Root-lesion nematodes of the genus Pratylenchus are among the most important nematode pests that limit production of small-grain cereals. Four Pratylenchus species, viz. P. thornei, P. neglectus, P. penetrans, and P. crenatus, are considered of major economic significance in cereals of which P. thornei and P. neglectus are the most important and widely distributed species in cereal crops worldwide. However, P. thornei is more destructive causing estimated yield losses of up to 50% in the USA and 85% in Australia. This paper provides information regarding the global distribution of Pratylenchus species, yield loss due to their attack, their biology and pathogenic relation to plants, the research cutting edges in nematode identification of different Pratylenchus species, and their control through cultural practices and resistant varieties as correct identification of root-lesion nematodes can be difficult to achieve, particularly if a quick diagnosis is needed. In this context, in recent years, several molecular techniques for these Pratylenchus species have been developed such as quantitative PCR assays which are able to produce precise and rapid identification of several root-lesion nematodes species. So far, many global attempts have been made to control root-lesion nematodes in cereals, including cultural practices and development of resistant varieties. The use of resistant accessions is considered the most economically feasible and environmentally sustainable method. Resistance genes in several lines have been identified and are being used in numerous breeding programmes against root-lesion nematodes species.

Soil organisms are a crucial part of the terrestrial biosphere. Despite their importance for ecosystem functioning, few quantitative, spatially explicit models of the active belowground community currently exist. In particular, nematodes are the most abundant animals on Earth, filling all trophic levels in the soil food web. Here we use 6,759 georeferenced samples to generate a mechanistic understanding of the patterns of the global abundance of nematodes in the soil and the composition of their functional groups. The resulting maps show that 4.4 ± 0.64 × 1020 nematodes (with a total biomass of approximately 0.3 gigatonnes) inhabit surface soils across the world, with higher abundances in sub-Arctic regions (38% of total) than in temperate (24%) or tropical (21%) regions. Regional variations in these global trends also provide insights into local patterns of soil fertility and functioning. The root lesion nematodes Pratylenchus penetrans and P. thornei cause high yield losses in rain-fed wheat fields in Morocco, as well as worldwide. Growing resistant varieties is one of the most effective methods for controlling nematodes. Therefore, a collection of 14 lines of spring wheat and 11 lines of winter wheat (Triticum aestivum and T. durum), provided by CIMMYT, were screened for resistance to P. penetrans and P. thornei in tubes (15 × 20 × 120 mm3) under greenhouse conditions. The resistance level was evaluated based on the numbers of nematodes extracted from roots and soil 9 weeks after infestation. Small grain cereals have served as the basis for staple foods, beverages, and animal feed for thousands of years. Wheat, barley, oats, rye, triticale, rice, and others are rich in calories, proteins, carbohydrates, vitamins, and minerals. These cereals supply 20% of the calories consumed by people worldwide and are therefore a primary source of energy for humans and play a vital role in global food and nutrition security. Global production of small grains increased linearly from 1960 to 2005, and then began to decline. Further decline in production is projected to continue through 2050 while global demand for these grains is projected to increase by 1% per annum. Currently, wheat, barley, and oat production exceeds consumption in developed countries, while in developing countries the consumption rate is higher than production. An increasing demand for meat and livestock products is likely to compound the demand for cereals in developing countries. Current production levels and trends will not be sufficient to fulfill the projected global demand generated by increased populations. For wheat, global production will need to be increased by 60% to fulfill the estimated demand in 2050. Until recently, global wheat production increased mostly in response to development of improved cultivars and farming practices and technologies. Production is now limited by biotic and abiotic constraints, including diseases, nematodes, insect pests, weeds, and climate. Among these constraints, plant-parasitic nematodes alone are estimated to reduce production of all world crops by 10%. Cereal cyst nematodes (CCNs) are among the most important nematode pests that limit production of small grain cereals. Heavily invaded young plants are stunted and their lower leaves are often chlorotic, forming pale green patches in the field. Mature plants are also stunted, have a reduced number of tillers, and the roots are shallow and have a “bushy-knotted” appearance. CCNs comprise a number of closely-related species and are found in most regions where cereals are produced.

