Purpose Wireless Capsule Endoscopy (WCE) is a minimally invasive diagnosis tool for lesion detection in the gastrointestinal tract, reaching places where conventional endoscopy is unable to. However, the significant amount of acquired data leads to difficulties in the diagnosis by the physicians; which can be eased with computer assistance. This paper addresses a method for the automatic detection of tumors in WCE by using a two-step based procedure: region of interest selection and classification. Methods The first step aims to separate abnormal from normal tissue by using automatic segmentation based on a Gaussian Mixture Model (GMM). A modified version of the Anderson method for convergence acceleration of the expectation-maximization (EM) algorithm is proposed. The proposed features for both segmentation and classification are based on the CIELab color space, as a way of bypassing lightness variations, where the L component is discarded. Tissue variability among subjects, light inhomogeneities and even intensity differences among different devices can be overcome by using simultaneously features from both regions. In the second step, an ensemble system with partition of the training data with a new training scheme is proposed. At this stage, the gating network is trained after the experts have been trained decoupling the joint maximization of both modules. The partition module is also used at the test step, leading the incoming data to the most likely expert allowing incremental adaptation by preserving data diversity. Results This algorithm outperforms others based on texture features selected from Wavelets and Curvelets transforms, classified by a regular support vector machine (SVM) in more than 5%. Conclusions This work shows that simpler features can outperform more elaborate ones if appropriately designed. In the current case, luminance was discarded to cope with saturated tissue, facilitating the color perception. Ensemble systems remain an open research field. The objective of the study was to examine the effects of the relative age effect (RAE) and predicted maturity status on body size and repeated sprint ability (RSA: 7 x 34.2 m / 25 s interval) in youth soccer. The sample was composed of 197 male players aged 13-14 years. Body mass, stature, and sitting height were measured, RSA was assessed in the field, and age at peak height velocity (APHV) was predicted. Factorial ANOVA tested the independent and combined effects of RAE given by birth quarters (BQs) and maturity status on dependent variables. Players born in the second birth quarter (BQ2) were significantly taller (F = 4.28, p < 0.01) than their peers born in BQ1 and BQ3. Additionally, players born in BQ2 performed better than players born in BQ4 in RSA total time and ideal time (F ranged between 4.81 and 4.90, p < 0.01), while players born in BQ1 exhibited a lower RSA fatigue index compared to those born in BQ4 (F = 2.90, p < 0.05). The interaction of the BQ and maturity status was a significant source of inter-individual variation for body size (F ranged between 64.92 and 105.57; p < 0.01) and RSA output (F ranged between 4.082 and 6.76; p < 0.05). In summary, being relatively older and, simultaneously, advanced in maturity status corresponds to a substantial advantage in characteristics that are related to soccer-specific fitness.

Asymptomatic and mildly symptomatic patients were forced to quarantine and closely monitor their symptoms and vital signs, most of the time at home. This paper describes e-CoVig, a novel mHealth application, developed as an alternative to the current monitoring paradigm, where the patients are followed up by direct phone contact. The e-CoVig provides a set of functionalities for remote reporting of symptoms, vital signs, and other clinical information to the health services taking care of these patients. The application is designed to register and transmit the heart rate, blood oxygen saturation (SpO2), body temperature, respiration, and cough. The system features a mobile application, a web/cloud platform, and a low-cost specific device to acquire the temperature and SpO2. The architecture of the system is flexible and can be configured for different operation conditions. Current commercial devices, such as oximeters and thermometers, can also be used and read using the optical character recognition (OCR) functionality of the system. The data acquired at the mobile application are sent automatically to the web/cloud application and made available in real-time to the medical staff, enabling the follow-up of several users simultaneously without the need for time consuming phone call interactions. Dyslipidemia is a major traditional risk factor for cardiovascular disease (CVD) in chronic kidney disease (CKD) patients, although the altered lipid profile does not explain the number and severity of CVD events. High-density lipoprotein (HDL) is a heterogeneous (size, composition, and functionality) population of particles with different atherogenic or atheroprotective properties. HDL-cholesterol concentrations per se may not entirely reflect a beneficial or a risk profile for CVD. Large HDL in CKD patients may have a unique proteome and lipid composition, impairing their cholesterol efflux capacity. This lack of HDL functionality may contribute to the paradoxical coexistence of increased large HDL and enhanced risk for CVD events. Moreover, CKD is associated with inflammation, oxidative stress, diabetes, and/or hypertension that are able to interfere with the anti-inflammatory, antioxidative, and antithrombotic properties of HDL subpopulations. How these changes interfere with HDL functions in CKD is still poorly understood. Further studies are warranted to fully clarify if different HDL subpopulations present different functionalities and/or atheroprotective effects. To achieve this goal, the standardization of techniques would be valuable.

Chronic inflammation plays an important role in the progression and outcome of chronic kidney disease (CKD). The circulating levels of the inflammatory biomarkers interleukin 6 (IL6) and pentraxin 3 (PTX3) are enhanced in CKD patients, and are associated with the progression of the disease and with higher risk for cardiovascular events, the major cause of death in CKD patients. Our aim was to study how specific polymorphisms of IL6 and PTX3 encoding genes affect the inflammatory response and outcome of end-stage renal disease (ESRD) patients on dialysis. Methodology included the analysis of two single nucleotide polymorphisms (SNP), namely the IL6 (rs1800795) polymorphism in the promoter region (-174G > C), and the PTX3 (rs2305619) polymorphism in the intron 1 (+ 281A > G), which were analyzed in ESRD patients on dialysis and in a group of heathy individuals. The allelic frequencies, genotype distribution and their association with circulating levels of the inflammatory markers C-reactive protein (CRP), IL6, growth differentiation factor 15 (GDF15) and PTX3, were determined in ESRD patients. Events of death were recorded along one year, to assess the association of the studied SNPs with all-cause mortality and the inflammatory biomarkers, in ESRD patients. Results showed that the allelic frequencies and genotype distribution for IL6 and PTX3 SNPs in the control group and ESRD patients were similar and in agreement with other European reports. For the IL6 polymorphism, we found a trend towards higher levels of high-sensitivity (hs) CRP, IL6 and PTX3 in the homozygous genotypes; the CC genotype also showed the highest levels of GDF15. The mortality rate after the 1-year follow-up was 10.4%. The CC genotype (IL6 SNP) was associated to a higher risk of mortality and deceased patients carrying this genotype also showed the highest levels of hsCRP. Regarding the studied PTX3 SNP, the AA genotype was linked to an enhanced inflammatory response, showing the highest values of hsCRP and IL6. Nevertheless, this genotype had no significant impact on the mortality rate. In conclusion, both studied SNPs seem to modulate the inflammatory response in ESRD and may, therefore, be determinant on disease progression and patients’ outcome. Our data also highlights the importance of research on genetic variants that, although less frequent, may have significant biological value.

