Abstract Liver metastasis remains the primary cause of mortality in patients with colon cancer. Identifying specific driver gene mutations that contribute to metastasis may offer viable therapeutic targets. To explore clonal evolution and genetic heterogeneity within the metastasis, we conducted single-cell exome sequencing on 150 single cells isolated from the primary tumor, liver metastasis, and lymphatic metastasis from a stage IV colon cancer patient. The genetic landscape of the tumor samples revealed that both lymphatic and liver metastases originated from the same region of the primary tumor. Notably, the liver metastasis was derived directly from the primary tumor, bypassing the lymph nodes. Comparative analysis of the sequencing data for individual cell pairs within different tumors demonstrated that the genetic heterogeneity of both liver and lymphatic metastases was also greater than that of the primary tumor. This finding indicates that liver and lymphatic metastases arose from clusters of circulating tumor cell (CTC) of a polyclonal origin, rather than from a single cell from the primary tumor. Single-cell transcriptome analysis suggested that higher EMT score and CNV scores were associated with more polyclonal metastasis. Additionally, a mutation in the TRPS1 (Transcriptional repressor GATA binding 1) gene, TRPS1 R544Q, was enriched in the single cells from the liver metastasis. The mutation significantly increased CRC invasion and migration both in vitro and in vivo through the TRPS1R544Q/ZEB1 axis. Further TRPS1 mutations were detected in additional colon cancer cases, correlating with advanced-stage disease and inferior prognosis. These results reveal polyclonal seeding and TRPS1 mutation as potential mechanisms driving the development of liver metastases in colon cancer.

The natural bone structure exhibits a radial gradient with dense cortical bone externally and porous cancellous bone internally. However, previous studies proposing scaffold designs have predominantly focused on homogeneous porous structures. Moreover, no consensus exists on the optimal structure for gradient porous scaffolds. Our scaffold closely imitated the natural bone structure by incorporating a gradient of pillar diameters. Additionally, we introduced a inverse gradient structure and three uniform diameter pillar structures (400 μm, 600 μm, and 1000 μm) for comparative analysis. Mechanical testing revealed that the compressive strength and elastic modulus of the Ti6Al4V porous scaffolds gradually increased with an increase in pillar diameter. In vitro experiments demonstrated that both the biomimetic gradient and inverse gradient scaffolds promoted osteogenic differentiation, with higher ALP activity (alkaline phosphatase) and osteogenesis-related gene expression compared to the uniform pillar structure scaffolds. The in vivo experiments confirmed these results, highlighting the superior ability of the biomimetic gradient Ti6Al4V porous scaffold to induce new bone formation. Based on our findings, we suggest that the optimal porous structure should have fine-diameter pillars (approximately 400 μm) internally to enhance cell penetration, while coarse-diameter pillars (approximately 800 μm) should be used externally to increase the cell attachment area and enhance mechanical strength. Overall, our study contributes to the field of bone tissue engineering by providing a biomimetic scaffold design that closely imitates the structure of natural bone and promotes osteogenic activity.

Abstract Cellular senescence (CS) is closely related to tumor progression. However, the studies about CS genes across human cancers have not explored the relationship between cancer senescence signature and telomere length. Additionally, single-cell analyses have not revealed the evolutionary trends of malignant cells and immune cells at the CS level. We defined a CS-associated signature, called “senescence signature”, and found that patients with higher senescence signature had worse prognosis. Higher senescence signature was related to older age, higher genomic instability, longer telomeres, increased lymphocytic infiltration, higher pro-tumor immune infiltrates (Treg cells and MDSCs), and could predict responses to immune checkpoint inhibitor therapy. Single-cell analysis further reveals malignant cells and immune cells share a consistent evolutionary trend at the CS level. MAPK signaling pathway and apoptotic processes may play a key role in CS, and senescence signature may effectively predict sensitivity of MEK1/2 inhibitors, ERK1/2 inhibitors and BCL-2 family inhibitors. Abstract Primary testicular lymphoma (PTL) is a rare lymphoma predominantly occurring in the elderly male population. It is characterized by a limited response to treatment and a heightened tendency towards relapse. Histologically, approximately 90% of PTL cases are classified as diffuse large B-cell lymphomas (DLBCL). Genetic features of PTL were delineated in a limited scope within several independent studies. Some of the articles which analyzed the genetic characterization of DLBCL have incorporated PTL samples, but these have been constrained by small sample sizes. In addition, there have been an absence of independent molecular typing studies of PTL. This report summarizes the common mutational features, copy number variations (CNVs) and molecular typing of PTL patients, based on whole-exome sequencing (WES) conducted on a cohort of 25 PTL patients. Among them, HLA, CDKN2A and MYD88 had a high mutation frequency. In addition, we found two core mutational characteristics in PTL including mutation in genes linked to genomic instability (TP53 and CDKN2A) and mutation in immune-related genes (HLA, MYD88, CD79B). We performed molecular typing of 25 PTL patients into C1 subtype with predominantly TP53 mutations and C2 subtype with predominantly HLA mutations. Notably, mutations in the TP53 gene predicted a poor outcome in most types of lymphomas. However, the C1 subtype, dominated by TP53 mutations, had a better prognosis compared to the C2 subtype in PTL. C2 subtype exhibited a worse prognosis, aligning with our finding that the mechanism of immune escape in PTL was primarily the deletions of HLA rather than PD-L1/PD-L2 alterations, a contrast to other DLBCLs. Moreover, we calculated the tumor mutation burden (TMB) and identified that TMB can predict prognosis and recurrence rate in PTL. Our study underscores the significance of molecular typing in PTL based on mutational characteristics, which plays a crucial role in prognostication and guiding therapeutic strategies for patients.

