OBJECTIVE: To describe the translation and transcultural adaptation of Hurria's chemotherapy toxicity score, a prediction tool to estimate chemotherapy toxicity in the elderly. METHODS: The original English version of the score was translated to Portuguese using the forward and backward translation technique, as described by Guilemin. RESULTS: The translation was performed by two Brazilians (a physician and a professional translator), both fluent in English. Two professionals from a specialized linguistic service, not related to the study, then carried out the backward translation. Finally, a medical committee composed of oncologists, geriatricians, and hematologists discussed the consistency of the score, choosing a final version of the instrument in Portuguese. This version was piloted-tested in medical charts in an Oncogeriatric service, with high reliability as tested by Kappa statistic test. CONCLUSION: The final version of Hurria's chemotherapy toxicity score in Portuguese proved to be an easy, clear, and quick tool, suitable for use in clinical practice.

Abstract Opioid use disorder (OUD) has been linked to macroscopic structural alterations in the brain. The monthly injectable, extended-release formulation of μ-opioid antagonist naltrexone (XR-NTX) is highly effective in reducing opioid craving and preventing opioid relapse. Here, we investigated the neuroanatomical effects of XR-NTX by examining changes in cortical thickness during treatment for OUD. Forty-seven OUD patients underwent structural magnetic resonance imaging and subjectively rated their opioid craving ≤1 day before (pre-treatment) and 11 ± 3 days after (on-treatment) the first XR-NTX injection. A sample of fifty-six non-OUD individuals completed a single imaging session and served as the comparison group. A publicly available [¹¹C]carfentanil positron emission tomography dataset was used to assess the relationship between changes in cortical thickness and μ-opioid receptor (MOR) binding potential across brain regions. We found that the thickness of the medial prefrontal and anterior cingulate cortices (mPFC/aCC; regions with high MOR binding potential) was comparable between the non-OUD individuals and the OUD patients at pre-treatment. However, among the OUD patients, mPFC/aCC thickness significantly decreased from pre-treatment to on-treatment. A greater reduction in mPFC/aCC thickness was associated with a greater reduction in opioid craving. Taken together, our study suggests XR-NTX-induced cortical thickness reduction in the mPFC/aCC regions in OUD patients. The reduction in thickness does not appear to indicate a restoration to the non-OUD level but rather reflects XR-NTX’s distinct therapeutic impact on an MOR-rich brain structure. Our findings highlight the neuroplastic effects of XR-NTX that may inform the development of novel OUD interventions.

Prickly pear (Opuntia ficus-indica L. Mill) is widely cultivated in arid regions of every continent for its nutritious fruits and various commercial applications, cactus pear cladodes are used as forage in the most arid regions, while recently prickly pear by-products are studied as ruminant feed. They are characterised by a high moisture and sugar content; therefore, it is necessary to study suitable conservation methods. The aim of this study was to verify the effectiveness of silage to preserve prickly pear peels (PP) in a mixture with other by-products at different inclusion levels (5% straw and 6% or 12% wheat bran). For each treatment, six under vacuum micro-silos were prepared and, after 40 d of storage, the state of preservation was evaluated. Subsequently, the aliquots were analysed for chemical composition and incubated with sheep rumen fluid to evaluate the fermentation kinetics. The use of wheat bran and straw added to PP resulted in an increase in dry matter (DM); PP silage with wheat bran was found to have higher protein content and nutritional value. The greater presence of lactic acid was found in PP, such as acetic acid, therefore the lactic/acetic ratio was 2.33, significantly lower than silages with the addition of wheat bran. Silage with 5% straw showed the lowest level of organic matter disappearance (OMD) and the cumulative volume of gas (OMCV) produced during the incubation. The PP showed the fastest fermentation rate achieved after 12 h of fermentation, while those containing wheat bran, presented a higher maximum fermentation rate (Rmax), which need longer time to be achieved. Canonical discriminant analysis (CDA) showed clear discrimination of different silages based on their chemical characteristics and parameters detected during in vitro incubation. Overall, the silages with the addition of wheat bran can be considered the best and, of the two, those produced with the addition of 12% bran. Finally, ensilage represents a conservation technique suitable for preserving the nutritional characteristics of PP.

