Lysophosphatidylcholine acyltransferase 1(LPCAT1), a key metabolic enzyme in the phosphatidyl-choline metabolism pathway, mediates phosphatidylcholine synthesis through deacylation-reacylation, which leads to alterations in the phospholipids composition of cell membranes and the remodelling of the cellular cytoskeleton. Lysophosphatidylcholine acyltransferase 1 has been shown to be highly expressed in gastric, breast and colorectal cancer, and can accelerate the alteration of the phospholipids composition of tumor cell membranes or interact with tumor driver genes, such as epidermal growth factor receptor and cell cycle-related genes. It might promote tumor development by affecting tumor cell proliferation, migration, invasion and resistance to chemotherapy.

Rail breakage can cause train derailment that leads to catastrophic consequences, prevention of which is worthy of a significant amount of investment in finance and time. Non-destructive evaluation can provide qualitative and quantitative inspection of rail tracks in-situ to detect dangerous faults, even though these defects are primarily invisible or hardly noticed from the surface of the rail. To make it efficient and convenient, infrared thermography technique offers a remote operation without interfering with rail transportation. The image processing for edge detection can further quantify the defect deterioration by depicting the contour of the defect. Raspberry ketone accounts for the characteristic aroma of the raspberry fruit. In order to explore the genes involved in raspberry ketone synthesis, the transcriptome in fruit tissues of two red raspberry varieties "Polka" and "Orange legend", were sequenced and 24213 single genes were obtained. As the red raspberry fruit ripening, genes involved in flavonoid and anthocyanin synthesis were up-regulated, while those associated with lignin synthesis were down-regulated. A gene (RinPKS4) highly related to raspberry ketone synthesis was identified by transcriptome analysis, and RinPKS4 gene was over-expressed in raspberry in order to further understand the function of RinPKS4 gene in raspberry ketone synthesis. The results showed that the gene expression level of RinPKS4 in the leaf tissues of a transgenic lines increased by about 4-fold and the content of raspberry ketone increased by 42.64% compared with the wide type. Contamination by zooplankton has to a certain extent limited the large-scale cultivation and industrial exploitation of microalgae. However, systematic research on these predators in microalgal culture is still lacking. The identification of zooplanktonic contaminants derived from microalgal cultures is a basis for conducting related studies. Moreover, knowledge of the ecological distribution of such predators is crucial for avoiding or reducing the risk of biological contamination in the management of large-scale microalgal cultures. Understanding the feeding behaviors of zooplanktonic contaminants contributes to the establishment of targeted prevention strategies and control methods. Early detection is essential to allow prevention and control measures to be implemented in a timely and effective way. Reducing the susceptibility of the cultured microalgae to predators through breeding strains selection, the potential of modern molecular methods, or a synthesis of these approaches will be indispensable to the management of zooplankton contamination. Furthermore, exploring the resource utilization of predators helps to understand this issue comprehensively and to turn hazard into wealth. The genus Poterioochromonas is a typical mixotrophic flagellate and has attracted increasing attention because of the dramatic damage it can inflict on a wide range of microalgal cultures, regardless of the culture system, season, or environment. This review explores our current understanding of the predator Poterioochromonas and the areas where further research is needed, which should stimulate reflection on what we still need to know about these predators from a microalgal culture perspective and how we can utilize them. Elimination of antibiotics such as tetracycline hydrochloride (TC) from wastewater is of great significance, but still faces challenges. In this regard, Cu2O decorated CuZnAl-layered double hydroxide (Cu2O/CuZnAl-LDHs) heterostructure photocatalysts were designed via a facile coprecipitation method and subsequent in situ reduction approach. The XRD, XPS, UV-vis DRS, PL, Mott–Schottky plots, and VB-XPS results together revealed that the construction of a heterostructure inhibits the recombination of relatively useful electrons and holes, providing a strong driving force for the photocatalytic degradation of TC. Additionally, the synergistic effects increased visible light absorption and the population of adsorption sites, which also played pivotal roles in the removal of TC. As a result, the degradation efficiency of TC catalyzed by