Abstract Background Unintentional injuries is the leading cause of death in children aged 6–18 in China. Previous studies on the association between the guardians’ educational levels and unintentional injuries in children have been inconclusive, and it remains unclear among the Chinese population. Therefore, this study aimed to identify the association between guardians’ educational levels and unintentional injuries in children aged 6–18 in Shenzhen, China. Methods This cross-sectional study enrolled 9,903 children aged 6–18 in Shenzhen in 2020 using a multistage cluster sampling method. Information on the children and guardians were collected, and unintentional injuries in the past year was examined by using two nested questions. Logistic regression analyses were used to test the association between the guardians’ educational levels and unintentional injuries in children aged 6–18, and the crude odds ratios (ORs) and adjusted ORs with 95% confidence intervals (95% CI) were calculated. Results 275 of the 9,903 children reported experiencing at least one unintentional injuries in the past year, and the weighted incidence of unintentional injuries was 6.3% (95% CI: 5.8–6.8%) in children aged 6–18 in Shenzhen, China. Abstract Long-term non-progressors (LTNPs) of HIV-1 infection may provide important insights into mechanisms involved in viral control and pathogenesis. Here, our results suggest that the ribosomal protein lateral stalk subunit P1 (RPLP1) is expressed at higher levels in LTNPs compared to regular progressors (RPs). Functionally, RPLP1 inhibits transcription of clade B HIV-1 strains by occupying the C/EBPβ binding sites in the viral long terminal repeat (LTR). This interaction requires the α-helixes 2 and 4 domains of RPLP1 and is evaded by HIV-1 group M subtype C and group N, O and P strains that do not require C/EBPβ for transcription. We further demonstrate that HIV-1-induced translocation of RPLP1 from the cytoplasm to the nucleus is essential for antiviral activity. Finally, knock-down of RPLP1 promotes reactivation of latent HIV-1 proviruses. Thus, RPLP1 may play a role in the maintenance of HIV-1 latency and resistance to RPLP1 restriction may contribute to the effective spread of clade C HIV-1 strains.

Abstract Background Direct-acting Antiviral Agents (DAAs) influence serum lipids of patients with Hepatitis C virus (HCV). This paper presents an analysis of the relevant literature to investigate the effects of DAAs in treating hepatitis C to achieve a sustained viral response (SVR) on lipid parameters. Methods PubMed,Web of science, Embase and Central databases were searched, with a deadline of September 2023. Studies on the effects of sustained viral response on lipid parameters after DAAs treatment for hepatitis C were selected. The required information was extracted from the included studies, and then the Stata 12.0 was used to analyze the data quantitatively. Results Of 32 studies, the results showed that total cholesterol (TC) levels increased from the end of treatment (WMD = 20.144, 95%CI = 3.404, 36.884,P = 0.018) to one year after treatment (WMD = 24.900, 95%CI = 13.669, 36.131, P < 0.001). From the end of treatment (WMD = 17.728, 95%CI = 4.375, 31.082, P = 0.009) to one year after treatment (WMD = 18.528, 95%CI = 7.622, 29.433, P < 0.001), the levels of low-density lipoprotein (LDL) were also increased. High-density lipoprotein (HDL) levels were elevated from 4 weeks after treatment (WMD = 6.665, 95%CI = 3.906, 9.424, P < 0.001) to 24 weeks after treatment (WMD = 3.159,95% CI = 0.176, 6.142, P = 0.038). Triglyceride (TG) levels showed no significant change after the treatment. Conclusions Hepatitis C patients who achieved SVR on DAAs showed the increase of lipid levels and the improvement of hepatic inflammation indicators AST and ALT. This may provide evidence-based medical evidence for the follow-up and monitoring of blood lipids and hyperlipidemia treatment. Registration PROSPERO CRD42020180793.

Intravascular large B-cell lymphoma (IVLBCL) is a rare subtype of non-Hodgkin lymphoma. Patients with hemophagocytic lymphohistiocytosis (HLH)-associated IVLBCL variants exhibit significantly poor survival. Cytokines play pivotal roles in malignancy-associated HLH as well as in capillary leak syndrome (CLS). The pathogenesis of CLS involves hyperpermeability and transient endothelial dysfunction. Here, we report the first case of HLH-associated IVLBCL variant complicated with CLS. The patient presented with fever, refractory hypoproteinemia, hypotension and severe edema, followed by telangiectasias. Treatment with etoposide and dexamethasone and hydroxyethyl starch-based artificial colloid led to transient improvement. The diagnosis of IVLBCL was confirmed after the sixth bone marrow biopsy. Subsequently, the R-CHOP (rituximab, cyclophosphamide, hydroxydaunorubicin, vincristine, and prednisolone) regimen was administered and resulted in prompt alleviation of CLS and HLH symptoms. The patient has survived for more than 6 years after combination of immunochemotherapy and autologous peripheral stem-cell transplantation. This case provides some insights into the mechanism and clinical management of IVLBCL complicated with HLH and CLS. Similar cases concerning lymphoma-associated CLSs were also reviewed.

Recycled concrete aggregate (RCA) presents several challenges such as irregular shape, high water absorption, high crushing value, and low apparent density, which restrict its application in concrete. Although carbonation technology offers a more effective solution, its effect on inferior RCA remains unclear. Compared to its common counterpart, inferior RCA is characterized by longer aging time, lower crushing value, and reduced carbonizable substances. This study focuses on low-quality RCA, employing a method of impregnation and carbonation with cement slurry to strengthen the properties of RCA. The investigation studied the carbonation system (hydration time + carbonation time), water–cement ratio of the cement paste, and carbonation effects and mechanisms via mercury injection, nanoindentation tests, X-ray diffraction, and scanning electron microscopy. The findings indicate that the optimal carbonation system involves hydration for 1 d and carbonation for 14 d (H1d + C14d). In this system, the water–cement ratio exhibits minimal influence on carbonation depth. After the impregnation–carbonation treatment, RCA displayed the best performance improvement at a water–cement ratio of 0.8 (0.8-RCA). This condition resulted in a 15.41% reduction in crushing value and an 18.00% decrease in porosity alongside a 21.42% increase in apparent density. Moreover, the interfacial transition zone (ITZ) between aggregate and mortar and the elastic modulus of adhesive mortar are improved after strengthening. Compared with the primitive RCA, the distribution of Ca2+ in the adhesive mortar of the strengthened RCA was more uniform and denser. The moderately fluid cement slurry penetrated the RCA pores easily, enabling CaCO3 produced by carbonation to efficiently fill defects.

BackgroundSensory symptoms linked to tic disorder (TD) are challenging to quantify via self- or parent-reported measures. The current study aimed to develop a novel observer-rated semi-structured interview, namely, the Sensory Phenomenon Assessment Scale (SPAS), to aid clinical evaluation on symptoms of TD among children.MethodsTo test its psychometric properties, tic, premonitory urge (PU), and obsessive–compulsive symptoms (OCS) were also assessed in 223 children via the Yale Global Tic Severity Scale (YGTSS), the Premonitory Urge for Tic Scale (PUTS), and the Children’s Yale–Brown Obsessive–Compulsive Scale (CY-BOCS). Factor analysis and internal consistency test were carried out using data from TD-diagnosed individuals.ResultsGood internal consistency and test–retest reliability were observed. Criterion validity was established by significant correlations between the PUTS, the YGTSS, the CY-BOCS, and scores of the SPAS. Factor analyses supported a single-factor model of the SPAS, in which the five items each showed a factor loading above 0.6.ConclusionThis study demonstrated that the SPAS is reliable and valid and, thus, can serve as a good and concise measure of clinical symptoms among children and adolescents with TD.

