Simple Summary: The role of mosquito eggs in disease transmission is not well understood, despite reports of virus vertical transmission and their ability to survive dry periods in a dormant stage. Additionally, field evidence shows that new generations of mosquitoes often reappear weeks or months later in certain city areas. This may be because both species follow an "R strategy" reproduction pattern, laying many eggs to ensure survival in a harsh environment. This study was conducted in southern Mexico in 2023 and focused on the egg-laying activity of Ae. aegypti and Ae. albopictus mosquitoes during the dry inter-epidemic season. Ovitraps were used in the backyards of the houses to identify egg species, and further larval containers were surveyed. Most of the larval habitats were containers smaller than 1 L, commonly not treated as they were considered trash. Even though homeowners volunteer to remove them during government cleaning community-based campaigns, city waste disposal trucks transporting these containers do not take any measures to destroy the tiny eggs. We conclude with the urgent need to develop an egg-killing treatment, which could significantly enhance vector reduction and achieve a total stage strategy within the Integrated Vector Control Programs. Although integrated management and control programs implement intense control measures for adult, pupal, larval, and breeding sites during outbreaks, there is a lack of studies to understand the role of the vector egg stage in disease dynamics. This study aimed to assess the dry season quiescent Aedes aegypti and Aedes albopictus egg populations in houses and backyards in Tapachula, southern Mexico. Two hundred and fifty ovitraps were placed in 125 homes in the Las Americas neighborhood. A total of 7290 eggs were collected from 211 (84.4%) ovitraps. Only 5667 (77.7%) hatched under insectary water immersion and food supply conditions, with 4031 (71.1%) identified as Ae. aegypti, and 1636 (28.8%) as Ae. albopictus, respectively. The remaining 1623 (22.3%) did not hatch due to Delayed Hatching and/or quiescence tropical stage. Eighty-three larval containers were sampled with desiccated eggs during the dry season; most of them were described as trash waste because larvicides are only used for larger containers of 5–10 L. Evolutionary characteristics for the two species including partial egg hatching, ambient-regulated quiescence, the ability of the embryo to survive for a more extended period intra-seasonally, the egg sticking to inner container walls, demands urgent operational research to achieve successful egg-proof larval container methods. To test whether Zika virus has adapted for more efficient transmission by Aedes aegypti mosquitoes, leading to recent urban outbreaks, we fed mosquitoes from Brazil, the Dominican Republic, and the United States artificial blood meals containing 1 of 3 Zika virus strains (Senegal, Cambodia, Mexico) and monitored infection, dissemination, and virus in saliva. Contrary to our hypothesis, Cambodia and Mexica strains were less infectious than the Senegal strain. Only mosquitoes from the Dominican Republic transmitted the Cambodia and Mexica strains. However, blood meals from viremic mice were more infectious than artificial blood meals of comparable doses; the Cambodia strain was not transmitted by mosquitoes from Brazil after artificial blood meals, whereas 61% transmission occurred after a murine blood meal (saliva titers up to 4 log 10 infectious units/collection). Although regional origins of vector populations and virus strain influence transmission efficiency, Ae. aegypti mosquitoes appear to be competent vectors of Zika virus in several regions of the Americas. Background:  After decades of obscurity, Zika virus (ZIKV) has spread through the Americas since 2015 accompanied by congenital microcephaly and Guillain-Barré syndrome. Although these epidemics presumably involve transmission by Aedes aegypti, no direct evidence of vector involvement has been reported, prompting speculation that other mosquitoes such as Culex quinquefasciatus could be involved.Methods:  We detected an outbreak of ZIKV infection in southern Mexico in late 2015. Sera from suspected ZIKV-infected patients were analyzed for viral RNA and antibodies. Mosquitoes were collected in and around patient homes and tested for ZIKV.Results:  Of 119 suspected ZIKV-infected patients, 25 (21%) were confirmed by RT-PCR of serum collected 1-8 days after the onset of signs and symptoms including rash, arthralgia, headache, pruritus, myalgia, and fever. Of 796 mosquitoes collected, A. aegypti yielded ZIKV detection by RT-PCR in 15 of 55 pools (27.3%). No ZIKV was detected in C. quinquefasciatus ZIKV sequences derived from sera and mosquitoes showed a monophyletic relationship suggestive of a point source introduction from Guatemala.Conclusions:  These results demonstrate the continued, rapid northward progression of ZIKV into North America with typically mild disease manifestations, and implicate A. aegypti for the first time as a principal vector in North America. Mosquitoes, especially Aedes aegypti, are becoming important models for studying invasion biology. We characterized genetic variation at 12 microsatellite loci in 79 populations of Ae. aegypti from 30 countries in six continents, and used them to infer historical and modern patterns of invasion. Our results support the two subspecies Ae. aegypti formosus and Ae. aegypti aegypti as genetically distinct units. Ae. aegypti aegypti populations outside Africa are derived from ancestral African populations and are