Cu2O/CuZnAl-LDHs under visible light reached 94.1% within 120 min. The capture experiments of active species and ESR analysis confirm that the superoxide radical (˙O2−), holes (h+) and hydroxyl radicals (˙OH) were responsible for the decomposition of TC. Mixotrophic flagellates play an important role in connecting the classical food chain and microbial food loop. The feeding characteristics of the mixotrophic flagellate Poterioochromonasmalhamensis have been well studied, but its role as a food source for other large zooplankton is less studied. This study focuses on the physiological and biochemical changes in P. malhamensis when using autotrophy, chemoheterotrophy, and phagotrophy, and the effect of these changes on the feeding ability of one of its predators, the ciliate Paramecium caudatum. The results showed that chemoheterotrophic P. malhamensis had a higher growth rate and larger cell size than autotrophic and phagotrophic P. malhamensis. The biochemical composition of P. malhamensis also varied greatly between the three nutritional modes. The protein, total absolute amino acid, and fucoxanthin contents were highest for autotrophic P. malhamensis, while chemoheterotrophic P. malhamensis had the highest contents of total sugar and total absolute fatty acid. The contents of most biochemical components in phagotrophic P. malhamensis fell between those in autotrophic and chemoheterotrophic P. malhamensis. A feeding experiment showed that the grazing ability of P. caudatum on chemoheterotrophic P. malhamensis was significantly higher than that on phagotrophic P. malhamensis and autotrophic P. malhamensis. This study showed that the transformation of nutritional modes can alter the biochemical composition of the mixotrophic flagellate P. malhamensis and, as a result, affect the grazing ability of its predator P. caudatum. Background and Aim: Metabolic reprogramming is characterized by dysregulated levels of metabolites and metabolic enzymes. Integrated metabolomic and transcriptomic data analysis can help to elucidate changes in the levels of metabolites and metabolic enzymes, screen the core metabolic pathways, and develop novel therapeutic strategies for cancer. Methods: Here, the metabolome of gastric cancer tissues was determined using liquid chromatography–mass spectrometry. The transcriptome data from The Cancer Genome Atlas dataset were integrated with the liquid chromatography–mass spectrometry data to identify the common dysregulated gastric cancer‐specific metabolic pathways. Additionally, the protein expression and clinical significance of key metabolic enzymes were examined using a gastric cancer tissue array. Results: Metabolomic analysis of 16 gastric cancer tissues revealed that among the 15 dysregulated metabolomic pathways, the aminoacyl‐tRNA biosynthesis pathway in the gastric tissues was markedly upregulated relative to that in the adjacent noncancerous tissues, which was consistent with the results of transcriptome analysis. Bioinformatic analysis revealed that among the key regulators in the aminoacyl‐tRNA biosynthesis pathway, the expression levels of threonyl‐tRNA synthetase (TARS) and phenylalanyl‐tRNA synthetase (FARSB) were correlated with tumor grade and poor survival, respectively. Additionally, gastric tissue array data analysis indicated that TARS and FARSB were upregulated in gastric cancer tissues and were correlated with poor prognosis and tumor metastasis. Conclusions: This study demonstrated that the aminoacyl‐tRNA biosynthesis pathway is upregulated in gastric cancer and both TARS and FARSB play key roles in the progression of gastric cancer. Background. Early diagnosis of hypoxic-ischaemic encephalopathy (HIE) is crucial in preventing neurodevelopmental disabilities and reducing morbidity and mortality. The study was to investigate the plasma metabolic signatures in the peripheral blood of HIE newborns and explore the potential diagnostic biomarkers. Method. In the present study, 24 newborns with HIE and 24 healthy controls were recruited. The plasma metabolites were measured by gas chromatography-mass spectrometry (GC-MS), and the raw data was standardized by the EigenMS method. Significantly differential metabolites were identified by multivariate statistics. Pathway enrichment was performed by bioinformatics analysis. Meanwhile, the diagnostic value of candidate biomarkers was evaluated. Result. The multivariate statistical models showed a robust capacity to distinguish the HIE cases from the controls. 52 metabolites were completely annotated. 331 significantly changed pathways were enriched based on seven databases, including 33 overlapped pathways. Most of them were related to amino acid metabolism, energy metabolism, neurotransmitter biosynthesis, pyrimidine metabolism, the regulation of HIF by oxygen, and GPCR downstream signaling. 