BackgroundOsteosarcoma (OS) is highly malignant and prone to local infiltration and distant metastasis. Due to the poor outcomes of OS patients, the study aimed to identify differentially expressed genes (DEGs) in OS and explore their role in the carcinogenesis and progression of OS.MethodsRNA sequencing was performed to identify DEGs in OS. The functions of the DEGs in OS were investigated using bioinformatics analysis, and DEG expression was verified using RT-qPCR and Western blotting. The role of SLC25A4 was evaluated using gene set enrichment analysis (GSEA) and then investigated using functional assays in OS cells.ResultsIn all, 8353 DEGs were screened. GO and KEGG enrichment analyses indicated these DEGs showed strong enrichment in the calcium signaling pathway and pathways in cancer. Moreover, the Kaplan-Meier survival analysis showed ten hub genes were related to the outcomes of OS patients. Both SLC25A4 transcript and protein expression were significantly reduced in OS, and GSEA suggested that SLC25A4 was associated with cell cycle, apoptosis and inflammation. SLC25A4-overexpressing OS cells exhibited suppressed proliferation, migration, invasion and enhanced apoptosis.ConclusionSLC25A4 was found to be significantly downregulated in OS patients, which was associated with poor prognosis. Modulation of SLC25A4 expression levels may be beneficial in OS treatment.

The formation and dynamics of individual estuarine turbidity maximum (ETM) in the Pearl River estuary (PRE) have been investigated but the temporal variability of the ETMs and interconnections among them remain poorly understood. To address these open questions, the distribution and transport of suspended particulate matter (SPM) in the PRE for the period of 2017–2020 are investigated by numerical modeling. The simulated sediment transport flux is decomposed into several major components associated with specific physical processes. Then, the relative contribution of each component to the formation of the ETMs is evaluated. Results suggest the coexistence of three prominent ETMs in the Lingding Bay of the PRE. They are formed by different physical mechanisms and characterized by remarkable seasonality in the spatial extension. In the two ETMs located at the west shoal and middle shoal, advection dominates the sediment transport flux, whilst tidal pumping plays a crucial role in maintaining the ETMs. A sharp bathymetric gradient leads to an entrapment of sediment flux within the bottom layer in the west channel ETM, a phenomenon referred to as topographical trapping. The interconnection analysis shows that the sediment transport between the ETMs varies with seasons, which is attributed to the variation of stratification driven by the monsoon-mediated river runoff. Our results provide new insights into the physical dynamics and interconnections of the ETMs in the PRE, which can serve as scientific base for estuarine sediment management and engineering.

A passively mode-locked Er-doped fiber laser employing ethylene glycol as a saturable absorber (SA) has been proposed and experimentally validated. The ethylene glycol-SA is fabricated by filling the space between the end facets of two optical patch cords inserted into a cannula with ethylene glycol. Utilizing such an SA with a modulation depth of 9.6 %, stable mode-locking operation are manifested in the laser cavity, including the production of bright pulse and bright-dark pulse pair. The generated bright pulse has a center wavelength at 1562.28 nm, a 3 dB bandwidth of 0.21 nm, a repetition frequency of 20.24 MHz, and a signal-to-noise ratio (SNR) of 57 dB. Through precisely manipulating the polarization state and the pump power in the laser cavity, the bright-dark pulse pairs can be successfully attained at the same repetition frequency. This is the first demonstration of a passively mode-locked fiber laser utilizing ethylene glycol as the saturable absorber. Due to their superior optical absorption characteristics, low cost, simplicity in fabrication and non-volatile nature, ethylene glycol-SAs have enormous potential in the field of nonlinear optics and ultrafast photonics.

Geopolymers, environmentally friendly materials, face application limitations due to their high drying shrinkage and propensity for cracking. The shrinkage reducing admixture (SRA) has shown promise in mitigating drying shrinkage in cement-based materials, yet its impact and mechanism in geopolymers remain uncertain. This study examines the influence of the small molecule polyol SRA-2-methyl-2,4-pentanediol on metakaolin-based geopolymer (MKG) properties and its multiscale structural development, aiming to understand the drying shrinkage behavior. The findings reveal that SRA significantly curbs the drying shrinkage of MKG while also reducing its mechanical properties. Analyses of macroscopic and microscopic properties indicate that while SRA does not alter the hydration products of MKG, it can impede the hydration process, particularly in the early stages. Specifically, SRA inhibits the formation of N-A-S-H gel, thereby reducing shrinkage caused by late-stage polycondensation. Furthermore, the alkaline environment in MKG aids SRA in lowering pore solution surface tension and increasing its contact angle, reducing drying shrinkage forces. Additionally, SRA modifies MKG's micro-structure, decreasing pore volume, particularly in the 10–50 nm range, further mitigating shrinkage due to surface tension.

Abstract Background Short stature, amelogenesis imperfecta, and skeletal dysplasia with scoliosis is a rare, autosomal recessive, skeletal disorder first described in 2018. This syndrome starts with pre- and postnatal developmental delay, and gradually presents with variable facial dysmorphisms, a short stature, amelogenesis imperfecta, and progressive skeletal dysplasia affecting the limbs, joints, hands, feet, and spine. Case presentation We identified a homozygous novel nonsense mutation in exon 1 of SLC10A7 (NM_001300842.2: c.100G > T / p.Gly34*) segregating with the typical disease phenotype in a Han Chinese family. We reviewed the 12-year surgical treatment history with seven interventions on spine. Conclusion To date, only 12 cases of the SLC10A7 mutation have been reported, mainly from consanguineous families. Our patient showed a relatively severe and broad clinical phenotype compared with previously reported cases. In this patient, annual check-ups and timely surgeries led to a good outcome.

Abstract The world is facing the problem of depletion of natural sand and gravel resources, and a large amount of coal gangue solid waste is produced in Inner Mongolia, China, which has low utilization rate and causes ecological pollution. In order to improve the gangue in the mining infrastructure construction of a wide range of application prospects, the use of coal gangue as the coarse aggregate of pumice concrete is of great significance. Inner Mongolia is a cold region, and gangue mixed aggregate concrete (MFC) will certainly face the damage caused by freeze–thaw cycles. Therefore, design gangue by different volume replacement rate (0%, 20%, 40%, 60%, 80%, 100%) to replace pumice coarse aggregate. The results show that with the increase of gangue substitution rate, the mass loss rate, relative dynamic elastic modulus, and peak stress of MFC decrease, but the trend of peak strain increases. It is mainly attributed to the less Al2O3 and SiO2 content of gangue, which makes the MFC hydration products decrease with the increase of substitution rate and more original microcracks and pores in the specimens. In addition, the damage model of MFC was established by using Weibull statistical distribution theory and the principle of LEMAITRE equivalent effect variation assumption, and the damage evolution characteristics were explored by combining the experimental results. It can provide the theoretical basis for the application of MFC in cold regions.

