The transformative impact of advancements in educational technology, particularly those powered by artificial intelligence (AI), on the landscape of education and the teaching profession has been substantial. This study explores the repercussions of AI-powered technologies on teachers' autonomous behavior, digital burnout, and professional development. The study involved a cohort of 320 high school teachers in China segregated into control and experimental groups. The experimental group received instructions on AI-integrated applications and how they might be used in education. However, the teachers assigned to the control group did not receive information on the use of AI educational applications. Three distinct questionnaires probing autonomous behaviors, digital burnout, and online professional development were administered, and the ensuing data were analyzed using independent sample t-tests. The findings elucidate a discernible positive impact of AI-integrated technology intervention on teachers' professional development and autonomous behaviors. The incorporation of AI-enhanced tools facilitated an augmentation in teachers' professional growth and bolstered their independent and self-directed instructional practices. Notably, using AI-integrated technology significantly reduced teachers' susceptibility to digital burnout, signifying a potential alleviation of stressors associated with technology- mediated teaching. This research provides valuable insights into the multifaceted effects of AI-powered technologies on educators, shedding light on enhancing professional competencies and mitigating digital burnout. The implications extend beyond the confines of this study, resonating with the broader discourse on leveraging technology to augment the teaching profession and optimize the learning environment.

Young children with early emotional, behavioral, and regulatory problems are at risk for long-term behavioral and academic challenges. This study examined the associations between O-classroom quality (i.e. emotional, organizational, and instructional supports) and problem behavior in six-year-old children who attend preschool. A correlation study design, under a quantitative approach, was used. Participants included 503 children (244 male and 259 female) living in the Wolaita communities of Southern Ethiopia. The Classroom Assessment Scoring System (CLASS; Pianta, La Paro, & Hamre, 2008) was used to assess classroom quality across three domains: emotional support, classroom organization, and instructional support by certified observers. The O- classrooms were observed three times during six cycles, twice in the first, second, and third observation. Furthermore, the Preschool and Kindergarten Behavior Scales (PKBS, Merrell, 1994, 2002) were used to measure children's problem behavior. Hierarchical linear modeling results showed that the O-classroom quality score had a significant negative correlation with problem behavior. Analyses revealed that the combined effects of observed O- classroom quality explained a 73% variance in children's problem behavior. Taken together, these findings indicate that classroom organization and emotional support significantly and negatively predicted children's problem behavior. Areas of further research and implications for policy and practice are advanced based on the major findings.

Abstract Background The genus Sapovirus in the family Caliciviridae comprises of a genetically diverse group of viruses that are responsible for causing acute gastroenteritis in both human and animals globally. As the number of sequences continues to grow and more recombinant sequences are identified, the classification criteria of genogroups and genotypes of sapovirus need to be further refined. Abstract Multi-constraint UAV path planning problems can be viewed as many-objective optimization problems that can be solved by meta-heuristic algorithms with good self-organizing optimization capabilities. However, such algorithms mostly use random initializing methods, resulting in low-quality initial paths that reduce the efficiency of subsequent algorithmic searches. Moreover, as the number of objective functions increases, meta-heuristic algorithms face inadequate selection pressure and convergence capability, which lead to poor solution. In order to address these issues, this paper proposes a UAV path planning method based on the framework of multi-objective jellyfish search algorithm (UMOJS). Firstly, an initializing strategy based on Rapidly-exploring Random Trees (RRT) is proposed to achieve higher quality initial paths. Secondly, a jellyfish updating strategy guided by the class-optimal individual is designed to enhance the convergence ability of the algorithm. Furthermore, a set of predefined reference points is imported to obtain Pareto optimal solutions with better convergence and distribution in many-objective optimization problems. To evaluate the superiority of the proposed UMOJS algorithm, three different difficulties of simulated flight environments are constructed to verify its performance. The experimental results show that UMOJS is not only able to gain more UAV paths with shorter length, but also more evenly distributed Pareto optimal solutions compared to five meta-heuristic algorithms when the constraint conditions are satisfied.

Abstract Questionnaires completed by 589 college students were used to investigate the impact of the risk perception of the COVID-19 pandemic on college students’ subjective well-being. The goal was to explore the relationship between college students’ risk perception and subjective well-being during the COVID-19 pandemic as well as to analyze the mediating role of emotional regulation and the moderating effect of parenting style. The results showed the following: First, significant negative correlations were found between risk perception and subjective well-being, as well as between the risk perception of the COVID-19 pandemic and emotional regulation. Emotional regulation was significantly positively correlated with subjective well-being. Second, emotional regulation played a mediating role between risk perception and subjective well-being. Third, a positive parenting style played a positive moderating role between risk perception and subjective well-being. We thus concluded that COVID-19 risk perception directly reduces subjective well-being and indirectly affects subjective well-being through emotional regulation. A positive parenting style reduces the negative impact of the risk perception of the COVID-19 pandemic on subjective well-being. This study provides guidance on how to improve college students’ well-being during major public health events.

