ABSTRACT Background Although pharmacological effects of SGLT2 inhibitors on the development of frailty and sarcopenia were known, the role of SGLT1 remained less clear. The present study investigated the possible effect of SGLT1 inhibition on these conditions and explored potential mediators. Methods A two‐sample Mendelian randomization (MR) analysis was performed to assess the effect of SGLT1 inhibition on frailty index (FI) and low grip strength in individuals aged 60 years and older using both the FNIH and EWGSOP criteria. Subsequently, a two‐step MR analysis was conducted to investigate the mediating role of insulin resistance phenotype and identify potential mediators of the effect of SGLT1 inhibition on the FI and low grip strength from 1558 plasma proteins and 1352 metabolites. Results Genetically predicted SGLT1 inhibition was associated with decreased FI (β: −0.290 [95% CI: −0.399, −0.181]) and reduced risk of low grip strength in individuals aged 60 years and older under both FNIH (β: −0.796 [95% CI: −1.216, −0.376]) and EWGSOP criteria (β: −0.287 [95% CI: −0.532, −0.041]). The two‐step MR analysis demonstrated the role of insulin resistance phenotype in mediating SGTL1 inhibition on alleviating frailty (mediation proportion = 19.56% [95% CI: 8.42%, 30.70%]). After screening, 24 proteins and 16 metabolites were identified as mediators of the impact of SGLT1 inhibition on FI. Additionally, 13 proteins and 16 metabolites were found to mediate the effect of SGLT1 inhibition on low grip strength according to FNIH criteria while 22 proteins and 6 metabolites were shown to mediate the impact of SGLT1 inhibition on low grip strength under EWGSOP criteria. Conclusions SGLT1 inhibition potentially mitigated frailty and sarcopenia through several biological mediators, shedding new light for therapeutic intervention.

Abstract Background Previous studies have established a correlation between the pathogenesis of oxidative stress and sarcopenia. The Oxidative Balance Score (OBS) is an integrated measure that reflects the overall balance of antioxidants and pro-oxidants in dietary components and lifestyle. However, there are limited reports on the association between OBS and lean mass and the impact of protein intake on the association between OBS and lean mass. Methods Using data from the National Health and Nutrition Examination Survey from 2011 to 2018, multivariate linear and logistic regression analyses were conducted to explore the associations between OBS and outcomes. The findings were then illustrated through fitted smoothing curves and threshold effect analyses. Results This study included 2,441 participants, demonstrating that higher OBS is significantly associated with an increased ratio of appendicular lean mass to body mass index. Key inflection points at OBS 31 mark pronounced changes in these associations, with age and protein intake notably affecting the association. The effect of OBS on lean mass varies among populations with high and low protein intake. Conclusions Our findings suggest that OBS is significantly and positively associated with lean mass. A high protein intake of more than 84.5 g/day may enhance the role of OBS in influencing muscle health to improve muscle outcomes.

Abstract Background The ratio between non-high-density lipoprotein cholesterol and high-density lipoprotein cholesterol (NHHR) is a reliable marker for assessing the risk linked to lipid metabolism disorders. Sarcopenia, characterized by age-related loss of muscle mass and strength/function, includes the assessment of muscle mass, muscle strength, and muscle-specific strength. However, research into NHHR’s relationship with low muscle mass risk remains unexplored. Methods Our study utilized a cross-sectional approach, examining data derived from the National Health and Nutrition Examination Survey (NHANES) from 2011 to 2018. Through multivariable linear and logistic regression, we investigated the relationships of the NHHR with muscle mass and low muscle mass. We visualized the results using smoothing curves and assessed threshold effects. We also performed various subgroup and sensitivity analyses. Results This research encompassed 9,012 participants and demonstrated significant nonlinear associations between NHHR and ALMBMI or low muscle mass risk in a generalized additive model (GAM), pinpointing critical NHHR values (3.328 and 3.367) where changes in NHHR significantly impacted ALMBMI and low muscle mass risk. Conclusions The NHHR demonstrates a significant association with an increased risk of low muscle mass among middle-aged Americans. This ratio has potential as a predictive marker for low muscle mass. Further exploration of NHHR is expected to aid in advancing preventive and therapeutic measures for this condition.