The atomization process plays a very important role because the success or failure of combustion in the combustion chamber is influenced by the size of the liquid fuel mist. The occurrence of good atomization from atomized is determined by the size of the fuel droplets and the size of the nozzle diameter. Reducing the droplet diameter can increase the velocity, which can affect the Reynolds number and cause turbulence effects. The effect of turbulence provides a vortex effect, which enhances the heat transfer process between hot air and fuel particles in the combustion chamber. In this research, the effect of fuel magnetization on droplet size is analyzed. The fuel used is E0, E10, E20, E30, and E100, all types of fuel are channeled through an electromagnetic field of 1500 Gauss. Sol LeWitt, a pioneer of conceptual art, created during his career over 1350 wall drawings, including the Wall Drawing #736 (1993) at the Center for Contemporary Art Luigi Pecci in Prato (Prato, Italy). The painting, executed by Andrea Marescalchi and Antony Sansotta under LeWitt’s instructions, features a grid of coloured rectangles obtained by overlapping different layers of inks. During a 2021 restoration by the Wall Paintings and Stuccoes Department of the Opificio delle Pietre Dure (Firenze, Italy), an in-depth investigation of the composition and the materials used by LeWitt’s assistants in producing Wall Drawing #736 was performed. A multi-analytical approach entailing Raman spectroscopy, high-performance liquid chromatography coupled to diode array and high-resolution mass spectrometry, gas chromatography–mass spectrometry (GC–MS), and pyrolysis coupled with GC–MS was applied. Our results revealed the use of animal glue, shellac resin, paraffin wax, linseed oil, and various organic pigments. The binder in the preparation layer was identified as poly(vinyl acetate), while poly(n-butyl methacrylate) was determined as a fixative. This research provided valuable insights into LeWitt’s techniques. The acquired knowledge on the paint technique is highly relevant in supporting conservators in restoration and consolidating the many wall drawings produced exploiting the same technique all over the world.

Sheep milk production is a complex process that is influenced by various factors. This study aims to investigate how the litter size (single vs. twins), birth type (male vs. female), age of the ewes and body weight affect the milk composition and fatty acid profile of milk fat. To face the ongoing issues related to global warming, a circular economy approach should be pursued, rethinking the waste management system and the recovery of organic waste. The main organic waste streams are Food Waste (FW) and municipal Sewage Sludge (SS). In the spirit of circularity, a commingled treatment of FW and SS could be a viable solution. To this end, the present work aims to review the technical and environmental aspects of the co-treatment of FW and SS through biological and thermal processes. Firstly, a detailed characterization of the two substrates is presented as well as the current and future treatment technologies. Then, the technical feasibility and the environmental impacts of conventional biological co-treatments of FW and SS (i.e., composting, anaerobic digestion, and a combination of them), as well as innovative thermal ones (i.e., incineration, gasification, pyrolysis, and hydrothermal carbonization), is summarized. The outcomes of this work could contribute to achieving a more sustainable way to approach organic waste treatment and to help policy-making authorities move toward sustainable planning.

Gastrotrichs of the genus Chaetonotus Ehrenberg, 1830 (Chaetonotida, Chaetonotidae) are known to be a speciose and pervasive group in both the marine and freshwater ecosystems, and recent investigations indicate that there is a significant amount of species diversity yet to be discovered. In the present study, a new species of this genus is described from two quarry lakes in northern Italy and characterized using an integrative morphological and molecular approach. Chaetonotus polites n. sp. is recognized by a rather stocky body, a five-lobed head, and the cuticular covering resolved in 13 alternating columns of partially imbricated scales, trilobed on the head and pentagonal on the trunk; each scale bears a peculiar simple, very thick, curved spine with a truncated apex. The new species is also distinguished by four putative molecular apomorphies at the 18S rRNA gene and two at the 28S rRNA gene. The location and type of these unique traits in the predicted secondary structure of the ribosomal genes is provided. The phylogenetic analysis based on concatenated sequences of three genes (the nuclear 18S and 28S rRNA genes and the mitochondrial COI gene) derived from 123 selected chaetonotidans, including the novel species, confirms that the genus Chaetonotus is a polyphyletic group, with several of its members resolved together with species of other genera. Abstract E3 ubiquitin protein ligase encoded by ARIH2 gene catalyses the ubiquitination of target proteins and plays a crucial role in posttranslational modifications across various cellular processes. As prior documented, mutations in genes involved in the ubiquitination process are often associated with autism spectrum disorder (ASD) and/or intellectual disability (ID). In the current study, a de novo heterozygous mutation was identified in the splicing intronic region adjacent to the last exon of the ARIH2 gene using whole exome sequencing (WES). We hypothesize that this mutation, found in an ASD/ID patient, disrupts the protein Ariadne domain which is involved in the autoinhibition of ARIH2 enzyme. Predictive analyses elucidated the implications of the novel mutation in the splicing process and confirmed its autosomal dominant inheritance model. Nevertheless, we cannot exclude the possibility that other genetic factors, undetectable by WES, such as mutations in non-coding regions and polygenic risk in inter-allelic complementation, may contribute to the patient's phenotype. This work aims to suggest potential relationship between the detected mutation in ARIH2 gene and both ASD and ID, even though functional studies combined with new sequencing approaches will be necessary to validate this hypothesis.

