Single-cell approaches are quickly changing our view on biological systems by increasing the spatiotemporal resolution of our analyses to the level of the individual cell. The field of plant biology has fully embraced single-cell transcriptomics and is rapidly expanding the portfolio of available technologies and applications. In this review, we give an overview of the main advances in plant single-cell transcriptomics over the past few years and provide the reader with an accessible guideline covering all steps, from sample preparation to data analysis. We end by offering a glimpse of how these technologies will shape and accelerate plant-specific research in the near future.

In the meat industry, hydrocolloids and phosphates are used to improve the quality attributes of meat products. However, latest research results revealed that the usage of exopolysaccharide (EPS)-forming lactic acid bacteria (LAB), which are able to produce EPS in situ during processing could be an interesting alternative. The current review aims to give a better understanding of bacterial EPS production in food matrices with a special focus on meat products. This includes an introduction to microbial EPS production (homopolysaccharides as well as heteropolysaccharides) and an overview of parameters affecting EPS formation and yield depending on LAB used. This is followed by a summary of methods to detect and characterize EPS to facilitate a rational selection of starter cultures and fermentation conditions based on desired structure-function relationships in different food matrices. The mechanism of action of in situ generated EPS is then highlighted with an emphasis on different meat products. In order to apply satellite data to guiding navigation in the Arctic more effectively, the sea ice concentrations (SIC) derived from passive microwave (PM) products were compared with ship-based visual observations (OBS) collected during the Chinese National Arctic Research Expeditions (CHINARE). A total of 3 667 observations were collected in the Arctic summers of 2010, 2012, 2014, 2016, and 2018. PM SIC were derived from the NASA-Team (NT), Bootstrap (BT) and Climate Data Record (CDR) algorithms based on the SSMIS sensor, as well as the BT, enhanced NASA-Team (NT2) and ARTIST Sea Ice (ASI) algorithms based on AMSR-E/AMSR-2 sensors. The daily arithmetic average of PM SIC values and the daily weighted average of OBS SIC values were used for the comparisons. The correlation coefficients (CC), biases and root mean square deviations (RMSD) between PM SIC and OBS SIC were compared in terms of the overall trend, and under mild/normal/severe ice conditions. Using the OBS data, the influences of floe size and ice thickness on the SIC retrieval of different PM products were evaluated by calculating the daily weighted average of floe size code and ice thickness. Our results show that CC values range from 0.89 (AMSR-E/AMSR-2 NT2) to 0.95 (SSMIS NT), biases range from -3.96% (SSMIS NT) to 12.05% (AMSR-E/AMSR-2 NT2), and RMSD values range from 10.81% (SSMIS NT) to 20.15% (AMSR-E/AMSR-2 NT2). Floe size has a significant influence on the SIC retrievals of the PM products, and most of the PM products tend to underestimate SIC under smaller floe size conditions and overestimate SIC under larger floe size conditions. Ice thickness thicker than 30 cm does not have a significant influence on the SIC retrieval of PM products. Overall, the best (worst) agreement occurs between OBS SIC and SSMIS NT (AMSR-E/AMSR-2 NT2) SIC in the Arctic summer.

The similarity of in-situ and ex-situ absorption/extinction properties was found by comparing data sets obtained in premixed flat ethylene flames (C/O = 0.77); both ex-situ absorption measurements at CNR of Naples and in-situ laser extinction/absorption measurements at Lund University. The optical band gap analysis was performed as a method to separate/evaluate the contributions to the UV–Visible absorption from organic carbon and soot by selecting two spectral regions; above 685 nm where only soot is assumed to absorb radiation and below 685 nm, where both soot and organic carbons absorb. Optical band gap for soot was analyzed separately from organic carbon, obtaining their individual contribution to the spectral absorption from inception, throughout the growth region up to soot aggregation. While the optical band gap of soot strongly decreased along the flame from 1 to 0.1 eV, the band gap for organic carbons remained constant at ∼1.7–1.8 eV. By using the relationship of the optical band gap with nanostructural parameters (layer length and aromatic rings number), the average molecular weight of organic carbon around 400 u resulted rather far away from the molecular weight of 1000–2000 mass units evaluated for incipient soot consistently with the view of PAH species sticking together for particle inception.

Montmorillonite (MMT) was modified with cetyl trimethyl ammonium bromide (CTAB) to obtain organomontmorillonite (OMMT) by stirring and pulsed ultrasonic mixing. Polyamide 6 (PA6)/OMMT nanocomposites were then prepared via in-situ polymerization.The resulting OMMT and PA6/OMMT nanocomposites were characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM) and scanning electron microscopy (SEM). The results suggested that the OMMT interlayer distance was greatly increased to 3.13 nm due to CTAB being inter-calated into the MMT galleries. The OMMT interlayer distance was further enlarged to 10-20 nm during the polymerization process. The OMMT layers were exfoliated into nanoscale layers and uniformly dispersed in the molten ∈-caprolactam and PA6 matrix, and exfoliated structure nanocomposites were formed.

Current time-resolvedin situapproaches limit the scope of mechanochemical investigations possible. Here we develop a new, general approach to simultaneously follow the evolution of bulk atomic and electronic structure during a mechanochemical synthesis. This is achieved by coupling two complementary synchrotron-based X-ray methods: X-ray absorption spectroscopy (XAS) and X-ray diffraction. We apply this method to investigate the bottom-up mechanosynthesis of technologically important Au micro and nanoparticles in the presence of three different reducing agents, hydroquinone, sodium citrate, and NaBH4. Moreover, we show how XAS offers new insight into the early stage generation of growth species (e.g.monomers and clusters), which lead to the subsequent formation of nanoparticles. These processes are beyond the detection capabilities of diffraction methods. This combined X-ray approach paves the way to new directions in mechanochemical research of advanced electronic materials.

Innovative and efficient hit‐identification techniques are required to accelerate drug discovery. Protein‐templated fragment ligations represent a promising strategy in early drug discovery, enabling the target to assemble and select its binders from a pool of building blocks. Development of new protein‐templated reactions to access a larger structural diversity and expansion of the variety of targets to demonstrate the scope of the technique are of prime interest for medicinal chemists. Herein, we present our attempts to use a protein‐templated reductive amination to target protein‐protein interactions (PPIs), a challenging class of drug targets. We address a flexible pocket, which is difficult to achieve by structure‐based drug design. After careful analysis we did not find one of the possible products in the kinetic target‐guided synthesis (KTGS) approach, however subsequent synthesis and biochemical evaluation of each library member demonstrated that all the obtained molecules inhibit MDM2. A large amount of research within organic biosensors is dominated by organic electrochemical transistors (OECTs) that use conducting polymers such as poly(3,4-ethylene dioxythiophene doped with poly(styrenesulfonate) (PEDOT:PSS). Despite the recent advances in OECT-based biosensors, the sensing is solely reliant on the amperometric detection of the bioanalytes. This is typically accompanied by large undesirable parasitic electrical signals from the electroactive components in the electrolyte. Herein, we present the use of in-situ resonance Raman spectroscopy to probe subtle molecular structural changes of PEDOT:PSS associated with its doping level. We demonstrate how such doping level changes of PEDOT:PSS can be used, for the first time, on operational OECTs for sensitive and selective metabolite sensing whilst simultaneously performing amperometric detection of the analyte. We test the sensitivity by molecularly sensing a lowest glucose concentration of 0.02 mM in phosphate buffered saline (PBS) solution. By changing the electrolyte to cell culture media, the selectivity of in-situ resonance Raman spectroscopy is emphasized as it remains unaffected by other electroactive components in the electrolyte. The application of this molecular structural probe highlights the importance of developing biosensing probes that benefit from high sensitivity of the material's structural and electrical properties whilst being complimentary with the electronic methods of detection.