Abstract The photocatalytic reduction of CO2 represents an environmentally friendly and sustainable approach for generating valuable chemicals. In this study, a thiophene‐modified highly conjugated asymmetric covalent triazine framework (As‐CTF‐S) is developed for this purpose. Significantly, single‐component intramolecular energy transfer can enhance the photogenerated charge separation, leading to the efficient conversion of CO2 to CO during photocatalysis. As a result, without the need for additional photosensitizers or organic sacrificial agents, As‐CTF‐S demonstrates the highest photocatalytic ability of 353.2 µmol g−1 and achieves a selectivity of ≈99.95% within a 4 h period under visible light irradiation. This study provides molecular insights into the rational control of charge transfer pathways for high‐efficiency CO2 photoreduction using single‐component organic semiconductor catalysts.

Abstract Background The interplay between physical activity (PA) and air pollution in relation to type 2 diabetes (T2D) remains largely unknown. Based on a large population-based cohort study, this study aimed to examine whether the benefits of PA with respect to the risk of T2D are moderated by exposure to air pollution. Methods UK Biobank participants (n = 359,153) without diabetes at baseline were included. Information on PA was obtained using the International Physical Activity Questionnaire short form. Exposure to air pollution, including PM2.5, PMcoarse (PM2.5−10), PM10, and NO2, was estimated from land use regression models. Cox regression models were used to estimate the hazard ratios (HRs) and 95% confidence intervals (95% CIs). Results During a median of 8.9 years of follow-up, 13,706 T2D events were recorded. Compared with a low PA level, the HRs for the risk of T2D among individuals with moderate and high PA were 0.82 (95% CI, 0.79–0.86) and 0.73 (95% CI, 0.70–0.77), respectively. Compared with low levels of air pollution, the HRs for risk of T2D for high levels of air pollution (PM2.5, PMcoarse, PM10, and NO2) were 1.19 (1.14–1.24), 1.06 (1.02–1.11), 1.13 (1.08–1.18), and 1.19 (1.14–1.24), respectively. There was no effect modification of the associations between PA and T2D by air pollution (all P-interactions > 0.05). Clean and efficient fire extinguishing technology has been a hot research topic in fire science. The research of acoustic extinguishing technology originates from the discovery that the different acoustic modes of combustion noise can lead to an unstable oscillation of the flame and local flame extinction. Recently, acoustic extinguishing technology has gradually entered the research field of scholars because it is clean and exhibits no secondary pollution. To study the fire extinguishing mechanism and analyze the specific control behavior of the acoustic wave on an unclosed flame, the flame shapes and the combustion characteristics of the pool flame with 3, 4, and 5 cm diameters under a 30–90 Hz acoustic force were analyzed. The experimental system includes a high-speed camera, signal generator, power amplifier, loudspeaker, and acoustic signal analysis device. The flame image analysis shows that the transverse sound wave intensifies the unsteady flow of the vortex, and the flame shape under an acoustic force could be divided into three types of state: intermittent, deflective, and stable. The numerical analysis of the flame geometry shows that the flame surface is highly twisted and wrinkled under the intermittent and deflective states with a higher fractal dimension. The frequency-domain signal analysis of the flame area, height, and width shows that the flame signal is very unstable in the intermittent state, and the peak frequency domain is concentrated in the range of 0–10 Hz. The acoustic frequency is always prominent in the frequency distribution of the flame width signal. Based on the relationship between the flame inclination angle and Richardson number, the form of the latter under the action of the acoustic wave was proposed. In the 50–70 Hz range, the response of the flame to the acoustic frequency was particularly considerable, and there may be a marginal effect when the acoustic frequency is higher or lower than the said range. The critical Ria−1 of the intermittent and deflective states are 10.32 and 2.92, respectively.