The unscientific application of nitrogen (N) fertilizer not only increases the economic input of pear growers but also leads to environmental pollution. Improving plant N use efficiency (NUE) is the most effective economical method to solve the above problems. The absorption and utilization of N by plants is a complicated process. Glutamine synthetase (GS) and glutamate synthase (GOGAT) are crucial for synthesizing glutamate from ammonium in plants. However, their gene family in pears has not been documented. This study identified 29 genes belonging to the GS and GOGAT family in the genomes of Pyrus betulaefolia (P.be, 10 genes), Pyrus pyrifolia (P.py, 9 genes), and Pyrus bretschneideri (P.br, 10 genes). These genes were classified into two GS subgroups (GS1 and GS2) and two GOGAT subgroups (Fd–GOGAT and NADH–GOGAT). The similar exon–intron structures and conserved motifs within each cluster suggest the evolutionary conservation of these genes. Meanwhile, segmental duplication has driven the expansion and evolution of the GS and GOGAT gene families in pear. The tissue–specific expression dynamics of PbeGS and PbeGOGAT genes suggest significant roles in pear growth and development. Cis–acting elements of the GS and GOGAT gene promoters are crucial for plant development, hormonal responses, and stress reactions. Furthermore, qRT–PCR analysis indicated that PbeGSs and PbeGOGATs showed differential expression under exogenous hormones (GA3, IAA, SA, ABA) and abiotic stress (NO3− and salt stress). In which, the expression of PbeGS2.2 was up–regulated under hormone treatment and down–regulated under salt stress. Furthermore, physiological experiments demonstrated that GA3 and IAA promoted GS, Fd–GOGAT, and NADH–GOGAT enzyme activities, as well as the N content. Correlation analysis revealed a significant positive relationship between PbeGS1.1, PbeGS2.2, PbeNADH–GOGATs, and the N content. Therefore, PbeGS1.1, PbeGS2.2, and PbeNADH–GOGATs could be key candidate genes for improving NUE under plant hormone and abiotic stress response. To the best of our knowledge, our study provides valuable biological information about the GS and GOGAT family in the pear for the first time and establishes a foundation for molecular breeding aimed at developing high NUE pear rootstocks.

Background Data on viral kinetics and variants affecting the duration of viral shedding were limited. Our objective was to determine viral shedding in distinct severe acute respiratory syndrome coronavirus 2 variants, including Omicron BA.4/5 and BF.7, and to identify the relevant influencing factors. Methods We carried out a longitudinal cohort study at Beijing Xiaotangshan Fangcang shelter hospital from May to June 2022 (Omicron BA.4/5) and from November to December 2022 (Omicron BF.7). Nucleocapsid protein (N) and open reading frame (ORF) genes were considered as the target genes of the reverse transcription PCR. The daily results of cycle threshold (CT), including lowest ORF1ab-CT values for days 1–3 post-hospitalisation and lowest N-CT values for days 1–3 post-hospitalisation (CT3minN) and demographic and clinical characteristics were collected. Results 1433 patients with coronavirus disease 2019 (COVID-19) were recruited from the Fangcang shelter hospital, in which 278 patients were diagnosed with Omicron BA.4/5 and 1155 patients with Omicron BF.7. Patients with BF.7 infection showed a longer duration of viral shedding. The duration of viral shedding was associated with the variants age, alcohol use, the severity of COVID-19 and CT3minN. Moreover, the nomogram had excellent accuracy in predicting viral shedding. Conclusions Our results indicated that patients with Omicron BF.7 had a longer period of contagiousness than those with BA.4/5. The duration of viral shedding was affected by a variety of factors and the nomogram may become an applicable clinical instrument to predict viral shedding. Furthermore, we developed a new COVID-19 viral shedding predicting model that can accurately predict the duration of viral shedding for COVID-19, and created a user-friendly website to apply this prediction model (https://puh3.shinyapps.io/CVSP_Model/).