Understanding nucleation phenomena is crucial across all branches of physical and natural sciences. Colloidal nanocrystals are among the most versatile and tunable synthetic nanomaterials. While huge steps have been made in their synthetic development, synthesis by design is still impeded by the lack of knowledge of reaction mechanisms. Here, we report on the investigation of the reaction intermediates in high temperature syntheses of copper nanocrystals by a variety of techniques, including X-ray absorption at a synchrotron source using a customized in situ cell. We reveal unique insights into the chemical nature of the reaction intermediates and into their role in determining the final shape of the metal nanocrystals. Overall, this study highlights the importance of understanding the chemistry behind nucleation as a key parameter to predict synthetic pathways for shape-controlled nanocrystals.

An efficient electrospun aligned surface enhanced Raman scattering (SERS) and maize-like substrate of polyvinyl alcohol (PVA) composite and Ag colloid nanofibers decorated with thermal evaporated Ag nanoparticles (AgNPs) has been developed by taking advantage of electrostatic interactions. The synergistic effects of the evaporated AgNPs (niblets) and the Ag colloid in PVA (corncob) could arouse strong electromagnetic field between the lateral and vertical nanogaps which has been demonstrated by experiment and finite-different time-domain (FDTD) simulation. In this experiment, the aligned nanofibers possesses an excellent sensitivity by detection of crystal violet (CV) and malachite green (MG) molecule at low concentration. Moreover, the proposed flexible SERS sensor was measured with outstanding uniformity and reproducibility. We also carried out in-situ electrospinning on a curved surface to detect the mixture of Sudan I, CV and MG molecule, which demonstrates that flexible SERS sensor, has enormous potential in accurate and in-situ detection on the complex geometric structure.

Bacterial nanocellulose (BNC) is a natural polymer composed of glucose units with an important application as a two and three-dimensional scaffold for tissue engineering. However, as a polysaccharide, BNC does not have the biological signals of protein biomaterials. Therefore, this paper aims to develop a novel methodology to biomimic soft extracellular matrix (ECM) chemistry on to 3D BNC using the bioengineering of fibroblasts (the cells responsible for producing and regenerating the ECM) to immobilise adhesion proteins such as collagen and fibronectin. Modified 3D BNC (Mod-BNC) biomaterials were morphologically, thermally, and chemically characterised, and furthermore, the cell response was analysed by adhesion studies using atomic force microscopy (AFM), XTT assay, and confocal microscopy.Cell-derived proteins were deposited on the BNC nanoribbon network to modify its surface. The contact angle was increased from 40 degrees to 60 degrees, reducing the wettability of the biomaterial, and during thermogravimetry, the proteins in Mod-BNC exhibited an enhanced thermal stability because of the interactions between themselves and BNC. Chemical and immunocytochemistry analyses confirmed the presence of collagen type I and fibronectin on 3D BNC. These proteins activate integrin adhesion pathways that generate stronger cell adhesions. AFM experiments showed higher forces and energies on modified biomaterials, and moreover, the cells that adhered on to Mod-BNC exhibited higher mitochondrial activity and higher cell populations per cubic millimetre than non-modified surfaces (NMod-BNC). Accordingly, it was established that this novel methodology is robust and able to biomimic the chemical surface of soft ECM and immobilise cell-derived adhesion proteins from fibroblast; moreover, the Mod-BNC exhibited better cell response than NMod-BNC because of the biological signals in 3D BNC.

This study aims at assessing the thermal nuclei motion effects on the multipole transition channels involved in two core-level spectroscopies, x-ray absorption spectroscopy (XAS) and x-ray Raman scattering (XRS). Temperature effects on the $1s\ensuremath{\rightarrow}s$ monopole, $1s\ensuremath{\rightarrow}p$ dipole, and $1s\ensuremath{\rightarrow}d$ quadrupole transitions are investigated using two reference systems for which we present original experimental data: $\ensuremath{\alpha}\text{\ensuremath{-}}{\mathrm{Al}}_{2}{\mathrm{O}}_{3}$ at the Al $K$ edge probed by XRS at room temperature and rutile $\mathrm{Ti}{\mathrm{O}}_{2}$ at the Ti $K$ pre-edge probed by XAS at temperatures ranging from 6 to 700 K. Through the latter, this work enlightens the part of the pre-edge peak enhancement due to temperature in the $K$ pre-edge region of $3d$ transition metal, which is known to be routinely used to determine the concentration, valence or symmetry of the probed element in a given sample. Nuclear thermal fluctuations are taken into account using a method based on density functional theory that consists in averaging spectra over atomic configurations, generated within the harmonic approximation and obeying quantum statistics at finite temperature. Since only a finite number of such configurations are used, the numerically averaged spectra generally lose the symmetry of the equilibrium crystal positions. In this paper, we demonstrate that the physical average has to be symmetric and propose a method to restore the physical angular dependence of the spectra. The approach is successfully applied to investigate the angular dependent XAS spectra in rutile as a function of temperature. The two systems under study allow to draw general conclusions regarding the effect of nuclear quantum fluctuations on the different transition channels available to both core-level spectroscopies.

Immense field enhancement and nanoscale confinement of light are possible within nanoparticle-on-mirror (NPoM) plasmonic resonators, which enable novel optically-activated physical and chemical phenomena, and render these nanocavities greatly sensitive to minute structural changes, down to the atomic scale. Although a few of these structural parameters, primarily linked to the nanoparticle and the mirror morphology, have been identified, the impact of molecular assembly and organization of the spacer layer between them has often been left uncharacterized. Here, we experimentally investigate how the complex and reconfigurable nature of a thiol-based self-assembled monolayer (SAM) adsorbed on the mirror surface impacts the optical properties of the NPoMs. We fabricate NPoMs with distinct molecular organizations by controlling the incubation time of the mirror in the thiol solution. Afterwards, we investigate the structural changes that occur under laser irradiation by tracking the bonding dipole plasmon mode, while also monitoring Stokes and anti-Stokes Raman scattering from the molecules as a probe of their integrity. First, we find an effective decrease in the SAM height as the laser power increases, compatible with an irreversible change of molecule orientation caused by heating. Second, we observe that the nanocavities prepared with a densely packed and more ordered monolayer of molecules are more prone to changes in their resonance compared to samples with sparser and more disordered SAMs. Our measurements indicate that molecular orientation and packing on the mirror surface play a key role in determining the stability of NPoM structures and hence highlight the under-recognized significance of SAM characterization in the development of NPoM-based applications.

Poly [(vinylbenzyl) trimethylammonium chloride] (PVBTMAC) has been used as a stabilizer in the polymerization-induced self-assembly polymerizations of diacetone acrylamide (DAAM). A whole spectrum of particle morphologies was obtained simply by adjusting the ionic strength of the reaction mixtures; no dilution of the cationic charges with noncharged comonomers or with noncharged polymers was needed. In addition to the ionic strength, the effects of solid content and the length of the PDAAM block on the morphologies of the particles were studied in detail. The experiments are a continuation to previous studies on solution properties of PVBTMAC. It has been shown earlier that the solubility of the polycation may be tuned with counterions. Hydrophobic triflate ions induce an upper critical solution temperature behavior. In the present case, the chains bound to hydrophobic cores of the particles show either a one-step phase separation in aqueous triflate solutions or under certain conditions, a two-step transition. The step-wise transition is typical for responsive polymers with limited mobility.