Light heterogeneity leads to anatomically and physiologically heterogeneous features in leaves. However, little attention has been paid to the effects of nonuniform illumination on the anatomical and photosynthetic performance on both sides along the leaf main vein. This study explored such effects by combining in situ determination in the field with shading simulation in the phytotron, on pima cotton that has cupping leaves. Photosynthetic characteristics and morphological structures were measured in the field on both sides along the main vein of eastward, westward, southward, and northward leaves. The results showed that the difference in photosynthetic capacity between the two sides along the main vein in different directions was closely related to the daily photo irridiance (DPI). This result indicates that the photosynthetic heterogeneity between the two sides is related to their intercepted light energy. The conclusion was further verified by the shading simulation experiments. Photosynthetic capacity and leaf thickness of the unshaded sides of leaves in the half-shaded treatment decreased, compared to those in the unshaded treatment. Therefore, it is conjectured that the development of photosynthetic characteristics on one side is systematically regulated by that on the other side. The study provides theoretical guidance on accessing the feasibility of sampling and directional planting.

Abstract Background To investigate whether the mitochondrial transcription factor A (TFAM) rs1937 single nucleotide polymorphism (SNP) is associated with longevity. Methods We conducted a case-control study among Chinese long-lived individuals (≥90 years). Data were obtained on 3294 participants who were able to voluntarily provided a saliva sample during 2008–2009 from the Chinese Longitudinal Healthy Longevity Survey (CLHLS). In this study, 1387 young elderly (65–74 years) were allocated to the control group, and 1907 long-lived individuals were recruited as the case group. SNP rs1937 on TFAM were genotyped. Logistic regression models were applied to evaluate the association between rs1937 SNP and longevity. Results The genotype frequency of the SNP of rs1937 in the two groups had a significant difference (p = 0.003). Binary logistic regression analysis showed that compared to younger elderly, the long-lived individuals with “CC genotype” of rs1937 were more closely related to increased longevity than those with “GG genotype” (OR: 1.989, 95% CI: 1.160–3.411). The positive association between rs1937 SNP and longevity was robust in stratified analyses and sensitivity analyses. Conclusions We found the SNP of rs1937 may be a potential biomarker for longer human life span. Further studies are necessary to elucidate the biological mechanism of rs1937 on TFAM with promoting longevity.

Secreted frizzled-related protein 5 (SFRP5) plays a pivotal role in regulating the development of many tissues and organs, however, as an inhibitor of Wnt signaling, the role of SFRP5 in vitiligo remains unknown. Hence, we speculated that SFRP5 might be associated with melanogenesis in melanocytes by regulating Wnt signaling in vitiligo. In this study, we found that SFRP5 was overexpressed in the skin lesions of patients with vitiligo. Compared with that in normal epidermal melanocytes (PIG1), the expression of SFRP5 was increased in vitiligo melanocytes (PIG3V). To investigate the effect of SFRP5 on melanin synthesis, PIG1 cells were infected with recombinant SFRP5 adenovirus (AdSFRP5), and PIG3V cells were infected with recombinant siSFRP5 adenovirus (AdsiSFRP5). The results showed that SFRP5 overexpression inhibited melanin synthesis in PIG1 cells through downregulation of microphthalmia-associated transcription factor (MITF) and its target proteins via suppression of the Wnt/β-catenin signaling pathway. Accordingly, SFRP5 silencing increased melanin synthesis and activated the Wnt signaling pathway in PIG3V cells. Moreover, SFRP5 overexpression also downregulated the transcriptional activity of T cell factor/lymphoid enhancer factor (TCF/LEF) in PIG1 cells. Furthermore, this inhibitory effect of SFRP5 on melanin synthesis was reversed by treatment with the β-catenin agonist, SKL2001. The inhibitory action of SFRP5 in pigmentation was further confirmed in vivo using a nude mouse model. Hence, our results indicate that SFRP5 can inhibit melanogenesis in melanocytes. Additionally, our findings showed that SFRP5 plays a vital role in the development of vitiligo, and thus may serve as a potential therapeutic target for vitiligo.

