[Objective] This study was aimed to investigate the effects of organic manure replacing chemical fertilizer with equal nitrogen under different nitrogen application rates on the growth, yield and water-fertilizer use efficiency of spring maize in the dry zone of northern Shaanxi. [Methods] Multiple regression analysis and three-dimensional fitting models were used to establishing an optimal nitrogen management model for spring maize cultivation in northern Shaanxi. In this experiment, three nitrogen application levels (240, 180, 120 kg/hm2 for N1, N2, N3, respectively) and five organic manure replacing chemical fertilizer with equal nitrogen ratios (R0, R12.5, R25, R37.5, R50 for 100% chemical fertilizer nitrogen, 12.5% organic fertilizer nitrogen+87.5% chemical fertilizer nitrogen, 25% organic fertilizer nitrogen+75% chemical fertilizer nitrogen, 37.5% organic fertilizer nitrogen+62.5% chemical fertilizer nitrogen, 50% organic fertilizer nitrogen+50% chemical fertilizer nitrogen, respectively), a total of 15 treatments. During the main reproductive period of spring maize, spring maize growth and yield and yield components were measured, and water consumption (ET), water use efficiency (WUE), nitrogen fertilizer partial productivity (NPFP) and economic benefits were calculated. [Results] The Logistic function had a high fit for dry matter accumulation in spring maize, the R12.5 treatment delayed the start, the end and the appearance of the maximum value of the period of rapid dry matter accumulation, and the N2 treatment enhanced the maximum daily growth rate of dry matter accumulation. Irrigation, fertilization, and variety are important factors affecting potato production in northwest China. Field experiments (2021 and 2022) were performed to investigate the effects of irrigation and fertilization on the plant growth and soil microbial population of different potato varieties. Three irrigation levels were used, i.e., 100% ETc (W1), 80% ETc (W2), and 60% ETc (W3), with ETc standing for crop evapotranspiration. Three fertilization levels were used (N-P-K), i.e., 240-120-300 kg ha−1 (F1), 180-90-225 kg ha−1 (F2), and 120-60-150 kg ha−1 (F3). Three variety types were used, i.e., Feiurita (V1), Longshu 7 (V2), and Qingshu 9 (V3). These factors significantly influenced tuber yield (TY), net income (NI), and water productivity (WP). TY, NI, WP, total nitrogen accumulation (TNA), and nitrogen use efficiency (NUE) peaked at F2. Fertilization significantly impacted soil bacteria quantity (SBQ), fungi quantity (SFQ), and actinomycetes quantity (SAQ). TY, NI, SBQ, SFQ, and SAQ were highest at W2. Soil microbial population was strongly correlated with TY, NI, WP, TNA, and NUE. Comprehensively, this study suggests that irrigation that is varied from 248 to 266 mm, and fertilization (N-P-K) that is varied from 149.09-74.55-186.36 to 212.73-106.36-265.91 kg ha−1 can promote the potato industry's sustainable development and provide important references for the optimal field management of potato cultivation in northwest China. Theanine, a unique and the most abundant non-proteinogenic amino acid in tea plants, endows tea infusion with the umami taste and anti-stress effects. Its content in tea correlates highly with green tea quality. Theanine content in new shoots of tea plants is high in mid-spring and greatly decreases in late spring. However, how the decrease is regulated is largely unknown. In a genetic screening, we observed that a yeast mutant, glutamate dehydrolase 2 (gdh2), was hypersensitive to 40 mM theanine and accumulated more theanine. This result implied a role of CsGDH2s in theanine accumulation in tea plants. Therefore, we identified the two homologs of GDH2, CsGDH2.1 and CsGDH2.2, in tea plants. Yeast complementation assay showed that the expression of CsGDH2.1 in yeast gdh2 mutant rescued the theanine hypersensitivity and hyperaccumulation of this mutant. Subcellular localization and tissue-specific expression showed CsGDH2.1 localized in the mitochondria and highly expressed in young tissues. Importantly, CsGDH2.1 expression was low in early spring, and increased significantly in late spring, in the new shoots of tea plants. These results all support the idea that CsGDH2.1 regulates theanine accumulation in the new shoots. Moreover, the in vitro enzyme assay showed that CsGDH2.1 had glutamate catabolic activity, and knockdown of CsGDH2.1 expression increased glutamate and theanine accumulation in the new shoots of tea plants. These findings suggested that CsGDH2.1-mediated glutamate catabolism negatively regulates theanine accumulation in the new shoots in late spring, and provides a functional gene for improving late-spring green tea quality. Simple Summary: Epigenetic modification plays an important regulatory role in breast cancer progression. However, the relationship between epigenetic modification with tumor metabolism has not yet been fully elucidated. PDK1, as a key enzyme in glucose metabolism, mediates multiple tumors development. But, the mechanism of epigenetic modification in regulating PDK1 remains unclear in breast cancer. Here, we demonstrated that HDAC2 and EZH2 upregulated PDK1 expression through inhibiting miR-148a. Importantly, miR-148a targeting PDK1 regulated