In this work, the composition, structural and morphological features, and particle size of the active phase of the catalyst (MoS2), synthesized in-situ during the heavy oil hydroconversion performed in continuous flow reactor on lab-scale pilot flow unit at T = 450 °C, P = 6.0–9.0 MPa, V = 1.0 h−1, H2/feed = 1000 nL/L, catalyst concentration C (Mo) = 0.01–0.08%wt have been studied. It has been shown that MoS2 formed during hydroconversion is represented by nanosized particles stabilized by polycondensation products as a result of strong adsorption and aggregation with the components of the hydroconversion reaction medium. The influence of morphological characteristics of catalyst nanoparticles on the feed conversion, the yield of gaseous and liquid products, and the quality of distillate fractions, as well as the yield of polycondensation products, have been studied. It has been established that an increase in MoS2 active site dispersion, both due to a decreased plate length and lower stacking numbers in MoS2 cluster, enhances hydroconversion effectivity, particularly, in suppressing polycondensation reactions.

Fishery of blue mussels Mytilus edulis constitutes a very important economic activity in Denmark, whereas mussel farming on long-lines or nets is a new, growing sector. Spawning from natural mussel beds takes place during early summer, and larvae disperse via water currents before settling on the bottom or on spat collectors in the farms. In the present study, we coupled a 3D physical model system (FlexSem) with an agent-based model in order to examine the connectivity of this marine system in terms of mussel larval dispersal and settling potential. To address this question, we (1) estimated the dispersal and connectivity between 17 areas in the Limfjorden, (2) identified the main donor and receiver areas of mussel larvae and (3) identified possible dispersal barriers. The results show that the central narrow strait in the Limfjorden was the main donor area in all the studied years, and that the adjacent eastern areas were the main receiver areas. Towards the inner basins of the Limfjorden, isolation increased and limited connectivity was observed. The results from the cluster analysis grouped the Limfjorden into 3 to 5 clusters, but there was still some exchange of simulated larvae observed among these clusters. Analysis of molecular markers revealed no genetic differentiation between areas and supports the model results, indicating that despite distinguishable hydrographic boundaries, the mussel populations in the Limfjorden are well connected. This study demonstrates how connectivity modeling can be used to support site selection processes in aquaculture.

Framed within the Copernicus Climate Change Service (C3S) of the European Commission, the European Centre for Medium-Range Weather Forecasts (ECMWF) is producing an enhanced global dataset for the land component of the fifth generation of European ReAnalysis (ERA5), hereafter referred to as ERA5-Land. Once completed, the period covered will span from 1950 to the present, with continuous updates to support land monitoring applications. ERA5-Land describes the evolution of the water and energy cycles over land in a consistent manner over the production period, which, among others, could be used to analyse trends and anomalies. This is achieved through global high-resolution numerical integrations of the ECMWF land surface model driven by the downscaled meteorological forcing from the ERA5 climate reanalysis, including an elevation correction for the thermodynamic near-surface state. ERA5-Land shares with ERA5 most of the parameterizations that guarantees the use of the state-of-the-art land surface modelling applied to numerical weather prediction (NWP) models. A main advantage of ERA5-Land compared to ERA5 and the older ERA-Interim is the horizontal resolution, which is enhanced globally to 9 km compared to 31 km (ERA5) or 80 km (ERA-Interim), whereas the temporal resolution is hourly as in ERA5. Evaluation against independent in situ observations and global model or satellite-based reference datasets shows the added value of ERA5-Land in the description of the hydrological cycle, in particular with enhanced soil moisture and lake description, and an overall better agreement of river discharge estimations with available observations. However, ERA5-Land snow depth fields present a mixed performance when compared to those of ERA5, depending on geographical location and altitude. The description of the energy cycle shows comparable results with ERA5. Nevertheless, ERA5-Land reduces the global averaged root mean square error of the skin temperature, taking as reference MODIS data, mainly due to the contribution of coastal points where spatial resolution is important. Since January 2020, the ERA5-Land period available has extended from January 1981 to the near present, with a 2- to 3-month delay with respect to real time. The segment prior to 1981 is in production, aiming for a release of the whole dataset in summer/autumn 2021. The high spatial and temporal resolution of ERA5-Land, its extended period, and the consistency of the fields produced makes it a valuable dataset to support hydrological studies, to initialize NWP and climate models, and to support diverse applications dealing with water resource, land, and environmental management. The SuperCam instrument suite provides the Mars 2020 rover, Perseverance, with a number of versatile remote-sensing techniques that can be used at long distance as well as within the robotic-arm workspace. These include laser-induced breakdown spectroscopy (LIBS), remote time-resolved Raman and luminescence spectroscopies, and visible and infrared (VISIR; separately referred to as VIS and IR) reflectance spectroscopy. A remote micro-imager (RMI) provides high-resolution color context imaging, and a microphone can be used as a stand-alone tool for environmental studies or to determine physical properties of rocks and soils from shock waves of laser-produced plasmas. SuperCam is built in three parts: The mast unit (MU), consisting of the laser, telescope, RMI, IR spectrometer, and associated electronics, is described in a companion paper. The on-board calibration targets are described in another companion paper. Here we describe SuperCam's body unit (BU) and testing of the integrated instrument. The BU, mounted inside the rover body, receives light from the MU via a 5.8 m optical fiber. The light is split into three wavelength bands by a demultiplexer, and is routed via fiber bundles to three optical spectrometers, two of which (UV and violet; 245-340 and 385-465 nm) are crossed Czerny-Turner reflection spectrometers, nearly identical to their counterparts on ChemCam. The third is a high-efficiency transmission spectrometer containing an optical intensifier capable of gating exposures to 100 ns or longer, with variable delay times relative to the laser pulse. This spectrometer covers 535-853 nm ( 105 - 7070 cm − 1 Raman shift relative to the 532 nm green laser beam) with 12 cm − 1 full-width at half-maximum peak resolution in the Raman fingerprint region. The BU electronics boards interface with the rover and control the instrument, returning data to the rover. Thermal systems maintain a warm temperature during cruise to Mars to avoid contamination on the optics, and cool the detectors during operations on Mars. Results obtained with the integrated instrument demonstrate its capabilities for LIBS, for which a library of 332 standards was developed. Examples of Raman and VISIR spectroscopy are shown, demonstrating clear mineral identification with both techniques. Luminescence spectra demonstrate the utility of having both spectral and temporal dimensions. Hypothesis: Water and oil inhibition treatment is essential for protecting natural and artificial stone surfaces. Bioinspired super-antiwetting surfaces with ''lotus effect", together with superoleophobic properties, can be achieved combining very low-surface-energy materials and suitable surface roughness. Exploiting the natural roughness of stone surfaces, the simple and inexpensive fabrication of superamphiphobic surfaces through the coating dispersion deposition is expected. It seems the ideal method for the safeguard of contemporary and historical constructions, since the physical, chemical and aesthetic properties can be maintained. Experiments: The new coating agent (3-perfluroether-amidopropylsilane) was synthesized via one-step amidation. Hydrophobicity, robustness and environmental durability were systematically studied on stone surfaces through several tests: contact angle (CA), contact angle hysteresis (CAH), water inhibition efficiency, vapor diffusivity, chemical and mechanical resistance, artificial and field-exposure ageing. Findings: The as-prepared coating demonstrated superamphiphobicity (oil and water CA > 150 with CAH < 10) on stones with low and high porosity. Moreover, it manifested very high water inhibition efficacy while maintaining high vapor diffusivity and aesthetic properties of substrates. The superhydrophobic coating showed good robustness towards corrosive chemical agents, peeling, mechanical abrasion, water immersion and environmental weathering, thereby permitting various outdoor applications, including stone protection in rainy regions where acid rain is also present.