Abstract Background Candidate genes of neuromuscular junction (NMJ) pathway increased risk of frailty, but the extent and whether can be offset by exercises was unclear. The aim of this study was to investigate the association between aerobic exercises and incident frailty regardless of NMJ pathway‐related genetic risk. Methods A cohort study on participants from Chinese Longitudinal Healthy Longevity Survey was conducted from 2008 to 2011. A total of 7006 participants (mean age of 80.6 ± 10.3 years) without frailty at baseline were interviewed to record aerobic exercise status, and 4053 individuals among them submitted saliva samples. NMJ pathway‐related genes were genotyped and weighted genetic risk scores were constructed. Results During a median follow‐up of 3.1 years (19 634 person‐years), there were 1345 cases (19.2%) of incident frailty. Persistent aerobic exercises were associated with a 26% lesser frailty risk [adjusted hazard ratio (HR) = 0.74, 95% confidence interval (CI) = 0.64–0.85]. This association was stronger in a subgroup of 1552 longevous participants (age between 90 and 111 years, adjusted HR = 0.72, 95% CI = 0.60–0.87). High genetic risk was associated with a 35% increased risk of frailty (adjusted HR = 1.35, 95% CI = 1.16–1.58). Of the participants with high genetic risk and no persistent aerobic exercises, there was a 59% increased risk of frailty (adjusted HR = 1.59, 95% CI = 1.20–2.09). Abstract Background Heat shock transcription factors (Hsfs) are present in majority of plants and play central roles in thermotolerance, transgenerational thermomemory, and many other stress responses. Our previous paper identified at least 82 Hsf members in a genome-wide study on wheat (Triticum aestivum L.). In this study, we analyzed the Hsf expression profiles in the advanced development stages of wheat, isolated the markedly heat-responsive gene TaHsfA2–10 (GenBank accession number MK922287), and characterized this gene and its role in thermotolerance regulation in seedlings of Arabidopsis thaliana (L. Heynh.). Results In the advanced development stages, wheat Hsf family transcription profiles exhibit different expression patterns and varying heat-responses in leaves and roots, and Hsfs are constitutively expressed to different degrees under the normal growth conditions. Overall, the majority of group A and B Hsfs are expressed in leaves while group C Hsfs are expressed at higher levels in roots. The expression of a few Hsf genes could not be detected. Heat shock (HS) caused upregulation about a quarter of genes in leaves and roots, while a number of genes were downregulated in response to HS. The highly heat-responsive gene TaHsfA2–10 was isolated through homeologous cloning. qRT-PCR revealed that TaHsfA2–10 is expressed in a wide range of tissues and organs of different development stages of wheat under the normal growth conditions. Compared to non-stress treatment, TaHsfA2–10 was highly upregulated in response to HS, H2O2, and salicylic acid (SA), and was downregulated by abscisic acid (ABA) treatment in two-leaf-old seedlings. Transient transfection of tobacco epidermal cells revealed subcellular localization of TaHsfA2–10 in the nucleus under the normal growth conditions. Phenotypic observation indicated that TaHsfA2–10 could improve both basal thermotolerance and acquired thermotolerance of transgenic Arabidopsis thaliana seedlings and rescue the thermotolerance defect of the T-DNA insertion mutant athsfa2 during HS. Compared to wild type (WT) seedlings, the TaHsfA2–10-overexpressing lines displayed both higher chlorophyll contents and higher survival rates. Yeast one-hybrid assay results revealed that TaHsfA2–10 had transactivation activity. The expression levels of thermotolerance-related AtHsps in the TaHsfA2–10 transgeinc Arabidopsis thaliana were higher than those in WT after HS. Conclusions Wheat Hsf family members exhibit diversification and specificity of transcription expression patterns in advanced development stages under the normal conditions and after HS. As a markedly responsive transcriptional factor to HS, SA and H2O2, TaHsfA2–10 involves in thermotolerance regulation of plants through binding to the HS responsive element in promoter domain of relative Hsps and upregulating the expression of Hsp genes.