Abstract We retrospectively investigated the relationship between cardiopulmonary exercise testing (CPET) parameters and coronary microvascular dysfunction (CMD) using a novel angiography-based index of microcirculatory resistance (AccuIMR) in patients with ST-elevation myocardial infarction (STEMI) who underwent primary percutaneous coronary intervention (PCI) with complete revascularization. In 418 patients, the culprit vessel AccuIMR was calculated after successful primary PCI. CPET was conducted 44.04 ± 19.28 days after primary PCI. Overall, 157 patients (37.6%) showed elevated AccuIMR (> 40 U) in the culprit vessels. The LVEF was significantly lower in the CMD group than in the Non-CMD group. The CMD group showed worse results in VO2peak, peak O2-pulse, and VE/VCO2 slope than the Non-CMD group. Abstract Background Exogenous gonadotropin-controlled ovarian stimulation is the critical step in animal reproductive management, such as pig, sheep, bovine and other species. It helps synchronize ovulation or stimulate multiple ovulations. However, a number of evidence indicated an unexpected decrease in pregnancy outcomes following ovarian stimulation. This study aimed to explore the underlying mechanism of the pregnancy defect and develop a practical rescue strategy. Results Compared with those in the control group, gilts that underwent ovarian stimulation showed a decrease in pregnancy rate, farrowing rate, and total number of piglets born. Stimulated gilts also showed an increase in estradiol (E2) levels. The supraphysiological E2 level was correlated with the decrease in the number of piglets born. Furthermore, we found that high levels of E2 impair uterine receptivity, as shown by the overproliferation of endometrial epithelial cells. In vitro mechanistic studies demonstrated that high levels of E2 hyperactivate FGF-FGFR-ERK signaling cascade in the uterine endometrium, and in turn induces overproliferation of endometrial epithelial cells. Of note, N-acetyl-L-cysteine (NAC) supplementation effectively inhibits ERK hyperphosphorylation and ameliorates endometrial epithelial overproliferation. Importantly, in vivo experiments indicated that dietary NAC supplementation, compared with ovarian stimulation group, improves the uterine receptivity in gilts, and significantly increases the pregnancy rate and total number of piglets born. Conclusions Ovarian stimulation-induced supraphysiological levels of E2 impairs uterine receptivity by hyperactivating FGF-FGFR-ERK signaling cascade, thereby reducing pregnancy rate and litter size. Supplementing NAC to a conventional diet for gilts ameliorates hyperactivated ERK signaling and improves uterine receptivity, thus rescuing adverse pregnancy outcomes following ovarian stimulation. Polysaccharides are one of the most important natural bioactive ingredients in pumpkin. Pumpkin polysaccharides have variable structure and complex monosaccharide composition, and possess a variety of biological activities such as hypoglycemia and antioxidant. In recent years, with the increasing incidence of diabetes, the hypoglycemic effect of pumpkin polysaccharides has attracted more and more attentions of scholars. The research focuses on the structure, monosaccharide composition and different mechanism of action of pumpkin polysaccharides. Therefore, this paper systematically summarizes the structural characteristics, mechanisms of hypoglycemic action and influence factors of hypoglycemic activity of pumpkin polysaccharides, and looks forward to its research direction and application prospects, and proposes the suggestions, with a view to providing reference for the subsequent research of pumpkin polysaccharides on diabetes.

Abstract Objective To verify whether the bleeding risk assessment guidelines from the 9th American College of Chest Physicians (ACCP) are prognostic for respiratory intensive care unit (RICU) patients and to explore risk factors for hemorrhages, we conducted a secondary data analysis based on our previously published cohort study of venous thromboembolism. Patients and methods We performed a secondary data analysis on the single-center prospective cohort from our previous study. Patients admitted to the RICU at Beijing Chao-Yang Hospital from August 1, 2014 to December 31, 2020 were included and followed up until discharge. Results The study enrolled 931 patients, of which 715 (76.8%) were at high risk of bleeding, while the remaining were at low risk. Of the total, 9.2% (86/931) suffered major bleeding, and no significant difference was found between the two risk groups (p = 0.601). High-risk patients had poor outcomes, including higher mortality and longer stays. Independent risk factors for major bleeding were APACHE II score ≥ 15; invasive pulmonary aspergillosis; therapeutic dose of anticoagulants; extracorporeal membrane oxygenation; and continuous renal replacement therapy. Abstract Stimulator of interferon genes (STING) is vital in the cytosolic DNA-sensing process and critical for initiating the innate immune response, which has important functions in host defense and contributes to the pathogenesis of inflammatory diseases. Zinc finger CCCH-type antiviral protein 1 (ZC3HAV1) specifically binds the CpG dinucleotides in the viral RNAs of multiple viruses and promotes their degradation. ZAPS (ZC3HAV1 short isoform) is a potent stimulator of retinoid acid-inducible gene I (RIG-I) signaling during the antiviral response. However, how ZC3HAV1 controls STING signaling is unclear. Here, we show that ZC3HAV1 specifically potentiates STING activation by associating with STING to promote its oligomerization and translocation from the endoplasmic reticulum (ER) to the Golgi, which facilitates activation of IRF3 and NF-κB pathway. Accordingly, Zc3hav1 deficiency protects mice against herpes simplex virus-1 (HSV-1) infection- or 5,6-dimethylxanthenone-4-acetic acid (DMXAA)-induced inflammation in a STING-dependent manner. These results indicate that ZC3HAV1 is a key regulator of STING signaling, which suggests its possible use as a therapeutic target for STING-dependent inflammation.

Abstract Background This study aims to assess the performance of an established an AI algorithm trained on conventional polychromatic computed tomography (CT) images (CPIs) to detect pulmonary ground-glass nodules (GGNs) on virtual monochromatic images (VMIs), and to screen the optimal virtual monochromatic energy for the clinical evaluation of GGNs. Methods Non-enhanced chest SDCT images of patients with pulmonary GGNs in our clinic from January 2022 to December 2022 were continuously collected: adenocarcinoma in situ (AIS, n = 40); minimally invasive adenocarcinoma (MIA, n = 44) and invasive adenocarcinoma (IAC, n = 46). A commercial CAD system based on deep convolutional neural networks (DL-CAD) was used to process the CPIs, 40, 50, 60, 70, and 80 keV monochromatic images of 130 spectral CT images. AI-based histogram parameters by logistic regression analysis. The diagnostic performance was evaluated by the receiver operating characteristic (ROC) curves, and Delong’s test was used to compare the CPIs group with the VMIs group. Results When distinguishing IAC from MIA, the diagnostic efficiency of total mass was obtained at 80 keV, which was superior to those of other energy levels (P 0.05). Conclusion The AI algorithm trained on CPIs showed consistent diagnostic performance on VMIs. When pulmonary GGNs are encountered in clinical practice, 80 keV could be the optimal virtual monochromatic energy for the identification of preoperative IAC on a non-enhanced chest CT.

Abstract Background The role of Claudin‐1 in tongue squamous cell carcinoma (TSCC) metastasis needs further clarification, particularly its impact on cell migration. Herein, our study aims to investigate the role of Claudin‐1 in TSCC cell migration and its underlying mechanisms. Methods 36 TSCC tissue samples underwent immunohistochemical staining for Claudin‐1. Western blotting and immunofluorescence analyses were conducted to evaluate Claudin‐1 expression and distribution in TSCC cells. Claudin‐1 knockdown cell lines were established using short hairpin RNA transfection. Migration effects were assessed through wound healing assays. Furthermore, the expression of EMT‐associated molecules was measured via western blotting. Results Claudin‐1 expression decreased as TSCC malignancy increased. Adenosine monophosphate–activated protein kinase (AMPK) activation led to increased Claudin‐1 expression and membrane translocation, inhibiting TSCC cell migration and epithelial–mesenchymal transition (EMT). Conversely, Claudin‐1 knockdown reversed these inhibitory effects on migration and EMT caused by AMPK activation. Conclusions Our results indicated that AMPK activation suppresses TSCC cell migration by targeting Claudin‐1 and EMT pathways.

