground-truth GRNs when evaluating the performance makes realistic simulations of GRNs necessary. One aspect of this is that local network motif analysis of real GRNs indicates that the feed-forward loop (FFL) is significantly enriched. To simulate this properly, we developed a novel motif-based preferential attachment algorithm, FFLatt, which outperformed the popular GeneNetWeaver network generation tool in reproducing the FFL motif occurrence observed in literature-based biological GRNs. It also preserves important topological properties such as scale-free topology, sparsity, and average in/out-degree per node. We conclude that FFLatt is well-suited as a network generation module for a benchmarking framework with the aim to provide fair and robust performance evaluation of GRN inference methods.