Abstract Increasing damage of pests in agriculture and forestry can arise both as a consequence of changes in local species and through the introduction of alien species. In this study, we used population genetics approaches to examine population processes of two pests of the tree‐of‐heaven trunk weevil (TTW), Eucryptorrhynchus brandti (Harold) and the tree‐of‐heaven root weevil (TRW), E. scrobiculatus (Motschulsky) on the tree‐of‐heaven across their native range of China. We analyzed the population genetics of the two weevils based on ten highly polymorphic microsatellite markers. Population genetic diversity analysis showed strong population differentiation among populations of each species, with FST ranges from 0.0197 to 0.6650 and from −0.0724 to 0.6845, respectively. Populations from the same geographic areas can be divided into different genetic clusters, and the same genetic cluster contained populations from different geographic populations, pointing to dispersal of the weevils possibly being human‐mediated. Redundancy analysis showed that the independent effects of environment and geography could account for 93.94% and 29.70% of the explained genetic variance in TTW, and 41.90% and 55.73% of the explained genetic variance in TRW, respectively, indicating possible impacts of local climates on population genetic differentiation. Our study helps to uncover population genetic processes of these local pest species with relevance to control methods.

Since impacts of rail transit on urban industries and space most obviously appear in metro station areas, correlations between the spatial distribution of producer services and urban built environment in metro station areas are studied in Xi’an, China. At first, the scope determining methods of a metro station area were separately proposed for both single-line station and multiline transfer station based on their construction and transportation modes. Then, when the producer services were divided into 6 categories of business, POI numbers of enterprises of each category in metro station areas were collected and weighted to calculate the distribution intensity for categories. Finally, on condition that the urban built environment of a metro station area included job and residence space, outdoor activity space, and municipal road space, correlation performances between the distribution intensity of categories and indexes representing 3 space elements of the built environment were calculated through second-order partial correlation analysis, while the corresponding mechanisms of correlations were explained too. The main conclusions of this paper indicate that there are significant positive correlations between the distribution intensity and the diversity index of job and residence space for most categories, whereas significant negative correlations between it and the openness index of outdoor activity space for most categories. Moreover, positive but weak correlations were appeared between the distribution intensity and the street network density index of municipal road space only for categories of intermediary consulting and leasing.

This study aims to develop a high-speed and nondestructive mildewed rice grain detection method. First, a set of microscopic images of rice grains contaminated by Aspergillus niger, Penicillium citrinum, and Aspergillus cinerea are acquired to serve as samples, and the mildewed regions are marked. Then, three YOLO-v5 models for identifying regions of rice grain with contamination of Aspergillus niger, Penicillium citrinum, and Aspergillus cinerea in microscopic images are established. Finally, the relationship between the proportion of mildewed regions and the total number of colonies is analyzed. The results show that the proposed YOLO-v5 models achieve accuracy levels of 89.26%, 91.15%, and 90.19% when detecting mildewed regions with contamination of Aspergillus niger, Penicillium citrinum, and Aspergillus cinerea in the microscopic images of the verification set. The proportion of the mildewed region area of rice grain with contamination of Aspergillus niger/Penicillium citrinum/Aspergillus cinerea is logarithmically correlated with the logarithm of the total number of colonies (TVC). The corresponding determination coefficients are 0.7466, 0.7587, and 0.8148, respectively. This study provides a reference for future research on high-speed mildewed rice grain detection methods based on MCV technology.

Abstract Prompt double-$$J/\psi $$ J/ψ production at high-energy hadron colliders can be considered as a golden channel to probe double parton scatterings (DPS)—in particular to study gluon–gluon correlations inside the proton—and, at the same time, to measure the distribution of linearly-polarised gluons inside the proton. Such studies, however, require a good control of both single parton scatterings (SPS) and DPS in the respective regions where they are carried out. In this context, we have critically examined two mechanisms of SPS that may be kinematically enhanced where DPS are thought to be dominant, even though they are either at higher orders in the strong-coupling or velocity expansion. First, we have considered a gauge-invariant and infrared-safe subset of the loop-induced contribution via colour-singlet (CS) transitions. We have found it to become the leading CS SPS contributions at large rapidity separation, yet too small to account for the data without invoking the presence of DPS yields. Second, we have surveyed the possible colour-octet (CO) contributions using both old and up-to-date non-perturbative long-distance matrix elements (LDMEs). We have found that the pure CO yields crucially depend on the LDMEs. Among all the LDMEs we used, only two result into a visible modification of the NRQCD (CS+CO) yield, but only in two kinematical distributions measured by ATLAS, those of the rapidity separation and of the pair invariant mass. These modifications, however, do not impact the control region used for their DPS study.