BackgroundThere is a dearth of feasibility assessments regarding using large language models (LLMs) for responding to inquiries from autistic patients within a Chinese-language context. Despite Chinese being one of the most widely spoken languages globally, the predominant research focus on applying these models in the medical field has been on English-speaking populations. ObjectiveThis study aims to assess the effectiveness of LLM chatbots, specifically ChatGPT-4 (OpenAI) and ERNIE Bot (version 2.2.3; Baidu, Inc), one of the most advanced LLMs in China, in addressing inquiries from autistic individuals in a Chinese setting. MethodsFor this study, we gathered data from DXY—a widely acknowledged, web-based, medical consultation platform in China with a user base of over 100 million individuals. A total of 100 patient consultation samples were rigorously selected from January 2018 to August 2023, amounting to 239 questions extracted from publicly available autism-related documents on the platform. To maintain objectivity, both the original questions and responses were anonymized and randomized. An evaluation team of 3 chief physicians assessed the responses across 4 dimensions: relevance, accuracy, usefulness, and empathy. The team completed 717 evaluations. The team initially identified the best response and then used a Likert scale with 5 response categories to gauge the responses, each representing a distinct level of quality. Finally, we compared the responses collected from different sources. ResultsAmong the 717 evaluations conducted, 46.86% (95% CI 43.21%-50.51%) of assessors displayed varying preferences for responses from physicians, with 34.87% (95% CI 31.38%-38.36%) of assessors favoring ChatGPT and 18.27% (95% CI 15.44%-21.10%) of assessors favoring ERNIE Bot. The average relevance scores for physicians, ChatGPT, and ERNIE Bot were 3.75 (95% CI 3.69-3.82), 3.69 (95% CI 3.63-3.74), and 3.41 (95% CI 3.35-3.46), respectively. Physicians (3.66, 95% CI 3.60-3.73) and ChatGPT (3.73, 95% CI 3.69-3.77) demonstrated higher accuracy ratings compared to ERNIE Bot (3.52, 95% CI 3.47-3.57). In terms of usefulness scores, physicians (3.54, 95% CI 3.47-3.62) received higher ratings than ChatGPT (3.40, 95% CI 3.34-3.47) and ERNIE Bot (3.05, 95% CI 2.99-3.12). Finally, concerning the empathy dimension, ChatGPT (3.64, 95% CI 3.57-3.71) outperformed physicians (3.13, 95% CI 3.04-3.21) and ERNIE Bot (3.11, 95% CI 3.04-3.18). ConclusionsIn this cross-sectional study, physicians’ responses exhibited superiority in the present Chinese-language context. Nonetheless, LLMs can provide valuable medical guidance to autistic patients and may even surpass physicians in demonstrating empathy. However, it is crucial to acknowledge that further optimization and research are imperative prerequisites before the effective integration of LLMs in clinical settings across diverse linguistic environments can be realized. ABSTRACT The accessory protein ORF6 of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a key interferon (IFN) antagonist that strongly suppresses the production of primary IFN as well as the expression of IFN-stimulated genes. However, how host cells respond to ORF6 remains largely unknown. Our research of ORF6-binding proteins by pulldown revealed that E3 ligase components such as Cullin 4B (CUL4B), DDB1, and RBX1 are potential ORF6-interacting proteins. Further study found that the substrate recognition receptor PRPF19 interacts with CUL4B, DDB1, and RBX1 to form a CRL4B-based E3 ligase, which catalyzes ORF6 ubiquitination and subsequent degradation. Overexpression of PRPF19 promotes ORF6 degradation, releasing ORF6-mediated IFN inhibition, which inhibits SARS-CoV-2 replication. Moreover, we found that activation of CUL4B by the neddylation inducer etoposide alleviates lung lesions in a SARS-CoV-2 mouse infection model. Therefore, targeting ORF6 for degradation may be an effective therapeutic strategy against SARS-CoV-2 infection.IMPORTANCEThe cellular biological function of the ubiquitin-proteasome pathway as an important modulator for the regulation of many fundamental cellular processes has been greatly appreciated. The critical role of the ubiquitin-proteasome pathway in viral pathogenesis has become increasingly apparent. It is a powerful tool that host cells use to defend against viral infection. Some cellular proteins can function as restriction factors to limit viral infection by ubiquitin-dependent degradation. In this research, we identificated of CUL4B-DDB1-PRPF19 E3 Ubiquitin Ligase Complex can mediate proteasomal degradation of ORF6, leading to inhibition of viral replication. Moreover, the CUL4B activator etoposide alleviates disease development in a mouse infection model, suggesting that this agent or its derivatives may be used to treat infections caused by SARS-CoV-2. We believe that these results will be extremely useful for the scientific and clinic communities in their search for cues and preventive measures to combat the COVID-19 pandemic.

Abstract Aims The aims are to describe the key components of family integrated care intervention for preterm infants in the neonatal intensive care unit (NICU) and assess the impact on breastfeeding outcomes for those infants. Design A scoping review. Methods We conducted a systematic study search based on the databases, including PubMed, Scopus, Cochrane, Web of Science, MEDLINE, CINAHL, CNKI and Wanfang Database in December 2022. The search time ranged from database establishment to 31 December 2022. Papers by manual searching were also listed on the references. We adopted Joanna Briggs Institute Reviewer's Manual methodology and followed the PRISMA guidelines for Scoping Reviews (PRISMA‐ScR) to conduct the review. Two independent reviewers filtered the papers, extracted data and synthesized the findings. A table was used to extract data and synthesize results. Results After systematic searching, 11 articles that implemented family integrated care (FIcare) were finally included in this scoping review. By analysing the implementation of this nursing model, we identified seven main components: NICU staff training, parent education, parent participation in infants' care, parent involvement in medical plans, peer support, NICU environmental support and mobile app for parents. Based on the extracted breastfeeding data, this scoping review concludes that family integrated care shows a positive effect on increasing breastfeeding rates at discharge. Through this scoping review, we find that family integrated care is feasible and it can support breastfeeding of preterm infants. Further studies will be needed to provide more evidence that family integrated care could facilitate breastfeeding of preterm infants. Impact This scoping review provides evidence for the positive role of family integrated care on breastfeeding outcomes. The analysis may contribute to the implementation of family integrated care. This study explores the health consequences of prolonged sitting by introducing a standing chair and examining the effects of seat tilt angles on comfort. Using synchronized pressure distribution testing and surface electromyographic (sEMG) signal analysis, we assessed pressure and sEMG responses at tilt angles of 0°, 20°, and 40° during tasks such as computer work, writing, and ironing. Initial measurements with body pressure distribution sensors targeted the buttocks and feet, while surface electromyographic equipment captured signals from the bilateral lumbar erector spinae, vastus lateralis, and gastrocnemius muscles. MATLAB processing facilitated the analysis of integrated electromyographic values and mean power frequencies, elucidating the effects of tilt angles on comfort. Our research findings indicate that a 20° tilt angle significantly enhances comfort during computer and writing tasks and noticeably increases the comfort of the erector spinae muscles during ironing. Conversely, a 0° tilt angle is more beneficial for the vastus lateralis and gastrocnemius muscles. These results underscore the importance of selecting appropriate tilt angles to improve comfort across various tasks. Furthermore, integrating pressure sensors and surface electromyographic signals enables a comprehensive evaluation of chair ergonomic quality, offering valuable insights for chair design optimization.

The node pressure and flow rate along the shale gas flow process are analyzed according to the characteristics of the shale gas flow pipe network, and the non-leaking and leaking processes of the shale gas flow pipe network are modeled separately. The changes in pressure over time along each pipe segment in the network provide new ideas for identifying leaking pipe sections. This paper uses the logarithmic value of pressure as the basis for judging whether the flow pipe network is leaking or not, according to the process of varying flow parameters resulting in the regularity of leakage. A graph of the change in pressure of the pipe section after the leak compared to the pressure of the non-leaking section of pipe over time can be plotted, accurately identifying the specific section of pipe with the leak. The accuracy of this novel method is verified by the leakage section and statistical data of the shale gas pipeline network in situ used in this paper.

The mathematical model of a flexible multistate switch (FMSS) exhibits nonlinear and strong coupling characteristics, whereas traditional power decoupling control makes it difficult to completely decouple the output power. The traditional proportional–integral control parameters are difficult to adjust, and their robustness and dynamic performance are poor, which affects the stability of the voltage of the power distribution network and feeder power. To address these problems, this study first converted the original system into a linear system via coordinate transformation using feedback-accurate linearization to decouple active and reactive currents. Thereafter, a super-twisting sliding mode control (ST-SMC) algorithm was introduced, and an adaptive quasi-super-twisting sliding mode control (AQST-SMC) algorithm comprising the quasi-sliding mode function and adaptive proportional term was proposed. An FMSS double closed-loop controller was designed to achieve improved vibration suppression and convergence speed. A three-port FMSS simulation model was developed using MATLAB/Simulink, and the simulation results show that the proposed control strategy enhances the robustness and dynamic performance of the system.