Currently, there are still many patients who require outpatient triage assistance. ChatGPT, a natural language processing tool powered by artificial intelligence technology, is increasingly utilized in medicine. To facilitate and expedite patients’ navigation to the appropriate department, we conducted an outpatient triage evaluation of ChatGPT. For this evaluation, we posed 30 highly representative and common outpatient questions to ChatGPT and scored its responses using a panel of five experienced doctors. The consistency of manual triage and ChatGPT triage was assessed by five experienced doctors, and statistical analysis was performed using the Chi-square test. The expert ratings of ChatGPT’s answers to these 30 frequently asked questions revealed 17 responses earning very high scores (10 and 9.5 points), 7 earning high scores (9 points), and 6 receiving low scores (8 and 7 points). Additionally, we conducted a prospective cohort study in which 45 patients completed forms detailing gender, age, and symptoms. Triage was then performed by outpatient triage staff and ChatGPT. BackgroundLianhuaqingwen (LHQW), a traditional Chinese medicine comprised of 13 herbal extracts renowned for their robust heat-clearing and detoxifying properties, has gained widespread utilization in China but has yet to garner similar recognition abroad. It is believed to exhibit efficacy in ameliorating symptoms in individuals afflicted with coronavirus disease 2019 (COVID-19). However, the precise impact of LHQW on viral shedding (VS), particularly in the context of mild or asymptomatic infections caused by the Omicron BF.4/5 or BF.7 variants of COVID-19, remained inadequately elucidated. Consequently, a real-world study was conducted, involving patients diagnosed with COVID-19, with the primary objective of ascertaining the effectiveness of LHQW in this specific clinical context.MethodsWe conducted an investigation on Omicron-infected patients through a single-center, propensity score-matched real-world study conducted at Xiaotangshan Fangcang Hospital from May to November 2022. A total of 3,368 COVID-19 patients were enrolled in the study, all of whom presented mild or asymptomatic infections caused by either BF.4/5 or BF.7 strains of the virus. Demographic and clinical data were systematically collected from medical records. Patients were allocated to receive treatment with LHQW (designated as the treatment group) or received no LHQW treatment (designated as the not-treated/no-treatment group). Viral load was quantified utilizing quantitative real-time PCR (qPCR), and the duration of VS was defined as the time interval between the initial negative test result and the date of COVID-19 diagnosis or symptom onset.ResultsThe study encompassed a cohort of 3,368 patients, and following propensity score matching, a subset of 296 patients was meticulously chosen for subsequent analysis. Notably, baseline characteristics exhibited disparities between the treatment and not-treated/no-treatment groups. However, post-matching, these characteristics achieved a commendable level of comparability. Our findings unequivocally demonstrated that there existed no statistically significant disparity in VS. This holds true when comparing patients subjected to LHQW treatment against those not administered LHQW, as well as when contrasting individuals presenting asymptomatic and mild COVID-19 manifestations.ConclusionNo statistically significant difference in VS was observed between patients who underwent LHQW treatment and those who did not. Additional investigations are imperative to provide a comprehensive assessment of LHQW’s efficacy, particularly in patients afflicted with severe COVID-19 or those infected with viral strains distinct from BF.4/5 or BF.7.

ABSTRACTObjective: Diffuse Large B-Cell Lymphoma (DLBCL) is a common and frequently occurring subtype of Non-Hodgkin Lymphoma (NHL). The effective treatment and prognosis of DLBCL are still urgently needed to be explored. This article aims to shed light on the connection between DLBCL survival and NR3C1 expression levels. Methods: First, we divided the 952 DLBCL patients into an NR3C1 high-expression group and an NR3C1 low-expression group and compared the baseline characteristics of the two groups. Second, we used multivariate analysis to predict the dependent variable for age, pathology, ECOG score, lactate dehydrogenase (LDH) ratio, and NR3C1 expression level. Abstract Background Cell cycle dysregulation characterized by cyclin D1 overexpression is common in mantle cell lymphoma (MCL), while mitotic disorder was less studied. Cell division cycle 20 homologue (CDC20), an essential mitotic regulator, was highly expressed in various tumors. Another common abnormality in MCL is p53 inactivation. Little was known about the role of CDC20 in MCL tumorigenesis and the regulatory relationship between p53 and CDC20 in MCL. Methods CDC20 expression was detected in MCL patients and MCL cell lines harboring mutant p53 (Jeko and Mino cells) and wild-type p53 (Z138 and JVM2 cells). Z138 and JVM2 cells were treated with CDC20 inhibitor apcin, p53 agonist nutlin-3a, or in combination, and then cell proliferation, cell apoptosis, cell cycle, cell migration and invasion were determined by CCK-8, flow cytometry and Transwell assays. The regulatory mechanism between p53 and CDC20 was revealed by dual-luciferase reporter gene assay and CUT&Tag technology. The anti-tumor effect, safety and tolerability of nutlin-3a and apcin were investigated in vivo in the Z138-driven xenograft tumor model. Results CDC20 was overexpressed in MCL patients and cell lines compared with their respective controls. The typical immunohistochemical marker of MCL patients, cyclin D1, was positively correlated with CDC20 expression. CDC20 high expression indicated unfavorable clinicopathological features and poor prognosis in MCL patients. In Z138 and JVM2 cells, either apcin or nutlin-3a treatment could inhibit cell proliferation, migration and invasion, and induce cell apoptosis and cell cycle arrest. GEO analysis, RT-qPCR and WB results showed that p53 expression was negatively correlated with CDC20 expression in MCL patients, Z138 and JVM2 cells, while this relationship was not observed in p53-mutant cells. Dual-luciferase reporter gene assay and CUT&Tag assay revealed mechanistically that CDC20 was transcriptionally repressed by p53 through directly binding p53 to CDC20 promoter from − 492 to + 101 bp. Moreover, combined treatment of nutlin-3a and apcin showed better anti-tumor effect than single treatment in Z138 and JVM2 cells. Administration of nutlin-3a/apcin alone or in combination confirmed their efficacy and safety in tumor-bearing mice. Conclusions Our study validates the essential role of p53 and CDC20 in MCL tumorigenesis, and provides a new insight for MCL therapeutics through dual-targeting p53 and CDC20.

One important parameter for evaluating the safety and reliability of a ship is o the dynamic ultimate load capacity of ship structures. Because of the importance of this parameter, its determination is essential. In this paper, a novel “two-step” approach for determining the dynamic ultimate load capacity of ship structures is proposed. The main idea of two-step approach is to determine the dynamic ultimate load capacity based on the static ultimate load capacity after accounting for impacts that cause strain on the ship structures. This approach is based on nonlinear finite element method. Here, taking stiffened plate as a case study, the practical application of thus two-step approach is discussed in detail. The results of this approach reveal that the static ultimate load capacity decreases by less than 3% after a stiffened plate is subjected to an impact load whose amplitude corresponds to the dynamic ultimate load capacity. Then, the influence of the impact duration on the failure mode and the effect of the impact load cycles and the impact load sequence on the dynamic ultimate load capacity of the stiffened plate were investigated. Finally, the applicability of the two-step approach to a hull girder is demonstrated. The two-step approach and the conclusions presented in this paper can provide guidance for the evaluation of dynamic ultimate load capacity.