Abstract Ubiquitin‐specific protease 44 (USP44) has been reported as a tumor suppressor or promoter in some tumors, but its function in non‐small cell lung cancer (NSCLC) is still unclear. In this study, USP44 was found significantly downregulated in both of NSCLC tissues and cell lines, and low expression of USP44 predicted a poor prognosis for NSCLC patients. Overexpression of USP44 markedly downregulated the expression levels of Cyclin D1 and CDK4, but upregulated p53 expression, as a result of which, suppressing the cell growth of NSCLC cells. Further studies indicated that overexpression of USP44 significantly inhibited the phosphorylation of AKT, and its down‐stream signals, including mTOR and P70S6K. Moreover, overexpression of USP44 increased PTEN protein but not its mRNA levels, which suggested that USP44 inhibited AKT signaling by stabilizing PTEN in NSCLC cells. In conclusion, we demonstrated that USP44 showed prior evidence of a tumor suppressive function in NSCLC cells, and inhibited NSCLC cell growth by suppressing AKT signaling, suggesting that USP44 could be as a novel target for NSCLC therapy.

Abstract. Background:. Low handgrip strength (HS) and declining gait speed (GS) are increasingly obvious with aging, requiring effective, and safe medication for treatment. Trimetazidine (TMZ) modified release tablets, a common anti-angina drug, has potential benefits for alleviating the condition, but this has not yet been fully studied and therefore is the aim of this study. Methods:. This is a prospective randomized controlled study. Fifty-eight eligible patients will be randomly assigned to one of two study groups: TMZ group or control group. For the TMZ group, a dose of 35 mg of oral TMZ will be administered with a meal twice a day for 3 months, in addition to any conventional treatments for angina. Only conventional treatments for angina will be administrated in the control group. The primary outcome will be the 6-min walking distance and the secondary outcomes will be: muscle strength (HS and pinch strength), GS, muscle endurance (five times sit-to-stand test), balance maintenance (tandem standing test), and the frequency of angina per week. Additionally, body mass index, circumferences (biceps, waist, hip, and calf), albumin levels, and the score on a five-question scale for sarcopenia will be obtained during the study. Discussion:. This study aims to evaluate the usefulness of TMZ in a population with poor muscle function. The results may provide an effective and safe medical treatment to people with low muscle strength or physical performance. Trial registration:. N6-methyladenosine (m6A), catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14, is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However, the roles and precise mechanisms of m6A modification in regulating neuronal development and adult neurogenesis remain unclear. Here, we examined the function of Mettl3, the key component of the complex, in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced m6A levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neuronal development and skewed the differentiation of aNSCs more toward glial lineage, but also affected the morphological maturation of newborn neurons in the adult brain. m6A immunoprecipitation combined with deep sequencing (MeRIP-seq) revealed that m6A was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically, m6A was present on the transcripts of histone methyltransferase Ezh2, and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. Collectively, our results uncover a crosstalk between RNA and histone modifications and indicate that Mettl3-mediated m6A modification plays an important role in regulating neurogenesis and neuronal development through modulating Ezh2. We study the Single-Parton-Scattering (SPS) production of double quarkonia (J/ψ+J/ψ, J/ψ+ϒ, and ϒ+ϒ) in pp and pp¯ collisions at the LHC and the Tevatron as measured by the CMS, ATLAS, LHCb, and D0 experiments in the Colour-Evaporation Model (CEM), based on the quark-hadron duality, including Next-to-Leading Order (NLO) QCD corrections up to αs5. To do so, we also perform the first true NLO –up to αs4– study of the pT-differential cross section for single-quarkonium production. This allows us to fix the non-perturbative CEM parameters at NLO accuracy in the region where quarkonium-pair data are measured. Our results show that the CEM at NLO in general significantly undershoots these experimental data and, in view of the other existing SPS studies, confirm the need for Double Parton Scattering (DPS) to account for the data. Our NLO study of single-quarkonium production at mid and large pT also confirms the difficulty of the approach to account for the measured pT spectra; this is reminiscent of the impossibility to fit single-quarkonium data with the sole S1[8]3 NRQCD contribution from gluon fragmentation. We stress that the discrepancy occurs in a kinematical region where the new features of the improved CEM are not relevant.

