Aspergillus nidulans is a fungal model organism extensively used in genetic approaches. It may reproduce sexually and asexually, with a well‐defined parasexual cycle. The current paper demonstrates that the limitation of nitrogen source facilitates the production of A. nidulans's nonmeiotic recombinants directly from heterokaryons, without the recovery of the diploid phase. Heterokaryons formed between master strains were inoculated in sodium nitrate‐low (basal medium [BM]) and sodium nitrate‐rich media (minimal medium [MM]). All mitotic segregants produced by the heterokaryons were tested for their mitotic stability in the presence of benomyl, the haploidizing agent. Only mitotically stable haploid segregants were selected for subsequent analysis. Phenotypic analyses of such haploids favored the characterization of nonmeiotic recombinants. As the number of such recombinants was higher in BM than in MM, nitrogen limitation may have facilitated the isolation of nonmeiotic recombinants from heterokaryons by stimulating nuclear fusion still inside the heterokaryotic mycelium as a survival strategy. Guariroba is a palm species native to central Brazil. Seedlings of guariroba with leaf spots of unknown aetiology were found in Patos de Minas, Minas Gerais, Brazil. The leaf spots were manifest as two different symptom types: the first lesion type consisted of necrotic spots with a rounded to elongate shape, with a light colour and dark edges; the second lesion type had a rounded shape, was dark brown in colour with a light brown edge. The objective of this study was to elucidate the aetiology of both diseases. The likely causal agents were isolated and Koch's postulate fulfilled. Subsequently, the ITS region of rDNA from both micro-organisms were amplified and sequenced. According to the morphological characteristics and molecular analyses, the fungal species were identified as Pestalotiopsis adusta (causing necrotic spots with a rounded to elongate shape, with a light colour and dark edges) , and Alternaria tenuissima (causing lesions with a rounded shape, dark brown in colour with a light brown edge). Identification of the causal organisms of these two diseases will help guide management approaches that might be tested to reduce impact of the disease on Guariroba, including the use of fungicides and cultural approaches. Huanglongbing (HLB) is considered one of the most destructive diseases of citrus because the plants rapidly become unproductive, enter a decline, and eventually die. HLB is caused by the phloem-limited bacterium 'Candidatus Liberibacter' spp. The objective of this study was to evaluate seasonal variation of the in planta population of 'Ca. Liberibacter asiaticus' in the foliage of citrus trees in Brazil using real-time polymerase chain reaction (qPCR). Eleven plants (naturally infected, then screened) in the field with very mild and localized symptoms of HLB were confirmed to be 'Ca. L. asiaticus' infected by conventional PCR, and the canopies were divided into four quadrants. The bacterial population in the trees was tested on a monthly basis for up to 20 months by quantifying 'Ca. L. asiaticus' DNA using qPCR 'Ca. L. asiaticus'-specific primers (As84F/As180R). The average cycle threshold (Ct) values, which relate to 'Ca. L. asiaticus' titer, were analyzed using a mixed model. Significant differences were observed in Ct values between seasons (F = 8.77, P = 0.0004), and abrupt changes were observed in Ct values in different quadrants of the trees. Autumn had the lowest Ct values, indicating the highest 'Ca. L. asiaticus' titer, and, thus, is considered the best period to detect 'Ca. L. asiaticus' infection in foliage of citrus trees in southern Brazil. In addition to the seasonal changes in Ct values, there was an initial decline in the Ct value in the months following initial detection, the rate of decline slowing with time. Concomitant with the increase of the bacterial population in the host, there was an increase in severity of HLB symptoms in the trees over time (Spearman's rank correlation, r = -0.4083, P < 0.0001). The results identify the optimal season to sample foliage for 'Ca. L. asiaticus' in southern Brazil (autumn) and confirm the importance of sample timing to maximize detection of 'Ca. Asiatic citrus canker (ACC, caused by the bacterium Xanthomonas citri subsp. citri) is a destructive disease of citrus in Brazil and in several other citrus-producing countries. ACC management is problematic, and bactericides such as copper can be reasonably efficacious but do not completely control the disease. Furthermore, injury by citrus leaf miner (CLM) can exacerbate severity of ACC. Host resistance is the most desirable solution for management of ACC; however, evaluations of germplasm indicate that resistance is limited in many popular species and cultivars that are grown commercially. Limited evaluations have been made of sweet orange (Citrus sinensis) selections. We evaluated resistance of 25 Pera sweet orange selections to X. citri subsp. citri by wound inoculation and measuring lesion diameter under greenhouse conditions (wound inoculation indicates mesophyll resistance which will be valuable in areas where CLM exists). ACC severity was assessed on the same 25 selections at three locations in the field in Brazil, relying on natural inoculum and conditions to cause disease. In the greenhouse experiments, the selections EEL. Bianchi/CC. Ipigua, Olimpia, IAC 2000/1, and Ovale Siracusa consistently had the smallest diameter lesions, indicating greatest resistance, although differences in lesion diameter were small. Results from the field experiments were less conclusive, although EEL and Ovale Siracusa were consistently numerically least affected by ACC. These results indicate selections of sweet orange that might be preferable to consider in canker-prone areas in Brazil and elsewhere. [ABSTRACT FROM AUTHOR]
