Fatty acids of forty-two samples of perirenal fat of goat kids reared on three different feeding systems: goat milk (B), milk replacer (R) and milk-based starter (F) have been analyzed by Gas Chromatography flame ionization detector. The lipids were extracted by melting of perirenal fat in a microwave oven. The fat was then filtered and dissolved in hexane. This analysis was performed on a column (100 m x 0.25 mm i.d. and 0.25 microm film thickness) coated with a polar stationary phase HP-88 and flame ionization detector was used. Hydrogen (25 psi inlet constant pressure) was used as carrier gas. Programmed temperature was kept at 175 degrees C and held isothermally for 10 min, and was then raised to 205 degrees C at a rate of 3 degrees C/min and held isothermally for 10 min. By using the fatty acids as chemical descriptors, pattern recognition techniques were applied to differentiate between goat milk, milk replacer and milk-based starter fattening diet of goat kid. C18:2 and C18:3 acids were found to be the most differentiating variables.

Alpine marmots (Marmota marmota) were maintained on a laboratory diet, and the fatty acid composition of gonadal and subcutaneous white adipose tissues (WAT) was studied during a yearly cycle. Fatty acids (FA) released from isolated adipocytes were also identified after stimulation of in vitro lipolysis. Analysis of the FA composition of WAT depots showed that marmot WAT mainly contained monounsaturated FA (65%, mostly oleic acid, 18:1n-9) although laboratory food contained 45% of linoleic acid (18:2n-6) and only 21% of 18:1n-9. During stimulated lipolysis, saturated FA were preferentially released from isolated adipocytes whereas unsaturated FAs were retained. Despite this selective release of FA from isolated WAT cells in vitro, and despite the FA composition of the food, marmots maintained a constant FA composition in both WAT depots throughout the year. Six months of hibernation and fasting as well as an intense feeding period did not affect this composition. The potential adaptive benefit of such regulation of WAT composition, based on a high level of monounsaturated FA, might be to maintain fat with appropriate physical properties allowing animals to accommodate to and survive the wide range of body temperatures experienced during hibernation.

The effect of chronic heat exposure on carcass quality of broilers: proportion of lean and fat tissues, fat content, and fatty acid composition, was investigated. One hundred and eight 4-wk-old male chickens were brooded in individual battery cages in two controlled-environment rooms at constant ambient temperature (22 or 32 C) until 7 wk of age. They were equally distributed into three treatments: 22 C, ad libitum feeding (22AL); 32 C, ad libitum feeding (32AL); and 22 C, pair-feeding on the daily feed intake of heat-exposed chickens (22PF). At 7 wk of age, heat-exposed chickens (32AL) had a lower body weight gain than the other birds: -47% compared to 22AL and -31% compared to 22PF. At 32 C, broilers exhibited a lower breast to body weight proportion: 11.9 vs 13.4% for 22AL. Abdominal, subcutaneous, and intermuscular fat deposits were enhanced in hot conditions, respectively, 15, 21, and 22% compared to 22AL and 58, 64, and 33% compared to 22PF. However, lipid contents of abdominal, subcutaneous, intermuscular, and intramuscular tissues were not affected by heat exposure but were significantly reduced in the 22PF birds. In heat-exposed birds, although saturated fatty acid proportions, particularly palmitic acid (C16:0), were increased, unsaturated fatty acids as a percentage of total fatty acids were decreased, especially oleic (C18:1) and linoleic (C18:2) acids in fat tissues. Consequently, under ad libitum feeding conditions, heat exposure significantly decreased the unsaturated to saturated fatty acid ratio in the abdominal and subcutaneous fat tissues, but not in intermuscular and intramuscular fats.

trans isomers of unsaturated fatty acids are formed by biological or industrial hydrogenation. A population case-control study of sudden cardiac death in mean was done to test the hypothesis that trans isomers of oleic acid and linoleic acid increase the risk of sudden cardiac death due to coronary artery disease. In adipose tissue obtained at necropsy from 66 cases of sudden cardiac death and taken from 286 healthy age and sex matched controls, the proportions of trans isomers of oleic and linoleic acid were measured by gas-liquid chromatography. In cases, the mean (SE) percentage of total trans fatty acids (C18:1 plus C18:2), expressed as a proportion of all fatty acids, was significantly lower (2.68 [0.08]%) than in healthy controls (2.86 [0.04]%; p < 0.05). trans C18:1 was 2.1 (0.7)% in cases compared with 2.27 (0.04)% (p < 0.05) in controls. The proportion of all trans isomers of linoleic acid was 0.58 (0.02)% in cases compared with 0.59 (0.01)% in controls (p = 0.98). The estimated relative risk for sudden cardiac death of trans C18:1 and C18:2 fatty acids combined did not differ significantly from 1.0 in relation to the distribution of these trans isomers by quintile in the control population. The relative risk (95% CI) of sudden cardiac death in the top quintile was 0.40 (0.15-1.02) for C18:1 and 1.08 (0.48-2.74) for C18:2 compared with the bottom quintiles of their respective control distributions. When these univariate relations for trans fatty acids were adjusted for coronary risk factors, smoking was the only factor that remained independently associated with risk of sudden cardiac death (2.27 [1.23-4.17]). Overall, there was no evidence of a relation between trans isomers of oleic and linoleic acids combined and sudden cardiac death. However, trans oleic acid was negatively associated with risk of sudden cardiac death, whereas no association with trans forms of linoleic acid was seen. This study does not support the hypothesis that trans isomers increase the risk of sudden cardiac death.