Abstract In this paper, the color-octet long distance matrix elements are studied through the inclusive $$J/\psi $$ J / ψ production in $$e^{+}e^{-}$$ e + e - annihilation within the framework of non-relativistic QCD factorization. The calculations are up-to next-to-leading order with the radiative and relativistic corrections in the energy region of the B-factory and the near-threshold region of $$4.6{-}5.6~\mathrm{GeV}$$ 4.6 - 5.6 GeV . A constraint of the long distance matrix elements ( $$\langle ^1S_0^{8}\rangle $$ ⟨ 1 S 0 8 ⟩ , $$\langle ^3P_0^{8}\rangle $$ ⟨ 3 P 0 8 ⟩ ) is obtained. Through our estimation, the P-wave color-octet matrix element $$(\langle 0|^3P^8_0|0\rangle )$$ ( ⟨ 0 | 3 P 0 8 | 0 ⟩ ) should be of the order of $$0.008m_\mathrm{c}^2~\mathrm{GeV}^3$$ 0.008 m c 2 GeV 3 or less. The constrained region is not compatible with the values of the long distance matrix elements fitted at hadron colliders.

In this study, we have sequenced and annotated the complete mitochondrial genome (mitogenome) of Hemisphaeriusrufovarius (Hemiptera: Fulgoroidea: Issidae) for the first time,the mitogenome is 15,955 bp (GenBankNo. MT210096), includes13 PCGs, 2 rRNAs, 22 tRNAs and one putative control region (D-loop). The AT content of this mitogenomeis 78.3% (A 47.7%,T30.6%, C 13.3%, and G 8.4%). Most the PCGs started with ATN or TTG(nad5), and ended with TANor single T. The result ofPhylogenetic tree showed a close relationship among the families Issidae, Flatidae and Ricaniidae.

Tumor cell migration is a critical step in cancer metastasis. Over-activated Notch pathway can promote the migration of cancer cells, especially in the breast cancer. However, the underlying mechanism of non-canonical Notch signaling in modulating the migration has not yet been clearly characterized. Here we demonstrated that DAPT, a gamma secretase inhibitor, inhibited protrusion formation and cell motility, and then reduced the migration of triple-negative breast cancer cells, through increasing the activity of Cdc42 by non-canonical Notch pathway. Phosphorylation of AKT on S473 was surprisingly increased when Notch signaling was inhibited by DAPT. Inhibition of PI3K and AKT by LY294002 and MK2206, respectively, or knockdown of AKT expression by siRNA blocked DAPT-induced activation of Cdc42. Moreover, immunofluorescence staining further showed that DAPT treatment reduced the formation of lamellipodia and induced actin cytoskeleton remodeling. Taken together, these results indicated that DAPT inhibited Notch signaling and consequently activated PI3K/AKT/Cdc42 signaling by non-canonical pathway, facilitated the formation of filopodia and inhibited the assembly of lamellipodia, and finally resulted in the decrease of migration activity of breast cancer cells.

Overproduction of oxidants (reactive oxygen species and reactive nitrogen species) in the human body is responsible for the pathogenesis of some diseases. The scavenging of these oxidants is thought to be an effective measure to depress the level of oxidative stress of organisms. It has been reported that intake of vegetables and fruits is inversely associated with the risk of many chronic diseases, and antioxidant phytochemicals in vegetables and fruits are considered to be responsible for these health benefits. Antioxidant phytochemicals can be found in many foods and medicinal plants, and play an important role in the prevention and treatment of chronic diseases caused by oxidative stress. They often possess strong antioxidant and free radical scavenging abilities, as well as anti-inflammatory action, which are also the basis of other bioactivities and health benefits, such as anticancer, anti-aging, and protective action for cardiovascular diseases, diabetes mellitus, obesity and neurodegenerative diseases. This review summarizes recent progress on the health benefits of antioxidant phytochemicals, and discusses their potential mechanisms in the prevention and treatment of chronic diseases.