Morphological and molecular diversity among 11 populations of cereal cyst nematodes from different wheat production areas in Morocco was investigated using light microscopy, species-specific primers, complemented by the ITS-rDNA sequences. Morphometrics of cysts and second-stage juveniles (J2s) were generally within the expected ranges forHeterodera avenae; only the isolate from Aïn Jmaa showed morphometrics conforming to those ofH. latipons. When using species-specific primers forH. avenaeandH. latipons, the specific bands of 109 bp and 204 bp, respectively, confirmed the morphological identification. In addition, the internal transcribed spacer (ITS) regions were sequenced to study the diversity of the 11 populations. These sequences were compared with those ofHeteroderaspecies available in the GenBank database (www.ncbi.nlm.nih.gov) and confirmed again the identity of the species. Ten sequences of the ITS-rDNA were similar (99–100%) to the sequences ofH. avenaepublished in GenBank and three sequences, corresponding with one population, were similar (97–99%) toH. latipons.

Small grain cereals, such as wheat, barley and oats are considered among the most important food sources. Plant-parasitic nematodes play a considerable role in decreasing cereal yields. The three-major species of cereal cyst nematodes (CCN) Heterodera avenae, H. latipons, and H. filipjevi are distributed worldwide and cause considerable damage. This review provides information regarding the global distribution of these nematode species, yield loss due to CCN, their biology and pathogenic relation to plants, identification and control through agricultural practices, biological agents and resistance breeding. As morphological identification of CCN is difficult and time-consuming, several molecular techniques for the identification of these CCN species have been developed in recent years. The restrictions on the use of nematicides demand for resistance to CCN. Resistance genes in several lines are known and are used in numerous breeding programmes against CCN; pyramiding these resistance genes into high yielding cultivars that could become commercially available for farmers is progressing.

WOS:000411576400005 The cereal cyst nematode, Heterodera filipjevi (Madzhidov) Stelter, was detected in Turkey in 1996 and since then it has been increasingly reported to cause losses in wheat yields. Cereal cyst nematode populations have a wide genetic diversity which makes it difficult to be managed. The objectives of this research were to investigate the genetic variation of 23 cyst nematode populations collected from wheat growing areas of Bolu Province, utilising sequenced ITS-rDNA regions, and monitoring main nematode isolate population dynamics in field conditions. The results of ribosomal DNA sequencing revealed that all cyst populations were H. filipjevi and that little genetic variation was present among populations. The highest number of second-stage juveniles was observed between April and beginning of May in a field experiment; white females were visible on the roots in late May. The hatching process was positively correlated with low temperatures, but there were inverse correlations between reproductive factors and initial nematode population densities.

As the most abundant animals on earth, nematodes are a dominant component of the soil community. They play critical roles in regulating biogeochemical cycles and vegetation dynamics within and across landscapes and are an indicator of soil biological activity. Here, we present a comprehensive global dataset of soil nematode abundance and functional group composition. This dataset includes 6,825 georeferenced soil samples from all continents and biomes. For geospatial mapping purposes these samples are aggregated into 1,933 unique 1-km pixels, each of which is linked to 73 global environmental covariate data layers. Nematodes are ideal biological indicators to monitor soil biodiversity and ecosystem functioning. For this reason, they have been receiving increasing attention from a broad range of scientists. The main method to characterize soil nematode communities until at least genus level is still based on microscopic observations of nematode morphology. Such an approach is time-consuming, labor-intensive, and requires specialized personnel. The first studies on the potential use of DNA-metabarcoding to characterize nematode communities showed some shortcomings: under- or overestimation of species richness caused by failure to detect a number of nematode species or caused by intraspecific sequence variants increasing the number of OTUs (operational taxonomic units) or &lsquo, molecular&rsquo, species, and flaws in quantification. We set up experiments to optimize this metabarcoding approach. Our results provided new insights such as the drastic effect of different DNA-extraction methods on nematode species richness due to variation in lysis efficacy. Our newly designed primer set (18S rRNA gene, V4-V5 region) showed in silico an improved taxonomic coverage compared with a published primer set (18S rRNA gene, V6-V8 region). However, results of DNA-metabarcoding with the new primer set showed less taxonomic coverage, and more non-nematode reads. Thus, the new primer set might be more suitable for whole soil faunal analysis. Species-specific correction factors calculated from a mock community with equal amounts of different nematode species were applied on another mock community with different amounts of the same nematode species and on a biological sample spiked with four selected nematode species. Results showed an improved molecular quantification. In conclusion, DNA-metabarcoding of soil nematode communities is useful for monitoring shifts in nematode composition but the technique still needs further optimization to enhance its precision.

