20. Exopeptidase combination enhances the degradation of isotopically labelled gluten immunogenic peptides in humans.

Abstract

Introduction: Celiac disease is a common autoimmune-like enteropathy caused by an aberrant response to incompletely digested dietary gluten. Gluten immunogenic peptides including the immunodominant 33-mer are thought to be resistant to proteolytic digestion by human gastrointestinal peptidases. We developed a novel enzyme therapy approach to support gluten peptide digestion using a combination of two tandem-acting exopeptidases, AMYNOPEP, that complement the intrinsic enzymatic activity of intestinal brush border enterocytes. Methods: We evaluated the effects of AMYNOPEP supplementation on 33-mer degradation in vitro and in vivo. In a cross-over clinical study, healthy volunteers with no gastrointestinal disorders were given stable isotope (SI) labelled 33-mer peptides in the presence of varying peptide substrates and caloric loads, with and without AMYNOPEP. 33-mer degradation products (SI-labelled single amino acids) were measured in the blood plasma using LC-MS/MS. Results: AMYNOPEP achieved rapid, complete amino-to-carboxyl terminal degradation of the 33-mer in vitro, generating single amino acids and dipeptides. In healthy volunteers, AMYNOPEP supplementation significantly increased 33-mer degradation and absorption of SI-labelled amino acids even in the presence of competing substrates. Specifically, we observed a 2.8-fold increase in the Cmax of stable isotope-labelled amino acids in the presence of wheat gluten. The absorption kinetics of labelled amino acids derived from 33- mer digestion with AMYNOPEP closely resembled that of SI-labelled X-Proline dipeptides administered without enzyme supplementation, highlighting the rapid hydrolytic activity of AMYNOPEP on polypeptides. Conclusions: AMYNOPEP achieved complete degradation of the 33-mer into single amino acids and dipeptides in vitro and significantly improved 33-mer degradation kinetics in healthy volunteers, as measured by labelled amino acid detection, warranting further investigation into the potential therapeutic benefits of exopeptidase combinations for patients with gluten-related health disorders including celiac disease. 32. A navigated, robot-driven laser craniotomy tool for frameless depth electrode implantation. An in-vivo recovery animal study.

Abstract

Objectives: We recently introduced a frameless, navigated, robot-driven laser tool for depth electrode implantation as an alternative to frame-based procedures. This method has only been used in cadaver and non-recovery studies. This is the first study to test the robot-driven laser tool in an in vivo recovery animal study. Methods: A preoperative computed tomography (CT) scan was conducted to plan trajectories in sheep specimens. Burr hole craniotomies were performed using a frameless, navigated, robot-driven laser tool. Depth electrodes were implanted after cut-through detection was confirmed. The electrodes were cut at the skin level postoperatively. Postoperative imaging was performed to verify accuracy. Histopathological analysis was performed on the bone, dura, and cortex samples. Results: Fourteen depth electrodes were implanted in two sheep specimens. Anesthetic protocols did not show any intraoperative irregularities. One sheep was euthanized on the same day of the procedure while the other sheep remained alive for 1 week without neurological deficits. Postoperative MRI and CT showed no intracerebral bleeding, infarction, or unintended damage. The average bone thickness was 6.2 mm (range 4.1-8.0 mm). The angulation of the planned trajectories varied from 65.5° to 87.4°. The deviation of the entry point performed by the frameless laser beam ranged from 0.27 mm to 2.24 mm. The histopathological analysis did not reveal any damage associated with the laser beam. Conclusion: The novel robot-driven laser craniotomy tool showed promising results in this first in vivo recovery study. Abstract

The USA is fast becoming a 'majority–minority' country in which Whites will no longer comprise the numerically dominant racial group. Prior studies have linked Whites' status decline to heightened in-group solidarity and the feeling that Whites, as a group, face growing discrimination. In the light of these findings, we examine the extent to which a social norm controlling anti-White prejudice is now discernible in the USA. Drawing from an original survey measuring Americans' reactions to racially-offensive speech, we examine second-order beliefs about the social inappropriateness of offensive statements targeting White Americans. We find that White Americans (in comparison to non-Whites) are indeed more likely to profess a social norm governing anti-white prejudice. The pattern is most discernible among white Republicans whom we expect to be most fearful of demographic change. Abstract

Today's modern world is characterized by an increasing shift from human-to-human interaction toward human-computer-interaction (HCI). With the implementation of artificial agents as inspectors, as can be seen in today's airports, supermarkets, or, most recently, within the context of the COVID-19 pandemic, our everyday life is progressively shaped around interacting with automated agents. While our understanding of HCI is evolving, it is still in nascent stages. This is particularly true in the sphere of non-cooperative strategic interactions between humans and automated agents, which remains largely unexplored and calls for further investigation. A deeper understanding of the factors influencing strategic decision-making processes within HCI situations, and how perceptions of automated agents' capabilities might influence these decisions, is required. This gap is addressed by extending a non-cooperative inspection-game experiment with a tax-evasion frame, implementing automated agents as inspectors. Here, a within-subject design is used to investigate (1) how HCI differs from human-to-human interactions in this context and (2) how the complexity and perceived capabilities of automated agents affect human decision-making. The results indicate significant differences in decisions to evade taxes, with participants more likely to evade taxes when they are inspected by automated agents rather than by humans. These resultsmay also be transferred to normviolationsmore generally, whichmay becomemore likely when participants are controlled by computers rather than by humans. Our results further show that participants were less likely to evade taxes when playing against an automated agent described as a complex AI, compared to an automated agent described as a simple algorithm, once they had experienced different agents. Abstract

This study examines video games' depictions of archival architectures in levels and practices of interacting with archival material such as looting. By doing this, the article proposes a processual idea of proper archives and virtual counterparts, both determined by various moments of interplay, such as opening treasure chests, boxes, or cells. Archived artifacts, hidden in games to be discovered, lure players to begin interplay, and are a crucial part of the labyrinthic archival ecosystem. Examining video game archives enhances the perception of archival ecosystems from the interplay with institutional archives, and the archived material in general.