The effect of ethanol feeding on the essential fatty acid content of tissues has been contradictory. To define the effect, we analyzed fatty acid profiles in various tissues from five miniature pigs fed daily 105 kJ basal diet/kg body wt and 146 kJ ethanol/kg body wt, and also five control pigs pair-fed the same amount of basal diet but with corn starch substituted for ethanol. After 12 mo, biopsy samples were taken, and tissue fatty acid profiles were analyzed. In the phospholipid fraction from the ethanol group there was a uniform decrease in arachidonic acid (AA) and an increase in oleic acid in liver, serum, and muscle. AA was consistently decreased in the triglyceride fractions of liver, serum and subcutaneous adipose of the ethanol group. Possible explanations for this general reduction in tissue AA with ethanol feeding include decreased activities of delta 6 and delta 5 desaturases, and a displacement of AA from lipid fractions by other fatty acids.

Three grazing experiments were conducted to determine the effect of level of endophyte infection, rate of N fertilization of tall fescue grass, grazing period, and paddock exchange on selected chemical properties of four bovine carcass tissues. Samples of semitendinosus and longissimus muscle and of subcutaneous and perinephric adipose tissue were excised from the left side of each carcass. In Exp. 1, percentage of stearic acid was higher (P less than .05) and percentages of palmitoleic and oleic acid were lower (P less than .05) in all tissues from steers grazed on LELN Au-Triumph fescue than in tissues from steers grazed on LELN KY-31 fescue. Grazing periods of 175 or 245 d or paddock exchange (Exp. 2) had no significant effect on proximate composition of semitendinosus and longissimus muscles; however, moisture content was higher (P less than .05) in the semitendinosus muscle from steers grazed on 100% (100E) endophyte-infected KY-31 fescue. Forage treatment, grazing period, and paddock exchange (Exp. 2 and 3) had no significant effect on percentage of saturated fatty acids in the subcutaneous, semitendinosus, and longissimus tissue samples. In Exp. 2 and 3, percentages of saturated fatty acids were higher (P less than .05) in subcutaneous and perinephric adipose tissue samples from steers grazed on 100E than in samples from steers grazed on zero endophyte (OE) fescue.(ABSTRACT TRUNCATED AT 250 WORDS)

Milk fat octadecenoic acids were isolated by preparative gas-liquid chromatography of their methyl esters and separated into cis and trans isomers by argentation thin-layer chromatography. The distribution of the double bonds in the two fractions was determined by reductive ozonolysis. Cis octadecenoic acids had double bonds in positions 6 to 14. The cis-9 isomer was the most abundant isomer representing approximately 95% of the total. Trans octadecenoic acids had double bonds in positions 6 to 16, the predominant isomer being trans-11 octadecenoic acid. Data illustrating the range of variation in the distribution of positional isomers of octadecenoic acid for herd milk fat and butter are presented. There was no obvious seasonal variation in the distribution pattern. Fat from perirenal and subcutaneous adipose tissue of a dairy cow had composition similar to that of butter samples.

The fatty acid composition of whole bone marrow and that of isolated, disaggregated adipose cells from red and yellow marrow was examined by gas chromatography. Consistent and significant shifts from myristic and palmatic acids (in red marrow) to their respective monounsaturated derivatives myristoleic and palmitoleic acids (in yellow marrow) were found. These differences in the fatty acids correlate with histochemical studies and lend further support to the concept that the composition of lipid in the adipose cells of bone marrow may determine their relative stability in relation to haematopoietic requirements.

To reexamine the role of albumin in cellular uptake of long chain fatty acids, we measured [3H]oleate uptake by isolated hepatocytes, adipocytes, and cardiac myocytes from incubations containing oleate/albumin complexes at molar ratios from 0.01:1 to 2:1. For each ratio the uptake was studied over a wide range of albumin concentrations. In all three cell types and at any given oleate/albumin ratio, the uptake appeared saturable with increasing concentrations of oleate:albumin complexes despite the fact that the unbound oleate concentration for each molar ratio is essentially constant. However, the "Km" but not the "Vmax" of these pseudosaturation curves was influenced by substrate availability. At low albumin concentrations, uptake velocities did not correlate with unbound oleate concentrations. However, observed and expected uptake velocities coincided at albumin concentrations approaching physiologic levels and were a saturable function of the oleate/albumin ratios and the consequent unbound oleate concentrations employed. Hence, under the experimental conditions employed in this study using a variety of suspended cell types, oleate uptake kinetics were consistent with the conventional theory at physiologic concentrations of albumin.