Objective: To estimate the bone protective effect of alpinumisoflavone, a natural prenylated isoflavonoid, against glucocorticoid-induced osteoporosis in rats. Methods: Male Wistar rats received intramuscular administration of dexamethasone (4 mg/mL) at a dose level of 7 mg/kg for 5 weeks, and then alpinumisoflavone (5, 10, and 15 mg/kg) and alendronate (2 mg/kg) from 2 weeks. Abstract Cardia gastric cancer (CGC) is prevalent in East Asia, and noninvasive, cost-effective screening methods are needed. This study investigated the diagnostic value of serum pepsinogen (PG), gastrin-17 (G-17), Helicobacter pylori (H. pylori) antibodies, and proteomic profiling for CGC and precancerous lesions. We conducted a case-control study involving biopsy-confirmed patients with CGC (n = 60), low-grade intraepithelial neoplasia (CLGD, n = 60), high-grade intraepithelial neoplasia (CHGD, n = 64), and healthy controls (n = 120) matched for age and sex from high-incidence areas in China. Serological markers including PGI, PGII, G-17, and H. pylori were measured using ELISA and Western blot, while plasma protein markers were assessed using Olink® technology. Abstract This research investigates the implications of incorporating blockchain technology into the process of making decisions for green supply chains, particularly under conditions of demand uncertainty. A model was formulated to encompass both environmentally friendly products enabled by blockchain technology and those without such enabling technology. The study further explores the optimal method of introducing green input in a duopoly market using game theory. It also examines how consumer uncertainty about green products and acceptance of the technical parameters of blockchain influence this strategy. The findings suggest that increased consumer uncertainty can, in some instances, motivate manufacturers to enhance the eco-friendliness of their products and improve supply chain performance. However, the universal adoption of blockchain does not necessarily ensure better results; on the contrary, it may compromise product sustainability while enhancing supply chain profitability. Moreover, research has indicated that products enabled by blockchain typically have lower prices, thereby offering potential benefits to consumers when acceptance of sustainable energy solutions is high or uncertain. Additionally, this paper analyzes the impact of changes in green supply chain decision-making on system reliability. This involves exploring the relationship between decision parameters and consumer reluctance towards sustainable products and the adoption of blockchain technology. Abstract Neutrophilic inflammation contributes to multiple chronic inflammatory airway diseases, including asthma and chronic rhinosinusitis with nasal polyps (CRSwNP), and is associated with an unfavorable prognosis. Here, using single-cell RNA sequencing (scRNA-seq) to profile human nasal mucosa obtained from the inferior turbinates, middle turbinates, and nasal polyps of CRSwNP patients, we identify two IL-1 signaling-induced cell subsets—LY6D + club cells and IDO1 + fibroblasts—that promote neutrophil recruitment by respectively releasing S100A8/A9 and CXCL1/2/3/5/6/8 into inflammatory regions. IL-1β, a pro-inflammatory cytokine involved in IL-1 signaling, induces the transdifferentiation of LY6D + club cells and IDO1 + fibroblasts from primary epithelial cells and fibroblasts, respectively. In an LPS-induced neutrophilic CRSwNP mouse model, blocking IL-1β activity with a receptor antagonist significantly reduces the numbers of LY6D + club cells and IDO1 + fibroblasts and mitigates nasal inflammation. This study implicates the function of two cell subsets in neutrophil recruitment and demonstrates an IL-1-based intervention for mitigating neutrophilic inflammation in CRSwNP.

Antibodies targeting non-structural protein 1 (NS1) confer protection against Zika virus (ZIKV). Although monoclonal antibodies (MAbs) 3G2 and 4B8 are more potent than MAb 4F10 in suppressing ZIKV infection in neonatal mice models, the epitopes are unclear. Herein, we determined the Cryo-electron microscopy (Cryo-EM) structures of ZIKV NS1 in complex with five human antibodies at 2.6–2.9 Å resolution. Group I antibodies (3G2 and 4B8) recognize the previously unreported epitopes on the outer surface of the NS1 dimer. The unique binding mode of Group I antibodies led to a stronger recognition of the cell surface form of NS1 and completely inhibited secreted form non-structural protein 1 (sNS1)-induced endothelial permeability via their immunoglobulin G (IgG) and Fab. Group II antibodies (4F10, 2E11, and 14G5) recognize common epitopes in the distal end of the β-ladder domain, with a blockade efficiency that may be related to their affinity for the sNS1 protein and the presence of full-length IgG. These findings elucidate the correlation between epitope recognition and protective efficacy of anti-NS1 antibodies and highlight the diagnostic and therapeutic potential of 3G2 and 4B8.

Abstract. Although COVID‑19 no longer constitutes a “public health emergency of international concern”, which still has being spreading around the world at a low level. Small molecule drugs are the main antiviral treatment for novel coronavirus recommended in China. Although a variety of small‑molecule antiviral drugs against COVID‑19 have been listed in China, there is no specific drug recommendation for special populations. Society of Bacterial Infection and Resistance of Chinese Medical Association, together with the National Clinical Research Center for Respiratory Disease, and the National Center for Respiratory Medicine, organized domestic experts in various fields such as respiratory, virology, infection, critical care, emergency medicine and pharmacy to release Expert Consensus on the Clinical Application of Oral Small‑Molecule Antiviral Drugs against COVID‑19. Abstract The Standardized Precipitation Evapotranspiration Index (SPEI) is a widely recognized and effective tool for monitoring meteorological droughts. However, existing SPEI datasets suffer from spatial discontinuity or coarse spatial resolution problems, which limits their applications at the local level for drought monitoring research. Therefore, we calculated the SPEI index at meteorological stations, combined with the Global Precipitation Measurement (GPM) Precipitation (Pre), Moderate Resolution Imaging Spectroradiometer (MODIS) Land Surface Temperature (LST), ERA5‐Land Shortwave Radiation (SR), Shuttle Radar Topography Mission (SRTM) Digital Elevation Model (DEM) datasets and Random Forest Regression (RFR) model, developed a high spatial resolution (1 km) SPEI (HSPEI) datasets with multiple time scales in mainland China from 2001 to 2022. Compared to other SPEI datasets, the HSPEI datasets have higher spatial resolution and can effectively identify the detailed characteristics of drought in mainland China from 2001 to 2022. Overall, the HSPEI datasets can be effectively applied to the research of different droughts in China from 2001 to 2022.