Melanocyte loss in vitiligo vulgaris is believed to be an autoimmune process. Macrophage migration inhibitory factor (MIF) is involved in many autoimmune skin diseases. We determined the possible role of MIF in the pathogenesis of vitiligo vulgaris, and describe the relationship between MIF expressions and disease severity and activity. Serum MIF concentrations and mRNA levels in PBMCs were measured in 44 vitiligo vulgaris patients and 32 normal controls, using ELISA and real-time RT-PCR. Skin biopsies from 15 patients and 6 controls were analyzed by real-time RT-PCR. Values are reported as median (25th-75th percentile). Serum MIF concentrations were significantly increased in patients [35.81 (10.98-43.66) ng/mL] compared to controls [7.69 (6.01-9.03) ng/mL]. MIF mRNA levels were significantly higher in PBMCs from patients [7.17 (3.59-8.87)] than controls [1.67 (1.23-2.42)]. There was also a significant difference in MIF mRNA levels in PBMCs between progressive and stable patients [7.86 (5.85-9.13) vs 4.33 (2.23-8.39)] and in serum MIF concentrations [40.47 (27.71-46.79) vs 26.80 (10.55-36.07) ng/mL]. In addition, the vitiligo area severity index scores of patients correlated positively with changes of both serum MIF concentrations (r = 0.488) and MIF mRNA levels in PBMCs (r = 0.426). MIF mRNA levels were significantly higher in lesional than in normal skin [2.43 (2.13-7.59) vs 1.18 (0.94-1.83)] and in patients in the progressive stage than in the stable stage [7.52 (2.43-8.84) vs 2.13 (1.98-2.64)]. These correlations suggest that MIF participates in the pathogenesis of vitiligo vulgaris and may be useful as an index of disease severity and activity.

Alcoholic beverages such as beer, wine and spirits are widely consumed around the world. However, alcohol and its metabolite acetaldehyde are toxic and harmful to human beings. Chronic alcohol use disorder or occasional binge drinking can cause a wide range of health problems, such as hangover, liver damage and cancer. Some natural products such as traditional herbs, fruits, and vegetables might be potential dietary supplements or medicinal products for the prevention and treatment of the problems caused by excessive alcohol consumption. The aim of this review is to provide an overview of effective natural products for the prevention and treatment of hangover and alcohol use disorder, and special emphasis is paid to the possible functional component(s) and related mechanism(s) of action.

Background. Lumbar disc herniation (LDH) is one of the most common diseases and is a global medical and socioeconomic problem characterized by leg or back pain, weakness in the lower extremities and paresthesia. Objectives. A multicenter, randomized, double-blinded, parallel, positive-controlled clinical trial was conducted to evaluate the efficacy and safety of Yaobitong capsules (YBT) for LDH. Materials and methods. Patients (n = 479) were recruited and randomized into YBT and Jingyaokang capsule (JYK) groups (the positive control), and received YBT or JYK at a dose of 3 capsules 3 times per day after a meal for 30 days. The primary efficacy outcome was the Oswestry Disability Index (ODI), with the visual analogue scale (VAS) used as the secondary efficacy outcome. The adverse events and adverse reactions were also evaluated. Results. There was no significant difference in baseline characteristics between YBT (n = 358) and JYK groups (n = 120), and no difference was observed between groups for mean ODI score at day 0 (p = 0.064) or day 7 (p = 0.196), but there were differences at days 14, 21 and 30 (p < 0.001). The YBT showed more decline from baseline, and the decreased ODI score was substantially different from JYK (p < 0.001). The differences in decreased VAS scores between YBT and JYK were also significant at each time point (days 7, 14, 21, and 30), with better scores in the YBT group than in the JYK group (p < 0.001). In terms of safety, there was no obvious disparity in adverse events or adverse reactions between the 2 groups (p > 0.05). Conclusions. Yaobitong was better than JYK for LDH treatment, with no significant difference in safety. The study suggests that YBT is a promising and effective treatment for LDH. [ABSTRACT FROM AUTHOR]
