8. Global research hotspots and trends in oxidative stress-related diabetic nephropathy: a bibliometric study

Abstract

BackgroundOxidative stress is widely acknowledged as a key pathogenic mechanism in diabetic nephropathy (DN). In recent years, the role of oxidative stress in DN has garnered increasing attention. However, no bibliometric analysis has yet been conducted on the relationship between oxidative stress and DN. This study aims to systematically analyze the relevant literature, identify trends in research, assess current hotspots, and predict future directions.MethodsWe retrieved literature related to oxidative stress and DN from the Web of Science Core Collection database. We analyzed data on publication volume, countries/regions, institutions, journals, keywords, and other relevant metrics using VOSviewer, the Bibliometrix R package, and CiteSpace.ResultsFrom 2014 to 2024, a total of 4076 publications related to oxidative stress and DN were published across 755 journals, showing a consistent upward trend each year. China and the United States are the leading contributors in this field and demonstrate close collaborative efforts. The top contributors by country, institution, journal, and author include: China (1919 publications), Jilin University and Central South University (69 publications each), BIOMEDICINE & PHARMACOTHERAPY (117 publications), and Prof. Sun Lin (33 publications). The most frequent keyword is “oxidative stress” (3683 occurrences). In the co-citation analysis, Alicic RZ’s 2017 study was the most cited (144 citations). These findings highlight the critical importance of investigating the pathogenesis of DN from the oxidative stress perspective.ConclusionThis study demonstrates a steady increase in research on oxidative stress in DN since 2014, highlighting its central role in the pathogenesis of DN. Future research should focus on the molecular mechanisms of oxidative stress in DN and explore its therapeutic potential, to provide new strategies for the prevention and treatment of DN.

Abstract

Atherosclerotic cardiovascular disease (ASCVD) is the leading cause of death among urban and rural residents in China, and elevated low-density lipoprotein cholesterol (LDL-C) is a risk factor for ASCVD. Considering the increasing burden of ASCVD, lipid management is of the utmost importance. In recent years, research on blood lipids has made breakthroughs around the world, hence a revision of Chinese guideline for lipid management is imperative, especially since the target lipid levels in the general population vary in respect to the risk of ASCVD. The level of LDL-C, which can be regarded as appropriate in a population without frisk factors, can be considered abnormal in people at high risk of developing ASCVD. As a result, the “Guidelines for the prevention and treatment of dyslipidemia" were adapted into the “Chinese guideline for Lipid Management" (henceforth referred to as the new guidelines) by an Experts’ committee after careful deliberation. The new guidelines still recommend LDL-C as the primary target for lipid control, with cardiovascular disease (CVD) risk stratification to determine its target value. These guidelines recommend that moderate intensity statin therapy in adjunct with a heart-healthy lifestyle, be used as an initial line of treatment, followed by cholesterol absorption inhibitors or/and proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors, as necessary. The new guidelines provide guidance for lipid management across various age groups, from children to the elderly. The aim of these guidelines is to comprehensively improve the management of lipids and promote the prevention and treatment of ASCVD by guiding clinical practice.

Abstract

α,β-Unsaturated ketone is one of the important building blocks of the plenty of alkaloids, spices and pharmaceutical agents. Herein, we describe a peroxide-free electrochemical strategy to realize site-selective allylic C − H oxidation for the synthesis of α,β-unsaturated ketones. This electrochemically driven oxidation exhibits a broad substrate scope. Mechanistic investigations of this reaction uncovered HOAc and H2O as the oxygen source to achieve peroxide-free electrochemical allylic C − H oxidation.

Abstract

Abstract Highly aligned aggregation state structures have great significance for effective charge‐carrier transport in organic semiconductors. Several methods have been developed to provide organic semiconducting molecules with highly oriented aggregation state structure; among these, using a water surface to form organic semiconductor films is a widely implemented strategy, wherein solutions are spread on the surface of water. However, common techniques of film formation on water surfaces generally result in a nonuniform orientation of the film owing to the isotropic spread process of droplets on the water surface. In this study, a spatially confined air/water interfacial assembly method is proposed to obtain uniformly aligned monolayer and multilayer poly(diketopyrrolopyrrole‐thieno[3,2‐b]thiophene) thin films with controlled thickness. The structural and morphological characterizations obtained using atomic force microscopy, high‐resolution transmission electron microscopy, and grazing incidence wide‐angle X‐ray scattering indicates the crystalline structure of the thin films and high alignment of the molecular chains. The maximum mobility of the thin films reaches up to 2.06 and 0.5 cm2 V−1 s−1 in the parallel and perpendicular direction, respectively, indicating apparent anisotropic electrical properties. Furthermore, an inverter based on these thin films exhibits a voltage gain of up to 70, demonstrating the potential of applying the proposed technique to logic circuits.