Amphimictic populations of root-lesion nematodes with numerous males and females having three lip annuli, a functional spermatheca and non-areolated lateral field occur on sword fern (Nephrolepis exaltata) in Florida. Identified for decades asPratylenchus penetrans, they appeared to be a morphologically separated species on the basis of a longer stylet (17.8-18.3 μm) thanP. penetrans(15-17 μm) and different lip pattern inen faceview (rectangularvsdumb-bell inP. penetrans). Morphologically similar amphimictic root-lesion nematodes have also been detected on flax lily in Costa Rica. Subsequent morphological observations indicated that these amphimictic root-lesion nematodes from fern and flax lily are closely related to the parthenogenetic speciesP. bolivianus, which has areolated lateral fields. In spite of the reproductive and morphological dissimilarities between these populations, their separation into different species was not supported by the results of molecular analyses of their DNA sequences. The populations used in these analyses included those that are amphimictic from Florida and Costa Rica and others that are parthenogenetic from the type locality in Bolivia, and geographically distant localities in Chile, China, Colombia and Europe. Phylogenetic analyses of the ITS and D2-D3 expansion segments of the 28S rRNA gene indicated that they belong to the same species,P. bolivianus, which consists of two morphotypes,P. bolivianus(am) amphimictic andP. bolivianus(pm) parthenogenetic, herein described and illustrated. Contradictory results were obtained by the analyses using a portion of thehsp90gene. The phylogenetic study, which included sequences of other root-lesion nematodes, a topotype and geographical distant populations ofP. zeae, revealed thatP. bolivianusandP. zeaeformed highly supported clades in the majority consensus trees. PCR with species-specific primers for rapid diagnostics ofP. bolivianusandP. zeaewere developed and tested.

Entomopathogenic nematodes belong to the families of Steinernematidae and Heterorhabditidae. They are obligate and lethal parasites of insects that can provide effective control of some important pests of commercial crops. A total of 53 isolates of EPN were molecularly characterised (ITS region-based) in the present study. Most of the studied isolates belong to theSteinernemagenus and only few isolates belong to theHeterorhabditisgenus. The phylogenetic relations ofSteinernemaandHeterorhabditisspecies were analysed by utilising the maximum likelihood method. In theSteinernemaphylogenetic tree, 99 isolates formed five major, moderately or highly reinforced clades: clade I:affine-intermediumgroup; clade II:carpocapsae-siamkayai-tami-scapterisci; clade III:bicornutum-riobrave-thermophilum; clade IV:glaseri-arenarium-karii-longicaudum; and clade V:feltiae-schliemanni-kushidai-kraussei-oregonense. The BLAST analysis of the ITS region of the rDNA of the steinernematid isolate PAL10 showed a rather low similarity of 93% withS. vulcanicum(accession number: GU929442), supporting the possible designation of a new species. In theHeterorhabditisphylogenetic tree, 25 isolates formed three main clades: clade I:bacteriophora-argentinensis-hepialius; clade II:baujardi-sonorensis-amazonensis; and clade III:indica-brevicaudis-hawaiiensis. All five studied isolates ofHeterorhabditiswere identified asH. indicaandH. bacteriophora. In both phylogenetic trees, the intra-specific variability level was different among clades for some species. The description of the new species (PAL10 isolate) would need further morphometric characterisation, morphologically identification and cross-breeding studies.

Root-lesion nematodes (Pratylenchus) have a worldwide distribution and cause severe production constraints on numerous important crops. During a survey of the wheat-growing area of Morocco, 17 populations of root-lesion nematode were collected. They were identified on the basis of their morphological and morphometric characters, and by molecular methods. Microscopic observations of females and males demonstrated the occurrence ofP. penetransin 13 of the 17 samples;P. thorneiandP. pseudocoffeaewere detected in four samples from Zaers and a single sample from Settat, respectively. A duplex PCR primer set was used to confirm the presence ofP. penetranswhile the species-specific forward primer PTHO and the common reverse primer D3B were used forP. thornei. For the remaining populations, the D2-D3 expansion segments of the 28S rRNA gene were amplified and the obtained sequences were compared with those ofPratylenchusspecies in the GenBank database. This comparison confirmed the morphological identifications and revealed a population ofP. pinguicaudatus. The study of the phylogenetic relationship of the MoroccanPratylenchuspopulations showed a high similarity (99-100%) between allP. penetranspopulations. The population dynamics of sixPratylenchuspopulations from Morocco were evaluated on carrot disk cultures at 4, 8 and 12 weeks after inoculation, and at 10, 15, 20 and 25°C. The optimum temperature for reproduction of all populations was 20°C. After 8 weeks at this temperature, nematode numbers increased up to 458-fold, 310-fold and 252-fold for the four populations ofP. penetrans, theP. thorneiand theP. pseudocoffeaepopulation, respectively.