Abstract Background Plasma amyloid-β (Aβ) peptides and tau proteins are promising biomarkers of Alzheimer’s disease (AD), not only for predicting Aβ and tau pathology but also for differentiating AD from other neurodegenerative diseases. However, reference intervals for plasma biomarkers of AD in healthy elderly Chinese individuals have not yet been established. Methods Biomarkers of AD were measured using single-molecule array (Simoa) assays in plasma samples from 193 healthy, cognitively unimpaired Chinese individuals aged 50–89 years. The 95% reference intervals for plasma Aβ42, Aβ40, t-tau, p-tau181, and derived ratios were calculated by using log-transformed parametric methods. Results Plasma Aβ42, Aβ40, and p-tau181 levels were positively correlated with age, while the Aβ42/Aβ40 ratio was negatively correlated with age. The 95% reference intervals for plasma Aβ42 and Aβ40 were 2.72–11.09 pg/mL and 61.4–303.9 pg/mL, respectively, and the 95% reference intervals for plasma t-tau and p-tau181 were 0.20–3.12 pg/mL and 0.49–3.29 pg/mL, respectively. The 95% reference intervals for the Aβ42/Aβ40 ratio, p-tau181/t-tau ratio, and p-tau181/Aβ42 ratio were 0.022–0.064, 0.38–6.34, and 0.05–0.55, respectively. Conclusion Reference intervals for plasma biomarkers of AD may assist clinicians in making accurate clinical decisions.

Aim: To explore the lactating nurses' experiences of return to work after lifting COVID-19 lockdown. Background: Return to work is a key reason for the low rates of breastfeeding. Especially after lifting COVID-19 lockdown, case counts reached recorded highs. So lactating nurses face more challenges when they return to work. Method: The empirical phenomenology method was used to conduct a qualitative study. Lactating nurses were recruited in a tertiary hospital through purposive and snowball sampling, and participated in semi-structured video interviews. Colaizzi's method was used to analyze the data. Results: Three themes and 10 sub-themes emerged from the interview data of 15 participants. The first theme was “preparation for return to work”, which helped lactating nurses adapt to return to work quickly. The second was “experiences of return to work”. The inconvenience of pumping was mentioned repeatedly. In addition, the flexible work schedule was highlighted. The third was “experiences of infection”. The attitudes toward breastfeeding differed due to different perceptions of COVID-19. Conclusions: Lactation nurses easily interrupted or stopped breastfeeding when they returned to work after lifting COVID-19 lockdown. Recommendations include the further provision of longer periods of leave, flexible working arrangements, separate facilities for breast pumping, and breastfeeding strategies for epidemics.

Traditional approaches to improving basketball players’ shooting skills rely on coaches’ experience in adjusting players’ biomechanical motions. However, such an approach cannot provide specific instructions or facilitate immediate feedback for improvement of the shooting motion. In this article, a method is presented to quantitatively evaluate four key action indicators of shooting basketballs using a machine‐learning model based on Bayesian optimization of a light gradient boosting machine (LightGBM). Important motion data for the model are collected by micro‐inertial measurement units embedded in a wrist motion sensor and an internet of things (IoT) smart basketball. Basketball shooting motion data are collected from 16 subjects and used for model training and data testing, and four important action indicators that influence the shot quality are selected for quantitative assessment. The LightGBM model is then developed for the regression prediction of the four action indicators of shooting. In the results, it is indicated that for an individual player, the highest correlation scores of the four indexes range from 97.6% to 99.3%. The proposed approach for quantitatively assessing shooting indexes can provide objective and data‐based guidance to improve players’ shooting performance. Foreseeably, the prediction model can be embedded into a chip of a wearable device to evaluate the real‐time shot quality quantitatively.

Background Tumor mutational burden (TMB) is a valuable prognostic biomarker. This study explored the predictive value of TMB and the potential association between TMB and immune infiltration in diffuse large B-cell lymphoma (DLBCL). Methods We downloaded the gene expression profile, somatic mutation, and clinical data of DLBCL patients from The Cancer Genome Atlas (TCGA) database. We classified the samples into high-and low-TMB groups to identify differentially expressed genes (DEGs). Functional enrichment analyses were performed to determine the biological functions of the DEGs. We utilized the cell-type identification by estimating relative subsets of RNA transcripts (CIBERSORT) algorithm to estimate the abundance of 22 immune cells, and the significant difference was determined by the Wilcoxon rank-sum test between the high- and low-TMB group. Hub gene had been screened as the prognostic TMB-related immune biomarker by the combination of the Immunology Database and Analysis Portal (ImmPort) database and the univariate Cox analysis from the Gene Expression Omnibus (GEO) database including six DLBCL datasets. Various database applications such as Tumor Immune Estimation Resource (TIMER), CellMiner, konckTF, and Genotype-Tissue Expression (GTEx) verified the functions of the target gene. Wet assay confirmed the target gene expression at RNA and protein levels in DLBCL tissue and cell samples. Results Single nucleotide polymorphism (SNP) occurred more frequently than insertion and deletion, and C > T was the most common single nucleotide variant (SNV) in DLBCL. Survival analysis showed that the high-TMB group conferred poor survival outcomes. A total of 62 DEGs were obtained, and 13 TMB-related immune genes were identified. Univariate Cox analysis results illustrated that CD1c mutation was associated with lower TMB and manifested a satisfactory clinical prognosis by analysis of large samples from the GEO database. In addition, infiltration levels of immune cells in the high-TMB group were lower. Using the TIMER database, we systematically analyzed that the expression of CD1c was positively correlated with B cells, neutrophils, and dendritic cells and negatively correlated with CD8+ T cells, CD4+ T cells, and macrophages. Drug sensitivity showed a significant positive correlation between CD1c expression level and clinical drug sensitivity from the CellMiner database. CREB1, AHR, and TOX were used to comprehensively explore the regulation of CD1c-related transcription factors and signaling pathways by the KnockTF database. We searched the GETx database to compare the mRNA expression levels of CD1c between DLBCL and normal tissues, and the results suggested a significant difference between them. Moreover, wet experiments were conducted to verify the high expression of CD1c in DLBCL at the RNA and protein levels. Conclusions Higher TMB correlated with poor survival outcomes and inhibited the immune infiltrates in DLBCL. Our results suggest that CD1c is a TMB-related prognostic biomarker.

Abstract The coronavirus disease 2019 (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused a severe global health crisis; its structural protein envelope (E) is critical for viral entry, budding, production, and induction of pathology which makes it a potential target for therapeutics against COVID-19. Here, we find that the E3 ligase RNF5 interacts with and catalyzes ubiquitination of E on the 63rd lysine, leading to its degradation by the ubiquitin-proteasome system (UPS). Importantly, RNF5-induced degradation of E inhibits SARS-CoV-2 replication and the RNF5 pharmacological activator Analog-1 alleviates disease development in a mouse infection model. We also found that RNF5 is distinctively expressed in different age groups and in patients displaying different disease severity, which may be exploited as a prognostic marker for COVID-19. Furthermore, RNF5 recognized the E protein from various SARS-CoV-2 strains and SARS-CoV, suggesting that targeting RNF5 is a broad-spectrum antiviral strategy. Our findings provide novel insights into the role of UPS in antagonizing SARS-CoV-2 replication, which opens new avenues for therapeutic intervention to combat the COVID-19 pandemic.