OBJECTIVE The purpose of the study was to evaluate the agreement between the Fels and Greulich-Pyle methods for the assessment of skeletal age (SA) in female youth soccer players. METHODS The sample included 441 Portuguese players 10.08-16.73 years of age who regularly participated in organized and competitive soccer. Standardized radiographs of the left hand-wrist were obtained and analyzed by an experienced examiner. SA was estimated with the Fels and Greulich-Pyle (GP) methods. Differences between SA and chronological age (CA) were used to define skeletal maturity groups: late, average and early maturing. In addition to descriptive statistics, Cohen's kappa and Lin concordance correlation coefficients were used to evaluate agreement between methods. RESULTS Intraindividual differences in SA based on the two methods varied between 0.10 to 1.47 years among age groups with larger mean differences at older ages. Agreement of maturity classifications between methods was 74% at younger ages (under-13: kappa = 0.48; under-14: kappa = 0.39; Lin CCC = 0.68) and declined with increasing CA (under-17: 19% agreement; kappa = 0.001; Lin CCC = 0.11). About 19% of the total sample was skeletally mature with the Fels method and an SA was not assigned; in contrast, no players were skeletally mature with the GP method. CONCLUSIONS GP SAs were systematically lower than Fels SAs among female soccer players. Intraindividual variability in SAs between methods was considerable. The findings highlight the impact of method on estimates of maturity status.

A maturity-associated gradient among adolescent athletes in many sports tends to favour early maturing participants. This study was aimed to describe inter-individual variance in growth and skeletal maturation among male youth table tennis. The sample included 99 players registered in competitive clubs for at least 2 years. Chronological age (CA) ranged 10.00-14.63 years. Stature and body mass were plotted relative to US references. Skeletal age (SA) was estimated with the Fels method. Maturity status was given as SA minus CA. Somatic maturation was derived from predicted age at peak height velocity. Mean values for stature and weight were above the reference medians from 10 to 13 years and at the median in the oldest group. In general, SA was in advance to CA (CA: 12.45 ± 1.28 years; SA: 13.44 ± 2.37 years). The majority of the players were early maturing according to SA (52%) and average maturing based on the somatic indicator (80%). The results suggested that table tennis tends to retain taller and heavier participants who are somewhat advanced in skeletal maturity. Participation in youth table tennis and opportunities to win need to consider age- and maturity-associated variations, particularly regarding bands for competitions.

Basketball is characterized by high-intensity episodes predominantly reliant on anaerobic metabolism. The force-velocity test enables individual determination of an optimal braking force and emerged as appropriate to estimate optimal peak power. It has rarely been used in youth basketball. This study aimed to examine the contribution of body size, composition, and biological maturation to interindividual variation in force-velocity test output among pre-pubertal basketball players. The sample consisted of 64 male participants (8.4–12.3 years). Stature, sitting height, body mass and two skinfolds were measured, and leg length estimated. Fat-free mass and lower limb volume were estimated from anthropometry. Age at peak height velocity was predicted from maturity offset. Optimal peak power was correlated with all body size descriptors (correlation: 0.541–0.700). Simple allometric models explained 30–47% of inter-individual variance, with fat-free mass being the best predictor of performance. Whole-body fat-free mass (as a surrogate for active muscle mass) plus the indicator of maturation emerged as the best proportional allometric model (53% explained variance). Even at pre-pubertal ages, the interpretation of the force-velocity test requires assessing the metabolically active component of body mass.

Objectives This study aims to report the changes and adaptations of a vascular tertiary center during a global pandemic and the impact on its activity and patients. Methods We conducted a retrospective cohort study within the Vascular Surgery ward in Centro Hospitalar Universitário Lisboa Norte, Portugal. All data from surgical, inpatient and outpatient activity were collected from February to June 2020 and compared to the same 5-month period in 2018 and 2019. We ran a descriptive analysis of all data and performed statistical tests for the variation of procedures and admissions between February and June 2018 and the same time period in 2020. Results During the outbreak, our staff had to be readapted. Six nurses were transferred to COVID-19 units (out of a total of 33 nurses) while 1 of the 7 residents was transferred to an intensive care unit and 1 senior surgeon was put on prophylactic leave. In the outpatient clinic, there was an increase in the number of telemedicine consultations with a greater focus on first-time referrals and urgent cases. There was a significant increase in the total number of elective admissions whereas there were significantly less admissions from an emergency setting (+57% and −54%, respectively, P < 0.001). The vascular surgery team performed a total number of 584 procedures between February and June 2020 (−17.8% compared to 2018 and 2019), with a significant increase in the number of endovascular procedures (P < 0.001) and in the use of local and regional anesthesia (P < 0.001), especially in the Angio Suite (+600%, P < 0.001). Comparing with 2018 and 2019, the surgical team performed less outpatient procedures in early 2020. We reported a significant increase in the total number of procedures for patients with a chronic limb-threatening ischemia (CLTI) diagnosis (+21%, P < 0.001). We did not report significant changes in the proportion of other vascular conditions. Regarding mortality, we observed a 16% decrease in the intraoperative mortality (P 0.67). Conclusions In this study, we assessed the impact of the COVID-19 outbreak in daily activity during the contingency period. During the outbreak, there was an overall decline in outpatient clinics and inpatient admissions. Nevertheless, and despite the restrictions imposed by the pandemic and health authorities, we managed to maintain most procedures for most vascular diseases, particularly for CLTI urgent cases, without a significant increase in the mortality rate. Stringent protective measures for patient and staff or higher use of endovascular techniques and local anesthesia are some of the successful changes implemented in the department. These learned lessons are to be pursued as the pandemic evolves with future outbreaks of COVID-19, such as the current second outbreak currently spreading through Europe.

The aim of this study was to determine intra-observer (reproducibility) and inter-observer agreement of SA values obtained via the Greulich-Pyle (GP) method. In addition, the variation in calculated SAs by alternative GP protocols was examined. Methods: The sample was composed of 100 Portuguese female soccer players aged 12.0–16.7 years. SAs were determined using the GP method by two observers (OB1: experience < 100 exams using GP; OB2: experience > 2000 exams using several methods). The radiographs were examined using alternative GP protocols: (wholeGP) the plate was matched to the atlas as an overall approach; (30-boneGP) bone-by-bone inspections of 30-bones; (GPpmb) bone-by-bone inspections of the pre-mature bones only. For the 30-boneGP and GPpmb approaches, SA was calculated via the mean (M) and the median (Md). Results: Reproducibility ranged 82–100% and 88–100% for OB1 and OB2, respectively. Inter-observer agreement (100 participants multiplied by 30 bones) was 92.1%. For specific bones, agreement rates less than 90% were found for scaphoid (81%), medial phalange V (83%), trapezium (84%) and metacarpal V (87%). Differences in wholeGP SAs obtained by the two observers were moderate (d-cohen was 0.79). Mean differences between observers when using bone-by bone SAs were trivial (30-boneGP: d-cohen less than 0.05; GPpmb: d-cohen less than 0.10). The impact of using the mean or the median was negligible, particularly when analyses did not include bones scored as mature. Conclusion: The GP appeared to be a reasonably reproducible method to assess SA and inter-observer agreement was acceptable. There is evidence to support a recommendation of only scoring pre-mature bones during later adolescence. Further research is required to examine whether these findings are consistent in younger girls and in boys. Sprint performance over short distances is a central component in young tennis players' development. This study aimed to examine the longitudinal development of sprint performance in young female tennis players, and to investigate differences between performance levels. Also potentially explanatory variables were investigated. Female tennis players aged 10-15 (N= 167) participated in a, mixed-longitudinal study (n= 48 elite;n= 119 sub-elite). Players were measured annually on the 5 m sprint as well as for possible explaining variables for 5 m sprint performance development (age, height, body mass, maturity status, lower limb explosive strength). Multilevel analysis was used to obtain a developmental model. Moreover, it was possible to predict sprint performance (5 m) based on chronological age, body size given by height, and lower limb strength performance (p . 05). Sprint performance is an important characteristic of young female tennis players and seemed to depend on growth and maturation in parallel to physical fitness.