14 candidate metabolites showed great diagnostic potential on HIE. Among them, alpha-ketoglutaric acid has the potential to assess the severity of HIE in particular. Conclusion. The blood plasma metabolic profile could comprehensively reflect the metabolic disorders of the whole body under hypoxia-ischaemic injury. Several candidate metabolites may serve as promising biomarkers for the early diagnosis of HIE. Further validation based on large clinical samples and the establishment of guidelines for the clinical application of mass spectrometry data standardization methods are imperative in the future. Background: This study examines the effects of sports drinks ingestion during high-intensity exercise for carbohydrate oxidation rate (CHO-O) among athletes. Methods: PubMed, Embase, and the Cochrane library were searched for available papers published up to November 2019. The primary outcome is the carbohydrate oxidation rate (CHO-O), and the secondary outcome is the fat oxidation rate (Fat-O). Statistical heterogeneity among the included studies was evaluated using Cochran's Q test and the I2 index. The random-effects model was used for all analyses, regardless of the I2 index. Results: Five studies are included, with a total of 58 participants (range, 8–14/study). All five studies are randomized crossover trials. Compared to the control beverages, sports drinks have no impact on the CHO-O of athletes [weighted mean difference (WMD) = 0.29; 95% CI, −0.06 to 0.65, P = 0.106; I2 = 97.4%, P < 0.001] and on the Fat-O of athletes (WMD = −0.074; 95% CI, −0.19 to 0.06, P = 0.297; I2 = 97.5%, P < 0.001). Carbohydrate–electrolyte solutions increase CHO-O (WMD = 0.47; 95% CI, 0.08–0.87, P = 0.020; I2 = 97.8%, P < 0.001) but not Fat-O (WMD = −0.14; 95% CI, −0.31 to 0.03, P = 0.103; I2 = 98.2%, P < 0.001). Caffeine has a borderline effect on Fat-O (WMD = 0.05; 95% CI, 0.00–0.10, P = 0.050). Conclusions: Compared with the control beverages, sports drinks show no significant improvement in CHO-O and Fat-O in athletes. Grazing behaviour between protozoa and phytoplankton exists widely in planktonic ecosystems. Poterioochromonas malhamensis is a well‐known and widespread mixotrophic flagellate, which is recognized to play an important role within marine and freshwater planktonic ecosystems and regarded as the greatest contamination threat for mass algal cultures of Chlorella. In this study, a comprehensive range of factors, including morphological characters, biochemical compositions, and specific growth rate of ten species or strains of Chlorella, were evaluated for their effect on the feeding ability of P. malhamensis, which was assessed by two parameters: the clearance rate of P. malhamensis on Chlorella spp. and the specific growth rate of P. malhamensis. The results showed that the clearance rate of P. malhamensis was negatively correlated with cell wall thickness and specific growth rate of Chlorella spp., while the specific growth rate of P. malhamensis was positively correlated with carbohydrate percentage and C/N ratio and negatively correlated with protein, lipid percentage, and nitrogen mass. In conclusion, the factors influencing feeding selectivity include not only the morphological character and chemical composition of Chlorella, but also its population dynamics. Our study provides useful insights into the key factors that affect the feeding selectivity of P. malhamensis and provides basic and constructive data to help in screening for grazing‐resistant microalgae. Hypothalamic dysfunction is a key pathological factor in inflammation-associated depression. In the present study, isobaric tags for relative-absolute quantitation (iTRAQ) combined with mass spectrometry and gas chromatography-mass spectrometry (GC-MS) were employed to detect the proteomes and metabolomes in the hypothalamus of the lipopolysaccharide (LPS)-induced depression mouse, respectively. A total of 187 proteins and 27 metabolites were differentially expressed compared with the control group. Following the integration of bi-omics data, pertinent pathways and molecular interaction networks were further identified. The results indicated altered molecules were clustered into Ephrin receptor signaling, glutamatergic transmission, and inflammation-related signaling included the LXR/RXR activation, FXR/RXR activation, and acute phase response signaling. First discovered in the hypothalamus, Ephrin receptor signaling regulates N -methyl-D-aspartate receptor (NMDAR)-predominant glutamatergic transmission, and further acted on AKT signaling that contributed to changes in hypothalamic neuroplasticity. Ephrin type-B receptor 2 (EPHB2), a transmembrane receptor protein in Ephrin receptor signaling, was significantly elevated and interacted with the accumulated NMDAR subunit GluN2A in the hypothalamus. Additionally, molecules involved in synaptic plasticity regulation, such as hypothalamic postsynaptic density protein-95 (PSD-95), p-AKT and brain-derived neurotrophic factor (BDNF), were