After an incremental development which took place over four decades, X-ray imaging has become an important tool for non-destructive testing and evaluation. Computed Tomography (CT) in particular beholds the power of determining the location of flaws and inclusions (e. g. in castings and composites) in three-dimensional object coordinates. Therefore, and thanks to a speed-up of the measurement, CT is now routinely considered for in-line inspection of electronics, castings and composites. When precision and not speed is important, Micro-CT (μCT) can be employed for Dimensional Measurements (DM, e. g. quality assurance and shape verification), as well as for in situ testing, and for characterizing micro-structures in metals and composites. Using appropriate image processing and analysis μCT can determine the local fibre orientation in composites, the granular morphology of battery cathodes or the inter-connectivity of certain phases in casting alloys. Today, the large variety of X-ray instruments and methods poses an application problem which requires experience and a lot of knowledge for deciding which technique applies best to the task at hand. Application-specific guidelines exist for X-ray radiography testing (RT) only, whereas standardization has been applied to CT, unfortunately leaving out high resolution sub μ CT, and nano-CT. For the latter exist an equally high number of NDT applications, however these instruments still necessitate a profound expertise. The task is to identify key industrial applications and push CT from system standardization to application specific automation.

This study reports a probabilistic insight into the stellar-mass and supernovae (SNe) explosion energy of five CEMP-no stars' possible progenitors. As such, a direct comparison between the abundance ratios [X/Fe] of the light-elements and the predicted nucleosynthetic yields of SN of high-mass metal-free stars has been performed. This comparison suggests that the possible progenitors have a stellar-mass range of 11 - 22 M-circle dot and explosion energies of 0.3 - 1.8 x 10(51) erg. In addition, we investigate the kinematic signatures, derived from Gaia DR2, to conclude that these five CEMP-no stars have halo-like kinematic and do not enter the outer-halo region. In addition, we link the abundance patterns with kinematic signatures to investigate the Gaia-Sausage and Gaia-Sequoia memberships. This chemo-dynamical analysis suggests that these CEMP-no stars are neither Gaia-Sausage nor Gaia-Sequoia remnant stars, but another accretion event might be responsible for the contribution of these stars to the Galactic Halo.

In this work, the interactions of atomic-scale chemistry and strain rate in affecting the deformation response of a Zr-based metallic glass was studied by varying the concentration of oxygen dissolved into the local structure. Compression of micropillars over six decades of strain rate uncovered a remarkable reversal of the strain rate sensitivity from negative to positive above ~ 5 s$^{���1}$ due to a delocalisation of shear transformation events within the pre-yield linear regime for both samples, while a higher oxygen content was found to generally decrease the strain rate sensitivity effect. It was also identified that the shear band propagation speed increases with the actuation speed, leading to a transition in the deformation behaviour from serrated to apparent non-serrated plastic flow at ~ 5 s$^{���1}$., Published by Cambridge Univ. Mountain snow is a fundamental freshwater supply in the arid regions. Climate warming alters the timing of snowmelt and shortens the snow cover duration, which greatly influences the regional climate and water management. However, a reliable estimation of snow mass in the Tianshan Mountains (TS) is still unclear due to the scarcity of extensive continuous surface observations and a complex spatial heterogeneity. Therefore, a long-time snow simulation from 1982 to 2018 was performed in WRF/Noah-MP to quantify snow mass in the TS, forced by ERA5 reanalysis data and real-time updated leaf area index and green vegetation fraction. Meanwhile, March snow mass (close to the annual peak snow mass), snow cover fraction (SCF), and their associated trends were investigated in the TS. The results indicated a good accuracy of the simulated snow water equivalent (root mean square error [RMSE]: 7.82 mm/day) with a slight overestimation (2.84 mm/day). Compared with ERA5 data set, the RMSE and mean bias of the daily snow depth from WRF/Noah-MP downscaling were significantly reduced by 95.74% and 93.02%, respectively. The climatological March snow mass measured 97.85 (+/- 16.60) Gt in the TS and exhibited a negligible tendency during the study period. The total precipitation during the cold season controlled the variations of March snow mass. The increased precipitation in the high-altitude regions contributed to an extensive snow mass, which could offset the loss in the TS lowland. In contrast, rapidly rising air temperature caused a significant reduction of March SCF, particularly in the Southern TS.

The transient nature of the internal pore structure of particulate wall flow filters, caused by the continuous deposition of particulate matter, makes studying their flow and filtration characteristics challenging. In this article we present a new methodology and first experimental demonstration of time resolved in-situ synchrotron micro X-ray computed tomography (micro-CT) to study aerosol filtration. We directly imaged in 4D (3D plus time) pore scale deposits of TiO2 nanoparticles (nominal mean primary diameter of 25 nm) with a pixel resolution of 1.6 μm. We obtained 3D tomograms at a rate of ∼1 per minute. The combined spatial and temporal resolution allows us to observe pore blocking and filling phenomena as they occur in the filter’s pore space. We quantified the reduction in filter porosity over time, from an initial porosity of 0.60 to a final porosity of 0.56 after 20 min. Furthermore, the penetration depth of particulate deposits and filtration rate was quantified. This novel image-based method offers valuable and statistically relevant insights into how the pore structure and function evolves during particulate filtration. Our data set will allow validation of simulations of automotive wall flow filters. Evolutions of this experimental design have potential for the study of a wide range of dry aerosol filters and could be directly applied to catalysed automotive wall flow filters.

© 2020 The Authors Fretting fatigue can significantly reduce the life of components, leading to unexpected in-service failures. This phenomenon has been studied for over a century, with significant progress being made during the past decade. There are various methods that have been used to study fretting fatigue cracks in order to gain a greater understanding of the effects of fretting fatigue. Destructive methods are traditionally used to observe fretting fatigue cracks. Although useful in determining crack location, crack length, crack propagation modes, crack path and shape, it is not efficient or reliable for time based measurements. Non-destructive testing has developed in recent years and now in-situ monitoring can be used during testing in order to increase the understanding of fretting fatigue. This paper presents a review of non-destructive testing techniques used in-situ during fretting fatigue testing, which are compared in order to conclude the suitability of each technique. Recent developments in non-destructive techniques that could be also applied for fretting fatigue tests are also discussed, as well as recommendations for future research made.

Orchids are highly dependent on mycorrhizal fungi for seed germination and subsequent growth to a seedling as they provide essential carbon, water, and mineral nutrients to developing seeds. Although there is mounting evidence that orchid seeds are often colonized by multiple fungi simultaneously, most in vitro germination experiments focus on mycorrhizal monocultures and little is known about how mycorrhizal assemblages affect seed germination and growth of seedlings. In this study, we compared the effects of mycorrhizal monocultures and co-cultures on seed germination and seedling growth of the epiphytic orchid Dendrobium nobile. In situ baiting was used to isolate mycorrhizal fungi from protocorms for germination experiments. Germination experiments were conducted under two light regimes for 90 days. In total, five fungal strains were isolated from protocorms of D. nobile, indicating that the species was not highly specific to its fungal partners. Four strains (JC-01, JC-02, JC-04, and JC-05) belonged to the Serendipitaceae and one (JC-03) to the Tulasnellaceae. In vitro germination experiments showed that germination percentages were higher under light-dark conditions than under complete dark conditions, supporting previous findings that light facilitates germination in epiphytic orchids. While all strains were able to induce protocorm formation and growth into the seedling stage, large differences between fungal strains were observed. Co-cultures did not result in significantly higher seed germination percentages and seedling development than monocultures. Taken together, these results demonstrate that effects of fungal assemblages are not predictable from those of component species, and that more work is needed to better understand the role of fungal assemblages determining seed germination and subsequent growth under natural conditions. ispartof: FRONTIERS IN PLANT SCIENCE vol:11 ispartof: location:Switzerland status: published