Background and objectives: Athletes from combat sports are grouped into a series of weight categories that are intended to promote fair competition. Differences in performance are partly attributable to differences in body size. Consequently, ratio standards in which a performance variable is simply divided by an anthropometric characteristic such as body mass are often used, although this application is not recommended. This study aimed to obtain allometric models to interpret Wingate Anaerobic Test (WAnT) outputs among male adult athletes from combat sports. Materials and Methods: The sample was composed of 64 participants aged 18&ndash, 39 years (24.2 &plusmn, 4.6 years). Stature and body mass (BM) were measured and air displacement plethysmography used to estimate fat mass and fat-free mass (FFM). Lower-limb lean soft tissue (LL-LST) was derived from dual energy X-ray absorptiometry. WAnT outputs were peak power (WAnT-PP) and mean power (WAnT-MP). Allometric models were obtained from simple and multiple linear regressions using log-transformed variables. Results: Models derived from a single three-dimension descriptor explained a large portion of variance: WAnT-PP (BM: 31.1%, FFM: 54%, LL-LST: 47.2%) and WAnT-MP (BM: 50.1%, FFM: 57.4%, LL-LST: 62.7%). Finally, the best proportional allometric models emerged from the combination of LL-LST and FFM (WAnT-PP: 55%, WAnT-MP: 65%). Conclusions: The relationship between weight categories and performance did not seem to be explained by the basic principles of geometric similarity.

Introduction: In the last two decades there have been advances in the diagnosis and management of neonatal cholestasis, which may have changed its epidemiology, diagnostic accuracy, outcomes, and survival. Our goal was to characterize these changes over time in our setting. Methods: Retrospective cohort study in a tertiary center, enrolling patients born between January 1985 and October 2019. The cohort was divided into two periods, before (A; n = 67) and after (B; n = 87) the year 2000; and in two groups, according to patient's outcome (favorable, unfavorable). Overall survival and survival with and without orthotopic liver transplant (OLT) were evaluated in the two periods (A and B) and in different subgroups of underlying entities. Results: We found that the age of cholestasis recognition decreased significantly from period A to period B [median 43 days and 22 days, respectively, (p < 0.001)]; the changes in epidemiology were relevant, with a significant decrease in alpha-1-antitrypsin deficiency (p < 0.001) and an increase in transient cholestasis (p = 0.004). A next-generation sequencing (NGS) panel available since mid-2017 was applied to 13 patients with contributory results in 7, but, so far, only in 2 patients led to conclusive diagnosis of underlying entities. The number of cases of idiopathic cholestasis did not vary significantly. Over time there was no significant change in the outcome (p = 0.116). Overall survival and survival without OLT had no significant improvement during the period of observation (in periods A and B, 86 vs. 88%, and 85 vs. 87%, respectively). However, in period B, with OLT we achieved the goal of 100% of survival rate. Conclusions: Our data suggest that transient cholestasis became a very important subset of neonatal cholestasis, requiring specific guidance. The NGS panels can provide important inputs on disease diagnosis but, if applied without strict criteria and expertise, they can open a Pandora's box due to misinterpretation. Despite all the advances in accurate diagnosis and timely management-including early recognition of cholestasis-the improvement in patient outcomes and survival were still not significant. Cardiovascular disease (CVD) events are the main causes of death in end-stage renal disease (ESRD) patients on dialysis. The number and severity of CVD events remain inappropriate and difficult to explain by considering only the classic CVD risk factors. Our aim was to clarify the changes and the relationship of lipoprotein subfractions with other CVD risk factors, namely, body mass index (BMI) and adipokines, inflammation and low-density lipoprotein (LDL) oxidation, and the burden of the most prevalent comorbidities, diabetes mellitus (DM) and hypertension (HT). We studied 194 ESRD patients on dialysis and 22 controls; lipid profile, including lipoprotein subpopulations and oxidized LDL (oxLDL), C-reactive protein (CRP), adiponectin, leptin, and paraoxonase 1 activity were evaluated. Compared to controls, patients presented significantly lower levels of cholesterol, high-density lipoprotein cholesterol (HDLc), LDLc, oxLDL, and intermediate and small HDL and higher triglycerides, CRP, adiponectin, large HDL, very-low-density lipoprotein (VLDL), and intermediate-density lipoprotein- (IDL) B. Adiponectin levels correlated positively with large HDL and negatively with intermediate and small HDL, oxLDL/LDLc, and BMI; patients with DM (n=17) and with DM+HT (n=70), as compared to patients without DM or HT (n=69) or only with HT (n=38), presented significantly higher oxLDL, oxLDL/LDLc, and leptin and lower adiponectin. Obese patients (n=45), as compared to normoponderal patients (n=81), showed lower HDLc, adiponectin, and large HDL and significantly higher leptin, VLDL, and intermediate and small HDL. In ESRD, the higher adiponectin seems to favor atheroprotective HDL modifications and protect LDL particles from oxidative atherogenic changes. However, in diabetic and obese patients, adiponectin presents the lowest values, oxLDL/LDLc present the highest ones, and the HDL profile is the more atherogenic. Our data suggest that the coexistence of DM and adiposity in ESRD patients on dialysis contributes to a higher CVD risk, as showed by their lipid and adipokine profiles.

Background: Soluble transferrin receptor (sTfR) is a biomarker of erythropoiesis, which is often impaired in dialysis patients. The aim of our study was to evaluate sTfR levels in chronically dialyzed patients and assess potential determinants of its levels. Methods: We performed a cross-sectional study by evaluating 246 end-stage renal disease patients undergoing dialysis and 32 healthy controls. Circulating levels of interleukin (IL)-6, C-reactive protein (CRP), tumor necrosis factor (TNF)-α, hepcidin, sTfR, growth differentiation factor 15 (GDF15), and traditional iron metabolism markers were measured, as well as hemogram parameters. Clinical data was obtained from all patients. Results: Compared to controls, patients presented similar values of sTfR, reticulocytes and reticulocyte production index (RPI), and significantly higher levels of IL-6, CRP, ferritin, hepcidin, TNF-α, and GDF15. Iron, transferrin, hemoglobin levels, erythrocyte count, mean cell hemoglobin (MCH), and mean cell hemoglobin concentration (MCHC) values were significantly lower in dialysis group. Within patients, sTfR values were higher in diabetic patients and were positively and significantly correlated with reticulocytes and erythrocytes, RPI, and therapeutic doses of erythropoiesis stimulating agents (ESA) and intravenous iron; and inversely and significantly correlated with circulating iron, ferritin, transferrin saturation, hepcidin, MCH, and MCHC. In multiple linear regression analysis, ESA dose, RPI, serum iron, diabetes, and hepcidin levels were independently associated with sTfR levels in dialysis patients and, thus, with erythropoiesis. Conclusion: Our data suggest that, besides RPI and ESA dose, diabetes and hepcidin are closely related to erythropoiesis in dialysis patients. The influence of diabetes on sTfR levels deserves further investigation.