The pattern of layered rock crack propagation in mining and geotechnical engineering directly influences the production efficient of coal mines and the safety of workers. To reveal the characteristics of crack propagation in layered rock, the three-point bending experiment and numerical investigation were carried out to study the crack propagation process of single sandstone samples, sandstone-sandstone-sandstone combined samples, and sandstone-mudstone-sandstone combined samples. Using digital speckle correlation methods and acoustic emission techniques, the crack propagation process of the specimens was examined. The results show that the main factors affecting crack propagation include rock layer strength, bedding plane strength, and the position of pre-existing fractures. The presence of the bedding plane hinders the propagation of cracks, and when the bedding plane’s strength decreases, cracks that originated from the bedding plane spread horizontally along it. The weakening of the intermediate rock layer strength of the combined samples can lead to the initiation of microcracks within it earlier than macroscopic cracks, allowing many options for the initiation and propagation of following cracks. Meanwhile, the load-time curves of the combined samples during the experiment displays a double-peak characteristic. Furthermore, the presence of pre-existing fractures inside the layered rock might result in the unpredictability of the crack starting point and subsequently influence the crack propagation course.

Because of the problem of gangue discharge and surface subsidence during coal mining, the current research on underground filling mainly focuses on the paste filling, solid filling, and grouting filling of the overburden separation layer after scaffolding. We proposed the technology of fluidization gangue grouting for filling the collapse area based on our previous research. The prediction method of residual space in the collapse area and the diffusion law of gangue fluidization filling slurry are two essential points for successfully implementing the technology and maximizing the use of goaf for gangue backfilling and reducing overburden settlement. To further explore the remaining space distribution law of the collapsed goaf in thick seam fully mechanized top coal caving mining and the effect of coal gangue fluidization filling, the caving goaf of the 3307 fully mechanized top coal caving face in Sangshuping No. 2 coal mine in the Weibei mining area was detected by the transient electromagnetic method. We studied the distribution law of the measured abnormal area in the caving goaf, which reflects the distribution law of the remaining space from one aspect. The coefficient of the remaining space was calculated to be 19.5%. Then, we applied COMSOL simulation software. The diffusion law of coal gangue fluidized slurry in the caving goaf was simulated and analyzed. It shows that the most obvious diffusion direction of the coal gangue slurry is the trend of the gradual expansion of the “cavity pore” multi-type residual space, indicating that with the increase in the diffusion distance, the diffusion resistance gradually increases, and the slurry morphology gradually presents the “claw” form. According to the space theory and fractal dimension theory, the prediction method of the remaining space in the caving goaf is given, and the design basis of the filling drilling parameters is determined. Finally, the field-filling test was carried out. The results show that the high- and low-level fluidized filling in the caving goaf can safely and efficiently handle the gangue in the mine, and the residual space characteristics and slurry diffusion law in the caving goaf are consistent with the above. The research results provide theoretical support for the fluidization filling technology of coal gangue in thick seam fully mechanized top coal caving areas.

Abstract Background Acute myeloid leukemia (AML) is a heterogeneous disease in terms of genetic basis, clinical, biological and prognostic, and is a malignant clonal disease of leukemia stem cells (LSCs). Nearly half of adult AML patients exhibit a cytogenetic normal acute myeloid leukemia (CN-AML). The expression level of NCALD gene was associated with the prognosis of ovarian cancer and non-small cell lung cancer (NSCLC). The expression level of NCALD gene is still unclear in the prognosis of patients with AML. Method We integrated 5 independent datasets totally 665 AML patients (497 CN-AML patients) to analyzed relation between NCALD gene expression and the clinical FAB classification, gene mutation, therapy, prognosis of CN-AML. Thallium (Tl) is a typical toxic metal, which poses a great threat to human health through drinking water and the food chain (biomagnification). China has rich Tl-bearing mineral resources, which have been extensively explored and utilized, leading to release of large amounts of Tl into the environment. However, research on Tl pollution and removal techniques is relatively limited, because Tl has not been listed within the scope of environmental monitoring in China for several decades. This paper reviewed Tl pollution in wastewater arising from various industries in China, as well as the latest available methods for treating Tl-containing industrial wastewater, in order to give an outlook on effective technologies for controlling Tl pollution. Conventional physical and chemical treatment technologies are efficient at removing trace amounts of Tl, but it proved to be difficult to achieve the stringent environmental standard (≤0.1–5 μg/L) cost-effectively. Adsorption by using newly developed nanomaterials, and metal oxide modified polymer materials and microbial fuel cells are highly promising and expected to become next-generation technologies for remediation of Tl pollution. With the potential for greater Tl contamination in the environment under accelerated growth of industrialization, researches based on lab-scale implementation of such promising treatment technologies should be further expanded to pilot and industrial scale, ensuring environmental protection and the safety of drinking water for sustainable development. Comprehensive insights into experiences of Tl pollution in China and in-depth perspectives on new frontier technologies of Tl removal from wastewaters will also benefit other nations/regions worldwide, which are susceptible to high exposure to Tl likewise. In subtropical Hong Kong, clear glass is normally used as the translucent material for external windows of residential buildings, but it has poor heat insulation performance especially the tremendous solar heat gain in summer. To improve the thermal performance of conventional clear glass, a new heat blocking coating was developed. The coating can effectively absorb the near infrared radiation, so it can reduce the solar heat gain and therefore the energy use from air-conditioning systems. In this paper, the preparation of the heat blocking coating was firstly introduced. Then the optical and thermal properties of different types of window glazing were tested in the laboratory. Based on the experimental data, a combined daylight, thermal environment and building energy numerical model of a public rental housing was established to fully simulate the performance of different types of windows glazing. The results indicated that the heat blocking coating can reduce about 275.4 kWh (14.4%) electricity use per year compared to conventional clear window.