breast cancer cells glycolysis, invasion, epithelial-mesenchymal transition (EMT) and Adriamycin resistance. Our results suggested that the HDAC2/EZH2/miR-148a/PDK1 axis may be a promising potential therapeutic strategy. Background: Breast cancer has one of highest morbidity and mortality rates for women. Abnormalities regarding epigenetics modification and pyruvate dehydrogenase kinase 1 (PDK1)-induced unusual metabolism contribute to breast cancer progression and chemotherapy resistance. However, the role and mechanism of epigenetic change in regulating PDK1 in breast cancer remains to be elucidated. Methods: Gene set enrichment analysis (GSEA) and Pearson's correlation analysis were performed to analyze the relationship between histone deacetylase 2 (HDAC2), enhancer of zeste homologue 2 (EZH2), and PDK1 in database and human breast cancer tissues. Dual luciferase reporters were used to test the regulation between PDK1 and miR-148a. HDAC2 and EZH2 were found to regulate miR-148a expression through Western blotting assays, qRT-PCR and co-immunoprecipitation assays. The effects of PDK1 and miR-148a in breast cancer were investigated by immunofluorescence (IF) assay, Transwell assay and flow cytometry assay. The roles of miR-148a/PDK1 in tumor growth were investigated in vivo. Results: We found that PDK1 expression was upregulated by epigenetic alterations mediated by HDAC2 and EZH2. At the post-transcriptional level, PDK1 was a new direct target of miR-148a and was upregulated in breast cancer cells due to miR-148a suppression. PDK1 overexpression partly reversed the biological function of miR-148a—including miR-148a's ability to increase cell sensitivity to Adriamycin (ADR) treatment—inhibiting cell glycolysis, invasion and epithelial–mesenchymal transition (EMT), and inducing apoptosis and repressing tumor growth. Furthermore, we identified a novel mechanism: DNMT1 directly bound to EZH2 and recruited EZH2 and HDAC2 complexes to the promoter region of miR-148a, leading to miR-148a downregulation. In breast cancer tissues, HDAC2 and EZH2 protein expression levels also were inversely correlated with levels of miR-148a expression. Conclusion: Our study found a new regulatory mechanism in which EZH2 and HDAC2 mediate PDK1 upregulation by silencing miR-148a expression to regulate cancer development and Adriamycin resistance. These new findings suggest that the HDAC2/EZH2/miR-148a/PDK1 axis is a novel mechanism for regulating cancer development and is a potentially promising target for therapeutic options in the future. Kiwifruit contains abundant nutritive compounds and is highly favored by the consumers worldwide. Therefore, detailed metabolic profiling is important to provide theoretic basis for the improvement of kiwifruit quality. In this study, the levels of volatiles, carotenoids, and mineral elements in the flesh of 17 kiwifruit accessions were evaluated. Acids and esters were the main volatiles in kiwifruit. During these 17 kiwifruit accessions, "Chenhong," three "Jinyan," and two "Guichang" germplasms were specifically rich in aromatic esters, which might be associated with their special taste. The main carotenoids were lutein, β‐carotene, and zeaxanthin, and their levels were also genotype specific, with the green‐fleshed "Guichang" having the highest level of carotenoids, and red‐fleshed "Fuhong" and "Chenhong" being rich in zeaxanthin. Partial correlation analysis showed that the contents of some mineral elements were significantly correlated with those of specific volatiles and carotenoids, indicating the impacts of mineral elements on the accumulation of volatiles and carotenoids in the kiwifruit flesh. These results indicated that the contents of carotenoids and volatiles seemed to be affected by mineral elements and also provided a new potential method for improving fruit flavor quality in production. The prevalence of nonalcoholic fatty liver disease (NAFLD) increases with increasing body mass index (BMI). However, approximately 40-50% of obese adults do not develop hepatic steatosis. The level of inflammatory biomarkers is higher in obese subjects with NAFLD compared to BMI-matched subjects without hepatic steatosis. We used a casein injection in high-fat diet (HFD)-fed C57BL/6J mice to induce inflammatory stress. Although mice on a HFD exhibited apparent phenotypes of obesity and hyperlipidemia regardless of exposure to casein injection, only the HFD+Casein mice showed increased hepatic vacuolar degeneration accompanied with elevated inflammatory cytokines in the liver and serum, compared to mice on a normal chow diet. The expression of genes related to hepatic fatty acid synthesis and oxidation were upregulated in the HFD-only mice. The casein injection further increased baseline levels of lipogenic genes and decreased the levels of oxidative genes in HFD-only mice. Inflammatory stress induced both oxidative stress and endoplasmic reticulum stress in HFD-fed mice livers. We conclude that chronic inflammation precedes hepatic steatosis by disrupting the balance between fatty acid synthesis and oxidation in the livers of HFD-fed obese mice. This mechanism may operate in obese individuals with chronic inflammation, thus making them more prone to NAFLD. [ABSTRACT FROM AUTHOR]