Background The aim of the study was to examine the contribution of chronological age (CA), skeletal maturation, training experience and concurrent body size descriptors, to inter-individual variance in left ventricular mass (LVM) among female adolescent soccer players. Methods The sample included 228 female soccer players 11.8–17.1 years. Training experience defined as years of participation in competitive soccer (range 2–9 years), was obtained by interview. Stature, body mass and skinfolds (triceps, medial calf) were measured. Fat mass was estimated; Fat-free mass was derived. LVM was assessed by echocardiography. Skeletal maturity status was as the difference of skeletal age (SA, Fels method) minus CA. Results Fat-free mass was the most prominent single predictor of LVM (R2 = 36.6%). It was associated with an allometric coefficient close to linearity (k = 0.924, 95%CI: 0.737 to 1.112). A significant multiplicative allometric model including body mass, fat-free mass, CA, training experience and skeletal maturity status was also obtained (R = 0.684; R2 = 46.2%). Conclusion Stature has limitations as a valid size descriptor of LVM. Body mass, fat-free mass, training experience, CA, body mass and skeletal maturity status were relevant factors contributing to inter-individual variability in LVM.

The purpose of the study was to model the longitudinal development of knee extension (KE) and flexion (KF) strength in adolescent soccer players. A mixed-longitudinal sample composed of 67 soccer players aged 11.0-13.9 years at baseline was followed on three-to-five occasions over 5 years. Stature, body mass and several skinfold thicknesses were measured. Fat mass was estimated from skinfolds and fat-free mass (FFM) derived. Skeletal age was estimated with the TW2-RUS protocol. An isokinetic dynamometer was used to obtain peak torque of KE and KF from concentric assessments at an angular velocity of 180°/s. Multilevel random effects regression analyses were performed. Among youth soccer players aged 11-16 years, isokinetic strength of the knee muscle groups was reasonably predicted from chronological age (CA), stature and FFM: KE = -66.170 + 5.353 × (CA) + 0.594 × (CA2) + 0.552 × (stature) + 1.414 × (FFM), and KF = -9.356 + 2.708 × (CA) + 1.552 × (FFM). In conclusion, CA per se accounted for annual increments of 5.4 Nm in KE and 2.7 Nm in KF.

Introduction: Biological maturation has been related to the level of physical activity and motor competence of children. Objectives: This study aimed to: 1) analyze the association between biological maturation and performance in muscular strength tests and 2) examine whether the relationship between maturation and performance in physical tests is mediated by anthropometric variables. Method: The sample was composed of 71 eight-year-old Brazilian girls. Anthropometry considered stature, body mass, waist circumference, estimated fat mass and fat-free mass. Biological maturation was assessed based on the percentage of predicted adult stature. The physical tests consisted of 2-kg medicine ball throw, handgrip strength, sit-ups and standing long jump. Pearson’s correlation test was conducted between the study variables and the last stage consisted of a causal mediation analysis. Results: Biological maturation was significantly associated with the 2-kg medicine ball throw (r=0.52) and handgrip strength (r=0.42) tests. The increasing prevalence of functionally-limited hypertensive individuals highlights the need for interventions to reduce the burden of hypertension-aging-disability and to maximize the chances of healthy aging. This study aims to compare the effects of multicomponent exercise and different pharmacological treatments on functional status and cardiovascular risk outcomes in hypertensive older adults with comorbidities. Participants (n = 96) engage in a 3 days/week multicomponent (aerobic + resistance) exercise program and for one of the following three conditions: (1) thiazide-related diuretics (TDs; n = 33, 69.9 ± 9.5 years); (2) calcium channel blockers (CCBs; n = 23, 67.0 ± 9.0 years); (3) and β-blockers (βBs; n = 40, 65.6 ± 7.2 years) medication. Baseline and 2-year follow-up evaluations included the Senior Fitness Test battery, anthropometrics and hemodynamic profile, health-related quality of life (HRQoL; Short-Form Health Survey 36) and health history questionnaires. Background Poor brain extraction in Magnetic Resonance Imaging (MRI) has negative consequences in several types of brain post-extraction such as tissue segmentation and related statistical measures or pattern recognition algorithms. Current state of the art algorithms for brain extraction work on weighted T1 and T2, being not adequate for non-whole brain images such as the case of T2*FLASH@7T partial volumes. New method This paper proposes two new methods that work directly in T2*FLASH@7T partial volumes. The first is an improvement of the semi-automatic threshold-with-morphology approach adapted to incomplete volumes. The second method uses an improved version of a current implementation of the fuzzy c-means algorithm with bias correction for brain segmentation. Results Under high inhomogeneity conditions the performance of the first method degrades, requiring user intervention which is unacceptable. The second method performed well for all volumes, being entirely automatic. Comparison with existing methods State of the art algorithms for brain extraction are mainly semi-automatic, requiring a correct initialization by the user and knowledge of the software. These methods can't deal with partial volumes and/or need information from atlas which is not available in T2*FLASH@7T. Also, combined volumes suffer from manipulations such as re-sampling which deteriorates significantly voxel intensity structures making segmentation tasks difficult. The proposed method can overcome all these difficulties, reaching good results for brain extraction using only T2*FLASH@7T volumes. Background This study aims to analyze the effects of anti-hypertensive monotherapy and combined therapy on functional status, and cardiovascular risk outcomes in older adults. Methods This longitudinal non-randomized cohort study, involved hypertensive older adults (n = 440) aged 60 or more years with comorbidities. Participants underwent a community exercise training program and one of the following 2 conditions: i) use of daily mono-dose angiotensin-converting enzyme inhibitors (ACEi; n= 232); ii) combined therapy including ACEi plus other class agent (Combined; n= 208). Baseline and 2-year follow-up evaluations included the functional fitness, health-related quality of life (HRQoL), health history questionnaires, anthropometric and hemodynamic profile. Results Both experimental groups have significantly improved physical functional status, and have significantly decreased blood pressure and waist circumference. ACEi group has significantly reduced body mass and body mass index, the Combined group significantly reduced the waist-to-hip ratio. Additionally, both groups perceived better physical HRQoL. Conclusions Functional status has improved with ACEi medication and exercise training, regardless the ACEi medication therapy. Exercise training plus ACEi antihypertensive therapy should be recommended into the standard prescription practice to reduce the rate of physical disability among hypertensive older adults.

Synthetic cathinones abuse remains a serious public health problem. Kidney injury has been reported in intoxications associated with synthetic cathinones, but the molecular mechanisms involved have not been explored yet. In this study, the potential in vitro nephrotoxic effects of four commonly abused cathinone derivatives, namely pentedrone, 3,4-dimethylmethcatinone (3,4-DMMC), methylone and 3,4-methylenedioxypyrovalerone (MDPV), were assessed in the human kidney HK-2 cell line. All four derivatives elicited cell death in a concentration- and time-dependent manner, in the following order of potency: 3,4-DMMC >> MDPV > methylone ≈ pentedrone. 3,4-DMMC and methylone were selected to further elucidate the mechanisms behind synthetic cathinones-induced cell death. Both drugs elicited apoptotic cell death and prompted the formation of acidic vesicular organelles and autophagosomes in HK-2 cells. Moreover, the autophagy inhibitor 3-methyladenine significantly potentiated cell death, indicating that autophagy may serve as a cell survival mechanism that protects renal cells against synthetic cathinones toxicity. Both drugs triggered a rise in reactive oxygen and nitrogen species formation, which was completely prevented by antioxidant treatment with N‑acetyl‑L‑cysteine or ascorbic acid. Importantly, these antioxidant agents significantly aggravated renal cell death induced by cathinone derivatives, most likely due to their autophagy-blocking properties. Taken together, our results support an intricate control of cell survival/death modulated by oxidative stress, apoptosis and autophagy in synthetic cathinones-induced renal injury.