Abstract

Rapid and accurate molecular diagnostics are crucial for preventing the global spread of emerging infectious diseases. However, the current gold standard for nucleic acid detection, reverse transcription polymerase chain reaction (RT-PCR), relies heavily on traditional magnetic beads or silica membranes for nucleic acid extraction, resulting in several limitations, including time-consuming processes, the need for trained personnel, and complex equipment. Therefore, there is an urgent need for fully integrated nucleic acid detection technologies that are simple to operate, rapid, and highly sensitive to meet unmet clinical needs. In this study, we developed a novel, integrated microfluidic cartridge featuring a unique needle-plug/piston microvalve, which enables stable long-term reagent storage and flexible liquid handling for on-site nucleic acid analysis. Coupled with in situ tetra-primer recombinase polymerase amplification (tp-RPA), we achieved highly sensitive nucleic acid detection with a remarkable limit of detection of 20 copies per mL (0.02 copies per μL) and a short turnaround time of less than 30 minutes. To validate this assay, we tested 48 oropharyngeal swab samples. The positive detection rate reached 64.58% (31/48), significantly exceeding the approximately 50% positive detection rate of the traditional RT-PCR method. Furthermore, our assay demonstrated a 100% concordance rate with RT-PCR in detecting positive samples. Thus, we believe our microfluidic nucleic acid analysis system represents a promising approach for enabling rapid and ultrasensitive nucleic acid detection of pathogenic microorganisms in resource-limited settings and low-income areas. Abstract

Background Tendon calcification is a common disease, and it could happen in the tendons of the shoulder, wrist, etc. However, tendon calcification in the superior and inferior gemellus is rare, and in this region is likely to be misdiagnosed. Case Presentation Here, our case report first reported a 53‐year‐old female patient with an unusual case of calcific tendinitis of the gemellus superior and gemellus inferior muscles. The patient presented with severe pain in the right hip and lower extremities, not relieved using nonsteroidal anti‐inflammatory drugs (NSAIDs). The preoperative physical examination indicated an abnormality in the hip joint. Preoperative imaging confirmed the results of the physical examination and showed a right hip lesion. We did not make a definite diagnosis preoperatively but considered that the patient might have an osteochondroma. However, surgical findings indicated that the lesion was not in the hip capsule on subsequent arthroscopic surgery, which suggested that the preoperative diagnosis might be wrong. We opened the posterior capsule and found a “toothpaste‐like” lesion in the superior and inferior gemellus muscles' tendon. We thought this might be the calcified tendon. Then the arthroscopic surgery was finished to remove the lesion, and the removed tissue was sent to the pathology department for pathological examination. The pathological report confirmed the diagnosis of the calcified tendon. Postoperative follow‐up was conducted. The effect of the operation was noticeable. Postoperative symptoms were relieved. Conclusions Calcification of the tendons of the superior and inferior gemellus muscles can be easily misdiagnosed, and the disease can be treated minimally with arthroscopy.