monophyletic. The two subspecies co-occur in both East Africa (Kenya) and West Africa (Senegal). In rural/forest settings (Rabai District of Kenya), the two subspecies remain genetically distinct, whereas in urban settings, they introgress freely. Populations outside Africa are highly genetically structured likely due to a combination of recent founder effects, discrete discontinuous habitats and low migration rates. Ancestral populations in sub-Saharan Africa are less genetically structured, as are the populations in Asia. Introduction of Ae. aegypti to the New World coinciding with trans-Atlantic shipping in the 16th to 18th centuries was followed by its introduction to Asia in the late 19th century from the New World or from now extinct populations in the Mediterranean Basin. Aedes mascarensis is a genetically distinct sister species to Ae. aegypti s.l. This study provides a reference database of genetic diversity that can be used to determine the likely origin of new introductions that occur regularly for this invasive species. The genetic uniqueness of many populations and regions has important implications for attempts to control Ae. aegypti, especially for the methods using genetic modification of populations. Understanding the processes bywhich species colonize and adapt tohuman habitats is particularly important in the case of disease-vectoring arthropods. The mosquito species Aedes aegypti, a major vector of dengue and yellow fever viruses, probably originated as a wild, zoophilic species in sub-Saharan Africa, where some populations still breed in tree holes in forested habitats. Many populations of the species, however, have evolved to thrive in human habitats and to bite humans. This includes some populationswithin Africa aswell as almost all those outside Africa. It is not clearwhether all domestic populations are genetically related and represent a single 'domestication' event, orwhether associationwith human habitats has developed multiple times independently within the species. To test the hypotheses above, we screened 24worldwide population samples of Ae. aegypti at 12 polymorphic microsatellite loci. We identified two distinct genetic clusters: one included all domestic populations outside of Africa and the other included both domestic and forest populations within Africa. This suggests that human association in Africa occurred independently from that in domestic populations across the rest of the world. Additionally, measures of genetic diversity support Ae. aegypti in Africa as the ancestral form of the species. Individuals from domestic populations outside Africa can reliably be assigned back to their population of origin, which will help determine the origins of new introductions of Ae. aegypti. Background: Aedes aegypti is the main mosquito vector of the four serotypes of dengue virus (DENV). Previous population genetic and vector competence studies have demonstrated substantial genetic structure and major differences in the ability to transmit dengue viruses in Ae. aegypti populations in Mexico. Methodology/Principal Findings: Population genetic studies revealed that the intersection of the Neovolcanic axis (NVA) with the Gulf of Mexico coast in the state of Veracruz acts as a discrete barrier to gene flow among Ae. aegypti populations north and south of the NVA. The mosquito populations north and south of the NVA also differed in their vector competence (VC) for dengue serotype 2 virus (DENV2). The average VC rate for Ae. aegypti mosquitoes from populations from north of the NVA was 0.55; in contrast the average VC rate for mosquitoes from populations from south of the NVA was 0.20. Most of this variation was attributable to a midgut infection and escape barriers. In Ae. aegypti north of the NVA 21.5% failed to develop midgut infections and 30.3% of those with an infected midgut failed to develop a disseminated infection. In contrast, south of the NVA 45.2% failed to develop midgut infections and 62.8% of those with an infected midgut failed to develop a disseminated infection. Conclusions: Barriers to gene flow in vector populations may also impact the frequency of genes that condition continuous and epidemiologically relevant traits such as vector competence. Further studies are warranted to determine why the NVA is a barrier to gene flow and to determine whether the differences in vector competence seen north and south of the NVA are stable and epidemiologically significant. We have demonstrated that the intersection of the NVA with the coast in Veracruz state is a barrier to gene flow in Ae. aegypti. This was unexpected because the intersection of the NVA with the Pacific Coast is not a barrier to gene flow. Further studies to identify the actual mechanism(s) that is(are) contributing to the lack of gene flow will provide important information on the trafficking potential of Ae. aegypti, which will be of great value to Ae. aegypti control programs. There are significant differences in vector competence for dengue virus between mosquitoes north and south of the NVA, but the epidemiological significance of these finding remains to be determined. Future studies will determine if, for example, the genes that condition midgut infection and vector competence of Ae. aegypti populations provide biomarkers for risk of dengue transmission. Such biomarkers could be of great value to control programs in resource limited environments by allowing targeting of vector control efforts to areas at most risk for epidemic dengue and dengue hemorrhagic fever. Evidence of vertical transmission of dengue virus (DENV) in yellow