Triply periodic minimal surface (TPMS) structures with excellent properties of stable energy absorption, light weight, and high specific strength could potentially spark immense interest for novel and programmable functions by combining smart materials, e.g. shape memory polymers (SMPs). This work proposes TPMS lattices with hybrid configurations and materials that are composed of viscoelastic and shape-memory materials with the aim to bring temperature-dependent mechanical properties and additional dissipation mechanisms. Different configurations and diverse materials of polylactic acid (PLA), fiber-reinforced PLA, and polydimethylsiloxane (PDMS) are induced, generating five types of TPMS lattices, including (Schoen’s I-WP) IWP uniform lattice, IWP lattice with density gradient, hybrid configurations, hybrid materials, and filled PDMS, which are fabricated by 3D printing. The fracture morphologies and the distribution of carbon fibers are demonstrated via scanning electron microscopy with a focus on the influence of carbon fiber on shape-memory and mechanical properties. Shape recovery tests are conducted, which proves good shape memory properties and reusable capability of TPMS lattice. The combined methods of experiments and numerical simulation are adopted to evaluate mechanical properties, which presents multi-stage energy absorption ability and tunable vibration isolation performances associated with temperature and hybridization designs. This work can promote extensive research and provide substantial opportunities for TPMS lattices in the development of functional applications.

Abstract Maintenance of power transmission lines is essential for the safe and reliable operation of the power grid. The use of deep learning‐based networks to improve the performance of power line defect detection faces significant challenges, such as small target sizes, shape similarities, and occlusion issues. In response to these challenges, a transformer‐based end‐to‐end power line detection network called Power‐DETR is introduced. Initially, building upon Deformable DETR, a large pre‐trained model (Swin‐large) is utilized to increase the number of multi‐scale features, and activation checkpoint technology is applied to ensure effective training within limited memory capacity. Subsequently, a contrastive denoising training strategy is integrated to combat ambiguity and instability of the Hungarian matching algorithm during training, aiming to expedite model convergence. Additionally, a hybrid label assignment strategy combining OHEM and cost‐based ATSS is proposed to provide the model with high‐quality queries, ensuring adequate training for the decoder and enhancing encoder supervision. Experimental results substantiate the efficacy of the proposed Power‐DETR model as a novel end‐to‐end detection paradigm, surpassing both one‐stage and two‐stage detection models. Furthermore, the model demonstrates a significant 15.7% enhancement in mAP0.5 compared to the baseline.

According to the Species Catalogue of China, Tibet, China, has more than 1,000 species of bryophytes, showing its remarkably high biodiversity. Amongst them, the Polytrichaceae is one of the largest family, with six genera and 25 species, one subspecies and two varieties reported previously.Based on a field survey and specimen identification, the following species have been newly recorded in the bryoflora of Tibet, namely Oligotrichum obtusatum Broth., Pogonatum contortum (Menzies ex Brid.) Lesq. and Polytrichum sphaerothecium (Besch.) Müll. Hal.

Abstract Tamoxifen (TAM) resistance is a major challenge in treating oestrogen receptor-positive (ER+) breast cancers. It is possible that the H2S synthase cystathionine-γ-lyase (CSE), which has been previously shown to promote tumour growth and metastasis in other cancer cells, is involved in this resistance. Therefore, we investigated CSE's role and potential mechanisms in TAM-resistant breast cancer cells. First, we examined the effect of CSE expression on TAM sensitivity and resistance in MCF7 (breast cancer) cells. The findings revealed that CSE was directly associated with TAM sensitivity and involved in TAM resistance in ER+ breast cancer cells, indicating that it may be useful as a biomarker. Next, we wanted to determine the molecular mechanism of CSE's role in TAM resistance. Using cell migration, co-immunoprecipitation, western blotting, and cell viability assays, we determined that the CSE/H2S system can affect the expression of PPARγ by promoting the sulfhydrylation of PPARγ, which regulates the transcriptional activity of ACSL1. ACSL1, in turn, influences STAT3 activation by affecting the phosphorylation, palmitoylation and dimerization of STAT3, ultimately leading to the development of TAM resistance in breast cancer. Finally, we examined the effect of CSE inhibitors on reducing drug resistance to determine whether CSE may be used as a biomarker of TAM resistance. We observed that the novel CSE inhibitor I194496 can reverse TAM resistance in TAM-resistant breast cancer via targeting the PPARγ/ACSL1/STAT3 signalling pathway. Overall, our data indicate that CSE may serve as a biomarker of TAM resistance and that the CSE inhibitor I194496 is a promising candidate for combating TAM resistance.

Abstract Pathogens detection is a crucial measure in the prevention of foodborne diseases. This study developed a novel multicolor colorimetric assay to visually detect Salmonella Typhimurium (S. Typhimurium), by utilizing the etching process of gold nanorods (AuNRs) with TMB2+. The strategy involved the construction of nanozyme by assembling magnetic covalent organic framework (MCOF) with aptamer-conjugated AuNPs (Apt-AuNPs), exhibiting remarkable peroxidase-like activity to catalyze the oxidation of TMB/H2O2 and inducing the etching of AuNRs. The presence of S. Typhimurium could inhibit this process, resulting in the generation of vivid colors. The multicolor colorimetric assay could specifically determine S. Typhimurium from 102 to 108 CFU mL−1 in 60 min with visual detection limit of 102 CFU mL−1, and instrumental detection limit of 2.3 CFU mL−1. Moreover, detecting S. Typhimurium in chicken, milk, pork and lettuce samples has shown promise in practical applications.

Purpose: This study aimed to investigate the effect of the N6-methyladenosine (m6A) dependent ferroptosis on cisplatininduced Sertoli cell injury. Materials and Methods: A cisplatin exposure mouse model was established by intraperitoneal injection of cisplatin in our study. TM4 cell lines was used for in vitro study. Ferroptosis was detected according to metabolomic analysis and a series of assays, including malondialdehyde, glutathione, and glutathione disulfide concentration detection, 2',7'-dichlorodihydrofluorescein diacetate and BODIPY 581/591 C11 probe detection, and transmission electron microscope imaging. Key ferroptosisrelated genes were identified via transcriptomic analysis, western blot and immunohistochemistry. The m6A modification was demonstrated via m6A RNA immunoprecipitation and luciferase reporter assays. Immune cell infiltration was detected by mass cytometry, and verified by flow cytometry and immunofluorescence. Results: Ferroptosis, but not other types of programmed cell death, is a significant phenomenon in cisplatin-induced testis damage and Sertoli cell loss. Ferroptosis induced by cisplatin in Sertoli cell/TM4 cell is GPX4 independent but is regulated by SLC7A11 and ALOX12. Both SLC7A11 and ALOX12 are regulated via m6A dependent manner by METTL3. Furthermore, overexpressed ALOX12-12HETE pathway may result in macrophage polarization and inflammatory response in cisplatin exposure testis. Conclusions: Cisplatin-induced Sertoli cell injury via ferroptosis and promoted ferroptosis in an m6A dependent manner. m6A modification of both SLC7A11 and ALOX12 mRNA could result in ferroptosis in our in vitro model. Further, overexpressed ALOX12 can cause more production of 12-HETE, which may be responsible for testis inflammation caused by cisplatin.