SUMMARY OBJECTIVE The current study aimed to examine the body composition of adult male ultra-trail runners (UTR) according to their level of participation (regional UTR-R, vs. national UTR-N). METHODS The sample was composed of 44 adult male UTR (aged 36.5±7.2 years; UTR-R: n=25; UTR-N: n=19). Body composition was assessed by air displacement plethysmography, bioelectrical impedance, and dual-energy X-ray absorptiometry. In addition, the Food Frequency Questionnaire (FFQ) was applied. A comparison between the groups was performed using independent samples t-test. RESULTS Significant differences between groups contrasting in the competitive level were found for chronological age (in years; UTR-R: 38.8±8.2 vs. UTR-N: 33.5±4.1); body density (in L.kg-1; UTR-R: 1.062±0.015 vs. UTR-N: 1.074±0.009); and fat mass (in kg; UTR-R: 12.7±6.8 vs. UTR-N: 7.6±2.7). CONCLUSION UTR-N were younger, presented higher values for body density, and had less fat mass, although no significant differences were found for fat-free mass. The current study evidenced the profile of long-distance runners and the need for weight management programs to regulate body composition. A composição corporal foi avaliada recorrendo à pletismografia de ar deslocado, bioimpedância elétrica e absorciometria de raios X de dupla energia. RESULTADOS Foram encontradas diferenças significativas por nível de competição para as seguintes variáveis dependentes: idade cronológica (em anos; UTR-R: 38,8±8,2 vs. UTR-N: 33,5±4,1); densidade corporal (em kg/L; UTR-R: 1,062±0,015 L/kg vs. UTR-N: 1,074±0,009); massa gorda (em kg; UTR-R: 12,7±6,8 kg vs. UTR-N: 7,6±2,7). CONCLUSÃO Os UTR-N tendem a ser mais jovens e apresentam valores superiores de densidade corporal e, consequentemente, valores menores de massa gorda, sendo a massa isenta de gordura semelhante entre os grupos. Cytidine deaminases (CDAs) catalyze the hydrolytic deamination of cytidine and deoxycytidine in the pyrimidine salvage pathway. In kinetoplastids, pyrimidine metabolism has been extensively studied as a source of potential drug targets, given the fact that many of the enzymes of the pathway are essential. Thymidylate (dTMP) synthesis in Trypanosoma brucei exhibits unique characteristics. Thus, it has been suggested that the production of dUMP, the substrate for dTMP formation, is solely dependent on cytidine deaminase and thymidine kinase. Here we characterize recombinant T. brucei CDA (TbCDA) and present evidence that indeed the alternative route for dUMP formation via deoxyuridine 5′-triphosphate nucleotidohydrolase does not have a prominent role in de novo dTMP formation. Furthermore, we provide a scheme for the compartmentalization of dTMP biosynthesis, taking into account the observation that CDA is located in the mitochondrion, together with available information on the intracellular localization of other enzymes involved in the dTTP biosynthetic pathway., Cytidine deaminase (CDA) is a pyrimidine salvage enzyme that catalyzes cytidine and deoxycytidine hydrolytic deamination to yield uridine and deoxyuridine. Here we report the biochemical characterization of Trypanosoma brucei CDA as an enzyme within the tetrameric class of the CDA family that efficiently deaminates cytidine, deoxycytidine, and the nucleoside analogue 5-methyl-2′-deoxycytidine. In line with previous studies, we show that RNA interference (RNAi)-mediated CDA depletion impairs T. brucei proliferation when grown in pyrimidine-deficient medium, while supplementation with thymidine or deoxyuridine restores growth, further underscoring the role of this enzyme in providing deoxyuridine for dUMP formation via thymidine kinase, the substrate required for de novo thymidylate biosynthesis. This observation contrasts with the existence in T. brucei of a dimeric deoxyuridine 5′-triphosphate nucleotidohydrolase (dUTPase), an essential enzyme that can produce dUMP via the hydrolysis of dUTP/dUDP. Thus, T. brucei dUTPase-null mutants are thymidine auxotrophs, suggesting that dUTPase might have a role in providing dUMP for thymidylate biosynthesis. We show that overexpression of human dCMP deaminase (DCTD), an enzyme that provides directly dUMP through dCMP deamination, does not reverse the lethal phenotype of dUTPase knockout cells, which further supports the notion that in T. brucei, CDA is uniquely involved in providing dUMP, while the main role of dUTPase would be the withdrawal of the excess of dUTP to avoid its incorporation into DNA. Furthermore, we report the mitochondrial localization of CDA, highlighting the importance of this organelle in pyrimidine metabolism. Objective: To evaluate cardiorespiratory fitness’ tracking from childhood to adolescence, as well as to test the moderation role of somatic maturation. Methods: Our sample was composed by 375 children (197 boys), with a baseline age between 7 and 10 years old. The children were followed-up over three years. Body mass and stature were measured as anthropometric indicators and were used to estimate maturity status through Moore’s method. Cardiorespiratory fitness was evaluated through 9-minute running test. VO2peak was derived from an incremental progressive maximal protocol using a motorized treadmill. Anthropometry included body mass, stature and skinfolds. Measurements were performed by a single trained observer. Sexual maturation was assessed as self-reported stage of pubic hair (PH) development. Static allometric models were explored as an alternative to physiological output per unit of size descriptors. Allometry also considered chronological age and sexual maturation as dummy variable (PH2 vs. PH3 and PH3 vs. PH4).ResultsScaling coefficients for stature, body mass and fat-free mass were 1.463 (95%CI: 0.476 to 2.449), 0.516 (95%CI: 0.367 to 0.666) and 0.723 (95%CI: 0.494 to 0.951), respectively. The inclusion of sexual maturation increased explained variance for VO2peak (55% for PH2 vs. PH3 and 47% for PH3 vs. PH4). The intermittent nature of match performance in youth soccer supports relevance of ability to repeatedly produce high-intensity actions with short recovery periods. This study was aimed to examine the reproducibility of a repeated dribbling ability protocol and, additionally, to estimate the contribution of concurrent tests to explain inter-individual variability in repeated dribbling output. The total sample comprised 98 players who were assessed as two independent samples: 31 players were assessed twice to examine reliability of the protocol; and 67 juveniles aged 16.1 ± 0.6 years were compared by the competitive level (local, n = 34; national, n = 33) to examine construct validity. All single measurements appeared to be reasonably reliable: total (ICC = 0.924; 95%CI: 0.841 to 0.963); ideal (ICC = 0.913; 95%CI: 0.820 to 0.958); worst (ICC = 0.813; 95%CI: 0.611 to 0.910). In addition, the percentage of the coefficient of variation was below the critical value of 5% for total (%CV = 3.84; TEM = 2.51 s); ideal (%CV = 3.90, TEM = 2.48 s). Comparisons between local and national players suggested magnitude effects as follows: moderate (d-value ranged from 0.63 to 0.89) for all repeated sprint ability scores; large for total (d = 1.87), ideal (d = 1.72), worst (d = 1.28) and moderate for composite scores: the fatigue index (d = 0.69) and the decrement score (d = 0.67). In summary, the dribbling protocol presented reasonable reproducibility properties and output extracted from the protocol seemed to be independent from biological maturation.