Abstract Our study presents the genetic landscape betel quid chewing‐associated tongue carcinomas (BQ‐TCs). We compared the genetic landscape and mutational signatures of 15 BQ‐TCs, five nonbetel quid chewing‐associated tongue carcinomas (nBQ‐TCs), and 82 tongue carcinomas in general population from the TCGA (TCGA‐TCs) project. The highlights of this research mainly include: (a) The genetic landscape of BQ‐TC was characterized with frequent mutations in RASA1 gene and in CpG islands throughout the genome. (b) The BQ‐TC had a distinct mutational signature from that of nBQ‐TC and tongue carcinomas in the general population, and this signature was associated with the mutations in RASA1 and in CpG islands. (c) Our study indicates that betel quid (BQ) chewing classifies a distinct group of tongue carcinoma. The BQ chewing might not contribute to the tumorigenesis of tongue carcinomas as a mutagen.

Abstract Background Multiple myeloma (MM) is the plasma cell tumor, which is characterized by clonal proliferation of tumor cells, with high risk of progression to renal impairment, bone damage and amyloidosis. Although the survival rate of patients with MM has improved in the past decade, most people inevitably relapse. The treatment and prognosis of MM are still urgent problems. Breast Cancer Antiestrogen Resistance 3 (BCAR3) is a protein-coding gene that is associated with many tumors. However, there have been few studies on the relationship of BCAR3 and MM. Methods We analyzed 1878 MM patients (1930 samples) from 7 independent datasets. First, we compared the BCAR3 expression level of MM patients in different stages and MM patients with different amplification of 1q21. Second, we analyzed BCAR3 expression levels in MM patients with different molecular subtypes. Finally, we explored the event-free survival rate (EFS) and overall survival rate (OS) of MM patients with high or low BCAR3 expression, including patients before and after relapse, and their therapeutic responses to bortezomib and dexamethasone. Results The expression of BCAR3 showed a decreasing trend in stages I, II and III (P = 0.00068). With the increase of 1q21 amplification level, the expression of BCAR3 decreased (P = 0.022). Abstract Background N6-methyladenosine (m6A) is an important epitranscriptomic mark with high abundance in the brain. Recently, it has been found to be involved in the regulation of memory formation and mammalian cortical neurogenesis. However, while it is now established that m6A methylation occurs in a spatially restricted manner, its functions in specific brain regions still await elucidation. Results We identify widespread and dynamic RNA m6A methylation in the developing mouse cerebellum and further uncover distinct features of continuous and temporal-specific m6A methylation across the four postnatal developmental processes. Temporal-specific m6A peaks from P7 to P60 exhibit remarkable changes in their distribution patterns along the mRNA transcripts. We also show spatiotemporal-specific expression of m6A writers METTL3, METTL14, and WTAP and erasers ALKBH5 and FTO in the mouse cerebellum. Ectopic expression of METTL3 mediated by lentivirus infection leads to disorganized structure of both Purkinje and glial cells. In addition, under hypobaric hypoxia exposure, Alkbh5-deletion causes abnormal cell proliferation and differentiation in the cerebellum through disturbing the balance of RNA m6A methylation in different cell fate determination genes. Notably, nuclear export of the hypermethylated RNAs is enhanced in the cerebellum of Alkbh5-deficient mice exposed to hypobaric hypoxia. Conclusions Together, our findings provide strong evidence that RNA m6A methylation is controlled in a precise spatiotemporal manner and participates in the regulation of postnatal development of the mouse cerebellum.

Rutile nano-titanium dioxide (RNTD) characterized by loose particles with diameter in 20–50 nm has a very large surface area for adsorption of Tl, a typical trace metal that has severe toxicity. The increasing application of RNTD and widespread discharge of Tl-bearing effluents from various industrial activities would increase the risk of their co-exposure in aquatic environments. The adsorption behavior of Tl(I) (a prevalent form of Tl in nature) on RNTD was studied as a function of solution pH, temperature, and ion strength. Adsorption isotherms, kinetics, and thermodynamics for Tl(I) were also investigated. The adsorption of Tl(I) on RNTD started at very low pH values and increased abruptly, then maintained at high level with increasing pH >9. Uptake of Tl(I) was very fast on RNTD in the first 15 min then slowed down. The adsorption of Tl(I) on RNTD was an exothermic process; and the adsorption isotherm of Tl(I) followed the Langmuir model, with the maximum adsorption amount of 51.2 mg/g at room temperature. The kinetics of Tl adsorption can be described by a pseudo-second-order equation. FT-IR spectroscopy revealed that -OH and -TiOO-H play an important role in the adsorption. All these results indicate that RNTD has a fast adsorption rate and excellent adsorption amount for Tl(I), which can thus alter the transport, bioavailability and fate of Tl(I) in aqueous environment.