Abstract The overexpression of the ecotropic viral integration site-1 gene (EVI1/MECOM) marks the most lethal acute myeloid leukemia (AML) subgroup carrying chromosome 3q26 abnormalities. By taking advantage of the intersectionality of high-throughput cell-based and gene expression screens selective and pan-histone deacetylase inhibitors (HDACis) emerge as potent repressors of EVI1. To understand the mechanism driving on-target anti-leukemia activity of this compound class, here we dissect the expression dynamics of the bone marrow leukemia cells of patients treated with HDACi and reconstitute the EVI1 chromatin-associated co-transcriptional complex merging on the role of proliferation-associated 2G4 (PA2G4) protein. PA2G4 overexpression rescues AML cells from the inhibitory effects of HDACis, while genetic and small molecule inhibition of PA2G4 abrogates EVI1 in 3q26 AML cells, including in patient-derived leukemia xenografts. This study positions PA2G4 at the crosstalk of the EVI1 leukemogenic signal for developing new therapeutics and urges the use of HDACis-based combination therapies in patients with 3q26 AML.

High-quality language interactions not only support children's language development, but also promote better long-term academic outcomes. Language learning is the single best predictor of later growth in language, literacy, mathematics, and social development. However, many families do not have access to educationally enriched spaces that spur high-quality language opportunities. This leads the authors to ask what educators and policymakers can do to expose children to high-quality interactions that promote literacy skills. The Playful Learning Landscapes (PLL) initiative, which is presented in this article, was founded on the premise that children from all communities should have access to beautiful, enriching, and culturally relevant play environments that help them thrive. Working with community members, the authors co-designed public spaces that promote the kinds of adult-child conversations that lead to literacy learning.

Thanks to their intrinsic flexibility, energy efficiency and high portability, soft piezoelectric thin films represent the most effective technological approach for wearable devices to monitor health conditions. In order to improve effectiveness and applicability, more and more innovative and high-performing soft piezoelectric materials are being developed and benchmarked through their piezoelectric d33 coefficient. However, most existing methods to measure the d33 were developed for ceramic or bulk materials and cannot be applied to soft materials because high force/pressure can deform and damage the material structure. This work introduces a simple, effective, and fast method to accurately measure the d33 of soft and thin piezoelectric films by applying weak sinusoidal forces to avoid any damage to the sample, and simultaneously measuring the charges produced by the direct piezoelectric effect. The approach is versatile as it can be used for different types of materials and sizes of the active area. This method represents an effective solution to speed up the process of material optimization, paving the way for the rapid development of novel wearable piezoelectric devices.

Abstract Despite increasing knowledge about small extracellular vesicle (sEV) composition and functions in cell–cell communication, the mechanism behind their biogenesis remains unclear. Here, we reveal for the first time that sEV biogenesis and release into the microenvironment are tightly connected with another important organelle, Lipid Droplets (LDs). The correlation was observed in several human cancer cell lines as well as patient‐derived colorectal cancer stem cells (CR‐CSCs). Our results demonstrated that external stimuli such as radiation, pH, hypoxia or lipid‐interfering drugs, known to affect the number of LDs/cell, similarly influenced sEV secretion. Importantly, through multiple omics data, at both mRNA and protein levels, we revealed RAB5C as a potential important molecular player behind this organelle connection. Altogether, the potential to fine‐tune sEV biogenesis by targeting LDs could significantly impact the amount, cargos and properties of these sEVs, opening new clinical perspectives.