significantly altered in the LPS-induced depressed group. It might be an underlying pathogenesis that the EPHB2-GluN2A-AKT cascade regulates synaptic plasticity in depression. EPHB2 can be a potential therapeutic target in the correction of glutamatergic transmission dysfunction. In summary, our findings point to the previously undiscovered molecular underpinnings of the pathophysiology in the hypothalamus of inflammation-associated depression and offer potential targets to develop antidepressants. BCG, the only registered vaccine against Mycobacterial Tuberculosis (TB) infection, has been questioned for its protective efficacy for decades. Although lots of efforts were made to improve the BCG antigenicity, few studies were devoted to understand the role of host factors in the variability of the BCG protection. Using the IL-10KO mice and pulmonary tuberculosis infection model, we have addressed the role of IL-10 in the BCG vaccination efficacy. The data showed that IL-10-deficient dendritic cells (DCs) could promote the immune responses through upregulation of the surface costimulatory molecule expression and play an orchestra role through activating CD4+T cell. IL-10-deficient mice had higher IFN γ, TNF α, and IL-6 production after BCG vaccination, which was consistent with the higher proportion of IFN γ+CD3+, IFN γ+CD4+, and IFN γ+CD8+ T cells in the spleen. Particularly, the BCG-vaccinated IL-10KO mice showed less inflammation after TB challenge compared to WT mice, which was supported by the promoted Th1 and Tc, as well as the downregulated Treg responses in IL-10 deficiency. In a conclusion, we demonstrated the negative relationship between Th1/Tc responses with IL-10 production. IL-10 deficiency restored the type 1 immune response through DC activation, which provided better protection against TB infection. Hence, our study offers the first experimental evidence that, contrary to the modulation of BCG, host immunity plays a critical role in the BCG protective efficacy against TB. [ABSTRACT FROM AUTHOR]

Hepatitis B virus (HBV) infection remains a severe clinical concern in China. Of note, the progression of HBV infection varies between different populations. To identify the factors that influence the disease progression and prognosis, a total of 478 chronic HBV-infected patients were enrolled, and liver function parameters, HBV DNA levels and hepatic fibrosis indices were analyzed. First, the results demonstrated a significant difference in hepatitis B e antigen (HBeAg) expression between male and female patients (χ2=4.061, P=0.044). Furthermore, when comparing either HBeAg-negative or -positive male and female patients, males exhibited a greater variation in HBV DNA levels. Although significant differences between male and female patients in certain abnormal ratios of liver function parameters were identified, a trend in the differences was observed in the HBeAg-negative and -positive groups. When considering age, the results of the present study confirmed that HBV DNA levels decreased with advanced age, and the values of the majority of biomarkers exhibited an evident decreasing trend with increasing age. In addition, it was demonstrated that all HBeAg seropositive patients had higher levels of hepatic fibrosis indexes and higher abnormal ratios of hepatic fibrosis values in their serum when compared with those of HBeAg seronegative patients, particularly with regard to serum IV collagen. The present results revealed that HBV DNA replication was closely associated with liver function; however, it was notable that in HBeAg-negative patients, the association between HBV DNA levels and liver function was particularly significant among subjects aged <61. Furthermore, this result was not observed in HBeAg-positive patients. In conclusion, the present study indicated the importance of host factors (including sex and age) and viral factors (including HBeAg expression pattern and HBV DNA levels) in the progression of chronic HBV infection, and its influence regarding prognosis and treatment. A C 3-symmetrical 4,4″,4⁗-nitrilotris(2′-methyl-[1,1′-biphenyl]-3-carboxylic acid) (4) derived from nitrilotriacetic acid (NTA) was found to selectively bind Zinc(ii) ions both in DMSO or MeOH. A synergistic effect of the anionic counter ion SO42− on the sensing behaviour of 4 to metal ions was clearly observed in DMSO. Interestingly, 4 showed a rapid hypochromatic shift in emission ascribed to the deprotonation and the concomitant formation of a 4 –metal complex upon the addition of Zn2+ ions, instead of the bathochromic shift and emission enhancement attributed to the SO42−-involved hydrogen-bonding interaction for Ni2+, Li+, Mg2+, and Na+ ions at ratios below 1:1 in DMSO. The observed sensing process of sulfate salts associated with the SO42−-involved hydrogen-bonding interaction, deprotonation, and the concomitant complexation can also be clearly monitored by titration methods utilising UV-vis, fluorescence, and NMR spectroscopy in solution. In comparison with 4 , compound 1 showed an obvious difference in the binding interaction with zinc sulfate in MeOH, probably owing to the decreased acidity. Anion-induced hydrogen-bonding interactions and deprotonation of the COOH protons in the excited state also endowed 4 versatile spectroscopic properties. The addition of F− and SO42− anions resulted in a remarkable enhancement probably related with a rigidifying effect. 