As a key component of meridional overturning circulation, mid-deep circulation plays a crucial role in the vertical and meridional distribution of heat. However, due to a lack of observation data, current knowledge of the dynamics of mid-deep circulation currents moving through basin boundaries and complicated seabed topographies is severely limited. In this study, we combined oceanic observation data, bathymetric data, and numerical modeling of the northwest continental margin of the South China Sea to investigate (i) the main features of mid-deep circulation currents traveling through the central depression belt and (ii) how atmospheric-forcing (winds) mesoscale oceanic processes such as eddies and current–topography interactions modulate the mid-deep circulation patterns. Comprehensive results suggest that the convergence of different water masses and current–topography interactions take primary responsibility for the generation of instability and enhanced mixing within the central depression belt. By contrast, winds and mesoscale eddies have limited influence on the development of local circulation patterns at mid-deep depths (>400 m). This study emphasizes that the intensification and bifurcation of mid-deep circulation; specifically, those induced by a large depression belt morphology determine the local material cycle (temperature, salinity, etc.) and energy distribution. These findings provide insights for a better understanding of mid-deep circulation structures on the western boundary of ocean basins such as the South China Sea.

The performance of the capacitive gap-sensing system plays a critical role in a satellite-based gravity gradiometer that is developed using an electrostatic accelerometer. The capacitive sensing gain mainly depends on the stabilized injection bias amplitude, the gain of the transformer bridge, and the trans-impedance amplifier. Previous studies have indicated that amplitude noise is the main factor influencing the noise of capacitive displacement detection. Analyzing the capacitive gap-sensing system indicates that the amplitude, frequency, phase, and broadband noises of the stabilized injection bias have varying levels of influence on the performance of the detection system. This paper establishes a model to clarify the mentioned effects. The validation of the sub-tests demonstrates that the analysis and evaluation results of various noise coefficients are highly consistent with the model’s predicted outcomes.

Previous studies have shown that anisotropy generally exists in geological bodies such as sedimentary rocks and fault zones, and more and more attention has been paid to the arbitrary conductivity media in surveys with the magnetotelluric sounding method. With the breakthrough development of computer hardware technology, large-scale 3D magnetotelluric modeling in anisotropic media has gradually become possible. At present, there are 3D magnetotelluric field simulation algorithms based on finite differences or finite elements for arbitrary anisotropic conductivity. In order to solve the common computational efficiency problems of the existing algorithms, we proposed a rapid 3D magnetotelluric forward approach for arbitrary anisotropic conductivity in the Fourier domain. Through the 2D Fourier transform, the governing equation can be converted from the space domain to the Fourier domain, thereby greatly reducing the calculation amount of the numerical simulation and improving the calculation efficiency. Then, the classical 1D anisotropy model is used to verify the correctness and the computational efficiency. Finally, the 3D anisotropic models of land and ocean are calculated, and the influence characteristics of the anisotropic medium on the magnetotelluric response are analyzed. The proposed algorithm will be used in the inverse imaging technique for large-scale 3D anisotropic data in future studies.

BackgroundExhaustion of CD8+ tumor-infiltrating lymphocytes (TILs), characterized by the overexpression of immune checkpoints (IC), is a major impediment to anti-tumor immunity. However, the exhaustion status of CD8+TILs in angioimmunoblastic T cell lymphoma (AITL) remains unclear. Therefore, we aimed to elucidate the exhaustion status of CD8+TILs in AITL and its influence on prognosis.MethodsThe correlation between CD8+TILs and IC expression in AITL was analyzed using single-cell RNA sequencing (n = 2), flow cytometry (n = 20), and RNA sequencing (n = 20). Biological changes related to CD8+TILs exhaustion at different cytotoxic T lymphocyte (CTL) levels (mean expression levels of CD8A, CD8B, GZMA, GZMB, and PRF1) in AITL were evaluated using RNA sequencing (n = 20) and further validated using the GEO dataset (n = 51). The impact of CD8 protein expression and CTL levels on patient prognosis was analyzed using flow cytometry and RNA sequencing, respectively.ResultsOur findings demonstrated that the higher the infiltration of CD8+TILs, the higher was the proportion of exhausted CD8+TILs characterized by the overexpression of multiple IC. This was accompanied by extensive exhaustion-related biological changes, which suggested severe exhaustion in CD8+TILs and may be one of the main reasons for the poor prognosis of patients with high CD8+TILs and CTL.ConclusionOur study comprehensively reveals the exhaustion status of CD8+TILs and their potential negative impact on AITL prognosis, which facilitates further mechanistic studies and is valuable for guiding immunotherapy strategies.

The morphological evolution of two mud depocenters in the southwestern Baltic Sea is investigated by comparison of numerical model results to geological and oceanographic data. The pathways of dense currents during episodic dense-water inflows from the North Sea are shown to correspond to current pathways inferred from contouritic depositional geometries in the flow-confining channels within the study area. A favorable comparison of model results to published current speed observations shows that the mesoscale dynamics of individual inflow events are reproduced by the model, indicating that external forcing and basin geometry rather than internal dynamics control the mesoscale dynamics of inflow events. The bottom current directions during inflows show high stability in the flow-confining channels and explain the contouritic depositional geometries. Asymmetric depositional features in the channels are qualitatively reproduced in the model. Bottom currents are less stable in areas without contouritic features, possibly resulting in an overall diffusive effect on sediment distribution in those areas. In a simulation of resuspension by bottom-contacting fishing gear, inter-basin sediment transport is increased by 4–30%, depending on the area, compared to the case of natural hydrodynamic resuspension. The model predicts an increased winnowing of the finest sediment fraction due to bottom trawling, leading to an overall coarsening-to-fining trend in the direction of net sediment transport. The results show that rather than hemi-pelagic background sedimentation, episodic events with high bottom current velocities as well as bottom-trawling induced resuspension are responsible for the present-day and future morphological configuration of the mud depocenters in the southwestern Baltic Sea.

Congenital anomalous origin of coronary artery is a rare cardiovascular malformation and the most common anomaly is the left circumflex (LCX) arising from the right sinus of Valsalva (RSV). Other forms include both coronary arteries from RSV, the left anterior descending coronary artery from RSV, and a single coronary artery from the left sinus of Valsalva. Despite being rare, anomalous origin of left main coronary artery (LMCA) from RSV carries a high risk of sudden cardiac death. Here, we report a case of 13-year-old boy with chest pain and acute extensive anterior ST-segment elevation myocardial infarction (STEMI) who was initially diagnosed as acute myocarditis in the emergency department. A bedside echocardiogram showed severe global hypokinesia of left ventricle (LV) and normal right ventricle (RV) function. Coronary computed tomography angiography (CCTA) examination showed LMCA originated from the RSV. The patient underwent coronary artery bypass grafting surgery and was discharged without complications. A timely correct diagnosis of an anomalous coronary artery is critical in symptomatic patients, CCTA plays an important role in clinical decision making.

Numerous viruses manipulate host factors for viral production. We demonstrated that human enterovirus A71 (EVA71), a primary causative agent for hand, foot, and mouth disease (HFMD), increased the level of the DNA damage response (DDR) marker γ-H2AX. DDR is primarily mediated by the ataxia telangiectasia mutated (ATM), ATM and Rad3-related (ATR), or DNA-dependent protein kinase (DNA-PK) pathways. Upregulation of γ-H2AX by EVA71 was dependent on the ATR but not the ATM or DNA-PK pathway. As a nuclear factor, there is no previous evidence of cytoplasmic distribution of γ-H2AX. However, the present findings demonstrated that EVA71 encouraged the localization of γ-H2AX to the cytoplasm. Of note, γ-H2AX formed a complex with structural protein VP3, non-structural protein 3D, and the viral genome. Treatment with an inhibitor or CRISPR/Cas9 technology to decrease or silence the expression of γ-H2AX decreased viral genome replication in host cells; this effect was accompanied by decreased viral protein expression and virions. In animal experiments, caffeine was used to inhibit DDR; the results revealed that caffeine protected neonatal mice from death after infection with EVA71, laying the foundation for new therapeutic applications of caffeine. More importantly, in children with HFMD, γ-H2AX was upregulated in peripheral blood lymphocytes. The consistent in vitro and in vivo data on γ-H2AX from this study suggested that caffeine or other inhibitors of DDR might be novel therapeutic agents for HFMD.