Abstract

Background and purpose: 18 F-prostate-specific membrane antigen (PSMA)-1007 positron emission tomography and computed tomography (PET/CT) is an advanced molecular imaging evaluation method for prostate cancer (PCa). This study aimed to explore the early detection rate of recurrence and metastasis of patients with biochemical recurrence (BCR) after radical prostatectomy (RP) by 18 F-PSMA-1007 PET/CT and its influence on clinical treatment decisions. Methods: From December 2018 to December 2020, the data of 51 PCa patients with BCR after RP by 18 F-PSMA-1007 PET/CT were summarized and analyzed. Radioactive uptake of tumors was calculated semi-quantitatively by region of interest method and expressed by the maximum standardized uptake value (SUV max ). We assessed the detection rate of clinical recurrence and metastasis in BCR patients [local recurrence (prostatic bed), lymph node metastasis (pelvic, retroperitoneal and diaphragmatic), bone metastasis and visceral metastasis (such as lung)], and the difference in detection rate between prostate-specific antigen (PSA) groups and Gleason evaluation group was further compared respectively. Results: The median age of 51 patients was 66 years (52-80 years), and the median PSA was 35 ng/mL (6-224 ng/mL) at the time of initial diagnosis. All of them were prostatic acinar adenocarcinoma, including 1 case with intraductal carcinoma, 1 case with ductal adenocarcinoma, 1 case with mucinous adenocarcinoma, 1 case with signet ring-like component and 1 case with neuroendocrine differentiation. We found Gleason score ≤7 in 22 cases (43.14%) and Gleason score ≥ 8 in 29 cases (56.86%). The median time of BCR was 15 months (5-62 months), and the median PSA was 0.58 ng/mL (0.20- 110.00 ng/mL), including 21 (41.18%) cases with 0.20 ng/mL≤PSA ＜ 0.50 ng/mL, 12 (23.53%) cases with 0.50 ng/mL≤PSA ＜ 1.00 ng/mL, 4 (7.84%) cases with 1.00 ng/mL≤PSA ＜ 2.00 ng/mL and 14 (27.45%) cases with PSA≥2.00 ng/mL. There were 7 cases (13.73%) with no local recurrence or metastasis, and 44 cases (86.27%) with local recurrence or metastasis, including 9 cases (20.45%) with recurrence in the operative area of prostate, 28 cases (63.64%) had lymph node metastasis at different sites, 31 cases (70.45%) had bone metastasis, and 2 cases (4.55%) had visceral metastasis. In addition, there were 2 cases of subcutaneous nodule metastasis and 1 case of penile root metastasis. The median SUV max was 17.9 (1.4-110.9) for all recurrence or metastasis, 14.0 (3.2-110.9) for local recurrence, 10.2 (2.0-90.1) for lymph node metastasis, and 5.4 (1.4-109.6) for bone metastasis. The detection rates of recurrence or metastasis were 71.43% (15/21), 100.00% (12/12), 75.00% (3/4) and 100.00% (14/14), respectively, in the groups with 0.20 ng/mL≤PSA ＜ 0.50 ng/mL (21 cases), 0.50 ng/mL≤PSA ＜ 1.00 ng/mL (12 cases), 1.00 ng/mL≤PSA ＜ 2.00 ng/mL (4 cases) and PSA≥2.00 ng/mL (14 cases), and there was no statistically significant difference in the detection rate between groups with different PSA levels (P＞0.05). The recurrence or metastasis detection rates of original Gleason score ≤7 group (22 cases) and Gleason score ≥8 group (29 cases) were 68.18% (15/22) and 100.00% (29/29), respectively, and there were statistically significant differences in the detection rate between groups with different Gleason scores (P ＜ 0.05). In clinical treatment, 4 cases (7.84%) were treated by observation, 18 cases (35.29%) by endocrinotherapy alone, 2 cases (3.92%) by salvage radiotherapy (SRT) alone, 24 cases (47.06%) by endocrinotherapy combined with SRT, 1 case (1.96%) by endocrinotherapy combined with docetaxel systemic chemotherapy, and 2 cases (3.92%) by salvage pelvic lymphadenectomy. Conclusion: 18 F-PSMA-1007 PET/CT has a good value and efficacy in early diagnosis of clinical recurrence or metastasis of BCR patients after RP, which is conducive to accurate evaluation and optimal treatment plan for such patients, and significantly affects clinical treatment decisions.

Abstract

Emerging diagnostic scenarios, such as population surveillance by pooled testing and on‐site rapid diagnosis, highlight the importance of advanced microfluidic systems for in vitro diagnostics. However, the widespread adoption of microfluidic technology faces challenges due to the lack of standardized design paradigms, posing difficulties in managing macro‐micro fluidic interfaces, reagent storage, and complex macrofluidic operations. This paper introduces a novel modular‐based mesoscopic design paradigm, featuring a core "needle‐plug/piston" structure with versatile variants for complex fluidic operations. These structures can be easily coupled with various microfluidic platforms to achieve truly self‐contained microsystems. Incorporated into a "3D extensible" design architecture, the mesoscopic design meets the demands of function integration, macrofluid manipulations, and flexible throughputs for point‐of‐care nucleic acid testing. Using this approach, an ultra‐sensitive nucleic acid detection system is developed with a limit of detection of ten copies of SARS‐CoV‐2 per mL. This system efficiently conducts large‐scale pooled testing from 50 pharyngeal swabs in a tube with an uncompromised sensitivity, enabling a truly "sample‐in‐answer‐out" microsystem with exceptional performance. Abstract