Objective: The present study aimed to: 1) examine the association of biological maturation effect on performance at a motor coordination battery and 2) to assess whether the association between biological maturation and scores obtained in motor coordination tests is mediated by some anthropometric measurement. Methods: The convenience sample consisted of 73 male children aged 8 years old. Anthropometric data considered the height, body mass, sitting height, waist circumference, body mass index, fat mass and fat-free mass estimates. Biological maturation was assessed by the percentage of the predicted mature stature. Motor coordination was tested by the Körperkoordinationstest für Kinder. A partial correlation between anthropometric measurements, z-score of maturation and the motor coordination tests were performed, controlling for chronological age. Finally, causal mediation analysis was performed. Results: Height, body mass, waist circumference and fat mass showed a slight to moderate inverse correlation with motor coordination. Biological maturation was significantly associated with the balance test with backward walking (r=-0.34). Total mediation of the waist circumference was identified in the association between biological maturation and balance test with backward walking (77%). Conclusions: We identified an association between biological maturation and KTK test performance in male children and also verified that there is mediation of waist circumference. It is recommended that studies be carried out with female individuals and at other age ranges. A antropometria considerou a estatura, massa corporal, altura sentado, o perímetro de cintura, índice de massa corporal, as estimativas de massa gorda e massa livre de gordura. A maturação biológica foi avaliada pelo percentual da estatura matura predita. A coordenação motora foi testada pela bateria Körperkoordinationstest für Kinder. Foi feita a correlação parcial entre as medidas antropométricas, z-escore da maturação e as provas de coordenação motora, com controle para idade cronológica. Por último, a análise de mediação causal foi feita. Resultados: Estatura, massa corporal, perímetro de cintura e massa gorda apresentaram correlação inversa de magnitude pequena a moderada com as provas de coordenação motora. A maturação biológica associou-se significativamente à prova de equilíbrio em marcha à retaguarda (r=-0,34). Foi identificada mediação total do perímetro de cintura na relação do estado maturacional com o equilíbrio em marcha à retaguarda (77%). Conclusões: Foi possível identificar a associação entre o estado físico maturacional e o desempenho em uma prova de bateria de coordenação motora em crianças do sexo masculino e, ainda, afirmar que há mediação do perímetro de cintura. Recomenda-se que estudos sejam feitos com indivíduos de outras idades e do sexo feminino.

ResumoObjetivoO presente estudo teve como objetivos: 1) analisar a associação do estado maturacional com o desempenho nas provas de coordenação motora em crianças e 2) examinar se a relação entre o estado maturacional e o desempenho no KTK é mediada por alguma medida antropométrica.MétodosA amostra de conveniência foi composta por 73 crianças do sexo masculino com 8 anos. A antropometria considerou a estatura, massa corporal, altura sentado, o perímetro de cintura, índice de massa corporal, as estimativas de massa gorda e massa livre de gordura. A maturação biológica foi avaliada pelo percentual da estatura matura predita. A coordenação motora foi testada pela bateria Körperkoordinationstest für Kinder. Foi feita a correlação parcial entre as medidas antropométricas, z‐escore da maturação e as provas de coordenação motora, com controle para idade cronológica. Anthropometric data considered the height, body mass, sitting height, waist circumference, body mass index, fat mass and fat‐free mass estimates. Biological maturation was assessed by the percentage of the predicted mature stature. Motor coordination was tested by the Körperkoordinationstest für Kinder. A partial correlation between anthropometric measurements, z‐score of maturation and the motor coordination tests were performed, controlling for chronological age. Finally, causal mediation analysis was performed.ResultsHeight, body mass, waist circumference and fat mass showed a slight to moderate inverse correlation with motor coordination. Biological maturation was significantly associated with the balance test with backward walking (r=−0.34). Total mediation of the waist circumference was identified in the association between biological maturation and balance test with backward walking (77%).ConclusionsWe identified an association between biological maturation and KTK test performance in male children and also verified that there is mediation of waist circumference. It is recommended that studies be carried out with female individuals and at other age ranges.

Background and Objective: Respiratory gating training is a common technique to increase patient proprioception, with the goal of (e.g.) minimizing the effects of organ motion during radiotherapy. In this work, we devise a system based on autoencoders for classification of regular, apnea and unconstrained breathing patterns (i.e. multiclass). Methods: Our approach is based on morphological analysis of the respiratory signals, using an autoencoder trained on regular breathing. The correlation between the input and output of the autoencoder is used to train and test several classifiers in order to select the best. Our approach is evaluated in a novel real-world respiratory gating biofeedback training dataset and on the Apnea-ECG reference dataset. Results: Accuracies of 95 ± 3.5% and 87 ± 6.6% were obtained for two different datasets, in the classification of breathing and apnea. These results suggest the viability of a generalised model to characterise the breathing patterns under study. Conclusions: Using autoencoders to learn respiratory gating training patterns allows a data-driven approach to feature extraction, by focusing only on the signal’s morphology. The proposed system is prone to be used in real-time and could potentially be transferred to other domains.

The aim of this study was to review the evolutionary tendencies of research regarding to the study of male Rink-Hockey players´ and game performance. A systematic search was conducted in PubMed, Web of Knowledge and Scopus databases according to PRISMA method. The initial search identified 815 titles, resulting in 19 articles being included within the review. Original papers (English language) contained relevant data regarding rink hockey players’ performance or morphological/physiological demands, anthropometry/body composition characteristics were eligible. Studies were classified into categories: (1) Physiological Demands, (2) Anthropometry and Body Composition, (3) Game Characterization/Patterns, (4) Injuries. Results indicated that Rink hockey requires high intensity effort which demands both short and long duration efforts requirements from players. Body composition analysis shows to be an important monitoring tool which complements the understanding of the athlete’s cardiac adaptation. Game patterns shows a combination of specific game momentums with different outcomes according to the game zone. The intense short-term movements, collision and contact between players, in addition to the weight and speed of the hard ball and the stick, can considerably increase the risk of moderate and severe injuries. Lack of literature in Rink-Hockey is remarkable, and research is mainly focused on children and adolescents’ players. Furthermore, the existing research with adult elite athletes was assessed with a small sample size.

Persistent inflammation in end-stage renal disease (ESRD) patients is known to underlie the progression of chronic kidney disease and to be associated with multiple risk factors including malnutrition, atherosclerosis, and cardiovascular disease (CVD). The acute-phase protein pentraxin 3 (PTX3) has a proven potential as a local inflammatory biomarker, but its clinical utility in ESRD remains unclear. Circulating levels of PTX3 and classical inflammatory mediators, including the clinical prototypical C-reactive protein (CRP), were assessed in 246 ESRD patients on dialysis and analysed in relation to the lipid profile, adipokine levels, and nutritional, cardiac, and renal fibrosis markers. Occurrence of deaths was recorded for the following year. Contrarily to the classical inflammatory markers, PTX3 levels were negatively correlated with nutritional markers and associated with a less atherogenic lipid profile. Levels of the cardiac and renal fibrosis markers and of the oxidized LDL/LDL-C ratio were found to be independent determinants of PTX3 concentration. When comparing inflammatory mediators, the increase in the PTX3 levels was the only predictor of all-cause mortality in dialysis patients in a survival model adjusted to all markers under study, other than the inflammatory ones, besides common confounding factors in dialysis. Data support the clinical applicability of PTX3 as a broader inflammatory biomarker than the classical ones, presenting a close association with inflammation, malnutrition, CVD, and renal fibrosis and a great potential to predict all-cause mortality in dialysis patients. The pleiotropic character of PTX3 may be of clinical relevance, and it could be targeted to ameliorate the high morbidity and mortality associated with ESRD.