Summary: Thyroid carcinoma represents the first malignancy among the endocrine organs. Investigating the cellular hierarchy and the mechanisms underlying the initiation of thyroid carcinoma is crucial in thyroid cancer research. Here, we present a protocol for deriving thyroid cell lineage from human embryonic stem cells. Abstract Objective Neurocysticercosis (NCC) is considered the major cause of epilepsy in endemic regions. In the rural areas of the Bolivian Chaco prevalence of NCC among people with epilepsy (PWE) was 27.4%, according to a population‐based survey carried out in 1994. The aim of the study was to estimate the prevalence of Epilepsy Associated with Tonic–Clonic Seizures (EATCS) and to evaluate the prevalence of NCC among PWE in the rural communities of the Bolivian Chaco after 30 years. Methods Twenty‐two rural communities (total population 12 852) were involved in the study. PWE in the study area were ascertained by multiple sources and the diagnosis was confirmed by a neurologist. All PWE identified were invited to undergo brain CT scan examination and diagnosis of NCC was sought according to the revised Del Brutto criteria. Results Seventy‐eight PWE (30 men, 38.4%; mean age at onset was 12.7 ± 13.2 years) with EATCS were identified giving a crude prevalence of 6.1/1000 (95% CI: 4.7–7.3). Due to the COVID‐19 lockdown, the study was interrupted in 2020 and only 36 PWE (46%) of the whole sample underwent CT scan examination. Of these, 8 (22.2%) fulfilled the criteria for NCC of whom 6 (75%) presented only single or multiple calcifications. Significance This is the first study reassessing the prevalence of NCC among PWE after 30 years, in the same rural area and using a population‐based design. T. solium is still endemic in the Bolivian Chaco where more than 20% of EATCS may be attributable to NCC. Our findings show a substantially unchanged prevalence of NCC over the past 30 years despite improved knowledge, underlining the need for active intervention programs to control T. solium transmission in this area. Plain Language Summary Neurocysticercosis is still endemic in the Bolivian Chaco. The proportion of epilepsy attributable to neurocysticercosis is about 22%. Systematic efforts towards elimination of neurocysticercosis in these areas should be implemented.

This study provides an overview of the forests of Basilicata, Southern Italy, including their recent history, dominant forest types, current management, and vulnerability to climate change and wildfire. It outlines silvicultural and management proposals that can be implemented in the new forest plan that the Basilicata Region is about to adopt. The proposals are based on the principle of adaptive management to support forest functionality, biodiversity and ecosystem services. Silvicultural methods include continuous cover forestry, natural regeneration, species richness and functional diversity, structural diversification, imitation of natural disturbances, tree retention to increase biodiversity. The characteristics of the forest-wood supply chain have been analyzed, highlighting weaknesses and possible improvements.

Abstract Metastasis arises from disseminated tumour cells (DTCs) that are characterized by intrinsic phenotypic plasticity and the capability of seeding to secondary organs. DTCs can remain latent for years before giving rise to symptomatic overt metastasis. In this context, DTCs fluctuate between a quiescent and proliferative state in response to systemic and microenvironmental signals including immune-mediated surveillance. Despite its relevance, how intrinsic mechanisms sustain DTCs plasticity has not been addressed. By interrogating the epigenetic state of metastatic cells, we find that tumour progression is coupled with the activation of oncogenic enhancers that are organized in variable interconnected chromatin domains. This spatial chromatin context leads to the activation of a robust transcriptional response upon repeated exposure to retinoic acid (RA). We show that this adaptive mechanism sustains the quiescence of DTCs through the activation of the master regulator SOX9. Finally, we determine that RA-stimulated transcriptional memory increases the fitness of metastatic cells by supporting the escape of quiescent DTCs from NK-mediated immune surveillance. Overall, these findings highlight the contribution of oncogenic enhancers in establishing transcriptional memories as an adaptive mechanism to reinforce cancer dormancy and immune escape, thus amenable for therapeutic intervention.