Indoor visual positioning is a key technology in a variety of indoor location services and applications. The particular spatial structures and environments of indoor spaces is a challenging scene for visual positioning. To address the existing problems of low positioning accuracy and low robustness, this paper proposes a precision single-image-based indoor visual positioning method for a smartphone. The proposed method includes three procedures: First, color sequence images of the indoor environment are collected in an experimental room, from which an indoor precise-positioning-feature database is produced, using a classic speed-up robust features (SURF) point matching strategy and the multi-image spatial forward intersection. Then, the relationships between the smartphone positioning image SURF feature points and object 3D points are obtained by an efficient similarity feature description retrieval method, in which a more reliable and correct matching point pair set is obtained, using a novel matching error elimination technology based on Hough transform voting. Finally, efficient perspective-n-point (EPnP) and bundle adjustment (BA) methods are used to calculate the intrinsic and extrinsic parameters of the positioning image, and the location of the smartphone is obtained as a result. Compared with the ground truth, results of the experiments indicate that the proposed approach can be used for indoor positioning, with an accuracy of approximately 10 cm. In addition, experiments show that the proposed method is more robust and efficient than the baseline method in a real scene. In the case where sufficient indoor textures are present, it has the potential to become a low-cost, precise, and highly available indoor positioning technology.

N6-methyladenosine (m6A) is the most abundant epitranscriptomic mark found on mRNA and has important roles in various physiological processes. Despite the relatively high m6A levels in the brain, its potential functions in the brain remain largely unexplored. We performed a transcriptome-wide methylation analysis using the mouse brain to depict its region-specific methylation profile. RNA methylation levels in mouse cerebellum are generally higher than those in the cerebral cortex. Heterogeneity of RNA methylation exists across different brain regions and different types of neural cells including the mRNAs to be methylated, their methylation levels and methylation site selection. Common and region-specific methylation have different preferences for methylation site selection and thereby different impacts on their biological functions. In addition, high methylation levels of fragile X mental retardation protein (FMRP) target mRNAs suggest that m6A methylation is likely to be used for selective recognition of target mRNAs by FMRP in the synapse. Overall, we provide a region-specific map of RNA m6A methylation and characterize the distinct features of specific and common methylation in mouse cerebellum and cerebral cortex. Our results imply that RNA m6A methylation is a newly identified element in the region-specific gene regulatory network in the mouse brain.

Heterosis has been widely exploited as an approach to enhance crop traits during breeding. However, its underlying molecular genetic mechanisms remain unclear. Recent advances in RNA sequencing technology (RNA-seq) have provided an opportunity to conduct transcriptome profiling for heterosis studies. We used RNA-seq to analyze the flower transcriptomes of two F1 hybrid soybeans (HYBSOY-1 and HYBSOY-5) and their parents. More than 385 million high-quality reads were generated and aligned against the soybean reference genome. A total of 681 and 899 genes were identified as being differentially expressed between HYBSOY-1 and HYBSOY-5 and their parents, respectively. These differentially expressed genes (DEGs) were categorized into four major expression categories with 12 expression patterns. Furthermore, gene ontology (GO) term analysis showed that the DEGs were enriched in the categories metabolic process and catalytic activity, while Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis found that metabolic pathway and biosynthesis of secondary metabolites were enriched in the two F1 hybrids. Comparing the DEGs of the two F1 hybrids by GO term and KEGG pathway analyses identified 26 common DEGs that showed transgressive up-regulation, and which could be considered potential candidate genes for heterosis in soybean F1 hybrids. This identification of an extensive transcriptome dataset gives a comprehensive overview of the flower transcriptomes in two F1 hybrids, and provides useful information for soybean hybrid breeding. These findings lay the foundation for future studies on molecular mechanisms underlying soybean heterosis.

Smartphone indoor positioning ground truth is difficult to directly, dynamically, and precisely measure in real-time. To solve this problem, this paper proposes and implements a robust smartphone high-precision indoor positioning dynamic real-time ground truth reference system using color visual scatter-encoded targets based on machine vision and photogrammetry. First, a kind of novel high-precision color vision scatter-encoded patterns with a robust recognition rate is designed. Then we use a smartphone to obtain a sequence of images of an experimental room and extract the base points of the color visual scatter-encoded patterns from the sequence images to establish the indoor local coordinate system of the encoded targets. Finally, we use a high-efficiency algorithm to decode the targets of a real-time dynamic shooting image to obtain accurate instantaneous pose information of a smartphone camera and establish the high-precision and high-availability smartphone indoor positioning direct ground truth reference system for preliminary real-time accuracy evaluation of other smartphone positioning technologies. The experimental results show that the encoded targets of the color visual scatter-encoded pattern designed in this paper are easy to detect and identify, and the layout is simple and affordable. It can accurately and quickly solve the dynamic instantaneous pose of a smartphone camera to complete the self-positioning of the smartphone according to the artificial scatter feature visual positioning technology. It is a fast, efficient and low-cost accuracy-evaluation method for smartphone indoor positioning.