fever mosquito, Aedes aegypti (L.), was found at Cancun, Quintana Roo, Mexico . Ae. aegypti larvae were collected from 729 containers in a total of 178 households between September and November 2012. Larvae collected were put into sterile bags and transported in thermoses. RT-PCR followed by semi-nested PCR for molecular serotyping was used to evaluate for dengue virus in larval pools. Of the 178 houses sampled, 79 (44.4%) were positive for at last one Ae. aegypti larva. The positive breeding sites were grouped into a total of 79 pools. Only three larval pools from three houses close together were positive for DENV-2. The infection rate was 0.32 per 1,000 mosquitos (95% CI 0.09-0.85 per 1,000 mosquitoes). The result of this report suggested that vertical transmission of dengue virus in Ae. aegypti might be a mechanism of viral perpetuation during inter-epidemic periods. [ABSTRACT FROM AUTHOR]

Monitoring and control programs for yellow fever mosquito, Aedes aegypti (L.), usually do not focus on the egg as a potential target for control. The egg is the most numerous life stage but is invisible to conventional inspection by a sticky pad that attaches it. This laboratory study evaluated the potential ovicidal effect of five commonly used plastics. Plastic liners in oviposition containers were exposed to gravid female mosquitoes in an insectary. The percentage of eggs that hatched was recorded. The plastic liners altered the places where eggs were laid, i.e., 27.0% were glued onto the plastic film, 70.0% remained floating, and 3.0% were submerged. Vinyl blocked most egg adhesion, with a mean of 7.05 ± 10.1 eggs, compared to 170.7 ± 68.6 eggs for the check. Pooled numbers of glued, floating, and submerged eggs showed fewest eggs hatched on vinyl or low-density polyethylene, resulting in the death of 94.7% of the embryos. Plastics waterproofing property might be blocking the hyaluronic acid, the component of the sticky substance of mosquito eggs. Results demonstrated the potential use of plastic strips as an ovicide. Traditionally the major risk environment for transmission of dengue virus has been assumed to be households. In Mexico, dengue outbreaks continue year after year despite intense control efforts. Nonresidential sites (public and private spaces) infested with Aedes aegypti (L.) were evaluated. In total, 141 nonresidential sites were sampled for the presence of potential and active oviposition sites and adult mosquitoes. Eighty percent of the sites were oviposition sites; Ae. aegypti adults were recovered at 94.7% of nonresidential sites. Most female Ae. aegypti, 21.6 and 10.4, were at schools and recreational sites, respectively. Chi-squared indicated no significant differences in the dengue vector to categories of sample sites (X² = 17.76, df = 9, P = 0.38). Indoor-use patterns of adult mosquitoes indicated bathrooms and classrooms were preferred resting sites. Reverse transcription polymerase chain reaction (RT-PCR) assay did not identify dengue virus nucleic acids from a group of 221 pools containing 1,521 female Ae. aegypti. In October 2011, the State Health Department announced that several laboratory-confirmed cases of dengue had occurred among residents in two neighborhoods of Benito Juarez, Quintana Roo State, Mexico. To identify the dengue virus serotype(s) temporally and spatially associated with the cases, entomologic-based virus surveillance was initiated in October 2011 in both neighborhoods. Adult mosquitoes were collected from 88 houses by CDCbackpack aspirator, and all female Aedes aegypti L. (n = 419) were individually homogenized and assayed in pools of as many as 10 by reverse transcriptionpolymerase chain reaction (RT-PCR) using dengue virus-specific primers. Five (12%) of 41 pools were positive for dengue virus RNA. The individual mosquitoes that comprised the pools were analyzed separately by RT-PCR using dengue virus serotype-specific primers. Six mosquitoes were positive for dengue virus serotype-2 (DENV-2) RNA, three of which were collected in the same house. The mean number of female Ae. aegypti collected in each house was 4.76 ± 6.19. The overall dengue virus-infection rate in female Ae. aegypti was 1.4%. Interestingly, most (60%) of mosquito females were collected only from 15 (17%) houses. Secondary dengue virus infections are a major risk for developing dengue hemorrhagic fever. Recent exposure to infectious bites of Aedes aegypti (L.) females in previously diagnosed dengue cases fulfills the epidemiological model of dengue hemorrhagic fever. A study was comprised of 357 (89.2%) dengue and 43 (10.8%) dengue hemorrhagic fever cases confirmed by laboratory tests and clinical manifestations. An entomological survey was done in homes and backyards. Concurrently, a questionnaire was used to assess the impact of healthpromotion campaigns through knowledge of the vector and its epidemiological role. Seventy-six (28.4%) of the 268 (67.0%) total wet or dry oviposition sites were positive for the presence of larvae or pupae, while adult Ae. aegypti were found in 32 (8.0%). One hundred thirty-two (33%) householders who formerly had dengue fever or dengue hemorrhagic fever had knowledge of either larval or adult dengue vector stages. According to gender distribution, 145 (36.2%) and 14 (3.5%) of the males confirmed with cases of dengue and dengue hemorrhagic fever lived in houses with 17.9 and 2% of the Ae. aegypti larval and pupal habitats. Houses with females who had dengue and dengue hemorrhagic fever were 212 (53%) and 29 (7.3%), with containers with immature Ae. aegypti in 19.4 and 7%, respectively.