AIM: To describe the distribution of ocular biometrics and to evaluate its associations with refractive error and to assess the contribution from ocular parameters to refractive error among Chinese myopic children. METHODS: This cross-sectional study evaluated subjects aged 8-12y. Keratometry, ocular biometry, and cycloplegic autorefraction were performed on each subject. Spherical equivalent refraction (SER) and ocular biometrics were assessed as a function of age and gender. The Pearson correlation analysis between SER and ocular biometrics was carried out. Multiple linear regression was performed to analyze the association between SER and ocular parameters. RESULTS: A total of 689 out of 735 participants (321 boys, 48.1%) were analyzed, with a mean SER of -2.98±1.47 diopter (D). Axial length (AL), anterior chamber depth (ACD), corneal radius of curvature (CR), horizontal visible iris diameter (HVID), central corneal thickness (CCT) and lens power (LP) showed normal distribution. The AL, AL/CR ratio, ACD and CR increased from 8 to 12y of age, while SER and LP decreased, HVID and CCT remained stable. There was no difference in gender. ObjectiveTo detect and analyze the contamination of hygiene indicator bacteria in bulk ready-to-eat foods in various regions of Jilin province, providing a reference for the safety supervision of the bulk ready-to-eat food industry in Jilin province. MethodsFrom 2011 to 2021, 5 664 ready-to-eat food samples were collected from 9 regions in Jilin province, including Baicheng city, Baishan city, Jilin city, Liaoyuan city, Siping city, Songyuan city, Tonghua city, Yanbian Korean Autonomous Prefecture, and Changchun city. Four types of hygiene indicator bacteria, including total bacterial count, coliform bacteria, Escherichia coli, and mold, were detected and analyzed. ResultsThe overall pass rate of total bacterial count was 82.19% (3563/4335), the overall pass rate of coliform bacteria was 63.34% (1617/2553), the overall pass rate of Escherichia coli was 96.5% (1655/1715), and the overall pass rate of mold was 72.07% (240/333). Among the 9 monitored cities, Tonghua city had the lowest pass rate of total bacterial count at 58.53% (367/627), with a mean value of (4.41 ± 1.51) lgCFU/g; Baishan city had the lowest pass rate of coliform bacteria at 43.48% (100/230), with a mean value of (3.47 ± 1.34) lgCFU/g; Tonghua city had the lowest pass rate of Escherichia coli at 92.54% (310/335), with a mean value of (2.11 ± 0.87) lgCFU/g; and Songyuan city had the lowest pass rate of mold at 54.17% (13/24), with a mean value of (2.13 ± 1.04) lgCFU/g. Among different types of foods, salads had the lowest pass rate of total bacterial count at 0, with a mean value of (5.70 ± 0.71) lgCFU/g; Chinese-style cold dishes had the lowest pass rate of coliform bacteria at 15.13% (23/152), with a mean value of (3.49 ± 1.08) lgCFU/g; salads had the lowest pass rate of Escherichia coli at 94.34% (150/159), with a mean value of (1.88 ± 0.61) lgCFU/g; and cakes had the lowest pass rate of mold at 32.26% (10/31), with a mean value of (3.08 ± 1.98) lgCFU/g. Among different sampling locations, small restaurants had the lowest pass rate of total bacterial count at 74.74% (494/661), with a mean value of (4.14 ± 1.35) lgCFU/g; The qualified rates of Escherichia coli and Escherichia coli in department stores are the lowest, at 28.95% (11/38) and 80.00% (4/5), respectively. The average value of Escherichia coli is (4.06 ± 1.21) lgCFU/g, and the number of positive samples for Escherichia coli is 2.04 lgCFU/g; and convenience stores had the lowest pass rate of mold at 53.33% (24/45), with a mean value of (2.41 ± 1.00) lgCFU/g. ConclusionThe contamination levels of hygiene indicator bacteria in bulk ready-to-eat foods that do not undergo a final heating step, such as raw foods, cold dishes, and filled foods, are relatively high in different regions of Jilin province and should be prioritized for prevention. Small and medium-sized restaurants, fast food restaurants, and department stores have higher contamination rates of hygiene indicator bacteria and require focused supervision and management.

Abstract Background Astaxanthin (ASX) has been documented to exert beneficial influence on various processes in fish. Largemouth bass (Micropterus salmoides) serves as a common model for studying glucose-induced liver disease, making it imperative to investigate the regulatory mechanisms underlying its liver health. Methods Largemouth bass were fed with a control diet (CON), a high carbohydrate diet (HC), or a HC diet supplemented astaxanthin (HCA) for 8-weeks, followed by the glucose tolerance test (GTT). Primary hepatocytes were treated with low glucose and high glucose combined with different concentrations of astaxanthin for 48 h. The histopathology, enzymology, transcriptomics, molecular biology and cell biology were combined to investigate the mechanism of liver injury. Results This study provides evidence for the protective effects of ASX against growth performance reduction and hepatic liver injure in largemouth bass fed HC diet. In GTT, HCA diet exhibited an improvement in glucose tolerance following glucose loading. Although HCA diet did not restore the expression of insulin resistance-related genes in livers at different time during the GTT, the addition of ASX in the long-term HC diet did improve the insulin resistance pathway by regulating the PTP1B/PI3K/Akt signaling pathway. Hepatic transcriptome analyses showed that ASX plays an essential role in the modulation of glucose homeostasis in response to treated with HC diet. In in vitro study, ASX treatment resulted in an exaltation in cell viability and a reduction in the rate of cell apoptosis and reactive oxygen species (ROS). Additionally, astaxanthin was observed to improve apoptosis induced by high-glucose via p38MAPK/bcl-2/caspase-3 signaling pathway. Conclusions Astaxanthin exhibited a protective effect against apoptosis by regulating p38MAPK/bcl-2/caspase-3 pathway, and ameliorated insulin resistance by activating the PTP1B/PI3K/Akt pathway. This study elucidated the mechanism of astaxanthin in the liver injury of largemouth bass from a new perspective and provided a new target for the treatment of insulin resistance. Graphical Abstract