Image mosaicking is one of the key technologies in data processing in the field of computer vision and digital photogrammetry. For the existing problems of seam, pixel aliasing, and ghosting in mosaic images, this paper proposes and implements an optimal seam-line search method based on graph cuts for unmanned aerial vehicle (UAV) remote sensing image mosaicking. This paper first uses a mature and accurate image matching method to register the pre-mosaicked UAV images, and then it marks the source of each pixel in the overlapped area of adjacent images and calculates the energy value contributed by the marker by using the target energy function of graph cuts constructed in this paper. Finally, the optimal seam-line can be obtained by solving the minimum value of target energy function based on graph cuts. The experimental results show that our method can realize seamless UAV image mosaicking, and the image mosaic area transitions naturally.

Ghosting and seams are two major challenges in creating unmanned aerial vehicle (UAV) image mosaic. In response to these problems, this paper proposes an improved method for UAV image seam-line searching. First, an image matching algorithm is used to extract and match the features of adjacent images, so that they can be transformed into the same coordinate system. Then, the gray scale difference, the gradient minimum, and the optical flow value of pixels in adjacent image overlapped area in a neighborhood are calculated, which can be applied to creating an energy function for seam-line searching. Based on that, an improved dynamic programming algorithm is proposed to search the optimal seam-lines to complete the UAV image mosaic. This algorithm adopts a more adaptive energy aggregation and traversal strategy, which can find a more ideal splicing path for adjacent UAV images and avoid the ground objects better. The experimental results show that the proposed method can effectively solve the problems of ghosting and seams in the panoramic UAV images.

Dislocation is one of the major challenges in unmanned aerial vehicle (UAV) image stitching. In this paper, we propose a new algorithm for seamlessly stitching UAV images based on a dynamic programming approach. Our solution consists of two steps: Firstly, an image matching algorithm is used to correct the images so that they are in the same coordinate system. Secondly, a new dynamic programming algorithm is developed based on the concept of a stereo dual-channel energy accumulation. A new energy aggregation and traversal strategy is adopted in our solution, which can find a more optimal seam line for image stitching. Our algorithm overcomes the theoretical limitation of the classical Duplaquet algorithm. Experiments show that the algorithm can effectively solve the dislocation problem in UAV image stitching, especially for the cases in dense urban areas. Our solution is also direction-independent, which has better adaptability and robustness for stitching images.

Recent evidence suggests that aberrant activation of Hedgehog (Hh) signaling by Gli transcription factors is characteristic of a variety of aggressive human carcinomas including ovarian cancer. Therefore, chemotherapeutic agents that inhibit activation of Gli transcription factors have emerged as promising novel therapeutic drugs for ovarian cancer.In this study, we show that activation of Hh signaling promoted cellular migration and invasion, whereas blockade of Hh signaling with GANT61 suppressed cellular migration and invasion in ovarian cancer cells. After treatment with GANT61, cDNA microarray analyses revealed changes in many genes such as Integrin β4 subunit (ITGB4), focal adhesion kinase (FAK), etc. Furthermore, ITGB4 expression was up-regulated by Sonic Hedgehog (Shh) ligand and down-regulated by Hh signaling inhibitor. The Shh-mediated ovarian cell migration and invasion was blocked by neutralizing antibodies to ITGB4. In addition, phosphorylations of FAK were increased by Shh and decreased by Hh signaling inhibitor. Inhibition of Gli1 expression using siRNA mimicked the effects of GANT61 treatment, supporting the specificity of GANT61. Further investigations showed that activation of FAK was required for Shh-mediated cell migration and invasion. Finally, we found that down-regulation of Gli reduced the expression of ITGB4 and the phosphorylated FAK, resulting in the inhibition of tumor growth in vivo.The Hh signaling pathway induces cell migration and invasion through ITGB4-mediated activation of FAK in ovarian cancer. Our findings suggest that the diminishment of crosstalk between phosphorylated FAK and ITGB4 due to the down-regulation of Gli family transcription factors might play a pivotal role for inhibiting ovarian cancer progression.