The research aims on thermophysical properties of an ancient silver fir wood using transient plane heat source method in three directions of wood defined under isotropic environment. Crucial parameters studied at room temperature include thermal conductivity, thermal diffusivity, thermal effusivity. Analysis of measured thermal responses were done with respect to the full range of time window interval and shortened interval chosen for fitting by model. The depth of heat penetration was investigated with an emphasis on the criteria for evaluating the thermal response published literature (Boháč et al., 2000). The findings support the challenges and understanding of the interaction of wood's orthogonality loaded when interacting with different heating powers (25, 40, 50, 70) (m W) with time (s). The inhomogeneous structure for different levels of thermal energy provided valuable insights into the complex nature of wood and essential for applications requiring heat management such as preservation, construction, material engineering, in specific direction of wood structure.

A pesquisa teve como objetivo adaptar e validar a escala de autorrelato de precariedade no trabalho para o contexto brasileiro de trabalho “gig”, ou trabalho independente. Análises fatoriais exploratória e confirmatória foram realizadas em uma amostra de 504 trabalhadores brasileiros. Além disso, para análise de validade de critério, foram realizadas correlações de ordem zero em uma amostra de 304 trabalhadores brasileiros. Os resultados indicam que a escala de autorrelato de precariedade no trabalho é uma medida multidimensional com estrutura hierárquica. As análises de confiabilidade indicam que a escala apresentou consistência interna adequada tanto para as quatro dimensões quanto para a escala completa. Os resultados relativos à validade de critério demonstram que a precariedade no trabalho está negativamente ligada ao bem-estar e positivamente associada ao mal-estar; além disso, as dimensões da precariedade e da remuneração do trabalho apresentam associações significativas com todas as variáveis de resultado do trabalho investigadas. Por fim, a versão brasileira da escala de autorrelato de precariedade no trabalho é um instrumento com qualidades psicométricas robustas para avaliar a percepção dos trabalhadores sobre as condições de trabalho precárias no contexto brasileiro de trabalho independente. Além disso, a escala alarga o âmbito da investigação sobre condições de trabalho precárias e o seu impacto nas experiências psicológicas e nos resultados do trabalho.

Abstract DNA damage resistance is a major barrier to effective DNA-damaging therapy in multiple myeloma (MM). To discover mechanisms through which MM cells overcome DNA damage, we investigate how MM cells become resistant to antisense oligonucleotide (ASO) therapy targeting Interleukin enhancer binding factor 2 (ILF2), a DNA damage regulator that is overexpressed in 70% of MM patients whose disease has progressed after standard therapies have failed. Here, we show that MM cells undergo adaptive metabolic rewiring to restore energy balance and promote survival in response to DNA damage activation. Using a CRISPR/Cas9 screening strategy, we identify the mitochondrial DNA repair protein DNA2, whose loss of function suppresses MM cells’ ability to overcome ILF2 ASO−induced DNA damage, as being essential to counteracting oxidative DNA damage. Our study reveals a mechanism of vulnerability of MM cells that have an increased demand for mitochondrial metabolism upon DNA damage activation.

New Italian data on the distribution of 17 Annex I Habitats are reported in this contribution. Specifically, 11 new occurrences in Natura 2000 sites are presented and 30 new cells are added in the EEA 10 km × 10 km reference grid. The new data refer to the Italian administrative regions of Apulia, Campania, Calabria, Lazio, Sardinia, Sicily and Tuscany.

L’articolo analizza la quaestio de veneficiis del 331 a. C., la prima nella storia di Roma (Liv. viii 18), sviluppatasi da una delle tante pestilentiae del periodo altorepubblicano, culminata nel processo per veneficium e nella condanna di un folto gruppo di matrone. Alla base della vicenda vi è l’interferenza di piani fra sicurezza pubblica, salute dei cittadini e comportamento femminile, che genera due soluzioni distinte, una giudiziaria e laica (la quaestio) e una religiosa e prodigiale (il clavus a dictatore fixus). Se nella versione liviana l’aspetto processuale si presenta oggettivo e cronachistico, più problematico appare quello religioso, anche per la dimensione politica che viene proiettata su tutta la vicenda dall’associazione delle matrone venefiche alle rivolte plebee, risolte tramite lo stesso piaculum.