Abstract. Background:. Ubiquitination plays an essential role in many biological processes, including viral infection, and can be reversed by deubiquitinating enzymes (DUBs). Although some studies discovered that DUBs inhibit or enhance viral infection by various mechanisms, there is lack of information on the role of DUBs in virus regulation, which needs to be further investigated. Methods:. Immunoblotting, real-time polymerase chain reaction, in vivo/in vitro deubiquitination, protein immunoprecipitation, immunofluorescence, and co-localization biological techniques were employed to examine the effect of ubiquitin-specific protease 3 (USP3) on APOBEC3G (A3G) stability and human immunodeficiency virus (HIV) replication. To analyse the relationship between USP3 and HIV disease progression, we recruited 20 HIV-infected patients to detect the levels of USP3 and A3G in peripheral blood and analysed their correlation with CD4+ T-cell counts. Correlation was estimated by Pearson correlation coefficients (for parametric data). Results:. The results demonstrated that USP3 specifically inhibits HIV-1 replication in an A3G-dependent manner. Further investigation found that USP3 stabilized 90% to 95% of A3G expression by deubiquitinating Vif-mediated polyubiquitination and blocking its degradation in an enzyme-dependent manner. It also enhances the A3G messenger RNA (mRNA) level by binding to A3G mRNA and stabilizing it in an enzyme-independent manner. Moreover, USP3 expression was positively correlated with A3G expression (r = 0.5110) and CD4+ T-cell counts (r = 0.5083) in HIV-1-infected patients. Conclusions:. USP3 restricts HIV-1 viral infections by increasing the expression of the antiviral factor A3G. Therefore, USP3 may be an important target for drug development and serve as a novel therapeutic strategy against viral infections.

Abstract Although diaphragmatic dysfunction is an important indicator of severity of illness and poor prognosis in ICU patients, there is no convenient and practical method to monitor diaphragmatic function. This study was designed to analyze diaphragmatic dynamic dysfunction by bedside ultrasound in septic patients and provide quantitative evidence to assess diaphragm function systematically. This prospective observational study was conducted from October 2019 to January 2021 in the Department of Respiratory and Critical Care Medicine. 74 patients suffered from sepsis were recruited and divided into two groups, sepsis group 1 (2 ≤ SOFA ≤ 5, n = 41) and sepsis group 2 (SOFA > 5, n = 33). 107 healthy volunteers were randomly recruited as the control group. In all participants, the diaphragmatic thickness and excursion were measured directly and the dynamic parameters including thickening fraction (TF), EQB/EDB, Contractile velocity, and area under diaphragmatic movement curve (AUDMC) were calculated by bedside ultrasound during quiet breathing (QB) and deep breathing (DB). Each parameter among three groups was analyzed separately by covariance analysis, which was adjusted by age, sex, body mass index, MAP, hypertension, and diabetes. First, contractile dysfunction occurred before diaphragmatic atrophy both in sepsis group 1 and sepsis group 2. Second, compared with the control group, the dynamic parameters showed significant decrease in sepsis group 1 and more obvious change in sepsis group 2, including TF, EQB/EDB. Third, the maximum contractile velocity decreased in sepsis group 1, reflecting the damage of intrinsic contraction efficiency accurately. Finally, per breathing AUDMC in two septic groups were lower than those in control group. However, per minute AUDMC was compensated by increasing respiratory rate in sepsis group 1, whereas it failed to be compensated which indicated gradual failure of diaphragm in sepsis group 2. Diaphragmatic ultrasound can be used to quantitatively evaluate the severity of sepsis patients whose contractile dysfunction occurred before diaphragmatic atrophy. As dynamic parameters, TF and EQB/EDB are early indicator associated with diaphragmatic injury. Furthermore, maximum contractile velocity can reflect intrinsic contraction efficiency accurately. AUDMC can evaluate diaphragmatic breathing effort and endurance to overcome resistance loads effectively.

Six kinds of dithiocarbamates (DTCs) were synthesized from three linear amines with different amino numbers, two polyether amines with different molecular weights, and one branched amine with benzene rings, respectively. The conditions affecting oil removal rate and floc rising time of DTC were studied using simulated oily wastewater. Furthermore, the effects of the molecular structure of DTC on oil removal efficiency, floc morphology, floc rising time, and floc adhesion were investigated. When the conditions were optimal, the oil removal efficiency of DTC synthesized from polyethylene polyamine was 95.14%, which was higher than other DTCs. Meanwhile, the ferrous ion was the most suitable chelating metal ion for DTC than other transition metal ions. The increase of amino groups in the initiators improves the oil removal efficiency of DTC, while the linear structural DTC exhibits a low oil removal efficiency due to a lack of network structural flocs. The introduction of polyether structure helps reduce the volume of the flocs and make them compact, but it also increases the adhesion of the floc on the metal surface. The introduction of bisphenol A phenol amino resin structure induces the generation of the flocs in oil wastewater and improves the oil removal efficiency. HIGHLIGHTS A DTC was synthesized from the bisphenol A phenolamine resin for the first time.; The properties of DTC were evaluated by oil removal efficiency and floc property.; Optimal DTC/Fe2+ molar ratios agree with amino groups per molecule of initiators.; The polyether structure helps reduce the volume of the flocs.; The polyether structure increases the adhesion of the flocs on the metal surface.;

Magnetotactic bacteria (MTB) within the Nitrospirota phylum play important roles in biogeochemical cycles due to their outstanding ability to biomineralize large amounts of magnetite magnetosomes and intracellular sulfur globules. For several decades, Nitrospirota MTB were believed to only live in freshwater or low-salinity environments. While this group have recently been found in marine sediments, their physiological features and ecological roles have remained unclear. In this study, we combine electron microscopy with genomics to characterize a novel population of Nitrospirota MTB in a coral reef area of the South China Sea. Both phylogenetic and genomic analyses revealed it as representative of a novel genus, named as Candidatus Magnetocorallium paracelense XS-1. The cells of XS-1 are small and vibrioid-shaped, and have bundled chains of bullet-shaped magnetite magnetosomes, sulfur globules, and cytoplasmic vacuole-like structures. Genomic analysis revealed that XS-1 has the potential to respire sulfate and nitrate, and utilize the Wood–Ljungdahl pathway for carbon fixation. XS-1 has versatile metabolic traits that make it different from freshwater Nitrospirota MTB, including Pta-ackA pathway, anaerobic sulfite reduction, and thiosulfate disproportionation. XS-1 also encodes both the cbb3-type and the aa3-type cytochrome c oxidases, which may function as respiratory energy-transducing enzymes under high oxygen conditions and anaerobic or microaerophilic conditions, respectively. XS-1 has multiple copies of circadian related genes in response to variability in coral reef habitat. Our results implied that XS-1 has a remarkable plasticity to adapt the environment and can play a beneficial role in coral reef ecosystems.

BackgroundUrinary tract infections (UTIs) are some of the most common bacterial infections in the world. Nevertheless, as uncomplicated UTIs are treated empirically without culturing the urine, adequate knowledge of the resistance pattern of uropathogens is essential. Conventional urine culture and identification take at least 2 days. Here, we developed a platform based on LAMP and centrifugal disk system (LCD) to simultaneously detect the main pathogens and antibiotic resistant genes (ARGs) of urgent concern multidrug-resistant among UTIs.MethodsWe designed specific primers to detect the target genes above and evaluated their sensitivity and specificity. We also assessed the result of our preload LCD platform on 645 urine specimens with a conventional culturing method and Sanger sequencing.ResultsThe results obtained with the 645 clinical samples indicated that the platform has high specificity (0.988–1) and sensitivity (0.904–1) for the studied pathogens and ARGs. Moreover, the kappa value of all pathogens was more than 0.75, revealing an excellent agreement between the LCD and culture method. Compared to phenotypic tests, the LCD platform is a practical and fast detection approach for methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococci, carbapenem-resistant Enterobacteriaceae, carbapenem-resistant Acinetobacter, carbapenem-resistant Pseudomonas aeruginosa (kappa value of all >0.75), and non-extended-spectrum β-lactamase producers.ConclusionWe developed a detection platform that has high accuracy and that meets the need for rapid diagnosis, which can be completed within 1.5 h from specimen collection. It may be a powerful tool for evidence-based UTIs diagnosis, which has essential support for the rational use of antibiotics. More high-quality clinical studies are required to prove the effectiveness of our platform.