In view of the problem of taking the loose layer as the uniform cloth load while ignoring thickly sticky loose layered arch (TSLLA) of the traditional mining rock overburden structure analysis, combined with the geological conditions of shallow buried thickly sticky loose layer in Shendong mining area, the mechanical model of the mining TSLLA bearing structure is established, the variation law of arch height under different loose layers is analyzed, and the formation conditions of TSLLA are given. The first theory deduced the dynamic evolution mechanism of TSLLA structure advancing. The dynamic evolution of TSLLA structure in 22207 coalface is simulated. The formula for the limit height of TSLLA is derived. A classification method of mining loose layer structure is proposed. The loose layer structure type is divided into load type, arch beam type damage type, and limit arch height type. On this basis, the mechanical model of "arch beam" is constructed. The influence mechanism of the TSLLA on the surface cracks is revealed. The methods and calculation steps for initial surface cracks are presented. Soil-rock mixture is a special geological material between homogeneous soil masses and fractured rock masses. In this study, the shear characteristics, movement and failure characteristics of particles and the evolution law of cracks were studied by direct shear tests and particle flow numerical simulations. The results show that the shear stress-shear displacement curves of the soil-rock mixture can be roughly classified into three stages: elastic stage, plastic stage and strain softening stage, and there was a "jump" phenomenon. The higher the rock content was, the more obvious the phenomenon. The shear strength and its indices of the soil-rock mixture did not increase with increasing rock content, but there was an "optimal rock content". According to the experimental and simulation results, particle breakage can be divided into three types: slight failure, partial failure and complete failure. The crack propagation characteristics can be divided into three stages, and the crack propagation depth increases with increasing shear displacement. Explorations of the AI-MIII-BIII-PV-O quinary system under solid-state reactions led to an anhydrous cesium-chromium borophosphate-phosphate and an anhydrous cesium-iron borophosphate, namely, Cs2Cr3(BP4O14)(P4O13) (1) and CsFe(BP3O11) (2). They both feature complicated three-dimensional (3D) frameworks and represent the first examples of borophosphate frameworks with a P-O-P connection. Compound 1 contains a novel anionic borophosphate partial structure [B(P2O7)2]5- with B:P = 1:4 besides an isolated [P4O13]6- anion. Its 3D structure is composed of a layer of [CrB(P2O7)2]n2n- and a neutral layer of [Cr2(P4O13)]n interconnected with the linking of P-O-Cr, and the Cs ions are located at the space. Compound 2 contains another novel borophosphate anionic unit of [B(PO4)(P2O7)]4- with B:P = 1:3, which further bridges the Fe cations to constitute the whole structure. Both compounds are of high thermal stable and transparent in the range of 0.75-7.1 μm. Magnetic measurements indicate that there exist antiferromagnetic interactions in both compounds. A novel method is reported here to attach ZnO nanoclusters accurately at the end of multi-walled carbon nanotubes (MWCNTs) in a hydrothermal system. The ends of MWCNTs were selectively functionalised using gas-phase functionalisation in ammonia. ZnO/MWCNT heterojunctions are formed accurately at the nanotube-ends through hydrothermal microemulsion synthesis. The reverse micelle of sodium dodecyl sulphate combines with the positive nitrogen-containing basic groups at the MWCNT end and act as the microreactors for ZnO nanoclusters. The sidewalls of the MWCNTs remain intact without any ZnO particles functionalisation. Poor seed-set limits potential for soybean hybrid seed production. The objective of this study was to evaluate effects of different pollinators on the rate of seed set in three soybean cytoplasmic male sterile (CMS) lines, JLCMS9A, JLCMS82A, and JLCMS89A. The following five treatments were applied under net room isolation-conditions: (i) insecticide application and release of alfalfa leafcutter bees (Megachile rotundata) during flowering; (ii) insecticide application in the absence of leafcutter bees; (iii) release of alfalfa leafcutter bees during flowering without insecticide application; (iv) the absence of insecticide or alfalfa leafcutter bees during flowering; (v) plants without a net cover and pesticide application. Results indicated that the rate of pod production and seed set differ significantly between CMS lines and between treatments. Alfalfa leafcutter bees were an effective pollinator as evidenced by increases in pod-bearing rate and seed-set rate respectively from 21.35 to 53.22% and from 19.91 to 58.77% in the three CMS lines. The three CMS lines also showed significant differences in seed set, with JLCMS82A exhibiting the highest rate in all five treatments, followed by JLCMS9A and then JLCMS89A. These results suggested that thrips (Sericothrips variabilis) might have some effect on soybean pollination, but did not elicit a major influence based on the low increases in seed production. In this article a brick wall model is developed to describe the relationship between the minimum intergranular corrosion path length and the aspect ratio of grains of high-strength wrought aluminum alloy AA2024-T3. We study the distribution of the horizontal distance that a corrosion path will travel in the metal and fit the model to an actual corrosion dataset using the method of moments. The distribution of the horizontal distance of a corrosion path along a given grain is assumed to be uniformly distributed given the length of the grain, which is itself modeled by a gamma distribution. A modified brick wall model is proposed that also imposes a distribution on the vertical distance traveled by the corrosion path. Computer simulation is used to evaluate the fit of these models. A new small-pore compound K4Fe4P5O20 was obtained by conventional solid-state reaction in a closed crucible. The crystal structure is constructed by Fe4P5O20 units forming chains along the c axis and elliptical eight-ring channels on the a--b plane in which K cations locate inside. Such structural characteristics seem to be quite similar to those seen in the natrolite family. However, Fe ions in K4Fe4P5O20 have trigonal--bipyramidal instead of common tetrahedral coordination. Furthermore, our experimental results combined from magnetic susceptibility and 57Fe Mössbauer spectrum measurements show mixed valence Fe3+/Fe2+ in die tided material. Thallium (Tl) is a typical trace metal of extreme high toxicity. As a concomitant element, Tl is widely found in various sulfide minerals and K-containing rock minerals. The outburst of Tl pollution in drinking water sources of the northern branch of the Pearl River in China as reported in 2010 has greatly aroused public concerns about Tl pollution in China. Apart from typical sources of Tl pollution such as Pb and Zn smelting and the mining and utilization of Tl-containing pyrite ores, the steel-making industry was discovered a new significant source that contributed to this Tl pollution incidence. Thallium contents in raw materials, fly ash and wastewater collected from a typical steel-making enterprise were determined by inductively coupled plasma mass spectrometry (ICP-MS). The results showed that Tl contents (0.02-1.03 mg/kg) are generally low in the raw materials, while fly ash samples have generally enriched Tl levels (1.31-6.45 mg/kg). Wastewater obtained from the dedusting process of the sintering furnace also exhibited excessive Tl levels (574-2130 μg/L). All these results suggested a possible release and gasification of Tl compounds from the raw materials under high temperatures (>800°C) during the sintering processes, which were then accumulated in the flue gas and fly ash and washed into the wastewater by wet dedusting. Lime precipitation method is not effective for removing Tl from wastewater, since Tl mostly is present as dissolved Tl+ in the water. "Candidatus Neoehrlichia mikurensis" was detected by PCR in 4.0% (34/841) of the rodents tested in this study. The 34 rodents represented nine species from seven regions of China.