Abstract Traumatic dental injuries (TDIs) of teeth occur frequently in children and adolescents. TDIs that impact the periodontal tissues and alveolar tissue can be classified into concussion, subluxation, extrusive luxation, intrusive luxation, lateral luxation, and avulsion. In these TDIs, management of injured soft tissue, mainly periodontal ligament, and dental pulp, is crucial in maintaining the function and longevity of the injured teeth. Factors that need to be considered for management in laxation injuries include the maturation stage of the traumatic teeth, mobility, direction of displacement, distance of displacement, and whether there are alveolar fractures. In avulsion, the maturation stage of the permanent tooth, the out-socket time, storage media/condition of the avulsed tooth, and management of the PDL should also be considered. Especially, in this review, we have subdivided the immature tooth into the adolescent tooth (Nolla stage 9) and the very young tooth (Nolla stage 8 and below). This consensus paper aimed to discuss the impacts of those factors on the trauma management and prognosis of TDI to provide a streamlined guide for clinicians from clinical evaluation, diagnostic process, management plan decision, follow-up, and orthodontic treatment for tooth luxation and avulsion injuries.

Abstract Background Axillary lymph node dissection (ALND) is the standard axillary management for breast cancer patients with positive sentinel lymph node biopsy (SLNB) after neoadjuvant therapy. Nevertheless, when that happens, the frequency of additional positive nodes is not properly evaluated. We aim to develop a prediction model to assess the frequency of additional nodal disease after a positive sentinel lymph node following neoadjuvant therapy. Methods We retrospectively analyzed the ultrasound and clinicopathological characteristics of breast cancer patients with 1–3 positive sentinel lymph nodes (SLN) undergoing mastectomy after neoadjuvant therapy (NAT) at our institution, and performed univariate and multivariate logistic analyses to confirm the factors affecting non-SLN metastasis. These factors were included to establish a nomogram, and the area under receiver operating characteristic curve (AUC) and decision curve analysis (DCA) were utilized to assess the validity of this model. Results A total of 126 breast cancer patients were ultimately included in our study, 38 (53.5%) patients were diagnosed with non-SLN metastases of all 71 patients in training set. The results of multifactorial logistic analysis suggested that lymph node metastasis ratio (LNR), short axis of lymph node and progesterone receptor (PR) were strongly associated with non-SLN metastasis. We established a nomogram using the above three variables as predictors, which yielded an area under the curve of 0.795, and validated with a favorable AUC of 0.876. Conclusion The nomogram we constructed can accurately predict the likelihood of non-SLN metastasis in our patients with 1–3 positive SLN after NAT, which may help guide decision making regarding axillary management.

Abstract Background Given the established link between obesity and hyperuricemia (HUA), the research want to investigate the relationship between different obesity indices and HUA, and further analyze which obesity index can better predict HUA. Methods The data were obtained from a longitudinal study involving middle-aged and elderly populations in Dalian, China. The research encompassed individuals who exhibited typical uric acid levels initially and tracked their progress over a three-year period. 8 obesity indices were evaluated retrospectively. Subgroup analyses were conducted to identify susceptible populations. Restricted cubic splines (RCS) were utilized to model the dose-response relationships between obesity indices and HUA. Receiver operating characteristic (ROC) curves were applied to visualize and compare the predictive value of both traditional and new obesity indices for HUA. Results Among 4,112 individuals with normal baseline uric acid levels, 950 developed HUA. Significant associations with HUA were observed for body mass index (BMI), waist circumference (WC), body roundness index (BRI), cardiometabolic index (CMI), visceral adiposity index (VAI), Chinese visceral adiposity index (CVAI), lipid accumulation product (LAP), and abdominal volume index (AVI). Subgroup analysis indicated that all obesity indices proved more effective in assessing the onset of HUA in women without Metabolic Syndrome (MetS). Further analysis using RCS revealed non-linear dose-response relationships between LAP, CMI, VAI, and HUA in males, with similar non-linear relationships observed for all indices in females. The results from the ROC curves indicate that LAP may serve as a better predictor of HUA in males, and CVAI may serve as a better predictor in females. Conclusion HUA is closely associated with obesity indices. Among females, CVAI emerges as the preferred predictive index for HUA. In males, LAP emerges as the preferred predictive index for HUA.

Abstract Background Telomeres are a critical component of chromosome integrity and are essential to the development of cancer and cellular senescence. The regulation of breast cancer by telomere-associated lncRNAs is not fully known, though. The goals of this study were to describe predictive telomere-related LncRNAs (TRL) in breast cancer and look into any possible biological roles for these RNAs. Methods We obtained RNA-seq data, pertinent clinical data, and a list of telomere-associated genes from the cancer genome atlas and telomere gene database, respectively. We subjected differentially expressed TRLs to co-expression analysis and univariate Cox analysis to identify a prognostic TRL. Using LASSO regression analysis, we built a prognostic model with 14 TRLs. The accuracy of the model’s prognostic predictions was evaluated through the utilization of Kaplan-Meier (K-M) analysis as well as receiver operating characteristic (ROC) curve analysis. Additionally, immunological infiltration and immune drug prediction were done using this model. Patients with breast cancer were divided into two subgroups using cluster analysis, with the latter analyzed further for variations in response to immunotherapy, immune infiltration, and overall survival, and finally, the expression of 14-LncRNAs was validated by RT-PCR. Results We developed a risk model for the 14-TRL, and we used ROC curves to demonstrate how accurate the model is. The model may be a standalone prognostic predictor for patients with breast cancer, according to COX regression analysis. The immune infiltration and immunotherapy results indicated that the high-risk group had a low level of PD-1 sensitivity and a high number of macrophages infiltrating. The cluster 2 subtype showed the highest immune infiltration, the highest immune checkpoint expression, and the worst prognosis among the two subtypes defined by cluster analysis, which requires more attention and treatment. Conclusion As a possible biomarker, the proposed 14-TRL signature could be utilized to evaluate clinical outcomes and treatment efficacy in breast cancer patients.