In this study, we aimed to evaluate the relationships between the constituents of bulk milk of Valle del Belice sheep. During two years of investigation, 693 milk samples were collected and analysed to assess the major physicochemical parameters. Comparative analysis of partial and simple correlations revealed a strong correlation between milk constituents without the effect of third-party parameters.

Colostrum provides newborns with nutrients and immunoglobulins that play a major role in the immune system. In this survey, the immunoglobulin G (IgG) content, gross composition, mineral content, and fatty acid composition were determined in the colostrum of Valle del Belice ewes with the aim of evaluating the effect of lambing of season. In total, we analysed 60 bulk tank colostrum samples taken after lambing during two seasons (summer and winter). The IgG content and Brix mean values were 40.35 g/L and 21.02%, respectively. The colostrum fat, protein, casein, and lactose percentages were 8.01, 12.10, 8.51, and 3.13%, respectively. The lambing season significantly influenced the pH and somatic cell count, with higher values observed in winter than in summer. The most common fatty acids were palmitic (27.75 vs. 24.84 g/100 g), oleic (24.57 vs. 18.15 g/100 g), myristic (12.55 vs. 12.16 g/100 g) and stearic (7.02 vs. 6.16 g/100 g) detected in the summer and winter seasons, respectively. The winter season significantly increased the polyunsaturated fatty acid values (11.18 vs. 6.41). Additionally, the lambing season determined different mineral composition levels in the colostrum, particularly copper (8.50 vs. 5.35 µmol L−1) and manganese (3.35 vs. 0.80 µmol L−1), which were higher in the summer. Based on IgG content, the quality of the colostrum was excellent.

The recycling of PET trays is highly challenging. The aim of this paper was to investigate the issues related to the mechanical recycling process and, the correlation between feedstock composition and the quality of the produced rPET. Four feedstocks with different degrees of impurity were mechanically recycled at a laboratory pilot scale. The optical and thermal properties of the rPET products were examined to determine the quality and to seek relations with the starting level of impurities. The final products of the PET trays’ mechanical recycling were found to be affected by the presence of impurities (organics) and multi-material (non-PET) elements in the feedstocks. The rPET products crystallised faster for contaminated feedstocks showed lower molecular mass and higher yellow index values due to thermal degradation. Yellowing is a crucial parameter in assessing the thermal degradation of rPET. Injection moulded samples corresponding to higher contamination levels, reported values of Yellow Index equal to 179 and 177 compared to 15 of mono-PET sample. The intrinsic viscosity decreased from 0.60 dL/g to just above 0.30 dL/g, and losses were more significant for soiled or multi-material feedstocks. A method of improving the final quality would involve the purification of the starting feedstock from impurities.

From a circular bio-economy perspective, biomass valorization requires the implementation of increasingly efficient extraction techniques to ensure the environmental and economic sustainability of biorefining processes. This research focuses on optimizing the specialized metabolite extraction of Turkey oak chips from Quercus cerris L. by applying a 3 levels Full Factorial Design (FFD). The goal is to obtain an extract with the highest antioxidant activity [evaluated by 1,1-diphenyl-2-picryl hydrazyl (DPPH) scavenging activity and ferric reducing antioxidant power (FRAP) assays] and specialized metabolites content [measured as total phenolic content (TPC), total flavonoid content (TFC), condensed tannin content (CTC), and hydrolysable tannins content (THC)]. With this objective, three different variables were investigated and compared: temperature (20 °C, 50 °C, 80 °C), solvents EtOH/H2O (0%, 20%, 40%), and time (3 h, 6 h, 24 h), resulting in 27 different extracts. Following the FFD analysis, the optimal extractive conditions were determined to be 80 °C, 40% EtOH/H2O, and 19.8 h. Finally, the prediction ability of FFD was compared with that of artificial neural network (ANN) for DPPH scavenging activity, FRAP, and TPC data based on the coefficient of determination (R2), mean absolute error (MAE), and root mean square error (RMSE). The results indicated that ANN predictions were more precise than FFD ones; however, both methods were useful in optimizing the extraction process as they returned comparable optimized extraction parameters.
