Antarctica is estimated to contain as much as a quarter of earth's marine methane, however we have not discovered an active Antarctic methane seep limiting our understanding of the methane cycle. In 2011, an expansive (70 m × 1 m) microbial mat formed at 10 m water depth in the Ross Sea, Antarctica which we identify here to be a high latitude hydrogen sulfide and methane seep. Through 16S rRNA gene analysis on samples collected 1 year and 5 years after the methane seep formed, we identify the taxa involved in the Antarctic methane cycle and quantify the response rate of the microbial community to a novel input of methane. One year after the seep formed, ANaerobic MEthane oxidizing archaea (ANME), the dominant sink of methane globally, were absent. Five years later, ANME were found to make up to 4% of the microbial community, however the dominant member of this group observed (ANME-1) were unexpected considering the cold temperature (−1.8°C) and high sulfate concentrations (greater than 24 mM) present at this site. Additionally, the microbial community had not yet formed a sufficient filter to mitigate the release of methane from the sediment; methane flux from the sediment was still significant at 3.1 mmol CH4m−2d−1. We hypothesize that this 5 year time point represents an early successional stage of the microbiota in response to methane input. This study provides the first report of the evolution of a seep system from a non-seep environment, and reveals that the rate of microbial succession may have an unrealized impact on greenhouse gas emission from marine methane reservoirs.

Methane seeps provide biogeochemical and microbial heterogeneity in deep-sea habitats. In the Northeast (NE) Pacific Ocean recent studies have found an abundance of seeps at varying spatial separations and within distinct biogeochemical environments ranging in oxygen, depth, and temperature. Here, we examine eight newly discovered seeps and two known seeps covering 800 km and varying across 2000 m water depth to identify: (1) novel megafaunal communities in this geographical region; (2) variations in the microbiome of seep habitats across the margin; (3) spatial and biogeochemical drivers of microbial diversity at seeps. In addition to authigenic carbonates, clam beds, microbial mats, and exposed hydrates - we also observed Siboglinidae tube worm bushes and an anomalous deep-sea barnacle adding to the overall habitats known from the NE Pacific. The microbial communities showed high variability in their spatial distribution and community structure. The seep communities formed distinct groups that included multiple groups of anaerobic methane oxidizing Archaea (ANME; 1, 2ab, 2c, and 3), often co-occurring within one site – however, there were also other sites with clearly dominant members (e.g. ANME-1s at Nehalem Bank). Sulfide oxidizers were dominated by the non-mat forming Campylobacterales and even though vertical gradients in redox potential typify seep sediments, in two cases there was not a significant change in community structure across the top five cm of sediment. We posit that these patterns were driven by ‘bubble-turbation,’ and bioirrigation by megafauna. A surprising latitudinal trend was observed in species diversity and richness with increasing richness significantly correlated to increasing latitude. Overall, our results demonstrate that heterogeneity is ubiquitous in the seep biome, spanning all faunal classes, and that the understanding of seeps and the drivers of the community structure can be improved by studying seeps at a range of spatial scales.

A critical barrier to effective management of deep-sea resources is a lack of understanding by society of the benefits received from the oceans. To address this knowledge gap, we applied an iterative design-based research methodology to evaluate (1) how to effectively use an exhibit to increase public literacy of the deep sea over the short and long-term and (2) how visitors to a public science center perceive the deep sea. Using observations of visitor interactions and surveys of visitors, we evaluated three iterations of an exhibit that highlighted deep-sea ecosystem services and habitats as a case study of exhibit efficacy. Exhibits containing video and interactive components were effective in communicating deep-sea information that was retained by visitors over the long-term. For many visitors, the exhibit was their first introduction to the deep sea. Visitors agreed it is important to learn about the deep sea and expressed interest in learning more about deep-sea animals, habitats, resources, and benefits to humans. Visitors tended to agree with protection-oriented value statements and disagree with use-oriented value statements towards the deep sea. This study provides insight into how to effectively communicate policy-relevant information about the deep sea to an audience that has little to no prior knowledge of the ecosystem, yet who will be increasingly responsible for making use decisions of this habitat.

Methane seep habitats are widespread chemosynthesis-based ecosystems that span continental margins and interact with surrounding marine systems. With many of these habitats occurring below 200 m, seeps can serve as an important source of nutrients in otherwise food limited deep-sea environments. However, the potential for marauding megafauna to assimilate seep derived nutrition has been difficult to quantify. Here, we provide the first evidence of a commercially harvested species, Chionoecetes tanneri (tanner crab), assimilating chemosynthetic production. Although bulk isotope analysis of C. tanneri tissue indicated no quantifiable incorporation of seep-derived carbon or sulfur (mean δ13C, −18.5‰; mean δ34S, 19.5‰), depletions in 13C (δ13C as light as −38.8‰) were noted in fatty acid (FA) compounds. In addition, diagnostic biomarkers for seep bacteria, including 16:1ω6 and 18:1ω8c FA's, were found to have been assimilated by C. tanneri. Futher supporting a trophic link between the seep and the C. tanneri, seep associated bacteria and archaea were, in certain cases, the dominant taxa in the gut contents of the crabs. This work provides the first insights into a link between seep production and deep-sea ecosystem services, specifically fisheries production. In addition, it reveals a methodological bias that could exist in some trophic studies where bulk isotopes under-represent the role of seep nutrition in the diet of marauding animals.