Abstract There is increasing awareness of radiotherapy’s potential side effects, such as lymphopenia. Therefore, this study aimed to establish the association between WBRT and the development of lymphopenia in patients with brain metastases undergoing brain radiotherapy (RT), along with evaluating the corresponding clinical outcomes. Including 116 patients with brain metastases undergoing brain radiotherapy, the study collected the absolute lymphocyte counts (ALC) within 2 weeks before brain radiotherapy (pre-radiotherapy, pre-RT), as well as ones at 1 and 2 months after completing RT (post-RT). Univariate and multivariate analyses were performed to identify associations between radiation modality and post-RT ALC. The relationships between post-RT ALC and overall survival were evaluated with Kaplan–Meier analysis and a multivariate Cox regression model. The median ALC definitely decreased at 1 month post-RT, but at 2 months post-RT, gradually rose but not to the pre-RT ALC. The multivariate analysis identified WBRT and lower pre-RT ALC as independent risk factors associated with the decrease in post-RT ALC at 1 month. It also revealed more than 4 brain metastases, G3-4 lymphopenia at 1 month and lower post-RT ALC at 2 months exhibited significantly worse prognosis regardless of the radiation modality. However, there was indeed an independent correlation between radiation modality and the outcome of intracranial progression-free survival (PFS). To approach the feasibility and reasonableness of treatment, clinicians should carefully consider various factors to achieve long-term survival of patients.

Abstract Objective To evaluate the potential utility of 18F-FDG PET/CT to assess response to neoadjuvant immunochemotherapy in patients with resectable NSCLC, and the ability to screen patients who may benefit from neoadjuvant immunochemotherapy. Methods Fifty one resectable NSCLC (stage IA–IIIB) patients were analyzed, who received two-three cycles neoadjuvant immunochemotherapy.18F-FDG PET/CT was carried out at baseline(scan-1) and prior to radical resection(scan-2). SULmax, SULpeak, MTV, TLG, T/N ratio, ΔSULmax%,ΔSULpeak%, ΔMTV%, ΔTLG%,ΔT/N ratio% were calculated. 18F-FDG PET/CT responses were classified using PERCIST. We then compared the RECIST 1.1 and PERCIST criteria for response assessment.With surgical pathology of primary lesions as the gold standard, the correlation between metabolic parameters of 18F-FDG PET/CT and major pathologic response (MPR) was analyzed. All metabolic parameters were compared to treatment response and correlated to PFS and OS. Results In total of fifty one patients, MPR was achieved in 25(49%, 25/51) patients after neoadjuvant therapy. Abstract Genome-wide association studies (GWAS) identified over fifty loci associated with lung cancer risk. However, underlying mechanisms and target genes are largely unknown, as most risk-associated variants might regulate gene expression in a context-specific manner. Here, we generate a barcode-shared transcriptome and chromatin accessibility map of 117,911 human lung cells from age/sex-matched ever- and never-smokers to profile context-specific gene regulation. Identified candidate cis-regulatory elements (cCREs) are largely cell type-specific, with 37% detected in one cell type. Colocalization of lung cancer candidate causal variants (CCVs) with these cCREs combined with transcription factor footprinting prioritize the variants for 68% of the GWAS loci. CCV-colocalization and trait relevance score indicate that epithelial and immune cell categories, including rare cell types, contribute to lung cancer susceptibility the most. A multi-level cCRE-gene linking system identifies candidate susceptibility genes from 57% of the loci, where most loci display cell-category-specific target genes, suggesting context-specific susceptibility gene function.

Abstract Purpose The purpose of this study is to retrospect and summarize clinical efficiency and experience of the free superficial palmar branch of radial artery (SPBRA) flap for soft-tissue reconstruction in distal digital injury. Method 13 patients with soft-tissue defect of finger, reconstructed by the free superficial palmar branch of radial artery (SPBRA) flap in our department from January 2020 to January 2022, were reviewed. After 6–12 months of follow-up, evaluated the treatment effect of the fingers reconstructed by SPBRA flap. Results All the flaps in our series application were survival uneventful, and all the donor sites were closed primarily without complications or obvious scarring. The flaps were soft in texture and satisfactory in appearance and function. The flaps with the median nerve palmar cutaneous branch had a good sensation recovery. Measurement of two-point discrimination (TPD) ranged from 6 to 10 mm. All patients were satisfied with the aesthetic appearance. According to the Evaluation Trial Standards of Upper Limb Partial Function of Hand Surgery of Chinese Medical Association, the results were graded as excellent in 11 cases and good in 2 cases. Conclusion The SPBRA perforator flap has the advantages of simple operation, soft texture, good appearance and function, and is credible and useful for reconstructing various finger injuries.

The effectiveness of AI-driven drug discovery can be enhanced by pretraining on small molecules. However, the conventional masked language model pretraining techniques are not suitable for molecule pretraining due to the limited vocabulary size and the non-sequential structure of molecules. To overcome these challenges, we propose FragAdd, a strategy that involves adding a chemically implausible molecular fragment to the input molecule. This approach allows for the incorporation of rich local information and the generation of a high-quality graph representation, which is advantageous for tasks like virtual screening. Consequently, we have developed a virtual screening protocol that focuses on identifying estrogen receptor alpha binders on a nucleus receptor. Our results demonstrate a significant improvement in the binding capacity of the retrieved molecules. Additionally, we demonstrate that the FragAdd strategy can be combined with other self-supervised methods to further expedite the drug discovery process.

Traditional treatments against advanced non-small cell lung cancer (NSCLC) with high morbidity and mortality continue to be dissatisfactory. Given this situation, there is an urgent requirement for alternative modalities that provide lower invasiveness, superior clinical effectiveness, and minimal adverse effects. The combination of photodynamic therapy (PDT) and immunotherapy gradually become a promising approach for high-grade malignant NSCLC. Nevertheless, owing to the absence of precise drug delivery techniques as well as the hypoxic and immunosuppressive characteristics of the tumor microenvironment (TME), the efficacy of this combination therapy approach is less than ideal. In this study, we construct a novel nanoplatform that indocyanine green (ICG), a photosensitizer, loads into hollow manganese dioxide (MnO2) nanospheres (NPs) (ICG@MnO2), and then encapsulated in PD-L1 monoclonal antibodies (anti-PD-L1) reprogrammed exosomes (named ICG@MnO2@Exo-anti-PD-L1), to effectively modulate the TME to oppose NSCLC by the synergy of PDT and immunotherapy modalities. The ICG@MnO2@Exo-anti-PD-L1 NPs are precisely delivered to the tumor sites by targeting specially PD-L1 highly expressed cancer cells to controllably release anti-PD-L1 in the acidic TME, thereby activating T cell response. Subsequently, upon endocytic uptake by cancer cells, MnO2 catalyzes the conversion of H2O2 to O2, thereby alleviating tumor hypoxia. Meanwhile, ICG further utilizes O2 to produce singlet oxygen (1O2) to kill tumor cells under 808 nm near-infrared (NIR) irradiation. Furthermore, a high level of intratumoral H2O2 reduces MnO2 to Mn2+, which remodels the immune microenvironment by polarizing macrophages from M2 to M1, further driving T cells. Taken together, the current study suggests that the ICG@MnO2@Exo-anti-PD-L1 NPs could act as a novel drug delivery platform for achieving multimodal therapy in treating NSCLC.