In the petroleum industry, the casing steel is fixed with a cement sheath to ensure reliable service in demanding conditions characterized by high temperature, high pressure, and exposure to multiple types of media. After the hydration of the cement, a porous material is produced with a highly alkaline solution filling the pores, commonly referred to as the pore solution. The casing will form a protective passive film when in contact with a highly alkaline pore solution. Nevertheless, once the cement sheath cracks, chloride ions in the stratum will pass through the cement sheath to the surface of the casing. When chloride ions accumulate to a certain concentration, the passive film will be destroyed, without exerting a protective influence on the substrate. After chloride ions come into direct contact with the casing, the casing is prone to severe failure due to corrosion perforation. The casing failure can cause a blowout outside the casing and even scrapping of the oil well. Controlling casing corrosion and ensuring casing integrity relies on understanding the critical chloride ion concentration that can cause the degradation of the passive film. Therefore, to assess the electrochemical properties and analyze the damage process of the passive film under varying chloride ion concentrations, several characterization techniques were employed. These included potential–time curves (E-t), polarization curves, electrochemical impedance spectroscopy (EIS), and Mott–Schottky curves. In addition, the composition of the passive film on the surface of the P110 casing steel was qualitatively analyzed using X-ray photoelectron spectroscopy (XPS). To further understand the surface morphology of the P110 casing steel, scanning electron microscopy (SEM) was used.

Illicium verum Hook. F., also known as star anise, is one of the most important plants of the genus Anise in the family Magnoliaceae. I. verum not only has the functions of warming Yang, dispersing cold, regulating Qi and relieving pain but can also be used as a condiment to increase flavor as well as reconcile and remove fish smells. Currently, 201 chemical constituents have been identified from star anise; among these, star anise oil and shikimic acid are the two most widely used and studied chemical components in star anise, with the oil accounting for a large proportion of the total. This review integrates, classifies and updates studies related to the botany, pharmacology, phytochemistry, traditional and modern uses and quality control of star anise, with a special reference to its phytochemical composition and pharmacological activity. It will provide a reference for further research on this important medicinal plant. In addition, the broad applications and research profiles of star anise essential oil and shikimic acid are highlighted. Our review indicates that the research prospects regarding star anise are very broad and worthy of further investigation.

The Pearl River Delta (PRD), where several megacities are located, has undergone drastic morphological changes caused by anthropogenic impact during the past few decades. In its main estuary, the water area has been reduced by 21% whilst the average water depth has increased by 2.24 m from 1970s to 2010s. The mainly human-induced morphological change together with sea level rise has jointly led to a remarkable change in the water stratification. However, the spatial and temporal variability of stratification in the estuary and associated driving mechanisms remain less understood. In this study, stratification in the Pearl River Estuary (PRE) in response to morphological change and external forcing is investigated by 3-dimensional numerical modeling. Simulation results indicate that stratification in the PRE exhibits distinct spatial and temporal variabilities. At a tidal-to-monthly time scale, variation of stratification is mainly driven by advection and straining through tidal forcing. At a monthly-to-seasonal scale, monsoon-driven river runoff and associated plume and fronts dominate the variation of stratification. Human-induced morphological change leads to an enhancement of stratification by up to four times in the PRE. Compared to an overwhelming human impact in the past few decades, future sea level rise would further enhance stratification, but to a much lesser extent than past human impacts. In addition, stratification in different areas of the estuary also responds differently to the driving factors. The western shoal of the estuary is most sensitive to changes in morphology and sea level due to its shallowness, followed by the channels and other parts of the estuary, which are less sensitive.

Owing to their composition-tunable and narrow emissions and high photoluminescence quantum yield (PLQY), inorganic halide perovskite quantum dots (IPQDs) are a promising option for wide color gamut displays. However, their practical applications have been limited by their lattice structure instability and surface defect states. Herein, CsPbBr3:KBF4@SiO2 with improved stability and optical properties is successfully synthesized with a two-step optimization of fluorine (F) anion doping and SiO2 in situ coating. Compared with bromide (Br), higher electronegativity and a smaller radius of F lead to stronger binding energy with Pb2+. Also, F anions can occupy surface Br vacancies. Then, benefiting from the acidic environment provided by BF4− hydrolysis, tetraethyl orthosilicate (TEOS) can be more easily hydrolyzed on the CsPbBr3:KBF4 surface to generate SiO2 coating, thus further passivating lattice defects and improving environmental stability. Importantly, the PLQY of CsPbBr3:KBF4@SiO2 achieves 85%, and the stability has been greatly improved compared with pure CsPbBr3. Finally, CsPbBr3:KBF4@SiO2/PDMS, CsPbI3/PDMS, and CsPbCl3/PDMS composites with narrow emissions are applied to replace traditional phosphors as color converters for direct-view light-emitting diode (LED) displays or liquid crystal display (LCD) backlights. The color gamut reaches 118.22% under the NTSC standard. Concerning the display field, it suggests likely applications in the future.

The bZIP (basic leucine zipper) proteins play crucial roles in various biological functions. Nitrogen (N) is an essential element for plant growth, especially in cucumber (Cucumis sativus) due to its shallow roots. However, the regulation of bZIP genes in cucumber nitrogen metabolism has not been studied yet. In this study, we identified a total of 72 bZIP genes (CsbZIPs) in the cucumber genome that could be classified into 13 groups. These genes were unevenly distributed on seven chromosomes, and synteny analysis showed that the CsbZIP genes were expanded in a segmentally duplicating manner. Furthermore, our genome-wide expression analysis suggested that CsbZIP genes had different patterns and that five CsbZIP genes were regulated by nitrogen treatment in both leaves and roots. Consistent with CsNPF, CsbZIP55 and CsbZIP65 were regulated by nitrogen treatment in leaves and roots. Moreover, the subcellular localization showed that CsbZIP55 and CsbZIP65 were specifically located in the nucleus, and the transcriptional activation assay showed that CsbZIP55 and CsbZIP65 have transcriptional activation activity. Additionally, in the CsbZIP55 and CsbZIP65 overexpression plants, most nitrogen-regulated CsNPF genes were downregulated. Taken together, our comprehensive analysis of the bZIP gene family lays a foundation for understanding the molecular and physiological functions of CsbZIPs.

Urban fugitive dust is a significant contributor to atmospheric PM2.5 and a potential risk to humans. In 2019, both road dust and construction dust were collected from four cities, including Xi’an, Xianyang, Baoji, and Tongchuan, in Guanzhong Plain, China. Elements, water-soluble ions, and carbonaceous fractions were determined to establish the chemical source profile. High enrichment degrees of Se, Sc, Cl, and Zn in both road dust and construction dust indicated that the industrial system and energy consumption influenced Guanzhong Plain strongly. According to the coefficient of divergence, the two datasets within Xianyang and Tongchuan were similar. Combined with the chemical profile, road dust was affected by more stationary emission sources than construction dust in Xi’an, while biomass burning and vehicle exhaust contributed more to road dust than construction dust in Baoji. Moreover, the health risk of heavy metal was assessed, and corresponding influencing factors were identified. Road dust in all cities showed a non-negligible non-carcinogenic risk for children. Ingestion and inhalation were the main exposure pathways to which As and Co contributed the most, respectively. The land-use regression model revealed that the first-class road in a 100 m radius impacted all high-risk level metals, and the commercial building material and enterprises weakly influenced Co and Pb, respectively.