Electrochemical water splitting is a promising approach to generate ‘green’ hydrogen. The efficiency of this process relies on the effectiveness of the electrocatalysts used. The electro-kinetics of the hydrogen evolution reaction (HER) is highly pH dependent and conventional catalysts typically are expensive and rare platinum-based materials. The development of low-cost, multi-component electrocatalysts, where each of the components has a synergistic effect, can be an effective approach to improve kinetics. Herein, a series of transition metal (Fe, Mn, Co, and Ni)-modified molybdenum carbides in a nitrogen-doped carbon matrix (TM-Mo2C@NCF) are synthesised to maximise exposed active sites. Among them, Fe-Mo2C@NCF delivers the best-in-class HER performance over a wide range of electrolytes. Tafel slopes of 76, 109 and 110 mV dec−1 and overpotentials of 65, 130 and 129 mV at 10 mA cm−2 were obtained in 1.0 M KOH, 1.0 M phosphate buffer solution (PBS) and 0.5 M H2SO4, respectively. The computational study further indicates that the synergistic electronic modulation co-activated by Fe and N dopants in Fe-Mo2C@NCF can reduce the Gibbs free energy of H adsorption (ΔGH*) and render the Mo–Mo bridge site the most energetically favorable adsorption site for the H* intermediate, which contributes to an increased HER performance.

Magnesium alloys are primarily associated with complex forming mechanisms, which yield ductility at high temperatures. In sheet metal forming, high triaxiality stress states that favor the ductile damage mechanisms of void formation and growth are known to malleable metals. The formulation of coupled damage models has so far failed, due to the incomplete experimental determination of damage parameters for magnesium AZ31 thin sheet. A quantitative investigation was conducted to determine the ductile damage behavior of twin-roll cast, hot rolled, and annealed AZ31 thin sheet. Results on the mechanisms of void nucleation-, coalescence- and growth-rate were established at temperatures ranging from room temperature to 350 &deg, C. In-situ tensile tests were carried out in a scanning electron microscope with three different specimen types: Simple tension specimens, notched specimens for high triaxiality stress state testing, and shear specimens. Through a comparative analysis of local strains measured by digital image correlation and local void volume fractions determined through post-mortem analysis of specimen cross-sections, GTN (Gurson&ndash, Tvergaard&ndash, Needleman) model-based material parameters were determined by experiment, representing a novel departure in the magnesium research landscape. The procedure developed in this context should also be transferable to other metals in the form of thin sheets.

International audience; The synchrotron radiation–computed tomography (SR-CT) and digital volume correlation (DVC) methods were used to investigate the damage micromechanisms of lost foam casting (LFC) A319 alloy in low-cycle fatigue (LCF). LCF tests with SR-CT in-situ observations allow visualizing the damage evolution process in the bulk. DVC measures the mechanical fields and, thus, allows establishing the relations between crack initiation and propagation, mechanical fields, and microstructure. Cracks initiate at and propagate along hard inclusions due to strain localizations. The damage process, i.e., crack initiation and propagation, can be considered as a series of failure events of hard inclusions under strain localizations. The pores’ size, shape, location, and number were observed to have an influence on crack initiation, while the interconnected hard inclusion networks guarantee the continuous failure events of hard inclusions and, thus, provide crack propagation paths.

Poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) is one of the most studied conductive polymers, holding great potential in many applications such as thermoelectric generators, solar cells, and memristors. Great efforts have been invested in trying to improve its mechanical and electrical properties and to elucidate the structure-property relationship. In this work, a systematic and quantitative study of the effect of solvent polarity and solution processing on the film structure and conductivity is presented. By using grazing-incidence wide-angle X-ray scattering (GIWAXS) together with atomic force microscopy (AFM), the importance of the quality of the PEDOT crystal packing is highlighted as a key factor to reach improved electrical conductivity, rather than the overall degree of crystallinity. Moreover, the (re)structuring mechanisms occurring during the film formation and film exposure processes are also studied by in situ GIWAXS. Different intermediate precursor stages and different pathways to reach improved crystallinity are reported depending on the used solvent. The structural results are interpreted looking at the solvent nature and the PSS/solvent affinity. With this contribution, a guidance is hoped to be given not only on how to improve the PEDOT:PSS electrical conductivity, but also on how to tune the film structural or electrical property for different applications.

ADIOS 2 addresses scientific data management needs ranging from scalable I/O in supercomputers, to data analysis in personal computer and cloud systems. Version 2 introduces a unified application programming interface (API) that enables seamless data movement through files, wide-area-networks, and direct memory access, as well as high-level APIs for data analysis. The internal architecture provides a set of reusable and extendable components for managing data presentation and transport mechanisms for new applications. ADIOS 2 bindings are available in C++11, C, Fortran, Python, and Matlab and are currently used across different scientific communities. ADIOS 2 provides a communal framework to tackle data management challenges as we approach the exascale era of supercomputing.

International audience; In this study, we describe an experimental setup and a new approach for operando investigation of structural evolution of materials during wear and friction. The setup is particularly suited for testing various friction pairs, including those in which both rubbing bodies are made of metals. The developed device allows circumventing the problems related to significant scattering of X-rays produced by metals and makes it possible using “real samples” in synchrotron beamlines operating in reflection mode. To demonstrate the capabilities of the device and the proposed new approach, an iron-based massive sample was subjected to thousands of friction cycles using a cemented carbide pin. The material was probed with synchrotron X-ray radiation within a few milliseconds after leaving the friction zone. The results of the microstructural and structural analysis, as well as results obtained from diverse mathematical models, allowed us to evaluate several features, including gradual accumulation of defects, microstructural refinement, dislocation density changes, surface layer oxidation, as well as several other phenomena caused by the dry sliding friction process. Mainly, it was possible to conclude that the process of wear occurred due to the cooperative action of oxidation and plastic deformation, which began during the first cycle of frictional interaction and was manifested in increasing the dislocation density, whose type was changed gradually during testing. The number of defects quickly reached a threshold value and subsequently fluctuated around it due to periodically repeated processes of defect accumulation and stress relaxation resulting from material wear. It was also observed that friction led to the quick formation of a mechanically mixed layer, consisting of the sample material and a mixture of two types of iron oxide – hematite and magnetite. The delamination of this layer was probably the primary wear mechanism.

Dissolved oxygen (DO) is an important parameter to monitor in processes applicable in, for example, water technology and fermentation. In this study, we report the manufacturing of a membrane-less amperometric sensor that is based on the electrocatalytic reduction of oxygen. The sensor was tested in pH-neutral KNO3 solutions and in a microbial fermentation to monitor the consumption of dissolved oxygen. The nitrogen and boron-doped reduced graphene oxide (N,B-HRGO) is used as an electrocatalyst for oxygen reduction. One step co-doping of nitrogen and boron on graphene oxide is performed using the hydrothermal method. The sensor responded linearly to the DO concentration. A sensitivity of 0.2 &micro, A/mg&middot, L&minus, 1 O2 is obtained for the DO concentration of 1.5 to 10 mg&middot, 1 O2. The membrane-less N,B-HRGO based DO sensor is successfully tested in an Amycolotopsis methanolica fermentation by monitoring the dissolved oxygen in real-time. The sensor detected the consumption of oxygen during the growth of A. methanolica, which shows the compatibility of N,B-HRGO as electrode material for amperometric measurement of dissolved oxygen in aerobic fermentation. This membrane-less amperometric sensor can be used to fabricate microdevices for microbioreactor applications.