Self-directed learning with mobile technology (SDLMT) is critical to students' learning success. However, only minimal research has been conducted on the manner by which significant aspects (e.g., selfefficacy, student engagement) are related to SDLMT. This study analyzed the answers of 485 Chinese students (seventh to ninth grades) who were surveyed, and evaluated the relationships among self-efficacy (Internet selfefficacy and online communication self-efficacy), student engagement (behavioral, cognitive, emotional, and social engagement), and SDLMT. Structural equation model revealed that SDLMT was positively predicted by self-efficacy, and SDLMT was positively impacted by student engagement. Moreover, the mediation analysis was performed using the PROCESS plugin in SPSS and determined that after controlling for gender and grade, the relationship between self-efficacy and SDLMT was partially mediated by student engagement. Findings established the value of self-efficacy for SDLMT, and further emphasized the vital mediating role of student engagement. Hence, developing K-12 students' SDLMT entails enhancing self-efficacy and student engagement.

Participants were 1535 grades 3, 4, and 5 students from three elementary schools in Xi'an, China. Children with and without RD were compared to their depression and anxiety. Additionally, children's self-ratings, parents' ratings, and teachers' ratings of depression and anxiety were compared to better understand mental health issues of Chinese children with RD. The findings showed that Chinese children with RD experienced more depression but not anxiety compared to their typically developing peers across all three grade levels. Reports from all three informants consistently reflected that children with RD experienced more depression. However, some inconsistencies were found between self-reported levels of depression and anxiety and the observations made by parents and teachers. Children reported experiencing more depression than their parents and teachers observed but less anxiety than their parents reported. This study emphasizes the importance of understanding the links between RD and mental health and the need for appropriate intervention programs to help children with RD cope with the mental health challenges.

Writing has been recognized as a core competency for postgraduate students. However, improving writing performance, particularly for first-year postgraduate students, remains an important and challenging task. This study aimed to explore the effects of student engagement in peer feedback on writing performance from cognitive, affective and behavioral perspectives. Specifically, the participants were 88 postgraduate students. With the support of an online writing and peer review system (Peerceptiv), participants were asked to write an essay, engage in the peer feedback process, and revise their essays. Correlation analysis and multiple regression analysis were utilized to analyze data on the quality of participants' essays, peer feedback procedures, and interviews. Results showed that cognitive engagement, affective engagement, and behavioral engagement were significantly associated with postgraduate students' writing performance. The analysis indicated that the helpfulness of provided comments was the strongest predictor of writing performance, followed by suggestion, integration, and positive affective engagement. These findings highlight the significance of student engagement in peer feedback to improve writing performance in higher education. Recommendations for future research on quality improvement in writing performance are suggested based on the findings.

This study evaluated the effect of black soldier fly (Hermetia illucens) larvae oil (BO) produced by a novel technique, subcritical butane extraction, on the flesh quality, lipid nutrients and muscle growth of rainbow trout (Oncorhynchus mykiss) fillet, and investigated the alleviating mechanisms of dietary astaxanthin (AST) supplementation. Two hundred and forty fish (215.16 ± 2.30 g) were distributed to three groups with four replicates. Fish were fed three experimental diets for 8 weeks: the control diet (CD diet), total fish oil of the CD diet was replaced with BO to formulate the BO100 diet, and then 1 g/kg AST was supplemented with the BO100 diet to formulate the AST diet. Long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) are closely related to the treatment of human diseases. Traditional biological experiments often require time-consuming and labor-intensive in their search for mechanisms of disease. Computational methods are regarded as an effective way to predict unknown lncRNA-miRNA interactions (LMIs). However, most of them complete their tasks by mainly focusing on a single lncRNA-miRNA network without considering the complex mechanism between biomolecular in life activities, which are believed to be useful for improving the accuracy of LMI prediction. To address this, a heterogeneous information network (HIN) learning model with neighborhood-level structural representation, called HINLMI, to precisely identify LMIs. In particular, HINLMI first constructs a HIN by integrating nine interactions of five biomolecules. After that, different representation learning strategies are applied to learn the biological and network representations of lncRNAs and miRNAs in the HIN from different perspectives. Finally, HINLMI incorporates the XGBoost classifier to predict unknown LMIs using final embeddings of lncRNAs and miRNAs. Experimental results show that HINLMI yields a best performance on the real dataset when compared with state-of-the-art computational models. Moreover, several analysis experiments indicate that the simultaneous consideration of biological knowledge and network topology of lncRNAs and miRNAs allows HINLMI to accurately predict LMIs from a more comprehensive perspective. The promising performance of HINLMI also reveals that the utilization of rich heterogeneous information can provide an alternative insight for HINLMI to identify novel interactions between lncRNAs and miRNAs.

The anti-Clostridium difficile infection (CDI) drug fidaxomicin is a natural polyketide metabolite mainly produced by Micromonosporaceae, such as Actinoplanes deccanensis, Dactylosporangium aurantiacum, and Micromonospora echinospora. In the present study, we employed a stepwise strategy by combining heterologous expression, chassis construction, promoter engineering, activator and transporters overexpression, and optimization of fermentation media for high-level production of fidaxomicin. The maximum yield of 384 mg/L fidaxomicin was achieved with engineered Streptomyces albus D7-VHb in 5 L-tank bioreactor, and it was approximately 15-fold higher than the native strain Actinoplanes deccanensis YP-1 with higher strain stability and growth rate. This study developed an enhanced chassis strain, and for the first time, achieved the heterologous synthesis of fidaxomicin through a combinatorial metabolic engineering strategy.