This study aims to explore the correlation between renal blood perfusion (RBP) and diabetic nephropathy (DN). Methods: A total of 72 mice included db/db and db/m mice at the ages of 6, 14, and 22 weeks, forming six groups. RBP was assessed using Laser Speckle Contrast Imaging (LSCI). Kidney function markers and the extent of pathological damage were evaluated. Pearson correlation analysis was employed to predict the relationship between RBP and various indicators of kidney damage. Results: Compared to db/m mice of all ages, 6-week-old db/db mice showed no significant difference in kidney function markers and had no apparent pathological damage. However, db/db mice at other ages showed deteriorating kidney functions and evident pathological damage, which worsened with age. The RBP in db/m mice of all ages and 6-week-old db/db mice showed no significant difference; however, RBP in db/db mice demonstrated a significant declining trend with age. The correlation between RBP and kidney damage indicators was as follows: 24 h urinary microalbumin (r=-0.728), urinary transferrin (r=-0.834), urinary ß2-microglobulin (r=-0.755), urinary monocyte chemoattractant protein-1 (r=-0.786), Masson's trichrome staining (r=-0.872), and Periodic Acid-Schiff staining (r=-0.908). Abstract

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by complex pathogenesis mechanisms. Among these, β‐amyloid plaques and hyperphosphorylated Tau protein tangles have been identified as significant contributors to neuronal damage. This study investigates thonningianin A (TA) from Penthorum chinense Pursh (PCP) as a potential inhibitor targeting these pivotal proteins in AD progression. The inhibitory potential of PCP and TA on Aβ fibrillization was initially investigated. Subsequently, ultra‐high performance liquid chromatography‐quadrupole time‐of‐flight tandem mass spectrometry and biolayer interferometry were employed to determine TA's affinity for both Aβ and Tau. The inhibitory effects of TA on the levels and cytotoxicity of AD‐related proteins were then assessed. In 3xTg‐AD mice, the therapeutic potential of TA was evaluated. Additionally, the molecular interactions between TA and either Aβ or Tau were explored using molecular docking. We found that PCP‐total ethanol extract and TA significantly inhibited Aβ fibrillization. Additionally, TA demonstrated strong affinity to Aβ and Tau, reduced levels of amyloid precursor protein and Tau, and alleviated mitochondrial distress in PC‐12 cells. In 3xTg‐AD mice, TA improved cognition, reduced Aβ and Tau pathology, and strengthened neurons. Moreover, molecular analyses revealed efficient binding of TA to Aβ and Tau. In conclusion, TA, derived from PCP, shows significant neuroprotection against AD proteins, highlighting its potential as an anti‐AD drug candidate. [ABSTRACT FROM AUTHOR]

Abstract

Abstract Small nucleolar RNA host gene 6 (SNHG6) is a newly discovered long non-coding RNA (lncRNA), while the regulatory mechanism of SNHG6 in chondrosarcoma is largely unknown. Here we found that SNHG6 expression was upregulated and showed positive correlation with the progression of chondrosarcoma. Functional assays demonstrated that SNHG6 was required for the proliferation, migration, and invasion of chondrosarcoma cells. Mechanistic study revealed that SNHG6 could recruit EZH2 and maintain high level of H3K27me3 to repress the transcription of tumor-suppressor genes, including KLF6. KLF6 was found to bind to the promoter region of SP1 and restrained its transcription, while SP1 could be recruited to the promoter region of SNHG6 and promoted its transcription to form a positive loop. In summary, this study reveals that SP1-induced SNHG6 forms a positive loop to facilitate the carcinogenesis of chondrosarcoma through the suppression of KLF6 by recruiting EZH2, which manifests the oncogenic function of SNHG6 in chondrosarcoma.

Abstract

Since December 2019, COVID-19, an acute infectious disease, has gradually become a global threat. We report a case of thoracolumbar fractures (T12 and L1) and incomplete lower limb paralysis in a patient with COVID-19. After a series of conservative treatment which did not work at all, posterior open reduction and pedicle screw internal fixation of the thoracolumbar fracture were performed in Wuhan Union Hospital. Three weeks later, the patient could stand up and the pneumonia is almost cured. We successfully performed a surgery in a COVID-19 patient, and to our knowledge it is the first operation for a COVID-19 patient ever reported.