Abstract Aged microglia display augmented inflammatory activity after neural injury. Although aging is a risk factor for poor outcome after brain insults, the precise impact of aging-related alterations in microglia on neural injury remains poorly understood. Microglia can be eliminated via pharmacological inhibition of the colony–stimulating factor 1 receptor (CSF1R). Upon withdrawal of CSF1R inhibitors, microglia rapidly repopulate the entire brain, leading to replacement of the microglial compartment. In this study, we investigated the impact of microglial replacement in the aged brain on neural injury using a mouse model of intracerebral hemorrhage (ICH) induced by collagenase injection. We found that replacement of microglia in the aged brain reduced neurological deficits and brain edema after ICH. Microglial replacement-induced attenuation of ICH injury was accompanied with alleviated blood-brain barrier disruption and leukocyte infiltration. Notably, newly repopulated microglia had reduced expression of IL-1β, TNF-α and CD86, and upregulation of CD206 in response to ICH. Our findings suggest that replacement of microglia in the aged brain restricts neuroinflammation and brain injury following ICH.

Urbanization not only profoundly alters landscape profiles, ecosystems and vertebrate faunal diversity but also disturbs microbial communities by increasing stochasticity, vulnerability, biotic homogenization, etc. However, because of the buffering effect of host species, microbial communities are expected to be influenced by both host species and urbanization stresses. Therefore, the impacts of urbanization on animals’ microbial symbionts could be more complex and uncertain. In this study, we quantified the urbanization degree of sampling sites and surveyed the gut and skin microbes of three amphibian host species in different sites in urban parks and nearby villages of Chengdu, Southwest China. Furthermore, a co-occurrence network analysis, the phylogenetic normalized stochasticity ratio and Sloan neutral community models were applied to infer the impact of urbanization on symbiotic microbial communities. For the three host species, urbanization increased the diversity of symbiotic microbes and the number of keystone microbial taxa. However, the negative effects of such increased diversification were evident, as the community stochasticity and co-occurrence network structure vulnerability also increased, while the network structure complexity and stability were reduced. Finally, the community stochasticity had positive associations with the network vulnerability, implying that the existence of many transient symbiotic rare microbial taxa in urban parks makes the symbiotic microbial community structure more fragile. Conclusively, urbanization increased the symbiotic microbial diversity at the cost of community stability; the results provide a new perspective for better understanding the complex triangulated environment–host–microbe relationship.

Abstract Background Acute pulmonary embolism (PE) is one of the leading causes of maternal mortality, and cesarean section is an established independent risk factor for PE. The diagnostic utility of D-dimer for PE in non-pregnant women has been well-established, but its role in women with suspected PE after cesarean section is unclear. Furthermore, the optimal threshold level in this patient population is unknown. Traditional D-dimer levels have low diagnostic specificity, resulting in many pregnant women being exposed to potentially harmful radiation despite negative diagnostic imaging results. This research aimed to optimize the clinical threshold for D-dimer to improve specificity while ensuring high sensitivity and to identify risk factors for PE after cesarean section. Methods This retrospective study of 289 women who underwent diagnostic imaging (ventilation/perfusion [V/Q] or computed tomographic pulmonary angiography [CTPA]) for suspected acute PE after cesarean delivery from 2010 to 2021 was conducted. Clinical data and laboratory indicators within 24 h postpartum including D-dimer levels were collected for analyses. Results The final analysis included 125 patients, among whom 33 were diagnosed with acute PE (incidence of 11.42%, 95% confidence interval 7.7–15.1). The receiver operating characteristic curve analysis suggested that a D-dimer cut-off value of 800 ng/mL had specificity of 25.26% and sensitivity of 100% for detecting PE. The cut-off value was adjusted to 1000 ng/mL with a specificity of 34.74% and a sensitivity of 96.67%. Using a D-dimer cut-off value of 800 ng/mL (instead of the conventional value of 500 ng/mL) increased the number of patients excluded from suspected PE from 9.6 to 18.4% without additional false-negative results. Abstract Background Extranodal NK/T-cell lymphoma of the breast (ENKTL-Breast) is rarely detected in clinical practice, and its clinicopathological features remain unclear. Results A consecutive 11-year (2010–2020) ENKTL-Breast case series was retrospectively analyzed. Eight cases of ENKTL-Breast were selected, with three primary and five secondary lesions, accounting for 3.5% (8/228) of all breast lymphomas. All patients were female with a median age of 46 years. Lesions presented as solid breast masses (8/8, 100%) and were usually located in the upper outer quadrant of the breast (7/8, 87.5%). B-symptoms were observed in seven (7/8, 87.5%) cases. Two primary ENKTL-Breast cases showed concomitant diseases (IgA nephropathy and chronic active hepatitis B). Histological and immunohistochemical features of ENKTL-Breast were similar to those of ENKTL at other sites. T-cell receptor rearrangement revealed clonality in all examined primary cases (2/2, 100%), but only in one secondary case (1/5, 20%). The disease progressed rapidly in two primary cases and both patients died within 3 and 9 months. For secondary cases, the disease manifested as a disseminated disease, with a median survival time of 6 months. Conclusions Our data suggested that ENKTL-Breast clinically mimics breast cancer to some extent, though B-symptoms might serve as a distinguishing factor. ENKTL-Breast is highly aggressive and patients with this disease exhibit a short survival time. Primary ENKTL-Breast tends to originate from activated cytotoxic T-cells, and immune-related diseases may be involved in its pathogenesis and development.

Interfacial charge separation is an essential issue to restrict catalytic activity of photocatalysts. In this work, we report a promising and easy-to-handle strategy to modulate the activity of a C60-based photocatalyst (C60@Pt) for visible-light-driven hydrogen evolution reaction (HER) with the coupling of magnetic substance that had weak ferromagnetism (Fe3O4 microbeads as an instance). It is found that Fe3O4 microbeads played a pivotal role in boosting photocatalytic activity and apparent quantum efficiency of C60@Pt for visible-light-driven HER, as the weak local magnetic field it induced around C60@Pt contributed to facilitating the separation of photo electrons from electron-donor in reaction system.

BackgroundAngioimmunoblastic T-cell lymphoma (AITL) has a rich tumor microenvironment (TME) that typically harbors plenty of CD4+tumor infiltrating lymphocytes, (TIL)-T-cells (so called common AITL). Nonetheless, AITL with large numbers of CD8+TIL-Ts that outnumber CD4+cells have been observed (CD8-predominant AITL). However, detailed comparison of CD8-predominant AITL and common AITL are still lacking.MethodsWe compared clinicopathological features, TIL subsets, TME T cell receptor-β (TRB), and immunoglobulin heavy chain (IGH) repertoires, and gene expression profiles in six CD8-predominant and 12 common AITLs using case-control matching (2014 to 2019).ResultsComparing with common AITLs, CD8-predominant AITLs showed more frequent edema (P = 0.011), effusion (P = 0.026), high elevated plasma EBV-DNA (P = 0.008), and shorter survival (P = 0.034). Moreover, they had more pronounced eosinophil increase (P = 0.004) and a higher Ki67 index (P = 0.041). Flow cytometry revealed an inverted CD4/CD8 ratio in TIL-Ts and lower TIL-B proportions (P = 0.041). TRB repertoire metrics deteriorated, including lower productive clones (P = 0.014) and higher clonality score (P = 0.019). The IGH repertoire was also narrowed, showing a higher proportion of the top 10 clones (P = 0.002) and lower entropy (P = 0.027). Gene expression analysis showed significant enrichment for upregulated negative regulation of immune system processes and downregulated T-cell activation and immune cell differentiation.ConclusionOur findings demonstrated that CD8-predominant AITL is a distinct immune pattern of AITL characterized by anti-tumor immunity impairment and an immunosuppressive microenvironment. These characteristics can interpret its severe clinical manifestations and poor outcomes.
