Wastewater carries different pathogenic and non-pathogenic microorganisms that can be dispersed in the surrounding environment. Workers who frequent sewage treatment plants can therefore be exposed to aerosols that contain a high concentration of potentially dangerous biological agents, or they can come into direct contact with contaminated material. This can lead to allergies, infections and occupational health-associated diseases. A characterization of biological risk assessment of bioaerosol exposure is necessary. The aim of this study was to evaluate the application of an interdisciplinary method that combines chemical and biological approaches for the analysis of a bioaerosol derived from a wastewater treatment plant (WWTP) situated in Italy. Sampled filters were analyzed by HPLC-MS/MS spectroscopy that searched for different chemical biomarkers of airborne microorganisms. The analytical quantification was compared to the biological cultural method that revealed an underrated microbial concentration. Furthermore, next generation sequencing analysis was used also to identify the uncultivable species that were not detected by the culture dependent-method. Moreover, the simple animal model Caenorhabditis elegans was used to evaluate the pathogenicity of two isolates- Acinetobacter iwoffii and Micrococcus luteus- that showed multidrug-resistance. This work represents a starting point for the development of a multidisciplinary approach for the validation of bioaerosol exposure on WWTP workplaces.

Airborne microorganisms, as bacteria and fungi, are ubiquitous components of the atmospheric aerosol particles. In this paper, we report a method for the simultaneous extraction, purification, separation, identification and quantification of ergosterol, mannitol and arabitol as biomarkers of fungal spores in bioaerosol particles. After sampling by a low volume sampler, filters were spiked with mannitol-(13)C and dehydrocholesterol as internal standards. Samples were then extracted by accelerated solvent extraction using pure ethanol. The extract was then passed through an amino cartridge and divided in two parts: the apolar fraction, released from the cartridge, was subjected to liquid liquid extraction (by n-hexane), while polar compounds, retained by the cartridge, were eluted by a mixture of methanol-water. The two fractions were joined and analyzed by HPLC equipped with two different columns in series, and coupled to a triple-quadrupole mass spectrometer with Atmospheric Pressure Chemical Ionization source. In addition, the same fractions were analyzed, after derivatization, by GC-MS. The results obtained by the two techniques were finally compared, showing good agreement between them. Last, the contents of the three biomarkers have been estimated in three atmospheric samples collected in a suburban/rural site and, using literature conversion factors, correlated to fungal biomass., (Copyright © 2012 Elsevier B.V. Both regulated and unregulated air pollutants were detected during an intensive seasonal sampling campaign in a mixed industrial/semi-rural area on the outskirts of Rome, Italy, at two sites located opposite a hospital waste incinerator, downwind according to the direction of the prevailing local winds. Concentrations of pollutants were significantly lower than in urban atmospheres. The composition of particulate organic material indicated a heavy biogenic impact, accompanied by a lower contribution from petroleum-related processes. Both PAH and nitro-PAH group compositions of particulates were used to assess the nature and relative importance of sources. Both sites showed that different and diffuse sources contributed to local pollution with a significant contribution from traffic, proving that the hospital waste incinerator was not the main pollution source in this area. Among unregulated compounds, a series of positional isomers of nitro-PAHs and other organic compounds associated with particulate matter were investigated. In particular, 1- and 3-nitrophenanthrene identification was carried out, and they proved to be the most abundant nitro-PAHs.

Potential harmful effects of pesticides include risks to human health of workers involved in the wet spray application in cultivated areas. Inhalation exposure depends on several factors including pesticide concentrations in the respirable fraction of airborne particulate matter (PM4). To ensure a high level of protection, the use of tractors with cabins provides protection against dust, aerosols, and vapors. Since tractors not providing maximum protection are still in use, PM4 was sampled during spreading operations in agricultural fields inside and outside tractor cabins. Sample preparation technique based on accelerated solvent extraction and solid-phase extraction cleanup was optimized before analysis of nine pesticides in PM4. Meptyldinocap, deltamethrin, myclobutanil, fluopyram, methoxyfenozide, dimethomorph, fluopicolide, cyflufenamid, and metrafenone were simultaneously determined by high-performance liquid chromatography–electrospray ionization–tandem mass spectrometry (HPLC–ESI–MS–MS). Abstract: Size-segregated atmospheric particles were collected in Rome, Italy, using a low-pressure impactor. Twelve sampling campaigns were conducted under different meteorological conditions over a whole year covering 155 not consecutive days. The samples were analyzed for polycyclic aromatic hydrocarbons (PAHs) and their nitro- and methyl- derivatives known for their toxicity. An assessment of the carcinogenic potency of the particles known to penetrate into lungs, liver, heart and nervous system was performed. The distribution of the classes of compounds was unimodal and centrated at 0.4 μm size fraction for PAHs and bimodal and centrated at 0.1 and 0.4 μm for methyl- and nitro- derivatives. The 18% of toxic organic compounds we analyzed was distributed into the ultrafine fraction (PM0.1) and 76% in the fine fraction; but substituted PAH distribution in the ultrafine particles shifted toward higher values during warm periods. In July, the 50% of the total nitro-PAHs was found in PM0.1 and an average of 42% of the total methyl-PAHs was found in the same fraction in summer and intermediate seasons. The aim of this study was to identify areas of potential relevant exposure to pollutants within Rome''s urban core. To meet this goal, intensive field campaigns were conducted and simulations were performed, using the flexible air quality regional model (FARM), to study winter and summer pollution episodes. The simulations were performed using a complete emission inventory that included traffic flow model results of the Roman street network to better describe, with respect to the available diffuse national emission inventory, the hourly variation of traffic emissions in the city. The meteorological reconstruction was performed by means of both prognostic and diagnostic models by using experimental data collected during the field campaigns. To evaluate the capability of the FARM model to capture the main features of the selected episodes, a comparison of modelled results against observed air quality data for different pollutants was performed at urban and rural sites. FARM performed well in predicting ozone (O3) and nitrogen dioxide (NO2) concentrations, showing a good reproduction of both daily peaks and their diurnal variations. The model also showed a good capability to reproduce the magnitude of volatile alkane, aromatic and carbonyl compound concentrations. PM10 model results revealed the tendency to under-predict the observed values. PM composition model results were compared with observed data, evidencing good results for elemental carbon (EC), nitrate (NO3 −) and ammonium (NH4 +), underestimation for sulphate (SO4 2−) and poor performance for organic matter (OM). The soil components of PM were found to be significantly under-predicted by the model, especially during Saharan dust episodes. Overall, the study results show large areas of high O3 and PM10 concentrations where levels of pollutants should be carefully monitored and population exposure evaluated. [Copyright &y& Elsevier]