Inherent substrate selectivity is reported for the thermal RuO4 (ToRuS)/H-2 gas atomic layer deposition (ALD) process on H-terminated Si (Si-H) versus SiO2. In situ spectroscopic ellipsometry (SE) detected Ru growth from the first cycle on blanket Si-H, whereas on blanket SiO2, 60 cycles were needed to detect growth. Area-selective growth was evaluated on a patterned substrate with 1-10 mu m wide Si H lines separated by 10 mu m wide SiO2 regions. Ex situ planar scanning electron microscopy and cross-sectional high resolution transmission electron microscopy measurements showed that a smooth, continuous Ru film of 4.5 nm could be deposited on Si-H, with no Ru detected on SiO2. The proposed mechanism behind the inherent substrate selectivity is the oxidation of the Si-H surface by RuO, which was confirmed by in vacuo X-ray photoelectron spectroscopy (XPS) experiments. A methodology to enhance the nucleation of the RuO4/H-2 gas process on oxide substrates is also reported. In situ SE and in vacuo XPS experiments show that the nucleation delay on SiO2 can be completely removed by exposing the surface to trimethylaluminum (TMA) just before the start of the ALD process. We found evidence that the TMA pulse makes the oxide surface reactive toward RuO4, by introduction of surface methyl groups, which can be combusted by RuO4. As TMA is known to be reactive toward many oxide substrates, this methodology presents a way to achieve Ru metallization of virtually any surface. Therefore, one can either (i) use the RuO4/H-2 gas process to coat nonoxidized surfaces selectively with Ru or (ii) use TMA-priming by which one can bypass the selectivity and coat a wide variety of surfaces nonselectively with Ru.

Highlights • Turbidity currents transport large amounts of reactive iron minerals and associated phosphorus into the Black Sea deep basin • Sediments on the southwestern shelf enriched in reactive ferrous and ferric iron are suggested as turbidite source area • Iron sulfidation in and phosphorus release from turbidity currents in deep waters affect iron, phosphorus and sulfur budgets • The original redox signature of the source sediment can be preserved in turbidites for longer than expected Abstract The biogeochemical cycles of iron, phosphorus and sulfur are intimately linked and the fate of these elements is highly redox-dependent. Under anoxic conditions, iron is reduced to Fe(II), for an important part driven by reaction with sulfide. Reduction and sulfidation diminish the affinity of iron for phosphorus, thereby affecting sedimentary phosphorus retention. The coupled cycling of iron-phosphorus-sulfur as a function of redox conditions thereby helps control nutrient availability and primary productivity in marine systems. The Black Sea is the world's largest permanently stratified basin with a strong gradient from oxic surface waters to anoxic and strongly sulfidic deep waters, and is therefore well-suited to investigate redox-dependent changes in coupled iron-phosphorus-sulfur cycling. The presence of sulfide in the deep Black Sea alters the chemical speciation of iron (Fe) and phosphorus (P) in particulate matter in the water column and sediment. These alterations and their impact are poorly constrained for turbidites, which are deposits formed by mass transport from the continental slope into the deep basin of the Black Sea through turbidity currents. Here, we compare the geochemistry of sediments on the southwestern (SW) Black Sea slope overlain by oxygenated waters (234 m below sea surface, mbss) with that of a sediment record with turbidite intervals from the sulfidic SW deep basin (2169 mbss). The aims were to investigate the potential of SW slope sediment as source material for turbidites, and to assess the geochemical impact of reaction with sulfide. The bulk chemistry (Al, CaCO3, Fe) of the SW deep Black Sea turbidite intervals was similar to that of SW Black Sea slope sediments (234 m water depth), suggesting that the latter are source material for deep SW Black Sea turbidites. The source sediment is characterized by strong enrichments in highly reactive Fe(III) and Mn phases, high rates of sulfate reduction and the presence of methane. Similar total Fe contents in the slope and deep turbidite sediments suggest that Fe is efficiently retained in the sediment. In contrast, Mn is depleted during down-slope transport and P contents in the turbidite intervals are about 50% lower than in the slope sediment, indicating significant P release during and after turbidite deposition. Calculations using the obtained data and conservative estimates of the areal extent of turbidite deposition in the SW Black Sea indicate that turbidity currents have the potential to impact coupled Fe-P-S biogeochemical cycling by boosting Fe input, dissolved P accumulation, sulfide oxidation and FeS2 burial in the deep Black Sea.

Duplex stainless steels (DSSs) consist of ferrite and austenite phases with approximately equal volume fraction. They exhibit combined superior mechanical strength and corrosion resistance, therefore are increasingly used in various applications. However, under certain conditions, passivity breakdown may occur, leading to corrosion initiation, which is often related to weak points in the heterogeneous microstructure. To understand the influence of microstructure on the passivity and breakdown of DSSs requires local probing techniques that can be used in situ, so that corrosion initiation process can be correlated to the microstructure. Recent studies employing advanced scanning probe microscopy and synchrotron-based techniques, in combination with electrochemical measurements, have contributed to a deep understanding of the passive film, passivity breakdown, and corrosion initiation of DSSs, as well as the influence of microstructural and environmental factors. This mini review presents a short summary of recent literature focusing on the studies utilizing local probing techniques and synchrotron-based analyses.

International audience; The hybridization chain reaction (HCR) is a simple and sensitive method for quantifying nucleic acids. Current approaches cannot combine a washing-free sensing format with multiplexed target quantification at low concentrations, which would be highly desirable for detection both in solution and in situ. Here, we demonstrate the implementation of time-gated Förster resonance energy transfer (TG-FRET) between terbium donors and dye acceptors into HCR for multiplexed quantification of microRNAs (miR-20a and miR-21) and their DNA analogues. HCR-TG-FRET provided washing-free nucleic acid quantification with very low limits of detection down to 240 amol (1.7 pM) of microRNA and 123 amol (0.88 pM) of DNA. Efficient distinction from very homologous microRNAs demonstrated high target specificity. Multiplexing with a single measurement, a single excitation wavelength, and a single FRET pair allowed for a simultaneous quantification of miR-20a and miR-21 at concentrations between 30 and 300 pM from the same sample. HCR-TG-FRET showed similar performance for serum-free and serum-containing samples without the use of RNase inhibitors. Our results present a significant improvement in current HCR approaches regarding simplicity, sensitivity, and multiplexing. The versatile diagnostic performance of HCR-TG-FRET even in challenging biological environments presents an important advantage for advanced nucleic acid biosensing.

Electromechanical actuation in piezoceramics is usually enhanced by creating chemically homogeneous materials with structurally heterogeneous morphotropic phase boundaries, leading to abrupt changes in ion displacement directions within the perovskite unit cell. In the present study, an alternative mechanism to enhance electromechanical coupling is found in both chemically and structurally heterogeneous BiFeO3−BaTiO3 lead-free piezoceramics. Such a mechanism is observed in a composition exhibiting core-shell type microstructure, associated with donor-type substitution of Ti4+ for Fe3+, and is primarily activated by thermal quenching treatment. Here, we describe the use of in-situ high-energy synchrotron X-ray powder diffraction upon the application of a high electric field to directly monitor the ferroelectric and elastic interactions between these composite-like components, formed as core and shell regions within grains. Translational short or long-range ordering is observed in the BiFeO3‒depleted shell regions which undergo significant structural alterations from pseudocubic Pm-3m relaxor-ferroelectric in slow-cooled ceramics to rhombohedral R3c or R3m with long-range ferroelectric order in the quenched state. The strain contributions from each component are calculated, leading to the conclusion that the total macroscopic strain arises predominantly from the transformed shell after quenching. Such observations are also complemented by investigations of microstructure and electrical properties, including ferroelectric behaviour and temperature-dependent dielectric properties.