This study aimed to examine longitudinal changes in isometric strength of the knee extensors (ImKE) and knee flexors (ImKF) at 30° and 60°. The sample was composed of 67 players aged 11.0–13.9 years at baseline over five years. Stature, body mass, skinfolds, and isometric strength (ImKE30°, ImKF30°, ImKE60° and ImKF60°) were measured. Fat mass and fat-free mass (FFM) were derived from skinfolds. Skeletal age was obtained using TW2 RUS. Multilevel random effects regression analyses extracted developmental polynomial models. An annual increment on chronological age (CA) corresponded to 5.6 N (ImKE30°), 2.7 N (ImKF30°), 4.6 N (ImKE60°) and 1.5 N (ImKF60°). An increment of 1 kg in FFM predicted isometric strength as follows: 1.2 N (ImKE30°), 2.1 N (ImKF30°), 3.1 N (ImKE60°) and 2.0 N (ImKF60°). The following equations were obtained: ImKE30°=5.759×CA+1.163×FFM; ImKF30°=−19.369+2.691×CA+0.693×CA2+2.108×FFM; ImKE60°=4.553×CA+3.134×FFM; and, ImKF60°=-19.669+1.544×CA+2.033×FFM. Although skeletal maturity had a negligible effect on dependent variables, age and body size, based on FFM, were relevant longitudinal predictors. During adolescence, systematic assessment of knee extensors and knee flexors are strongly recommended to prevent impairment of knee muscle groups.

Year-to-year changes in sprinting in youth tennis players were examined in a mixed-longitudinal study (256 male players, aged 10–15 years: 993 measurements). Height (h), body mass (BM), lower limb explosive strength (LLES), and a 5-m sprint were measured over five years. During that period, players were classified as elite or sub-elite. To account for the repeated measurements within the individual nature of longitudinal data, multilevel random effects regression analyses were used. Sprint performance improved with age at each additional 1 year of age, thus predicting ∼.016 sec improvement in five-meter sprint time by all variables of the model. It was possible to predict the performance of elite tennis players in the 5-m sprint (sec) for elite players (1.1493 – (0.0159 ċ centered age) – (0.009 ċ BM) – (0.044 ċ LLES) and sub-elite players (1.1493 – (0.0159 ċ centered age) + 0.0135 – (0.009 ċ BM) – (0.044 ċ LLES) – (0.0557 ċ centered age). Sprint performance differences between elite and sub-elite players was related to longitudinal changes in body size and lower limb strength up until age 13.

This study aimed to predict motor coordination from a matrix of biocultural factors for 173 children (89 boys, 84 girls) aged 7–9 years who were assessed with the Körperkoordinationtest für Kinder test battery. Socioeconomic variables included built environment, area of residence, mother’s educational level, and mother’s physical activity level (using the International Physical Activity Questionnaire [short version]). Interrelationships among skeletal maturity status, body size, ventilator thresholds (VT) and peak oxygen uptake (VO2peak) were considered in 47 adolescent male soccer players aged 12.5–15.4 years. Body mass, stature, and the triceps and subscapular skinfolds were measured. The latter were used to estimate fat mass and fat-free mass. Skeletal age was assessed with the Fels method. VO2peak and VO2 at the first (VT1) and second (VT2) ventilatory thresholds were determined during an incremental maximal exercise test on a motorized treadmill. Ratio standards and allometric models were used in the analysis. Scaling exponents suggested linearity for all combinations between size descriptors and physiological variables, except between log-transformed values of VT1 and body mass (mL·kg-0.801·min, 95%CI: 0.649 to 0.952). Early maturing players attained greater values than players classified as “on-time” in skeletal maturity for the three ventilatory parameters expressed in absolute terms (d ranged from 0.65 to 0.71). The differences were attenuated after normalizing for mass descriptors using ratio standards and scaled variables (d ranged from 0.00 to 0.31). The results suggested significant variability between maturity groups when moving from VT1 to maximal metabolic conditions expressed by unit of stature (VT1: t = -2.413, p = 0.02, d = 0.60; VT2: t = -2.488, p = 0.02, d = 0.65; VO2peak: t = -2.475, p = 0.02, d = 0.65). Skeletal maturity status and associated variation in overall body size affects VT1, VT2 and VO2peak. The observed scaling of ventilatory outputs for body size may be related to the better running economy and smaller body size of average maturing athletes.

The current study aimed to examine the reproducibility of estimated peak power and estimated pedal velocity in a multi-trial 10-s all-out cycling test among adult athletes (n = 22; aged 23.50±4.73 years). Stature, sitting height and body mass were measured. Leg length was estimated as stature minus sitting height. Body volume was obtained from air displacement plethysmography and was subsequently used to calculate body density. Fat mass and fat-free mass were derived. The short-term power outputs were assessed from the force-velocity test (FVT), using a friction-braked ergometer on two separated occasions. Differences between repeated measurements were examined with paired t-test and effect sizes calculated. No significant differences were found between session 1 (898 W, 142 rpm) and session 2 (906 W, 142 rpm). Test-retest procedure showed acceptable reliability for estimated peak power output [technical error of measurement (TEM) = 31.9 W; % coefficient of variation (CV) = 3.5; intra-class correlation coefficient (ICC) = 0.986] and pedal velocity (TEM = 5.4 rpm, %CV = 3.8, ICC = 0.924). The current study demonstrated a reasonable reproducibility of estimated peak power and pedal velocity outputs in non-elite male athletes and supports that a familiarization session including a complete FVT protocol is not required.

The current study aimed to examine the reliability of the conventional and functional ratios derived from peak torques (PTs) and those obtained from the combination of knee flexors torque at the angle of knee extensors PT. Twenty-six male athletes (mean of 24.0±0.7 years) from different sports completed a test-to-test variation in isokinetic strength (Biodex, System 3) within a period of one week. Anthropometry and body composition assessed by Dual Energy X-ray Absorptiometry were also measured. The proposed isokinetic strength ratio measurements appeared to be highly reliable: conventional ratio at PT angle (intra-class correlation, ICC = 0.98; 95% confidence interval; 95%CI: 0.95 to 0.99); functional extension ratio at PT angle (ICC = 0.98; 95%CI: 0.96 to 0.99); and, functional flexion ratio at PT angle (ICC = 0.95; 95%CI: 0.89 to 0.98). Technical error of measurement (TEM) and associated percentage of the coefficient of variation (%CV) were as follows: conventional ratio at PT angle (TEM = 0.02; %CV = 4.1); functional extension ratio at PT angle (TEM = 0.02; %CV = 3.8); and, functional flexion ratio at PT angle (TEM = 0.03; %CV = 3.6). The current study demonstrated that the traditional and new obtained simple and combined isokinetic indicators seem highly reliable to assess muscle strength and function in adult male athletes. A single testing session seems to be sufficiently to obtain these isokinetic strength indicators.