2,2′,2″-Nitrilotribenzoic acid can be utilised as a potential scaffold to build a series of conjugated fluorescent sensors by its chelation effect owing to the rigid cavity pre-organised by the triphenylamine moiety and the carboxylic groups and the conjugation extension in the 4,4′,4″ positions. Although the Ficoll-Paque method is classically used to isolate peripheral blood mononuclear cells (PBMCs), modifications in this method are required for a more rapid and economic output for biobanks and clinical laboratories, particularly in developing countries. In this study, we addressed this issue by modifying the Ficoll-Paque method for the isolation of PBMCs or mononuclear cells from the peripheral and the umbilical cord blood of healthy and diseased (infected, anemic, and chronic obstructive pulmonary disease) adult individuals. In the modified method, we initiated the cell isolation process from the buffy coat layer, which appears in the interface between the plasma and sediments after centrifugation, instead of using the whole blood as described in the classic method. Although the PBMC yield by the modified method was about 12% less than in the classic method, the number of PBMCs isolated by the modified method was more than one million, which is enough for different research/diagnostic purposes, such as multi-omics detection. Assessment of cell viability and purity by hematology analyzer and trypan blue showed no significant difference between the viability and purity of the PBMCs isolated by these two methods in almost all groups, except samples from the infected and cord blood groups, where lower PBMC purity with higher granulocyte contamination were observed. In addition, at delayed processing time points, all parameters for the two methods were decreased in a time-dependent manner, especially at 8, 12, or 24 hours after the sample collection. In summary, the performance of PBMC isolation by the classic and modified methods mainly relies on the PBMC ratio in original samples. Diptychus maculatus(Cypriniformes: Cyprinidae), the sole species of genusDiptychus, is an economically important freshwater fish widely distributed in Asia. In this study, we first sequenced the complete mitochondrial genome ofD. maculatus.The genome is 16,835 bp in length, consisting of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes as well as 2 non-coding regions: origin of light–strand replication (OL) and control region (D-loop). The gene composition and order are identical with most bony fishes. The overall nucleotide composition of the heavy strand is 27.7% A, 26.7% T, 26.5% C and 19.1% G. The mitogenome data would be useful for further genetics studies, phylogenetic analysis, and resource protection ofD. maculatusand phylogenetic analysis of Schizothoracine fishes. There is an urgent need to identify the potential risk factors for activating latent Mycobacterium tuberculosis infection. In this study, we evaluated the immune function of Rv1737c, which is a latency‐associated antigen of dormancy survival regulator (DosR) of M. tuberculosis in a mouse model. Our data showed that mice pretreated with recombinant Rv1737c (rRv1737c) exhibited higher levels of antigen‐specific antibodies (IgG, IgM and IgA) than sham‐treated mice. Following Bacilli Calmette‐Guerin (BCG) challenge, rRv1737c adjuvanted with cholera toxin subunit B (CTB) induced diffuse lung inflammation and fibrosis compared to the control mice. The inflammatory pathogenesis due to rRv1737c pre‐exposure was associated with a switch in the macrophage phenotype from M1 to activated M2 and was characterized by IL‐10 production. Intracellular cytokine analysis further showed that the rRv1737c‐pretreated mice exhibited an increased frequency of Th2 cells in the lungs, lymph nodes and spleen after BCG challenge. Furthermore, IFN‐γ expression increased in the lungs after rRv1737c pretreatment compared to that in the sham mice. Accordingly, lung cells from rRv1737c‐immunized mice stimulated with killed BCG produced higher levels of multiple cytokines, such as IFN‐γ, IL‐10 and IL‐6. The results confirmed that the pathological features of rRv1737c promoted inflammation. Overall, our findings provide direct evidence of the pro‐inflammatory function of rRv1737c in a murine model of BCG infection, indicating that Rv1737c is a pathogenic antigen of M. tuberculosis and may be key to the recurrence of latent infection.