The synthesis of Au nanowires (NWs) in hexane solution of HAuCl4·3H2O, oleylamine and triisopropylsilane at room temperature was "in situ" monitored by means of X-ray absorption fine structure spectroscopies and small-angle X-ray scattering to determine, under identical synthesis conditions, both the changes in the oxidation state of gold atoms and the evolution of the size and shape of the objects involved in the formation of Au NWs. We propose a multistage process for the formation of the NWs: first, Au(III) atoms form a planar-square geometry complex that is continuously reduced to give Au(I) disk-like structures with diameters bigger than that of the final NWs. In a second stage, characteristic length/thickness ratio of these disk-like objects increases to form cylinders, presumably through aurophilic interactions between Au(I) centers and stacking of the disks. When most of the Au atoms have been reduced to Au(I), the reduction to Au(0) begins (third stage), and the NWs grow to form an hexagonal arrangement, separated by a bilayer of oleylamine molecules (fourth stage). The cycling of carbon (C) by benthic organisms is a key ecosystem function in the deep sea. Pulse-chase experiments are designed to quantify this process, yet few studies have been carried out using abyssal (3500-6000 m) sediments and only a handful of studies have been undertaken in situ. We undertook eight in situ pulse-chase experiments in three abyssal strata (4050-4200 m water depth) separated by tens to hundreds of kilometers in the eastern Clarion-Clipperton Fracture Zone (CCFZ). These experiments demonstrated that benthic bacteria dominated the consumption of phytodetritus over short (similar to 1.5 d) time scales, with metazoan macrofauna playing a minor role. These results contrast with the only other comparable in situ abyssal study, where macrofauna dominated phytodetritus assimilation over short (2.5 d) time scales in the eutrophic NE Atlantic. We also demonstrated that benthic bacteria were capable of converting dissolved inorganic C into biomass and showed that this process can occur at rates that are as high as the bacterial assimilation of algal-derived organic C. This demonstrates the potential importance of inorganic C uptake to abyssal ecosystems in this region. It also alludes to the possibility that some of the C incorporation by bacteria in our algal-addition studies may have resulted from the incorporation of labeled dissolved inorganic carbon initially respired by other unstudied organisms. Our findings reveal the key importance of benthic bacteria in the short-term cycling of C in abyssal habitats in the eastern CCFZ and provide important information on benthic ecosystem functioning in an area targeted for commercial-scale, deep-sea mining activities.

Dendritic microstructural evolution during the solidification of Mg-Zn alloys was investigated as a function of Zn concentration using in situ synchrotron X-ray tomography. We reveal that increasing Zn content from 25 wt% to 50 wt% causes a Dendrite Orientation Transition (DOT) from a six-fold snow-flake structure to a hyper-branched morphology and then back to a six-fold structure. This transition was attributed to changes in the anisotropy of the solid-liquid interfacial energy caused by the increase in Zn concentration. Further, doublon, triplon and quadruplon tip splitting mechanisms were shown to be active in the Mg-38 wt%Zn alloy, creating a hyper-branched structure. Using the synchrotron tomography datasets, we quantify, for the first time, the evolution of grain structures during the solidification of these alloys, including dendrite tip velocity in the mushy zone, solid fraction, and specific surface area. The results are also compared to existing models. The results demonstrate the complexity in dendritic pattern formation in hcp systems, providing critical input data for the microstructural models used for integrated computational materials engineering of Mg alloys.

Arctic Ocean surface sea-ice conditions are linked with the deep sea benthic oxygen fluxes via a cascade of interdependencies across ecosystem components such as primary production, food supply, activity of the benthic community, and their functions. Additionally, each ecosystem component is influenced by abiotic factors such as light availability, temperature, water depth, and grain size structure. In this study, we investigated the coupling between surface sea-ice conditions and deep-sea benthic remineralization processes through a cascade of interdependencies in the Fram Strait. We measured sea-ice concentrations, a variety of different sediment characteristics, benthic community parameters, and oxygen fluxes at 12 stations of the LTER HAUSGARTEN observatory, Fram Strait, at water depths of 275–2500 m. Our investigations reveal that the Fram Strait is bisected into two long-lasting and stable regions: (i) a permanently and highly sea-ice-covered area and (ii) a seasonally and low sea-ice-covered area. Within the Fram Strait ecosystem, sea-ice concentration and water depth are two independent abiotic factors, controlling the deep-sea benthos. Sea-ice concentration correlated with the available food and water depth with the oxygen flux. In addition, both abiotic factors sea-ice concentration and water depth correlate with the macrofauna biomass. However, at water depths > 1500 m the influence of the surface sea-ice cover is minimal with water depth becoming more dominant. Benthic remineralization across the Fram Strait on average is ∼ 1 mmol C m−2 d−1. Our data indicate that the portion of newly produced carbon that is remineralized by the benthos is 5 % in the seasonally low sea-ice-covered eastern part of Fram Strait but can be 14 % in the permanently high sea-ice-covered western part of Fram Strait. Here, by comparing a permanently sea-ice-covered area with a seasonally sea-ice-covered area, we discuss a potential scenario for the deep-sea benthic ecosystem in the future Arctic Ocean, in which an increased surface primary production may lead to increasing benthic remineralization at water depths

Chip-package interaction (CPI) and the related thermomechanical stress in microchips increase the risk of failure in on-chip interconnect stacks, caused by delamination along Cu/dielectrics interfaces (adhesive failure) and fracture in dielectrics (cohesive failure). High-resolution transmission X-ray microscopy (TXM) is a unique technique to image crack propagation in on-chip interconnect stacks. The visualization of crack evolution in Cu/low-k Backend-of-Line (BEoL) structures is demonstrated using an experimental setup which combines high-resolution X-ray imaging with mechanical loading. The application of an indenter manipulator at the TXM beamline of the synchrotron radiation source BESSY II provides an unprecedented level of details on the fracture behavior of microchips. This in-situ experiment allows to identify the weakest layers and interfaces and to evaluate the robustness of the BEoL stack against CPI.

Metal-organic frameworks (MOFs) have revolutionized the potential applications of nanoporous materials. One of the most recent and promising applications of these materials is their use as supports for enzyme immobilization. In this context, the in-situ (one-step) methodologies, which do not require the use of MOFs with pores larger than the enzyme to be immobilized, seem to be particularly encouraging. This work presents a systematic study of the semi-crystalline Fe-BTC MOF material (commercialized as Basolite F300) employed as support of the enzymes laccase and lipase through either in-situ or post-synthesis methodology. The presence of the enzyme in the resultant solid biocatalysts was proved by CHNS chemical analysis, thermogravimetric analysis, Bradford assays and by SDS-PAGE electrophoresis. The enzymatic activity of the resultant Fe-BTC-based biocatalysts was also tested. The in-situ approach is particularly relevant due to various reasons: (i) the enzyme immobilization is given in one step; (ii) it is rapid (10 min); (iii) it is very efficient in terms of encapsulation capacity (≥98% for laccase and ≥87% for lipase); (iv) the enzymes are fully retained and no leaching is observed after an initial release of only around 10% of the enzyme molecules weakly immobilized; and (v) the activity of the retained enzyme can be substantially maintained (97% with respect to the free enzyme in the case of lipase). Any of these parameters systematically improves these given by the post-synthesis (two-step) approach. Sufficient liquid feeding under constrained solidification conditions like, those experienced in welding and casting, is vital to avoid solidification cracking. We present the results of unique in-situ experimental observations of liquid feeding in a solidifying steel melt pool. Liquid feeding was observed in the inter-cellular regions during the terminal stage of solidification. An average liquid flow speed of 450–500 μm s−1 was found. A pressure difference of the order of 104 Pa is calculated to cause the liquid flow. The rate of solidification shrinkage and the rate of deformation were found to be less than the rate of liquid feeding.