The final sample included 49 boys (12.3 ± 0.8 years). V[Combining Dot Above]O2peak was derived from an incremental progressive maximal protocol using a motorized treadmill. In addition, maximal oxygen uptake was estimated from a 20-m shuttle run test. Static allometric models were obtained as an alternative to performance output per unit of size descriptors. Weight status had a significant effect on V[Combining Dot Above]O2peak using simple ratio standards per unit of body mass (BM) with adolescents classified as overweight and obese (OWOB) attaining lower values of V[Combining Dot Above]O2peak. A similar trend was noted for the allometric models adopting body mass (ml·kgBM·min), stature (L·m·min), and fat-free mass (FFM; ml·kgFFM·min). Findings also suggest the influence and interaction of pubertal and weight status on absolute values of V[Combining Dot Above]O2peak. Considering the data obtained, linear equations to estimate V[Combining Dot Above]O2peak from the 20-m shuttle run test should not be applied to boys who are OWOB because it will produce inaccurate assessments of cardiorespiratory fitness and penalize those who are heavier. Equations for V[Combining Dot Above]O2peak prediction need to be specific for pubertal status and preferably consider FFM as a body size descriptor.

Background Exploring the osteogenic effect of different bone-loading sports is particular relevant to understand the interaction between skeletal muscle and bone health during growth. This study aimed to compare total and regional bone and soft-tissue composition between female adolescent swimmers (n=20, 15.71±0.93 years) and volleyball players (n=26, 16.20±0.77 years). Methods Dietary intake was obtained using food frequency questionnaires. Body size was given by stature, sitting height, and body mass. Six skinfolds were measured. Bone mineral content (BMC) and density (BMD), lean soft tissue, and fat tissue were assessed using dual-energy X-ray absorptiometry. Pearson’s product moment correlation coefficients were calculated to examine the relationships among variables, by type of sport. Comparisons between swimmers and volleyball players were performed using student t-tests for independent samples and multivariate analysis of covariance (controlling for age, training history and body size). Results Swimmers (BMC: 2328±338 g) and volleyball players (BMC: 2656±470 g) exceeded respectively by 2.1 and 2.8 standard deviation scores the average of international standards for whole body BMC of healthy adolescents. SUMMARY OBJECTIVE: The current study was aimed to examine intra-individual variation on indicators of bone health in addition to whole-body plus appendicular tissue measurements using two concurrent assessments based on pencil beam and fan beam dual energy X-ray absorptiometry (DXA) systems in adult athletes from several sports. METHOD: Thirty-two male participants (27.6±10.1 years) were measured on anthropometry including multifrequency bioelectric impedance and air-displacement plethysmography. Bone mineral content (BMC), bone area, fat and lean soft tissue were derived using pencil beam (Lunar DPX-MD+) and fan beam (Lunar iDXA) absorptiometry. Bone mineral density (BMD) was obtained for the femoral neck, trochanter and triangle of ward. Finally, the right thigh was defined as a region of interest (ROI). Analyses comprised intra-class correlation (ICC), Effect size (d) from mean differences of repeated measurements, coefficient of variation (CV) RESULTS: ICC were >0.900 for all measurements. Intra-individual differences were large for BMC (d=1,312; CV=2,7%), bone area (d=1,761; CV=2,7%), fat tissue (d=1,612; CV=11%) and all indicators of appendicular lean soft tissue (d=1,237-1687; CV=2,0-4,1%). A very large difference (d=4,014; CV=8.4%) was diagnosed for lean soft tissue of the ROI. CONCLUSION: Although differences among concurrent instruments for BMC and bone area, the effect size of mean differences was negligible for BMD. Fat and lean soft tissue derived from DXA should be interpreted as reference values (not criterion) due to equipment-related variation, more apparently in the ROI values. RESUMO OBJETIVO: O presente estudo examinou a concordância entre os indicadores de saúde óssea e composição tecidual resultantes da aplicação de equipamentos concorrentes de absorciometria de raios X de dupla energia (DXA). MÉTODO: A amostra (n = 32), com 27,6 ± 10,1 anos de idade avaliados antropometricamente, inclui impedância bioelétrica com multifrequência e pletismografia de ar deslocado. O conteúdo mineral ósseo (CMO), a área de tecido ósseo, o tecido magro e o tecido gordo de corpo inteiro foram obtidos considerando o modo pencil beam (Lunar DPX-MD+) e o fan beam (Lunar iDXA). Para cada um dos equipamentos, foi efetuado um scanner proximal do fêmur, sendo produzida informação sobre a densidade mineral óssea (DMO) do colo, nomeadamente triângulo de Ward, trocanter e haste. Na fase de processamento, foi definida uma região de interesse (ROI; coxa direita). As análises compreenderam a diferença de médias de medidas repetidas com cálculo da magnitude de efeitos (d), coeficiente de correlação intraclasse (CCI), coeficiente de variação (CV). RESULTADOS: Foram obtidos CCI>0,900 para todas as medidas, com diferenças intraindividuais largas apenas para CMO (d = 1,312; CV = 2,7%), área de tecido ósseo (d = 1,761; CV = 2,7%), tecido gordo total (d = 1,612; CV = 11%) e tecido magro em todos os segmentos (d = 1,237-1,687; CV = 2,0-41%). A massa magra da ROI apresentou uma variaçāo intraindividual muito larga (d = 4,014; CV = 8,4%). CONCLUSÃO: Foram encontradas diferenças negligenciáveis para a DMO de corpo todo. As medidas de massa gorda e massa magra obtidas por DXA nāo devem ser tidas como critério, mas antes como referenda, muito especialmente quando se delimita uma ROI.

We report a measles outbreak in two Portuguese health regions (Algarve and Lisbon and the Tagus Valley) since February 2017, and which by 31 May resulted in 28 confirmed cases, of which 16 were unvaccinated. Thirteen cases were healthcare workers. One unvaccinated teenager died. Genotype B3 was identified in 14 cases from both regions. Synthetic cathinones (β-keto amphetamines) act as potent CNS stimulants similarly to classical amphetamines, which raise concerns about their potential neurotoxic effects. The present in vitro study aimed to explore and compare the mechanisms underlying the neurotoxicity of two commonly abused cathinone derivatives, 3,4-methylenedioxymethcathinone (methylone) and 3,4-methylenedioxypyrovalerone (MDPV), with those of 3,4-methylenedioxymethamphetamine (MDMA), using undifferentiated and differentiated SH-SY5Y cells. Following a 24 h exposure period, methylone and MDPV induced loss of cell viability in a concentration-dependent manner, in the following order of potency: MDPV ≈ MDMA > methylone. Dopaminergic differentiated cells evidenced higher sensitivity to the neurotoxic effects of both cathinones and MDMA than the undifferentiated ones, but this effect was not inhibited by the DAT inhibitor GBR 12909. Intracellular oxidative stress mediated by methylone and MDPV was demonstrated by the increase in reactive oxygen and nitrogen species (ROS and RNS) production, depletion of intracellular reduced glutathione and increased oxidized glutathione levels. All three drugs elicited mitochondrial impairment, characterized by the mitochondrial membrane potential (Δψm) dissipation and intracellular ATP depletion. Apoptosis was found to be a common mechanism of cell death induced by methylone and MDPV, with evident chromatin condensation and formation of pyknotic nuclei, and activation of caspases 3, 8, and 9. In conclusion, the present data shows that oxidative stress and mitochondrial dysfunction play a role in cathinones-induced neuronal damage, ultimately leading to cell death by apoptosis.