Cereal cyst nematodes (CCN) are important plant-parasitic nematodes of wheat and barley, and exist in most of the cereal growing regions of the world. As there is limited information on the nature and distribution of CCN species in Syria, a survey was conducted in north-eastern Syria to assess the distribution of CCN in the main wheat and barley growing areas. In the summer of 2009, a total of 167 composite soil samples were collected from 167 wheat and barley fields. Cysts were extracted from soil using the Fenwick can technique, then quantified and identified up to the species level by using both morphological and molecular methods. The study revealed that 62% of the fields were infested with the three Heterodera species: H. avenae, H. filipjevi and H. latipons. However, the most prevalent species was H. latipons, which was present in 76% of the infested samples, while it occurred alone in 67% of those samples. Heterodera avenae was detected singly in 20% of the samples, while 11% consisted of mixed populations with other species. Heterodera filipjevi was never detected alone; it was found mixed with H. avenae and/or H. latipons in 9% of the samples. The high number of extracted cysts (up to 116 cysts × 200 g−1 of soil) is most likely related to the monoculture practiced in that region and the fact that growers are unaware of the existence of resistant varieties.

Summary Among the recognised species groups of Heterodera, the Avenae group is one of the largest with a total of 12 species. Ten of them, H. arenaria, H. aucklandica, H. australis, H. avenae, H. filipjevi, H. mani, H. pratensis, H. riparia, H. sturhani and H. ustinovi, are morphologically closely related and represent the H. avenae species complex, and the other two, H. hordecalis and H. latipons, are morphologically more distinct from this complex. In this study we provide comprehensive phylogenetic analyses of several hundred COI and ITS rRNA gene sequences from the Avenae group using Bayesian inference, maximum likelihood and statistical parsimony. Some 220 COI and 11 ITS rRNA new gene sequences from 147 nematode populations collected in 26 countries were obtained in this study. Our study showed that the COI gene is a powerful DNA barcoding marker for identification of populations and species from the Avenae group. A putatively new cyst nematode species related to H. latipons was revealed from the analysis of COI and ITS rRNA gene datasets. COI gene sequences allow distinguishing H. arenaria, H. australis and H. sturhani from each other and other species. Problems of species delimiting of these species are discussed. The results of the analysis showed that COI haplotypes corresponded to certain pathotypes of the cereal cyst nematodes. It is recommended that information on COI haplotypes of studied populations be included in research with these nematodes. Based on the results of phylogeographical analysis and age estimation of clades with a molecular clock approach, it was hypothesised that several species of the Avenae group primarily originated and diversified in the Irano-Anatolian hotspot during the Pleistocene and Holocene periods and then dispersed from this region across the world. Different geographic barriers, centres and times of origin might explain current known distribution patterns for species of the Avenae group. Possible pathways, including a long distance trans-Atlantic dispersal, and secondary centres of diversification are proposed and discussed.

The pine wood nematode (PWN), Bursaphelenchus xylophilus, causes pine wilt disease and is transmitted by Monochamus beetles. An efficient trapping system for these organisms is essential for their early detection in threatened regions. A wet catching method, using monoethylene glycol (MEG) in a collecting cup to preserve captured insects, was suitable for catching longhorn beetles. As a second step in developing this technique, we studied the influence of MEG on nematode detection. When M. galloprovincialis carrying PWN were submerged in MEG, nematodes died within 24 h. The preservative altered certain morphological features of nematodes immersed in solutions of 10, 30 or 60% MEG, impeding their morphological identification. However, molecular identification was possible as long as the MEG concentration was below 70%. At higher concentrations, a rinsing step with water before DNA extraction was enough to allow molecular detection.