RationaleLaser microdissection-liquid vortex capture/electrospray ionization mass spectrometry (LMD-LVC/ESI-MS) has potential for on-line classification of tissue but an investigation into what analytical conditions provide best spectral differentiation has not been conducted. The effects of solvent, ionization polarity, and spectral acquisition parameters on differentiation of mouse brain tissue regions are described.MethodsIndividual 40 x 40m microdissections from cortex, white, grey, granular, and nucleus regions of mouse brain tissue were analyzed using different capture/ESI solvents, in positive and negative ion mode ESI, using time-of-flight (TOF)-MS and sequential window acquisitions of all theoretical spectra (SWATH)-MS (a permutation of tandem-MS), and combinations thereof. Principal component analysis-linear discriminant analysis (PCA-LDA), applied to each mass spectral dataset, was used to determine the accuracy of differentiation of mouse brain tissue regions.ResultsMass spectral differences associated with capture/ESI solvent composition manifested as altered relative distributions of ions rather than the presence or absence of unique ions. In negative ion mode ESI, 80/20 (v/v) methanol/water yielded spectra with low signal/noise ratios relative to other solvents. PCA-LDA models acquired using 90/10 (v/v) methanol/chloroform differentiated tissue regions with 100% accuracy while data collected using methanol misclassified some samples. The combination of SWATH-MS and TOF-MS data improved differentiation accuracy.ConclusionsCombined TOF-MS and SWATH-MS data differentiated white, grey, granular, and nucleus mouse tissue regions with greater accuracy than when solely using TOF-MS data. Using 90/10 (v/v) methanol/chloroform, tissue regions were perfectly differentiated. These results will guide future studies looking to utilize the potential of LMD-LVC/ESI-MS for tissue and disease differentiation.

Understanding and:controlling the. olymorphism of manganese dioxide (MnO2) is of vital importance in many nanoscale applications. Here in situ powder Xray diffraction (PXRD) in 'combination with in situ total X-ray scattering are used to reveal the formation mechariisin as well as polymorph-evolution of MnO2 under hydrothermal synthesis conditions. A "PXRD invisible" amorphous phase with a local structure :resembling alpha-MnO2 (denoted alpha-MnO2:(A)) is observed at all reaction stages, and it never fully disappears from the reaction solution. The MnO2 phase evolution involves initial formation:of delta-MnO2, which transforms to alpha-MnO2, and then subsequently to beta-MnO2. The phase transformations between-different polymorphs, do not involve dissolution-recrystallization, but they occur via solid-state mechanisms. However, the amorphous alpha-MnO2(A) phase, plays a key role since it is consumed in growing both the alpha-and beta-MnO2 polymorphs. Overall, the polymorphism of the crystalline product can be controlled through reaction time and temperature to forth either nanocrystalline and-disordered delta-MnO2, nanocrystalline alpha-MnO2, or nanocrystalline beta-MnO2. At the lowest temperature (200 degrees C) the very early growth of alpha-MnO2 appears to be by oriented attachment along (110) crystal planes of primary nanorbds, but this is quickly followed by rapid growth along the c-direction supported by consumption of alpha-MnO2(A).

Accurate assessments of forest resources rely on ground truth data that are collected via in-situ measurements, which are fundamental for all other statistical- and/or remote-sensing-based deductions on quantified forest attributes. The major bottleneck of the current in-situ observation system is that the data collection is time consuming, and, thus, limited in extent, which potentially biases any further inferences made. Consequently, conventional field-data-collection approaches can hardly keep pace with the coverage, scale and frequency required for contemporary and future forest inventories. In-situ measurements from mobile platforms seem to be a promising technique to solve this problem and are estimated at least 10 times faster than static techniques (e.g., terrestrial laser scanning, TLS) at the plot level. However, the mobile platforms are still at the very early stages of development, and it is unclear which three-dimensional (3D) forest measurements the mobile systems can provide and at what accuracy. This study presents a quantitative evaluation of the performance of mobile platforms in a variety of forest conditions and through a comparison with state-of-the-art static in-situ observations. Two mobile platforms were used to collect field data, where the same laser-scanning system was both mounted on top of a vehicle and wore by an operator. The static in-situ observation from TLS is used as a baseline for the evaluation. All point clouds involved were processed through the same processing chain and compared to conventional manual measurement. The evaluation results indicate that the mobile platforms can assess homogeneous forests as well as static observations, but they cannot yet assess heterogeneous forest as required by practical applications. The major challenge is twofold: mobile-data coverage and accuracy. Future research should focus on the robust registration techniques between strips, especially in complex forest conditions, since errors of data registration results in significant impacts ontree attributes estimation accuracy. In cases that the spatial inconstancy cannot be eliminated, attributes estimation in single strips, i.e., the multi-single-scan approach, is an alternative. Meanwhile, operator training deserves attention since the data quality from mobile platforms is partly determined by the operators’ selection of trajectory in the field.

The Coupled Model Intercomparison Project (CMIP) is now moving into its sixth phase and aims at a more routine evaluation of the models as soon as the model output is published to the Earth System Grid Federation (ESGF). To meet this goal the Earth System Model Evaluation Tool (ESMValTool), a community diagnostics and performance metrics tool for the systematic evaluation of Earth system models (ESMs) in CMIP, has been developed and a first version (1.0) released as open source software in 2015. Here, an enhanced version of the ESMValTool is presented that exploits a subset of Essential Climate Variables (ECVs) from the European Space Agency's Climate Change Initiative (ESA CCI) Phase 2 and this version is used to demonstrate the value of the data for model evaluation. This subset includes consistent, long-term time series of ECVs obtained from harmonized, reprocessed products from different satellite instruments for sea surface temperature, sea ice, cloud, soil moisture, land cover, aerosol, ozone, and greenhouse gases. The ESA CCI data allow 'extending the calculation of performance metrics as summary statistics for some variables and add an important alternative data set in other cases where observations are already available. The provision of uncertainty estimates on a per grid basis for the ESA CCI data sets is used in a new extended version of the Taylor diagram and provides important additional information for a more objective evaluation of the models. In our analysis we place a specific focus on the comparability of model and satellite data both in time and space. The ESA CCI data are well suited for an evaluation of results from global climate models across ESM compartments as well as an analysis of long-term trends, variability and change in the context of a changing climate. The enhanced version of the ESMValTool is released as open source software and ready to support routine model evaluation in CMIP6 and at individual modeling centers. (C) 2017 Elsevier Inc. A novel high-vanadium based hard composite coating was synthesized from premixed powders (V, Cr, Mo, Ti, Nb) on ductile iron (DI) substrate via atmospheric plasma beam surface alloying process. The graded coating can be divided into three distinct zones: upper alloyed zone (AZ) rich with spherical primary and eutectic submicron carbides, middle melted zone (MZ) with fine white iron structure embedded with high-carbon martensite and lower heat affected zone (HAZ) where martensite/ledeburite double shells were substantially formed. Spherical or bulk-like primary carbides with diameter
